    RotoForm, Robotic Sequential Casting : Toward Material System and Material Programming in Architecture

    This dissertation provides a step toward enhancing technology-driven design-to-fabrication procedures in the architecture, engineering, and construction industries. More specifically, it contributes to sustainable manufacturing and innovative robotic fabrication methods in concrete construction. The architecture, engineering, and construction (AEC) industries are experiencing a growing need to adapt emerging technologies to increase productivity, promote sustainable construction methods, and integrate automation. Although AEC is part of the production sector, it lags far behind other manufacturing industries in technological innovation. Research on computational design and robotic fabrication has examined the ability to build with emergent technologies and data-driven procedures, showing significant progress not only in technological development but also in the knowledge of its use. However, the implementation of these advances and the transfer of knowledge to the industrial sector remain bottlenecks in this field. This need is especially high given the demand for sustainable construction, the scarcity of construction materials, and the AEC industry's significant contribution to carbon emissions. The effective use of technology in AEC requires research methods that, firstly, conceive the implementation of this technology as an integral research element, and secondly, develop integral models that cover all phases from design to construction. This dissertation considers knowledge transfer as a methodological challenge and subsequently explores technology implementation and automation in architecture and construction through a systematic investigation methodology. The methodology addresses the disciplinary issue of separation between design and fabrication, as well as the bottleneck of technology implementation in the AEC industries, given the heterogeneous landscape of fragmented enterprises in this field. More precisely, it focuses on precast concrete, robotic fabrication, sustainability, and automation. Since concrete is the most widely used material in the construction industry but also the most destructive, it calls for changes in how architects design, fabricate, and maintain buildings with this material. The methodology enables the systematic development of four interrelated case studies, which generate a heterogeneous palette of solutions within different setups and objectives. The case studies are developed and evaluated based on the criteria of design simulation, machinery and robotic fabrication setup, material development, and implementation framework. The first case study aims to define the boundaries of RotoForm, a robotic fabrication method for dynamically casting hollow concrete elements. RotoForm is a material system that synthesizes a dynamic concrete casting method, the phase-changing state of concrete, and a hyperelastic formwork system into an integral model. Following this, Case Studies Two, Three, and Four each address specific challenges in the field of precast concrete. These studies present innovative solutions through technical developments and design-to-fabrication frameworks, all centered around RotoForm. The second case study explores the implementation of RotoForm in collaboration with an industry partner. It targets the development of a robotic fabrication process for large-scale hollow concrete elements as lost formwork. The third case study describes an intersectoral collaboration with a spinoff company in the field of robotic fabrication. The study aims to increase the level of automation through a continuous robotic fabrication setup, from formwork shaping to reinforcement winding to sequential robotic dynamic casting. The fourth case study deals with the challenges of onsite assembly and the need for non-permanency, adaptation, and reconfiguration in architecture. It utilizes the ability of RotoForm for uneven material distribution inside the hollow concrete elements and proposes a material programming system for guided self-assembly. This dissertation presents fundamental studies on RotoForm, a novel robotic fabrication framework for the sequential dynamic casting of hollow concrete elements. The case studies offer a systematic approach for studying the implementation of this technology as an integral element of research on innovation in AEC. The work proposes a model that explores the relationships among all aspects of the design-to-fabrication process. The research is the result of interdisciplinary collaboration with other engineering fields for technical development and with industry partners for intersectoral implementation. The emphasis on integrated research has brought fundamental architectural design questions closer to the technical challenges of implementing new technologies, paving the way for solutions that can meet our current sustainability needs

    Ein Beitrag zur Instandhaltung von Brandschutzmaßnahmen in Immobilien

    Die vorliegende Dissertation befasst sich mit der Instandhaltung von Brandschutzmaßnahmen während des längsten Abschnitts im Lebenszyklus von Immobilien: der Nutzungsphase. Ausgangspunkt ist unter anderem die hohe Mängelquote bei wiederkehrenden Prüfungen von Brandschutzeinrichtungen. Diese Mängel gefährden nicht nur die Sicherheit von Personen, Sachwerten und der Umwelt, sondern können auch erhebliche rechtliche und finanzielle Konsequenzen für Immobilieneigentümer*innen nach sich ziehen. Vor diesem Hintergrund liefert die vorliegende Arbeit eine Analyse von Schlüsselprozessen der Instandhaltung von Brandschutzmaßnahmen und baut dabei auf den Grundlagen des baulichen Brandschutzes auf. Diese bestehen aus den grundlegenden Anforderungen an bauliche, technische und organisatorische Brandschutzmaßnahmen, der Verankerung des Brandschutzes im Lebenszyklus einer Immobilie sowie der Betrachtung der Instandhaltung. Im Zuge der Untersuchung wurden die Aspekte des Immobilienübergangs, verschiedene Soll- und Ist-Zustände sowie die Betreiberverantwortung erarbeitet. Darauf aufbauend erfolgte die Entwicklung eines Prozessmodells, das durch eine systematische Betrachtung, Strukturierung und Priorisierung der relevanten Abläufe aus der Perspektive von Immobilieneigentümer*innen die notwendigen Prozesse der Instandhaltung abbildet. Der Fokus liegt dabei auf der Einordnung in das polyzyklische Lebenszyklusmodell von Immobilien, wobei das Prozessmodell aus drei zentralen Ketten besteht: der Integrationsphase, der Instandhaltungsphase und der Mängelbeseitigung. Weiter ist die Dokumentation der Instandhaltung von zentraler Bedeutung innerhalb des Modells und für die Instandhaltung an sich. Anhand dieser erfolgt die Prüfung der Instandhaltung, sowohl intern als auch durch die Behörden. Ebenfalls findet über die Rückkopplung mit der Unternehmensstrategie die geplante Verwendung einer Immobilie durch den Eigentümer bzw. die Eigentümerin Eingang in die Instandhaltung. Insgesamt wird ein Beitrag zur Praxis des Immobilienmanagements geleistet, indem ein ganzheitlicher und methodisch fundierter Ansatz für die Instandhaltung von Brandschutzmaßnahmen entwickelt wird

    On the Benefit of FMG and EMG Sensor Fusion for Gesture Recognition Using Cross-Subject Validation

    Hand gestures are a natural form of human communication, making gesture recognition a sensible approach for intuitive human-computer interaction. Wearable sensors on the forearm can be used to detect the muscle contractions that generate these gestures, but classification approaches relying on a single measured modality lack accuracy and robustness. In this work, we analyze sensor fusion of force myography (FMG) and electromyography (EMG) for gesture recognition. We employ piezoelectric FMG sensors based on ferroelectrets and a commercial EMG system in a user study with 13 participants to measure 66 distinct hand movements with 10ms labelling precision. Three classification tasks, namely flexion and extension, single finger, and all finger movement classification, are performed using common handcrafted features as input to machine learning classifiers. Subsequently, the evaluation covers the effectiveness of the sensor fusion using correlation analysis, classification performance based on leave-one-subject-out-cross-validation and 5x2cv-t-tests, and its effects of involuntary movements on classification. We find that sensor fusion leads to significant improvement (42% higher average recognition accuracy) on all three tasks and that both sensor modalities contain complementary information. Furthermore, we confirm this finding using reduced FMG and EMG sensor sets. This study reinforces the results of prior research about the effectiveness of sensor fusion by performing meticulous statistical analyses, thereby paving the way for multi-sensor gesture recognition in assistance systems

    Prozessmodell für die bedarfsgerechte Bestandsentwicklung des Portfolios von Wohnungsbaugenossenschaften

    Wohnen stellt ein Grundbedürfnis sowie ein Grundrecht der Menschheit dar. In Deutschland werden von den rund 43,1 Mio. Wohnungen mehr als die Hälfte (53,49 % der Wohnungen) vermietet. Der Mietwohnungsmarkt in Deutschland ist daher von großer Bedeutung. Insbesondere in den urbanen Regionen in Deutschland ist derzeit ein Anstieg der Nachfrage nach Wohnraum erkennbar. Demzufolge wird es für einkommensschwache Haushalte und Familien immer schwerer bedarfsgerechten, also bezahlbaren Wohnraum mit einer bedarfsgerechten Nutzungsqualität insbesondere in den urbanen Regionen in Deutschland zu finden. Wohnungsbaugenossenschaften zählen zu den professionell-gewerblichen Anbietern und agieren als Not-for-Profit Unternehmen. Sie sind aufgrund ihrer Satzung bestrebt, ihren Mitgliedern bedarfsgerechten sowie bezahlbaren Wohnraum zur Verfügung zu stellen. Derzeit stehen die Wohnungsbaugenossenschaften vor der Herausforderung, ihr heterogenes Immobilienportfolio gemäß den Bedürfnissen der Mitglieder zu sichern und fortzuentwickeln. Denn im Rahmen der Bestandsentwicklung sind neben den technischen, wirtschaftlichen und ökologischen Anforderungen insbesondere die sozialen Ziele und Werte zu berücksichtigen. Im Rahmen der vorliegenden Dissertation wurde ein Prozessmodell zur bedarfsgerechten Bestandsentwicklung des Portfolios von Wohnungsbaugenossenschaften entwickelt. Das Prozessmodell besteht aus drei Teilprozessstrukturelementen: Nutzeranforderungen, Bestandsentwicklungsmanagement und Bestandsentwicklungskosten. Im Kontext der Nutzeranforderungen sind Experteninterviews und Mitgliederbefragungen durchzuführen und die Kano-Methode ist anzuwenden. Im Rahmen des Bestandsentwicklungsmanagement ist als strategisches Instrument das von der Verfasserin entwickelte genossenschaftliche Portfoliomodell anzuwenden. Gegenstand der Bestandsentwicklungskosten ist die Bestimmung einer wirtschaftlichen Strategie zur langfristigen Unterhaltung der Immobilien unter Anwendung eines mathematischen Modells. Dieses Modell basiert auf der Graphentheorie. Das entwickelte Prozessmodell unterstützt die Wohnungsbaugenossenschaften im multikriteriellen Entscheidungsprozess, eine transparente und nachvollziehbare Entscheidung über die Fortentwicklung des Portfolios festzulegen. Durch Anwendung des Prozessmodells wird sichergestellt, dass das Portfolio bedarfsgerecht, qualitätsvoll und betriebswirtschaftlich fortentwickelt wird

    Discovery and Characterization of Allosteric Antibodies

    Antibodies are the fastest-growing class of treatments, with a majority of them being approved in the oncology area. Proteins are inherently flexible, and this flexibility can be exploited to modulate ligand-binding and target activity, a phenomenon referred to as allostery. This doctoral thesis focuses on the discovery and characterization of allosteric antibodies for cancer immunotherapy, specifically targeting the epidermal growth factor receptor (EGFR). EGFR, a transmembrane protein belonging to the receptor tyrosine kinase family, can be dysregulated and overexpressed, leading to cancer proliferation. As a widely pursued target, four therapeutic antibodies are approved for the treatment of EGFR+ cancers. These antibodies all compete with the natural ligand EGF for binding. Here, alternative strategies were explored for the discovery of allosteric antibodies to leverage EGFR biology and develop the next generation of anti-EGFR antibodies. The first aim of this doctoral thesis was to develop a robust strategy for the direct identification of non-ligand-competitive antibody fragments from yeast surface display (YSD) libraries. Traditionally, the identification of allosteric binders is performed using in vitro assays or by structural studies. Conventional screening of YSD libraries typically employs fluorescence-activated cell sorting (FACS) in the presence of the target protein using a two-color staining strategy. By leveraging the precise control over several parameters of FACS, an additional criterion was introduced during the sorting: the labeled ligand. Two YSD libraries, one displaying fragment-antigen binding (Fab) fragments and the other variable heavy domain of heavy chain only antibodies (VHHs), previously enriched for EGFR-binders, were utilized to develop and optimize this approach. These libraries were incubated with various concentrations of either biotinylated EGF or EGF Fc fusion protein, followed by secondary detection using either streptavidin-fluorescence or anti Fc antibody fluorescence conjugates. These libraries were screened using FACS with a three-color gating strategy, to simultaneously detect functional display, EGFR binding, and EGF presence. Conditions were meticulously compared to ensure the identification of the most promising candidates. By comparing sequencing results with the traditional sorting method, a greater number of hit candidates were obtained, demonstrating that this platform can significantly streamline the discovery process for candidates with the desired properties. The second study focused on the discovery and characterization of allosteric antibodies that inhibit EGFR allosterically. To this end, an immune camelid yeast surface display library was generated and screened by FACS in the presence of EGF Fc fusion protein, based on the optimized sorting conditions established in the first study. After two rounds of sorting, eleven binders were identified and reformatted as Fc fusion proteins. Binding to four different allosteric sites was confirmed through competition assays via biolayer interferometry. The first aspect of allosteric modulation was examined using a fluorescence polarization assay to evaluate the impact of antibodies on the affinity of EGFR for EGF, revealing various modulations. The second aspect of allosteric modulations involved the inhibition of EGFR-mediated signaling pathways. The impact on the MAPK/ERK pathway was assessed using a cell line reporter, which demonstrated that all the antibodies inhibited this pathway, with inhibitory concentrations ranging from two-digit picomolar to two-digit nanomolar. Subsequent homogeneous time resolved fluorescence (HTRF) assays determined the impact of antibodies on the PI3K/AKT pathway, revealing that one binder was able to inhibit this critical pathway. This binder exhibited greater inhibitory properties for both pathways compared to the commercially approved EGF-competitive antibody cetuximab. This investigation demonstrated that EGFR can be allosterically inhibited by an antibody more effectively than by EGF-competitive antibodies. Additionally, as none of these antibodies bind to paralogue proteins, they suggest targeting allosteric sites of EGFR presents a better selective profile and may represent a potential new mechanism of action. Bispecific natural killer cell engagers (NKCEs) represent an alternative approach for tumor cell killing by bringing into close proximity NK cells with tumor cells. Previous studies demonstrated the successful engagement of NKp30 on NK cells with tumor cells expressing EGFR. These studies were conducted on tumor cells that did not express one of the natural ligands of NKp30: B7-H6, striking out natural ligand recognition. In the third study presented here, NKp30-based NKCEs were engineered to avoid the competition with B7-H6. First, an immune camelid yeast surface display library was generated. This library was incubated simultaneously with NKp30 and B7-H6 to obtain VHHs that would bind to allosteric sites of NKp30, based on the method established in the first section. Eight candidates were identified, and BLI assays confirmed binding to two different allosteric sites of NKp30. Intriguingly, antibodies that did not cross-react with the rat ortholog also exhibited poor affinity to the endogenous NKp30 on NK92 cells. Subsequently, rat cross-reactive antibodies were reformatted as DuoBody with either cetuximab (anti-EGFR moiety) or trastuzumab (anti-HER2 moiety). These NKCEs were tested for their killing capacities on HeLa cells (B7-H6+/EGFR+) or SK-BR-3 cells (B7-H6+/HER2+). These constructs demonstrated a significant enhancement of killing capacities correlated with B7-H6 expression. An enhanced-targeting approach was proposed for allosteric NKCEs, wherein the natural recognition of B7-H6 complements the induced close proximity facilitated by NKCEs. The last part of this work presents a literature review of targets modulated allosterically by antibodies. This review highlights the growing interest for antibodies in targeting allosteric sites, which opens up novel avenues for druggability. Additionally, allosteric antibodies have great potential as tools in the drug discovery process, such as structural studies or to facilitate high-throughput screening. Finally, we intend to illuminate a roadmap for their design using contemporary computational approaches

    Advancing Machine Learning Algorithms for Object Localization in Data-Limited Scenarios : Techniques for 6DoF Pose Estimation and 2D Localization with limited Data

    Recent successes of Machine Learning (ML) algorithms have profoundly influenced many fields, particularly Computer Vision (CV). One longstanding problem in CV is the task of determining the position and orientation of an object as depicted in an image in 3D space, relative to the recording camera sensor. Accurate pose estimation is essential for domains, such as robotics, augmented reality, autonomous driving, quality inspection in manufacturing, and many more. Current state-of-the-art pose estimation algorithms are dominated by Deep Learning-based approaches. However, adoption of these best in class algorithms to real-world tasks is often constrained by data limitations, such as not enough training data being available, existing data being of insufficient quality, data missing annotations, data having noisy annotations, or no directly suitable training data being available at all. This thesis presents contributions on both 6D object pose estimation, as well as on alleviating the restrictions of data limitations, for pose estimation, and for related CV problems such as classification, segmentation, and 2D object detection. It offers a range of solutions to enhance quality and efficiency of these tasks under different kinds of data limitations. The first contribution enhances a state-of-the-art pose estimation algorithm to predict a probability distribution of poses, instead of a single pose estimate. This approach allows to sample multiple, plausible poses for further refinement and outperforms the baseline algorithm even when sampling only the most likely pose. In our second contribution, we drastically improve runtime and reduce resource requirements to bring state-of-the-art pose estimation to low power edge devices, such as modern augmented and extended reality devices. Finally, we extend a pose estimator based on dense-feature prediction to incorporate additional views and illustrate its performance benefits in the stereo use case. The second set of two contributions focuses on data generation for ML-based CV tasks. High quality training data is a crucial component for best performance. We introduce a novel yet simple setup to record physical objects and generate all necessary annotations in a fully automated way. Evaluated on the 2D object detection use case, training on our data performs favourably with more complex data generation processes, such as real-world recordings and physically-based rendering. In a follow-up paper, we further improve upon the results by introducing a novel postprocessing step based on denoising diffusion probabilistic models (DDPM). At the intersection of 6D pose estimation and data generation methods, a final group of three contributions focuses on solving or circumventing the data problem with a range of different approaches. First, we demonstrate the use of physically-based, photorealistic, and non-photorealistic rendering to localize objects on Microsoft HoloLens 2, without needing any real-world images for training. Second, we extend a zero-shot pose estimation method by predicting geometric features, thereby improving estimation quality with almost no additional runtime. Third, we demonstrate pose estimation of objects with unseen appearances based on a 3D scene representation, allowing robust mesh-free pose estimation. In summary, this thesis advances the fields of 6D object pose estimation and alleviates some common data limitations for pose estimation and similar Machine Learning algorithms in Computer Vision problems, such as 2D detection and segmentation. The solutions proposed include several extensions to state-of-the-art 6D pose estimators and address the challenges of limited or poor quality training data, paving the way for more accurate, efficient, and accessible pose estimation technologies across various industries and fields

    Geometrisch-optische Strukturmodellierung zur Simulation von Pflanzenbelichtung

    Elektrische Energie für Belichtung ist ein wesentlicher Kostenpunkt beim geschützten Anbau von Pflanzen. Die Erhöhung der Effizienz der Lichtquellen ist limitiert, weswegen das Potential einer Optimierung der Abstrahlcharakteristik, der spektralen Zusammensetzung sowie des emittierten Photonenstroms von Pflanzenleuchten ausgeschöpft werden muss. Feldexperimente mit realen Pflanzen sind zeit- und kostenintensiv. Daher wird ein modellbasierter Ansatz (virtuelle Pflanzen) gewählt, um Erkenntnisse zur Auswirkung von Leuchteneigenschaften simulativ abzuleiten. Virtuelle Pflanzen bestehen aus einem geometrischen und einem optischen Modell, zu deren Erstellung Prozesse entwickelt werden. Im ersten Teil erfolgt eine Analyse der optischen Eigenschaften der Blätter von jungen Schlangengurken. Dabei zeigt sich, dass die Reflexionsgrade für die beiden Blattseiten separat gemessen und modelliert werden müssen. Auch wird ein Zusammenhang zwischen den spektralen Reflexionsgraden und dem Blattalter in manchen Spektralbereichen nachgewiesen. Die spektralen Transmissionsgrade hingegen weisen eine weniger ausgeprägte Abhängigkeit vom Blattalter auf als die spektralen Reflexionsgrade. Altersabhängige Effekte sind insbesondere in ihrer Auswirkung auf den spektralen Absorptionsgrad von Bedeutung, da dieser direkt die berechnete Assimilationsrate beeinflusst. Neben diesen Größen werden die richtungsabhängigen Reflexionseigenschaften spektral bestimmt. Diese weisen spektrale Abhängigkeiten auf. Die Reflexion ist im grünen und fernroten Spektralbereich diffus, wohingegen sie im blauen und roten Spektralbereich gestreut ist. Ein Modell zur Modellierung dieser Eigenschaften wurde angewandt, um sie in Lichtsimulationen berücksichtigen zu können. Insbesondere im Hinblick auf die Modellierung von richtungsabhängigen Reflexionseigenschaften ist bisweilen unklar, inwieweit diese das Ergebnis beeinflussen. Die Einflussanalyse zeigt, dass der Einfluss auf die absorbierte Photonenstromdichte gering, auf die spektrale Photonenstromdichte in der Blattumgebung jedoch von Bedeutung ist. Eine Optimierung der Leuchteneigenschaften erfolgt unter Verwendung der erstellten virtuellen Pflanzen. Dabei wird gezeigt, dass mit virtuellen Pflanzen Leuchteneigenschaften bestimmt werden können, die den Wirkungsgrad der Belichtung erhöhen. Berücksichtigt werden auch Vergleiche mit Sonnenlicht, sodass unter Zuhilfenahme von ausgewählten Metriken eine negative Beeinflussung der Pflanzenentwicklung durch eine ungünstige Wahl der Leuchteneigenschaften verhindert wird. Einen großen Einfluss auf den Belichtungswirkungsgrad hat die Abstrahlcharakteristik (LVK) der Leuchte. Ebenfalls kann eine Erhöhung des Reflexionsgrades des Bodens die Assimilationsrate in den niedrigen Blatträngen erhöhen. In der spektralen Optimierung kann ein Spektrum ermittelt werden, das bei einem Abgleich mit dem Stand der Forschung nicht nur effizient im Hinblick auf den Belichtungswirkungsgrad, sondern auch im Hinblick auf die Effizienz der Lichtquelle ist

    Synthese und Evaluation neuer biogener Stabilisatoren für Polypropylen

    Angesichts der wachsenden Bedeutung nachhaltiger Materialien und der begrenzten Verfügbarkeit fossiler Ressourcen untersucht diese Arbeit die Entwicklung und Evaluierung biobasierter Stabilisatoren für Polypropylen. Der Schwerpunkt lag auf der Synthese neuartiger, biotechnologisch zugänglicher Phenolsäure-Derivate sowie der Analyse ihrer Struktur-Eigenschafts-Beziehungen hinsichtlich Verarbeitungs- und Langzeitstabilisierung. Durch systematische Modifikation von 4-Hydroxybenzoesäure, 4-Hydroxyzimtsäure und 3-(4-Hydroxyphenyl)propionsäure wurden verschiedene Ester, ein Amid und ein Imin synthetisiert und hinsichtlich ihrer thermischen Stabilität sowie antioxidativen Wirkung charakterisiert. Die Ergebnisse zeigen, dass insbesondere hydroxylierte Cinnamate und Phenylpropionate eine hohe Effizienz in der Verarbeitungsstabilisierung von Polypropylen aufweisen. DFT-Berechnungen lieferten dabei grundlegende Erkenntnisse über die zugrunde liegenden Stabilisierungseffekte, wobei sich eine Rangfolge der Stabilisatoreffizienz ergab: Phenylpropionate > Cinnamate > Benzoate. Zudem wurde ein weitgehend biobasiertes Phosphit als sekundäres Antioxidans untersucht. Die Kombination dieses Phosphits mit biobasierten Phenolen zeigte eine synergistische Wirkung, die insbesondere die Langzeitwärmestabilisierung erheblich verbesserte und industriellen Benchmarks mit tert-Butylsubstituenten sogar überlegen war. Die gewonnenen Erkenntnisse tragen zur Entwicklung nachhaltigerer polymerer Materialien bei und können zukünftig insbesondere für Anwendungen in Lebensmittelverpackungen und Kreislaufwirtschaftskonzepten weiter erforscht werden. Die Kombination biobasierter Antioxidantien mit Phosphiten stellt einen vielversprechenden Ansatz zur Optimierung der Stabilisierungsleistung dar

    Among all mammalian cellular organelles, the nucleus gets special attention as it contains the genetic information of the cell, the DNA. This is also the site where all DNA-dependent functions, such as genome duplication prior to cell division, DNA transcription, and damage repair upon various DNA breakages undergo. The nucleus also contains the subcompartment, an assembly site for the ribosome, the nucleolus. Among all the DNA-templated events, genome duplication is the most eventful, involving a large array of proteins simultaneously unwinding and synthesizing a new pair of DNA at thousands of locations of the same long polymer, yet tightly regulated in time and space. It is an event on a scale hitherto undreamt of. While the faithful reconstitution of cellular identity involves the accurate inheritance of epigenetics, the information that sits atop the genetic information, the dynamic nature of epigenetics also influences the chromatin structure and regulates various aspects of DNA-templated events, including genome replication programs. The progression of development also triggers changes in the epigenetic landscape, thereby influencing the replication program. During genome replication, the chromatin decondenses, the DNA unwinds, and new DNA is synthesized and rewrapped around the histone core. The spatio-temporal regulation of the replication program also ensures the faithful inheritance of the epigenetic nature, specifically the histone post-translational modifications. However, the intricate interplay between epigenetics and the genome replication program remains a mystery, awaiting further exploration. The present work first sheds light on the developmental changes in genome replication programs, features of replication forks, and underlying chromatin dynamics in human cells. It also uncovered the change in replication time of less-studied tandem and interspersed repeats, which constitute a large chunk of the genome, using repli-FISH. As a prerequisite, a novel approach to analyze the repli-FISH was developed. A switch in the replication timing of ribosomal DNA and differential replication program of Alu, LINE1, and Centromere were observed. Interactions between genome replication machinery and RNA polymerase I responsible for rDNA transcription were observed in pluripotent stem cells. Overall, the study also complements and expands our understanding of the developmentally regulated genome replication program in human cells. High-throughput image analysis was used to quantify the histone modification marks in different cell cycle phases to characterize the relationship between genome replication and epigenetics. Furthermore, the statistical tool nucim was used to map the dynamic localization of histone marks to different compaction classes with cell cycle and sub-S phase progression in pluripotent stem cells. Dynamic localization in constitutive heterochromatin and its replication timing regulation by H3K36me3 was unveiled. Furthermore, genome-wide dynamics of H3K27me3 and H3K4me3, which also mark the poised/bivalent chromatin in embryonic stem cells, unveiled a peculiar pattern. Using synthetic biology, the replication program was disrupted by localizing the constitutive heterochromatin next to nuclear lamin, and its effect on epigenetics was studied. Furthermore, the earliest replication origins and their (epi)genetic/chromatin characteristics are explored to validate the Domino model of replication progression. The approach to finding the cells with the earliest origins is developed in living and fixed cells was established for a detailed exploration. The ribosomal DNA tandem repeat was discovered as one of the preferred locations to start the replication across mammalian cell lines/species. To investigate the genetic nature of these origins, methods to identify, collect, and amplify single-cell genomes linearly were established to measure the micro-copy number gain.     Strömungssimulation im Dienste der Mobilität: Fortschritte bei CFD-Anwendungen in der Automobilaerodynamik-Entwicklung

    Die vorliegende Arbeit fasst die Forschungstätigkeiten des Autors in den Jahren 2009 bis 2023 aus dem Feld der numerischen Strömungssimulation (CFD-Simulation) in der Fahrzeugaerodynamik zusammen. Dabei wird insbesondere gezeigt, dass CFD sich auf allen gezeigten Feldern als verlässliches und zum Windkanal konkurrenzfähiges Analyse- und Prognosewerkzeug in der Fahrzeugentwicklung etabliert hat. Alle gezeigten Ergebnisse stammen aus Arbeiten des Autors im Rahmen seiner Tätigkeit bei den beiden Pkw-Herstellern AUDI AG und BMW Group, aus Kooperationsprojekten mit der TU München und der Universität Stuttgart sowie aus Forschungsvorhaben und Studentenprojekten im Rahmen seiner Lehrtätigkeit an der TU Darmstadt. Sie wurden bereits in diversen Zeitschriftenartikeln sowie Tagungs- und Buchbeiträgen veröffentlicht. Zunächst werden die notwendigen Grundlagen der Fahrzeugaerodynamik kontextbezogen beschrieben und die zu Grunde liegenden numerischen Methoden zusammengefasst. Die Schwerpunkte der Forschung liegen im Anschluss auf der Beschreibung des Strömungsfelds und der aerodynamischen Kräfte bei realistischer Straßenfahrt, aber auch bei Kurvenfahrt oder Überhol- und Windschattenmanövern. Außerdem werden Strömungsprobleme mit Wärmeübergang, Untersuchungen zur aktiven Strömungsbeeinflussung und zur Cabriolet-Zugfreihaltung, Ansätze zur automatischen Geometrieoptimierung, Simulation von Zweiphasenströmungsproblemen bei Regenfahrt und die Verwendung der Methoden im Rahmen der Produktentwicklung behandelt
