    Mechanisms of t follicular regulatory cell differentiation

    Respostas imunológicas humorais protetoras coordenadas pelas células B, quer em resposta a infeções ou à vacinação, requerem a participação de células T. Entre estas, as células T foliculares auxiliares (Tfh) emergiram como elementos-chave, fornecendo ajuda às células B e facilitando a produção de anticorpos de alta afinidade. Por outro lado, as células T foliculares reguladoras (Tfr), originárias de células T reguladoras (Treg) tímicas nos tecidos linfoides periféricos, assumem o papel de orquestradoras. Elas modulam a magnitude das recções dos centros germinativos (GC) para evitar o aparecimento de clones de células B auto-reactivas, ao mesmo tempo que a produção de anticorpos protetores é otimizada. Deste modo, manter este delicado equilíbrio entre a produção de anticorpos e a autoimunidade depende imensamente das subpopulações celulares Tfh e Tfr. Apesar da extensa investigação sobre células Tfh desde a sua descoberta, as células Tfr só ganharam destaque recentemente. Mais de uma década de investigação sobre as células Tfr tem vindo a revelar algumas disparidades na sua biologia em comparação com as células Tfh, desafiando a suposição da sua estreita semelhança. Apesar da sua importância (uma vez que a sua ausência está associada ao desencadeamento de doenças autoimunes), persistem lacunas significativas na compreensão da biologia das células Tfr, proporcionando um vasto campo para exploração e descoberta. A presente tese pretende iluminar o mundo intrincado das células Tfr, procurando refinar a sua definição e elucidar o seu processo de diferenciação. A nossa investigação sobre as células Tfr durante a resposta imunitária em murganho proporcionou uma caracterização mais completa do seu fenótipo e das moléculas que governam a sua trajetória de desenvolvimento. Apesar da ontogenia reguladora partilhada, descobrimos diferenças significativas no fenótipo e na diferenciação das células Tfr em comparação com as células T reguladoras efetoras. Simultaneamente, demonstrámos que duas moléculas expressas na membrana e amplamente utilizadas para identificar as células Tfh e Tfr, o ICOS e o PD-1, exibem padrões distintos durante a diferenciação destas populações de células em humanos. Enquanto o ICOS e o PD-1 podem ser utilizados de forma intercambiável para a identificação das células Tfh, o ICOS é expresso antes da expressão de PD-1 nas células Tfr. Esta observação levou à delimitação de três fases distintas de desenvolvimento das células Tfr nos tecidos linfoides humanos. Adicionalmente, as nossas descobertas sugerem que o OX40L e o IL-6 podem desempenhar papéis cruciais na indução do programa de diferenciação das células Tfr nas células Treg humanas. No seu conjunto, os resultados obtidos nesta tese fornecem novas perspetivas sobre a caracterização e diferenciação das células Tfr. Além disso, proporcionam oportunidades para investigações futuras na modulação da resposta dos GC e no papel de moléculas específicas, identificadas neste estudo, na função das células Tfr.Protective humoral immune responses orchestrated by B cells, whether in response to infection or vaccination, require the involvement of T cells. Among these, T follicular helper (Tfh) cells have emerged as key players, enhancing B cell help and facilitating the production of high-affinity antibodies. Conversely, T follicular regulatory (Tfr) cells, arising from thymic T regulatory (Treg) cells within peripheral lymphoid tissues, assume the role of orchestrators. They modulate the magnitude of the germinal center (GC) reactions to prevent the emergence of autoreactive B cell clones while optimizing the generation of protective antibodies. Maintaining this delicate equilibrium between antibody production and autoimmunity relies heavily on both Tfh and Tfr subsets. Despite extensive research on Tfh cells since their discovery, Tfr cells have only recently come into focus. Over a decade of research on Tfr cells has uncovered intriguing disparities in their biology compared to Tfh cells, challenging the assumption of their close resemblance. Despite their critical importance (as their absence is associated with the onset of autoimmune conditions), significant gaps persist in our understanding of Tfr cell biology, offering a rich avenue of exploration and discovery. The present thesis endeavors to shed light on the intricate world of Tfr cells, seeking to refine their definition and elucidate their developmental pathways. Our investigation into Tfr cells during the immune response in mice has provided a more comprehensive characterization of their phenotype and the molecules governing their developmental trajectory. Despite their shared regulatory ontogeny, we have uncovered significant differences in the phenotype and differentiation of Tfr cells compared to effector Treg cells. Simultaneously, we have shown that two widely used cell-surface molecules for identifying Tfh and Tfr cells, ICOS and PD-1, exhibit distinct patterns during the differentiation of these cell populations in humans. While ICOS and PD-1 can be used interchangeably for the identification of Tfh cells, ICOS is upregulated prior to PD-1 expression in Tfr cells. This observation has led to the delineation of three distinct developmental stages within Tfr cells from human lymphoid tissues. Furthermore, our findings suggest that OX40L and IL-6 may play pivotal roles in inducing the Tfr differentiation program in human Treg cells. Altogether, the results obtained in this thesis provide fresh insights on the characterization and generation of Tfr cells. Additionally, they pave the way for further research into modulating the GC response and investigating the role of specific molecules discovered in this study in the function of Tfr cells

    Freshwater fish community assessment using eDNA metabarcoding vs. capture-based methods: Differences in efficiency and resolution coupled to habitat and ecology

    Environmental DNA (eDNA) metabarcoding has revolutionized ecological and environmental research by describing communities without relying on direct observations, making it a powerful, non-invasive, and cost-effective tool in biodiversity monitoring. However, the adoption of eDNA as a standard protocol in long-term monitoring programs, which have traditionally relied on capture-based methods, presents challenges in terms of data comparability. Here, we compared freshwater fish communities assessed through eDNA metabarcoding and electrofishing, across 35 sampling sites in the lower Tagus River basin, Portugal. For the majority of species or species-groups analysed individually (13 out of 17), a significant correspondence was observed between electrofishing and eDNA metabarcoding detections. A weaker correspondence was found between the number of specimens captured by electrofishing with the number of eDNA metabarcoding reads, with seven out of 13 taxa showing significant relationships. Species richness estimates based on the two methods were very similar at the basin level. The methods yielded significantly different species compositions, although these differences were driven by samples collected in the Tagus main channel, which is wider and has higher flow rates than tributaries. Benthic and shoreline fish communities showed similar species composition in the two methods, but this was not the case for pelagic communities, probably due to the higher water turnover of the pelagic zone and electrofishing inefficiency. Our results highlight the high potential of eDNA metabarcoding as a complementary method to electrofishing for freshwater fish monitoring, though further validation is needed to assess biases related to site-specific hydrological conditions and the ecology of the target species.info:eu-repo/semantics/acceptedVersio

    Enterococci susceptibility to biocides used in the Biological Isolation and Containment Unit of a Veterinary Teaching Hospital

    Tese de Mestrado, Microbiologia Aplicada, 2024, Universidade de Lisboa, Faculdade de CiênciasAs bactérias pertencentes ao género Enterococcus são microrganismos gram-positivos ubíquos, capazes de colonizar não só o trato gastrointestinal de humanos e outros animais, como também o solo, água e comidas fermentadas, podendo ainda originar infeções oportunistas. Nos últimos anos, têm sido associados a infeções nosocomiais, verificando-se que Enterococcus faecalis e Enterococcus faecium são as duas espécies mais frequentemente associadas a este tipo de infeções. A capacidade destas espécies bacterianas para causarem doença está relacionada com a plasticidade do seu genoma, o que lhes permite adquirir e disseminar determinantes genéticos, e também com a sua elevada capacidade de resistência a um grande número de antibióticos. Enterococcus spp. possuem a capacidade de expressar um elevado número de fatores de virulência, incluindo proteínas de resposta a stress, toxinas e moléculas da superfície celular, que lhes permitem estabelecer infeções graves. De facto, estas características fazem com que as bactérias deste género sejam um dos principais agentes responsáveis por Infeções Adquiridas em Ambiente Hospitalar (IAAHs), estando frequentemente associadas a infeções do trato urinário, endocardites, e ainda a infeções relacionadas com cateteres ou outros aparelhos implantados. Para além disto, algumas espécies de Enterococcus também possuem a capacidade de formarem biofilme, o que aumenta a sua capacidade de sobrevivência em ambientes extremos. Os biofilmes são comunidades de células bacterianas com aptidão para aderirem facilmente a superfícies, graças à produção de uma matriz hidratada constituída por substâncias exo-poliméricas. A estrutura dos biofilmes bacterianos pode variar entre camadas simples de células bacterianas a estruturas macroscópicas de grande densidade. Esta variação depende de vários fatores, como a expressão de certos genes, a composição do substrato ou ainda alterações aleatórias associadas a fatores bióticos e abióticos. Esta característica permite que as células bacterianas promovam o desenvolvimento de infeções no hospedeiro através da colonização de diversos tecidos e da evasão ao sistema imunitário do mesmo. Encontra-se também demonstrado que os agentes antimicrobianos possuem uma maior dificuldade em penetrar a matriz do biofilme, o que resulta na tolerância destas comunidades bacterianas a estes compostos, e consequentemente ao agravamento e cronicidade das infeções associadas. Uma forma de se combater as infeções nosocomiais é a aplicação de biocidas ou desinfetantes nas superfícies de hospitais e outras instalações onde estes microrganismos sejam comuns. Os biocidas são agentes químicos ou biológicos capazes de eliminar microrganismos potencialmente patogénicos e que, ao contrário dos antibióticos, não têm um modo de ação específico. Estes antimicrobianos podem ser utilizados em inúmeras situações, com vista a controlar a carga microbiana presente em ambientes diversos, incluindo na indústria farmacêutica e em instalações de saúde humana ou veterinária. Contudo, a presença de bactérias na forma de biofilme pode prejudicar a ação dos biocidas, pois os desinfetantes têm dificuldade em penetrar a matriz que envolve estas estruturas, não eliminando a totalidade das células bacterianas nelas presentes. Assim, as concentrações de biocidas aplicadas podem não ser letais, promovendo o desenvolvimento de bactérias com suscetibilidade reduzida a estes agentes, incluindo à clorexidina. A clorexidina é um exemplo de biocida utilizado na Unidade Biológica de Isolamento e Contenção da Faculdade de Medicina Veterinária da Universidade de Lisboa. Este desinfetante tem atividade bacteriostática e bactericida, ligando-se à parede celular bacteriana e causando disrupções a nível do equilíbrio osmótico, ou levando mesmo à morte celular. Este biocida é utilizado muito frequentemente, devido ao seu amplo espetro de ação, sendo normalmente aplicado em concentrações entre os 0,5% e os 4%, dependendo da indicação clínica. No entanto, tal como observado relativamente a outros desinfetantes, a capacidade inibitória deste biocida parece diminuir quando na presença de matéria orgânica, como solo ou sangue. Pouco se sabe sobre os mecanismos que conferem suscetibilidade reduzida à clorexidina em Enterococcus spp. De acordo com o que se encontra descrito na literatura, a maior parte das espécies com suscetibilidade reduzida apresenta bombas de efluxo de várias famílias, que expelem as moléculas do biocida para fora da célula, permitindo a sobrevivência bacteriana. Estas bombas estão muitas vezes associadas a resistências a vários antimicrobianos, incluindo não só a biocidas, mas também antibióticos. O objetivo deste estudo foi avaliar a capacidade para permanecer no ambiente de Enterococcus, incluindo em superfícies, animais e pessoas. Para tal, analisou-se a capacidade de formação de biofilme de 34 isolados de Enterococcus, obtidos a partir de amostras de superfícies da Unidade Biológica de Isolamento e Contenção da Faculdade de Medicina Veterinária da Universidade de Lisboa. Após quantificação com Cristal Violeta, concluiu-se que 82% dos isolados em estudo são produtores de biofilme, sendo a maioria produtores de baixo nível. Por outro lado, no sentido de avaliar se a capacidade de permanência no ambiente se encontra associada a uma suscetibilidade reduzida a biocidas, as concentrações mínimas inibitória e bactericida de clorexidina relativamente aos isolados em estudo foram também determinadas, uma vez que este composto é amplamente usado nas instalações onde foram recolhidas as amostras. Estas concentrações foram também determinadas na presença de matéria orgânica de baixo e alto nível, permitindo constatar que, quanto mais elevado for o nível presente, a matéria orgânica tem um efeito negativo na atividade da clorexidina. De facto, observou-se um aumento dos valores médios das concentrações mínimas inibitória e bactericida, especialmente na presença de matéria orgânica de alto nível, com um aumento dos valores médios de 2,2 mg/L para 4,7 mg/L no caso da concentração mínima inibitória e de 6,3 mg/L para 20,9 mg/L no caso da concentração mínima bactericida. No entanto, nos ensaios relacionados com matéria orgânica de baixo nível não se observou alteração dos valores médios da concentração mínima inibitória, apenas havendo um aumento dos valores médios da concentração mínima bactericida, de 6,3 mg/L para 12,0 mg/L. Para terminar, foi avaliada a influência da presença de concentrações sub-inibitórias de clorexidina na expressão génica de biofilmes formados por um dos isolados em estudo, selecionado com base no seu perfil de resistência. Para tal, a expressão génica de amostras de biofilme sem qualquer contacto com este biocida e de amostras de biofilme em contacto com concentrações sub-inibitórias de clorexidina foram analisadas através da técnica RNASeq, tendo-se observado alteração nos níveis de expressão de 99 genes. Foi possível verificar que a maioria dos genes com níveis de expressão alterados encontra-se envolvidos em processos biológicos relacionados com transportadores ABC, e também com a entrada de maltose nas células bacterianas. Os resultados obtidos neste estudo servem para confirmar a grande capacidade de permanência no ambiente que as bactérias pertencentes ao género Enterococcus possuem. Desde a elevada produção de biofilme às alterações genéticas que fornecem uma suscetibilidade reduzida a desinfetantes, estas bactérias possuem uma plasticidade que lhes permite superar fatores de stress que existem em ambientes extremos. O facto de elevados graus de matéria orgânica permitirem que estes microrganismos sobrevivam em concentrações mais elevadas de clorexidina vem salientar a importância de garantir uma boa limpeza e desinfeção de todas as áreas de instalações hospitalares que possam encontrar-se contaminadas por estas bactérias. Sendo um dos principais agentes associados com IAAHs nos últimos anos, Enterococcus spp. podem ser extremamente perigosos se permanecerem não só em superfícies, mas também se tiverem oportunidade de colonizar os pacientes e trabalhadores que frequentem este tipo de instalações. Mais estudos devem ser desenvolvidos nesta área, de forma a melhor compreender como todos estes mecanismos se interligam. Sugere-se, por exemplo, a realização de estudos com isolados de outra espécie, ou ainda com isolados que sejam fortes produtores mais biofilme, de modo a avaliar o efeito destas variações. Seria também importante avaliar o papel do dissacarídeo maltose na suscetibilidade reduzida a biocidas, de modo a verificar se este fenómeno é apenas um efeito secundário da ativação e funcionamento de bombas de efluxo.Enterococci are microorganisms that colonize the gastrointestinal tract of humans, while also being associated with Hospital-Acquired Infections (HAIs). The capacity of these bacteria to cause disease is related to the plasticity of their genome and to their reduced susceptibility to many antibiotics. Enterococci also form biofilms, which promotes their capacity to survive in extreme environments. One way to fight nosocomial infections is the applications of biocides on hospital facilities, where these microorganisms are common. However, the formation of biofilms by bacteria seems to difficult the action of biocides, which could lead to the use of sublethal concentrations of biocide and to the development of bacteria with reduced susceptibility to antimicrobials. The objective of this work was to evaluate the capacity of enterococci to remain in health-related facilities, like the Biological Isolation and Containment Unit (BICU) of the Veterinary Teaching Hospital of the Faculty of Veterinary Medicine, University of Lisbon. For this, 34 enterococci isolates were tested for their biofilm-forming capacity and their susceptibility to the biocide chlorhexidine when in the absence and presence of organic matter. The effect of sub-MIC concentrations of chlorhexidine in gene expression of enterococci in the biofilm form was also evaluated. Of the 34 isolates under study, 82% were found to be biofilm producers. In terms of chlorhexidine activity, it decreased in the presence of interfering substances, of low and high level, as seen by an increase in Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) values. Sub-MIC concentrations of this biocide seem to upregulate genes related to ABC transporters in biofilm cells. These results confirm that enterococci are very capable of remaining in the hospital environment, which is dangerous to both humans and animals in there. Further studies are needed to better understand all mechanisms that confer this concerning ability to this bacterial group

    Science archives: a literature review and international projects

    The connection between 'archives' and 'science' is not immediately apparent, but scientists experience this relationship in a particular way. However, this link materializes in ‘science archives’, which began gaining importance as scientific activity took on a more prominent role in society, particularly over the last four decades. This research aims to establish the context between 'archives' and 'science', which gave rise to ‘science archives’, define this type of archive, understand what kind of records they preserve, and, finally, present some of the projects and initiatives surrounding this type of collection. To achieve this, a narrative literature review was conducted, developing qualitative research based on documentary investigation.     targeting predictive and descriptive tasks for biomedical

    Triclustering, targeting the discovery of coherent subspaces within three-way data, is becoming increasingly relevant in data science, especially for pattern discovery and knowledge acquisition from complex datasets in the biomedical field. This technique can reveal hidden patterns such as putative regulatory modules, disease progression profiles, and individuals with coherent behaviors. When applied to labeled data, triclustering aids in class differentiation and supports real-world decision-making. However, learning from 3W biomedical data is typically challenged by the rich temporal and heterogeneous nature, having mixed-type features and different structure compositions. In response to these challenges, this thesis establishes the foundations for pattern-centric 3W data analysis, focusing on triclustering for temporal and heterogeneous three-way data, targeting both descriptive and predictive tasks. In this context, this thesis includes six major contributions. It provides a literature review and comparative study of current triclustering algorithms for temporal data, highlighting the strengths and weaknesses of existing methods. It presents new tools to support the development and assessment of pattern discovery approaches in descriptive and predictive contexts, including a new data generator capable of creating heterogeneous three-way datasets with annotated triclustering solutions and benchmark datasets for comparative evaluation. It proposes a novel approach to capture time-contiguous triclusters, enhancing the search for temporal coherence. It introduces a new triclustering approach able to handle heterogeneous data by applying sequential pattern mining principles to identify relevant patterns and derive triclusters capturing temporal data dynamics. Additionally, it presents a new method for learning pattern-centric predictors. Finally, it proposes an extension and integration of principles for learning from static and temporal data structures. The developed methods were comprehensively validated in concrete real-world clinical scenarios, showing promising results concerning two progressive diseases. They were used to predict clinically relevant endpoints and identify disease-specific progression patterns, supporting medical decisions and identifying significant patient profiles.BRAINTEASER (H2020-GA101017598

    A systems medicine approach

    Autism Spectrum Disorder (ASD) is a heterogeneous neurodevelopmental disorder characterized by deficits in social communication and interaction and repetitive, restricted behaviors. Individuals with ASD may present a variety of clinical and behavioral symptoms, as well as a number of comorbidities, leading to variable degrees of clinical severity. ASD segregate in families and genetics has a significant contribution to the disease’s development. In this PhD thesis, we focused on exploring the role of neurotransmitter and synaptic genes (NS genes) in ASD. Going beyond the identification of ASD risk genes, the main objective was to clarify the biological processes affected by gene variants in NS genes in ASD and the global networks that link those processes together, using a systems medicine approach. A genetic similarity disease network approach was developed to explore the shared genetics between ASD and comorbid brain diseases. A Knowledge Graph was developed to take advantage of novel gene-disease predictions for disease terms related to ASD to identify and characterize biological pathways implicated in the disorder. In this thesis, it was also performed a community detection analysis using protein–protein interactions involved in ASD, defined by variants in NS genes found in large exomic datasets of ASD subjects. A literature review was also carried out, targeting studies that integrate data related with genetic alterations and their implications in changes in brain structure and function, in individuals with ASD. This PhD thesis also showed that brain imaging data obtained through magnetic resonance and electroencephalography, combined with genetic analysis, has the potential to help in the understanding of the neurobiological mechanisms that contribute to the development of ASD, and in the definition of new therapeutic targets.     Sustentabilidade versus qualidade: água de consumo humano e emulsões

Contudo, a indústria cosmética também é dos setores industriais com elevado impacto ambiental e com uma pegada hídrica consideravelmente grande. Considerando o aumento da procura pelos consumidores de produtos mais sustentáveis, a indústria cosmética desenvolveu produtos sólidos anidros ou com baixo teor de água. No entanto, a reconstituição do produto na casa dos consumidores coloca algumas questões, no que concerne à qualidade e eficácia do produto reconstituído. O presente trabalho teve como principal objetivo estudar a influência da qualidade da água para consumo humano na qualidade dos produtos cosméticos reconstituídos em casa pelo consumidor. Dos vários parâmetros de qualidade da água, selecionaram-se os parâmetros com potencial impacto na qualidade das formulações, nomeadamente, pH, condutividade, sólidos dissolvidos totais, turvação e dureza. Estes parâmetros podem variar consideravelmente com a zona geográfica e consequentemente originar formulações cosméticas com características físico-químicas distintas. O efeito dos vários parâmetros de qualidade foi previamente avaliado com soluções padrão (dureza e soluções tampão de vários valores de pH) estudando o impacto individual de cada parâmetro sobre alguns constituintes da formulação, nomeadamente os corantes. O estudo completou-se com o impacto das várias águas sobre uma formulação desenvolvida (emulsão), a qual foi reconstituída com os vários tipos de água. As formulações foram caracterizadas quanto ao pH, tamanho da gotícula, viscosidade e estabilidade. A emulsão reconstituída com águas macias e médias apresentavam menor tamanho de gotículas e maior viscosidade em relação à emulsão reconstituída com águas duras ou muito duras. Para o mesmo produto, foi obtida uma cosmeticidade diferente, dependendo dos parâmetros físico-químicos da água. A utilização crescente de produtos sem água visa responder às preocupações de sustentabilidade na indústria, mas ainda é necessário um longo caminho para garantir um produto sustentável sem comprometer a qualidade do produto.Cosmetic products have a significant economic value and an important role in the people's daily life. However, the cosmetics industry is also one of the industrial sectors with the biggest environmental impact, with a significant water footprint. Given the increased consumer demand for more sustainable products, the cosmetic industry has developed anhydrous or low water content solid products. However, the reconstitution of the product in the consumer’s home raises some questions regarding the quality and efficacy of the reconstituted product. The main aim of this work was to study the influence of drinking water quality on the quality of cosmetic products reconstituted at home by the consumer. Of the several parameters of water quality those with a potential impact on the quality of formulations were selected, namely, pH, conductivity, total dissolved solids, turbidity, and hardness. These parameters can vary considerably from geographical area to geographical area and consequently give rise to cosmetic formulations with different physicochemical characteristics. The effect of the various quality parameters was previously assessed with standard solutions (hardness and buffer solutions of various pH values), studying the individual impact of each parameter on certain constituents of the formulation, particularly the colorants. The study was completed with the impact of the various waters on a developed formulation (emulsion), which was reconstituted with the various types of water. The formulations were characterised in terms of pH, droplet size, viscosity, and stability. The emulsion reconstituted with very soft and medium water had smaller droplet size and higher viscosity than the emulsion reconstituted with hard or very hard water. Thus, for the same product, a different cosmeticity was obtained, depending on the physicochemical parameters of water. The increasing use of waterless products aims to respond to sustainability concerns in the industry, but there is still a long path to ensure a sustainable product without compromising product quality.Faculdade de Farmácia da Universidade de Lisboa

Quando descontrolada ou prolongada, a inflamação pode tornar-se crónica e contribuir para o desenvolvimento de várias doenças humanas, incluindo doenças autoimunes, doenças intestinais e, nos piores casos, tumorigénese e progressão tumoral. A exposição a disruptores endócrinos (EDCs) é um dos fatores ambientais que contribuem para a inflamação, e estudos recentes trouxeram o grupo dos bisfenóis (BP) para o centro das atenções científicas. Estes compostos têm sido fortemente associados a várias patologias, incluindo inflamação crónica, e o seu efeito na saúde intestinal humana é um tema de grande interesse na atualidade científica. Tendo isto em conta, o objetivo deste trabalho foi analisar os efeitos de quatro bisfenóis – BPA, BPS-MAE, BPAP e BPP – no stress da barreira intestinal e nos efeitos pró-inflamatórios associados. Para tal, foi otimizado e estabelecido um sistema de co-cultura, consistindo num protocolo melhorado de células epiteliais polarizadas Caco-2, semeadas em filtros de inserção PET no compartimento apical, juntamente com macrófagos derivados de monócitos THP-1 no compartimento basolateral. Posteriormente, foram testados os efeitos da exposição aos BPs na integridade da barreira, no stress celular e nas citocinas pró-inflamatórias, numa ampla gama de concentrações (de 100 µM a 0,1 µM). Experimentalmente, verificámos que o modelo foi capaz de fornecer dados específicos sobre os potenciais efeitos dos BPs na saúde. Em termos de resistência transepitelial e stress epitelial, conseguimos identificar algumas tendências claras que precisam de ser consolidadas com mais réplicas experimentais independentes. Em particular, o BPA foi o indutor menos potente de respostas de stress celular e alterações na polarização epitelial, enquanto os análogos de BP testados mostraram ser mais disruptivos do que o BPA, com o BPP a revelar-se potencialmente o mais perigoso, seguido do BPAP e por último o BPS-MAE. Para avaliar os efeitos moduladores da inflamação desses compostos, testámos macrófagos, tanto diretamente como em co-cultura, para a expressão do marcador pró-inflamatório IL-1β, utilizando uma abordagem de RT-PCR semi-quantitativa. Uma otimização importante foi a sua pré-ativação com IFN-γ para aumentar a sensibilidade do modelo e permitir maior relevância fisiológica. As nossas observações mostraram que, mais uma vez, os novos BPs induziram efeitos mais severos em comparação com o BPA. O BPP revelou-se o indutor mais potente de inflamação, seguido do BPS-MAE. Ambos apresentaram níveis elevados do marcador IL-1β em todas as concentrações testadas. O BPAP e o BPA produziram efeitos mais atenuados, embora significativos em concentrações mais altas. Em conclusão, este trabalho forneceu-nos resultados fundamentais sobre estes análogos do BPA e os seus efeitos na saúde intestinal, acrescentando novos conhecimentos sobre a "nova geração" emergente de BPs e os seus potenciais efeitos adversos na saúde.Inflammation has reached epidemic proportions in industrialized countries, mainly due to unhealthy habits, poor diet, environmental pollution and other factors not yet understood. If uncontrolled or prolonged, inflammation can become chronic and contribute to the development of a number of human diseases, including autoimmune diseases, intestinal diseases and, in the worst cases, tumorigenesis and tumor progression. Exposure to endocrine disrupting chemicals (EDCs) is one environmental factor contributing to inflammation, and recent studies have brought the bisphenol (BP) group of EDCs into the scientific spotlight. They have been strongly linked to various pathologies, including chronic inflammation, and their effect on human gut health is a hot topic in the scientific community. With this in mind, the aim of this work was proposed to analyze the effects of four bisphenols, BPA, BPS-MAE, BPAP and BPP, on intestinal barrier stress and associated pro-inflammatory effects. To achieve this, a co-culture system was optimized and established, consisting of an improved protocol of polarized Caco-2 epithelial cells seeded on PET insert filters in an apical compartment, together with THP-1 derived macrophages in a basolateral compartment. Subsequently, the effects of BPs exposure on barrier integrity, cellular stress and pro-inflammatory cytokine were tested in a wide range of concentrations (from 100 µM to 0.1 µM). Experimentally, we found that the model was capable of delivering BP-specific data on potential health effects. In terms of transepithelial resistance and epithelial stress, we were able to identify some clear trends that need to be consolidated with more independent experimental replicates. In particular, BPA was the least potent inducer of cellular stress responses and changes in epithelial polarization, whereas the BP analogues tested proved to be more disruptive than BPA, with BPP appearing to be the most potentially hazardous, followed by BPAP and then BPS-MAE. To access the inflammation-modulating effects of these compounds, we tested macrophages, either directly or as co-cultured cells, for expression of the pro-inflammatory marker IL-1β using a semiquantitative RT-PCR approach. An important optimization was their priming with IFN-γ to increase the sensitivity of the model and allow for more physiological relevance. Our observations showed that, once again, the BP analogues induced greater effects compared to BPA. BPP appeared to be the more potent inducer of inflammation, followed by BPS-MAE. Both showed elevated levels of the IL-1β marker at all concentrations tested. BPAP and BPA produced more attenuated effects, although significant at higher concentrations.     Coastal areas face increasing pressure from urbanization and human activities, exacerbating erosion and environmental challenges. Coastal engineering structures, such as breakwaters, groins, seawalls, and jetties, have been used to stabilize beaches, but often prove inadequate. Beach nourishment has emerged as a promising alternative; however, significant challenges remain in assessing sediment transport associated with shoreface nourishments, and in understanding their efficiency and longevity. This thesis aims to enhance the understanding of sediment dynamics in the upper shoreface of wave-dominated coastal environments, with a focus on optimizing sediment management and improving the long-term sustainability of shoreface nourishments. To achieve these objectives, this research employs a multi-faceted approach combining physical experiments, field data collection, and numerical modelling. Chapter 3 describes a physical model experiment using magnetic tracers to measure sediment transport in a controlled environment. The effectiveness of the magnetic tracers is compared to fluorescent tracers in an experiment conducted in the large wave flume (Großer Wellenkanal, GWK)in Hannover. Results showed similar dispersion patterns and net transport rates for both tracers, though the magnetic tracers exhibited lower recovery rates, indicating the need for further research. Wave streaming emerged as a key process driving sediment transport under non-breaking waves in the direction of wave propagation. In chapter 4, field measurements, including a tracer experiment, are combined with numerical modelling to assess sediment transport in the shoreface of Costa Nova, Aveiro. The Delft3D model is used to simulate the interaction of waves, currents, sediment transport, and morphology in this complex, energetic environment. Data from the tracer experiment validate the model’s performance. A Lagrangian model is also developed to estimate sediment particle pathways based on the simulated transport fields. The results reveal that onshore bed load transport dominates during calm conditions, while longshore suspended transport prevails during moderate wave energy events. These findings improve the understanding of shoreface sediment dynamics. In chapter 5, the implemented numerical model is applied to analyse the processes and dynamics of a shoreface nourishment conducted in Costa Nova in the summer of 2020. A short- and seasonal-term analysis of the nourishment’s evolution highlights the critical role of incident wave conditions, particularly during moderate to energetic events. These conditions not only protect the beach by dissipating wave energy but also nourish adjacent areas through sediment spreading. A conceptual model is presented that illustrates the short- and seasonal-term impacts of the nourishment. In the short term, the nourishment behaves as a detached breakwater, affecting hydrodynamics; at a seasonal term, sediment redistribution contributes to both the subaerial and submerged beach. The thesis develops tools to support the optimization of nourishment placement to enhance its effectiveness. Future research should focus on refining model accuracy through better sediment characterization and improved acceleration techniques. This work contributes to the understanding of shoreface nourishment and supports sustainable coastal protection strategies

    Hidroxitirosol: estratégias de estabilização recorrendo à nanotecnologia

O azeite representa um dos principais componentes da dieta mediterrânea, sendo por isso largamente consumido e, por isso mesmo, produzido em grande escala. Com o intuito de reduzir a geração de resíduos e maximizar todo o potencial que a azeitona, a oliveira e o azeite apresentam, são utilizadas estratégias que permitem extrair componentes que apresentem atividades pertinentes do ponto de vista cosmético, nascendo assim o interesse pelo Hidroxitirosol – principal composto fenólico presente no azeite e subprodutos da sua produção. Deste modo, com o intuito de usufruir das inúmeras propriedades que este composto apresenta têm de ser aplicadas estratégias que permitam a sua estabilização e posterior incorporação em formulações cosméticas. Aqui, a nanotecnologia destaca-se por oferecer a possibilidade de, não só estabilizar o ativo, como também por permitir uma entrega mais eficiente do mesmo, de um modo mais controlado e personalizado, permitindo ainda aumentar a durabilidade do produto.The growing consumer demand for cosmetic products with sustainable and cost-effective origins has led the cosmetics industry to focus on new raw materials. Olive oil is one of the main components of the Mediterranean diet and is therefore widely consumed and, therefore, produced on a large scale. In order to reduce the generation of waste and maximize the full potential of olives, olive trees and olive oil, there are some strategies that are being used to extract components that have relevant activities from a cosmetic point of view, thus giving rise to interest in hydroxytyrosol - the main phenolic compound present in olive oil and by-products of its production. In order to take advantage of the numerous properties this compound presents, strategies must be applied to allow its stabilization and then its incorporation into cosmetic formulations. Here, nanotechnology stands out for offering the possibility of not only stabilizing the active ingredient, but also allowing it to be delivered more efficiently, in a more controlled and personalized way, while also increasing the product's durability
