Agrostemma githago increases caspase 3/7 activity in Leishmania major promastigotes
Introduction and objectives: Acquired resistance to anti-leishmanial drugs are increasing worldwide. Anti-leishmanial effect of Agrostemma githago seed extract on Leishmania major promastigotes has already been shown in our laboratory. This study was conducted to assess the induction of apoptosis in Leishmania major promastigotes after treatment with Agrostemma githago seed extract.
Methods: The activity of caspase 3/7 as the key executioners of apoptosis were measured using the Caspase-Glo 3/7 Assay kit. Following treatment with EC50 dose of Agrostemma githago (0.4 mg/ml), 1:1 ratio of Caspase-Glo 3/7 reagent and Leishmania major promastigotes suspension containing 5 × 106 cells were added into white-walled 96 well plates and incubated at room temperature for 3 hours. The cleavage of tetrapeptide sequence DEVD by caspase enzymes was measured at 570 nm emission using plate-reading luminometer (Synergy HT, BioTek). Data are reported as the relative caspase activity in comparison to control group.
Results: Our data showed that the activity of caspase 3 and 7 were increased in Agrostemma githago received group in comparison to untreated control.
Discussion and Conclusion: Aqueous extract of Agrostemma githago seed induced apoptosis in Leishmania major promastigotes via increasing caspases 3 and 7 enzymes activity
Frequency of Ocular Complications and its Effective Factors in Type 1 Diabetic Patients Referred to Eye Clinic of Imam Reza Hospital from September 2017 to August 2018
Background and objective: Visual complications are one of the major health problems in diabetic patients, especially in undevelopedand developing countries, due to the lack of similar studies in Ardabil province, this study was conducted with the aim of evaluating the frequency of ocular complications and its effective factors in a sample of patients with type 1 diabetes in Ardabil.
Methods: In this one-year cross-sectional study, all type 1 diabetic patients referred to the internal clinic of Imam Khomeini Hospital during September 2017 to August 2018 were referred to the ophthalmology clinic of Imam Reza Hospital and were examined for the presence or absence of ocular complications. On arrival at the eye clinic, the necessary clinical tests and the eye examinations were performed for all the patients and the required information was entered into the pre-prepared checklists. Patients' eyes were examined by an eye specialist.
Results: A total of 66 type 1 diabetic patients with the mean age of 25.6 ± 10.8 years (range 5 to 49 years) were studied that the number of males was slightly higher (54.5% vs. 45.5%). The most common ocular complications were lens opacity, persistent corneal epithelial defect, diabetic retinopathy, staphylococcal blepharitis, and meibomian gland dysfunction that are seen in 54.5%, 27.2%, 10.6%, 6.1%, and 3% of patients, respectively. Mean HbA1c was higher among patients with diabetic retinopathy compared to patients without retinopathy. The frequency of retinopathy was higher among male patients compared to female patients (13.9% vs. 6.7%), among patients older than 20 years compared to younger patients (14.3% vs. 0%), and among patients with a disease duration of more than 10 years compared to patients with a shorter disease duration (20% vs. 4.9%).
Conclusion: The present study showed the types of ocular complications among a sample of patients with type 1 diabetes in Ardabil that the lens opacity, persistent corneal epithelial defect, and retinopathy were the most common ones. Retinopathy was associated with longer disease duration and higher age. These findings highlight the need for regular screening of ocular complications in patients with type 1 diabetes, especially in cases who are at higher risk, namely patients older than 20 years of age and patients with a disease duration of more than 10 years
The effectiveness of social skills training in decreasing social anxiety in students with Specific learning Disorder (SLD)

The aim of this study was to determine the effectiveness of social skills training in decreasing social
anxiety in students with specific learning disorder.This research was Study quasi-experimental with the
pre-test, post-test and a control group. The statistical population of the present study included all fifth
grade elementary school students with specific learning disorder who were studying in Ardabil in 2015.
The research sample consisted of 40 students with specific learning disabilities, selected by multistage
cluster sampling and assigned to two experimental and control groups (20 subjects per group).To collect
data, Kay Math mathematic, Raven Intelligence Test, Reading Test of Shafiei et al, Falahchay writing
Expression, social anxiety scales and Diagnostic Interview based on DSM-5 were used. Test Data were
analyzed by multivariate of Covariance analysis (MANCOVA) model in the SPSS software version 22.
The results showed that social skills training positive effect in decreasing on the social anxiety in
students with specific learning disorder.Based on the findings of this study can be considered Because
of that social skills training can develop to adaptive behaviors and Adjustment appropriate responses to
the environment. Thus, This can result in positive effect in decreasing on social anxiety in this students
Comparing the Effect of Mentalization-Based Therapy and Cognitive Analytical Therapy on Ego Strength and Defense Mechanisms of People with Borderline Personality Disorder
ims Mentalization-Based Therapy (MBT) and Cognitive Analytic Therapy (CAT) are psychotherapy ap-
proaches offering specific methods for the treatment of Borderline Personality Disorder (BPD). There-
fore, this study compared the effectiveness of MBT and CAT on improving the ego strength and defense
mechanisms in people with BPD.
Methods & Materials This was a quasi-experimental study with a pretest-posttest and a control group de-
sign and follow-up stage. In total, 40 men with BPD were selected by purposive sampling technique. The
samples were selected from males admitted to Isar psychiatric Hospital and the psychiatric department of
Fatemi Hospital in Ardabil City, Iran, in 2018. The samples were randomly assigned into two experimental
groups and one control group. The first experimental group received 20 sessions of MBT, and the second
experimental group received 16 sessions of CAT. To collect data, we used psychological inventory ego
strength and defense styles questionnaire. The obtained data were analyzed using mixed model Analysis
of Variance (ANOVA) and Scheffe posthoc test.
Findings The collected results suggested that both interventions were effective in improving the ego
strength and defense mechanisms of people with BPD (P<0.001). However, the effect of MBT on the over-
all score of the ego strength and the immature defenses was higher than that of the other intervention
(P<0.05). Moreover, there was no significant difference in neurotic defense between the two interven-
tions (P>0.05).
Conclusion The present research results can support the use of these therapies as part of a non-pharma-
ceutical treatment program in different treatment centers or psychiatric hospitals
Effect of Nurse-led care on Quality of Care and Level of HbA1C in Patients with Diabetic Foot Ulcer
Background: Diabetic foot ulcer (DFU) is a serious and costly complication in diabetes which affects different aspects of life and can reduce patient’s quality of life. Various views to manage DFU have been introduced. The Nurse-led care as a multidisciplinary team can be effective due to a holistic approach to some disease management but patients with DFU have no assess. Therefore, this study was designed to determine the effect of Nurse-led care on quality of care and improvement of HbA1C in patients with Diabetic Foot Ulcer.
Methods: This was a randomized clinical trial study performed on 52 patients with DFU. Patients were randomly assigned into two groups: Nurse-led care intervention and standard care. The study was conducted in one of the hospitals affiliated to Tehran University of Medical Sciences. Data was collected using two questionnaires: demographic characteristics and Quality Patient Care Scale (QUALPACS) and taking a blood sample for HbA1C. The Nurse-led care interventions were conducted in three stages: Integrated, Interdisciplinary, and Comprehensive. Descriptive and analytic statistical methods were used to analyze the data. p<0.05 was considered significant.
Results: The results demonstrated that according to repeated measures test, (before, 4 and 12 weeks after the intervention), the level of Quality of Care dimensions (psychosocial, communication and physical aspect) in the intervention group had a significant difference with control group (P<0.0001). Also, according to the results of Independent T test, there was a significant difference in total Quality of Care scoring and HbA1c between intervention and control groups after the intervention and follow up (P<0.0001).
Conclusion: Considering the burden of diabetes and diabetic foot ulcers, it seems that the establishment of a nurse-led care approach can be an effective strategy to manage and treat these patients, and eliminate the disruption of care and achieve optimal care quality
Investigation of Electro-coagulation and Advanced Oxidation (UV/Na2S2O8) Processes efficiency in Reactive Blue52 removal from aqueous solutions
Background and & Objectives: Colored wastewaters are mainsources of aquatic pollution which will cause considerable impacts to the environment if discharge without treatment. Therefore, the purpose of this study was investigation of Electro-coagulation and Advanced Oxidation (UV/Na2S2O8) Processes efficiency in Reactive Blue52 removal from aqueous solutions
Methods: In this experimentall study a glass beaker equipped with 2 iron electrodes with total effective surface area of 60 cm2, as a electro-coagulation reactor, and a cylindrical reactor equipped with 2 UVc lamps (6wat), as a Advanced Oxidation reactor, were used to remove Reactive Blue52. The operating parameters of electro- coagulation process such as initial Reactive Blue52 concentration, pH, current density and reaction time were investigated. Then obtained effluent was introduced to UV/Na2S2O8 process and operating parameters such as sodium persolfat dosage and pH were examined
Results: The results showed that Reactive Blue52 removal efficiency of 82/4% was achieved in pH=7, current density=10 mA/cm2, and reaction time of 30 min. In combination of electro- coagulation and UV/Na2S2O8 (pH=7 and Na2S2O8=15mM/L and reaction time of 90 min) processes the dye and COD removal efficiency of 100% and 76.9%was attained. (respectively)
Conclusion: The results revealed that combination of electro- coagulation and Advanced Oxidation (UV/ Na2S2O8) processes is an efficient method to remove Reactive Blue52 from aqueous solution
Design, synthesis and evaluation of cytotoxic activity of novel heterocyclic compounds with 1,2,3,4-terahydropyrimidine scaffold against A549, Hep-G2 and MCF-7 cell lines
Introduction
Cancer is one of the main problems worldwide and is the second leading cause of death after cardiovascular diseases. Cancer is the due to a series of molecular events that by inactivating of cell proliferation control systems, lead to excessive cell growth and division and attacks other tissues. Cancer is mainly treated by three methods, surgery, radiation therapy and systemic treatment. Despite advances in cancer prevention and treatment, problems such as resistance to existing drugs and severe side effects of drugs due to their low selectivity require researchers to design and develop new structures with more potency.
Materials and Methods
According to studies on the cytotoxicity of some compounds, in this project a series of new 1,2,3,4-tetrahydropyrimidine derivatives were synthesized using the Biginelli reaction and after purification, structures of the synthesized compounds were confirmed by 1H-NMR, FT-IR and MS spectra. Thus, the cytotoxicity of the synthesized compounds evaluated against three A549, Hep-G2 and MCF-7 human cancer cell lines using MTT assay.
Results
Our results showed, compound benzyl 6-methyl-4- (4-nitro phenyl) -2-thioxo-1,2,3,4-tetrahydropyrimidine-5-carboxylate (S8) having the electron withdrawing group (NO2) at para position of phenyl ring on C4 position, thioxo group at C2 and benzyl carboxylate group at C5 was the most potent compound among all compounds against three cell lines (IC50 = 65.54 µM in MCF-7 cell line, IC50 = 73.71µM in Hep-G2 cell line and IC50 = 43.97 µM in A549). According to the results of the statistical analysis, the cytotoxic effect of all the derivatives synthesized at all concentrations was significantly different compared to the negative control group (P< 0.05) and all concentrations of the synthesized compounds had significant inhibitory effects in all three periods of 24, 48 and 72 hours compared to similar concentrations of doxorubicin as positive control (P˂0.05). Also, the viability of cells treated with four concentrations of synthetic compounds in cancer cells (P<0.05) and normal cells at all concentrations was significantly different and at 100 μM concentration more significant for most compounds (P< 0.01). The cytotoxicity results of these compounds on Human umbilical vein endothelial cell (HUVEC) as normal cell line also showed that they were partially selective. Among compounds, compound S1 was more selective against three cell lines at the 100 μM concentration.
Discussion and conclusion
Investigation of the structure-activity relationship of the synthesized compounds indicates that the increase lipophilicity and the presence of strong electron withdrawing group at the C4 position of phenyl ring increases cytotoxicity
Synthesis and characterization of plasmonic quantum dots for the development of MicroRNA-134 optical biosensors
Introduction and goal
MicroRNAs are non-coding sequences that are 18 to 28 nucleotides in length. These biological molecules play an important role in cellular processes. In particular, they affect the expression of proteins in the human body and the formation of cellular structures. one of the diseases that severely affect human societies are mental illnesses. Bipolar disorder is one of the most common and debilitating diseases that is mostly diagnosed on the basis of clinical symptoms, MicroRNA 134 has been shown to play a key role in the formation of the disease and in the manic phases its concentration in plasma is strongly increased. Researchers are using this microRNA as a diagnostic biomarker to enable more accurate diagnosis and follow-up of treatment. The aim of this thesis is to synthesize microRNA 134 biosensor and record the optical signal in the presence of miR-134 spiked into the blood plasma.
Methods
The dot quantum nanoparticles were first synthesized by the aqueous method and a thin gold coating was formed after mapping around it. After preparing the aforementioned nanoparticles, the particle surface properties and absorption and emission spectra are studied. Finally, the hybrid bonding of plasmonic nanoparticles with microRNAs and its optical properties are studied in buffer and plasma environments.
Findings
The dot quantum nanoparticles were obtained at 3.2 nm and increased to 6.7 nm after coating, allowing the coating of biomolecules, especially DNA strands. Various optimizations were made for the present biosensor, and the biosensor with a very low detection limit of 150 pg / ml in the appropriate linear range was able to generate a suitable and repeatable response within 21 minutes.
Discussion and conclusion
The biosensor was tested in serum and was able to detect microRNA 134 present in the serum, meaning it could detect microRNA 134 from blood samples taken from mania patients and detect the disease in a faster time
Effects of Chrysin in Inhibiting the Toxicity Induced by Aluminum Phosphide in Isolated Mitochondria Obtained from Rat Heart Muscle
introduction
Apart from the anticancer, antioxidant, anti-inflammatory effects and inhibition of aromatase, chrysin is involved in the protection of cardiovascular disorders. Cardiovascular complications are the main cause of death induced by aluminum phosphide (AIP) which are related to oxidative stress and mitochondrial damages. For this purpose, we investigated the effect of chrysin as an antioxidant and mitochondrial protective agent against AIP-induced toxicity in isolated cardiomyocytes and mitochondria obtained from rat heart ventricular.
Material and method
Using by biochemical and flowcytometry, mitochondrial toxicity parameters such as mitochondrial NADH/succinate dehydrogenase activity, mitochondrial swelling, reactive oxygen species formation, mitochondria membrane potential and lipid peroxidation were analyzed in isolated mitochondria.
Discussion and conclusion
our finding in isolated mitochondria showed that chrysin (up to 10 µM) significantly (P < 0.001) decreased AIP induced mitochondrial toxicity. These findings demonstrated that chrysin as an antioxidant and mitochondrial protective agent exert protective effect in wild-type cardiomyocyte treated with aluminum phosphide. It was concluded that chrysin significantly reduced the toxicity of AlP in isolated cardiomyocytes and mitochondria. Due to the very low toxicity of chrysin to humans it could be a promising agent in treatment of AlP poisoning
