Clozapine-induced Toxicity via Induction of Oxidative Stress and Mitochondrial Dysfunction in Human Blood Lymphocytes and Protecting role of L-Carnitine

    No full text
Clozapine is a useful antipsychotic drug but with serious, life threatening toxicity effects. The aim of this study is to assess the direct cytotoxicity effect of clozapine (CLZ) on human blood lymphocytes and investigate the protective effect of L‐carnitine (LC) against clozapine‐induced cytotoxicity. Clozapine at 70 μM concentration induced cytotoxicity following 12 h. The Clozapine induced cytotoxicity was associated with intracellular reactive oxygen species (ROS) generation, mitochondrial membrane potential (MMP) collapse, lysosomal membrane injury, lipid peroxidation, and depletion of glutathione (GSH) and raising of oxidized glutathione (GSSG). We showed that LC (1 mM) has a beneficial cytoprotective effect against clozapineinduced toxicity. Summery, clozapine causes organelles damages and triggers oxidative stress in lymphocytes. These data suggest that using of L‐carnitine could be useful for prevention and treatment of clozapine toxicity. Finally, it could be concluded that LC exerts a beneficial antioxidant and mitochondrial protective effects against oxidative stress induced by CLZ

Introduction: Implant-supported over-dentures (IODs) are an accepted and predictable form of treatment for the edentulous jaw. The purpose of this prospective clinical study was to evaluate the effect of mandibular implant overdenture on Peri-implant Tissues Materials and Methods: A prospective study was carried out with 24 edentulous patients (n = 24; mean age 67.13 years). In all patients, two dental implants were placed in the interforaminal region of the mandible and after a 3-month healing period overdentures were fabricated .Clinical and radiographic parameters were evaluated 6 and 12 month after completion of the prosthetic treatment . Implant loss, bleeding on probing , gingival recession , Attachment level and probing depth were assessed as clinical parameters. Peri-implant bone loss was assessed on dental radiographs taken with a paralleling technique . Peri-implant tissue health was assessed, and the presence of mucositis and peri-implantitis was recorded Results: 24 patients who received 48 implants were evaluated. The average marginal bone loss of patients was 0.33mm after 6 month of evaluation and 0.63mm after one years . the mean probing depth was 1.89mm and 2.19mm respectively after 6 and 12 month of evaluation . No implants were lost and no peri implantitis occurred. the survival rate was 100% after one year of function. Conclusion: Considering the limitations of this short term clinical study, two implants to retain mandibular overdentures seems to be a feasible treatment option with a high survival rate for patients with insufficient jawbone volume

Background & Objective: The serum decoy receptor 3 (DcR3) level increases in chronic inflammatory diseases. The present study aimed to examine serum DcR3 and IL-6 levels in patients with stable and acute exacerbation of chronic obstructive pulmonary disease (COPD) and the correlation of this markers with airflow limitation. Methods: We measured serum DcR3 and IL-6 levels in 60 COPD patients (30 stable COPD (SCOPD), and 30 acute exacerbation of COPD (AECOPD)), and 30 control subjects and compared them with airflow limitation according to the COPD stage in the Global Initiative for Chronic Obstructive Pulmonary Disease (GOLD) criteria, peripheral O2 saturation (SpO2), and COPD Assessment Test (CAT) score. We also tested the association of serum DcR3 level with COPD patients’ clinical parameters. Results: Both serum DcR3 and IL-6 levels increased with increasing severity of airflow limitation in SCOPD and AECOPD groups (P < 0.01 to P < 0.001, respectively). It also increased in patients with AECOPD group compared with SCOPD group in GOLD stages III-IV (P < 0.05 to P < 0.001, respectively). In addition, there was a significant positive correlation between serum DcR3 level and IL-6, CAT score and Smoking history (per year). Conclusion: The study showed that serum DcR3 level increased with an increasing severity of airflow limitation in COPD patients, markedly in acute exacerbation phase. This increase was associated with a reduced quality of life and increased severity of hypoxia. These results suggest that DcR3 in COPD patients may be participate with its pathophysiology

Introduction: The complications of fixed partial dentures and single crowns have a great importance to be evaluated. The objectives of this study were to determine the prevalence of complications associated with crowns and bridges and the effect of service duration on the prevalence of complications among a Population Presenting for Treatment to a Dental School in Ardebil. A total of 40 patients were selected. Qualities of the present fixed partial dentures were clinically and radiographically assessed. Clinical examinationwas performed. The results of this study showed that the percentage of the failures were Periodontal disease (53.8%), Open contact (25.6%), porcelain fracture (20.5%), open margins (10.3%), Occlusal problem (7.7%), Poor esthetics (7.7%) ,prostheses loose (5.1%) and endodontic complications (0%). The duration of service was found to influence most of the assessed complications especially periodontal disease and open contact. With limitation of this study, the periodontal disease and gingival bleeding are the most frequent complications responsible for failure of fixed partial denture and single crowns that recommend an effective method to control dental diseases and to improve dental awareness. Method and Materials: A total of 40 patients (age range of 20-60 years, 15 of the participants were females and 25 males) were selected randomly from the dental clinic at Ardebil Medical College, Dentistry Program. A sample questionnaire will prepared in order to collect the relevant data. All prostheses will be performed by several general dental practitioners with a minimum of one year service duration. Patients will informed and written consents will be obtained. Age, gender and the time of prostheses placement will be recorded. Complete clinical and radiographic examinations will be conducted. Statistical analysis was done using a Statistical Package for Social Science (SPSS, version 22). Descriptive statistics, including means and standard deviations were calculated for continuous variables, frequency and percentage for categorical variables, were calculated Results: With limitation of this study, the periodontal disease and open contact are the most frequent complications responsible for failure of fixed partial denture and single crowns

Background and Objective: Postoperative shivering is characterized by involuntary movements of one or more muscle groups in the early stages of recovery after a general anesthesia. This complication can lead to many problems for patients. In this study, we evaluated the efficacy of intravenous Acetaminophen and Ondansetron in preventing of shivering in patients undergoing general anesthesia with appendicitis diagnosis and compared the efficacy of each. Methods: This study is a triple blind clinical trial. It was conducted on 150 patients with ASA class I and II undergoing appendectomy with general anesthesia in Fatemi Hospital affiliated by ARUMS during 2017-18. These patients were randomly divided into three groups of Acetaminophen and Ondansetron and placebo. Patients were unaware of the type of drug received. Half an hour before surgery, drugs were injected in groups, then the endotracheal tube was removed and the patients were transferred to the recovery room and shivering was measured up to half an hour after the operation. Results: Survey of demographic data showed that there was no significant difference between sexes, mean age, weight average and ASA distribution in study groups in three groups(p>0.05). Incidence and severity of shivering in recovery room in Ondansetron group were significantly lower than Acetaminophen group and placebo group (p<0/05). Evaluation of the incidence of nausea and vomiting in the recovery room in the group receiving ondansetron was significantly lower than the other two groups (p<0/05). VAS-based pain assessment was indicated at recovery room, where pain score in ondansetron group was significantly higher than other groups and acetaminophen had the greatest effect on pain relief (p<0/05). coclusion: This study showed that ondansetron is a suitable and effective option in the prevention of postoperative shivering. According to the results, decreasing nausea and vomiting in the ondansetron group could also be another advantage of ondansetron

Particulate matters can be transferred distances from their sources. Due to different chemical and microbial characteristics, they can potentially impact on public health and ecosystems. Therefore, in this study we aimed to study the concentrations of particulate matters (PM2.5 and PM10) and types of bacteria in the ambient air of Ilam city during dusty (PM10 ≥ 150 µg/m3) and non-dusty days from February 2012 to February 2013. Totally, there existed more dust storm events during warm and dry months. The Mann-Whitney test confirmed a significant relationship between dusty and non-dusty days (P < 0.05). Totally, by increasing the PM concentrations, the number of bacterial colonies increased. Kruskal-Wallis test showed that there was a significant relationship between the number of bacterial colonies and season. The mostly observed bacterial colonies were Staphylococcus, Cryptococcus, Corynebacterium, Bacillus, Actinosynnema, Nocardioides, Arthrobacter, Flavimonas, Paenibacillus, Pseudomonas, Microbacterium, Planococcus, Streptomyces, Kurthia, Neisseria Agrococcus, Curtobacterium, Duganella, Ancylobacter, Paracoccus, Propionibacterium, Pseudomonas, Rhizobium and Enterococci. The predominant direction of dust plumes in Ilam city was from the west of Ilam from neighboring countries. The higher number of microbial colonies during dusty days in this city obviously showed the important role of these events on microbial characteristics
