Search for Lead Compounds and In Vitro Assay of Potential Inhibitors of Plasmodium falciparum Prolyl-tRNA Synthetase from Natural Compound Database
Background: Malaria continues to be a serious problem in several countries, marked by an increase in the
number of cases and a high morbidity rate. One of the commonly adopted strategies in drug discovery is by
performing compound screening using computational tools, known as virtual screening. This technique allows
one to screen multitudes of chemical compounds in silico, thus saving cost and time by reducing the amount of
tested compound in vitro. Recently, P. falciparum prolyl-tRNA synthetase (PfPRS) is one of the top priority
targets to be explored of potent inhibitors. This enzyme plays an important role in attaching L-proline into tRNA,
which then will be incorporated into protein sequence. Its inhibition would halt the protein synthesis and kill the
parasite. Methods: Hierarchical virtual screening was performed against PfPRS enzyme using 2D followed by
3D similarity method implemented in Infinisee 3.2.0 and SeeSAR 12.1.0, respectively. 1-(pyridin-4-yl)
pyrrolidin-2-one based analog, which was previously discovered as potent antimalarial agent, was used as
template to screen potential hits from Molport Database of Purchasable Natural Product Compounds.
Compounds with high similarity value were evaluated by molecular docking using SeeSAR 12.1.0 approach.
The best scoring compounds were subjected into ADMET prediction, molecular dynamics simulation, and in
vitro assay against P. falciparum. Results: Two compounds were obtained from virtual screening and molecular
docking process, with predicted IC50 value lies on micromolar and nanomolar range. These compounds also
satisfy ADMET characteristics in general as well as showing stability during 100 ns molecular dynamics
simulation. Bioassay study showed that both compounds yielded < 10 μg/mL inhibitory concentration.
Conclusion: This study has discovered two novel compounds using in silico approach, which can be further
developed as potential antimalarial agents

implementasi service quality pada loket pengurusan SKA di Dinas Perindustrian
dan Perdagangan (Disperindag) Provinsi Jawa Timur memiliki peran penting
dalam mendukung pertumbuhan ekonomi, salah satunya melalui pengurusan Surat Keterangan Asal (SKA) dalam bidang Pengembangan Perdagangan Luar Negeri (PPLN). SKA sangat penting untuk memperoleh preferensi tarif di negara tujuan ekspor. Namun, proses pengurusan SKA masih menghadapi berbagai kendala, seperti antrean panjang dan proses administrasi yang lambat. Laporan ini disusun berdasarkan praktik kerja yang dilakukan selama dua bulan (September- November 2024) untuk mengevaluasi kualitas layanan SKA. Pendekatan yang digunakan meliputi observasi dan wawancara. Hasil evaluasi menunjukkan bahwa lima dimensi kualitas layanan tangible, reliability, responsiveness, assurance, dan empathy perlu perbaikan signifikan. Upaya perbaikan yang dilakukan mencakup penerapan sistem antrean manual, penataan parkir, pelatihan petugas, dan peningkatan transparansi informasi. Meskipun langkah-langkah ini berhasil meningkatkan efisiensi, pengembangan lebih lanjut seperti penerapan sistem antrean elektronik masih dibutuhkan untuk meningkatkan kualitas layanan dan mendukung kelancaran ekspor
Effect of antioxidant dietary behaviour in smokers with chronic obstructive pulmonary disease risk during COVID-19 pandemic
Background: Smokers are at risk of Chronic obstructive pulmonary disease (COPD)which has a risk of decreasing in quality of life because of lung function decline and smoking habits. Decreased lung function due to an increase in free radicals. Intake of foods that contain antioxidants can help reduce free radicals.
Objectives: The research aimed to determine the effect of antioxidant intake (vitamins A, C, and E) on the development of COPD risk in active smokers.
Methods: This study was conducted in May-July 2023 in Surabaya. This study used a cross-sectional design, with purposive sampling and snowball sampling methods. The subjects were adult active smokers. Antioxidant intake consisted of the intake consumed by subjects daily containing vitamins A, vitamin C, and vitamin E using the Food Frequency Questionnaire (FFQ) method to describe nutritional intake in a certain period. COPD risk assessment was measured using a pulmonary function test with spirometry, with an assessment of the ratio of forced expiratory volume in one second (FEV1) / forced vital capacity (FVC) below 0.70 defined as airflow limitation. The data were tested using the Spearman test to see the relationship between antioxidant intake from food and lung function.
Results: This study involved 49 active smokers with COPD. Most subjects experienced deficiencies in vitamin A (77.55%), and vitamin C (87.76%), and all subjects experienced vitamin E deficiencies. The most consumed food containing vitamin A was palm oil, the most consumed food containing vitamin C was papaya, and the most consumed food containing vitamin E was chicken. There was no relationship between vitamin A and the development of COPD risk (Sig.=0.187; r=-0.192), but there was a significant relationship (weak negative) between vitamin C and the risk of developing COPD (Sig.=0.031; r=-0.309). All subjects had a deficiency of vitamin E.
Conclusions: The lower the intake of vitamin C, the greater the risk of COPD which is characterized by decreased lung function

Concerns about sustainability are shared by the corporate and academic groups. Organizations have become more knowledgeable about sustainability-related issues during the past 20 years, including what sustainability is, why it's significant, and how to manage and measure it. In 2016, the Dutch government unveiled its policy plan, “Circular Economy in the Netherlands by 2050.” The Netherlands will use fewer abiotic (mineral, metal, and fossil) resources thanks to the policy strategy. How Wageningen University & Research maintains its success in the field of higher education is the subject that this study seeks to address. We use a narrative approach. Wageningen University & Research's synergy has improved since the merger. It is the focus of many academic publications and has a substantial impact on environmental sustainability. Because we examine the story of Wageningen University & Research's success, a qualitative study is suited for this subject. The University of Indonesia, National Taiwan University, The Higher Education, the Government of the Netherlands, and particularly Wageningen University & Research are the primary targets of this study's narrative methodology, which focuses on published materials. The coordination of environmental, technological, and financial factors has been done by Wageningen University & Research. The importance of education in preserving the deteriorating environment that societal injustices have caused is growing. This study shows how Wageningen University & Research has expanded and continued to be successful. Environmental quality, sustainability, and governance have all been strategically managed
Perlindungan Hukum bagi Importir yang Menggunakan Metode Pembayaran Advance Payment atas Status Barang Impor yang ditahan oleh Otoritas Bea Cukai
Transaksi jual beli internasional atau transaksi ekspor impor semakin berkembang pesat dari tahun ke tahun. Transaksi ekspor impor ini dilakukan menggunakan perjanjian elektronik. Perjanjian elektronik tersebut harus memenuhi syarat sahnya suatu perjanjian yang tercantum dalam Pasal 1320 Kitab Undang-Undang Hukum Perdata dan Pasal 46 Ayat (2) Peraturan Pemerintah Nomor 71 Tahun 2019 tentang Penyelenggaraan Sistem dan Transaksi Elektronik. Namun pada kenyataannya masih banyak yang tidak dapat memenuhi peraturan-peraturan yang berlaku dalam transaksi ekspor impor, seperti yang dilakukan oleh Usaha Dagang NaSabe dan Date Palm Development, dimana pihak eksportir tidak dapat memenuhi kewajibannya atau tidak sesuai dengan yang diperjanjikan sehingga menyebabkan kerugian bagi pihak importir. Tujuan penulisan ini untuk menganalisis akibat hukum atas transaksi jual beli yang dilakukan oleh pihak eksportir. hasil penulisan ini menunjukkan bahwa perjanjian yang dilakukan dapat dibatalkan dikarenakan tidak sesuai dengan yang di perjanjikan sehingga pihak importir mendapatkan perlindungan hukum
Ketidakpastian Hukum Mengenai Syarat Usia Gubernur dan Wakil Gubernur: Analisis Putusan Mahkamah Agung Nomor 23/p/hum/2024
Berdasarkan Pasal 1 Ayat (3) Undang-Undang Dasar Negara Republik Indonesia Tahun 1945 ditegaskan bahwa Indonesia merupakan negara hukum, lebih lanjut di dalam Pasal 28D Ayat (1) Undang-Undang Dasar Negara Republik Indonesia Tahun 1945 telah diatur bahwa Setiap orang berhak atas pengakuan, jaminan, perlindungan, dan kepastian hukum yang adil serta perlakuan yang sama di hadapan hukum. Namun dalam dinamika bernegara terkadang terjadi hal-hal yang menyebabkan terjadinya ketidakpastian hukum. Seharusnya di dalam Negara Kesatuan Republik Indonesia yang merupakan negara hukum tidak terjadi ketidakpastian hukum karena hukum merupakan dasar acuan dalam bernegara sebagaimana telah diamanatkan di dalam Pasal 1 Ayat (3) Undang-Undang Dasar Negara Republik Indonesia Tahun 1945. Lembaga kehakiman seperti Mahkamah Agung dan Mahkamah Konstitusi memiliki peran yang paling penting dalam mewujudkan kepastian hukum. Activity Management System with Automatic Priority Adjustment using Simple Multi-Attribute Rating Technique
Activity management is needed to increase a person's productivity in arranging a more efficient activity schedule. To help organize activities, especially for workers or students who have many activities, the use of a planner is very necessary. However, the determination of inappropriate priorities in scheduling activities and difficulties in rescheduling are obstacles in using the planner. Therefore, this study aims to create an activity management system that is able to calculate the priority of each user activity and provide automatic rescheduling recommendations. The system implements the Simple Multi-Attribute Rating Technique (SMART) method for the priority configuration process. If the system's priority order does not match the user's priority order, then the weight changes for each criterion are carried out by a learning process.
The resulting replacement priority is used as a limitation in generating new schedule recommendations. Based on the
experiments, it can be concluded that the system has succeeded in helping to determine activity priorities automatically using the SMART method. From the validation results, the average value of the suitability of the ranking between the SMART calculation results and the user was 63.84% , indicating that the automatic ranking system has met the average expectations of respondents, although it is not yet optimal. The reschedule feature in the system based on a survey conducted resulted in a usability value of 68.75%, which means that on average this feature can help facilitate rescheduling
Pilot Study Efektivitas Single-session Intervention Berbasis Growth Mindset Terhadap Indikator Gangguan dan Kesehatan Mental Remaja Indonesia
Penelitian ini merupakan pilot study untuk mengevaluasi efektivitas single-session intervention berbasis growth mindset (GM-SSI) dalam mengurangi kecemasan, depresi, keputusasaan, dan meningkatkan keyakinan terkait kemampuan ketika menghadapi berbagai tugas pada remaja Indonesia. Intervensi GM-SSI dilakukan dalam satu sesi secara daring menggunakan desain randomised controlled trial, dengan melibatkan 76 remaja yang dibagi secara random ke dalam kelompok eksperimen dan kelompok kontrol, masing-masing terdiri dari 38 partisipan. Partisipan kelompok eksperimen menerima modul GM-SSI yang menekankan pada kepercayaan bahwa kemampuan dan keterampilan sseorang dapat dikembangkan dan dibentuk. Perbedaan skor indikator gangguan kesehatan mental remaja, seperti depresi dan kecemasan antara kedua kelompok dianalisis pada baseline, post-intervention, serta tiga dan enam bulan follow-up. Hasil penelitian menunjukkan bahwa GM-SSI belum menunjukkan efektivitas signifikan dalam menurunkan kecemasan, depresi, dan keputusasaan, serta meningkatkan keyakinan diri remaja untuk menyelesaikan suatu tugas. Temuan ini mengindikasikan bahwa GM-SSI perlu disesuaikan dan diuji kembali pada penelitian selanjutnya dengan mempertimbangkan faktor literasi sains dan motivasi remaja Indonesia. Studi lanjutan yang menyesuaikan konteks remaja Indonesia dan mode penyampaian modul intervensi diharapkan dapat meningkatkan efektivitas intervensi ini
AnalisisPerforma Algoritma BLAKE2b dan SHA-256 pada Implementasi Blockchain
The use of cryptographic algorithms in blockchain systems plays a vital role in ensuring data security and integrity. Among various algorithm types, hash functions serve a crucial purpose in linking blocks and detecting even the slightest changes in data. One of the most widely used algorithms in this context is SHA-256, which is well-known for its strong security features. However, it has limitations in terms of processing efficiency, particularly when implemented in large-scale systems or on devices with limited resources. On the other hand, the BLAKE2b algorithm offers faster performance with comparable security levels. In the study, tests were conducted and the results showed that the BLAKE2b algorithm completed the process in 0.067 ms, while SHA-256 took 0.162 seconds. This time difference indicates that BLAKE2b is 58.64% more efficient than SHA-256 in the same testing scenario. In addition to speed, memory consumption was also measured, and SHA-256 was found to consume 23.71% less memory than BLAKE2b. This demonstrates that the choice of algorithm for blockchain depends on resource requirements and technical aspects
Lobster Puerulus Trading and Cultivation in Situbondo: A Financial Feasibility and Risk Analysis
Lobster (Panulirus spp.) aquaculture holds significant economic potential for coastal Indonesia, including Situbondo, East Java, relying heavily on wild puerulus harvest. However, developing sustainable cultivation and trading ventures faces considerable financial uncertainties due to biological, market, operational, and policy risks. This study assesses the financial feasibility and identifies key financial risks for lobster cultivation and puerulus trading in Situbondo. An integrated framework combining financial analysis with Monte Carlo Simulation for risk assessment was used, utilizing primary data and secondary sources. Probabilistic analysis identified key risk factors impacting profitability through sensitivity analysis. The financial modelling and analysis resulted positive valuation for both trading (48 billion rupiah NPV, B/C ratio 1.12) and cultivation (0.99 billion rupiah NPV, B/C ratio 1.52). The risk analysis identified 4 main risk factors, including selling price, survival rate, exchange rate, and interest rates. Ultimately, this risk-integrated feasibility assessment provides crucial insights for investors, farmers, and policymakers, supporting informed decisions for sustainable lobster aquaculture development in Situbondo
