    Addressing Non-Point Source Agricultural Pollution in the Chesapeake Bay Watershed through Legal Policies

    Close to one quarter of land in the Chesapeake Bay Watershed is devoted to cropland. The primary pollutants that come out of farming are nutrient from fertilizers and animal waste (nitrogen and phosphorus) and sediment from the loose soil exposed from farming. To combat this, legislation has been put in place on both the federal and state level to reduce the level of pollutants in the waterways. The primary piece of legislation on the federal level is the Clean Water Act(CWA) which covers a wide array of topics all dealing with water pollution. The state level (Pennsylvania) involves both the Clean Streams Law, Non-Point Source Management Program (2008) and Best Management Practices(BMP). The main purpose of this project is to address the current tools in place to help improve the quality of the Bay given the current political situation and the pressure to dismantle the Environmental Protection Agency (EPA)

    Vortex Tube

    This discussion will be explaining the Hilsch-Ranque Vortex Tube and show experimental data about the model that was built for this experiment. In 1928, George Ranque invented the device accidentally while he was working on a vortex pump. It was later discovered that the vortex tube could be used on train cars to keep water warm and cold for passengers. The tube itself uses the compressed air along with the shape of the tube to create a vortex of air in which the air on the outside of the vortex is subjected to much more friction, making it much warmer. This air is then funneled out on side of the device. The air in the middle of the vortex gives off heat to the outer air, making the cold air come out the other side of the device

    Analyzing Susquehanna University’s Utility and Greenhouse Gas Data via EPA Webtools

    This project is dedicated to updating the collection methods and processing of utility information for all building Susquehanna University owns by use of Environmental Protection Agency (EPA) certified webtools. Utilities tracked for this project include Electricity, Natural Gas, Fuel Oil, Water, Coal, and more tracked all the way back to 2013 by using the platform called Portfolio Manager. The total greenhouse gas emissions from the university was also determined for the 2016 year, categorized by three different scopes. Buildings needing large improvements to infrastructure and energy use were highlighted by project and given suggestions for improvement. All data was collected by records kept by Facilities Management. Preliminary results for project have shown a total CO2 emissions of all scopes is over 10,000 Metric tons per year from the university. Also depicted by the utility information is the massive drop in energy use from Susquehanna switching from coal burning to natural gas

    Comparison of Computational Methods to Determine the Crystal Structure of Elm1p

    Elm1p, a serine/threonine protein kinase, is required for coordination of cell growth and nuclear division in budding yeast. The primary sequence of Elm1p is only 30 to 40% identical to sequences of kinases with solved crystal structures. Thus, determining the crystal structure of Elm1p will expand current knowledge of kinase structures as well as help elucidate how this enzyme functions to coordinate cell growth and nuclear division. Computational methods were used to accomplish this goal. Many tools have been developed to predict three dimensional protein structure based on the primary sequence such as ab initio modeling (Robetta), homology modeling (Phyre2 original), and threading (RaptorX). The primary sequence of Elm1p was submitted to several servers using these methods and the resulting predicted structures were compared. The results indicate that homology modeling and threading produced the most complete or fully folded predicted crystal structure

    Transfer of Latent Inhibition Memory Cues to a New Context: A Case of Erase and Update?

    Previous research has demonstrated that contextual cues for latent inhibition can be transferred from one context to another by mere exposure while the memory is still in an active state. Using a latent inhibition paradigm, the present experiment was conducted to determine what would happen when the subject was tested back in the original context, i.e., would the memory for latent inhibition transfer or would the memory also remain in the original context? Using Long-Evans rats, we evaluated the transfer of latent inhibition and whether the memory remained in the original context following the transfer. Results revealed that the transfer of latent inhibition cues was obtained, as well as that the memory for the original context remained. This finding indicates that when tested back in the original context, the subjects treated the two contexts similarly

    Response of Benthic Macroinvertebrate Communities to Variations in Discharge during the Fall Months of 2015 and 2016 in the Upper Main Stem of the Susquehanna River

    In this eighth year of a long-term study of benthic macro invertebrate communities (BMI) in the upper main stem of the Susquehanna River, we use BMI communities to assess the river health to reflect both water quality and habitat variation. Flow regimes of 2015 during summer showed 317% increase from 79 year average (July 2015, using the Sunbury USGS gage). Bray-Curtis showed 47% similarity during summer 2015, but fell to 38% during the same period in 2016. Low flow had the greatest negative impact on the individuals of he %EPT, with the exception of Heptageniidae. The Hilsenhoff Biotic Index (HBI) score rose at sites 1 and 2 from 4.024 and 3.188 in 2015 to 5.615 and 3.746 in 2016. Shannon Diversity Index (SDI) increased from 2015 (1.59 and 1.03) to 2016 (1.78 and 1.41). These results underscore the importance of multiyear sampling in assessments of environments as variable as large rivers

    Polyphenolic compounds from Hypericum punctatum leaves

    Hypericum perforatum (St. John’s wort), the most well-known member of the Hypericum genus, produces a variety of natural products including flavonoids, naphthodianthrones and acylphloroglucinol derivatives. These constituents have medicinal benefits such as antibacterial, anticancer, and anti-HIV activities. There are 19 Hypericum species in Pennsylvania and two occur in the Hypericumtaxonomic section: H. perforatum and H. punctatum. A HPLC analysis of extracts of H. perforatum and H. punctatum revealed similarities in chemical composition. This study is focused on the phytochemical examination of an acetone extract of the flowers of H. punctatum, with the aim of comparing its chemical composition to that of H. perforatum. Purification resulted in the isolation of two acylphloroglucinol derivatives, and several flavonoids. The structures of the flavonoids and acylphloroglucinol derivatives were elucidated using 1H-, 13C-, DEPT- and 2D-nuclear magnetic resonance (NMR) spectroscopic analyses

    Comparison of Pointed RNAi Knockdown Phenotypes and the Quantification of Resulting mRNA Levels

    Pointed is an ETS family transcriptional activator that functions downstream of the EGFR RTK signaling pathway and is essential in the development and differentiation of several cell types. Previous research has shown that Pnt is needed for the differentiation of cell types within the eyes, somatic follicle cells, and heart cells of Drosophila melanogaster. We used three RNAi lines: Valium 20, Valium 10, and VDRC, to knockdown pnt mRNA expression and examined the phenotypic effects. Val20 is distinct since it generates a short hairpin RNA. Valium 10 and VDRC on the other hand produce long hairpin loop dsRNA. We found that Val20 produced the strongest phenotypes while Val10 and VDRC produced varying levels of severity. Based on these results the goal of our study is to determine whether the severity of phenotypes produced by the three RNAi lines correlates with the extent to which the pnt mRNA level is depleted
