    Methods for identification of the opportunistic gut mycobiome from colorectal adenocarcinoma biopsy tissues

    Colorectal cancer poses a significant threat to global health, necessitating the development of effective early detection techniques. However, the potential of the fungal microbiome as a putative biomarker for the detection of colorectal adenocarcinoma has not been extensively explored. We analyzed the viability of implementing the fungal mycobiome for this purpose. Biopsies were collected from cancer and polyp patients. The total genomic DNA was extracted from the biopsy samples by utilizing a comprehensive kit to ensure optimal microbial DNA recovery. To characterize the composition and diversity of the fungal mycobiome, high-throughput amplicon sequencing targeting the internal transcribed spacer 1 (ITS1) region was proposed. A comparative analysis revealed discrete fungal profiles among the diseased groups. Here, we also proposed pipelines based on a predictive model using statistical and machine learning algorithms to accurately differentiate colorectal adenocarcinoma and polyp patients from normal individuals. These findings suggest the utility of gut mycobiome as biomarkers for the detection of colorectal adenocarcinoma. Expanding our understanding of the role of the gut mycobiome in disease detection creates novel opportunities for early intervention and personalized therapeutic strategies for colorectal cancer. •Detailed method to identify the gut mycobiome in colorectal cancer patients using ITS-specific amplicon sequencing. •Application of machine learning algorithms to the identification of potential mycobiome biomarkers for non-invasive colorectal cancer screening. •Contribution to the advancement of innovative colorectal cancer diagnostic methods and targeted therapies by applying gut mycobiome knowledge

    Sensitization to oil palm pollen associates with risks and severity of allergic diseases

    Background: Elaeis guineensis (Ela g, oil palm) pollen is one of the most predominant species of inhalant allergens in the tropical Southeast Asia region; however, its association with the manifestation of allergic diseases remains largely unexplored. This study aimed to determine the sensitization pattern of oil palm pollen and associate this with the risk and severity of allergic diseases. Methods: Participants were recruited as a part of the Singapore and Malaysia cross-sectional genetic and epidemiological study (SMCSGES). Two independent cohorts were recruited: n = 564 serum samples were collected and serological assessment was performed against a panel of 16 crude inhalant allergens including house dust mite, pet, insect, pollen, and fungal allergens; n = 13 652 Singapore/Malaysia Chinese young adults were recruited and skin prick test was used to assess oil palm sensitization, which was tested for its association with the risk and severity of asthma, allergic rhinitis (AR), and atopic dermatitis (AD). Results: The sensitization rate of oil palm pollen is 9.6% in the n = 564 Singapore/Malaysia cohort. In the n = 13 652 Singapore/Malaysia Chinese cohort, oil palm sensitization significantly associates with increased risks of asthma (p = 1.34x10-4), AR (p = 2.91x10-13), and AD (p = 6.95x10-7). Asthmatic patients with oil palm sensitization have increased risks of wheezing (p = 0.00995), nocturnal cough (p = 0.0122), and exacerbations (p = 0.00139) in the past 12 months. AR patients with oil palm sensitization also have an increased risk of developing moderate-to-severe symptoms (p = 0.00113). Conclusions: We have identified significant associations of oil palm sensitization with increased risks, exacerbations, and the severity of symptoms of allergic diseases in the tropical Southeast Asian region (Singapore/Malaysia)

    Kenaf-based activated carbon: A sustainable solution for high-performance aqueous symmetric supercapacitors

    This study presents an innovative method for synthesizing activated carbon with an exceptionally high surface area (3359 m2 g-1) using kenaf fiber-based biochar through chemical activation. The achieved specific surface area surpasses activated carbon derived from other reported fiber-based precursors. The resulting activated carbon was investigated as electrodes for supercapacitors, revealing a remarkable maximum capacitance of 312 F g-1 at a current density of 0.5 A g-1. An aqueous symmetric supercapacitor employing these high-surface-area electrodes exhibited an outstanding energy density of 18.9 Wh kg-1 at a power density of 250 W kg-1. Notably, the supercapacitor retained exceptional capacitance, maintaining 93% of its initial capacitance even after 5000 charge-discharge cycles

    Edible beetles (Coleoptera) as human food – a comprehensive review

    The consumption of edible Coleoptera, or beetles, is examined in this review as a viable remedy for the rising world food demand brought on by the population’s predicted expansion to over 9 billion by 2050. The review illustrates the nutritional profile of beetles, highlighting their high protein content, good fats, and oils, while being low in saturated fats and high in omega-3. Beetles have a rich history of consumption, with over 2 billion people, particularly in regions like Africa, Asia, and the Americas, incorporating them into their diets. They contribute significantly to human nutrition while also playing essential ecological roles, including soil fertilization and pollination. Beetles represent a promising solution to combat climate change, as traditional livestock production is a major greenhouse gas emitter, and beetle farming boasts lower emissions, reduced resource requirements, shorter life cycles, and superior feed conversion rates. Notwithstanding their advantages in terms of nutrition and the environment, there are obstacles including customer adoption, safety worries, and legal limitations. The review also covers how beetles are processed into different food items, such as liquids, pastes, and powders, and how these products are used in the feed, food, and nutraceutical industries. In general, edible beetles present a viable substitute food source with noteworthy nutritional and ecological benefits; yet, additional investigation and endeavors are required to surmount obstacles to their extensive integration

    Enhanced energy recovery of non-hazardous organic wastes via moderate pyrolysis with natural calcium- and potassium-based additives

    The current research focuses on the efficiency of energy recovery from non-hazardous organic wastes. It entails the generation of low-chloride refuse-derived fuel (RDF) from municipal solid waste (MSW) through moderate pyrolysis of combustible components. The moderate pyrolysis settings and composition ratio of biogenic/non-biogenic content from MSW combination (food waste, plastic waste, and paper waste) had a significant impact on the thermochemical characteristics and fuel behavior of RDF. Furthermore, pyrolysis with optimal MSW composition was studied to reduce toxic elements (chlorides, sulphides, and nitrides) and enhance the energy value in RDF by utilizing potassium-rich (waste orange peels (OP); banana peels (BP); and maize cob (MC)) and calcium-rich species (waste animal bone meal (ABM); egg shells (ES); and mussel shells (MS)) as natural additives. The results showed that increasing the pyrolysis temperature maximized the carbon concentration with reduced oxygen moieties (low O/C ratio), indicating a high energy value as the oxygenates were eliminated during moderate pyrolysis. The RDF treated at 400 °C (SF4-400) with MSW composition of 55% non-biogenic carbon and 45% biogenic carbon resulted in HHV of 29.22 MJ kg−1, the lowest ash concentration (0.96%), and a densified microstructural surface. The addition of natural maize cob additive into MSW (SF4-400-MC) yielded the maximum HHV (33.07 MJ kg−1) with considerable reductions in chlorides (71.43%), sulphides (87.50%), and insignificant nitrides content, all of which meet ASTM RDF quality criteria Grade I

    The aim of this study is to evaluate the current water quality status of one of the urban rivers in Malaysia, called Sungai Air Hitam. The river's water supply is not only unsuitable for the inhabitants but also hazardous to the aquatic species that depend on it. In order to simulate the water quality formulation of the river, the Model for Urban Stormwater Improvement Conceptualization (MUSIC) was used. The effects of various best management practices (BMPs) components have been examined to improve the river's water quality. This study also investigated different scenarios of the expected future changes in the land cover and the quality of the river. As the proportion of impervious surfaces increases, the urban hydrology cycle can be significantly altered, resulting in an increase in volumes and peak flows, and a decrease in storage, infiltration, and interception. The MUSIC results have shown significant reductions in biochemical oxygen demand (BOD), total suspended solids (TSS), total phosphorus (TP), and total nitrogen (TN) after introducing BMPs. It was also noticed that the prediction of pollutants falls within the acceptable range set by the Urban Stormwater Management Manual for Malaysia (MSMA) 2nd edition. For the land cover, it was found that the total reduction of BOD, TSS, TP, and TN for existing land use is 92.5 %, 94.5 %, 90.7 % and 91.9 %. Meanwhile, the total reduction in future land use is 81.6 % for BOD, 86.2 % for TSS, 80.9 % for TP and 80.8 % for TN. From the simulation results, it was observed that the application of BMPs has successfully reduced the observed mean BOD concentration from 92.38 mg/L (Class V) to 6.93 mg/L (Class IV) of the national water quality standards, NWQS, water quality index. As a result, the water quality index of the overall catchment has improved from Class IV to Class III (WQ1, WQ3, and WQ4) and from Class V to IV (WQ2) with the application of the BMPs. This assessment aims to raise awareness within the Sungai Air Hitam community regarding the importance of preserving river cleanliness and understanding the long-term environmental impact of water quality. These findings underscore the importance of an integrated system in managing urban water systems, which can offer valuable insight to the decision-makers

Plastics are utilized in the packaging of food items, medications, detergents, and cosmetics. The overconsumption of plastics presents a significant peril to both the ecosystem and human existence on Earth. The accumulation of plastics on land and in the sea has sparked interest in finding ways to breakdown these polymers. It is necessary to employ suitable biodegradable techniques to decrease the accumulation of plastics in the environment. To address the environmental issues related to plastics, it is crucial to have a comprehensive understanding of the interaction between microorganisms and polymers. A wide range of creatures, particularly microbes, have developed techniques to survive and break down plastics. This review specifically examines the categorization of plastics based on their thermal and biodegradable properties, as well as the many types of degradation and biodegradation. It also discusses the various types of degradable plastics, the characterization of biodegradation, and the factors that influence the process of biodegradation. The plastic breakdown and bioremediation capabilities of these microbes make them ideal for green chemistry applications aimed at removing hazardous polymers from the ecosystem

    Preclinical Development of a Novel Epitope-based DNA Vaccine Candidate against SARS-CoV-2 and Evaluation of Immunogenicity in BALB/c Mice

    The protective efficacies of current licensed vaccines against COVID-19 have significantly reduced as a result of SARS-CoV-2 variants of concern (VOCs) which carried multiple mutations in the Spike (S) protein. Considering that these vaccines were developed based on the S protein of the original SARS-CoV-2 Wuhan strain, we designed a recombinant plasmid DNA vaccine based on highly conserved and immunogenic B and T cell epitopes against SARS-CoV-2 Wuhan strain and the Omicron VOC. Literature mining and bioinformatics were used to identify 6 immunogenic peptides from conserved regions of the SARS-CoV-2 S and membrane (M) proteins. Nucleotide sequences encoding these peptides representing highly conserved B and T cell epitopes were cloned into a pVAX1 vector to form the pVAX1/S2-6EHGFP recombinant DNA plasmid vaccine. The DNA vaccine was intranasally or intramuscularly administered to BALB/c mice and evaluations of humoral and cellular immune responses were performed. The intramuscular administration of pVAX1/S2-6EHGFP was associated with a significantly higher percentage of CD8+ T cells expressing IFN-γ when compared with the empty vector and PBS controls. Intramuscular or intranasal administrations of pVAX1/S2-6EHGFP resulted in robust IgG antibody responses. Sera from mice intramuscularly immunized with pVAX1/S2-6EHGFP were found to elicit neutralizing antibodies capable of SARS-CoV-2 Omicron variant with the ACE2 cell surface receptor. This study demonstrated that the DNA vaccine construct encoding highly conserved immunogenic B and T cell epitopes was capable of eliciting potent humoral and cellular immune responses in mice
