Changes in polyphenols and expression levels of related genes in 'Duke' blueberries stored under high CO2 levels

    No full text
Blueberries are highly perishable fruits, and consequently, storage under high CO2 and low O2 levels is recommended to preserve the highly appreciated polyphenols. However, high CO2 levels might be detrimental for certain cultivars. The aim of this study was to investigate the impact of storage conditions on various quality parameters, including polyphenol composition in ‘Duke’ berries. Results show that storage under 18 kPa CO2, coupled with 3 kPa O2, resulted in accelerated softening of berries, which was accompanied by lower levels compared to other conditions of hexosides and arabinosides of malvidin, petunidin, cyanidine, and delphinidin. However, this storage condition had no negative impact on chlorogenic acid levels. Expression data of key polyphenol-biosynthesis genes showed higher expression levels of all investigated genes at harvest time compared to all storage conditions. Of particular importance is the expression level of chalcone synthase (VcCHS), which is severely affected by storage at 18 kPa CO2

A highly recurrent RPS27 5’UTR mutation in melanoma

    No full text
The incidence of melanoma continues to rise globally and is increasing at a rate greater than any other cancer. To systematically search for new genes involved in melanomagenesis, we collated exome sequencing data from independent melanoma cohort datasets, including those in the public domain. We identified recurrent mutations that may drive melanoma growth, survival or metastasis, and which may hold promise for the design of novel therapies to treat melanoma. These included a frequent recurrent (i.e. hotspot) mutation in the 5’ untranslated region of RPS27 in ~10% of samples. We show that the mutation expands the 5’TOP element, a motif known to regulate the expression of most of the ribosomal protein family, to which RPS27 belongs, and thus might sensitize the mutated transcript to growthmediated regulation. This finding highlights not only the important role of non-protein coding genetic aberrations in cancer development but also their potential as novel therapeutic target

Design and implementation of an ontology for the computable representation of clinical prediction rules

    No full text
A lack of acceptance has hindered the widespread adoption and implementation of clinical prediction rules (CPRs). The use of clinical decision support systems (CDSSs) has been advocated as one way of facilitating a broader dissemination and validation of CPRs. This requires computable models of clinical evidence based on open standards rather than closed proprietary content. Methods: The on-going TRANSFoRm project has developed ontological models of CPRs suitable for providing CPR based decision support. Results: This paper describes the design and implementation of a generic ontology model for the representation of computable CPRs. The conceptual validity and implementation of the ontology is discussed using an illustrative example of a CPR in the form of the Alvarado Score for acute appendicitis. Conclusions: We demonstrate how the model is used to query the structure of this particular rule, providing a generic computable representation suitable for the representation of CPRs in genera

An article in : Zeitschrift des Deutschen Palastina-Vereins, 132, 2016, 2, pp. 146-150The first season (August 7th – September 7th 2014) of a joint project of archaeological surveys and excavations at Hirbet Qila was undertaken by Birzeit University, the Palestinian Department of Antiquities and Cultural Heritage, and Montpellier University 1 . In search for archaeological remains and in order to carry out a site-assessment study for the destruction at the site , two areas were chosen for excavations. Area A is located on the western part of the top of the hill. Three squares (5 * 5m each) were opened in Sector 14 in Area A. Area B was set on the southeastern slope of the hill and contained 4 squares (5 * 5m each). After excavating in Square 1 in Area A, the data gained from that square indicated that the area served for food production or was part of an industrial area. A grain silo, part of a wine press and grinding mortars have been uncovered here in addition to twenty loom weights found scattered in the fill. In Area B a section of 20m of the city wall as well as a rectangular tower (5 4m) were unearthed. The preliminary investigation of the material from the survey and the excavations at the site date the material to the Middle Bronze Age, Late Bronze Age, Iron Age II as well as to the Classic and Islamic periods

    No full text
