Einseitige Stimmlippenparese: Charakterisierung des Patientenkollektivs der HNO Uniklinik Köln: Epidemiologie und Verlauf

    Get PDF

Learning the Topology of Latent Signaling Networks from High Dimensional Transcriptional Intervention Effects

Biological systems rely on complex networks of interacting molecules, genes, proteins, and other regulators to govern cellular processes. However, inferring the causal structure of these networks is challenging. Particularly when important regulatory factors are only indirectly measured or entirely hidden. This difficulty is combined with the need to distinguish direct from indirect interactions and to capture dynamic responses under different perturbations such as gene knockouts or drug treatments. Consequently, developing a method that can incorporate both time resolved data and partial observability is critical for inference of the intricate architectures underlying signal transduction and gene regulation. To address these challenges, we propose an Ordinary Differential Equation based Nested Effects Model (odeNEM). Our approach combines the established Nested Effects Model framework, which maps hidden regulatory nodes to observable downstream effects, with a continuous time ODE formulation. This combination can capture saturable, non-linear kinetics. By explicitly modeling the propagation of perturbation signals over time, odeNEM infers which hidden regulators are causally upstream of others. Additionally, we developed mixture models for the downstream expression data which is considering the noisy high throughput nature of biological experiments. We validate our method on two key applications. First, we reconstruct signaling pathways in breast cancer cells using phosphoprotein time course data from the HPN-DREAM challenge. This shows how odeNEM captures context specific interactions among kinases such as AKT, mTOR, and MEK. Second, we infer a gene regulatory network in pluripotent stem cells from CRISPR single cell transcriptomics (RENGE), highlighting both canonical regulators like POU5F1 and SOX2 and novel interactions that warrant further biological investigation. Across both use cases, our results align with known biology and external data sources (e.g., ChIP-seq). This also confirms that a continuous time hidden node model can robustly uncover causal relationships. Overall, odeNEM expands the applicability of NEM approache by explicitly modeling. The model’s synergy with perturbation data, prior knowledge, and advanced inference strategies (e.g., MCMC) enables a more robust and biologically realistic reconstruction of latent signaling networks. This contributes a promising picture for unraveling the complex interplay of molecular interactions in systems biology, with broad implications for identifying therapeutic targets and understanding disease mechanisms

Subthreshold Depression und Promotionsabbrüche: Eine Studie zur psychischen Gesundheit von Kölner Doktorand*innen

    Get PDF

Superconducting Proximity Effect in Quantum Anomalous Hall Insulators

When a thin film of a topological insulator (TI) is doped with magnetic impurities, it can exhibit the quantum anomalous Hall effect (QAHE). This phenomenon emerges from the breaking of time-reversal symmetry (TRS) in a system with strong spin-orbit coupling, resulting in a vanishing longitudinal resistance and a quantized Hall resistance of h/e² (where h is the Planck constant and e is the charge of an electron), even in the absence of an external magnetic field. Such a magnetic topological insulator is called a quantum anomalous Hall insulator (QAHI), where electrical current is carried by one-dimensional (1D) chiral edge states that propagate along the sample boundaries, while the two-dimensional (2D) bulk remains insulating. When superconducting pairing correlations are induced in such a material via the proximity to an s-wave superconductor, the resulting topological superconductivity is predicted to host chiral Majorana edge modes. In a Hall-bar device configuration, where a superconducting strip lies across the full width of a QAHI thin film, a quantized two-terminal conductance of 0.5(e²/h) was proposed as the smoking-gun evidence of the topological superconducting phase associated with a single chiral Majorana mode. This reduction in two-terminal conductance by a factor of two, compared to e²/h observed in a bare QAHI without the superconducting strip, has been experimentally reported. However, the origin of this 0.5(e²/h) conductance feature remains a topic of active debate, as alternative trivial mechanisms have also been proposed. This emphasizes the need for more robust experimental evidence to confirm the superconducting proximity effect in QAHIs. In this thesis, narrow superconducting electrodes of Nb on top of a QAHI thin film are investigated with widths ranging from 160 to 520 nm. By measuring the nonlocal `downstream' resistance with respect to the grounded superconducting electrode, a negative resistance contribution of -400Ω is observed for the narrowest superconducting electrode. This contribution decreases exponentially as the width of the SC increases. This negative nonlocal resistance is attributed to crossed Andreev reflection (CAR) taking place across the superconducting electrode. In the CAR process, an electron in the chiral edge state, arriving at the superconducting electrode with an energy eV smaller than the SC gap Δ, is converted into a hole in the chiral edge state leaving from the SC, carrying a potential of -V. Simultaneously, a Cooper pair is formed in the superconductor. The observation of a negative edge potential, measurable as a negative `downstream' resistance with respect to the grounded SC in our experiment, is a compelling signature of induced superconducting pair correlation in the chiral edge state of the QAHI. Moreover, the characteristic length over which the CAR process is suppressed with increasing the width of the SC is found to be significantly longer than the superconducting coherence length of Nb. This implies that the CAR process is mediated by the superconductivity induced in the QAHI film underneath the Nb electrode, rather than by the Nb superconductor itself. These findings are supported by a detailed Landauer-Büttiker analysis of the experimental set-up, accounting for all possible processes at the SC-QAHI interface, and by KWANT simulations that incorporate charge disorder in the QAHI film, as well as the metallization effects by the superconducting electrode. Having established a reliable method to proximitize a QAHI thin film using a superconducting Nb, the second part of this thesis re-evaluates the 0.5(e²/h) feature in two-terminal conductance measurements. Rather than only characterizing the device's two-terminal conductance, the potentials of all chiral edge states arriving at and leaving from the superconducting Nb electrode are determined in this thesis, providing a comprehensive understanding of the transport through the proximitized QAHI thin film. Two Hall-bar devices were fabricated for this purpose. In the first device, a Nb superconducting electrode spans the full width of the Hall-bar, forming a proximitized QAHI region beneath the SC. The second device serves as a control experiment, where the QAHI film is interrupted underneath the superconducting electrode, creating two separate QAHI Hall-bars connected in series through the Nb electrode. For both devices, a quantized resistance of h/e² is measured across the Nb electrode in a four-terminal configuration when the current flows through the QAHI film. This quantized resistance of h/e² in the four-terminal set-up employed in this thesis corresponds to a two-terminal conductance of 0.5(e²/h) for the device. No difference is observed between the devices with and without a continuous QAHI under the superconducting electrode. This indicates that the 0.5(e²/h) conductance feature is unrelated to chiral Majorana edge mode transport, as the interrupted QAHI in the control device prevents Majorana transmission underneath the superconducting electrode. In addition, the potentials of all chiral edge states arriving at and leaving from the superconducting electrode remain unchanged when the external magnetic field exceeds the upper critical field of Nb, which points to a trivial effect unrelated to (induced) superconductivity. By using the Landauer-Büttiker formalism, the experimental data are shown to be consistent with a model in which the superconductor equilibrates all the chiral edge states arriving at and leaving from the superconducting electrode. Lastly, no negative nonlocal edge potentials are observed in Hall-bar devices with a Nb strip, in contrast to the narrow Nb electrodes studied in the first part of this thesis. This suggests that signatures of the superconducting proximity effect in QAHI thin films are only observable within a length scale comparable to the superconducting coherence length. The findings in this thesis provide critical insights into the chiral edge transport at superconducting electrodes interfaced with QAHIs and prove the existence of the SC proximity effect in QAHIs, offering a foundation for future studies of topological superconductivity, Majorana physics, and the search for non-Abelian zero modes

Postoperative Cochlear Obliteration after Retrosigmoid Approaches in Patients with Vestibular Schwannoma

    Get PDF
Vestibular schwannomas (VSs) are the most common tumors located in the cerebellopontine angle (CPA). While facial nerve function is often preserved, the cochlear nerve is frequently affected. Magnetic resonance imaging (MRI) is the gold standard for diagnosis. Treatment options include observation, microsurgical resection, fractionated radiotherapy, and radiosurgery. The main goals of surgery are to preserve facial nerve function and, ideally, cochlear nerve function. However, hearing loss is often unavoidable due to the vulnerability of the cochlear nerve. To restore hearing postoperatively, cochlear implants (CI) and auditory brainstem implants (ABI) are available. CI is the preferred option when the cochlear nerve is preserved and the cochlea remains patent. However, cochlear obliteration presents a challenge for CI implantation. It is assumed that the surgical approach (retrosigmoid vs. translabyrinthine) may influence the risk of cochlear obliteration. This monocentric, retrospective study evaluated the frequency of cochlear obliteration after retrosigmoid VS surgery using high-resolution, T2-weighted MRI scans. Cochlear obliteration was observed in 32% of cases. Factors such as age, tumor size, operation duration, and reoperation were not significantly associated with higher obliteration rates. However, obliteration occurred more frequently in female patients and was significantly associated with obliteration of the semicircular canals. The proximity of these canals to the posterior lip of the internal auditory canal may contribute to inner ear susceptibility. Future studies should explore surgical technique differences and the ideal timing for CI implantation before cochlear obliteration develops

Social Networks, Health Behaviour, and Well-Being in Middle-Aged and Older Adults

Social epidemiological research has shown that social embeddedness is crucial for physical and mental health and overall well-being, especially among older adults. This dissertation examines the links between social relationships, health behaviour and well-being in middle-aged and older adults and is divided into two pillars: The first pillar focuses on social support and health behaviour, in particular the relationship between social support and physical activity. The second pillar analyses the relationship between social networks and well-being using an empirical-qualitative network approach. For Pillar 1 (Study 1), a scoping review of 116 studies was conducted to examine the relationship between social support and physical activity in older adults. For Pillar 2 (Studies 2 and 3), a qualitative network analysis was conducted based on 28 qualitative interviews and 25 unstructured egocentric network maps. Study 2 analysed middle-aged and older adults' perceptions of well-being and the contribution of their personal networks to their well-being. Study 3 analysed the use of personal networks to maintain or improve well-being. Overall, the dissertation shows a predominantly positive relationship between social networks, health and well-being in middle-aged and older adults. Each of the three studies examined different aspects of social integration (social support, social engagement and social networks) and their relationship with health outcomes. The findings underline the importance of social integration for health and well-being and highlight the interrelatedness of different components of social integration, underlining the complexity of this area of research. While the extensive evidence from all three studies generally points to a positive relationship, each study also identified instances where social relationships can be detrimental to health and well-being. This highlights the importance of considering possible negative aspects of social relationships

Patterns of iconicity across sign languages and gestures

    Get PDF
Human communication is inherently multimodal. It expands into any available modality, for example in gesture and sign where properties of meaning are displayed through visual iconicity: resemblance-based mappings between form and referent. Iconicity relies on embodiment and has therefore been explored in concrete rather than abstract semantic domains in the past. This dissertation expands on this by exploring how the body is used for iconic depiction of both concrete and abstract concepts in the visuo-manual modality. It compares lexical signs from two unrelated sign languages, British Sign Language (BSL) and German Sign Language (DGS), with silent gestures produced by non-signing speakers of English and German. The study examines how iconic strategies are employed across modalities and languages through a multi-methods approach. Lexical signs in BSL and DGS showed highly similar distributions of iconic strategies, with the acting and entity strategies emerging as most prevalent. Lexical overlap across the two languages was not predicted by concreteness, but by shared iconic strategy. In gesture, non-signers showed a strong preference for the acting strategy and struggled more with abstract concepts. Interestingly, the same iconic strategies used in sign languages were also found in gesture, though proportions differed both compared to the sign languages and between concrete and abstract concepts. The findings suggest that the potential of iconic mappings for meaning-making in the visuo-manual modality is not limited to conventionalised signs but extends to spontaneous gesture. This iconic potential is structured and shared across user groups, and even abstract concepts can be expressed using these same iconic strategies. Signers and gesturers thus appear to draw from a shared cognitive capacity for form-meaning mapping, enabling humans to flexibly harness the expressive affordances of the body across contexts and communicative needs

Übertragbarkeit eines Künstlichen-Intelligenz-Algorithmus zwischen zwei Chirurgen bei der Descemet Membran Endothelialen Keratoplastik

    Get PDF
