On Combinatorial Properties of Subsequences
A word, also known as string, is a sequence of letters, while a subsequence of a word w is another word u which can be obtained by deleting some of the letters of w.
Words in general, and subsequences in particular, play an important in role in many fields of theoretical and applied computer science, being the primary way to represent information: files are strings of bits, while DNA is represented as sequences of nucleotides, that is, letters A,C,G and T. Subsequences can be seen as lossy-representations of a word: they are natural mathematical model for situations where one has to deal with strings with missing symbols, such as it is quite often the case when processing DNA data, when dealing with digital signals communicated through lossy channels, or when dealing with corrupted data files.
Natural problems when dealing with subsequences are either related to matching (deciding whether a longer word contains a shorter one as subsequence) and analysis (e.g., identifying the absent subsequences of a given length from a word, or checking whether two words have the same set of subsequences).
In this thesis, in Chapters 3 and 4, we first focus on the set of strings which appear as subsequences in given words; and on the set of strings which do not occur in a given word as subsequences, that is, are absent. We provide efficient algorithms to determine whether two words have exactly the same set of subsequences (up to a predefined length k) and compact data structures to store shortest and minimal subsequences not occurring in a word.
Further, in Chapter 5, we extend the classic setting to subsequences of formal languages, e.g., regular languages given by a finite automaton. We investigate the lower and upper bounds for the universality index of words occurring in a language and provide a polynomial time algorithm computing the former and, as well, show NP-completeness for the latter (in case this upper bound is finite).
In a different line of research, in Chapter 6, we impose constraints on the factors of w bounded by the first and last letter of a subsequence u and, respectively, on the factors occurring between two consecutive letters of u. in this setting, we present optimal algorithms to determine whether u occurs as subsequence or, respectively, whether it is a minimal absent subsequence. Furthermore, we show NP-completeness for the problem to determine whether all words of length k occur as subsequence or to determine whether a given string u is a shortest absent subsequence, which is in stark constrast to the case of unconstrained subsequences presented in the previous chapters.
Likewise, in Chapter 7, we investigate the longest common subsequence problem (LCS) in the framework of Chapter 6. Further, we impose length constraints, called gap constraints, on the factors occurring between the embedding of the letters of the subsequences in the the two input words. We define several natural possibilities to define a sequence of gap constraints and investigate the computation of the LCS for each of these possibilities.
In Chapter 8, we introduce Soliton Automata and give combinatorial insights leading to sufficient conditions for a Soliton Automaton to be strongly connected and, respectively, complete, answering, in this way, an open question from the literature.2024-08-2
Effects of overexpression of the ALS associated protein TDP-43 and its mutant on the axonal and somal proteome in compartimentalized primary cortical neurons of rats
Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disease of the motor nervous system. During its course, the function of the upper and lower motor neurons is predominantly lost. It is a very severe disease with a life-time risk of 2.6 in 1000 and a rapid disease progression, that results in death in median within four years. Symptoms of the disease include a diverse picture such muscle weakness, atrophy, fasciculation, hyperreflexia, muscle spasms and more. Most ALS cases are of sporadic nature, however about 10% of the cases can be traced back to distinct gene mutations e.g., in the TAR DNA binding protein (TDP-43). No distinct pathological mechanism has been found thus far, that explains the very diverse picture of ALS. However, many pathological mechanisms are in question. Because of the uncertainty regarding ALSs origin, therapeutic options are very limited, with only a few symptomatic therapies available.
TDP-43 is a heterogenous nuclear ribonucleoprotein, that is very diverse in its functions. In health, it is predominantly located in the nucleus and supports the cell in various ways. It is essential for RNA related pathways such as transcription, translation, transportation, and splicing. Furthermore, it takes part in nucleocytoplasmic shuttling, the formation of stress granules and transportation of this granules. In ALS, TDP-43 is shifted towards the cytosol and forms protein inclusion bodies, that can be found in 97% of all ALS cases including sporadic and familial ALS. Other pathological TDP-43 related pathways include a disrupted nucleocytoplasmic shuttling, RNA alterations, mitochondrial dysfunctions, and degeneration of axons. The latter is highly relevant because axonal degeneration is an early indicator of the disease. TDP-43 can be dysregulated in its wildtype form and in mutations such as M337V, that leads to an even higher number of aggregates in the cytoplasm and increased velocity.
This study aims to give a comprehensive analysis of the somal and axonal proteome in wildtype and mutant TDP-43 overexpressing cortical neurons of embryonic rats in comparison to a control. The results show changes and similarities between the wildtype and mutant TDP-43 and differences between the axonal and the somal compartment. They confirm that there are in fact differences between wildtype and mutant TDP-43 overexpression, especially in the axonal compartment and show effects that the overexpression of TDP-43 has on different aspects of cellular homeostasis. For the proteomic analysis, cortical neurons of embryonic rats were cultivated in microfluidic chambers, that made a distinct separation of the proteome of cell bodies and the axons possible. Afterwards they were transduced with viral adeno-associated vectors that overexpressed wildtype and mutant TDP-43 and only EGFP as control. TDP-43 overexpression has been confirmed through immunocytochemistry, Western Blot and proteomic data. Bioinformatical analysis has been performed using the bioinformatical software DAVID, KEGG and STRING, that make visualization of the highly complex datasets possible.
In the comparison of the somal and axonal proteome for wildtype and mutant TDP-43 overexpressing cells, mostly proteins were altered regarding lysosome and synapse related pathways. Comparing wildtype and mutant in the somal compartment revealed most statistical relevant dysregulated proteins behave in a similar way, therefore being equally higher or lower abundant. The higher abundant proteins in the somal compartment for wildtype and mutant were predominantly connectable to Isopeptide bonds, messenger RNA metabolism and fatty acid metabolism. Especially mRNA metabolism and fatty acid metabolism are subjects closely linked to ALS pathology. The lower abundant proteins in the somal compartment in wildtype and mutant were linked to synaptic alterations. Comparing wildtype and mutant in the axonal compartment highlighted the differences between those two TDP-43 altering groups. Here, the majority of significantly altered proteins were higher abundant in the wildtype and lower abundant in the mutant. They were mostly related to axon guidance, synapse and microtubules. Similar behaving higher abundant proteins for wildtype and mutant in the axon were connected to isopeptide bond and endocytosis. Lower abundant proteins in the axon regarding wildtype and mutant pointed to the respiratory chain in the mitochondria, where proteins related multiple complexes were lower abundant. Throughout all analyzed groups hints towards cytoskeletal impairment were found, especially regarding actin, intermediate filaments, and microtubules. This study confirms previous findings of several different aspects of TDP-43 pathology and ALS. However, it also identifies more distinct research targets like the fatty acid metabolism and SUMOylation.2024-09-2
Retrospektiver und prospektiver Vergleich der klinischen Performance des CertasPlus- mit dem proGAV 2.0-Shuntventil zur Behandlung von Hydrocephalus-Patientenf
Implantation of a vp-shunt represents the current standard of care in hydrocephalus patients. Both combination valves implanted in this study contain a proprietary differential pressure valve, which is combined with a gravity-regulated anti-siphon device (ASD) in the proGAV valve and with a flow-controlled ASD in the CertasPlus valve. We sought to investigate whether the risk and performance profile differs in the two combination valves. Methods: Assignment of patients to valve type was block randomized in this prospective observational study. The sample consisted of 79 adult patients with hydrocephalus who received a vp-shunt for the first time. The two largest diagnostic subgroups (66 patients in total) were analyzed, namely iNPH (44%, n=35) and secondary communicating hydrocephalus (39%, n=31). Analyzed frontal occipital horn ratio (FOHR) and Evans index were CT derived. FOHR, Evans index and Stein-Langfitt (SL) scores were assessed prior to valve implantation and at follow-up. Black grading scale was assessed at follow-up only. Results: In this intermediate analysis, patients with iNPH compared to patients with secondary communicating hydrocephalus were significantly younger (75 ± 6 years vs. 58 ± 15 years, p<0.01). In patients with secondary communicating hydrocephalus , FOHR and Evans index tended to improve due to valve-treatment (p= 0.10 and p= 0.24) while SL scores improved significantly (p=0.015). No difference between valve types was found, neither on level (p≥ 0.29) nor on time-course (p≥ 0.37). In patients with iNPH, FOHR and Evans score changed significantly due to valve treatment (p≤ 0.01 and show a tendency for improvement of Stein-Langfitt (p= 0.067). Of note, in patients with iNPH, FOHR, Evans, and SL scores prior to valve-implantation had been significantly lower in CP compared to PG (p = 0.0021 multivariate joint test). Postoperative change in Evans (p=0.95) and FOHR did not differ significantly (p=0.39) and no differences were observed in median SL nor Black scale between valves (p=0.64). There were 2 revisions in the PG group (1 wound dehiscence, 1 cranial catheter occlusion) and none in the CP group. No difference in number of valve adjustments between CP and PG (p=0.24) was observed in both hydrocephalus subtypes. Conclusion: VP-shunt insertion for treatment of hydrocephalus can be performed safely and effectively with both valve types. No difference in outcome was noted between both valve types2024-11-0

An examination procedure for the detection of breast cancer (“MARIA”) is presented, which has been in the testing and optimization phase for years. In an intra-individual comparative study, 109 consecutive women who were scheduled to undergo breast MRI for medical reasons were examined in advance using the MARIA system between July and December 2020.
A major reason for the unsatisfactory results of the MARIA system is obviously that the procedure is based on the different impedance between the carcinoma and the surrounding tissue, assuming fat as the surrounding tissue and a high water content of malignant tumors. These conditions are obviously not fulfilled due to the high variability of both the carcinoma in the tissue and the healthy surrounding tissue.2024-11-0
Growth and Silviculture of Northern Red Oak (Quercus rubra L.) in Northwest Germany
Die vorliegende Arbeit zur nordamerikanischen Roteiche (Quercus rubra L.) widmet sich 70 Jahre nach der Veröffentlichung erster Monografien für Westdeutschland (Bauer 1953a) und Ostdeutschland (Göhre u. Wagenknecht 1955) erneut dem Wachstum, der waldbaulichen Behandlung, der Verjüngungsökologie und darüber hinaus Aspekten der ökologischen Integration dieser vor 250 Jahren nach Deutschland eingeführten und im forstlichen Anbau bewährten Baumart.
Im Mittelpunkt des waldwachstumskundlichen Teils stehen neu erstellte bzw. neu parametrisierte Modelle des Einzelbaum- und Bestandeswachstums der Roteiche für Nordwestdeutschland. Damit werden angesichts einer aktuell zunehmenden Berücksichtigung der Roteiche in waldbaulichen Planungen und in der praktischen Walderneuerung aufgrund der von ihr erwarteten Anpassungsfähigkeit an projizierte Klimaänderungen wichtige Planungs- und Bewirtschaftungsgrundlagen auf dem neuesten Stand zur Verfügung gestellt. Die Datengrundlage der Modellparametrisierungen bilden Einzelbaum- und Bestandesdaten langer Zeitreihen von 19 ertragskundlichen Versuchen in Rheinland-Pfalz, Nordrhein-Westfalen, Hessen, Niedersachsen und Schleswig-Holstein sowie Datenergänzungen, die das repräsentierte Standortspektrum und den abgedeckten Altersbereich gezielt erweitern. Neben zahlreichen einmaligen Probeflächenaufnahmen, v. a. in jüngeren Beständen, handelt es sich um die Neuanlage einer 12 Versuche umfassenden Versuchsserie im bisher unterrepräsentierten niedersächsischen Tiefland und einen Roteichen-Durchforstungsversuch mit abgestuften Eingriffsstärken in Bezug auf den natürlichen Bestockungsgrad einer Nullfläche (Assmann 1956). In den neueren und einem Teil der alten Versuchsanlagen wurden neben BHD-Vollaufnahmen und repräsentativen Höhen- und Kronenansatzmessungen zusätzlich Stammfußkoordinaten und Kronenprojektionen erhoben. Die Standortverhältnisse umfassen eine weite Spanne von sommertrockenen bis hin zu betont frischen bzw. grundwasserbeeinflussten Verhältnissen des Bodenwasserhaushaltes und schließen hinsichtlich der Nährstoffversorgung arme bis reiche Böden ein. Die klimatischen Bedingungen reichen von subatlantischen bis in subkontinentale Bereiche einschließlich trocken-warmer Bedingungen, jedoch nur weniger höherer Lagen des Berglandes (Gauer u. Aldinger (Hrsg.) 2005). Diese Spanne edaphischer und klimatischer Verhältnisse der Datengrundlage erreicht nicht die standörtlichen Extrema des ausgedehnten Heimatareals der Roteiche. Trotzdem führt sie bereits zu großen Unterschieden in Nordwestdeutschland beobachteter Höhenwuchsleistungen mit Site Index (SI)-Werten zwischen 17,3 und 31 m bei einem Bezugsalter von 50 Jahren.
Die Modellierung des Einzelbaumwachstums zielt auf robuste Prognosemodelle zur Verwendung in einem Einzelbaumwachstumssimulator und setzt deshalb zunächst bei den bestehenden, bewährten Modellformen von BWinPro/TreeGrOSS (Nagel 1999, Döbbeler et al. 2003) an. Für die Modellierung der Alters-Höhenbeziehung, im Wachstumssimulator gleichzeitig die Grundlage des potenziellen Höhenzuwachses der Einzelbäume, wird weiter die dreiparametrische Chapman-Richards-Funktion verwendet (Nagel 1994, Gadow 2003). Ihre Neuparametrisierung führt zu steileren, mit den Edellaubbaumarten Bergahorn (Acer pseudoplatanus L.) und Esche (Fraxinus excelsior L.) vergleichbaren Höhenzunahmen sehr junger Bestände und anschließend zu einem eher gestreckten, jedoch bis in hohe Alter anhaltenden Höhenwachstum. Ein alternatives, auf der Zuwachsfunktion von Sloboda (1971) beruhendes Modell zeigt bezüglich der Lage des Wendepunktes der Höhenentwicklung ein biologisch sehr plausibles Jugendwachstum. Verworfen wird dieses Modell aufgrund von Unsicherheiten bestimmter Höhenmessungen, die möglicherweise zu Überschätzungen der Höhenzunahmen in hohen Altern führen, sowie von Einschränkungen der nutzbaren Datenbasis, da nur Differenzen wiederholter Aufnahmen verwendet werden können.
Ein Kronenbreitenmodell wird für die Roteiche in Deutschland erstmals parametrisiert, nachdem BWinPro/TreeGrOSS bisher ein Kronenbreitenmodell der in Deutschland heimischen Stiel- (Quercus robur L.) und Traubeneiche (Quercus petraea [Matt.] Liebl.) verwendete. Es handelt sich um ein verallgemeinertes lineares Modell mit dem BHD als einziger erklärender Variablen. Nach dem neuen Modell liegt der Standraumbedarf der Roteiche bis zu einem BHD von ca. 50 cm unter dem der heimischen Eichen, erst zu höheren BHD etwas darüber und durchgängig unter dem der Rotbuche (Fagus sylvatica L.). Weitere Einflussfaktoren auf die Kronenbreite von Roteichen deuten sich an, so die Zugehörigkeit zum Ober- oder Unterstand sowie die unterschiedliche Freistellung von Z- und Füllbestandsbäumen. Für ein einfaches und robustes Prognosemodell wird die modellhafte Einbeziehung dieser Faktoren aufgrund des zusätzlichen Datenbedarfes jedoch zunächst als unzweckmäßig erachtet.
Ein überarbeitetes Kronenansatzmodell enthält an unabhängigen Variablen den BHD, das H/BHD-Verhältnis als Ausdruck der bisherigen Konkurrenzsituation des Baumes und die Spitzenhöhe des Bestandes zur Berücksichtigung des Standortes. Mathematisch besitzt das neue Modell die Form eines verallgemeinerten nichtlinearen Modells (Generalized nonlinear least squares fit; Pinheiro et al. 2019) mit einer zusätzlichen Varianzmodellierung mithilfe der Varianzfunktion „Power-of-the-Mean“. Gegenüber dem früheren Kronenansatzmodell besitzen Roteichen jetzt im Durchschnitt längere Kronen. Außerdem ist die Kronenlänge nunmehr sensitiver gegenüber dem Standort, indem Bäume in wüchsigeren Beständen bei unveränderten Werten der anderen Einflussgrößen deutlich kürzere Kronen aufweisen.
Die Konkurrenz für die Modellierung des Einzelbaumwachstums wird auch künftig durch den bewährten Konkurrenzindex c66 abgebildet und die Freistellung im Zuge von Durchforstungen durch dessen Änderung c66c. Verschiebungen der Konkurrenzberechnung durch die neue Kronenbreiten- und Kronenansatzfunktion bleiben gering, da sich deren Effekte mit über ein großes BHD-Spektrum jetzt schmaleren, dafür aber längeren Kronen größtenteils neutralisieren.
Die modellhafte Ableitung maximaler Dichten bzw. maximaler Grundflächenhaltungen von Roteichenbeständen verfolgt verschiedene methodische Ansätze. Eine als Referenz für die Dichtesteuerung im Rahmen der Bestandesbehandlung angestrebte Funktion der maximalen Grundfläche in Abhängigkeit der Oberhöhe und damit der Höhenbonität kann nach dem Ansatz von Sterba (1975, 1981, 1987) mit den zur Verfügung stehenden Daten, die nur wenige Nullflächenbeobachtungen enthalten, nicht plausibel hergeleitet werden. Einfachere Ansätze, basierend auf dem Stand Density Index (SDI) nach Reineke (1933) oder in Roteichenbeständen beobachteten maximalen c66-Werten, ergeben plausiblere Ergebnisse. Sie bestätigen im Wesentlichen die bisher in BWinPro/TreeGrOSS zur Initialisierung dichtebedingter Mortalität verwendete Überschirmungsdichte und Bestandesgrundfläche (Döbbeler et al. 2003).
Die erste Version eines Grundflächenzuwachsmodells der Roteiche als Kern des Einzelbaumwachstumsmodells stützt sich auf die bewährte Modellkonstruktion von BWinPro/TreeGrOSS mit der Mantelfläche der Lichtkrone als Hauptprädiktor des Grundflächenzuwachses und den Effekten der unabhängigen Variablen Baumalter, abstandunabhängige Konkurrenzindex c66 und Freistellung c66c als Konkurrenzänderung bei Durchforstungen. Die Mantelfläche der Lichtkrone wird mit den neuen Funktionen von Kronenbreite und -ansatz weiter nach der Modellkrone von BWinPro/TreeGrOSS in Form eines elliptischen Rotationsparaboloides berechnet. Ebenfalls wie bisher unveränderlich und damit baumarten- und behandlungsunabhängig bleiben die Kronenproportionen mit einem relativen Lichtkronenanteil von stets zwei Dritteln der Kronenlänge von der Baumspitze aus, obwohl andere Autoren gerade für Laubbäume Abweichungen von dieser Annahme feststellen (Pretzsch 2002, Sprauer 2013). Die Neuparametrisierung des Referenzmodells nach der BWinPro/TreeGrOSS-Modellkonstruktion unter Anwendung der neuen Funktionen von Kronenbreite und -ansatz bei der Berechnung von Kronenmantelfläche, Konkurrenz und Freistellung führt zu deutlichen Veränderungen der Funktionsparameter gegenüber dem vorherigen Stand (Nagel 1999, Döbbeler et al. 2003). Gerichtete Residualabweichungen der Zuwachsschätzungen dieses Modells und Abweichungen der Effekte wichtiger Prädiktoren von einem linearen Verlauf motivierten die Parametrisierung eines alternativen Grundflächenzuwachsmodells. Dafür wird ein generalisiertes additives Modell der Familie GAULSS (Sprauer 2013) mit Beschreibung heterogener Varianz bzw. Standardabweichung bei Normalverteilungsannahme der Fehler verwendet (Schmidt 2020). Gleichzeitig wird in diesem Modell die komplexe Lichtkronenmantelfläche durch die einfachen Dimensionsgrößen BHD und Bekronungsgrad ersetzt, während die anderen erklärenden Variablen des Referenzmodells beibehalten werden. Die anschließende Linearisierung der Effektverläufe durch geeignete Näherungsterme dient vorrangig der technischen Implementierung in BWinPro/TreeGrOSS. Sowohl das neu parametrisierte Referenzmodell als auch das neue alternative Grundflächenzuwachsmodell zeigen grundsätzlich plausible Zuwachsschätzungen und eine hinreichende Konkurrenzsensitivität. Eine Validierung an zunächst nur begrenzt zur Verfügung stehenden unabhängigen Daten erlaubt noch keine abschließende Entscheidung zugunsten der einen oder anderen Modellform, so dass weitere Überprüfungen auch unter Implementierung in die vollständige Software-Umgebung des Wachstumssimulators BWinPro/TreeGrOSS empfohlen werden.
Das Bestandeswachstumsmodell einer neuen Reinbestands-Ertragstafel der Roteiche für Nordwestdeutschland, erstellt nach der klassischen Top-down-Methode (Pretzsch 2002), ausgehend von den Grundbeziehungen zwischen Alter und Spitzenhöhenentwicklung sowie Spitzenhöhe und Gesamtwuchsleistung, gibt als Behandlungsnorm eine gestaffelte Hochdurchforstung vor. Die relativen Höhenbonitäten orientieren sich an den Spitzenhöhen im Alter 100 Jahre der Ertragsklassen der Roteichen-Ertragstafel für Westdeutschland von Bauer (1955), ergänzt um eine 0. Ertragsklasse. Durch die neue Alters-Höhenbeziehung ergeben sich innerhalb dieses neuen Bonitätsfächers Veränderungen der Verläufe des Höhenwachstums gegenüber der alten Ertragstafel. Weitere Merkmale der neuen Ertragstafel sind über einen weiten Altersbereich geringere Grundflächen- und Stammzahlhaltungen des verbleibenden Bestandes, womit die angestrebten stärkeren Durchmesser erreicht werden. Dies geht einher mit einem gegenüber der bisherigen Ertragstafel nicht erhöhten Niveau der Gesamtwuchsleistung vergleichbarer Höhenbonitäten. Dafür tritt eine standörtliche Differenzierung in der Grundflächenhaltung und Produktivität der neuen Ertragstafel wesentlich stärker hervor. Angesichts nicht stationärer Wuchsverhältnisse infolge der Dynamik von Standortsfaktoren, die sich in Änderungen des Wuchsverhaltens langfristiger Versuchsflächen offenbaren und gewisse methodische Schwierigkeiten verursachen, werden durch die neue Ertragstafel gegenwärtige Wuchsverhältnisse der jeweiligen Altersbereiche abgebildet. Die Ergebnisse eines jüngeren Durchforstungsversuches auf einem für die Roteiche geeigneten, verbreiteten Standortstyp bestätigen die Behandlungsvorgabe und Grundflächenhaltung der neuen Ertragstafel in Bezug auf die angestrebten Produktionsziele für die erste Durchforstungsphase.
Gegenüber den in mehreren Versuchsanlagen integrierten standorts- und altersgleichen Stiel- und Traubeneichenbeständen erreichen Roteichenbestände zum Alter von ca. 100 Jahren um 30 bis 80 % höhere Volumenleistungen sowie um 10 cm größere Mitteldurchmesser des Oberstandes.
Ein einfaches Standort-Leistungsmodell auf der Basis eines allgemeinen linearen Modells mit dem SI als Zielvariable stellt eine Beziehung zwischen der Leistung der Roteiche nach der neuen Ertragstafel und den Boden- und Klimaverhältnissen her. Innerhalb der durch Daten abgedeckten Amplitude der Standortbedingungen erlaubt das Modell eine quantifizierte Abschätzung der Standorteignung und des zu erwartenden Wachstums von Roteichenbeständen. Neben der starken Wirkung der Wasserhaushaltsstufe des Bodens geht eine positive Wirkung auf die Höhenwuchsleistung von einer höheren Jahresmitteltemperatur, einer besseren klimatischen Wasserbilanz in der Vegetationszeit sowie einem weniger weit zurückliegenden Keimjahr aus, während die Nährstoffstufe der Standortskartierung kaum eine Wirkung auf das Höhenwachstum erzielt.
Bezüglich der Standortinteraktion als Merkmal der ökologischen Zuträglichkeit einer eingeführten Baumart (Otto 1993) ist die Humusform in Roteichenbeständen nicht durchgehend ungünstig, sondern eng mit der Nährstoffstufe des Standortes korreliert. Ein pauschal negativer Einfluss einer Roteichenbestockung auf Boden- und Humuszustände konnte nicht bestätigt werden. Bodenchemische Kennwerte unter etablierten Roteichenbeständen des niedersächsischen Tieflandes liegen für gleiche Bodensubstratgruppen innerhalb durchschnittlicher Bodenzustände aller Bestockungstypen unterschiedlicher Baumarten der Bodenzustandserhebung II (Evers et al. 2019) und bieten daher keine Anhaltspunkte für eine auffällige systematische Verschlechterung der Bodeneigenschaften. Die Kohlenstoffbindung der untersuchten Böden unter Roteichenbeständen des Tieflandes ist eher überdurchschnittlich. In einem Baumartenvergleich zwischen gleichalten Roteichen- und Traubeneichenbeständen auf einem nährstoffkräftigen, frischen und einem ziemlich armen, mäßig trockenen Standort des Wuchsbezirkes Ostheide zeigt die Roteiche in der Summe von Boden und Bestand einer höhere Gesamtkohlenstoffspeicherung. In einigen bodenchemischen Kennwerten wie der Basensättigung und den Stickstoffvorräten besitzt die Traubeneiche auf beiden Standorten gewisse Vorteile, auf dem schwächeren Standort darüber hinaus in den Gesamtvorräten an Kalium und Phosphor. Auf dem besseren Standort ebenfalls etwas günstiger sind der Zustand der Humusauflage sowie die Kohlenstoffumsätze und -verfügbarkeit unter der Traubeneichenbestockung. Dagegen zeigt der Boden des schwächeren Standortes die höhere biologische Aktivität unter der Roteiche, was sich in einem größeren Vorrat an mikrobiellem Kohlenstoff und einer deutlich besseren Kohlenstoffverfügbarkeit dokumentiert. In einem schwach wüchsigen Kiefernbestand auf einem nährstoffschwachen, früher streugenutzten Standort führten ein Roteichenvoranbau und die gleichzeitige Kompensationskalkung mit 6 t je ha kohlensaurem Dolomitkalk nach 20 Jahren zu einem deutlich verbesserten Humus- und Bodenzustand.
In Verjüngungsversuchen aus Pflanzung auf der Freifläche und unter dem Schirm von Kiefernbeständen unter trocken-warmen Klimaverhältnissen auf nährstoffschwachen Sandböden zeichnet sich die Roteiche durch eine geringe Mortalität in der Etablierungsphase aus und ist darin heimischen Laubbaumarten und mediterranen Eichenarten überlegen. In einem Versuch auf der Freifläche trifft das auch auf die hier mit untersuchte Freisaat der Roteiche zu, die sich als erfolgreiches Kulturbegründungsverfahren erweist. Vor allem kommt die Roteiche mit trotz fortschreitender Temperaturerhöhung noch immer regelmäßig auftretenden Spätfrosten besser zurecht als Trauben-, Stiel-, Flaum- (Quercus pubescens Willd.), Stein- (Quercus ilex L.) und Ungarische Eiche (Quercus frainetto Ten.) sowie Rotbuche. Dies führt neben den geringeren Ausfällen zu einem gegenüber diesen Baumarten überlegenen Jugendwachstum. Die längere Überschirmung durch einen Kiefernbestand hat auf das Höhenwachstum der Roteiche relativ geringere Auswirkungen als auf das der Sandbirke (Betula pendula Roth.), der Stieleiche und der Rotbuche, die als einzige den Schirm bevorzugt. Von sehr starken Auflichtungen eines Kiefernschirms profitiert das Wachstum der Roteiche mittelfristig weniger als jenes der Douglasie (Pseudotsuga menziesii [Mirb.] Franco). Unter Freiflächenbedingungen sind gleichalte Waldkiefern (Pinus sylvestris L.) und Sandbirken als heimische Pionierbaumarten der Roteiche gegenüber mindestens gleich-, in der Regel vorwüchsig und Roteichenpflanzungen lassen deren natürliches Ankommen zu. In einzelstammweiser, gleichalter Mischung mit Rotbuche bleibt die Roteiche selbst unter einem Kiefernschirm mittlerer Überschirmungsdichte lange vorwüchsig, ohne die später potenziell im Wachstum aufholende Rotbuche zu verdrängen. Die vorteilhaften Eigenschaften im Hinblick auf die Etablierung unter schwierigen Standortverhältnissen und die Behauptung in Mischbeständen sprechen unter Berücksichtigung der letztgenannten Aspekte nicht für die Invasivität der Roteiche im Sinne einer nicht kontrollierbaren Ausbreitung und Verdrängung heimischer Arten.
Die Untersuchungsergebnisse von Naturverjüngungen in drei Roteichenbeständen lassen sich in den bestehenden Kenntnisstand zur Verjüngungsökologie der Baumart in ihrem natürlichen Verbreitungsgebiet und in Deutschland einordnen. Sie bestätigen für Roteichenaltbestände grundsätzlich hohe Erfolgsaussichten einer natürlichen Verjüngung mit hohen Sämlingsdichten aus mehreren Samenjahren. Als wichtigste Grundlage der Verjüngungsetablierung erweist sich eine gegenüber den heimischen Eichenarten höhere Überschirmungstoleranz. Eine wirksame Senkung des Wildverbisses, dem die Roteiche in besonderer Weise unterliegt, und die Lichtung eines ggf. vorhandenen Unterstandes aus Schattbaumarten sind Voraussetzungen des Verjüngungserfolges. Die Ergebnisse der vorliegenden Arbeit unterstützen die Klassifizierung der Roteiche als Lückenspezialistin mit einer Anpassungsstrategie, die eher auf ihrer Stresstoleranz beruht als auf der Fähigkeit zu extrem starken Zuwachsreaktionen bei veränderter Ressourcenverfügbarkeit. Trotz gewisser Überlebensfähigkeit auch unter dunkleren Bedingungen setzt eine stärkere Zunahme des Höhenwachstums der Roteichennaturverjüngung erst bei Lichtverhältnissen von etwa 15 % Indirect Site Factor (ISF) ein, während ein Wendepunkt zu degressiv steigenden Höhenzunahmen bei ca. 25 % ISF auftritt. Diese Angaben bleiben orientierende Richtwerte, da das Monitoring der drei Naturverjüngungsversuche weitere wichtige Einflussfaktoren nicht in eine systematische Versuchssteuerung einbezog. Ein Modell zum Höhen- und Wurzelhalsdurchmesserwachstum als Fallstudie an einem der Naturverjüngungsversuche bezieht eine pflanzenindividuelle Altersbestimmung mit ein und identifiziert die Ungleichaltrigkeit im Zusammenwirken mit unterschiedlichem Lichtgenuss als wichtigste Treiber der starken Höhen- und Durchmesserdifferenzierung der Naturverjüngung.
Langjährig dokumentierte Beobachtungen der zahlreichen Versuchsflächen fügen sich in die Ergebnisse einer Literaturübersicht ein, wonach die Einschleppung von auf die heimische Flora und Fauna nachteilig wirkenden Organismen durch die Roteiche bisher trotz einer langen Anbaugeschichte nicht nachgewiesen ist und ebenso wenig ihre eigene existenzielle Bedrohung durch hiesige abiotische und biotische Gefährdungen. Nach Spätfrostereignissen, Blattfraß durch die Eichenfraßgesellschaften und Bodenfeuern besitzt die Roteiche ein gutes Regenerationsvermögen aufgrund ihrer starken Wiederausschlagfähigkeit. Ansätze einer ökologischen Integration in Mitteleuropa beruhen auf Adaptionsmöglichkeiten heimischer Arten an die Strukturen lebender Roteichen sowie an deren Totholz, das durch zahlreiche Destruenten erschlossen werden kann. Aus einer fehlenden Koevolution resultierende Nachteile für bestimmte mitteleuropäische Nahrungsspezialisten können durch die Mischung mit heimischen Baumarten gemindert werden können. Eine Fallstudie im Umfeld eines Roteichenbestandes innerhalb eines Stieleichen-Hainbuchen-Waldes findet keine unkontrollierte horizontale Ausbreitung von Roteichennaturverjüngung, wofür als Hauptursachen geringe Verbreitungsentfernungen des Samens und die sehr starke Verbisspräferenz der Roteiche identifiziert werden. Das Ausbreitungsverhalten in lichten Wäldern, vor allem Kieferbeständen, war nicht Gegenstand eigener Untersuchungen und bleibt weiterer Forschung vorbehalten.
Ein waldbauliches Behandlungskonzept für Roteichen-Rein- und Mischbestände berücksichtigt die im Rahmen dieser Arbeit erzielten neuen Erkenntnisse und stellt deren Synthese an der Schnittstelle zu einer unmittelbaren praktischen Anwendung dar. Es kennzeichnet die Roteiche als eine anpassungsfähige, ertragreiche Baumart mit vorteilhaften Eigenschaften, wozu in erster Linie ihre Robustheit bei der Etablierung unter schwierigen Ausgangsbedingungen, die Integrationsfähigkeit in strukturreiche Mischbestände und die Möglichkeiten einer einzelstammweisen Nutzung und waldbaulich steuerbaren natürlichen Regeneration gehören.Seventy years after the first monographs on the Northern Red Oak (Quercus rubra L.) for West Germany (Bauer 1953a) and East Germany (Göhre and Wagenknecht 1955) were published, this work is once again devoted to growth, silvicultural treatment, regeneration ecology and, beyond that, ecological aspects of integration of this tree species, introduced to Germany 250 years ago in forest cultivation.
The focus of the forest growth part is newly created or newly parameterized models of the single tree and stand growth of Northern Red Oak for northwest Germany. Mechanistic Studies on flat and stepped Palladium Single Crystal Surfaces: Experimental Evidence of Reaction Intermediates for the Decomposition of Formic Acid and Oxidation of Ammonia
In this work, experimental and theoretical investigations of formic acid desorption, decomposition and ammonia oxidation on single crystal palladium surfaces are presented. The formic acid system is investigated using a helium seeded molecular beam of formic acid, isotopic labeling and universal femtosecond laser ionization. Ammonia oxidation is investigated using crossed molecular beams, resonanced-enhanced multiphoton ionization and laser-induced desorption. Ion-imaging detection is used for both systems. All experimental results are supported by density functional theory
(DFT) based kinetic modeling.
First, the thermal desorption rate of formic acid from Pd(111) between 228 to 273K using four isotopologoues (HCOOH, DCOOH, HCOOD, DCOOD) is investigated. Upon molecular adsorption, formic acid undergoes decomposition to CO2 and H2 and thermal desorption. The contribution of these processes to the measured rates is disentangled by quantification of the desorption probability using mass conservation. From the experimentally derived thermal desorption rate constants, the isotope-independent binding energy of 0.639 ± 0.008 eV is obtained using the detailed balance rate model. The experimental binding energy is compared to predictions of different DFT functionals.
Second, the intermediates in the formic acid decomposition reaction on Pd(111) and Pd(332) are identified using isotopologue specific CO2 formation rates. At surface temperatures around ∼400K CO2 formation shows on both facets two temporally distinct channels which exhibit unique and large kinetic isotope effects (KIEs) upon isotopic labeling. Supported by rate constants derived from transition-state theory (TST) using DFT, the fast channel is assigned to the carboxyl intermediate (C*OOH) and the slow channel to the bidentate formate intermediate (HCO*O*). This is the first
experimental observation of the carboxyl intermediate in any surface science experiment. On Pd(332) CO formation is observed which is only possible from the carboxyl intermediate. Furthermore, the experimental derived rate constants for bidentate formate dissociation show a large KIE. A new pathway based on TST-DFT derived rate constants is presented which predicts the correct KIE.
Third, a mirco-kinetic model (MKM) within the mean-field (MF) approximation based on the identified reaction intermediates is presented. The MF-MKM is constructed using a one-dimensional grid which is separated into terrace and step-sites. The model is evaluated against the experimental CO2 formation rates and desorption yields of HCOOH, CO2 and CO on Pd(111) and Pd(332). While the initial prediction based on TST-DFT rate constants shows qualitatively correct results, a fitting routine to adjust the DFT calculated barrier heights is employed. The fitting routine improves the quantitative agreement by a factor of 2. The results of the MF-MKM are analyzed using the degree of rate control analysis. Predictions about other reaction products that have not experimentally been investigated and the high coverage regime are made.
Last, results on ammonia oxidation on Pd(332) are presented. Ammonia is oxidized by atomic oxygen on the surface to form NO and H2O. NO formation rates feature a temporally resolved onset and decay which is fitted using the simple model
I∗→ N∗O→ NO. Novel experimental data on the temporal evolution of the concentration of nitrogen atoms on the surface using laser-induced desorption restrict the simple kinetic model to formation of N*O being slower than desorption. A more complex kinetic model is developed based on oxygen assisted hydrogen abstraction from NHx (x = 3, 2, 1) intermediates which indicates that the rate determining step of ammonia oxidation is N*O formation from atomic nitrogen and atomic oxygen. Scaling of the effective rate constant of N*O formation indicates that the active species is oxygen bound to the up-step site. TST-DFT rate constant modeling shows reasonable agreement with the experimental second order rate constant but indicates an error in the energy barrier of at least 0.19 eV.2024-10-1
Development of mass spectrometry-based proteome analysis workflows for the systematic study of cardiac disease research models
A specific variant of genetic mutation in the phospholamban gene expresses a cardiac phospholamban protein lacking the arginine amino acid at position 14 (R14Δ-PLN). The mutation is believed to have a founder in the Netherlands, with over 1500 cases detected. It is also present in Greece, the United States, Canada, China, and Japan. The variant potentially leads to dilated cardiomyopathy and/or arrhythmogenic cardiomyopathy. A clinical study in the Netherlands suggests that there is a 12–15% correlation between the R14Δ-PLN mutation and dilative cardiomyopathy or arrhythmogenic cardiomyopathy, respectively. The clinically documented cases for the R14Δ-PLN mutation exhibit heterozygosity. Patient carriers are at risk of developing biventricular abnormalities, and fatty and fibrofatty infiltration of the outer compact layer of the ventricular myocardium, with a poor prognosis. Under unknown treatment, efforts are focused on detecting early disease events in cardiomyopathy prior to the manifestation of the initial symptoms.
Complexome profiling and crosslinking mass spectrometry are two systems wide mass spectrometry-based proteome methods with proven capabilities for the detection of protein protein interactions. Complexome profiling was applied to study atrial fibrillation by Alsina et al., 2019. Crosslinking mass spectrometry was studied in mouse cardiac tissue in a pioneering study by Chavez et al., 2018; there are, however, only a few reports of the application of complexome profiling and crosslinking mass spectrometry to whole-cell or tissue studies in cardiovascular research. While less explored in cardiology, the methods hold promise for revealing system-wide changes with further development and optimization.
We hypothesized that the R14Δ-PLN mutation causes a systemic affection that leads to cardiomyopathy through changes or alterations in the composition and function of different protein-protein complexes and critical cardiac pathways. Therefore, we developed novel systems for wide mass spectrometry-based proteome analysis workflows and investigated the R14Δ-PLN cardiomyopathy in a well-established heterozygous mouse disease-causing variant research model of the R14Δ-PLN mutation.
Here, I extend the state-of-the-art of complexome profiling and crosslinking mass spectrometry workflows with enhanced application to the cardiology field. I introduce two platforms for complexome profiling and one for crosslinking mass spectrometry, along with their respective bioinformatics analysis approaches: 1- mini-Complexome Profiling (mCP), a precast blue native polyacrylamide gel electrophoresis data independent acquisition mass spectrometry workflow. Along with software for FDR-controlled global targeted detection of protein complexes. 2- Extended-range SEC-DIA/MS workflow, utilizing size exclusion chromatography (SEC). Along with a bioinformatics workflow for unbiased differential identification of altered protein profiles between two conditions. 3- A crosslinking mass spectrometry workflow. Along with an integrative workflow for mutual validation of crosslinking and complexome profiling of the detected protein protein interactions.
I conducted a systematic analysis of high-molecular weight protein complexes and supercomplexes in young, presymptomatic R14Δ/+ mouse hearts by CP workflows. We identified alterations in inner mitochondrial membrane complexes and preliminary intercalated disk supercomplexes, which are involved in cardiomyocyte-to-cardiomyocyte electrical contacts, as early indicators of disease. Additionally, our findings suggest MICOS complexes alterations, rather than changes in calcium handling components, are associated with disruptions at ER-Mitochondria contact sites. The impaired cardiomyocyte connections and resulting energetic imbalance could explain the arrhythmic events in the disease. These discoveries highlight the impact of mitochondrial dysfunction and ICD cell-cell contact disruption in the early stages of R14Δ-PLN cardiomyopathy, offering potential early-disease therapeutic targets, particularly crucial given the lack of effective treatments for advanced stages of the disease.2024-10-0
Influence of therapeutic inhibition of anaplastic lymphoma kinase on the regulation of mitochondrial apoptosis in ALK+ anaplastic large cell T-cell lymphomas
T-Zell Lymphome sind eine seltene, heterogene Gruppe an Erkrankungen reifer post-thymischer T- oder natürlicher Killer-Zellen mit primär nodaler oder extranodaler Manifestation. Der Subtyp anaplastisches, großzelliges Lymphom (ALCL) kann anhand einer Translokation der anaplastischen Lymphomkinase (ALK) in ALK+ und ALK- unterschieden werden. In ALK+ ALCL-Erkrankungen ist der häufigste Translokationspartner Nucleophposmin (NPM). Aufgrund der beschriebenen Translokation kommt es zu einer konstitutiven Autophosphorylierung des NPM-ALK Fusionsproteins. Daraus resultieren downstream-Effekte verschiedener intrazellulärer Signalkaskaden mit den Folgen einer unabhängigen Zellproliferation und Apoptoseresistenz. Für junge erkrankte Patienten ist die Prognose gut. Die 5-Jahres progressionsfreie Überlebensrate liegt für rezidivierte Patienten und Patienten mit initialem hohen Prognoseindex jedoch bei nur 20 bis 30 %. Daraus ergibt sich die Notwendigkeit neuer Therapieansätze. Eine Möglichkeit eine ALK+ ALCL-Erkrankung zu adressieren ist eine Inhibition der NPM-ALK Aktivität mit einem Tyrosinkinasen-Inhibitor. Der Tyrosinkinasen-Inhibitor der ersten Generation Crizotinib erzielte in klinischen Studien vielversprechende Ergebnisse, dennoch erreichten nicht alle Patienten eine Remission. Deswegen sollen in dieser Arbeit die Effekte von zwei Tyrosinkinasen-Inhibitoren – Crizotinib und Alectinib - in vitro untersucht und Strategien zur Verstärkung der therapeutischen Effektivität durch Kombinationsstrategien entwickelt werden. Hierfür sollen insbesondere mögliche Mechanismen der Apoptosedysregulation, die zum Überleben der ALCL-Zellen unter ALK-Inhibition beitragen, analysiert werden. Darauf aufbauend sollen rationale Kombinationsstrategien zur Steigerung der therapeutischen Effektivität entwickelt werden, besonders soll die Kombination mit dem bereits für diese Entität zugelassenen CD30 Wirkstoff-Antikörper-Konjugat Brentuximab Vedotin (BV) geprüft werden.
In Viabilitätsmessungen wurde in vitro die Beeinflussung der Zellviabilität unter aufsteigenden Konzentrationen von Crizotinib und Alectinib beschrieben. Durch die Zellzyklus-Analysen wurden Erkenntnisse zu Zellzahl/Zelltod und Stadium des Zellzyklus gewonnen. Mittels dynamischem BH3-profiling konnten Einblicke in die Mechanismen der mitochondrialen Apoptose ermöglicht werden und eine gesteigerte funktionelle Abhängigkeit von Bcl-xL und Mcl-1 gezeigt werden. Abschließend wurden mit Western Blot-Analysen die Effekte auf die Proteinexpression der Proteine der Bcl-2 Familie näher beschrieben und eine on-target Aktivität nachgewiesen.
Nicht alle getesteten ALK+ ALCL-Zelllinien zeigten eine starke Abnahme der Zellviabilität nach Behandlung mit den Inhibitoren, teils konnte die Ausbildung einer residuellen Population beobachtet werden. Nach Crizotinib Inkubation nahm in allen getesteten Zelllinien der prozentuale Anteil der G0/G1-Phase ab. Es konnte eine gesteigerte Abhängigkeit der verwendeten ALK+ ALCL-Zelllinien von den anti-apoptotischen Proteinen Mcl-1 und Bcl-xL nach Crizotinib Inkubation festgestellt werden. In Western Blot-Analysen wurde anhand einer abnehmenden pALK-Proteinexpression die on-target Aktivität nachgewiesen werden. Darüber hinaus zeigten die Western Blot-Analysen eine abnehmende Proteinexpression des pro-apoptotischen Mcl-1 Antagonisten NOXA trotz Persistenz des Proteins Mcl-1. Für die Kombination Bcl-xL Inhibitor ABT263/Crizotinib wurden geringe viabilitätshemmende und synergistische Effekten beschrieben. Die Kombination Mcl-1 Inhibitor/Crizotinib zeigte starke antiproliferative Effekte und synergistische Effekte. Im Hinblick auf eine klinische Anwendbarkeit sowie putative Effekte des Antiköper-Wirkstoff-Konjugats BV auf anti-apoptotische Bcl-2 Familienmitglieder wurde nachfolgend eine Kombinationsstrategie von BV mit Crizotinib oder Alectinib untersucht. BV reduzierte die Viabilität der getesteten Zelllinie in Kombination mit Crizotinib und Alectinib deutlich. In weiterführenden Zellzyklus-Analysen konnte gezeigt werden, dass mehr Zelltod induziert werden konnte als durch die Medikamente Crizotinib und BV einzeln betrachtet. Aufgrund des unterschiedlichen Angriffspunkts im Zellzyklus der beiden Medikamente wurde das Konzept der sequenziellen Behandlung entwickelt. Die Effektivität der Kombination Crizotinib/BV konnte in einer Zelllinien im Vergleich zur einzeitigen Behandlung gesteigert und mehr Apoptose induziert werden.
Zusammenfassend kann die Kombination AZD-5991/Crizotinib und BV/Crizotinib als vielversprechend gewertet werden. Es benötigt eine in vivo Prüfung der Kombination BV und Crizotinib zum Verifizieren dieser Ergebnisse und Testung der klinischen Umsetzbarkeit v. a. bezogen auf das Konzept der sequenziellen Behandlung.T-cell lymphomas are a rare, heterogeneous group of diseases of mature post-thymic T- or natural killer cells with primarily nodal or extranodal manifestation. The anaplastic large cell lymphoma (ALCL) can be classified into two subtypes, ALK-positive and ALK-negative, based on the presence or absence of a translocation involving the anaplastic lymphoma kinase (ALK). In ALK+ ALCL diseases, the most common translocation partner is nucleophposmin (NPM). The translocation results in the continuous autophosphorylation of the NPM-ALK fusion protein. This leads to downstream effects in various intracellular signaling pathways, resulting in independent cell proliferation and resistance to apoptosis. The prognosis is good for young patients. However, the 5-year progression-free survival rate for relapsed patients and patients with an initial high prognosis index is only 20 to 30 %. This results in the need for new therapeutic approaches. One approach to target ALK+ ALCL is to inhibit NPM-ALK activity using a tyrosine kinase inhibitor. The first-generation tyrosine kinase inhibitor Crizotinib achieved promising results in clinical trials, but not all patients achieved remission. Consequently, we will investigate the effects of two tyrosine kinase inhibitors, Crizotinib and Alectinib, in vitro, while also developing strategies to enhance therapeutic efficacy through combination approaches. Specifically, we will examine the potential mechanisms of apoptosis dysregulation that enable ALCL cells to survive despite ALK inhibition. Based on this, rational combination strategies to increase therapeutic efficacy will be developed, in particular the combination with the CD30 antibody-drug conjugate Brentuximab Vedotin (BV), which is already approved for this entity, will be tested.
In viability measurements, the influence of ascending concentrations of Crizotinib and Alectinib on cell viability was described in vitro. Cell cycle analyses provided insights into cell number/cell death and cell cycle. Dynamic BH3-profiling provided insights into the mechanisms of mitochondrial apoptosis and showed an increased functional dependence on Bcl-xL and Mcl-1. Finally, the effects on the protein expression of the Bcl-2 family proteins were described in detail using Western blot analyses and on-target activity was demonstrated.
Not all tested ALK+ ALCL cell lines showed a strong decrease in cell viability after treatment with the inhibitors; in some cell lines a residual population was observed. After Crizotinib incubation, the percentage of the G0/G1 phase decreased in all tested cell lines. An increased dependence of the ALK+ ALCL cell lines on the anti-apoptotic proteins Mcl-1 and Bcl-xL was observed after Crizotinib incubation. Western blot analyses demonstrated on-target activity by decreasing pALK protein expression. In addition, the Western blot analyses showed a decrease in protein expression of the pro-apoptotic Mcl-1 antagonist NOXA despite persistence of the Mcl-1 protein. The combination of the Bcl-xL inhibitor ABT263 and Crizotinib demonstrated low viability-inhibiting and synergistic effects. In contrast, the combination of the Mcl-1 inhibitor AZD5991 and Crizotinib exhibited significant antiproliferative and synergistic effects. Considering the clinical relevance and potential effects of the antibody-drug conjugate BV on anti-apoptotic Bcl-2 family members, a combination strategy involving BV and either Crizotinib or Alectinib was subsequently explored. BV significantly reduced the viability of the tested cell line in combination with Crizotinib and Alectinib. Further cell cycle analyses showed that more cell death was induced than by the drugs Crizotinib and BV individually. Due to the distinct mechanisms of action of the two drugs within the cell cycle, a sequential treatment approach was developed. The effectiveness of the combination Crizotinib/BV was increased in one cell line compared to single-drug treatment and more apoptosis was induced.
In summary, the combination of AZD-5991/Crizotinib and BV/Crizotinib can be considered promising. In vivo testing of the BV and Crizotinib combination is necessary to verify these results and assess their clinical applicability, particularly in relation to the concept of sequential treatment.2025-10-2
Evaluation of the diagnostic accuracy of B-scan ultrasonography for the grading of hepatic steatosis in patients with chronic liver disease
Abstract
Fatty liver disease is characterised by excessive storage of fat. The prevalence in Germany is about 30%. The worldwide prevalence is approximately 25%. This means that about 1.8 billion people are affected. Steatosis hepatis is diagnosed when 5% or more of the liver parenchyma are fatty. Based on the intracellular fat content, four stages are differentiated.
The spectrum of manifestations of steatosis is very diverse. Also the clinical presentation of patients with the disease is quite non-specific.
The aim of this work was to investigate the diagnostic accuracy of B-Mode sonography in determining the degree of steatosis in patients with hepatopathy. A retrospective analysis of data from a total of 231 patients between 03/2016 and 05/2019 was performed. 157 patients were included in the study after applying the exclusion criteria. B-Mode ultrasound of the liver was performed with high-end ultrasound devices such as the Logiq E9 (GE Medical Systems, Wauwatosa, USA; software R1.0.6) or Hitachi ALOKA (Pro Sound Alpha 7).
B-Mode ultrasonography showed a sensitivity of 75.6% and a specificity of 76% in discrimination between steatosis and no steatosis (AUROC 0.758). This resulted in a positive predictive value of 0.775 and a negative predictive value of 0.74. The diagnostic accuracy was 0.758 (0.680 - 0.836). For higher grades of steatosis (>II°), the specificity for the presence of histological steatosis was >98.7%. The sensitivity was 90.9%. The agreement between the two investigators was 89.2% (κ=0.783) for the presence of steatosis and 80.9% (κ=0.704) for the individual grades of steatosis. We were able to show that B-Mode ultrasonography has excellent diagnostic accuracy in the diagnosis of moderate and massive steatosis. In the presence of mild steatosis, the sensitivity was somewhat lower. As a result, high-end equipment may have increased sensitivity for steatosis detection, but also decreased specificity, resulting in more false positives compared to older sonography equipment. However, higher grade steatosis (grade 2 or higher) is detected very reliably, as shown in our work. The results were also detectable in patients with aetiology other than NAFLD.
Furthermore, we were able to show that the presence of fibrosis has no decisive influence on the sonographic diagnosis of fatty liver disease. Also, we were able to record very good results in the interobserver agreement. Overall, the results of this work show that B-scan sonography is a very good method to detect higher grade steatosis, independent of the degree of fibrosis or concomitant liver disease. Thus, sonography can be considered the diagnostic tool of first choice for the detection and graduation of steatosis hepatis.2024-11-2
