PENGARUH TEKNIK SINTESIS TERHADAP KUALITAS PRODUK FATTYAMINA SEKUNDER
Secondary fattyamines has been synthesized by reacting primary fatty amines with acylchlorides and continued with reducing the corresponding secondary fattyamides formed to secondary fattyamines using LiAIH<I'Fatty amines are raw material of natural-based surfactants that can be derived from fatty acids, olefins, or alcohols, of which can be synthesized from natural sources such as palm oil. Conversion of secondary fatty amides to secondary fatty amines was evaluated through the quality of FTIR spectra on wave number of 1639-1645 em' (C=O vibration) and 1544-1555 em" (vibrations of C-H and of secondary amine N-H). Method of synthesis by using closed reflux syncore reactor was better than those of using microwave teflon tubes, and open reflux. The yield of 9 different secondary fatty amines obtainedfrom 5 to 27 replicates by the closed reflux syncore reactor method varied from 17%to 96%.Keywords: Natural-based surfactant, secondary fatty amine
PENGARUH SUMBER KARBON TERHADAP PRODUKSI ENZIM INULINFRUKTOTRANSFERASE DARI Nonomuraea sp
The aims of this research is to produce inulinfructotransferase enzyme with potentially benefit of bacteria Actinomycetes which was isolated from Indonesia soils to produce Difructoanhydride-Ill (DFA III), a compound has physiology activities to enhance calsium absorption of rats. cows and human colons so that the materials can be applied as antiosteoporosis. The processing of inulinfructotransferase enzyme from Nonomuraea sp by usingshakeflask level and study the influence of carbohydrate source were conducted. This enzyme has optimum temperature at 65rtC,higher than optimum temperature which has found before and the activities was stable at 70dc for 20 minutes. So that the enzyme has potentially applicable for industry scale. The results indicated that optimation condition for processing inulinfructotransferase enzyme of Nonomuraea sp : temperature sore. incubation time 48 hours.carbon source of inulin 20 g/L, nitrogen source 5g/L, noyeast extract added, the enzyme activities was 7,3 Unit/ml; However,when medium contains yeast extract 0,2 gIL,the inulinfructotransferase enzyme activity was 14,6 Unit/mL.Keywords: Inulinfructotransferase enzyme, Nonomuraeasp.inulin. yeast extract, DFAI
UJI AKTIVITAS ANTIOKSIDAN EKSTRAK KASARAIR BUNGA KECOMBRANG (Etlingera elatior) SEBAGAI BAHAN PANGAN FUNGSIONAL
A research of antioksidant activity from water crude extract of kecombrang flower (Etlingera elatior) have been reported. This Research aim to give erudite evidence of kecombrang flower as a functional food. Antioksidant was examined by activity diphenyl picryl hidrazyl ( DPPH) methode and chemicalcomponent analyses using GCMS. Water crude extract of kecombrang flower shows antioxidan activity ( ICso = 61. 6497 ppm) and waspredicted to have component of l-dodekanol, S-metil-l-oxo-lbuten 1-(21,41, 51-trihidroxyl phenil} and 1tetradecene (tR= 13,26, area=6,03, similarity at 98%).Keyword: Kecombrang ( Etlingera elatior), functional food, antioxidan
PROSES STERILISASI SARI BUAH TERONG UNGU (Solanum melongena) DENGAN SISTEM OZONISASI
One effort in the preservation of food is processed through a sterilization process using ozonization system with ozone molecule (0). The purpose of this study is to review the process of sterilization of juice eggplant (Solanum melongena) using ozonation systems with a concentration of 0.6 ppm ozone. In this study, eggplant juice made in 6 ratio of water: sugar(1:10%, 1:15%, 1:20%, 2:10%, 2:15%, 2:20%) and sterilize with 0.6 ppm ozone The ozonation time studiedfor (0. 6ppm) 0 min, 2 min, 4 minutes, 6 minutes, 8 min and long storage 0 days, 2 days, 4 days, 6 day. Test parameters obtained included the juice organoleptic test (preference level) and test of vitamin C. The results showed that ozonation can be used as a process of sterilization of juice eggplant, ozonation time with optimal and durable power offruit quality is 4minutes with long storage for 2 days. Response panelists on organoleptic test as a whole eggplant juice between 2.8 (preferred), and 3.13 (preferred). Levels of vitam in C, protein, carbohydrate and total microbial best eggplant juice are 1.35 mg1100mL, 1.1%, 3.43 mg1100mL, 8.36x1 rI cfulmL, respectively.Key words: Eggplant, ozonization, ozone,sterilization
STUDI AWAL PADA PREPARASI KATALIS BERBASIS LEMPUNG BERPILAR UNTUK REAKSI ETANOL MENJADI GASOLINE (ETG)
Research on catalytic reaction of ethanol to gasoline (ETG) was investigated in this study. The catalyst with a high acidity is required for the ETG reaction. Therefore, this study focused on the catalyst preparation of ETG reaction by using natural clays which prepared by the pillarization techniquewithseveral metal oxides, such as:AI, Zr, Fe and Cr as the pillared. The prepared of pillared clay were characterized by x-ray diffraction, specific surface area and pore distribution (BET) measurements and catalytic activity testing on the ETG reaction. It is observed that after the pillarization; the surface area and basal spacing (d001) of the natural clays were significantly increased. thus the ptllarization process was successfully prepared on natural clay. However,the acidity of theprepared ofpillared clay is still lower,probably due to the cations between layers of the natural clay were not homogeneous, thus although the pillarization process was happen but still not completely. Moreover, IR measurement showed only the acid type of Lewis is increased after the pillarization. Thistype is not favorable for the ETG reaction, and thus only the lower hydrocarbon products (<Cl2) were observed after the ETG reaction, even aromatics and cyclic hydrocarboncompounds wereobtainedKeyword : preparation, pillared clay, catalyst, ETG reaction
PENGARUH KONSENTRASI PENGINDUKSI METANOL SERTA"SUMBER KARBON SORBITOL DAN MONITOL TERHADAP PRODUKSI a-AMILASE Saccharomycopsis fibuligera R64 DALAM Pichia pastoris
Pichia pastoris has been widely used as host for heterologous protein expression for commercial purposes. The advantages of P. pastoris as protein expression host is its ability to grow with high cell density and the presence of a gene promoter that canbe induced tightly, named AOXI encoding alcohol oxidase. Improvement of recombinant protein production under control of AOXI promoter in P. pastoris expression system is still a major concern. Optimization of methanol concentration as inducerand addition of carbon sources, is one of the strategies to improve the expression level. This research aims to study the effect of methanol as inducer as well as sorbitol and mannitol as additional carbon source to the expression level of recombinantSaccharomycopsis fibuligera a-amylase (Sfamy) by P. pastoris (Mut). Sorbitol and manuot known as non-repressive carbon source, to increase the growth ofP. past oris, but not inhibit the AOX1 promoter and foreign proteins expression. Sfamy was expressed in P. pastoris GS115 (His-, Mut) with addition ofcarbon source, sorbitol and mannitol saparately to the expression medium. The result showed that, the optimum concentration" of methanol inducer for Sfamy production is 0.75%. The addition of sorbitol or mannitol increased Sfamy production. Concentrations of sorbitol and mannitol 2% with0.75% methanol inducer increase the secretion level of recombinant Sfamy 2.13 times and 1.94 times respectively, compared with no additional carbon source. This result indicated that addition of carbon source can improved recombinant protein production by P. pastoris, and the used of sorbitol as additionalcarbon source is more effective compared to mannitol.Key words: a-amylase. UJI AKTIVITAS EKSTRAK DAN FRAKSINASI DAUN BRUCEA JAVANICA (Merril) SECARA INVITRO
Brucea javanica (Merril) is a tropical plant widely grown in Indonesia, Brucea javanica has a bitter taste efficacious to reduce the heat and remove toxins, lethal parasites, anti-anti-dysentery and malaria. Chemical compounds contained in many of which have bioactivity. Antidiabetic test has been performed with a-glucosidase method, anti-oxidant test method "DPPH free radical scavenger" and BSLT toxicity test method (Brine Shrimp Lethality Test) of ethanol extractB.javanica. In the ethanol extract of leaves of the partition B.javanica performed using hexane, ethyl acetate and butanol. Then test each fraction in vitro activity. Test results showed that activity of ethylacetate is active as an anti-oxidant with IC50 value of 89,75 ppm and 7,52 ppm LC50 toxic values while antidiabetic did not have activites.Keyword : Antidiabetic, anti-oxidant, toxicity, a-glukosidase, DPPH, Brine Shrimp Lethality Test, Brucea Javanica(Merril)
ISOLASI DAN IDENTIFIKASI SENYAWA TRITERPENOID DAN ASAM 3-HIDROKSINIKOTINAT DARI EKSTRAK DIKLOROMETANA AKAR Garcinia picrorrhiza Miq
Have been isolated chemical constituent from medicinal plants of Garcinia picrorrhiza root from Cluciaceaefamily. Two pure compounds have been isolatedfrom dichloromethane extract by flash column chromatography in silica gel and mixture of hexane and ethyl acetate (7:3) eluant (Chromatographic separation of n-hexane extract of dried G. picrorrhiza Miq. root furnished two triterpenoids, identified as Eupha-8-23-dien-7-okso-260ac acid (1) and 3-hydroxy-isonicotinic acid (2).
