    No full text
Background and purpose Inflammation is the body's defense mechanism against pathogens, which can lead to many diseases. Cyclooxygenase 2 enzyme is one of the main enzymes involved in the inflammatory process and its inhibition reduces and eliminates inflammation. In this study, To Achieve compounds with the potential for anti-inflammatory effects and restriction of pathological conditions connected to inflammation , chemical compounds with a structural scaffold similar to the curcumin compound, wich showed inhibitory effects on cyclooxygenase 2, were investigated. Materials and Methods In the present project, 2063 chemical compounds based on 70% structural similarity with curcumin obtained from the PubChem database and then they was saved. Then, the various steps and filters of the virtual screening process was performed by the servers of Molinspiration, SwissADME, admetSAR and PASS. Finally, the energy and interactions of the selected compounds were investigated using Autodock 4.2 software. Results Compounds CID_54676038, CID_337736, CID_628942 and CID_337738 with free binding energies of -10.49, -10.34, -9.65 and -9.40 kcal /mol, respectively showed the highest free binding energy among all selected compounds. These four compounds formed hydrogen and hydrophobic interactions with key amino acids of cyclooxygenase 2 enzyme active site. According to molecular docking results, amino acids Gln178, Phe504, Ser339, Ile503, Ala502, Val509, Trp373, Met508 and Ala513 may be important in maintaining the stability of compounds in cyclooxygenase 2 active site. Conclusion In recent study to identify and introduce new cyclooxygenase 2 inhibitors, it was observed that a number of important amino acids establish hydrogen and hydrophobic interactions with 4 selected compounds, curcumin and Celecoxib on cyclooxygenase 2 active site. Also, in studies to investigate the specific effects of these compounds on cyclooxygenase 2, it was observed that one of the main selected compounds has a greater affinity to form a complex and bind to cyclooxygenase 2 than cyclooxygenase 1

BACKGROUND AND OBJECTIVE: High infiltration of glioma cells into the surrounding brain tissue is the cause of treatment failure in glioblastoma. Glioblastoma interactions with endothelial cells increase the migration of cancer cells. Co- culture systems try’s to prepare a situation similar to physiology of body. The aim of this study was to investigate the effect of tranexamic acid (TXA) on reducing the migration of glioblastoma and endothelial cells and the levels of metalloproteinases-2 and -9 (MMP-2/9) in co-culture conditions. METHODS: In this experimental study, cells prepared from Pasteur Institute were treated with different concentrations of TXA in mono-culture and co-culture. Cell survival was determined by MTT assay. The rate of migration was assessed by making grooves in the culture dishes and comparing the groove area at different time intervals in the control and treated groups with 6 and 24 mM TXA. The levels of MMP-2/9 enzymes in the control group and cells treated with 6 mM TXA were assessed by zymography. FINDINGS: Decreased survival of T98G and HUVECs cells was observed in mono- and co-culture from 60 mM TXA and above. Groove area in HUVECs group treated with TXA was significant more than control group. No difference in groove size after 36 hours of threatment was observed in T98G cells. TXA significantly reduced the levels of MMP-2 and MMP-9 in co-culture conditions and the reduction level in MMP-2 is more than MMP9. CONCLUSION: The results showed that TXA could reduce the migration of glioblastoma and endothelial cells as well as the levels of MMP-2/9 in co-culture condition

    No full text
Introduction: Periodontal disease is a local inflammatory process that leads to alveolar bone resorption and if left untreated and the disease progresses, it can lead to the loss of a dental unit. Numerous studies have supported this hypothesis that; Inflammatory factors related to periodontal disease enter the bloodstream and lead to systemic inflammation as a risk factor for coronary heart disease. The aim of this study was to determine the extent and severity of periodontal disease in patients with obstructive CHD and to compare them in patients with one-vessel, two-vessel and three-vessel involved. Methods and Materials: The present study is a cross-sectional descriptive analytical study on 96 patients who underwent coronary angiography and obstructive coronary heart disease; Based on the angiographic result, patients were classified into three categories: one involved vessel, two involved vessels and three involved vessels. Then, periodontal indices Plaque Index, Bleeding On Probing, Periodontal Pocket Depth and Clinical Attachment Loss were measured in these three groups and together Were compared. Results: In this study, a comparison of periodontal indices between the three groups showed that only the Plaque Index increases with increasing number of vessels involved and this relationship is statistically significant(P<0.05), but the indices of Bleeding On probing, Periodontal Pocket Depth and Clinical Attachment Loss was not statistically significant between the three groups. Conclusion: According to the results of the present study and due to the strong relationship between poor oral hygiene and the degree of coronary artery occlusion; The need to improve oral care has become more apparent among patients with cardiovascular disease, and physicians and dentists should pay special attention to oral health care in these patient
