Asp73 and Asp420 play a critical role in ATP hydrolysis of yeast mitochondrial Hsp60 --Research works accomplished by using materials analysis facilities: XXXI(1)--
application/pdfType I chaperonins (Cpn60/Hsp60) are essential proteins that mediate the correct folding of newly translated and stress-denatured proteins in bacteria, chloroplast and mitochondria. Despite the high sequence homology among chaperonins, there are some critical differences at the mechanistic level between the bacterial and the mitochondrial chaperonins with respect to their oligomeric state and their ability to function with co-chaperonins from different sources. Asp52 and Asp398 in Escherichia coli chaperonin GroEL are known as the critical residues on ATP hydrolysis because GroELD52A/D398A mutant is severely deficient in ATP hydrolysis but not in ATP binding. Here, yeast mitochondria Hsp60 (mHsp60) and Thermus thermophilus Hsp60 (TCpn60) were used to investigate the functional commonality of these two well-conserved Asp in ATP hydrolysis. These residues in GroEL correspond to Asp73 and Asp420 in mHsp60 and to Asp51 and Asp396 in TCpn60, respectively. We found that ATPase activity of mHsp60D73A and mHsp60D420A mutants were ~20% and < 4% of wild type, respectively, indicating that functional commonality of two Asp was shown in yeast mHsp60, despite suggesting different roles of Asp51 in TCpn60, surprisingly.departmental bulletin pape

Developing High Performance CAD for Depression by Employing Image and Voice Information
application/pdfThe number of depressed people is on the rise in Japan. In 2011, depression was added to the four major diseases, making it one of the five major diseases. This suggests the need for reliable detection of depressed people. Currently, the diagnosis of depression is mainly based on the interview. However, this lacks objective indicators, and some have questioned the accuracy of the diagnostic results. In order to solve this problem, we are developing an AI-based system that calculates the degree of certainty that a patient is depressed by focusing on facial images and voice information. The purpose of this study is to use this as an objective indicator to assist in the diagnosis of depression. This paper describes the construction of a voice discriminator that constitutes the system. A total of 42 features were created based on the patient's speech and the characteristics of the depressed person. The SVM was used to create and evaluate the discriminator, and the accuracy was 90% for the female data. However, the accuracy for male data was about 55%. Hence, these features were selected by 18 feature selection methods. The discriminator was made and evaluated again, and the accuracy was 80%, even for male data.departmental bulletin pape
Goodput analysis for wireless networks during message segmentation: Case of independent bit error prone links
application/pdfThis paper investigates the effect of payload size on goodput for wireless networks where packets created from a message through a segmentation function are lost due to independent bit errors and these lost packets are recovered by a stop-and-wait protocol. To achieve this, we derive the exact analytical form of goodput from a given message size distribution and payload size. In previous work, packet sizes are assumed to be constant, which are payload size plus header size, although actual segmented packets are not constant in size. From numerical results, we show that the constant packet-size assumption is not justified in some cases. In addition, we propose the approximate payload size to achieve the maximum goodput. Finally, we demonstrate that the approximate goodput models which capture the message segmentation simply yields good accuracy.departmental bulletin pape
Study on the reaction behavior of limonenediols and dimethyl carbonate
application/pdfTo develop biomass-derived chemicals, reaction behavior of dimethyl carbonate (DMC) with four types of limonene-1,2-diol (LMdiol) was studied in the presence of catalysts such as 4-dimethylaminopyridine. Each of the reaction was carried out without any addition of organic solvents at 90℃ for 6-48 hours, in which the feed molar ratio of LMdiol : DMC : catalyst was fixed to 1.0 : 2.0 : 0.2. In all cases, the reactions produced no polymeric products, poly(limonene carbonate). The 1H NMR spectroscopic analysis revealed that the reaction products were dependent on the stereochemical configuration at two hydroxy groups of starting LMdiols, resulting in formation of the corresponding fused five-membered cyclic carbonates, monoalcohol-monocarbonate, and trans-limonene oxide.departmental bulletin pape
Synthesis of chlorosilicate under reducing atmosphere --Research works accomplished by using materials analysis facilities: XXXI(2)--
application/pdfIn this study, we attempted to synthesize Ca10Si6O21Cl2 in a reducing atmosphere using CaCO3, SiO2, and CaCl2‧2H2O as starting materials and by firing in a 5% H2 stream or with activated carbon. In order to confirm that the reducing atmosphere was realized, CaSO4‧2H2O or Eu2O3 was added as a starting material, and the introduction of sulfur or europium into the sample and their states in the sample were examined. Ca10Si6O21Cl2 phase was produced by both firing methods, but there were many impurities when firing in a 5% H2 stream, while there were few impurities when firing with activated carbon. Sulfur was detected by energy dispersive spectroscopy in the sample obtained by adding CaSO4‧2H2O as a starting material and firing with activated carbon, but SO42- could not be confirmed by Raman scattering spectroscopy. The sample obtained by adding Eu2O3 as a starting material and firing with activated carbon showed green fluorescence originated from Eu2+ by ultraviolet irradiation. When activated carbon is used, the Ca10Si6O21Cl2 phase can be successfully synthesized in a reducing atmosphere.departmental bulletin pape

Development of a novel evaluation system of tip growth for antifungal compounds with use of S. cerevisiae and application of the system for the screening in herbal medicine.
application/pdfThe infection by fungus such as yeast or mold is called mycoses. Among mycoses, deep mycoses are most likely to have more severe symptoms. To cure those deep mycoses, antifungal agents is used as a chemotherapy. However, the number of antifungal agents which can be used for treatment are only 4 classes 11 types. In addition, side effects and the appearance of resistant fungi have been confirmed for these drugs. Therefore, it is necessary to develop an antifungal agent different from the conventional antifungal agent.
Thus, we focused on a tip growth, which is a unique growth system of fungi. Tip growth is a method of growth by repeating the decomposition and the resynthesis of the cell wall. It has been reported that the cell wall-degrading enzyme β-1,3-D-Glucanase (BGL2) involved in this tip growth is vesicular transported from the inside of the cell to the growth point.
We consider that targeting vesicle transport during tip growth will provide selective toxicity to fungi.
The purpose of this study is to develop novel antifungal agents targeting vesicle transport of fungal
tip growth. Therefore, we constructed an evaluation system for visualizing vesicle transport using
Saccharomyces cerevisiae.
This evaluation system can visualize the vesicle transport of BGL2 during tip growth by fusion of the
green fluorescent protein (EmGFP) to BGL2. The constructed tip growth evaluation systems were applied to search for vesicle transport inhibitor in herbal medicine. As a result, vesicle transport inhibitory activity was observed in 8 Kinds of herbal medicine extract.departmental bulletin pape
