Macrophage-mediated phagocytosis of bacteria adhering on biomaterial surfaces
Biomateriaal-geassocieerde infecties (BAI) vormen een belangrijk klinisch probleem dat moeilijk te behandelen is en waardoor implantaatvervanging vaak noodzakelijk is. Zowel tijdens een operatie als postoperatief kunnen pathogene bacteriën op het oppervlak van een implantaat geïntroduceerd worden en de concurrentie aangaan met gastheercellen die proberen het implantaat te integreren. Het uiteindelijke lot van een geïmplanteerd biomateriaal wordt beschreven als een wedstrijd tussen bacterie-adhesie en biofilmgroei op het oppervlak van een implantaat versus weefselintegratie (“race for the surface”). Tot op heden zijn in vitro studies naar infectierisico’s van biomaterialen of functionele coatings voor orthopedische en tandheelkundige implantaten gericht op òf het tegengaan van bacterie-adhesie aan een implantaat, òf de bevordering van adhesie en proliferatie van weefselcellen. Hoewel het “race for the surface”-concept uitgebreid onderzocht is in vivo, tot voor kort bestond er geen in vitro-methode voor dit doel. Op dit moment hebben verscheidene onderzoeksgroepen methoden ontworpen waarmee de simultane respons van bacteriën en weefselcellen op een oppervlak onderzocht kan worden. In dit proefschrift, beschrijven we kweekexperimenten waarbij twee, en drie entiteiten gelijktijdig gekweekt worden met als doel de evaluatiemogelijkheden van multifunctionele coatings in vitro te verbeteren en daarmee het kennishiaat tussen in vitro en in vivo onderzoeken te overbruggen. Verschillende biomateriaal oppervlakken onderzocht te voorkomen of te elimineren bacteriële infectie terwijl het bevorderen weefselintegratie . Daarnaast, een visie op de behandeling van biomateriaal-geassocieerde biofilms in de toekomst op basis van de experimenten in dit proefschrift beschreven, wordt gegeven in de algemene discussie.
Biomaterial-associated infections (BAI) constitute a major clinical problem that is difficult to treat and often necessitates implant replacement. Pathogens can be introduced on an implant surface during surgery or postoperatively and compete with host cells attempting to integrate the implant. The fate of a biomaterial implant has been depicted as a race between bacterial adhesion and biofilm growth on an implant surface versus tissue integration. Until today, in vitro studies on infection risks of biomaterials or functional coatings for orthopedic and dental implants were performed either for their ability to resist bacterial adhesion or for their ability to support mammalian cell adhesion and proliferation. Even though the concept of the race for the surface in BAI has been intensively studied before in vivo, until recently no in vitro methodology existed for this purpose. Currently, various groups have proposed co-culture experiments to evaluate the simultaneous response of bacteria and mammalian cells on a surface. In this thesis, we describe biculture, and tri-culture experiments that allow better evaluation of multi-functional coatings in vitro and therewith bridge the gap between in vitro and in vivo studies. Several biomaterial surfaces were investigated to prevent or eliminate bacterial infection while promoting tissue integration. In addition, from the experiments described in this thesis, an outlook on future treatment of biomaterial-associated biofilms is given in the general discussion.
Home-making of older Antillean migrants in the Netherlands
<p>A group of 141,345 immigrants from the Netherlands Antilles, a former colony, live in the Netherlands. An increasing number of these migrants are at or above retirement age, and for them, the question of where they want to grow old becomes relevant. It is important for people to age in a place where they feel at home, as attachment to place increases wellbeing in old age. In this article we discuss how older Antillean migrants in the Netherlands make their house and immediate living environment into a home. We focus on home-making practices in a broader cultural context, and in relation to wellbeing. These topics are addressed by drawing on qualitative life-history interviews with Antillean older people, who live in a co-housing community for older adults. It turns out that objects which remind the participants of their home country play an important role in making a home. Also, the community, with people from similar backgrounds, contributes to a sense of home. Finally, the presence of children and other family members is a key motivation for the participants' decision to age in the Netherlands.</p>
The Electrophysiology of Language Comprehension: a Neurocomputational Model
Ongeveer tien jaar geleden stuitten onderzoekers die gebruik maken van elektro-encefalografie (EEG) op resultaten die leken te wijzen op een "Semantische Illusie" bij het begrijpen van taal: Zinnen die qua betekenis vreemd zijn, maar verder grammaticaal correct, hadden geen effect op de aan betekenis gerelateerde "N400 component" in het EEG signaal, maar juist op de aan structuur gerelateerde "P600 component".
Op basis van deze bevindingen zijn er maar liefst vijf nieuwe modellen van taalverwerking voorgesteld.
Al deze modellen claimen dat er niet een enkele verwerkingsroute, maar twee of meer verwerkingroutes zijn binnen het taalverwerkingsysteem, waarvan bij één route de structuur van een zin volstrekt niet van belang is, maar enkel de betekenis van de woorden waaruit de zin bestaat.
In dit proefschrift laat ik zien dat er een veel simpelere uitleg is voor de 'problematische' bevindingen en toon ik met computersimulaties aan dat deze alternatieve uitleg alle relevante data kan verklaren.
One decade ago, researchers using event-related brain potential (ERP) measurements stumbled upon what looked like a Semantic Illusion in language comprehension: Semantically anomalous, but otherwise well-formed sentences did not affect the meaning-related N400 component, but instead increased the amplitude of the structure-related P600 component. This finding spawned five new models of language comprehension, all of which claim that instead of a single comprehension process, there are two or even more separate processing streams, one of which is not driven by structure, but by word meaning alone.
In this thesis, I argue that there is a much simpler way to account for these data, and present evidence from neurocomputional simulations showing that this alternative explanation is able to predict all relevant ERP patterns found in the literature.
Interactive Participation in Rapid Onset Natural Disasters: An Analysis of ACFs emergency responses between July 2012 and July 2013
The purpose of this thesis is to investigate how programs that are emergency responses to rapid onset natural disasters incorporate interactive participation. Interactive participation in the context of rapid onset natural disasters is an unexplored topic within literature, providing a clear research gap for this study. By complementing theoretical knowledge gathered through the literature review with an analysis of the actual practices of a humanitarian actor, the aim is for the findings of the study to be helpful for humanitarian professionals regarding the design of future participatory approaches that aim to be sustainable and empowering.
Four emergency response programs of Action Contre la Faim (ACF) serve as case studies for this thesis. Eligible programs for analysis had to have a level of participation embedded within them, and constitute an emergency response to a rapid onset natural disaster between July 2012 and July 2013. Each case is analysed individually on the presence of the six factors that constitute interactive participation. The statistical process used to analyse the data in this thesis is a qualitative content analysis, based on a deductive category application.
After the data analysis, the main research result is that not all factors of interactive participation are present within ACFs responses to rapid onset natural disasters, with one of the six factors missing in its entirely. However, a quite significant finding is that five of the six factors are present within ACFs programs. The current research thus presents empirical evidence that proves that it is possible to a certain extent to implement factors of interactive participation that facilitate empowerment and sustainability in such contexts. The main conclusion drawn at the end of the study is that further research is necessary in order to continue both theoretical and practical advancement on the topic of interactive participation in the context of rapid onset natural disasters, in order to assure the accountability to beneficiaries and affected communities in the these contexts. The current research aims to increase the understanding, both on a theoretical and practical level, regarding interactive participation, its constraints and most importantly, its possibilities in the context of rapid onset natural disasters.
Sigma-1 Receptor Imaging in the Brain
De sigma-1 receptor is een unieke “orphan receptor” die sterk tot expressie komt in neuronen en glia cellen. Sigma-1 receptoren zijn betrokken bij verschillende aandoeningen van het centraal zenuwstelsel (CZS) zoals depressie, angststoornissen, psychose, schizofrenie, de ziekte van Parkinson, de ziekte van Alzheimer, verslaving en neuropathische pijn. Vele geneesmiddelen voor het CZS zoals haloperidol, donepezil, rimcazol, fluvoxamine, sertraline en clorgyline hebben een middelmatige tot hoge affiniteit voor sigma-1 receptoren. Tot de farmaceutische bedrijven die zich momenteel met onderzoek, ontwikkeling en klinische trials van sigma-1 liganden bezighouden behoren Anavex Life Sciences Corporation, M’s Science Corporation en Laboratorios del Dr.Esteve S.A.
Het onderzoek dat in dit proefschrift wordt beschreven heeft aangetoond dat 11C-SA4503 bruikbaar is om sigma-1 receptoren in de hersenen van de rat te bestuderen met behulp van microPET. Een afname van de binding van deze tracer werd waargenomen tijdens normale veroudering en een toename in spontane hypofyse tumoren. REM slaapdeprivatie bleek eveneens tot een verlaging van de expressie van sigma-1 receptoren in het brein te leiden, terwijl behandeling van proefdieren met cutamesine (SA4503) het geheugenverlies dat door REM slaap deprivatie was ontstaan volledig kon opheffen. De bezetting van sigma-1 receptoren door de geneesmiddelen donepezil en cutamesine in doseringen die werkzaam zijn in studies van cognitie en gedrag konden met behulp van 11C-SA4503 worden bepaald. Een model van de kinetiek van het geneesmiddel cutamesine in plasma and brein kon worden gemaakt met behulp van klassieke en “non-linear mixed effects modeling” (NONMEM) op basis van gegevens voor respectievelijk de koude stof en de radioactieve verbinding (11C-SA4503).
Samenvattend kan worden gezegd dat de sigma-1 agonist 11C-SA4503 een bruikbare tracer is voor het onderzoek van verschillende aandoeningen, zowel van het CZS als van perifere organen, en bovendien gebruikt kan worden voor de evaluatie van het effect van geneesmiddelen.
The sigma-1 receptor is a unique orphan receptor, strongly expressed in neurons and glia. Sigma-1 receptors are involved in several central nervous system (CNS) disorders like depression, anxiety, psychosis, schizophrenia, Parkinson’s disease, Alzheimer’s disease, addiction and neuropathic pain. Several CNS drugs like haloperidol, donepezil, rimcazole, fluvoxamine, sertraline and clorgyline have moderate to high sigma-1 receptor affinity. Pharmaceutical companies currently involved in R&D and clinical trials of sigma-1 drugs include Anavex Life Sciences Corp., M’s Science Corp. and Laboratorios del Dr. Esteve S.A.
The studies presented in this thesis demonstrated the usefulness of 11C-SA4503 and microPET to evaluate sigma-1 receptors in the rat brain. Decreased and increased binding of the tracer to sigma-1 receptors was estimated in normal aging brain and spontaneous pituitary tumor respectively. REM sleep deprivation was found to cause reduced sigma-1 expression in the brain, while treatment with cutamesine (SA4503) was shown to overcome REM sleep deprivation induced cognitive deficits. The receptor occupancy by agonist drugs, donepezil and cutamesine, at doses effective in behavioral studies of cognition could be estimated using 11C-SA4503. The kinetics of the drug cutamesine in plasma and brain could be modeled using classical and non-linear mixed effects modeling (NONMEM) for the cold drug and its radioactive form (11C-SA4503) respectively.
In conclusion, the sigma-1 agonist tracer 11C-SA4503 has potential uses in the study of various disorders, both of the CNS and periphery, as well as in the evaluation of potential treatments and drug effects.
The role of economic integration for European cities and border regions
De groei van steden is afhankelijk van hun locatie en toegang tot markten. Geografische en institutionele obstakels – zoals nationale grenzen – belemmeren die toegang. Dit proefschrift laat zien hoe verschillende vormen van economische integratie de groei van steden beïnvloedt. Het onderzoek richt zich in het bijzonder op grensregio’s. Het laat zien dat nationale grenzen een negatief effect hebben op de ontwikkeling van steden in de grensregio’s. Omgekeerd hebben juist steden in de grensregio’s baat bij maatregelen die de economische integratie bevorderen. Overigens is er daarbij wel sprake van asymmetrie; soms profiteren de steden aan de ene kant van de grens, terwijl de steden aan de andere kant negatieve gevolgen ondervinden van verdergaande economische integratie. Tot slot geeft dit proefschrift aanwijzingen voor de effectiviteit van grensoverschrijdende stedenbanden; ook paren van individuele steden kunnen op die manier grensbarrières slechten.
The growth of cities depends on their location and access to markets. Geographical and institutional obstacles – such as national borders – limit that access. This thesis shows how different measures to stimulate economic integration affects the growth of cities. The study focuses in particular on border regions. It shows that national borders have a negative effect on the development of cities in the border regions. On the other hand, towns in the border regions benefit most from measures that try to stimulate economic integration. These effects are asymmetric; sometimes towns on one side of the border profit, while towns on the other side of the border are negatively affected. Finally, this thesis provides some evidence for the effectiveness of cross-border twinning as an alternative way to break down border barriers.
