Wie beeinflusst Bestrahlung der extrazellulären Matrix das Invasionsverhalten von Nierenzellkarzinomen?
Renal cell carcinoma often forms bone metastases and is often only discovered in late stages due to metastasis-associated symptoms. Affected patients suffer from pain, the survival rates are poor. Thanks to new techniques such as stereotactic radiation, radiation therapy is becoming increasingly important in the treatment of renal cell carcinoma. In radiation therapy the cancer cells surrounding tissue is irradiated as well as the cancer cells. The surrounding tissue mostly consists of extracellular matrix (ECM). The ECM functions as a barrier to cancer cell metastasis by forming basement membranes. In this study we built an artificial basement membrane by using Matrigel. We studied in how far the invasion of renal cell cancer cells through an artificial basement membrane was effected by radiation of said membrane.
Our invasion experiments using the xCELLigence Real Time Cell Analysis Dual Purpose System show that radiation of a Matrigel membrane can inhibit the invasion of the renal cell carcinoma cell line 786-O through this membrane. This effect occurred at doses of 25 Gy and above - no significant effect was observed at 12.5 Gy. The exact reason for the inhibition is yet to be discovered. Previous research results suggest an interaction between radiation and collagen present in ECM. Collagen appears to be softened by the radiation, which seems to have a negative effect on the invasion of cancer cells.
In our experiments, we focused on laminin-1, which was analysed using Western Blot. As a heterotrimer, laminin-1 consists of three subunits: α1, β1, and γ1. The analyses showed that the band pattern of irradiated laminin intensified with increasing radiation dose. We suspect radiation to destroy laminin’s structure. This effect could only be observed in Western Blot analyses in which the used antibody was directed against the entire laminin protein. The analyses with antibodies against the subunits α1, β1, and γ1 showed no significant differences due to radiation. Furthermore, a significant effect of radiation in the Western Blot could only be found in relation to laminin within Matrigel. No significant change was observed in pure laminin. Laminin could be blocked by collagen in non-irradiated Matrigel and thus not be accessible to the Western Blot antibodies. Radiation can lead to unmasking, making laminin accessible to the antibodies and allowing it to exert an inhibitory effect on the renal cell carcinoma cells.
Understanding the effects of radiation on tissue surrounding tumors and the extent to which it has a positive or negative impact on the metastasis of various tumor entities can contribute to optimizing individual tumor therapy for patients and improve survival rates in the future.2026-01-0
Measurement of the Production Cross-Section in the Non-Resonant Four Lepton Final State in Collisions at with the ATLAS Detector
This thesis presents the measurements of the associated production of a top quark pair with a Standard Model Higgs boson in multi-lepton final states, focusing on the measurement of the inclusive cross-section in the channel. The analysis uses the full Run 2 data set, corresponding to an integrated luminosity of , collected in proton-proton collisions at a centre-of-mass energy of . The analysis covers six final states, defined by the number and flavour of charged-lepton candidates. Machine learning techniques are used for all channels to improve signal sensitivity. Leading prompt background contributions from , , and diboson production are constrained by additional regions populated with those backgrounds. Moreover, contributions from fake and non-prompt leptons are estimated using data-driven methods such as the fake factor or template method. The observed signal strength, the ratio of the measured signal yield over the Standard Model prediction, is extracted using a profile likelihood fit. For the channel, the observed signal strength is , corresponding to an observed (expected) significance of (). The measured signal strength for the combination of all six channels is . It corresponds to an observed (expected) significance of (). The measured inclusive cross-section is found to be . The compatibility is with the Standard Model prediction of . The mitochondrial calcium metabolism in the biology of malignant melanoma
Das maligne Melanom zählt zu den aggressivsten Hauttumoren und viele Patient*innen sprechen trotz Fortschritten im Bereich von Immuntherapien sowie der Anwendung von BRAF- und MEK-Inhibitoren nicht ausreichend auf bestehende Behandlungsoptionen an oder entwickeln Resistenzen im Verlauf. Vor diesem Hintergrund der Resistenzentwicklung gewinnt die biologische Funktion mitochondrialer Stoffwechselwege und Ca²⁺-abhängiger Signalprozesse zunehmend an Bedeutung. Eine zentrale Rolle spielt dabei der mitochondriale Calcium-Uniporter (MCU), der die Ca²⁺-Aufnahme in die Mitochondrien vermittelt und essenziell für deren Funktion ist. Diese Arbeit untersucht, wie eine Herunterregulation des MCU die mitochondriale Funktion, Proliferation, Therapiesensitivität und Resistenzentwicklung in Melanomzellen beeinflusst. Durch Transfektion-vermitteltem Knockdown des MCU sowie pharmakologische Hemmung mit Mitoxantron wurde dessen Rolle im malignen Melanom untersucht. MCU-herunterregulierte/knockdown Melanomzellen zeigten reduzierte basale ROS-Level, jedoch nach Vemurafenib-Behandlung eine verstärkte ROS-Antwort. Zudem wiesen sie ein schnelleres Wachstum auf, reagierten jedoch sensitiver auf Vemurafenib und Trametinib. Insgesamt zeigte sich der MCU als ein wichtiger Regulator mitochondrialer Funktion, der Proliferation und Therapieantwort von Melanomzellen. Weiterhin zeigt die vorliegende Arbeit, dass der mitochondriale Stoffwechsel und insbesondere der MCU interessante Ansatzpunkte für potenzielle neue Therapiestrategien sein könnten.Melanoma remains one of the deadliest skin cancers, and despite significant progress through immunotherapies and targeted treatments such as BRAF and MEK inhibitors, many patients develop therapeutic resistance. Increasing evidence highlights the importance of mitochondrial metabolism, redox regulation, and calcium signaling in melanoma biology. The mitochondrial calcium uniporter (MCU), a key mediator of mitochondrial Ca²⁺ uptake, plays a central role in these processes, however its impact on melanoma progression and treatment response remains incompletely understood. This study investigates how MCU downregulation influences mitochondrial function, proliferation, drug sensitivity, and resistance development in melanoma cells. Using stably transfected MCU-deficient melanoma cells and the specific MCU inhibitor mitoxantrone reveal that MCU silencing reduces mitochondrial ROS levels but leads to a stronger ROS increase following vemurafenib treatment. MCU downregulation further enhanced sensitivity to targeted therapies, with vemurafenib and trametinib causing greater growth inhibition in MCU-deficient cells. Taken together, this study shows that the MCU modulates mitochondrial function, proliferation, therapeutic sensitivity, and resistance in melanoma cells. Targeting mitochondrial calcium signaling might provide a novel opportunity to optimize melanoma treatment strategies.2026-01-1
Epitaxy of 2D Materials on Gallium Nitride
In this thesis, the epitaxial growth of 2H-TaS few layers onto GaN(0001) is demonstrated. A growth study is presented, in which the influence of substrate temperature and reactant fluxes is investigated via electron diffraction and scanning probe microscopy techniques. The best structural properties are found at a high growth temperature of C and are attributed to the observed transition from a screw dislocation driven growth mode to layer-by-layer growth with increasing substrate temperature. From the first monolayer, the 2H-TaS grows unstrained with an epitaxial relation of 2H-TaS () GaN (). The growth conditions can be directly transfered to hexagonal boron nitride and epitaxial graphene substrates. The composition as well as the structural and electronic properties within the grown overlayers and at the interface to the GaN substrate are further studied by x-ray photoemission spectroscopy and scanning transmission electron microscopy. The latter reveals a layered growth of 2H-TaS, but also the formation of pits in the substrates, which most probably form due to the thermal decomposition of GaN at the elevated growth temperatures. However, no indication of a chemical reaction between the substrate and overlayer is found. Observed core level shifts in photoelectron spectra are related to an electron transfer from the unintentionally doped n-GaN to the 2H-TaS and to the formation of nitrogen vacancies at the interface. With low temperature low-energy electron diffraction and temperature-dependent resistivity measurements, no clear evidence of the charge density wave transition expected in bulk TaS below K is found. Whether this is due to the GaN induced electron doping remains to be cleared. In conclusion, these findings illustrate that GaN(0001) is a promising substrate for the MBE growth of uniform, substrate-scale transition metal dichalcogenide layers with high orientation.2025-12-2
Single Step Genomics: Methods and Implementation in German Cattle Populations
Genomik und genomische Methoden haben entscheidend zum Züchtungsfortschritt vieler Pflanzen- und Nutztierpopulationen beigetragen. Der Einfluss auf die Fleischrinderpopulationen, insbesondere in Deutschland, war bisher jedoch begrenzt. Diese Dissertation konzentriert sich auf die Implementierung und Weiterentwicklung der Zuchtwertschätzung mittels der Single-Step Methode für deutsche Rinderpopulationen, wobei der Schwerpunkt auf den Fleischrinderrassen liegt.
Kapitel 2 enthält beschreibt das Manuskript mit dem Titel „Single-step SNPBLUP Evaluation in Six German Beef Cattle Breeds“, veröffentlicht im Journal of Animal Breeding and Genetics. Dieses Kapitel beschreibt die erstmalige Anwendung der Single-Step-SNPBLUP-Genomzuchtwertschätzung für sechs deutsche Fleischrinderrassen. Ziel war es, die Vorteile der Integration von Pedigree- und Genominformationen in ein gemeinsames Zuchtwertschätzsystem aufzuzeigen, dass eine simultane Zuchtwertschätzung aller Tiere ermöglicht, unabhängig davon, ob genotypische Daten vorliegen oder nicht. Dies verbessert die Genauigkeit und Effizienz der Zuchtwertschätzung erheblich. Seit der Erstimplementierung wurde die Methode für die Rassen Hereford und Salers erweitert und ist nun Teil des nationalen Zuchtwertschätzsystem in Deutschland. Kapitel 3 untersucht die Integration internationaler Zuchtwerte in nationale genomische Zuchtwertschätzungen. Im Manuskript „Impact of Deregressed Foreign Breeding Values on the Single-Step Genomic Evaluation of German Beef Cattle Populations“ werden Methoden zur Integration der Interbeef-Zuchtwerte entwickelt und evaluiert. Interbeef bietet international geschätzte Zuchtwerte (EBVs), die die Sicherheit nationaler Zuchtwerte durch die Einbeziehung von Verwandtschaftsinformationen aus mehreren Ländern erhöhen. Dieses Kapitel entwickelt und validiert Ansätze zur Kombination nationaler und internationaler Zuchtwerte, einschließlich skalaren und matrixbasierten Deregressionsmethoden für direkte und maternale genetische Effekte anhand der Zuchtwertschätzung für das Merkmal 200-Tage Gewicht. Kapitel 4 widmet sich der Identifizierung von genomischen Loci, die für phänotypische Merkmale und deren genetische Architektur von Bedeutung sind. Traditionelle GWAS-Ansätze sind zwar für einfache Merkmale effektiv, stoßen jedoch bei komplexen Merkmalen aufgrund von Linkage disequilibrium, polygenicity, und unvollständigen Genotyp-Phänotyp-Daten an ihre Grenzen. Dieses Kapitel stellt eine neuartige Methode im Rahmen von ssSNPBLUP vor, um die Unsicherheit von SNP-Effekten zu schätzen und Signifikanzschwellen festzulegen. Mit Datensätzen von Limousin-Fleischrindern und Holstein-Milchkühen aus Deutschland wird der Einfluss der Referenzpopulationsgröße auf die Signifikanz der SNPs untersucht und mit konventionellen GWAS-Methoden verglichen. Die Ergebnisse erweitern das Verständnis der SNP-Effekte in genomischen Zuchtwertschätzungen und verbessern die Anwendbarkeit von ssSNPBLUP auf kleine und große Datensätze.
Kapitel 5 analysiert die genetische Diversität deutscher und irischer Fleischrinderpopulationen im Kontext der genomischen Selektion. Während die genomische Selektion den Zuchtfortschritt beschleunigt, birgt sie das Risiko einer erhöhten Inzucht und einer Verringerung der genetischen Vielfalt, was eine nachhaltige Tierzucht gefährden kann. Dieses Kapitel bewertet die genomische Vielfalt von sechs Rassen – Angus, Blonde d’Aquitane, Charolais, Simmental, Limousin und Uckermärker – und liefert grundlegende Kennzahlen zur Überwachung der genetischen Diversität. Ziel ist es, Strategien zu entwickeln, die den genetischen Fortschritt mit dem Management von Inzucht in Einklang bringen.
Mit der Entwicklung und Anwendung von Single-Step-Genomik-Methoden leistet diese Dissertation einen bedeutenden Beitrag zur Verbesserung der Genauigkeit, Effizienz und Nachhaltigkeit der Zuchtwertschätzung in deutschen FleischrinderpopulationenGenomics and genomic technologies have significantly advanced the development of many plant and livestock populations. However, their impact on beef cattle populations, particularly in Germany, has been limited. This thesis focuses on the implementation and development of single-step genomic methodologies for German beef cattle breeds. Chapter 2 presents the paper titled “Single-step SNPBLUP Evaluation in Six German Beef Cattle Breeds,” published in the Journal of Animal Breeding and Genetics. This chapter details the first implementation of the single-step Single Nucleotide Polymorphism Best Linear Unbiased Prediction (ssSNPBLUP) genomic evaluation for six German beef cattle breeds. The primary aim is to demonstrate the advantages of integrating pedigree and genomic information into a single genomic evaluation framework, which enables simultaneous evaluation of all animals, regardless of the availability of genomic information. Single step approaches improve the accuracy and efficiency of genetic evaluations. Since the initial implementation for 6 beef cattle breeds, the method has been extended to include Hereford and Salers breeds, further expanding its relevance and utility in Germany's national genetic evaluation system. Building on this, Chapter 3 investigates the integration of international breeding values into national genomic evaluations. The manuscript, “Impact of Deregressed Foreign Breeding Values on the Single-Step Genomic Evaluation of German Beef Cattle Populations,” published in the Journal “Genetics Selection Evolution” explores methods to incorporate Interbeef evaluations, which provide internationally estimated breeding values (EBVs) into the national single step genomic evaluation of four beef cattle breeds. By blending international and national EBVs, this chapter addresses challenges in blending information derived from overlapping data sources while preserving valuable genetic information for increased reliability of breeding values. In this study, scalar and matrix deregression methods for direct and maternal genetic effects were developed and validated using forward validation, with the trait 200-day weight as a case study. Chapter 4 focuses on identifying genomic loci influencing phenotypes and their genetic architecture. Traditional single marker regression (Genome Wide Association Studies (GWAS)) methods, while effective for simple traits, face limitations with complex traits due to factors such as linkage disequilibrium, polygenicity, and incomplete genotype-phenotype records. To address these challenges, this chapter introduces a novel approach based on the ssSNPBLUP framework for estimating SNP effect reliability and defining significance thresholds for SNP markers in a genomic evaluation. Using datasets from German beef and dairy cattle, the method evaluates the influence of reference population size on SNP significance and compares results with conventional GWAS. This work enhances the applicability of ssSNPBLUP for both small and large datasets, bridging gaps in our understanding SNP effects in genomic evaluations. Finally, Chapter 5 examines the genetic diversity of German and Irish beef cattle populations under genomic selection. While genomic selection accelerates genetic gains, it also poses risks of increased inbreeding and reduced genetic diversity, which can hinder long-term sustainability. This chapter evaluates the genomic diversity of six breeds—Angus, Blonde d’Aquitane, Charolais, Simmental, Limousin, and Uckermärker—providing baseline metrics for monitoring diversity and informing strategies to balance genetic improvement with inbreeding management.
Through the development and application of single-step genomic methodologies, this thesis contributes to improving the accuracy, efficiency, and sustainability of genetic evaluations in German beef cattle populations.2026-01-1
Influence of Selective Androgen and Estrogen Receptor Modulators in Mono- and Combination Therapy on the Bone Substance of Osteopenic Male Rats
Die Osteoporose gehört zu den systemischen Skeletterkrankungen, bei der es zu einem
Verlust von Knochen kommt, welcher zu einer erhöhten Fragilität der Knochen mit Frak-
turneigung führt. Die Osteoporose wird von der WHO als Volkskrankheit bewertet und
zeigt auch in Deutschland zuletzt eine steigende Inzidenz und Prävalenz. Die häufigste
Form ist die postmenopausale Osteoporose. Allerdings zeigt auch die Osteoporose bei
Männern in den letzten Jahren eine zunehmende Tendenz. Ein wichtiger Grund ist das
zunehmende Alter der Gesellschaft und folglich der progrediente Verlust der Hodenfunk-
tion im höheren Alter. Studien, mit dem Ziel, medikamentöse Therapieoptionen bei Män-
nern mit Osteoporose zu erforschen, gibt es wenige. Da die altersassoziierte Osteoporose
beim Mann mit einem Testosteronmangel vergesellschaftet ist, lag die Idee nahe, diesen
durch Substitution auszugleichen. Einige Studien zeigten, dass dies effektiv zu einer Ver-
besserung der Knochenbeschaffenheit führt. Die Testosteronsubstitution führt jedoch oft
zu vielen unerwünschten Nebenwirkungen. Ein neuer Ansatz ist die Anwendung von se-
lektiven Androgenrezeptormodulatoren (SARM). Diese haben idealerweise eine gewebs-
selektive Wirkung auf den Knochen. Eine wichtige Erkenntnis der letzten Jahre war, dass
auch im männlichen Organismus Östrogene eine Rolle für die gesunde Knochenhomöo-
stase spielen. Der Östrogenmangel bei der postmenopausalen Osteoporose wird therapeu-
tisch schon durch den Einsatz selektiver Östrogenrezeptormodulatoren (SERM) adres-
siert. Zum Einsatz dieser Substanzen bei Frauen gibt es zahllose Studien, während der
Einsatz bei Männern wenig untersucht wurde. Ziel unserer Studie war es, den Einfluss
einer Kombinationstherapie aus dem SARM Enobosarm und dem SERM Raloxifen auf
den männlichen Knochenstoffwechsel zu untersuchen. Gleichzeitig erfolgte ein Ver-
gleich mit den beiden Substanzen in Einzeltherapie. In dieser Studie untersuchten wir, ob
die genutzten Medikamente die Ausbildung einer Osteoporose verhindern können und
somit prophylaktisch eingesetzt werden könnten.
In unserer Studie induzierten wir mit Hilfe der Orchiektomie einen Testosteronmangel,
welcher bei den Ratten konsekutiv zur Ausbildung einer Osteoporose führte. Hierfür wur-
den jeweils fünfzehn acht Monate alte, männliche Ratten in fünf Versuchsgruppen einge-
teilt. Bei vier Gruppen erfolgte die Orchiektomie zur Induktion einer Androgenmangel-
assoziierten Osteoporose. Eine Gruppe wurde nicht orchiektomiert und diente als gesunde
Kontrollgruppe (NON-ORX), während eine orchiektomierte Gruppe (ORX) als kranke
Kontrollgruppe vorgesehen war. Drei der orchiektomierten Gruppen wurden prophylak-
tisch medikamentös behandelt. Die EN-Gruppe wurde mit Enobosarm behandelt
(0,4 mg/kg Körpergewicht). Die RLX-Gruppe erhielt Raloxifen (0,7 mg/kg Körperge-
wicht) und eine Gruppe erhielt Enobosarm (0,4 mg/kg Körpergewicht) kombiniert mit
Raloxifen (0,7 mg/kg Körpergewicht). Direkt nach Orchiektomie erfolgte die prophylak-
tische Gabe der Substanzen mit dem sojafreien Futter. Die Gabe wurde über 18 Wochen

fortgesetzt. Anschließend erfolgte die Tötung der Versuchstiere und die Gewinnung der
Versuchsproben. Der vierte Lendenwirbelkörper und das rechte Femur wurden mittels
Mikrocomputertomographie, biomechanischer Testung und einem Veraschungsversuch
untersucht. Weiter wurden im Serum der Ratten die Knochenumbauparameter erhoben.
Die statistische Analyse wurde mit einer one-way-ANOVA und einem Tukey-Test durch-
geführt.
Die kombinierte Behandlung aus Raloxifen und Enobosarm führte zur deutlichsten Ver-
besserung der Knochenparameter. Die Effekte waren ausgeprägter als unter den jeweili-
gen Monotherapien. Die Knochenumbauparameter zeigten sich signifikant verbessert.
Bei der RLX+EN-Gruppe kam es auch zu einer deutlichen Gewichtsabnahme der Tiere.
Die kombinierte Therapie führt wahrscheinlich zu einer Abnahme des viszeralen Fettge-
webes.
Enobosarm zeigte eine partielle Verbesserung der Mikroarchitektur, während wir keinen
Effekt auf die biomechanischen Eigenschaften, oder die Knochenumbauparameter mes-
sen konnten. Die Körpergewichte der Tiere blieben ohne signifikante Änderungen.
Unter der Behandlung mit Raloxifen zeigten sich die biomechanischen und trabekulären
Parameter gegenüber der ORX-Gruppe verbessert. Die Knochenumbauparameter blieben
ohne nennenswerte Änderung. Weiter kam es zu einer Körpergewichtsabnahme in der
RLX-Gruppe. Diese ist bei Ratten bereits vorbeschrieben worden und resultierte aus einer
verminderten Nahrungsaufnahme.
In dieser Studie konnten wir zum ersten Mal einen synergistischen positiven Effekt einer
Kombination aus dem osteoanabolen Enobosarm und dem antiresorptiven Raloxifen auf
das Knochengewebe orchiektomierter Ratten nachweisen.
Die meisten der bestimmten Knochenparameter blieben unter der Kombinationstherapie
erhalten, oder zeigten sich sogar verbessert gegenüber der gesunden Kontrollgruppe, was
ein Hinweis auf einen positiven Einfluss auf den altersbedingten Knochenverlust sein
könnte. Die Monotherapien führten ebenfalls zu einer partiellen Verbesserung der unter-
suchten Parameter, blieben aber insgesamt in ihrem Effekt hinter der Kombinationsthe-
rapie zurück. Die von uns genutzte Kombinationstherapie stellt möglicherweise eine prä-
ventive Behandlungsoption für die altersbedingte Osteoporose bei Männern dar. Neben-
effekte und weitere Kombinationen mit anderen Vertretern der genutzten Substanzklas-
sen sollten allerdings weiter untersucht werden.Osteoporosis is one of the systemic skeletal diseases characterized by a loss of bone mass, leading to increased bone fragility and susceptibility to fractures. The World Health Organization (WHO) classifies osteoporosis as a widespread disease, and its incidence and prevalence have been rising in Germany in recent years. The most common form is postmenopausal osteoporosis. However, osteoporosis in men has also shown an increasing trend in recent years. One important reason for this is the aging population and the consequent progressive loss of testicular function in older age. There are few studies aimed at exploring pharmacological treatment options for men with osteoporosis. Since age-related osteoporosis in men is associated with testosterone deficiency, the idea emerged to address this deficiency through substitution therapy. Some studies have shown that this can effectively improve bone quality. However, testosterone substitution often results in numerous undesirable side effects.
A novel approach is the application of selective androgen receptor modulators (SARMs), which ideally have tissue-selective effects on bone. An important finding in recent years has been that estrogens also play a role in maintaining healthy bone homeostasis in the male body. Estrogen deficiency in postmenopausal osteoporosis is already therapeutically addressed through the use of selective estrogen receptor modulators (SERMs). There are numerous studies on the use of these substances in women, but their application in men has been scarcely investigated.
The aim of our study was to examine the influence of a combination therapy of the SARM enobosarm and the SERM raloxifene on male bone metabolism. Additionally, we compared the combination therapy with the individual therapies using each substance. This study investigated whether the medications used could prevent the development of osteoporosis and thus be applied prophylactically.
In our study, testosterone deficiency was induced through orchiectomy, which led to the development of osteoporosis in the rats. Fifteen eight-month-old male rats were divided into five experimental groups. In four groups, orchiectomy was performed to induce androgen deficiency-associated osteoporosis. One group was not orchiectomized and served as the healthy control group (NON-ORX), while one orchiectomized group (ORX) served as the diseased control group. Three of the orchiectomized groups received prophylactic pharmacological treatment. The EN group was treated with enobosarm (0.4 mg/kg body weight). The RLX group received raloxifene (0.7 mg/kg body weight), and one group received a combination of enobosarm (0.4 mg/kg body weight) and raloxifene (0.7 mg/kg body weight). Prophylactic administration of the substances began immediately after orchiectomy using soy-free feed and continued for 18 weeks. Subsequently, the animals were euthanized, and samples were collected. The fourth lumbar vertebra and the right femur were analyzed using micro-computed tomography, biomechanical testing, and ashing experiments. Additionally, bone turnover markers were measured in the serum of the rats. Statistical analysis was performed using a one-way ANOVA and Tukey's test.
The combined treatment with raloxifene and enobosarm resulted in the most significant improvement in bone parameters, with effects being more pronounced than with the respective monotherapies. Bone turnover markers showed significant improvement. In the RLX+EN group, there was also a marked reduction in the animals' body weight. The combined therapy likely led to a decrease in visceral fat tissue.
Enobosarm showed partial improvement in microarchitecture, but no effect on biomechanical properties or bone turnover markers was observed. The body weights of the animals remained unchanged.
Treatment with raloxifene improved biomechanical and trabecular parameters compared to the ORX group. However, bone turnover markers showed no significant changes. Additionally, a reduction in body weight was observed in the RLX group. This has been previously described in rats and is attributed to reduced food intake.
In this study, we demonstrated for the first time a synergistic positive effect of a combination of the osteoanabolic enobosarm and the antiresorptive raloxifene on the bone tissue of orchiectomized rats.
Most of the measured bone parameters were preserved or even improved under combination therapy compared to the healthy control group, suggesting a positive influence on age-related bone loss. The monotherapies also led to partial improvement of the investigated parameters but were overall less effective than the combination therapy. The combination therapy we used may represent a preventive treatment option for age-related osteoporosis in men. However, side effects and additional combinations with other representatives of the utilized substance classes should be further investigated.2025-02-2
Cortical PV and VIP neurons are necessary for texture discrimination performance of freely behaving mice
Rodents gather information about their surroundings through their whiskers, enabling them to differentiate textures, recognize objects, and navigate. Each whisker is represented in the barrel cortex (S1BF) by a single cortical column in an isomorphic manner. The microcircuits within each barrel-related column comprise both excitatory and inhibitory neurons. Extensive literature exists on genetically defined GABAergic neurons—parvalbumin (PV), vasoactive intestinal polypeptide (VIP), and somatostatin (SST)—and their involvement in tactile perception through whisking, including both passive stimulation of the whiskers and active touch. S1BF is well established as the core node for whisker-based texture discrimination. However, how PV, VIP, and SST interneurons contribute to the behavioral outcome of information processing during tactile perception in texture discrimination remains unclear.
This study aims to further investigate the role of PV and VIP neurons in S1BF through chemogenetic activation in awake, freely behaving mice trained in a texture discrimination task. For this purpose, a novel whisker-based texture discrimination paradigm, the textured T-maze (TT), was developed, allowing mice to explore the maze freely while being trained on texture discrimination. PV-cre and VIP-cre mice exhibited comparable baseline performance. An activating Cre-dependent chemogenetic virus was bilaterally injected into the S1BF of PV-cre and VIP-cre mice to express “activating” DREADDs, enabling selective manipulation of PV and VIP cells via intraperitoneal administration of clozapine-N-oxide (CNO). Chemogenetic activation of either PV or VIP neurons impaired texture discrimination, reducing performance to chance level. Whole-cell patch-clamp experiments demonstrated that the intrinsic properties of transfected cells were comparable to those of non-transfected counterparts. The depolarizing effect of CNO on transfected cells was tested via bath application. Recordings confirmed that PV cells exhibited depolarization as expected from the viral construct, whereas VIP cells underwent depolarization block.
In conclusion, although the chemogenetic virus did not produce equivalent depolarization effects in PV and VIP cells, the activation of PV cells or the putative deactivation of VIP cells in S1BF disrupted perceptual discrimination in freely behaving mice during the texture discrimination task.2025-04-0
NTRK alterations in non-small cell lung cancer
Alterationen, insbesondere Genfusionen, der NTRK-Genfamilie, bestehend aus NTRK1, NTRK2 und NTRK3, stellen einen seltenen, aber therapeutisch potentiell adressierbaren Treiber in nicht-kleinzelligen Lungenkarzinomen dar. Durch Alterationen bedingte konstitutive Aktivierung oder Überexpression der durch sie kodierten Tropomyosin-related-Kinasen (respektive TrkA, TrkB und TrkC) können deren physiologische Effekte wie Zellproliferation und Apoptosehemmung zu einem unkontrollierten Zellwachstum und damit zur Kanzerogenese führen. Die niedrige Prävalenz der therapeutisch adressierbaren NTRK-Genfusionen bedingt die Herausforderung der Molekularpathologie, dieses seltene Phänomen zuverlässig zu erkennen, um den entsprechenden Patient*innen die hocheffektive Therapie mit einem spezifischen Tyrosinkinaseinhibitor zukommen zu lassen. Alterationen anderer Art wie Spleißvarianten, Amplifikationen oder Punktmutationen sind ebenfalls beschrieben, in ihrer Expression und ihrem Effekt auf die Onkogenese jedoch nicht eindeutig klassifiziert. Für diese Arbeit wurde eine Kohorte von 1.080 konsekutiven, nicht vorselektierten Fälle nicht-kleinzelliger Lungenkarzinome, die zwischen Dezember 2016 und Juni 2018 im Institut für Pathologie der Universitätsmedizin Göttingen bearbeitet wurden, mittels unterschiedlicher Methoden auf NTRK-Alterationen untersucht. Der Großteil der Fälle erhielt zunächst eine immunhistochemische Testung auf eine Trk-Expression, die bei Positivität durch eine Testung der Ursache mit RNA-basierter next generation-Sequenzierung und/ oder Fluoreszenz-in situ-Hybridisierung ergänzt wurde. Auch einige immunhistochemisch negative Fälle wurden zur Vergleichbarkeit mit diesen Methoden untersucht. Neben der vorausgegangenen Klassifizierung der Immunhistochemie in „positiv“ und „negativ“ wurden Intensität und subzelluläre Lokalisation der Färbung im Rahmen dieser Arbeit mittels eines Scores beurteilt und sortiert. Ein anderer Teil der Fälle wurde als einzige Methode per Fluoreszenz-in situ-Hybridisierung untersucht. In 23 Fällen (2,1%) konnte eine Alteration jeglicher Art von NTRK1, -2- oder -3 ermittelt werden. Genfusionen traten mit zwei Fällen zu 0,19% auf: Eine SQSTM1-NTRK1- und eine EPS15-NTRK1-Fusion wurden in zwei Adenokarzinomen männlicher Patienten identifiziert und ihre Träger einer Tyrosinkinaseinhibitortherapie zugeführt. Immunhistochemisch zeigten sich beide Fälle stark positiv (Rang 1 und 7 unter 169 positiven Fällen) und wiesen in anderen prominenten Treibern (EGFR, KRAS, ALK, ROS1, RET, MET) keine Mutationen auf, was die NTRK-Fusion als primären Treiber wahrscheinlich macht. Zehn Fälle wiesen in der Fluoreszenz-in situ-Hybridisierung eine Vermehrung der Kopienzahl (über vier) eines oder mehrerer NTRK-Gene auf, die zu 90% mit einer immunhistochemischen Positivität einherging. In insgesamt zwölf Fällen konnten alternative Spleißvarianten der drei Trk-Proteine nachgewiesen werden, darunter sieben Fälle eines Exon 12-skippings (zu 85,7% immunhistochemisch positiv) und drei Fälle eines Exon 10-skippings (100% immunhistochemisch positiv) jeweils von NTRK2. Der Krankheitswert der Spleißvarianten und Genkopienzahlerhöhungen lässt sich mit den vorliegenden Daten nicht eindeutig bestimmen und ist auch im Literaturkontext Gegenstand von Diskussionen. Während sich die alleinige Testung auf Translokationen per Fluoreszenz-in situ-Hybridisierung als zu zeit- und materialaufwändig in einer Kohorte mit so niedriger Prävalenz zeigte, erwies sich ein Vorgehen mit primärer Immunhistochemie und sequentieller RNA-basierter next generation-Sequenzierung als zuverlässig, um die vorliegenden NTRK-Fusionen und -Spleißvarianten zu erkennen. Für das Ermitteln von Amplifikationen war die FISH eine geeignetere, wenn auch nicht ideale Methode. Im Kontext der aktuellsten Literatur wird für NTRK-Alterationen in nicht-kleinzelligen Lungenkarzinomen eine RNA-basierte Nachweismethode favorisiert. Spleißvarianten und Amplifikationen von NTRK sollten als potentiell adressierbare Mutationen näher auf ihr onkogenes Potenzial und therapeutische Adressierbarkeit untersucht werden.Alterations, especially gene fusions, of the NTRK gene family, consisting of NTRK1, NTRK2 and NTRK3, represent a rare, but therapeutically potentially addressable driver in non-small cell lung carcinomas. Constitutive activation or overexpression caused by alterations of the tropomyosin-related kinases encoded by them (respectively TrkA, TrkB, and TrkC) can lead to uncontrolled cell growth and thus to carcinogenesis due to their physiological effects such as cell proliferation and apoptosis inhibition. The low prevalence of therapeutically addressable NTRK gene fusions makes it challenging for molecular pathology to reliably detect this rare phenomenon in order to offer the corresponding patients the highly effective therapy with a specific tyrosine kinase inhibitor. Alterations of other kinds, such as splice variants, amplifications, or point mutations, are also described, but their expression and effect on oncogenesis are not clearly classified.
For this study, a cohort of 1080 consecutive, non-preselected cases of non-small cell lung carcinomas that were treated at the Institute of Pathology at the University of Göttingen between December 2016 and June 2018, were examined for NTRK alterations using different methods. Most of the cases initially underwent immunohistochemical testing for Trk expression, which, if positive, was supplemented by testing the cause with RNA-based next generation sequencing and/or fluorescence in situ hybridization. Some immunohistochemically negative cases were also examined using these methods for comparability. In addition to the prior classification of immunohistochemistry into "positive" and "negative", the intensity and subcellular location of the staining were evaluated and sorted using a score in the context of this work. Another part of the cases was examined by fluorescence in situ hybridization as the only method. An alteration of any kind of NTRK1, -2, or -3 was determined in 23 cases (2.1%). Gene fusions occurred in two cases at 0.19 %: A SQSTM1-NTRK1 and EPS15-NTRK1 fusion was identified in two adenocarcinomas in male patients and their carriers were given tyrosine kinase inhibitor therapy. Immunohistochemically, both cases showed strongly positive (rank 1 and 7 among 169 positive cases) and did not show mutations in other prominent drivers (EGFR, KRAS, ALK, ROS1, RET, MET), which likely makes the NTRK fusion the primary driver. Ten cases showed an increase in copy number (above four) of one or more NTRK genes in fluorescence in situ hybridization, which was associated with immunohistochemical positivity in 90 %. In a total of twelve cases, alternative splice variants of the three Trk proteins could be detected, including seven cases of exon 12 skipping (85.7% immunohistochemically positive) and three cases of exon 10 skipping (100% immunohistochemically positive) each of NTRK2. The disease value of splice variants and increased gene copy numbers cannot be determined conclusively with the available data and is also a subject of discussion in the literature. While testing only for translocations by fluorescence in situ hybridization proved to be too time-consuming and material-consuming in a cohort with such low prevalence, an approach with primary immunohistochemistry and sequential RNA-based next generation sequencing proved to be reliable in detecting the present NTRK fusions and splice variants. For the detection of amplifications, FISH was a more suitable, if not ideal method. In the context of the latest literature, an RNA-based detection method is favored for NTRK alterations in non-small cell lung carcinomas. Splice variants and amplifications of NTRK should be closely examined for their oncogenic potential and therapeutic addressability as potentially addressable mutations.2025-04-2
Evaluation of treatment with KAN0438757 in combination with different chemotherapies in the glycolytic and lipogenic subtypes of pancreatic ductal adenocarcinoma
Despite current treatment options, the 5-year survival rate for patients with pancreatic ductal adenocarcinoma is below 10%.
This highlights the relevance of new therapeutic approaches. Pancreatic ductal adenocarcinoma, which shows metabolic dependence, can be divided into different tumor groups, of which the glycolytic subtype has a more aggressive phenotype with shorter survival.
The aim of the project was to combine and evaluate the glycolytic inhibitor KAN0438757 with the chemotherapies gemcitabine, 5-FU, FOLFIRINOX, and gemcitabine + paclitaxel. Both the cell viability and colony formation assays showed that the combination treatment of all tested chemotherapies with KAN0438757 significantly reduced the viability and clonogenic capacity of the pancreatic cancer cell lines HPAF-II and MIA PaCa-2. The reduction in cell viability and clonogenic capacity was more pronounced in the more aggressive cell line MIA PaCa-2, as it belongs to the glycolytic subtype.
As the influence of the combination treatment on the marker pH2AX could not be demonstrated by Western blotting, the project leaves open how the significant results of the combination of chemotherapy and KAN0438757 in the viability and colony formation assays can be explained at protein level.
In summary, the data with significant effects of the combination treatment of chemotherapy with KAN0438757 show promising new therapeutic approaches for targeted treatment of ductal pancreatic carcinoma, depending on the metabolic subtype.2025-06-0
The effect of 8-prenylnaringenin and alendronate in combination with vertical whole-body vibration on the vertebral body of the osteopenic rat model
Die Osteoporose ist eine systemische Erkrankung des Skeletts mit verminderter
Knochenstabilität und erhöhtem Frakturrisiko. Aufgrund steigender Osteoporose-bedingter
Gesundheitskosten stellen die Erkrankung und ihre Therapie ein zunehmendes
gesundheitsökonomisches Problem der modernen Gesellschaft dar und rücken daher
vermehrt in den Fokus des wissenschaftlichen und öffentlichen Interesses. Insbesondere die
postmenopausale Östrogenmangel-induzierte Osteoporose steht im Mittelpunkt vieler
Projekte. Vertikale Ganzkörpervibration (WBVV) hat bei verschiedenen Frequenzen in
klinischen und experimentellen Studien eine positive Wirkung auf das Knochengewebe
gezeigt. Alendronat (4-Amino-1-Hydroxybutyliden-1,1-Bisphosphonat; AL) ist ein
zugelassenes Antiosteoporose-Medikament. 8-PN ist der stärkste Agonist unter den
Phytohormonen am Östrogenrezeptor α, der in früheren Studien eine signifikante
Verbesserung der Knochenmineraldichte (bone mineral density, BMD) bewirkte. Ziel dieser
Studie war es, mögliche synergistische Effekte von AL und 8-PN in Kombination mit
WBVV auf Wirbelkörper von ovariekomierten (OVX) Ratten als Modell für
postmenopausale Osteoporose zu untersuchen.
Drei-Monate alte weibliche Ratten (ngesamt =90) wurden in sieben Gruppen eingeteilt: (i)
NON-OVX, (ii) OVX, (iii) OVX+WBVV, (iv) OVX+AL, (v) OVX+AL+WBVV, (vi)
OVX+8-PN, (vii) OVX+8-PN+WBVV. OVX wurde in den Gruppen ii-vii durchgeführt.
Die WBVV in den Gruppen iii, v und vii (35 Hz, 0,5 mm) begann am Tag 35 nach der OVX,
fünfmal pro Woche, zweimal pro Tag für 15 min. AL wurde ab dem 35. Tag nach der OVX
in den Gruppen iv und v in einer Konzentration von 10 mg/kg Futter im Pelletfutter
verabreicht, was zu einer durchschnittlichen täglichen Dosis von 0,58 mg/kg Körpergewicht
(KG) führte. Subkutane Injektionen von 1,77 mg/kg KG des Phytohormons 8-PN wurden
ab dem 35. Tag nach OVX in den Gruppen vi und vii appliziert. Die Behandlungsdauer
betrug zehn Wochen. Am Ende des Versuchs wurden Blutproben entnommen und die
Lendenwirbelkörper (LWK I-IV) präpariert. Es wurde die Serumaktivität der alkalischen
Phosphatase (AP) bestimmt. Im Knochen wurden Genexpressionsanalysen für die
Alkalische Phosphatase (AP), Osteocalcin (OC), Rezeptor Aktivator des Kernfaktors (nuclear
factor)-κB Ligand (RANKL), Osteoprotegerin (OPG) und die Tartrat-resistente saure
Phosphatase (TRAP) durchgeführt. Außerdem wurden nach der Veraschung der Calciumund
Phosphatanteil bestimmt und die organische und anorganische Substanz gemessen. Die
Analysen der Knochenmineraldichte (bone mineral density, BMD), der Kortikalis und des
Trabekelnetzwerks wurden mittels Mikrocomputertomographie (mCT) durchgeführt. Die
Knochenqualität wurde durch biomechanische Tests bewertet.
Der Erfolg der OVX zeigte sich in der vorliegenden Untersuchung in einer Reduktion des
Uterusgewichtes und des anorganischen Knochenanteiles. Die biomechanischen
Eigenschaften des Knochens und die Zusammensetzung des Knochens hinsichtlich des
Calciumanteils wurden durch die OVX jedoch nicht beeinträchtigt. Auf die Aktivität der AP
und die Genexpression der untersuchten Knochenmarker hatte die OVX in der vorliegenden
Untersuchung keinen nachweisbaren Effekt. Anhand der mCT-Analyse konnte gezeigt
werden, dass das trabekuläre Netzwerk und die Kortikalis durch die OVX nicht beeinträchtigt
wurden, obwohl die BMD signifikant reduziert war.
Im Rahmen einer wirksamen Osteoporosetherapie sollte ein positiver Effekt auf den
Knochenumsatz nachweisbar sein und die biomechanischen Eigenschaften des Knochens
optimiert werden. Dies konnte in der vorliegenden Arbeit weder für die Einzelbehandlungen
mit WBVV, AL und 8-PN noch für die Kombinationsbehandlungen von AL+WBVV und
8-PN+WBVV gezeigt werden. Die Anwendung von WBVV reduzierte die BMD und
erhöhte AP im Serum. Der Einsatz von AL erhöhte zwar den Calciumanteil, hatte aber
keinen Effekt auf biomechanischen und strukturellen Eigenschaften der OVX-Ratten. Die
Kombination aus AL+WBVV erhöhte die OC-Genexpression und AP im Serum, was auf
einen positiven Einfluss auf die Osteoblasten hinweist, hatte aber ansonsten keine positive
Auswirkung auf die Knocheneigenschaften des LWKs im Vergleich zu AL allein. 8-PN
wurde den OVX-Ratten in der vorliegenden Arbeit in einer nicht-karzinogenen Dosis
verabreicht und zeigte konsekutiv keinen Anstieg des Uterusgewichtes, führte jedoch auch
zu keiner signifikanten Verbesserung der Knochenqualität. Die Kombination von 8-
PN+WBVV führte zur Reduktion des TV und einem Anstieg des BV/TV-Verhältnisses,
aber zu keiner Steigerung der BMD.
Analysen der Skelettmuskulatur im Rahmen anderer Untersuchungen zeigten eine Zunahme
der Muskelfasergröße durch WBVV in diesem Tierversuch. AL und 8-PN erhöhten die
Kapillardichte, was zu einer verbesserten Muskeldurchblutung führte und sich letztlich
positiv auf die Knochenstruktur auswirken könnte.
Osteoporose wird auch in Zukunft ein medizinisch relevantes Forschungsgebiet bleiben und
sollte weiterhin Gegenstand randomisierter Studien sein. Für zukünftige Studien wäre eine
Kombination von 8-PN und AL interessant. Weiterhin könnten mögliche synergistische
Effekte von Sclerostininhibitoren in Kombination mit AL, 8-PN oder WBVV untersucht
werden.Osteoporosis is a systemic disease of the skeleton with reduced bone stability and increased
fracture risk. Due to rising osteoporosis-related healthcare costs, the disease and its treatment
represent an increasing health economic problem in modern society and are therefore
increasingly becoming the focus of scientific and public interest. In particular, postmenopausal
oestrogen deficiency-induced osteoporosis is at the centre of many projects.
Whole-body vertical vibration (WBVV) has shown a positive effect on bone tissue at various
frequencies in clinical and experimental studies. Alendronate (AL) is an authorised antiosteoporosis
drug. 8-Prenylnaringenin (8-PN) is the strongest agonist among the
phytohormones at the estrogen receptor α, which caused a significant improvement in bone
mineral density (BMD) in previous studies. The aim of this study was to investigate possible
synergistic effects of AL and 8-PN in combination with WBVV on vertebral bodies of
ovariectomised (OVX) rats as a model for postmenopausal osteoporosis. WBVV at different
frequencies showed a positive effect on the biomechanical properties of the osteopenic rat.
Previous studies also have shown that alendronate (AL) has a positive effect on the
biomechanical properties of the vertebral bones of OVX rats. 8-Prenylnaringenin (8-PN),
the strongest agonist among the phytohormones at the oestrogen receptor α, showed a
significant improvement in BMD in previous studies.
The aim of the present study was to investigate synergistic effects of the combination of
AL+WBVV and the combination of 8-PN+WBVV on the lumbar spine in the osteopenic
rat model.
Three-month-old female rats (ntotal =90) were divided into seven groups: (i) NON-OVX, (ii)
OVX, (iii) OVX+WBVV, (iv) OVX+AL, (v) OVX+AL+WBVV, (vi) OVX+8-PN, (vii)
OVX+8-PN+WBVV. OVX was performed in groups ii-vii. WBVV in groups iii, v and vii
(35 Hz, 0.5 mm) started on day 35 after OVX, five times per week, two times per day for 15
min. AL was administered from day 35 after OVX in groups iv and v at a concentration of
10 mg/kg feed in the pellet diet, resulting in an average daily dose of 0.58 mg/kg body weight
(bw). Subcutaneous injections of 1.77 mg/kg body weight of the phytohormone 8-PN were
administered from the 35th day after OVX in groups vi and vii. The duration of treatment
was ten weeks. At the end of the trial, blood samples were taken and the lumbar vertebral
bodies (LWK I-IV) were dissected. Serum alkaline phosphatase (AP) activity was determined.
In bone, gene expression analyses were performed for alkaline phosphatase (AP),
Osteocalcin (OC), receptor activator of nuclear factor-κB ligand (RANKL), Osteoprotegerin
(OPG) and tartrate-resistant acid phosphatase (TRAP). In addition, the calcium and
phosphate content was determined after ashing and the organic and inorganic substance was
measured. Analyses of bone mineral density (BMD), cortical bone and the trabecular network
were carried out using micro-computed tomography (mCT). Bone quality was assessed by
biomechanical tests.
Female rats (ntotal =90) were divided into seven groups: (i) NON-OVX, (ii) OVX, (iii)
OVX+WBVV, (iv) OVX+AL, (v) OVX+AL+WBVV, (vi) OVX+8-PN, (vii) OVX+8-

Ovariectomy (OVX) was performed in groups ii-vii. Bilateral osteotomy of the tibial
metaphysis with osteosynthesis was performed in all groups on day 63 post OVX. WBVV
in groups iii, v and vii (35 Hz /amplitude 0.5 mm) started on day 35 post OVX, five times
/week, twice/day, 15 min for ten weeks. AL in the pellet feed from day 35 post OVX
onwards in groups iv and v resulted in an average daily dose of 0.58 mg/kg bw. Subcutaneous
injections of 1.77 mg/kg body weight of the phytohormone 8-PN from day 35 post OVX
in groups vi and vii. The animals were anaesthetised with CO2 110 days after OVX and
euthanised by decapitation. Blood samples were taken and the lumbar vertebral bodies (LWK
I-IV) were dissected. The blood samples were subjected to serum alkaline phosphatase (AP)
analysis. Gene expression analyses for AP, OC, RANKL, OPG and TRAP were performed
on the bone samples. Furthermore, the calcium and phosphate content was determined after
ashing, and the organic and non-organic matter was measured. Biomechanical tests and
analyses of the BMD, the cortical bone and the trabecular network were carried out using
three-dimensional reconstructions of microtomographies.
In the present study, the success of OVX was reflected in a reduction in uterine weight and
the inorganic bone content. However, the biomechanical properties of the bone and the
composition of the bone in terms of calcium content were not affected by OVX. In the
present study, OVX had no detectable effect on the activity of the AP and the gene
expression of the bone markers analysed. Micro-CT analysis showed that the trabecular
network and cortical bone were not affected by OVX, although BMD was significantly
reduced.
As part of an effective osteoporosis therapy, a positive effect on bone turnover should be
demonstrable and the biomechanical properties of the bone should be optimised. This could
not be demonstrated in the present study either for the individual treatments with WBVV,
AL and 8-PN or for the combination treatments of AL+WBVV and 8-PN+WBVV. The use
of WBVV reduced BMD and increased AP in serum. The use of AL increased the calcium
content but had no effect on the biomechanical and structural properties of the OVX rats.
The combination of AL+WBVV increased OC gene expression and AP in serum, indicating
a positive effect on osteoblasts, but otherwise had no positive effect on the bone properties
of the LWK compared to AL alone. 8-PN was administered to the OVX rats in the present
study at a non-carcinogenic dose and consecutively showed no increase in uterine weight,
but also did not lead to a significant improvement in bone quality. The combination of 8-
PN+WBVV led to a reduction in TV and an increase in the BV/TV ratio, but no increase
in BMD. Analyses of skeletal muscle in other studies showed an increase in muscle fibre size
with WBVV in this animal study. AL and 8-PN increased capillary density, which led to
improved muscle perfusion and could ultimately have a positive effect on bone structure.
Osteoporosis will remain a medically relevant area of research in the future and should
continue to be the subject of randomised trials. A combination of 8-PN and AL would be
interesting for future studies. Furthermore, possible synergistic effects of sclerostin inhibitors
in combination with AL, 8-PN or WBVV could be investigated.2025-05-2
