    Isolation and primary culture of chick embryonic neural crest cells

    Aim: we aimed at presenting a simple and efficient method for isolating and characterizing the neural crest cells. Material and Methods: The hen’s fertilized eggs were incubated for about 35h at 38°c and 55-60% humidity until the embryos reached to stages 10-12 according to Hamburger-Hamilton developmental stage table. Then the embryos were removed from the egg’s yolk and the neural tube was isolated and cultured for 24 h in a tissue culture dish to release neural crest cell. Then after, the neural tube was removed and allowed to NCC to expand for further 5 days. Finally, the cells were collected and subjected to PCR to study their gene expression profile. Results: The neural tube released NCC and these cells proliferated in culture condition. They also expressed markers including Slug, Sox9 ,and Sox10 by the RT-PCR method. Conclusion: The neural tube can release NCC in culture condition and these cells can proliferate in the presence of an appropriate medium

    Investigation of Cause of Abnormal Uterin Bleeding in Postmenopusal women : A review study

    Background and Aim: Abnormal uterin bleeding (AUB) is a common gynecologic problem in postmenopusal women. It can be caused by uterine abnormalities or due to other reasons. Any bleeding with excessive duration, frequency and amount is AUB that occur based on the several etiology: polyp; malignancy; coagulopathy; iatrogenic; etc. ACOG recomends endometrial sampeling as first diagnostic test in women with AUB who are older than 45 years. The aim of this study was to Investigat the cause of abnormal uterin bleeding in postmenopusal women. Materials and Methods: This study was a simple review and reasons of abnormal uterin bleeding in postmenopusal women was investigated from 2009-2019 using several databases including Cochrane, Scopus, Medline, Web of Science, Embase, Google Scholar, Iran medex, and Scientific Information Database with keywords: AUB; postmenopusal bleeding and menopause. 14 articles were selected based on the research criteria. Results: Results showed that the most common finding in Postmenopusal women with AUB was atrophic endometrium. Other commen causes were hyperplastic endometrium; endometrial carcinoma and endometrial polyp, respectively. The most abnormal pathologies were seen in women with diabetes, hypertension and hypothyroidism. Conclusion: The results of the present study underscore endometrial sampling should be performed to detect endometrial cancer and hyperplasia in postmenopausal women with AUB, specially in women who are 60< and with endometrial volume ≥3 ML. In all cases with postmenopausal bleeding, focal lesions and endometrium 5 mm≤ operative hysteroscopy should be performed

    Wharton’s Jelly-derived Mesenchymal Stem Cells and Polycaprolactone/Hydroxyapatite (PCL/HA) Scaffold for Bone Tissue Engineering

    Background: Polycaprolactone (PCL) is a suitable polymer in the field of drug delivery and tissue engineering scaffolds. Aim of Work: This study aimed to design a Polycaprolactone /Nano hydroxyapatite (PCL/nHA10%) hybrid nanofiber scaffold and evaluate differentiation of Wharton’s jelly-derived mesenchymal stem cells (WJ-MSCs) to osteogenic lineage on scaffolds for bone tissue engineering. Materials and Methods: In this study, PCL/nHA(10%) composite polymeric nano scaffolds were prepared by electrospinning. Mesenchymal stem cells were isolated from the Warton’s Jelly and cultured in the PCL/nHA 10% scaffold. Biocompatibility of scaffolds was confirmed by MTT assay. Characterization of umbilical cord mesencymal stem cells were performed using flow cytometry. The morphological and cell adhesion characteristics of mesenchymal stem cells (MSCs) on the scaffolds were performed using scanning electron microscopy (SEM) imaging. Finally, the cells were treated with osteogenic differentiation medium for 21 days to investigate their differentiation potential on the scaffolds and then differentiated cells were stained with alizarin red and von Kossa stains. Results: The largest pore size of PCL/nHA (10%) nanofibers was about 22 μm. The culture of WJ-MSCs on the scaffolds showed that the addition of nHA 10% to PCL scaffold caused further attachment and proliferation of the cells. The stained mineral deposit scaffold with alizarin red and von Kossa were also compared. In both types of staining, nano-composite scaffold showed higher calcium deposits. Conclusion: The results showed that the PCL/nHA 10% scaffolds for bone tissue engineering is a good choice, and can be used for bone tissue repair

    Comparison of the Effect of Kalsitrapine and Metformin as an adjunct therapy on infertility treatment in women with PCOS referring to Alavi Hospital in Ardebil

    Background: Chronic anovulation is one of the most common causes of infertility. In women with PCOS, other factors related to oocyte quality or endometrial and nesting disorders may also play a role. Infertile and non-ovulatory women who tend to become pregnant are candidates for ovulation induction. Aim: Considering the importance of PCOS syndrome and the importance of its treatment, the aim of this clinical trial was to evaluate the effect of kalsitrapine and metformin on the treatment of patients with PCOS. Methods and material: The study was performed as a clinical trial. A total of 80 female patients with PCOS who had referred to the obstetrics clinic of Alavi Hospital for infertility treatment were randomly selected with personal consent and entered the study. Results: The mean age of the subjects was 21.5±08.21 years. There was no statistically significant difference between the two groups in terms of age of patients (P >0.05). The mean age of the subject’s husbends was 31.49±3.7 years. There was no statistically significant difference between the two groups in terms of subject’s husbends age (P >0.05). There was no statistically significant difference between the two groups in terms of BMI (P >0.05). There was no statistically significant difference between the two groups in terms of primary and secondary infertility (P >0.05). The mean duration of infertility was 3.68±3.49 years. There was no statistically significant difference between the two groups in terms of infertility duration (P >0.05). There was no statistically significant difference between the two groups in terms of the type of menstrual cycle (P >0.05). The response rate to treatment was 17.5% in the metformin group and 10% in the kalsitrapinegroup. There was no statistically significant difference between the two groups in terms of response to treatment (P >0.05). In comparison, none of the studied factors in the two groups was significantly associated with the response to treatment (P >0.05). Conclusion: The success rate of treatment with metformin as an adjunct in the treatment of infertility was 17.5% and this rate was obtained in kalsitrapine10. There was no significant difference between the two findings, but the high success rate of metformin could indicate that calcitropine, as a new herbal medicine, did not have a significant advantage over existing drugs

    Vitamin D3 and erythropoietin protect against renal ischemia‑reperfusion injury via heat shock protein 70 and microRNA‑21 expression

    Kidney ischemia reperfusion (IR) contributes to the development of acute kidney injury. The hypoxic conditions in ischemic damage lead to oxidative stress and apoptotic cell death. We investigated the effects of vitamin D3 (Vit D) and erythropoietin (EPO) on microRNA-21(miR-21) expression in renal IR. Wistar rats were divided into five groups including the control, vehicle + IR, Vit D + IR, EPO + IR, and Vit D + EPO + IR groups. The animals were unilaterally nephrectomized and subjected to 45 min of renal pedicle occlusion followed by 24 h reperfusion. Vitamin D3 and EPO were administered prior to ischemia. After 24 h reperfusion, the kidney samples were collected for the detection of miR-21, heat shock protein 70 (hsp70) and caspase-3 expression levels. Kidney IR significantly increased the expression of miR-21, hsp70 and capase-3 and blood urea nitrogen (BUN)-Cr levels. Treatment with vitamin D3 and EPO significantly decreased the BUN-Cr levels and hsp70 and caspase-3 expression. Also, the co-administration of two drugs significantly increased miR-21 expression. It seems that vitamin D3 or EPO administration could protect the kidney against IR injury. However, vitamin D3 and EPO co-treatment was the most effective compared with the other treatment groups

    Association between Molar Incisor Hypomineralization and Both Prenatal and Postnatal Factors in 8-10-Year-Old Children in Ardebil

    Molar incisor hypo-mineralization is a malformation with a systemic background for which aetiology is not well defined. Prevalence of these lesions varies in different countries, but few studies have been conducted on its prevalence in the Iranian society. The aim of this study was to determine the prevalence of molar incisor hypo-mineralization among 8-10-year-old children in Ardabil and its relationship with prenatal and postnatal factors. METHODS In this analytical cross-sectional study, participants included 520 children from public schools of Ardabil. In the first stage, the frequency of MIH was determined by a clinical examination based on the index introduced by Tourino et al. Then, they were evaluated in two groups (with or without incisor hypo mineralization) to determine the causative factors. To collect information, a questionnaire including information about factors before and after birth was used. Data were analysed using SPSS 18 software. Chi-square and T-test and logistic regression were used to test the relationship between different factors. RESULTS The prevalence of molar-incisor hypo mineralization in children aged 8-10-year-old in Ardabil was estimated to be 24 %. The results also showed that there is a significant relationship between latarerp and postnatal factors (p < 0.05). CONCLUSIONS Due to the high prevalence of MIH in Ardabil, preventive measures, informing the parents and periodic examination of the teeth are recommended

    The health effects of quarantine during the COVID-19 pandemic

    BACKGROUND: Quarantine is considered as an effective solution in the early stages of an epidemic. In the case of the coronavirus epidemic, quarantine was also recommended and implemented as a significant guideline to prevent the disease. However, despite the benefits of quarantine, there are also complications and problems. OBJECTIVE: The present study aimed to investigate the health effects of quarantine during the COVID-19 pandemic. METHODS: This study was conducted as a literature review through searching the databases Google Scholar, PubMed, and Science Direct for papers published before July 2020. The research was conducted based on the keywords 'Coronavirus,' 'COVID-19,' and 'quarantine.' The references of the papers were also reviewed to find the ones not found in the databases. The guidelines published by reputable organizations such as the World Health Organization (WHO) and Centers for Disease Control and Prevention (CDC) were used in this study. RESULTS: Although quarantine is applied as an important and primary solution in the outbreak of epidemics, in cases of pandemics, it may not be free of negative effects on individuals and public health. However, because of the need to reopen and restart social and economic activities, some changes should be made in lifestyles and work activities. Using cyberspace and telework can be helpful. As the findings showed, COVID-19 bubbles can be used to restore social communications. CONCLUSION: Using masks, avoiding unnecessary gatherings, complying with personal and social hygiene, and respecting social distancing can be valuable solutions that, if implemented properly, can decrease the rate of the disease significantly. It is also emphasized that quarantine is still necessary and important as the best solution for sick people and individuals who are suspected carriers of the disease

    Global and regional burden of cancer in 2016 arising from occupational exposure to selected carcinogens: a systematic analysis for the Global Burden of Disease Study 2016

    Objectives This study provides a detailed analysis of the global and regional burden of cancer due to occupational carcinogens from the Global Burden of Disease 2016 study. Methods The burden of cancer due to 14 International Agency for Research on Cancer Group 1 occupational carcinogens was estimated using the population attributable fraction, based on past population exposure prevalence and relative risks from the literature. The results were used to calculate attributable deaths and disability-adjusted life years (DALYs). Results There were an estimated 349 000 (95% Uncertainty Interval 269 000 to 427 000) deaths and 7.2 (5.8 to 8.6) million DALYs in 2016 due to exposure to the included occupational carcinogens—3.9% (3.2% to 4.6%) of all cancer deaths and 3.4% (2.7% to 4.0%) of all cancer DALYs; 79% of deaths were of males and 88% were of people aged 55 –79 years. Lung cancer accounted for 86% of the deaths, mesothelioma for 7.9% and laryngeal cancer for 2.1%. Asbestos was responsible for the largest number of deaths due to occupational carcinogens (63%); other important risk factors were secondhand smoke (14%), silica (14%) and diesel engine exhaust (5%). The highest mortality rates were in high-income regions, largely due to asbestos-related cancers, whereas in other regions cancer deaths from secondhand smoke, silica and diesel engine exhaust were more prominent. From 1990 to 2016, there was a decrease in the rate for deaths (−10%) and DALYs (−15%) due to exposure to occupational carcinogens. Conclusions Work-related carcinogens are responsible for considerable disease burden worldwide. The results provide guidance for prevention and control initiatives

    Evaluation of pulmonary function tests in male bakeries in Ardabil city in 1398

    Background and purpose: The respiratory system is one of the areas that are most vulnerable to unhealthy workplace injuries. One of the most common occupational diseases, are illnesses and respiratory complications in employees who are exposed to harmful pollutants in the workplace. Flour powder is a complex organic particle with different components. These ingredients include wheat bran, pollen, starch particles and a variety of bacteria. Given the importance of respiratory illness and the impact it has on the quality of life of these people, in this study we compared the pulmonary function of bakery workers with that of healthy people. Materials and Methods: A total of 51 bakers who had been in working in bakeries in Ardabil for at least 5 years and 51 control individuals with the same age and gender and no confounding factors after informed consent and completing the questionnaire including demographic information etc. were studied and underwent spirometry. Finally, the raw data were extracted and processed by SPSS software. In addition to the mean and standard deviation, t-test was used for comparing the means and at all stages the error probability less than 5% was deemed to be significant. Results: In the present study, there was a significant difference between the case group and the control group in pulmonary parameters. Also, there was no significant difference between the level of education, type of bakery, number of working hours per day and BMI of bakers. There was a significant relationship between work experience (year) and FVC, FEV1, FEV1% and FEF50%. There was also a significant relationship between the age of bakers and the all parameters above, except FVC, and there was a significant difference between different tasks in bakeries only in the case of FEF50%. Conclusion: In healthy people, pulmonary parameters decrease with age, but in bakers this process is more severe and needs to be followed
