    Mikroservis Tabanlı E-Ticaret Uygulamalarında Sipariş Odaklı Endpoint Yönetimi

    Günümüz teknolojisindeki hızlı gelişmeler, bireylerin gereksinimlerini ciddi oranda dönüştürmektedir. Teknolojinin bireyler üzerindeki etkisi baz alınarak dünyada her gün yeni gelişmeler yaşanmaktadır. Modern yazılım projelerinde ihtiyaçlara sürekli yenilerinin eklenmesi, ölçeklenebilirlik, hata izolasyonu, teknoloji çeşitliliği, esneklik gibi konularda sorunlara yol açmaktadır. Monolitik mimari’den günden güne mikroservis mimariye geçiş kaçınılmaz hale gelmektedir. Projelerin büyümesi ile mikroservis mimarilerin kullanım avantajlarından daha fazla yararlanılır hale gelinmiştir. Bu nedenle, monolitik mimari yerine mikroservis mimarisine geçiş, projelerin geleceğe yönelik hale getirilmesine imkan sunmaktadır. Tüketicilerin kolayca ihtiyaçları olan ürünlere ulaşımını kolaylaştırmak amacıyla, hızlı kargo, temassız teslimat ve temassız ödeme gibi seçenekler hayatımıza dahil olmuştur. Bu kapsamda tüketicilerin siparişlerinin hataya yer vermeden işlenilmesi ve teslimatının sağlanması büyük önem arz etmektedir. Bu çalışmadaki amaç, e-ticaret uygulamalarında sipariş adımında meydana gelen bir takım hataların önüne geçilerek, maliyet, sistem karmaşıklığı, performans kaybı, veriler arası tutarlılık gibi konuların iyileştirilmesi sağlanıp tüketicilerin ihtiyaçlarına daha iyi yanıt vermek, daha optimize edilmiş bir deneyim sunulması amaçlanmıştır. Çalışma kapsamında, mikroservis mimari ile oluşturulmuş e-ticaret uygulamasında yapılan bir istek, anında servislerden herhangi birinde alınacak olan hata, bütün isteği başarısız kılıyor olup, tüketiciler tarafından bakıldığında kabul edilebilir bir sonuç değildir. Oluşan problem üzerinde hata mekanizmalarının uygulanabilirliği, uygulamaların nasıl işleneceği, mekanizmaları kullanmanın kazanımları, kullanılması ve kullanılmaması durumlarında yaşanacak etkenlerin analizlerine yer verilmiştir

    MCDM MODEL PROPOSAL AND SOLUTION FOR EVALUATION OF MEDICAL WASTE DISPOSAL TECHNIQUES WITHIN THE SCOPE OF ZERO WASTE APPROACH

    This study is carried out in order to evaluate the disposal techniques of medical wastes, which contain various viruses, bacteria, microorganisms and have negative effects on both the environment and human health. Within the scope of the study, a multi-criteria decision-making model consisting of medical wastes and disposal techniques have been proposed and this model has been solved with a two-stage methodology. In the first stage of the solution, medical wastes have been prioritized with the ENTROPY method, and in the second stage alternative disposal techniques have been evaluated with the WASPAS and EDAS methods. According to ENTROPY method, it was found that the most important medical waste generated as a result of the processes in health institutions was Radioactive waste with an importance weight of 0.1751. This medical waste was followed by Amalgam waste and Chemical waste medical wastes with the importance weights of 0.1478 and 0.1408, respectively. After these findings, alternative disposal techniques have been evaluated with the WASPAS and EDAS methods. According to WASPAS method results the Mechanical operations technique was found to be the most appropriate technique for the disposal of medical wastes in the proposed model with 0.7761 relative importance value. This alternative technique was followed by Irridation methods and Sterilization techniques, respectively. In addition, according to the results of the EDAS method, it was determined that the mechanical processes technique was the most suitable disposal technique with a value of 0.9328.     This paper examines the complex relationships among technological innovation, technological anxiety, perceived value and customer experience within the leisure services environment. Its main objectives are to explore the direct impact of technological innovation on customer experience and perceived value, as well as the direct impact of technological anxiety on customer experience and perceived value. Additionally, the study aims to examine the mediating role of perceived value and assess the moderating effects of technological anxiety. Data were gathered from 423 respondents using a survey questionnaire between May and July 2024 and examined using SEM. The main findings show that technological anxiety significantly increases both customer experience and perceived value. Technological anxiety positively influences both customer experience and perceived value, suggesting that overcoming technological challenges can increase customer satisfaction. Perceived value acts as a mediator in the relationships between technological anxiety and customer experience and between technological innovation and customer experience. Furthermore, the interaction between technological anxiety and technological innovation positively influences perceived value. The main findings highlight the need to continuously invest in advanced technologies, effectively manage technological anxiety and emphasize value-added benefits to increase customer satisfaction with entertainment services.     Short and long-term blockades of adenosine 2A, 5-HT2A, and 5-HT7 receptors induce apoptosis, reduce proliferation, and show differential effects on miR-27b-3p expression in neuroblastoma cell lines

    The first of our aims in this study was to investigate the effects of 5-HT2AR, 5-HT7R, and A2AR blockades on miR-27b-3p expression in the short and long-term in neuroblastoma cells. Our second aim was to reduce the expression of pERK and suppress proliferation by blocking the 5-HT2AR with ketanserin. Our third aim was to reduce the expression of pAKT and induce apoptosis by blocking the A2AR and 5-HT7R with MSX3 and SB269970. Thus, we aimed to investigate the therapeutic efficacy of ketanserin, MSX3 and SB269970, individually or in combination, on neuroblastoma cells. We found that short and long-term blockades of A2A, 5-HT2A, and 5-HT7 receptors had different effects on miR-27b-3p expression. Blockade of A2AR and 5-HT7R with MSX3 and SB269970 decreased miR-27b-3p expression in the short term while increasing it in the long term. Ketanserin increased miR-27b-3p expression in both the short and long term. When 5-HT2AR was blocked with ketanserin, no significant difference was observed in pERK expression and proliferation in the short term. In contrast, a substantial decrease in pERK expression and proliferation was detected in the long term. Our findings show that the MSX3 + SB269970 dual combination and ketanserin + MSX3 + SB269970 triple combination are especially critical in suppressing pAKT expression in the long term. These findings showed that pAKT protein expression induced apoptosis due to decreased in neuroblastoma cells. Our study provides the first evidence for the relationships between ketanserin/miR-27b-3p/pERK, MSX3/miR27b-3p/pAKT, and SB269970/miR-27b-3p/pAKT in neuroblastoma cells.     Activation of epidermal growth factor receptors in triple-negative breast cancer cells by morphine; analysis through Raman spectroscopy and machine learning

    Triple negative breast cancer (TNBC) is a very aggressive form of breast cancer, and the analgesic drug morphine has been shown to promote the proliferation of TNBC cells. This article investigates whether morphine causes activation of epidermal growth factor receptors (EGFR), the roles of ?-opioid and EGFR receptors on TNBC cell proliferation and migration. While examining the changes with molecular techniques, we also aimed to investigate the analysis ability of Raman spectroscopy and machine learning-based approach. Effects of morphine on the proliferation and migration of MDA.MB.231 cells were evaluated by MTT and scratch wound-healing tests, respectively. Morphine-induced phosphorylation of the EGFR was analyzed by western blotting in the presence and absence of ?-receptor antagonist naltrexone and the EGFR-tyrosine kinase inhibitor gefitinib. Morphine-induced EGFR phosphorylation and cell migration were significantly inhibited by pretreatments with both naltrexone and gefitinib; however, morphine-increased cell proliferation was inhibited only by naltrexone. While morphine-induced changes were observed in the Raman scatterings of the cells, the inhibitory effect of naltrexone was analyzed with similarity to the control group. Principal component analysis (PCA) of the Raman confirmed the epidermal growth factor (EGF)-like effect of morphine and was inhibited by naltrexone and partly by gefitinib pretreatments. Our in vitro results suggest that combining morphine with an EGFR inhibitor or a peripherally acting opioidergic receptor antagonist may be a good strategy for pain relief without triggering cancer proliferation and migration in TNBC patients. In addition, our results demonstrated the feasibility of the Raman spectroscopy and machine learning-based approach as an effective method to investigate the effects of agents in cancer cells without the need for complex and time-consuming sample preparation.     Memorable gastronomy tourism experiences of Polish tourists visiting Turkey

    Türkiye’yi ziyaret eden yabancı turistlere yönelik istatistikler incelendiğinde son beş yılda en önemli artış gösteren ülkelerden birisinin Polonya olduğu görülmektedir. Türkiye’yi ziyaret eden Polonyalı turist sayısı beş yıl öncesine kıyasla %100’ün üzerinde artışla 1.5 milyona yaklaşmıştır. Bu kapsamda bu araştırma, Türkiye’yi ziyaret eden Polonyalı turistlerin unutulmaz gastronomi turizmi deneyimlerini keşfetmek amacıyla gerçekleştirilmiştir. Bu amaçtan hareketle Türkiye’ye seyahat eden ve ülkelerine geri dönen 18 Polonyalı ziyaretçi ile kartopu örneklem tekniği kullanılarak yarı yapılandırılmış görüşme formu vasıtasıyla görüşmeler gerçekleştirilmiştir. Görüşmeden elde edilen veriler MAXQDA 2024 programı ile betimsel analize tabii tutulmuştur. Elde edilen bulgular, Türkiye seyahatinden en çok hatırlanan yemeklerin “baklava, ayran, Adana kebap, döner, tantuni” olduğunu, yemeklerin “lezzet, yenilik ve otantiklik” bağlamında unutulmaz deneyim yaşattığı ve bu anlamda, unutulmaz gastronomi deneyimi yaşanmasında en çok ürüne dayalı unsurların etkili olduğu görülmüştür. Katılımcıların en kötü unutulmaz yemek deneyimlerinin “kebap ve mumbar” tüketimi ile ve “sağlık, alışkanlık, hijyenik olmaması ve bayat olması” etkenleri ile meydana geldiği görülmüştür. Diğer yandan katılımcıların Türk mutfağı hakkındaki düşünceleri “lezzetli, oldukça iyi, otantik, oldukça çeşitli” gibi olumlu görüşler ve “çok baharatlı, çok yağlı, sağlıksız, kaliteli değil” gibi olumsuz görüşlerden oluşmuştur. Katılımcıların Türk mutfağı hakkındaki önerileri “çalışanların nitelik ve niceliği, tadımlık porsiyon sunumu, daha az baharat kullanımı, kaliteli ürün kullanımı, adil fiyatlandırma ve eldiven kullanımı” etrafında şekillenmiştir. Bu bulguların Türkiye’yi ziyaret eden farklı milletlerden turistlerin Türk mutfağına yönelik olumlu ve olumsuz görüşleri ile benzerlik gösterdiği görülmektedir.The statistics for foreign travelers visiting Turkey indicate that Poland has experienced a substantial increase in the past five years. The number of Polish tourists visiting Turkey has increased by over 100% compared to five years ago, approaching 1.5 million. We conducted this research to explore the memorable gastronomy tourism experiences of Polish tourists visiting Turkey. Based on this purpose, 18 Polish visitors who travelled to Turkey and returned to their country were interviewed through a semi-structured interview form using the snowball sampling technique. The data obtained from the interviews were subjected to descriptive analysis through MAXQDA 2024 program. The results showed that “baklava, ayran, Adana kebab, döner, and tantuni” were the most memorable dishes from the trip to Turkey, providing a memorable experience in terms of “taste, novelty, and authenticity.” In this sense, product-based elements were most effective in creating a memorable gastronomy experience. Notably, the participants’ most memorable food experiences were associated with the consumption of “kebab and mumbar” and the factors of “health, habit, unhygienic, and stale”. On the other hand, the participants’ opinions about Turkish cuisine consisted of positive ones such as “delicious, quite good, authentic, and quite diverse” and negative ones such as “too spicy, too oily, unhealthy, and not of good quality.” The participants shaped their suggestions about Turkish cuisine around “quality and quantity of employees, tasting portions, proper use of spice, quality products, fair pricing, and use of gloves.

    Impact of microwave heating on product safety and quality in meatballs

    The effects of microwave heating at different processing time and power levels (360 W, 600 W and 900 W) on Salmonella Typhimurium , Escherichia coli O157:H7, Listeria monocytogenes and Staphylococcus aureus in Turkish type meatballs were evaluated. Besides, the changes in the textural and sensorial properties of meatballs were determined to reveal the cooking time and temperatures that would make the product safety without affecting the consumer preferences. In this direction, the experimentally contaminated meatballs were subjected to microwave heating at low, medium and high power levels for different processing times and analysed for the survival of the relevant pathogenic bacteria. The total aerobic bacteria count in meatballs could be reduced approximately 3 - 4 log CFU/g by medium and high microwave powers, whereas 5 - 7 log CFU/g inhibition was obtained in meatballs inoculated with pathogens. The increase in power level together with the increasing process time resulted in a decrease in the counts of bacteria and an undesirable effect on the textural and sensorial qualities of meatballs. Microwave heating at medium and high power levels as from 3 min, caused excessive drying and blackening of the product, making it difficult to consume due to crust formation.     Fabrication of a Microfluidic Test Device with a 3D Printer and Its Combination with the Loop Mediated Isothermal Amplification Method to Detect Streptococcus pyogenes

    New rapid, reliable, and cost-effective alternative systems are needed for the rapid diagnosis of Streptococcus pyogenes. The aim of this study was to fabricate a microfluidic test device to detect Streptococcus pyogenes by combining the Loop-mediated isothermal amplification method via a 3D printer. Microfluidic test devices were designed in CATIA V5 Release 16 software, and data were directly transferred to a 3D printer and produced using the FDM method with biocompatible PLA filament. The S. pyogenes ATCC 19615 and different ATCC strains was used. Following identification by classical culture methods, a 0.5 McFarland suspension was prepared from the colonies, and DNA isolation was performed from this liquid by a boiling method. S. pyogenes specific speB gene was used to desing LAMP primer sets in PrimerExplorer V5 software and tested on a microfluidic device. LAMP reactions were performed on microfluidic device and on a microcentrifuge tube separately. Both results were analyzed using the culture method as the standard method to diagnostic values. Melting curve analysis of the amplicons of the LAMP reactions performed on a LightCycler 480 system to detect amplification. Among the 50 positive and 100 negative samples, only four samples were found to be false negative by LAMP reaction in a microcentrifuge tube, while eight samples were found to be false negative by LAMP reaction on a microfluidic device. Six samples were found to be false positive by the LAMP reaction in the microcentrifuge tube, while ten samples were found to be false positive by the LAMP reaction on a microfluidic chip. The sensitivity, specificity, positive predictive value, and negative predictive value of the LAMP reactions performed in the microcentrifuge tube and on the microfluidic device were 92-84%, 94-90%, 88.46-80.77%, and 95.92-91.84%, respectively. The limit of detection (LOD) was found to be the same as 1.5 x 10(2) CFU/mL and the limit of quantification (LOQ) values of the LAMP reactions were performed on the microcentrifuge tube and on the microfluidic device were 2.46 x 102-7.4 x 10(2) CFU/mL, respectively. Cohen's kappa (kappa) values of the LAMP reactions were performed on the microcentrifuge tube and on the microfluidic device were 0.620-0.705, respectively. In conclusion, our data showed that the LAMP method can be combined with microfluidic test device to detect S. pyogenes, this microfluidic device can be manufactured using 3D printers and results are close to gold standard methods.     The Correlation between Economic Convergence and Health Indices in Developed Countries

    Background: Economic convergence signifies diminishing income disparities among global or regional economies and their eventual disappearance. It is also linked to economic growth and key health indicators. We aimed to assess the association between economic convergence and key health indicators in developed countries called G7 (USA, UK, Germany, France, Italy, Japan, and Canada). Methods: We examined G7 health and economic indicators from 2000 to 2021 using panel data analysis. We compared balanced and unbalanced panel datasets to address missing data and applied suitable methods to handle missing health indicators. Results: Little's MCAR test (X2 = 3.2872, P ? value = 0.3494) confirmed random missing data in the unbalanced panel, enabling us to impute missing values as missing observations were below 5%. Unit root tests on balanced and unbalanced panel data validated the health convergence hypothesis, showing no unit roots in economic growth rate, current health expenditure, and female and male population indicators (P<0.05). Interestingly, the hypothesis for hospital bed counts in the unbalanced panel, differing from the balanced panel, offers new insights into addressing incomplete health data. Conclusion: While G7 have economic similarities, their health indicators diverge (excluding hospital bed counts). Variations in health indicators stem from healthcare system structures, funding mechanisms, resource allocation, and health investments, even among economies of similar size. Therefore, G7 member states should develop tailored national health policies based on their specific circumstances and priorities, utilizing economic convergence data for effective health resource planning. © 2024 Mutlu et al.
