Phytochemical screening, total flavonoid and phenolic content assays of various solvent extracts of musa sapientum

    No full text
Flavonoids represent the most common and widely distributed group of plant phenolics and are abundant in foods; quercetin and rutin are the flavonoids most abundantly consumed. Musa sapientum is one of the well-known plants of the Musaceae family that have been used in traditional medicine since hundred years to alleviate various diseases and health problems. The objective of this research is to conduct the preliminary phytochemical screening, total flavonoid and phenolic contents assays of various solvent extracts of tepal of Musa sapientum. Phytochemical screening was carried out according to the method of Trease and Evans, total flavonoid content was measured by the aluminium chloride colorimetric assay and total phenolic content was estimated spectrophotometrically by FolinCiocalteau method. Preliminary phytochemical screening reveals the presence of phenolics, flavonoids, alkaloids, tannins, terpenoids in all the three different extracts (methanolic, ethanolic and aqueous). Polar solvents are frequently used for recovering polyphenols from plant matrices. The most suitable solvents are aqueous mixtures containing ethanol, methanol, acetone, and ethyl acetate. Tepal methanolic extract has the richest content of both phenolics and flavonoids i.e. (5.17 mg GAE/g and 0.36 mg QE/g) respectively, and aqueous extract was the least i.e. (2.02 mg GAE/g and 0.214 mg QE/g). All the extracts were not significantly different with one another (p>0.05). It can be hypothesised that the high contents of phenolic compounds of tepals of Musa sapientum indicated that these compounds contribute to the antioxidant activity and can be regarded as promising plant species for natural sources of radical scavenging activity with potential value for treatment of many life threatening diseases. The process of extraction and identification of active principles responsible for the free radical scavenging property of tepal extract of Musa sapientum through bioactivity guided fraction is under progress in order to understand the possible mechanism of action. Utilization of this tepal will be of advantage to mankind and increased in its consumption will help in prevention of chronic life style diseases

Thymoquinone effects on dopamine level and lactate dehydrogenase activity in differentiated SH-SY5Y cells exposed to amphetamine

Aims 4/ Background: Amphetamine (AT) is used to treat some medical conditions and also is highly abused. It is a potent central nervous system stimulant that is capable of producing damaging effects to the central dopaminergic pathway. Thymoquinone (TQ), an active compound of Nigella sativa is known for its cellular protective effects. The objectives of this study were to determine the ICso value of AT and TQ on differentiated 51-1-5\15Y neuronal cells and to evaluate the changes of dopamine (DA) levels and lactate dehydrogenase (LDH) activity in the cells exposed to AT after co-administering with TQ. Methodology: Differentiated 511-1-5Y5Y cells were grown in cell culture flask containing DM EM/F12 with 10% (v/v) fetal bovine serum and 1% (w/v) penicillin/streptomycin. Cell viability was counted to determine the IC50 value of AT and TQ. Serially diluted solutions of AT (7396, 3698, 1849, 925, 462, 231, 116 and 0 pM) and TQ (6090, 3045, 1523, 761, 381, 190, 95 and 0 OA) were added to the cells and incubated for 72 hours.Subsequently, 20 pi of 3-(4,5-dimethylthiazol-2-y1)2,5- diphenyltetrazolium bromide and 100 pi of DM50 were added. The DA levels and LDH activity were determined by using the ELISA and LDH activity assay kits respectively. Results: The ICso values of AT and TQ were determined to be 1596 OA and 926 pM respectively. Co-administration of 30 phi of TQ and 37 of AT demonstrated a significant increase in DA levels when compared to administration of AT alone (P5.0.05). There was also a significant decrease in LDH activity in the cells exposed to both AT and TQ compared to administration of AT alone (iPLO.001).Conclusion: These findings suggested that TQ has a role in maintaining the DA activity and also possesses a neuroprotective effect against cell damage after long term AT exposure

Complexity approximation of classification task for large dataset ensemble artificial neural networks

    No full text
In this paper, operational and complexity analysis model for ensemble Artificial Neural Networks (ANN) multiple classifiers are investigated. The main idea behind this, is lie on large dataset classification complexity and burden are to be moderated by using partitioning for parallel tasks and combining them to enhance the capability of a classifier. The complexity of the single ANN and ensemble ANN are obtained from the estimates of upper bounds of converged functional error with the partitioning of dataset. The estimates derived using Apriori method shows that the use of an ensemble ANN with different approach is feasible where such problem with a high number of inputs and classes can be solved with time complexity of O(n ) for some k, which is a type of polynomial. This result is in line with the importance of good performance achieved by diversity rule applied with the use of reordering technique. As a conclusion, an ensemble heterogeneous ANN classifier is practical and relevance to theoretical and experimental of combiners for ensemble ANN classifier systems for large dataset

Meta-Model Interaction for Collaborative School Subject Management

    No full text
Subject management is process of developing and coordinating of teaching materials. A collaborative subject module management is used to allow participants to share and manage course modules via Internet technology. Currently, Malaysian school does not have collaborative platform to manage subject materials. Teachers usually used personalized tools to manage their subject modules. This article will discuss the modelling of cloud-based subject module management. Raquel's interaction meta-model is adopted as the central framework of the model. Based on developed model, a prototype has been develop which implemented under cloud computing platform. In the experiment, The prototype is evaluated using four cloud quality of service metrics. The results show that the model has accomplished the metrics of efficiency of 62%, effectiveness of 64% and learnability of 62%
