Six-Year Follow-Up of an Abstinence-Based, Food Addiction Recovery Approach to Weight Management
Introduction: Research shows that the average person has a slightly addictive relationship with food, manifesting two or more symptoms of food addiction, and more than one in eight people have clinically diagnosable food addiction. Meanwhile, obesity and food addiction share some neurological mechanisms and are correlated in the general population. Could an abstinence-based approach to food addiction recovery be a viable long-term weight loss or weight maintenance strategy? Methods: The current study presents six-year retrospective follow-up data from a cohort of participants who started an abstinence-based food addiction recovery program for weight loss in October of 2017. Survey responses from 267 participants were analyzed and compared to baseline self-reported data from six years prior. Results: At six years, 71.8% of participants were maintaining greater than 5% weight loss. There was a statistically significant association between sustained weight loss and both current program membership (p\u3c0.001) and degree of adherence to the abstinence-based food plan (p\u3c0.001). Adherence was associated with weight loss outcomes in a dose-response manner. The average sustained weight loss for current members who followed the program was 13.9%. Discussion: In spite of the methodological challenges with this type of study, the results do suggest the long-term efficacy of a food addiction recovery approach to weight loss. They also help validate the notion that food addiction may be a significant contributor to the multi-factorial etiology of obesity and indicate the need for further research into the viability of abstinence-based food plans as tools for weight management
A Novel Splicing Graph Allows a Direct Comparison Between Exon-Based and Splice Junction-Based Approaches to Alternative Splicing Detection
There are primarily two computational approaches to alternative splicing (AS) detection using short reads: splice junction-based and exon-based approaches. Despite their shared goal of addressing the same biological problem, these approaches have not been reconciled before. We devised a novel graph structure and algorithm aimed at mapping between the exonic parts and splicing events detected by the two different methods. Through simulations, we demonstrated disparities in sensitivity and specificity between splice junction-based and exon-based methods. When applied to empirical data, there were large discrepancies in the results, suggesting that the methods are complementary. With the discrepancies localized to individual events and exonic parts, we were able to gain insights into the strengths and weaknesses inherent in each approach. Finally, we integrated the results to generate a comprehensive list of both common and unique AS events detected by both methodologies

Repurposing of a Gill Gene Regulatory Program for Outer-Ear Evolution
How new structures emerge during evolution has long fascinated biologists. An example is how the diminutive bones of the mammalian middle ear arose from ancestral fish jawbones1. By contrast, the evolutionary origin of the outer ear, another mammalian innovation, remains a mystery, partly because it is supported by non-mineralized elastic cartilage, which is rarely recovered in fossils. Whether the outer ear arose de novo or through the reuse of ancestral developmental programs has remained unknown. Here we show that the outer ear shares gene regulatory programs with the gills of fishes and amphibians for both its initial outgrowth and the later development of the elastic cartilage. Comparative single-nucleus multiomics of the human outer ear and zebrafish gills reveals conserved gene expression and putative enhancers enriched for common transcription factor binding motifs. This is reflected by the transgenic activity of human outer-ear enhancers in gills, and of fish gill enhancers in the outer ear. Furthermore, single-cell multiomics of the cartilaginous book gills of horseshoe crabs reveals a developmental program shared with the distal-less homeobox (DLX)-mediated gill program of vertebrates, with a book-gill distal-less enhancer driving expression in zebrafish gills. DOGMA-Seq and Multimodal, Single-Cell Analysis in Acute Myeloid Leukemia
Acute myeloid leukemia (AML) is a complex cancer, yet advances in recent years from integrated genomics methods have helped improve diagnosis, treatment, and means of patient stratification. A recent example of a powerful, multimodal method is DOGMA-seq, which can measure chromatin accessibility, gene expression, and cell-surface protein levels from the same individual cell simultaneously. Previous bimodal single-cell techniques, such as CITE-seq (Cellular indexing of transcriptomes and epitopes), have only permitted the transcriptome and cell-surface protein expression measurement. DOGMA-seq, however, builds on this foundation and has implications for examining epigenomic, transcriptomic, and proteomic interactions between various cell types. This technique has the potential to be particularly useful in the study of cancers such as AML. This is because the cellular mechanisms that drive AML are rather heterogeneous and require a more complete understanding of the interplay between the genetic mutations, disruptions in RNA transcription and translation, and surface protein expression that cause these cancers to develop and evolve. This technique will hopefully contribute to a more clear and complete understanding of the growth and progression of complex cancers
American Lung Association Pulmonary Hypertension Roundtable: Executive Summary
The American Lung Association and Pulmonary Hypertension Association convened a scientific roundtable of pulmonary hypertension experts to discuss the latest recommendations from the European Guidelines for the Diagnosis and Treatment of Pulmonary Hypertension (PH) and from the 7th World Symposium on Pulmonary Hypertension (WSPH). The aim of the roundtable was to discuss changes that were made compared to earlier recommendations and guidelines set out by the European Society of Cardiology and the European Respiratory Society in 2015, the 6th World Symposium on Pulmonary Hypertension in 2018, and the CHEST Guideline on Therapy for Pulmonary Arterial Hypertension in 2019. The overall objectives were to: 1) Create an educational resource for providers that summarizes currently available PAH guidelines, 2) Provide an expert critique of current guidelines, outlining strengths and weaknesses and resolving differences where guidelines do not agree. 3) Provide guidance for the incorporation of the recently approved drug, sotatercept into current guidelines. An executive summary was drafted following the roundtable meeting on April 8, 2024, and revised by the panel in September 2024, following publication of the proceedings from the 7th WSPH held in Barcelona June 29-July 1, 2024. The Executive Summary reviews changes to the hemodynamic criteria for defining pre- and post-capillary PH, exercise-induced PH, and PH associated with lung disease. Recommendations are given for proper diagnosis and clinical classifications of the various forms of PH. The role of screening for PH in high-risk populations and the use of risk scores for disease stratification are discussed. Finally, treatment algorithms for managing pulmonary arterial hypertension, PH associated with lung disease, and chronic thromboembolic pulmonary hypertension are presented
