Determination of recognition sequence of DNA methyltransferase M. Ape from Hyperthermophilic archaea Aeropyrum pernix
Get PDFapplication/pdfDNA methylation, one of the epigenetic modifications, is mainly reported N6-methyladenine(m6A), N5-methylcytosine(m5C) and N4-methylcytosine(m4C) and there exist in all of the domain of eukaryote, bacteria and archaea. The significant function of DNA methylation is gene regulation, control of cell cycle and Restriction-Modification system (R-M system) of bacteria. The Type II of R-M system is constituted by restriction enzyme and DNA methyltransferase and there recognize same sequence. In the R-M system of hyperthermophilic archaea Aeropyrum pernix, restriction enzyme already has been sale from New England Biolabs Inc., however, the DNA methyltransferase of the pair of restriction enzyme is only demonstrated by homology analysis on genome. In this report, we clarified that the properties of DNA methyltransferase M. Ape of R-M system Type II from Aeropyrum pernix.
The ORF sequence of M. Ape was introduced into pCold I vector and expression in E. coli JM109 strain. And then, it was purified by His-tag. The recognition sequence of M. Ape was decided by methylation activity and bisulfite sequence. As the result, it is clarified that M. Ape is cytosine-specific methyltransferase and it modify the second cytosine in GCTGC sequence. And also, it was suggested that the optimum temperature of M. Ape was 60 degrees Celusius.departmental bulletin pape
Novel methylation analysis method with use of recombinant maintenance methyltransferase
Get PDFapplication/pdfDNA methylation is one of epigenetics, and aberrant DNA methylation is related to various diseases such as cancer, neurological diseases, and allergic diseases. It is necessary to understand the methylation status of DNA in order to elucidate the fundamental cause of these diseases and to make an accurate diagnosis. Currently, the bisulfite sequencing method (BS-seq) is generally widely used as a DNA methylation analysis method. However, BS-seq has a problem that it cannot distinguish between hydroxymethylated cytosine (hmC) and methylated cytosine (mC).
Therefore, our laboratory is trying to develop a new methylation analysis method that combines DNA (cytosine-5) methyltransferase 1 (Dnmt1) and Polymerase.
In this study, recombinant Dnmt1 with low de novo activity and low activity against hmC in vitro was prepared and applied to the methylation analysis of the tyrosinase promoter of B16 melanoma. As a result, the difference in the methylation site was observed between the analysis result of only BS-seq and the analysis result of the novel methylation analysis method.
From this result, it is considered that by combining BS-seq with recombinant Dnmt1, only methylation could be detected excluding hydroxymethylation.departmental bulletin pape
Affirmative Concepts toward the Linguistic Diversity and the Sociolinguistic Study in Japan.
Get PDFapplication/pdfIn this paper, I aim to reveal quantitatively how Japanese sociolinguists have studied the concept of multilingualism by comparing the number of articles, reviews, and lecture texts published in the three academic journals, Sociolinguistics, The Japanese Journal of Language in Society and Language Policy, which are the most representative media among Japanese sociolinguists. Analyzing the article trends in these publications reveals how Japanese scholars have accepted the concept of multilingualism. At least within the scope of the journals examined in this study, European multilingual policies have often been discussed; however, Canadian or Australian language policies have not been a central topic, although these two states have implemented their own multilingual policies earlier than most European states. Effects of Neurofeedback Training on Nursing Students -Verification of attention control improvement-

Drafting Education Using Additive Manufacturing Products

In addition, I install plural FDM method 3D printers in the point, and a production section manufactures a 3D printer molding product with a designer each, and the method to design it with a videophone, and to review is reported. Handwritten drafting time decreased for CAD education time, and it has been said that improvement of the ability for space grasp was difficult. The inflection of the FDM method 3D printer molding product was tried as a help of the understanding of the three-dimensional structure. It was concluded that understanding of the trigonometry advanced than the one that the presentation time of the 3D printer molding product showed at the time of explanation before the drafting start showed it during drafting. As for the scoring average of the understanding degree of each method, the latter was 48% 73.5% of former. In addition, handwritten drawing quality tended to be downplayed as CAD education got into full swing. Numerical scheme based on the spectral method for calculating nonlinear hyperbolic evolution equations

The detail discretization processes are discussed in case of one-dimensional Klein-Gordon equations. In conclusion, a numerical scheme with the order of total calculation cost: O(N log2 N) is proposed. A pilot Study of the Efficacy of a Support Program for Families with a Young Child with a Developmental Disorder
Get PDFapplication/pdfAim: A support program for family of a child with a developmental disorder based on the family systems theory (Wright & Leahey, 2005) and previous studies by our research group, was designed to foster the family’s problem-solving ability. This study aimed to examine the efficacy of this program.
Methods: The effectiveness of the program was evaluated with the one-group pretest and posttest design using “the ability to problem-solving rating scale for families” developed by research group. This scale consists of 26 items and 4 subscales and its reliability and validity were largely confirmed (Hamazono & Moriyama, 2011), so that this scale can be considered useful for the measurement of ability to problem-solving of families with a young child with a developmental disorder. The participants consisted of 13 families, all of whom attended a four-day training session.
Results: The total score increased significantly immediately after the program compared with before the program and was maintained after one month. After the program, the average values for each subscale at three time points were scores for “Sense of self-control regarding parenting” tended to increase significantly.
Conclusions: The results reported here demonstrate that our support program for families is effective in enhancing their problem solving abilities. It is necessary, however, to examine why intervention did not produce significant improvement in some aspects of the program and to perform a follow-up for spousal relationships one month after the intervention.departmental bulletin pape
Characteristics of Interpersonal Communication Use of SNS for Nursing College Students
Get PDFapplication/pdfThe study aims was to clarify the relationship between SNS utilization and interpersonal communication among nursing college students. The survey was conducted by creating a questionnaire of 46 questions composed of the subject's personal background, SNS utility recognition, and social skills. The survey was conducted on 156 nursing college students enrolled in the Faculty of Nursing at the University the Kanto region, and 150 subjects with valid results were analyzed.
As a result, SNS was used by all subjects and was a necessary tool for the daily life of nursing college students. Nursing college students used social networking services as an easy and convenient way to interact, rather than seeking to build intimate relationships with others and satisfy them. Nursing college students also recognized that they use SNS to understand others and control their emotions. In modern nursing education where ICT spreads, it was considered highly necessary to study learning methods using non-face-to-face communication techniques to enhance social skills.departmental bulletin pape
