Caffeine doses of 3 mg/kg increase unilateral and bilateral vertical jump outcomes in elite traditional Jiu-Jitsu athletes
Caffeine increases vertical jump, although its effects on kinetics and kinematics during different phases of bilateral and unilateral jumps remain unclear. The aim of this study was to identify the effects of 3 mg/kg on kinetic, kinematic and temporal variables in the concentric and eccentric phases of bilateral and unilateral countermovement jumps. A total of 16 Spanish national team traditional Jiu-Jitsu athletes took part in two experimental trials (3 mg/kg caffeine or placebo) in a randomized, double-blind crossover study. Sixty minutes after ingestion, bilateral and unilateral jumps were performed on a force platform. Compared to the placebo, caffeine increased bilateral jump height (p = 0.008; Δ% = 4.40), flight time (p = 0.008; Δ% = 2.20), flight time:contraction time (p = 0.029; Δ% = 8.90), concentric impulse (p = 0.018; Δ% = 1.80), peak power (p = 0.049; Δ% = 2.50), RSI-modified (p = 0.011; Δ% = 11.50) and eccentric mean braking force (p = 0.045; Δ% = 4.00). Additionally, caffeine increased unilateral RSI-mod in both legs (Left: p = 0.034; Δ% = 7.65; Right: p = 0.004; Δ% = 11.83), left leg flight time (p = 0.044; Δ% = 1.91), left leg jump height (p = 0.039; Δ% = 3.75) and right leg FT:CT (p = 0.040; Δ% = 9.72). Caffeine in a dose of 3 mg/kg BM in elite Jiu-Jitsu athletes is a recommended ergogenic aid as it increased performance of bilateral and unilateral vertical jumps. These increases were also accompanied by modified jump execution during the different phases of the countermovement prior to take-off
Lack of Parkinsonian Pathology and Neurodegeneration in Mice After Long-Term Injections of a Proteasome Inhibitor in Olfactory Bulb and Amygdala
Proteinaceous inclusions, called Lewy bodies (LBs), are used as a pathological hallmark for Parkinson’s disease (PD). Recent studies suggested a prion-like spreading mechanism for α-synucleinopathy where early neuropathological deposits occur, among others, in the olfactory bulb (OB) and amygdala. LBs contain insoluble α-synuclein and many other ubiquitinated proteins, suggesting a role of protein degradation system failure in PD pathogenesis. Therefore, we wanted to study the effects of a proteasomal inhibitor, lactacystin, on the aggregability and transmissibility of α-synuclein in the OB and amygdala. We performed injections of lactacystin in the OB and amygdala of wild-type mice. Motor behavior, markers of neuroinflammation, α-synuclein, and dopaminergic integrity were assessed by immunohistochemistry. Overall, there were no differences in the number of neurons and α-synuclein expression in these regions following injection of lactacystin into either the OB or amygdala. Microglial and astroglial labeling appeared to be correlated with surgery-induced inflammation or local effects of lactacystin. Consistent with the behavior and pathological findings, there was no loss of dopaminergic cell bodies in the substantia nigra and terminals in the striatum. Our data showed that long-term lactacystin injections in extra nigrostriatal regions may not mimic spreading aspects of PD and reinforce the special vulnerability of dopaminergic neurons of the substantia nigra pars compacta (SNc)
Therapeutic Drug Monitoring of Antifungal Drugs: Another Tool to Improve Patient Outcome?
Introduction: This study aimed to examine the relationship among adequate dose, serum concentration and clinical outcome in a non-selected group of hospitalized patients receiving antifungals.
Methods: Prospective cross-sectional study performed between March 2015 and June 2015. Dosage of antifungals was considered adequate according to the IDSA guidelines, whereas trough serum concentrations (determined with HPLC) were considered adequate as follows: fluconazole > 11 µg/ml, echinocandins > 1 µg/ml, voriconazole 1-5.5 µg/ml and posaconazole > 0.7 µg/ml.
Results: During the study period, 84 patients (65.4% male, 59.6 years) received antifungals for prophylaxis (40.4%), targeted (31.0%) and empirical therapy (28.6%). The most frequent drug was micafungin (28/84; 33.3%) followed by fluconazole (23/84; 27.4%), voriconazole (15/84; 17.9%), anidulafungin (8/84; 9.5%), posaconazole (7/84; 8.3%) and caspofungin (3/84; 3.6%). Considerable interindividual variability was observed for all antifungals with a large proportion of the patients (64.3%) not attaining adequate trough serum concentrations, despite receiving an adequate antifungal dose. Attaining the on-target serum antifungal level was significantly associated with a favorable clinical outcome (OR = 0.02; 95% CI 0.01-0.64; p = 0.03), whereas the administration of an adequate antifungal dosage was not.
Conclusions: With the standard antifungal dosage, a considerable proportion of patients have low drug concentrations, which are associated with poor clinical outcome.

Impulsivity and the Experience of Desire in the Choice of Erotic Stimuli
(1) Background: the relationship between erotic desire and personality factors is still relatively understudied. (2) Objective: to study the influence of the experience of desire, as well as impulsivity in the choice of videos, as the behavioral variable in the experimental trial. (2) Method: the sample consisted of 48 adult subjects, who took part in an experimental study that involved watching videos. (3) Results: the linear regression analysis revealed that the behavior involved in choosing videos is predicted by the sexual desire felt at the time of the trial, and not by stable personality factors, such as impulsivity or general self-report levels of sexual desire. (4) Conclusion: it is observed that the specific moment or situation and the behavior have a bigger impact on the erotic desire experienced at the time of the test than certain personality traits, as well as the previous and habitual levels of erotic desire of which an individual reports
Individual differences in anger and displaced aggression: the role of metacognitive beliefs and anger rumination.
Metacognitive model is a theoretical approach aimed to explain emotion dysregulation and others emotion-related issues, such anger, and aggressive behavior. From this model, people having higher maladaptive metacognitive beliefs (e.g., “I can't control my thoughts”) are more likely to activate and maintain anger rumination and, in turn, to experience higher levels of anger and to act aggressively. Preliminary evidence shows the role of metacognitive beliefs on anger rumination and anger levels, whereas no studies have examined its association with aggressive behavior. This study first examined the associations between metacognitive beliefs, anger rumination, anger levels, and the propensity to engage in displaced aggression, and second, the mediation role of anger rumination in the relations among metacognitive beliefs and anger and displaced aggression. Participants were 947 students and non-students from general population recruited in two different countries (Australia and Spain). Correlational analyses revealed a similar pattern of results in the Australian and Spanish sample, with participants having dysfunctional metacognitive beliefs also showing higher anger rumination, higher levels of anger and a higher tendency to act aggressively. Structural equation analyses revealed the associations of metacognitive beliefs with anger levels and displaced aggression was fully mediated by anger rumination in both samples. These results suggest that metacognitive beliefs should be considered in comprehensive models and in the therapy of anger problems and aggression
Emotional regulation processes: influence on pain and disability in fibromyalgia patients
Objectives: Fibromyalgia (FM) is a chronic syndrome characterised by widespread musculoskeletal pain associated with other symptoms, including psychological distress. While negative mood (anxiety, depression, and anger) has been widely explored in FM, few studies have investigated emotional dysregulation. Our purpose was to evaluate problems in the processes of emotional regulation and to explore their influence on the severity of pain and disability.
Methods: Emotional regulations, anxiety, depression, anger, pain and disability were evaluated in 47 FM patients and 47 healthy subjects. Regression analyses were performed to evaluate the role that emotional regulation processes have on pain severity and disability of FM patients.
Results: Results showed that although FM patients do not differ in terms of the attention paid to their emotional states, FM patients had greater difficulties in the emotional regulation process. In addition, emotional rejection and interference are two variables that influence the pain severity and disability.
Conclusions: FM patients need to be trained in strategies for regulating their emotions, in order to achieve a reduction in negative mood states, as well as their impact in pain and disability
