    High glutamate levels in the bone marrow of multiple myeloma patients promote osteoclast formation: a novel target for osteolytic bone disease

    Multiple Myeloma (MM) is a glutamine (Gln)-addicted cancer. Consequently, the MM bone microenvironment (BM) is characterized by lower Gln and higher glutamate (Glu) levels than those in pre-malignant monoclonal gammopathies. Such MM-dependent metabolic perturbation impairs osteoblast differentiation in the bone microenvironment, but its effect on osteoclast (OCL) bone resorption is still unknown. We first show that bone marrow mononuclear cells from MM patients release higher levels of Glu compared to those from patients with monoclonal gammopathy of undetermined significance (MGUS) or smoldering multiple myeloma (SMM). This increased Glu production correlates with elevated bone resorption activity. We then demonstrate that Glu stimulates OCL differentiation via the activation of NF-κB-NFATc1 pathway in low-Glu BM samples from pre-malignant patients but not in high-Glu samples of MM patients. Secondly, the early phase of OCL formation was associated with high Glu intracellular content and induction of the Glu transporter EAAT1. Consistently, the pharmacological inhibition of EAAT1 hinders OCL differentiation by blocking the RANKL-dependent signaling pathway and actin cytoskeleton reorganization. Overall, our data indicate that high Glu levels in MM bone marrow are involved in OCL formation, suggesting that targeting Glu transport may represent a novel approach for the prevention of osteolytic lesions in MM patients

    Public Administration and Agromafia: Administrative Law Profiles between Coordination, Prevention, and Control in the Agri-food Sector

    The phenomenon of “agromafias” – the infiltration of organized crime into the agri-food supply chain – represents one of the most insidious threats to economic legality and environmental protection. In this context, administrative law emerges as a key instrument not only in repressing illegality but, above all, in preventing it. This analysis highlights how the Italian administrative apparatus – spanning both central and local levels – is currently facing a dual challenge: effectively coordinating the actors involved in managing, monitoring, and planning the agri-food sector, while simultaneously ensuring transparency, legality, and sustainability in public policy. Tools such as anti-mafia interdiction orders, collaborative prevention measures, whitelists, legality protocols, and Minimum Environmental Criteria (MEC) applied to public food procurement demonstrate the anticipatory and strategic role that administrative law plays in combating agromafias. Preventive action is not limited to excluding colluding economic operators but extends to promoting sustainable practices, overseeing the use of EU funds (CAP), and integrating environmental regulations, food safety, and labor protection. The fight against agromafias requires multi-level, integrated governance based on the principles of subsidiarity, coordination, and precaution, with public administration playing a crucial role in shaping effective and resilient policies. Ultimately, administrative law – far from being a mere regulatory framework – acts as a dynamic tool for building legality and safeguarding the public interest in a sector vital to the country’s sustainable development

    Primary delusional experiences in schizophrenia and the sacred: a qualitative study

    Psychopathology has highlighted the pivotal role of revelation in schizophrenic primary delusions. However, empirical studies on the topic remains limited. Therefore, this research aimed to explore the basic forms of subjectivity underpinning delusions in schizophrenia and to compare the experiential structure of these delusional phenomena with the experience of the sacred. The study involved eleven participants diagnosed with schizophrenia and adopted the qualitative methodology of Interpretative Phenomenological Analysis (IPA). Participants’ accounts revealed four core experiential themes: 1) fragility of presence, 2) disposition to delusion, 3) apophany, and 4) eccentric existential reorientation. Primary delusional experiences may constitute a state-like phenomenon unfolding in four structural phases toward the formation of delusional contents: 1) a weakened bodily resonance with the world, 2) a predominance of the “physiognomic” properties, 3) an imposition of idiosyncratic and self-referential meanings, and 4) an active elaboration through delusion-like ideas. Formal similarities may be found with the “numinous” experience and with the specific articulations of the manifestation of the sacred or “hierophany.” Disembodiment may be posited as the common underlying scaffolding for both delusions in schizophrenia and the sacred, albeit persistent and forced upon the subject in the former, and transient, voluntarily sought, and socio-culturally integrated in the latter
