Near-field photocurrent in correlated 2D moiré materials
(English) Since the discovery of graphene, two-dimensional (2D) materials have garnered significant attention from the condensed matter physics community owing to their potential to engineer new physical, optical, and mechanical properties. The 2D material class now includes insulators (hexagonal boron nitride, hBN), semiconductors (transition metal dichalcogenides, TMDs), superconductors (NbSe2), topological insulators (Bi2Te3), and ferromagnets (CrI3). Beyond their inherent properties, layered materials allow for new characteristics through vertical stacking. Recent developments have led to the discovery of moiré materials, in which electronic properties are significantly altered by twisting adjacent 2D layers.
The discovery of superconductivity in magic-angle twisted bilayer graphene (MATBG) marked a milestone in moiré physics, initiating a rapidly growing field. The resulting phase diagrams of other high-Tc superconductors, MATBG, serve as a platform for exploring highly tunable strongly correlated states. At a twist angle of approximately 1.1°, the "magic angle,” MATBG shows significant band flattening near the Dirac points, reducing the Fermi velocity and making the kinetic energy smaller than the repulsive Coulomb interactions. This results in superconductivity and various emergent phases dominated by many-body physics, including correlated insulators, orbital magnetism, nematic orders, and topological states.
Moiré materials with large superlattice unit cells facilitate the exploration of strongly correlated phenomena at low charge carrier densities. Local back-gate electrodes enable capacitive tuning between strongly correlated states in-situ, a unique feature not available in other high-Tc superconductors. Advances in scanning probe techniques have allowed researchers to determine local properties at the sub-nanometer scale. Scattering-type scanning near-field optical microscopy (s-SNOM) is particularly suited for exploring MATBG because it can measure scattering and photovoltage signals at the nanometer scale while simultaneously probing mesoscopic electron transport.
Utilizing a groundbreaking cryo-near-field nanoscopy method, we will conduct s-SNOM measurements at cryogenic temperatures (as low as 8 K) to assess the optical and photovoltage near-field responses. This approach employs energies in the mid-infrared (MIR) and terahertz (THz) ranges, which align with the anticipated optical transition energies in the band structures of these materials. The primary objectives of this thesis are to ascertain the pertinent optical and thermoelectric coefficients in twisted moiré materials, evaluate the impact of inhomogeneities through gate-tuned near-field photovoltage and optical measurements, visualize correlated phenomena and broken symmetry states, and comprehend the nature of dephased signals in various measurements. This dissertation seeks to highlight crucial advancements in quantum phases, quantum nano-optoelectronics, and thermoelectricity, while supporting interest in unresolved questions, such as the characteristics of low-temperature correlated states. Additionally, it outlines future objectives for near- and far-field photovoltage experiments.(Català) Des del descubriment del grafè, els materials bidimensionals (2D) han acaparat una atenció significativa de la comunitat de física de la matèria condensada pel seu potencial de controlar noves propietats físiques, òptiques i mecàniques. La clase de materials 2D ara inclou aïllants (nitrur de bor hexagonal, hBN), semiconductors (metalls de transició dicalcogenoides, TMDs), superconductors (NbSe2), aïllants topològics (Bi2Te3), i ferroimans (CrI3). Més enllà de les seves propietats inherents, els materials a capes també permeten noves característiques a través de la seva construcció vertical. Els desenvolupaments recents han portat al descubriment dels materials moiré, on les propietats electròniques es veuen significativament alterades per la rotació de les capes 2D adjacents.
El descubriment de la superconductivitat en el grafè rotat d’angle màgic (MATBG) va marcar un hite en la física de moiré, iniciant un camp de ràpid creixement. Els materials moiré, amb grans cel·les de super-xarxes, faciliten l’exploració de fenòmens de fortes correlacions a baixes densitats de portadors. Els elèctrodes de les portes locals permeten l’ajust capacitiu entre estats de forta correlació in situ, una característica única no disponible en altres superconductors d’alta-Tc. Avenços en les tècniques d’escombratge ara permeten als investigadors determinar propietats locals en escales sub-nanomètriques. La microscòpia d’escombrament de camp proper de tipus dispersió (s-SNOM) està particularment preparada per a explorar MATBG, donat que pot mesurar el senyal de dispersió i fotovoltatge en escales nanomètriques al mateix temps que permet l’estudi mesoscòpic del transport electrònic.
Utilitzant el mètode innovador de nanoscòpia criogènica de camp-proper, realitzarem medicions s-SNOM a temperatures criogèniques (fins a 8 K) per evaluar la respota òptica i de fotovoltage en camp-proper. Aquest enfoc utilitzarà energies en el rang de l’infrarroig-mitjà (MIR) i els teraherzs (THz), que s’alineen amb les anticipades transicions òptiques de les estructures de bandes d’aquests materials. El principal objectiu d’aquesta tesis es determinar els coeficients òptics i termoelèctrics pertinents en materials moiré, evaluar l’impacte de les inhomogeneitats a través de mesures de fotovoltatge de camp-proper ajustant el voltatge de la porta, visualitzar fenòmens de correlacions i estats amb ruptures de simetria, i comprendre la naturalesa del retràs de la senyal de les mesures. Aquesta disertació busca subratllar els avenços crucials en fases quàntiques, nano-optoelectrònica quàntica, i termoelectricitat, mentres també dóna suport a l’interès en preguntes sense resoldre com les característiques dels estats correlacionats de baixes temperatures. Dynamics and function of the 3D chromatin structure in the germ line of vertebrates
How genomes are organized and regulated within the cell nucleus varies among cell types and taxa. In germ cells of eutherian mammals, chromatin undergoes extensive remodelling to accommodate key meiotic events such as chromosome pairing and recombination. Yet, whether these patterns are conserved in deeper phylogenetic branches of the Vertebrate Tree of Life remains unknown. Exploring the dynamics and function of the three-dimensional organization of the genome during the generation of germ cells across taxa is essential to understand the mechanisms underlying the heritability of chromatin architecture, and its role in fertility and evolution.
The main goal of this thesis was to investigate the dynamics of meiosis progression and chromatin folding during spermatogenesis in representative vertebrates (eutherian mammals, marsupials and reptiles), with a special interest in the structural and functional roles of meiotic cohesins. For this aim, three specific objectives were established: (i) characterize meiosis dynamics among vertebrates, (ii) examine the spatial organization of the genome during spermatogenesis, and (iii) identify the structural and functional roles of cohesins during spermatogenesis. This was archived by combining comparative cytological analysis, fluorescence activated cell sorting (FACS) and high-throughput sequencing technologies, including chromosome conformation capture (Hi-C) and single cell RNA sequencing (sc-RNAseq). We generated new Hi-C data from flow-sorted germ cells from representative vertebrates (tammar wallaby and bearded dragon) along with mice lacking the meiotic cohesin subunit RAD21L. This was combined with single cell transcriptomics profiles throughout spermatogenesis. The data generated was compared with available data from other mammals to provide a comprehensive view of the structural and functional roles of chromatin during germ line formation.
We observed that the testicular organization of germ cells follows a non-cystic organization in amniotes, with meiotic dynamics being highly conserved among vertebrates, including canonical events such as bouquet formation, double strand break (DSB) formation and repair, homologous chromosomes synapsis, and meiotic sex chromosome inactivation (MSCI). Despite this conservation, lineage-specific dynamics were detected, including the presence of a prominent bouquet in reptiles and a transient configuration in most marsupials, the early pair and clustering of micro-chromosomes in reptiles, and the elongation of telomeres during male prophase I in dasyurids. Additionally, different sex chromosome systems (i.e. XY, XY1Y2 and ZW) revealed distinct strategies of sex chromosome pairing and silencing, with implications for the evolution of sex chromosomes.
Our findings also unveiled fundamental principles of chromatin architecture during vertebrate spermatogenesis. At the hierarchical level of DNA loops, we identified a correlation between synaptonemal complex length, DNA loop size and DSB rates that was conserved between species. DNA loop size was likely determined by genome size, with species with large genomes organized into short chromosomal axes with long meiotic DNA loops, but short DNA loops at post-meiotic stages. We also observed a conserved remodelling of chromatin architecture across species, with both compartments and TADs being attenuated during prophase I. Despite these conserved dynamics, our comparative analysis revealed lineage-specific patterns of chromatin organization during meiosis, where chromosome length, chromosome morphology and DNA loop size are key factors.
This thesis also uncovered the structural and functional roles of cohesins during mouse spermatogenesis. Loss of the meiotic subunit RAD21L impacts both chromatin architecture and gene expression, resulting in changes at different levels, including longer DNA loops, loss of TADs, compartmentalization strengthening, generation of aberrant interactions, misregulation of key meiotic genes, and partial disruption of the MSCI.
Overall, our findings provide new insights into the evolutionary dynamics of meiosis progression and chromatin architecture in vertebrate germ cells, along with the structural and functional roles of meiotic cohesins. Untangling lineages: population genetics and phylogenomics of Auxis Rochei and Euthynnus Alletteratus
ENG- The Little Tunny (Euthynnus alletteratus) and the Bullet Tuna (Auxis rochei) are two fish species within the tuna family that are closely related to larger tunas. Although they have a lower market value, thousands of tonnes of these species are captured each year, making the study of their populations essential for ensuring sustainable exploitation. Genetic tools play a vital role in this research by allowing us to examine genetic diversity, assess its distribution across their geographic ranges and trough time, and identify distinct populations. Additionally, molecular methods can validate the correct identification of captured species.
This thesis investigates these aspects for both species, focusing on population differentiation, the levels of genetic diversity, and the accuracy of visual species identification through genetic assignment. Given their evolutionary relationship and similar ecological behaviours, this thesis also aimed to compare the dynamics between species. This study both classical genetic markers and high-throughput sequencing techniques have been analysed.
For the Little Tunny, results revealed significant genetic differences between individuals in the North Atlantic and those in the tropical Atlantic, indicating that they may represent two distinct species. Within each species, we observed shallower genetic differences among localities, suggesting the presence of multiple populations.
Regarding the Bullet Tuna, we identified two populations in the Mediterranean Sea: one around the Iberian Peninsula and another in North Africa. Notably, our study revealed that some captures of Mediterranean Bullet Tuna were actually Frigate Tuna, a closely related species previously believed to be absent from the region.
The results of this thesis should guide the implementation of fishery management policies that ensure sustainable fisheries. These practices include accurate capture and stock identification. Recognizing the two Little Tunny species as separate fishery stocks is the most urgent policy that should be implemented by the relevant agencyCAT- La bacoreta (Euthynnus alletteratus) i la melva (Auxis rochei) són dues espècies de peixos que pertanyen al grup dels petits túnids. Espècies molt properes en termes evolutius a les tonyines. Com elles, també són explotades com a recurs pesquer i, tot i que tenen un valor de mercat més baix, cada any tenen unes captures molt elevades. Per això, l’estudi i seguiment de les seves poblacions és essencial per tal de garantir una explotació sostenible. En aquest sentit, les eines genètiques juguen un paper fonamental, ja que permeten estudiar la diversitat genètica, determinar si aquesta es distribueix de manera diferencial en l’espai (i el temps) i, per tant, saber si existeixen poblacions genèticament diferenciades. A més, els mètodes moleculars també permeten validar que l’espècie capturada sigui realment l’espècie objectiu.
En aquesta tesi s’ha estudiat aquests aspectes per ambdues espècies, investigant si hi havia més d’una població en cadascuna d’elles, quina era la diversitat genètica de les seves poblacions i si l’assignació visual de les espècies coincidia amb la informació genètica. A més, donat que són dues espècies molt properes evolutivament i amb comportaments ecològicament similars, també s’han comparat les seves dinàmiques poblacionals. L’estudi s’ha realitzat tant amb marcadors genètics clàssics com amb tècniques de seqüenciació massiva.
Pel que fa a la bacoreta, els resultats han mostrat que les diferències genètiques entre els individus del nord de l’Atlàntic i els del tròpic són tan elevades que, en realitat, podrien correspondre a dues espècies separades. A més, hem observat que també hi ha diferenciació genètica entre les diferents localitats estudiades, indicant l’existència de poblacions diferents dins de cadascuna de les dues possibles espècies.
En relació amb la melva, hem identificat dues poblacions dins del Mediterrani, separant els individus al voltant de la península Ibèrica dels del nord d’Àfrica. Els nostres estudis també han revelat que part de les captures de melva al Mediterrani són en realitat de melva llisa, una espècie propera que es pensava absent de la regió.
Els resultats obtinguts en aquesta tesi haurien de servir per implementar mesures de gestió de les pesqueries que garanteixin la sostenibilitat per a les espècies estudiades, incloent el correcte reconeixement de les captures i dels estocs. Especialment en el camp de l'aigua per la seva vulnerabilitat, multifuncionalitat i rellevància, així com per la seva vinculació amb l'emergència climàtica. La seguretat hídrica s'ha convertit en una prioritat en les polítiques relacionades amb el desenvolupament sostenible. Aquesta investigació se suma als plantejaments sobre la necessitat d'un nou paradigma en la gestió dels recursos hídrics, amb un enfocament sistèmic, que realça la resiliència i la gestió de l'anomenada “aigua verda” i, per tant, el nexe amb la gestió dels sols. En un adequat maneig del sòl agropecuari rau una bona part de la solució per a la seguretat hídrica, alimentària i climàtica. S'ha evidenciat en múltiples experiències i estudis que mitjançant pràctiques d'agricultura restaurativa es pot revertir la degradació de terres. No obstant això, representa un gran repte estendre (escalar) aquestes pràctiques a tot un territori, més enllà de la vida dels projectes demostratius. La complexitat dels factors limitants requereix mecanismes de governança robusts; cosa que també és un gran repte en si. La present investigació demostra avanços en agricultura restaurativa i gestió hídrica en fomentar processos de governança local en el Cacahuatique (El Salvador, Amèrica Central). Utilitzant la teoria dels sistemes socioecològics, la metodologia d'Investigació-Acció-Participativa (IAP) i els projectes de l'ONG CRS com a instrument d'acció, es va impulsar un procés de enfortiment de la governança hídrica durant 6 -10 anys (segons la zona). Es va basar en implicar als operadors comunitaris d'aigua i les alcaldies en la protecció de les àrees de recàrrega hídrica, i va incloure la creació de tres Fons d'Aigua i Agricultura (FOAG), com a plataformes de finançament i governança multiactors, una experiència inèdita en contextos rurals de la regió. La investigació aporta reflexions sobre èxits, frustracions i reptes, tant en la generalització de l'adopció de pràctiques d'agricultura restaurativa com en els processos de governança, particularment en el model de Fons d'Aigua. Entre les principals conclusions, cal esmentar que la utilització de l'enfocament de protecció de fonts ha estat molt útil per a la mobilització social. No obstant això, la creació d'estructures i xarxes de treball no obté els resultats esperats sense suficient lideratge i cohesió social. La restauració ambiental efectiva necessita de la restauració del capital i teixit social (i viceversa), i això requereix temps i intervencions integrals. La investigació conclou que ha d'haver un canvi en els models de projectes de desenvolupament i en el disseny dels seus programes de formació. La ciència de l'escalament defensa la consideració de tots els factors, especialment els estructurals, treballant la dimensió col·lectiva i la individual, per tal de modificar adequadament la realitat. Tornar a la font és una crida a un retorn a l'essencial: la protecció de les terres d'on prové l'aigua i forjar persones i societats que siguin motors de canvi positiu.La interrelación, complejidad, variabilidad e incertidumbre creciente del mundo actual requieren de enfoques de gestión más sistémicos. Muy especialmente en el campo del agua por su vulnerabilidad, multifuncionalidad y relevancia, así como por su vinculación con la emergencia climática. La seguridad hídrica se ha convertido en una prioridad en las políticas vinculadas al desarrollo sostenible. Esta investigación se suma a los planteamientos de la necesidad de un nuevo paradigma en la gestión de los recursos hídricos, con enfoque sistémico, realzando la resiliencia y la gestión de la denominada “agua verde” y, por tanto, el nexo con la gestión de los suelos. La presente investigación demuestra avances en agricultura restaurativa y gestión hídrica al fomentar procesos de gobernanza local en el cerro Cacahuatique (El Salvador, Centro América). Entre las principales conclusiones, cabe mencionar que la utilización del enfoque de protección de fuentes ha sido muy útil para la movilización social. Sin embargo, la creación de estructuras y redes de trabajo no obtienen los resultados esperados sin liderazgos y cohesión social suficiente. La restauración ambiental efectiva necesita de la restauración del capital y tejido social (y viceversa), y ello requiere de tiempo y de intervenciones integrales. La investigación concluye que debe de haber un cambio en los modelos de proyectos de desarrollo y en el diseño de sus formaciones. La ciencia del escalamiento aboga por considerar todos los factores, especialmente los estructurales, trabajando la dimensión colectiva y la individual, para poder modificar la realidad adecuadamente. Volver a la fuente es un llamado a un regreso a lo esencial; la protección de las tierras de dónde proviene el agua y forjar personas y sociedades que sean motor de cambio positivo.The interrelatedness, complexity, variability and increasing uncertainty of today's world require more systemic management approaches. This is especially true in the field of water due to its vulnerability, multifunctionality and relevance, as well as its link with climate emergency. Water security has become a priority in policies linked to sustainable development. This research adds to the need for a new paradigm in the management of water resources, with a systemic approach, enhancing resilience and the management of the so-called “green water” and, therefore, the link with soil management. A good part of the solution for water, food and climate security lies in adequate agricultural soil management. Multiple experiences and studies have shown that land degradation can be reversed through restorative agricultural practices. However, it is a great challenge to extend (scale up) these practices to an entire territory, beyond the life of the demonstration projects. The complexity of the limiting factors demands robust governance mechanisms, which presents a major challenge in itself. This research demonstrates advances in restorative agriculture and water management by fostering local governance processes in Cerro Cacahuatique (El Salvador, Central America). Using social-ecological systems theory, participatory action-research (PAR) methodology and CRS NGO projects as a tool for action, a process of strengthening water governance was promoted for 6-10 years (depending on the area). This research engaged community water operators and municipalities in the protection of water recharge areas and fostered the creation of three Water and Agriculture Funds (FOAG), as financing and multi-stakeholder governance platforms, an unprecedented experience in rural contexts in the region. The research provides reflections on achievements, failures and challenges, both in the generalization of the adoption of restorative agriculture practices and in governance processes, particularly in the Water Funds model. Among the main conclusions, it is worth mentioning that the use of the source protection approach has been very useful for social mobilization. However, the creation of structures and networks does not achieve the expected results without sufficient leadership and social cohesion. Effective environmental restoration relies on the restoration of social capital and social fabric (and vice versa), and this requires time and comprehensive interventions. This research concludes that there must be a change in the models of development projects and in the design of their capacity strengthening processes. The scaling-up approach emphasizes taking all factors into account, particularly structural ones, and addressing both collective and individual dimensions to effectively bring about meaningful change. Evolutionary dynamics of cancer drug resistance and prediction of tumor modes of growth
Cancer remains a significant challenge in healthcare and is a leading concern worldwide. Despite advances in cancer therapy and the identification of numerous actionable events, relapse and drug resistance continue to complicate treatment outcomes. This thesis investigates the evolution of cancer from the perspectives of population genetics, computational simulations, and data analysis .We present a novel method developed to infer patterns of tumor growth in both primary tumors and metastatic sites using approximate Bayesian computation. By considering the sampling noise in DNA sequencing that occurs with the construction of the variant allele frequency distribution, we enable the reconstruction of tumor growth modes in samples with both high and low coverage distributions. Furthermore, we investigate the phenomenon of drug resistance using numerical simulations. We show that using the variant allele frequency distribution of a cancer sample, together with a target of drug resistance genes, informs about the time to relapse.
Next, we analyze genomic patient data to identify genes that, when mutated, may contribute to either drug resistance or promote the development of metastases. This involves comparing cohort-wise selection estimates between primary tumors and their metastatic counterparts. Finally, we analyze the influence of genetic heterogeneity on the level of gene expression counts from single-cell RNA data in patients with acute myeloid leukemia. This work advances the field of cancer evolution by providing a new method to infer tumor modes of growth and helping to understand the measurability of cancer drug resistance in current sequencing methods.El càncer segueix sent un repte significatiu en l'àmbit de la salut i una de les principals preocupacions a nivell mundial. Malgrat els avenços en teràpies i la identificació d’objectius rellevants, les recaigudes i la resistència a fàrmacs segueixen complicant els pronòstics dels tractaments. Aquesta tesi investiga l'evolució del càncer des de les perspectives de la genètica de poblacions, les simulacions computacionals i l'anàlisi de dades.Presentem un mètode nou desenvolupat amb l’objectiu d’inferir patrons de creixement tumoral tant en tumors primaris com en metastàtics utilitzant el càlcul bayesià aproximat. Considerant el soroll de mostreig en la seqüenciació de l'ADN que es produeix en la construcció de la distribució de freqüència de l’al·lel alternatiu, possibilitem la reconstrucció dels modes de creixement del tumor en mostres amb distribucions de cobertura tant altes com baixes.A més, investiguem el fenomen de la resistència a fàrmacs utilitzant simulacions numèriques. Mostrem que utilitzant la distribució de freqüència de l’al·lel alternatiu d'una mostra de càncer juntament amb un conjunt de gens de resistència a fàrmacs obtenim informació sobre el temps fins a la recaiguda.A continuació, analitzem dades genòmiques de pacients per identificar gens que al mutar poden contribuir tant a la resistència a fàrmacs com a promoure el desenvolupament de metàstasis. Això implica comparar estimacions de selecció per cohorts de tumors primaris i els seus equivalents metastàtics.Finalment, analitzem la influència de la heterogeneïtat genètica en el nivell de recomptes d'expressió gènica a partir de dades d'ARN de cèl·lula única en pacients amb leucèmia mieloide aguda.Programa de Doctorat en Biomedicin
Association between the degree of immunosuppression measured by nuclear factor of activated T- cells regulated genes and cytomegalovirus infection in lung transplant recipients
El trasplantament de pulmó (TP) és l’última opció terapèutica per a pacients amb malalties respiratòries en fase terminal, però presenta l’esperança de vida més baixa entre els trasplantaments d’òrgans sòlids (TOS). L’exposició constant dels pulmons a l’entorn extern augmenta les infeccions, responsables del 37% de les morts durant el primer any després del TP. A llarg termini, el principal obstacle és el desenvolupament de la disfunció crònica de l’empelt pulmonar (CLAD), amb una taxa de mortalitat del 25-30% en els primers 3-5 anys. Aquests problemes estan interrelacionats, ja que les infeccions són un factor de risc conegut per a la CLAD.
Determinar la dosi òptima d’immunosupressors és crucial per evitar infeccions i tumors per una immunosupressió excessiva, així com el rebuig agut i la CLAD causats per una immunosupressió insuficient. L’ajustament de la dosi és complicat a causa de l’alta variabilitat interindividual. Actualment, es controlen els nivells mínims de tacrolimus, però aquesta mesura farmacocinètica no reflecteix amb precisió l’estat immunosupressor del pacient. Per això, s’investiguen biomarcadors no invasius i eines farmacodinàmiques per avaluar millor la immunosupressió i el risc d’infecció.
El citomegalovirus (CMV) és una infecció oportunista freqüent i clínicament rellevant després del TOS, augmentant el risc de morbiditat i mortalitat. Els seus efectes immunomoduladors incrementen la susceptibilitat a altres infeccions, el rebuig agut i la CLAD. Les taxes de malaltia per CMV en TP oscil·len entre el 8% i el 55%, segons el tipus i durada de la profilaxi antiviral, que és l’estàndard terapèutic. Tot i que la profilaxi redueix el risc d’infecció, el seu ús prolongat pot provocar efectes adversos. Encara que els pacients seropositius es consideren immunitzats, un 25% desenvolupen infeccions i un 15% pateixen la malaltia en els primers 18 mesos després del TP. L’estratificació actual del risc es basa només en marcadors humorals, amb pocs estudis que avaluïn la resposta immune cel·lular específica contra el CMV (CMV-CMI) en receptors de TP (RTP).
L’objectiu d’aquesta tesi va ser avaluar dues proves immunològiques prometedores: (1) la mesura de l’expressió gènica residual de tres gens regulats pel factor nuclear de cèl·lules T activades (NFAT) (IL-2, IFN-γ i GM-CSF) i (2) el test ELISPOT per al CMV, per millorar el monitoratge dels fàrmacs immunosupressors en RTP i identificar pacients amb més risc d’infeccions, especialment per CMV.
El primer estudi va trobar que els nivells mínims de tacrolimus no es correlacionaven amb el nivell d’immunosupressió mesurat per NFAT-RGE, ni predeien el risc d’infecció o rebuig. En canvi, els nivells màxims de tacrolimus es van correlacionar amb el NFAT-RGE. Un valor de NFAT-RGE <30% es va associar amb un risc més alt d’infeccions bacterianes greus. Per tant, la introducció del NFAT-RGE a la pràctica clínica podria convertir-se en una eina no invasiva valuosa. També es va suggerir la genotipificació dels enzims CYP3A4 i CYP3A5 per identificar metabolitzadors ràpids, amb més risc d’infeccions, i metabolitzadors lents, amb més risc de rebuig.
En el segon estudi, el test CMV ELISPOT no va predir de manera fiable quins pacients desenvoluparien infecció per CMV. No obstant això, quan es van utilitzar dos antígens combinats, es va observar que pacients amb una resposta cel·lular feble o intermèdia tenien una taxa d’infecció més alta entre els 6 i els 18 mesos post-TP. Aquells amb una resposta forta tenien un 49% menys de probabilitat de patir una infecció significativa per CMV.
En conclusió, aquests estudis contribueixen a la monitorització personalitzada de la teràpia immunosupressora amb tacrolimus i a l’avaluació del risc d’infecció per CMV en pacients de TP, obrint noves perspectives per a futures investigacions.El trasplante de pulmón (TP) es la última alternativa para pacientes con enfermedades respiratorias en fase terminal, pero sigue siendo el trasplante de órgano sólido (TOS) con menor esperanza de vida. El principal desafío es la disfunción crónica del injerto (DCI), responsable de una alta mortalidad entre tres y cinco años post-TP. Además, las infecciones representan el 37% de las muertes durante el primer año y son un factor de riesgo para desarrollar DCI debido a la constante exposición de los pulmones al ambiente externo.
El citomegalovirus (CMV) es una infección oportunista común tras el TOS, que agrava el riesgo de complicaciones como infecciones, rechazo agudo y DCI. A pesar de la profilaxis antiviral estándar, que reduce el riesgo de enfermedad por CMV, persisten riesgos significativos. Las tasas de enfermedad por CMV oscilan entre el 8% y el 55%, según la duración y el tipo de profilaxis. Aunque se asume que los pacientes seropositivos tienen inmunidad, un 25% desarrolla infección y un 15% enfermedad por CMV en los primeros 18 meses post-TP. Actualmente, la estratificación del riesgo se basa en marcadores humorales, mientras que la evaluación de la respuesta inmune celular es limitada.
La tesis se centra en mejorar el manejo inmunológico de los receptores de trasplante de pulmón (RTP) mediante dos pruebas inmunológicas: la medición de la expresión génica regulada por NFAT (NFAT-RGE) y el test de ELISPOT para CMV. El objetivo es identificar a los pacientes con mayor riesgo de infecciones y mejorar la personalización de la inmunosupresión.
En la primera parte, se evaluó la relación entre los niveles de tacrolimus y NFAT-RGE. Se encontró que los niveles valle de tacrolimus no se correlacionan con la inmunosupresión ni predicen infecciones o rechazo, mientras que los niveles pico sí lo hacen. Valores de NFAT-RGE inferiores al 30% se asociaron con un mayor riesgo de infecciones bacterianas graves, sugiriendo su posible uso como herramienta de monitorización clínica. Además, la genotipificación de los citocromos P450 (CYP3A4 y CYP3A5) podría ayudar a identificar a pacientes con mayor susceptibilidad a infecciones o peores resultados en el injerto, según su capacidad de metabolizar el tacrolimus.
En la segunda parte, se evaluó el test de CMV ELISPOT, que mide la respuesta inmune celular específica. Aunque no predijo de manera precisa qué pacientes desarrollarían infección por CMV, sí mostró su utilidad al combinar ambos antígenos analizados. Los pacientes con respuesta celular débil o intermedia frente a CMV presentaron una mayor tasa de infección entre seis y 18 meses después del TP, mientras que aquellos con una respuesta celular fuerte tuvieron un 49% menos de riesgo de infección significativa. Esto permite una mejor evaluación del riesgo, facilitando ajustes en la inmunosupresión y la profilaxis antiviral.
En conclusión, los estudios de esta tesis proponen estrategias para personalizar el manejo inmunológico en RTP. La medición de NFAT-RGE podría convertirse en una herramienta no invasiva para ajustar la inmunosupresión, mientras que el test de ELISPOT podría ayudar a valorar el riesgo de infección por CMV en pacientes seropositivos. Estos hallazgos abren nuevas vías para futuras investigaciones clínicas.Lung transplantation (LT) is the last therapeutic option for patients with end-stage respiratory diseases. Despite advancements, LT has the lowest survival rate among all solid organ transplants (SOT), with 37% of deaths in the first year due to infections caused by the lungs’ exposure to the environment. Chronic lung allograft dysfunction (CLAD) remains the main barrier to long-term survival, causing 25-30% mortality within 3-5 years post-LT. Infections also increase the risk of CLAD, highlighting the need for better post-transplant management.
Proper immunosuppressant dosing is critical to balance infection prevention and rejection control. However, due to significant inter- and intra-patient variability, monitoring tacrolimus trough levels remains insufficient for assessing immunosuppressive states. Research is focused on identifying non-invasive biomarkers and pharmacodynamic tools to improve personalized therapy.
Cytomegalovirus (CMV) is a significant opportunistic infection after SOT, raising the risk of infections, acute rejection, and CLAD through its immunomodulatory effects. CMV disease rates range from 8% to 55%, depending on prophylaxis duration and type. Universal antiviral prophylaxis, usually given for six to 12 months, reduces CMV disease risk but can cause side effects. Notably, 25% of CMV-seropositive patients still develop CMV infection, and 15% develop CMV disease within 18 months post-LT. Although CMV-specific cellular immune response (CMV-CMI) is essential for infection control, current risk assessment focuses only on humoral markers, leaving a gap in evaluating CMV-CMI in LT recipients (LTR).
This thesis aimed to evaluate two promising immunological tests: (1) measuring residual gene expression of three NFAT-regulated genes (IL-2, IFN-γ, and GM-CSF), and (2) using the CMV enzyme-linked immunosorbent spot (ELISPOT) test. These tests were designed to improve therapeutic drug monitoring and identify LTRs at higher infection risk, especially for CMV.
The first study found that tacrolimus trough levels, currently used for drug monitoring, did not correlate with immunosuppression levels measured by NFAT-regulated gene expression (NFAT-RGE) or predict infection or rejection risk. In contrast, tacrolimus peak levels showed a strong correlation with NFAT-RGE. Importantly, NFAT-RGE values below 30% were linked to a higher risk of severe bacterial infections, suggesting its potential as a non-invasive infection risk assessment tool. Additionally, pre-LT genotyping of cytochromes P450 3A4 (CYP3A4) and P450 3A5 (CYP3A5) could help identify fast metabolizers (FM), prone to infections, and poor metabolizers (PM), at risk of worse graft outcomes due to low tacrolimus levels.
The second study assessed the CMV ELISPOT test, which measures CMV-specific immune responses. Although it could not reliably predict which patients would develop CMV infection, its predictive value improved when both antigens were tested together. Among seropositive patients, those showing intermediate (response to one antigen) or weak (response to neither antigen) CMV-CMI had significantly higher infection rates between six and 18 months post-LT. In contrast, patients with strong CMV-specific responses were 49% less likely to develop severe CMV infections. This approach could guide personalized immunosuppression adjustments and CMV prophylaxis modifications.
In conclusion, this thesis highlights the potential of NFAT-RGE measurement as a tool for personalized immunosuppressive therapy and the CMV ELISPOT test for assessing CMV infection risk in LTRs. Oligonucleotide-Drug Conjugates and Alternative Methods to Selectively Reactivate the Inactive X-Chromosome in Female Somatic Cells
En aquesta tesi, proporciono evidència que un inhibidor de DNMT1 pot ser enriquit en un únic cromosoma mitjançant la conjugació amb un oligonucleòtid antisentit. Aquest nou tipus de bioconjugat, anomenat conjugat oligonucleòtid-fàrmac (ODC, per les seves sigles en anglès), pot ser utilitzat per a l'edició dirigida de l'epigenoma. Un ODC basat en XIST pot reactivar el cromosoma X inactiu en línies cel·lulars de diversos trastorns genètics lligats al cromosoma X. S'ha desenvolupat una variant d'aquest enfocament en què s'empra química bioortogonal per permetre la separació espacial de l'ASO i la petita molècula durant l'administració, amb posterior conjugació en condicions fisiològiques.En esta tesis, presento evidencia de que un inhibidor de DNMT1 puede ser enriquecido en un solo cromosoma mediante la conjugación con un oligonucleótido antisentido. This novel type of bioconjugate , termed oligonucleotide-drug conjugate (ODC), can be used for targeted epigenome editing. An XIST-based ODC can reactivate the inactive X-chromosome in cell lines of various X-linked genetic disorders. An alteration of this approach is developed in which bioorthogonal chemistry is employed to allow spatial separation of the ASO and small molecule at administration with subsequent conjugation under physiological conditions.Universitat Pompeu Fabra. Through this rigorous study, the reader will become familiar not only into the latest legislative developments —such as the introduction of ADR as a procedural requirement and its impact on the order for payment procedure— but also, with how other controversial issues and interpretative gaps can be resolved.

Aquesta tesi explora la interacció de la frustració magnètica en òxids de perovskita en capes, amb l'objectiu de descobrir nous mecanismes per estabilitzar el magnetisme espiral d'alta temperatura i la multiferroicitat. La recerca se centra principalment en YBaCuFeO (YBCFO).
YBCFO emergeix com un sistema remarcable que exhibeix espirals d'espín quirals d'alta temperatura impulsades per un mecanisme no convencional d'"ordre per desordre". Tot mantenint un desordre Cu/Fe constant, vam explorar múltiples estratègies per millorar la frustració magnètica i l'estabilitat espiral. Els enfocaments físics van incloure estudis de pressió hidrostàtica, que van revelar una reducció significativa de (12 K/GPa) mitjançant difracció combinada de raigs X/neutrons, magnetometria i espectroscòpia Raman. Els enfocaments químics es van centrar en la modificació de la geometria piramidal i el gruix de la capa mitjançant substitucions estratègiques. El dopatge de Zn al lloc B per a Cu va millorar l'estabilitat espiral reduint l'elongació piramidal sense generar fases magnètiques addicionals per sota del punt tricrític. A concentracions més altes de Zn, es va descobrir un segon estat magnètic espiral amb un vector de propagació diferent = (1/2, 1/2, ), que ampliava el diagrama de fases més enllà del punt triple. Les substitucions combinades de Sr al lloc A i Co al lloc B van revelar efectes no additius del dopatge de doble lloc, amb tres regions diferents identificades com a contingut de Sr variat, demostrant una interacció complexa entre les modificacions estructurals i l'ordenació magnètica.
Els estudis de dispersió de neutrons de monocristall sota camps magnètics van demostrar la capacitat de modificar el vector de propagació magnètica mitjançant la manipulació del camp. Aquest descobriment, combinat amb l'observació de transicions induïdes pel camp entre els estats magnètics espiral i col·lineal, va mostrar un procediment per millorar la temperatura d'ordenació espiral dels cistals YBCFO tal com han crescut. Hem obtingut un cistal YBCFO amb l'ordre espiral inconmensurable per sobre de la temperatura ambient. Això representa un avenç significatiu cap a les aplicacions a temperatura ambient.
En paral·lel, la caracterització estructural i magnètica i l'anàlisi de simetria dels compostos magnetoelèctrics RCrTiO₂ (Nd, Tb) van proporcionar informació per comprendre el mecanisme impulsor de la multiferroicitat en aquests compostos. Es van investigar i descriure els efectes impulsats per la frustració que il·lustren la importància de l'ordre no col·lineal en aquesta estructura. Donen compte de les transicions magnetoelèctriques, multiferroiques i estructurals i magnètiques en aquests titanats frustrats on l'acoblament espín-xarxa és clau per a l'estabilització de les diferents fases.Esta tesis explora la interacción de la frustración magnética en óxidos de perovskita estratificados, con el objetivo de descubrir nuevos mecanismos para estabilizar el magnetismo espiral de alta temperatura y la multiferroicidad. YBCFO emerge como un sistema notable que exhibe espirales de espín quirales de alta temperatura impulsadas por un mecanismo no convencional de "orden por desorden". Manteniendo constante el desorden de Cu/Fe, exploramos múltiples estrategias para mejorar la frustración magnética y la estabilidad espiral. Los enfoques físicos incluyeron estudios de presión hidrostática, que revelaron una reducción significativa en (12 K/GPa) mediante difracción combinada de rayos X/neutrones, magnetometría y espectroscopia Raman. Los enfoques químicos se centraron en modificar la geometría piramidal y el espesor de capa mediante sustituciones estratégicas. El dopaje de Zn en el sitio B para Cu mejoró la estabilidad espiral al reducir la elongación piramidal sin generar fases magnéticas adicionales por debajo del punto tricrítico. A concentraciones más altas de Zn, se descubrió un segundo estado magnético espiral con un vector de propagación = (1/2, 1/2, ) distinto, lo que extiende el diagrama de fases más allá del punto triple. Las sustituciones combinadas de Sr en el sitio A y Co en el sitio B revelaron efectos no aditivos del dopaje de doble sitio, con tres regiones distintas identificadas con variación en el contenido de Sr, lo que demuestra una compleja interacción entre las modificaciones estructurales y el ordenamiento magnético.
Los estudios de dispersión de neutrones de monocristales bajo campos magnéticos demostraron la capacidad de modificar el vector de propagación magnética mediante la manipulación del campo. Este descubrimiento, combinado con la observación de transiciones inducidas por el campo entre estados magnéticos espirales y colineales. Además, mostramos un procedimiento para mejorar la temperatura de ordenamiento espiral de cristales de YBCFO en estado bruto de crecimiento. Hemos obtenido un cristal de YBCFO con un orden espiral desproporcionado por encima de la temperatura ambiente. Esto representa un avance significativo hacia las aplicaciones a temperatura ambiente.
Paralelamente, la caracterización estructural y magnética, así como el análisis de simetría de los compuestos magnetoeléctricos RCrTiO₄₃ (Nd, Tb), proporcionaron información para comprender el mecanismo que rige la multiferroicidad en estos compuestos. Se investigaron y describieron los efectos de frustración que ilustran la importancia del orden no colineal en esta estructura. Estos efectos explican las transiciones magnetoeléctricas, multiferroicas, estructurales y magnéticas en estos titanatos frustrados, donde el acoplamiento espín-red es clave para la estabilización de las distintas fases.This thesis explores the interplay of magnetic frustration in layered perovskite oxides, aiming to uncover novel mechanisms for stabilizing high-temperature spiral magnetism and multiferroicity. YBCFO emerges as a remarkable system exhibiting high-temperature chiral spin spirals driven by an unconventional 'order-by-disorder' mechanism. While maintaining constant Cu/Fe disorder, we explored multiple strategies to enhance magnetic frustration and spiral stability. Physical approaches included hydrostatic pressure studies, which revealed a significant reduction in (12 K/GPa) through combined X-ray/neutron diffraction, magnetometry, and Raman spectroscopy. Chemical approaches focused on modifying pyramidal geometry and layer thickness through strategic substitutions. B-site Zn doping for Cu enhanced spiral stability by reducing pyramidal elongation without generating additional magnetic phases below the tricritical point. At higher Zn concentrations, a second spiral magnetic state with distinct propagation vector = (1/2, 1/2, ) was discovered, extending the phase diagram beyond the triple point. Combined A-site Sr and B-site Co substitutions revealed non-additive effects of dual-site doping, with three distinct regions identified as Sr content varied, demonstrating complex interplay between structural modifications and magnetic ordering.
Single crystal neutron scattering studies under magnetic fields demonstrated the ability to modify the magnetic propagation vector through field manipulation. This discovery, combined with the observation of field-induced transitions between spiral and collinear magnetic states. Furthermore, we show a proceduce to enhance the spiral ordering temperature of as-grown YBCFO cystals. We have obtained an YBCFO cystal with the incommensurate spiral order above ambient temperature. This represents a significant advance towards room temperature applications.
In parallel, structural and magnetic characterization and symmetry analysis of the magnetoelectric RCrTiO compounds (Nd, Tb) provided insights to understand the driving mechanism of multiferroicity in these compounds. Frustration driven effects were investigated and described that illustrate the importance of the non-collinear order in this structure. They give account of the magnetoelectric, multiferroic and structural and magnetic transitions in these frustrated titanates where spin-lattice coupling is key for the stabilization of the distinct phases.Universitat Autònoma de Barcelona. Transformative insights and evolving roles of women managers in Iran’s tourism industry
ENG- The thesis consists of three research articles: two research papers and one research note. It examines the transformative impacts of tourism leadership roles on Iranian women managers, the evolving trajectories of their managerial self-perception, and their potential to create broader workplace and social change. These themes are explored across three distinct yet interconnected research pieces. Utilizing qualitative methodologies together with a feminist epistemology (Aitchison, 2005; Pritchard, 2014), these three studies collectively generate a multi-level perspective on women managers in tourism, culminating in a conceptual framework that establishes a model for understanding how women's leadership development in restrictive contexts generates valuable shifts in organizational cultures and societal practices. By mapping the interplay between internal transformation and structural change, this study offers a new theoretical lens while equipping tourism institutions with actionable insights for fostering equitable ecosystems.
Iran is the chosen case study for this thesis, offering valuable and insightful perspectives for several reasons. While recent studies on gender in tourism have increasingly focused on women in developing countries (Efthymiadou & Farmaki, 2023), in the field of women in tourism management research has predominantly centered on Western contexts and fewer studies have explored this topic in developing regions. Examining Iran is not just another different geographical setting for a case study, as this case amplifies the voices of women in a restrictive society. Iran’s gender apartheid (ranked 143/146 in the Global Gender Gap Report 2024) and the widespread protests of 2022 serve as strong evidence of a unique case of systemic gender discrimination and inequality across the world. These factors make Iran a compelling and necessary setting for an in-depth exploration of women in tourism management.
To articulate the complex and layered nature of change revealed in the study, the thesis proposes a transformational framework grounded in existing multidimensional models. This framework traces the evolving internal traits and values of Iranian women in tourism management, demonstrating how leadership roles in the tourism industry can extend far beyond financial empowerment to shape their personalities and perspectives on gender. The shifting self-perceptions of these women, particularly during moments of social unrest in Iran, reveal that even within a restrictive patriarchal society, women continue to evolve. These women’s negotiation of gendered power within organizational structures and their capacity to act as transformative agents is further expressed through subtle yet powerful shifts in language, offering deep insight into how personal change can influence broader social transformationCAT- La tesi consta de tres articles de recerca: dos articles i una nota de recerca. S’hi examinen els impactes transformadors dels rols de lideratge en el turisme sobre les dones directives iranianes, l’evolució de la seva autopercepció com a gestores, i el seu potencial per generar canvis més amplis en l’àmbit laboral i social. Aquests temes es desenvolupen a través de tres peces de recerca distintes però interconnectades. Mitjançant metodologies qualitatives i una epistemologia feminista (Aitchison, 2005; Pritchard, 2014), els tres estudis aporten una perspectiva multinivell sobre les dones directives en el turisme, culminant en un marc conceptual que estableix un model per entendre com el desenvolupament del lideratge femení en contextos restrictius pot generar canvis valuosos en les cultures organitzatives i les pràctiques socials. En cartografiar la interacció entre la transformació interna i el canvi estructural, aquest treball ofereix una nova mirada teòrica alhora que proporciona a les institucions turístiques eines útils per fomentar ecosistemes més equitatius.
L’Iran ha estat escollit com a estudi de cas per aquesta tesi, oferint una perspectiva valuosa i reveladora per diversos motius. Tot i que els estudis recents sobre gènere i turisme han començat a posar el focus en dones de països en desenvolupament (Efthymiadou & Farmaki, 2023), el camp de recerca sobre dones en la gestió turística ha estat predominantment centrat en contextos occidentals, i pocs estudis han abordat aquesta temàtica en regions en desenvolupament. Analitzar l’Iran no és simplement treballar amb un escenari geogràfic diferent, sinó donar veu a dones que viuen en una societat restrictiva. L’apartheid de gènere a l’Iran (classificat 143 de 146 al Global Gender Gap Report 2024) i les protestes massives del 2022 són proves contundents d’un cas únic de discriminació i desigualtat sistèmica de gènere a escala mundial. Aquests factors fan de l’Iran un escenari especialment rellevant i necessari per a una exploració profunda del lideratge femení en el sector turístic.
Per articular la naturalesa complexa i estratificada del canvi revelada en l’estudi, la tesi proposa un marc transformacional basat en models multidimensionals ja existents. Aquest marc traça l’evolució de trets interns i valors de les dones iranianes en la gestió turística, demostrant com els rols de lideratge en aquest sector poden anar molt més enllà de l’empoderament econòmic i influir profundament en la seva manera de ser i de veure el gènere. Els canvis en l’autopercepció d’aquestes dones, especialment en moments de tensió social a l’Iran, evidencien que, fins i tot dins d’una societat patriarcal restrictiva, les dones continuen evolucionant.
