    Olfactory Processing and Learning in Drosophila: Models Across Biological Scales and Computational Abstractions

    Olfaction is a fundamental and universal sensory modality across the animal kingdom. It enables animals to locate food sources, avoid danger, and identify mating partners. Moreover, odors can acquire predictive value through experience, allowing animals to infer and anticipate relevant aspects of their environment. Thus, olfactory perception and learning are critical for both survival and reproduction. Understanding how the brain processes and learns olfactory information provides a powerful window into the core principles of neural function. In this thesis, we investigate mechanisms of olfactory processing and learning in the compact yet sophisticated brain of the fruit fly, Drosophila melanogaster. Using computational modeling across multiple levels of abstraction, we aim to formalize and explore the biological principles that shape sensory representation, learning, and memory. At the circuit level, we examine how specific network motifs support memory formation, updating, and extension, including mechanisms underlying second-order conditioning and extinction learning. At the cellular level, we investigate the role of inhibitory interactions in binary odor mixture processing in early olfactory circuits. Based on ultrastructural connectome data, we analyze how sensory information is integrated and develop computational models to explore how these circuits shape odor representations. At the molecular level, we develop a phenomenological model of synaptic transmission dynamics shaped by the differential expression of synaptic protein isoforms. This allows us to investigate how molecular composition tunes short-term plasticity and modulates information flow at synaptic resolution. Through these interdisciplinary approaches, we aim to advance the understanding of olfactory computation and memory by bridging molecular mechanisms, circuit dynamics, and behavioral outcomes. Ultimately, this work seeks to uncover general principles of brain function that extend beyond the olfactory system and offer transferable insights into neural computation

    Successful Natalizumab Treatment of Two Female Individuals With Susac Syndrome

    Background: Susac syndrome is a rare autoimmune endotheliopathy that affects the central nervous system, retina, and inner ear, characterized by encephalopathy, branch retinal artery occlusions, and sensorineural hearing loss. Due to the heterogeneity of its presentation, early diagnosis, and treatment remain challenging. Objective/Methods: To evaluate the clinical outcomes and radiological responses in two patients with Susac syndrome treated with natalizumab in an off‐label therapeutic approach, clinical assessments and serial magnetic resonance imaging (MRI) were performed over a follow‐up period of up to 22 months to monitor disease progression and treatment response. Results: Both patients demonstrated clinical stabilization with reduced MRI and retinal angiography disease activity. Treatment was well tolerated, and no significant adverse events were reported during observation. Discussion: Natalizumab may constitute a potential off‐label therapeutic for Susac syndrome. Further studies are warranted to assess its efficacy and safety in this rare condition

    Doing Memory. Digitale Erinnerungskulturen an nationalsozialistische Verbrechen auf TikTok

    In den letzten Jahren hat sich TikTok zu einer interessenbasierten Plattform entwickelt, auf der Informationen zu einer Vielzahl von Themen von Politik über Sexualität bis zum Holocaust kommuniziert und verhandelt werden. Allein im deutschsprachigen Raum gibt es mittlerweile rund 26 Profile, die sich mit Inhalten über die nationalsozialistische Verfolgung, ihre Opfer und deren Erinnerung befassen, darunter Gedenkstätten, Stiftungen und einzelne Aktivist*innen. Content-Creator*innen wie Susanne Siegert (@keine.erinnerungskultur), die Medienkampagne @zumfeindgemacht und die Gedenkstätte KZ Neuengamme (@neuengamme.memorial) erzielen dabei regelmäßig mehrere tausend Aufrufe für ihre Inhalte, indem sie die Mediensprache von TikTok nutzen, um das Bewusstsein um NS-Geschichte, Opfergruppen sowie Kontinuitäten zu diversifizieren, zu schärfen und neue Zielgruppen zu erreichen. Die Forschungsarbeit konzentriert sich darauf, wie diese Creator*innen TikTok als Medium für die Erinnerung an NS-Verbrechen reichweitenstark nutzen und wie ihr Ansatz die Dynamiken von Erinnerungskulturen als öffentlicher Geschichte in den sozialen Medien beeinflusst. Auf Basis eines creator*innenzentrierten Ansatzes wurde dafür die Webformat-Analyse von Martina Schuegraf und Anna Janssen als methodischer Rahmen angepasst, um die spezifische Nutzung von Profilfunktionen und Videostilen in Bezug auf diskursive Erinnerungspraktiken und Erinnerungskulturen im digitalen Umfeld zu bestimmen. Die Methode wurde darüber hinaus um Aspekte der historischen Quellenkritik erweitert, um die Hintergründe und Kontexte zu erarbeiten, in denen die Profile erstellt wurden. Aufgrund der digitalen Quellenbasis wurden letztlich auch Impulse digitaler Hermeneutik an jeder Arbeitsebene zur Reflexion des Forschungsprozesses aufgenommen. Auf der Grundlage der Forschungen zu digitalen Kulturdynamiken von Hoskins und Poell/van Dijck behaupt die Arbeit, dass Erinnerung bei der Nutzung von TikTok als soziokultureller digitaler Raum in flachen Konfigurationen zwischen Nutzern und Empfehlungssystemen der vermittelnden Plattform selbst hyperkonnektiv verhandelt wird. Durch ihre Algorithmen wird die Plattform zu einem kommerziell orientierten Diskursrahmen und Vermittler in der performativen Konstruktion von Erinnerung. In Anlehnung an Erll und Hoskins schränken diese das Gedächtnis ein, bieten aber gleichzeitig multidirektionales und interkulturelles Potenzial und schaffen neue thematische Räume, in denen Nutzer populäre Mediensprachen aneignen müssen, um Einfluss auszuüben und innerhalb von Gedächtnisdiskursen Reichweite zu erzielen. Dies führt zu verständlichen Mikro-Lerninhalten, die sich durch eine multimodale, aufmerksamkeitsstarke Ästhetik auszeichnen und sich auf opferzentrierte, bottom-up-Perspektiven konzentrieren, die oft von jungen Content-Erstellern selbst als Gleichaltrige des Publikums präsentiert werden. Die Profile laden die Nutzer zur Teilnahme und Reflexion ein, während sie bewusst versuchen, den Diskurs zu moderieren und negativer Multidirektionalität entgegenzuwirken, beispielsweise in Form von Whataboutism und Holocaustleugnung

    Immune Phenotypes in Patients With Invasive Mould Infection Support the Use of PD ‐1 Inhibition as Potential Treatment Option

    Background: Invasive mould infections (IMI) cause substantial morbidity and mortality in populations at risk. Novel treatment approaches are urgently needed. Targeting immune checkpoints may reverse hyporesponsiveness of the innate and adaptive immune systems. Methods: In this prospective, observational study, we investigated immune checkpoint expression levels on immune cells in patients with invasive aspergillosis (IA; n = 25) and mucormycosis (MU; n = 7). Healthy controls (HC; n = 5) and patients with matched haematological diseases but without IMI served as control populations (CP; n = 10). Multicolour flow cytometry analysis was used to compare immune cell subsets and the expression of immune‐regulatory molecules in peripheral blood mononuclear cells (PBMCs). Results: Lymphocyte subsets and immune phenotypes in PBMCs were similar between patients with IMI and haematological CP, except for regulatory T cells, which were increased in PBMCs of patients with IA and MU compared to HCs. In IA and MU, PBMCs showed increased expression of immune checkpoint molecules compared to healthy controls and matched haematological CP, with this effect being more pronounced in IA than in MU. We found heterogeneous, disease‐, molecule‐, and patient‐specific expression patterns of immune checkpoint molecules. For example, PD‐1 expression was highest in MU PBMCs, followed by IA PBMCs, while HC PBMCs showed lower expression levels. Overall mortality in our patient population was 44.0% (IPA) and 80.0% (MU). Conclusions: We report an immune phenotype consistent with T‐cell exhaustion in IMI, indicating potential contributions from haematological treatment, underlying disease, and infection. However, the primary underlying cause remains unclear and requires further investigation. A marker that was notably higher in IMI patients was PD‐1, and treatment approaches specifically targeting this molecule may be promising

    Adaptive learning in bionics: transforming science education

    Introduction: Adaptive learning platforms offer innovative teaching approaches by tailoring educational content to individual learner’s needs, abilities, and paces. Methods: This study investigates the effects of an adaptive digital learning platform on user experience, motivation, and learning outcomes among 56 sixth-grade students from two German grammar schools. Students completed three bionics-focused modules— “polar bear”, “heat transfer”, and “temperature and heat”—integrated into science lessons. Data from questionnaires and performance tests assessed prior knowledge, learning success, cognitive activation, and situational interest. Results: The findings indicate that 98% of students found digital media motivating, with 81% favoring a hybrid mix of traditional and digital teaching methods. Positive emotional responses were reported by 62% of participants, though 38% experienced uncertainty. The “polar bear” module achieved the highest learning gain (+41%), followed by “heat transfer” (+23%) and “temperature and heat” (+13%) module. Discussion: These results suggest that adaptive digital learning platforms can enhance learning outcomes and cognitive engagement, particularly when the content aligns with student interests and needs

    An organelle pathogen sensor reveals factors required for mediating trans-kingdom membrane contact sites

    Toxoplasma gondii (Toxoplasma) is a human parasite that establishes a life-long infection in one-third of the world’s population although prevalence can vary depending on the country. In Germany for example more than 50% of the population over age 50 are estimated to be seropositive for Toxoplasma. Thus, defining the mechanisms by which Toxoplasma engages with the host cells can lead to the development of better therapeutics. A common consequence of infection with Toxoplasma is the formation of trans-kingdom membrane contact sites (MCS) between the vacuole of Toxoplasma and host mitochondria and host endoplasmic reticulum (ER). Although the association of host ER with the vacuole was first described in the early 1970s neither of the molecular components that mediate this interaction have been identified thus far. Here, we investigated the molecular machinery that mediates the MCS between Toxoplasma and ER. To this end, we developed a split-GFP based sensor where GFP reconstitution indicates successful formation of MCS between the pathogen and host organelles. To validate our sensor for FACS based CRISPR-Cas9 screening, I first applied it to monitor the known mitochondria- Toxoplasma MCS. As expected, GFP reconstitution occurred at these contact sites but failed to do so in the absence of the Toxoplasma tether TgMAF1 or the host counterpart TOM70, confirming the sensors specificity. I then adapted our sensor to study host ER-Toxoplasma MCS and performed a Toxoplasma effector protein targeted loss-of-function CRISPR screen. I found that Toxoplasma rhoptry protein 1 (TgROP1) is required for mediating Toxoplasma-ER MCS. Interestingly, TgROP1 contains putative FFAT [(two phenylalanines (FF) in an acidic tract (AT)] motifs, that are regions known to interact with ER membrane proteins VAPA/VAPB (VAPs). Subsequent work identified VAPs as required to mediate MCS with the Toxoplasma vacuole and mutating the FFAT-binding domain of VAPs reduced this interaction. Our findings reveal that Toxoplasma exploits a mechanism like host proteins to establish MCS with host organelles. This work advances our current understanding of host pathogen MCS and lays the foundation for future studies investigating their functional consequences on infection outcomes

    Comparison of airway pressure release ventilation (APRV) versus biphasic positive airway pressure (BIPAP) ventilation in COVID-19 associated ARDS using transpulmonary pressure monitoring

    Background: APRV has been used for ARDS in the past. Little is known about the risk of ventilator- induced lung- injury (VILI) in APRV vs. BIPAP in the management of in COVID19-associated ARDS (CARDS). This study aimed to compare transpulmonary pressures (TPP) in APRV vs. BIPAP in CARDS in regard to lung protective ventilator settings. Methods: This retrospective, monocentric cohort study (ethical approval: 21-1553) assessed all adult ICU- patients with CARDS who were ventilated with BIPAP vs. APRV and monitored with TPP from 03/2020 to 10/2021. Ventilator-settings / -pressures, TPP, hemodynamic and arterial blood gas parameters were compared in both modes. Results: 20 non- spontaneously breathing patients could be included in the study: Median TPPendexpiratory was lower / negative in APRV (-1.20mbar; IQR − 4.88 / +4.53) vs. positive in BIPAP (+ 3.4mbar; IQR + 1.95 / +8.57; p < .01). Median TPPendinspiratory did not differ. In APRV, mean tidal- volume per body- weight (7.05 ± 1.28 vs. 5.03 ± 0.77 ml; p < .01) and mean airway- pressure (27.08 ± 1.67 vs. 22.68 ± 2.62mbar; p < .01) were higher. There was no difference in PEEP, peak-, plateau- or driving- pressure, compliance, oxygenation and CO 2 - removal between both modes. Conclusion: Despite higher tidal- volumes / airway-pressures in APRV vs. BIPAP, TPPendinspirator y was not increased. However, in APRV median TPPendexpiratory was negative indicating an elevated risk of occult atelectasis in APRV- mode in CARDS. Therefore, TPP- monitoring could be a useful tool for monitoring a safe application of APRV- mode in CARDS

    Diese Studie untersucht, ob routinemäßig erhobene Blutparameter bei Patienten mit Malignem Melanom im Stadium III mit mikroskopischen Sentinel-Lymphknotenmetastasen etwas über das rezidivfreie Überleben und Gesamtüberleben aussagen können. Ein hoher CRP-Wert plus eine niedrige LMR zeigten das größte Potential als prognostischer Marker für das rezidivfreie Überleben. Dies sollte in größeren prospektiven Studien weiter überprüft werden

    Die Studie „The Incidence and Economic Burden of CDI in Inpatient Settings of the German Health Care System” wurde durchgeführt, um die ökonomischen Auswirkungen der Clostridioides-difficile-Infektion (CDI) im stationären Bereich des deutschen Gesundheitssystems zu untersuchen. CDI stellt eine erhebliche finanzielle Belastung für das Gesundheitswesen dar, insbesondere durch verlängerte Krankenhausaufenthalte, intensive Therapiemaßnahmen und die hohen Kosten für rezidivierende und refraktäre Fälle. Ziel der Studie war es, direkte und indirekte Kosten von CDI zu analysieren, um ein besseres Verständnis für die wirtschaftliche Belastung und die wichtigsten Kostentreiber zu gewinnen. Die Studie wurde retrospektiv und prospektiv in 10 deutschen Krankenhäusern zwischen August 2017 und September 2020 durchgeführt. Die Beobachtung umfasste 535 Patienten, die in drei Gruppen unterteilt wurden: Indexgruppe (einmalige CDI-Episode), Refraktärgruppe (therapieresistente Fälle) und Rezidivgruppe (wiederkehrende Infektionen). Die Kostenevaluation erfolgte gemäß den CHEERS-Standards (2022) und umfasste die Analyse von Behandlungskosten in verschiedenen Krankenhausstationen (z. B. allgemeine Station, Intermediate-Care, Intensivstation), Kosten für gezielte Antibiotikatherapien (Fidaxomicin, Metronidazol, Vancomycin) sowie Produktivitätsverluste durch krankheitsbedingte Arbeitsausfälle. Die Ergebnisse zeigten, dass die Gesamtkosten mit der Schwere und dem Verlauf der Infektion signifikant anstiegen. Während Patienten in der Indexgruppe durchschnittliche Gesamtkosten von 13.607 € verursachten, lagen diese in der Refraktärgruppe bei 19.953 € und in der Rezidivgruppe bei 22.671 €. Die Hauptkostentreiber waren längere Krankenhausaufenthalte und intensive Maßnahmen wie Isolation und gezielte Antibiotikatherapie. Refraktäre und rezidivierende Fälle erforderten signifikant mehr Ressourcen, darunter durchschnittlich 24 Tage Antibiotikatherapie in der Rezidivgruppe im Vergleich zu 11 Tagen in der Indexgruppe. Indirekte Kosten, die durch Produktivitätsverluste entstanden, waren ebenfalls bei schwereren Fällen erhöht, jedoch nicht statistisch signifikant. Limitationen der Studie umfassen den Fokus auf den stationären Bereich, ohne ambulante Kosten oder langfristige Folgen wie chronische Gesundheitsprobleme und wiederholte Krankenhausaufenthalte zu berücksichtigen. Auch die geringe Anzahl von Patienten, die mit Fidaxomicin behandelt wurden (n=18), schränkt die Aussagekraft der Ergebnisse zu den Kosten und der Wirksamkeit dieses Medikaments ein. Zudem wurde keine Differenzierung zwischen verschiedenen C.-difficile-Stämmen vorgenommen, wie dem virulenten Ribotyp 027, der bekanntermaßen mit schwereren Verläufen und höheren Kosten verbunden ist. Die Studie kommt zu dem Schluss, dass refraktäre und rezidivierende CDI-Fälle die Haupttreiber für die hohen Kosten im Zusammenhang mit CDI sind. Sie unterstreicht die Notwendigkeit neuer präventiver und therapeutischer Ansätze, um Rezidive zu verhindern und die wirtschaftliche Belastung durch CDI zu reduzieren. Langfristige Analysen, die die gesamte Versorgungskette sowie die langfristigen klinischen und wirtschaftlichen Auswirkungen berücksichtigen, sind für zukünftige Forschungen entscheidend, um fundierte gesundheitspolitische Maßnahmen zu entwickeln. Die Ergebnisse liefern eine solide Grundlage für die Verbesserung der Behandlung und Prävention von CDI im deutschen Gesundheitssystem
