    DUNE Phase II: scientific opportunities, detector concepts, technological solutions

    The international collaboration designing and constructing the Deep Underground Neutrino Experiment (DUNE) at the Long-Baseline Neutrino Facility (LBNF) has developed a two-phase strategy toward the implementation of this leading-edge, large-scale science project. The 2023 report of the US Particle Physics Project Prioritization Panel (P5) reaffirmed this vision and strongly endorsed DUNE Phase I and Phase II, as did the European Strategy for Particle Physics. While the construction of the DUNE Phase I is well underway, this White Paper focuses on DUNE Phase II planning. DUNE Phase-II consists of a third and fourth far detector (FD) module, an upgraded near detector complex, and an enhanced 2.1 MW beam. The fourth FD module is conceived as a Module of Opportunity, aimed at expanding the physics opportunities, in addition to supporting the core DUNE science program, with more advanced technologies. This document highlights the increased science opportunities offered by the DUNE Phase II near and far detectors, including long-baseline neutrino oscillation physics, neutrino astrophysics, and physics beyond the standard model. It describes the DUNE Phase II near and far detector technologies and detector design concepts that are currently under consideration. A summary of key R&D goals and prototyping phases needed to realize the Phase II detector technical designs is also provided.     Cynarine Exhibits Antiproliferative Activity and Bcl-2-Mediated Apoptotic Cell Death in Breast Cancer Cells

    Cynarine is a biologically active compound that is a derivative of hydroxycinnamic acid and can exhibit a structure-function relationship and therapeutic potential due to its bioactive properties. This study aimed to investigate the antiproliferative and apoptosis activity of the Cynarine compound in breast cancer cells in vitro. MDA-MB-231 MCF7 breast cancer cells and MCF 10A non-cancer cells were used. The MTT method was used to evaluate cell viability. Acolony-forming assay was employed on the cells treated with cynarine to assess their colony-forming capability. Western blotting and annexin-propidium iodide methods with flow cytometry were used to clarify its role in apoptosis. Cell proliferation and colony-forming abilities were reduced in cynarine-treated cells at 56.2%, 67, and 3% in MCF7 and MDA-MB 231, respectively. Apoptotic cell rates after 72 hours of cynarine treatment were found at 7.15%, 42.94%, and 68.67% in MCF 10A, MCF7, and MDA-MB-231 cell lines, respectively. Apoptosis was significantly different in MDA-MB-231 and MCF7 cells treated with cynarine (p < 0.05) It has been concluded that cynarine can be used as a combination therapeutic or a precursor compound to develop new drug combinations to treat breast cancer

    The therapeutic role of Aronia melanocarpa on cyclophosphamide-induced premature ovarian failure

    The purpose of the study was to evaluate the effects of Aronia melanocarpa (A. melanocarpa) extract on ovarian cell apoptosis, vascularization, and follicle reserve in the premature ovarian failure (POF) induced by cyclophosphamide (CYC). After creating a premature ovarian failure model with 100 mg/kg CYC, A. melanocarpa was given to Wistar albino female rats by gavage 15 times every other day (200 mg/kg).Vaginal smears were taken to evaluate the estrus cycles before, during and at the end of the experiment. Histological ovarian follicle count was performed by examining the ovaries with Hematoxylin&Eosin staining. Immunohistochemical evaluation of apoptosis markers of caspase-3 (cas-3), caspase-9 (cas-9) and vascular endothelial growth factor (VEGF) was performed. A. melanocarpa reregulated the estrus cycles in a similar way to the controls by reducing cas-3 and cas-9 in ovarian cells and increasing the number of ovarian cells. While CYC disrupted ovarian vascularization, A. melanocarpa contributed to ovarian reserve by increasing VEGF immunopositivity. In chemotherapy-induced POF, A. melanocarpa extract increases ovarian reserve by providing vascularization, decreasing apoptosis and providing cycle control in female rats

    Effect of napabucasin and doxorubicin via the Jak2/Stat3 signaling pathway in suppressing the proliferation of neuroblastoma cells

    Purpose: Napabucasin (NP) is a natural compound that can suppress cancer cell proliferation and cell cycle by inhibition of the signal transducer and activator of transcription 3 (STAT3) gene. We examined the effects of NP on the proliferation and invasion of neuroblastoma cells (SH-SY5Y). Methods: Human neuroblastoma SH-SY5Y cell line was used in this study. NP was administered to groups at the doses of 0.3-1 mu M. Cell viability was analyzed by MTT assay. Real-time quantitative reverse transcription polymerase chain reaction and western blot analysis assessed the expressions of interleukin (IL)-6 dependent Jak2/Stat3 signaling pathway. The MTT cell viability method was applied to determine the antagonistic-synergistic effects and inhibitory concentration (IC50) doses of doxorubicin (DX) and NP. Results: It was determined that 0.3-1 mu M doses of NP killed the cells almost completely after 48 hours, and also that Jak2/Stat3 expressions decreased dose-dependently via IL-6. At the protein level, NP and DX were found to reduce Jak2 and Stat3 levels. Conclusion: NP showed that it suppresses the proliferation of neuroblastoma cells. Due to its inhibitory effect on Jak2 and Stat3, it can be used to prevent invasion of SH-SY5Y cells. NP, which can inactivate Jak2/Stat3, can be used as a treatment agent by combining with DX in proliferation pathway in neuroblastoma

They are one of the most played games of all time, which is why it has been the focus of academic studies since its first release. Within the scope of this research, a comprehensive survey was conducted. The first phase of the survey was conducted with 54 participants and the participants who graduated from different departments were asked whether there is a relationship between The Sims game experiences, interest in playing SIMS and choosing a profession. The following phase was conducted with the focus of architects, interior architects and industrial design graduates and students, who are called design faculty members.     The Effect of Mindfulness-Based Stress Reduction on Pediatric Oncology Nurses’ Stress Level, and Pediatric Oncology Patients’ Psychosocial Symptoms and Care Satisfaction: A Prospective Non-Randomized Trial

    Studies investigating the effects of relaxation practices on nurses and patient outcomes are limited. This study aimed to evaluate the impact of Mindfulness-Based Stress Reduction (MBSR) on pediatric oncology nurses’ stress levels, as well as the psychosocial symptoms and care satisfaction of pediatric oncology patients. A non-randomized prospective study with a pretest-posttest design was conducted, involving 112 children and 8 pediatric oncology nurses between November 2021 and August 2022. The study was conducted in 3 stages. First, the children’s nursing care satisfaction and psychosocial symptoms were evaluated during the first 3 months before the MBSR intervention. Second, the nurses participated in the MBSR program for 2 months. Third, the children’s nursing care satisfaction and psychosocial symptoms were assessed again 3 months after the MBSR intervention. The results showed that children who received nursing care post-MBSR had lower scores in the Psychosocial, Hopelessness, and Communication Difficulty subscales compared to those who received care pre-MBSR. Additionally, these children had higher scores in Evaluation of Quality of Care from the Children’s Point of View, Psychosocial, Physical, and Information subscales. There was a significant reduction in the stress levels of nurses between the pre-MBSR and post-MBSR assessments. MBSR is an effective method for reducing pediatric oncology nurses’ perceived stress levels and should be utilized to support both pediatric oncology nurses and patients in enhancing their psychosocial functioning.     Investigation of the mediator role of anxiety levels in the relationship between the general self-efficiency and expat adaption of citizens of the republic of Turkey living in the Netherlands

    Mevcut çalışmanın amacı, göç eden bireylerin expat uyumlarının yordayıcıları olarak bireylerin genel öz yeterliliklerinin ve anksiyete düzeylerinin incelenmesidir. Bu amaç kapsamında genel öz yeterlilik ve expat uyumu arasındaki ilişkide anksiyetenin aracı rolü test edilmiş ayrıca araştırma değişkenleri arasındaki ilişki incelenmiştir. Araştırma verileri 402 kişinin katılımı ile toplanmıştır. Kaıtılımcılara, Demografik Bilgi Formu, Expat Uyum Ölçeği, Genel Öz Yeterlilik Ölçeği ve Bütünleşik Anksiyete Stres Ölçeği uygulanmıştır. Katılımcılardan elde edilen veriler SPSS paket programı ile analiz edilmiştir. Yapılan analizler neticesinde, genel öz yeterlilik ile expat uyumu arasındaki ilişkide anksiyetenin kısmi kısmi aracı rolünün bulunduğu, expat uyumunun bileşenleri olan etkileşimsel uyum, genel yaşam uyumu ve iş yeri uyumu ile genel öz yeterlilik arasındaki ilişkilerde de anksiyetenin kısmi aracı rolünün olduğu sonuçlarına varılmıştır. Ayrıca, demografik değişkenler ile araştırma değişkenleri arasındaki ilişkide yapılan analizler vasıtası ile ortaya koyulmuştur. Elde edilen bulgular literatür çerçevesinde tartışılmış ve gelecekteki araştırmalar için önerilerden bulunulmuştur.The purpose of the current study is to examine the predictors of expatriate adjustment in migrating individuals, with focusing on the roles of general self-efficacy and anxiety levels. For this purpose, the mediating role of anxiety in the relationship between general self-efficacy and expat adjustment was tested, and the relationships among the research variables were analyzed. The research data were collected from 402 participants. Participants were taken a Demographic Information Form, an Expat Adjustment Scale, a General Self Efficacy Scale, and an Integrated Anxiety Stress Scale. The data obtained from participants were analyzed using the SPSS. The analyses revealed that anxiety plays a partial mediating role in the relationship between general self efficacy and expat adjustment. This was also the case for the dimensions of expat adjustment, namely interaction adjustment, general life adjustment, and workplace adjustment, with general self-efficacy where anxiety was found to play a partial mediating role. In addition, the relationships between demographic variables and research variables were elucidated through analyses. The findings have been discussed within the framework of the literature and suggestions have been made for future research

    THE INFLUENCE OF SPORTS HOTEL EMPLOYEES'POLITICAL SKILL ON ROLE STRESS: THE MODERATING ROLE OF SELF-EFFICACY

    Political skill is one of the most important personal resources that employees have. The presence of political skill can cause employees to manage situations such as conflict, ambiguity and overload caused by their roles more effectively. In addition, depending on whether self-efficacy is high or low, the strength of the influence between the relevant variables may increase or decrease. In this context, the study's main purpose is to investigate the moderating role of self-efficacy in the effect of political skill on employees' role stress. The study population of the study consists of those working in sports tourism hotels. The data obtained from 308 sports tourism hotel employees were analyzed with the help of SPSS and Smart PLS programs. According to the findings, it was determined that political skill did not significantly negatively affect role stress and its sub-dimensions, such as role conflict, role overload and role ambiguity. However, when the analysis was made on the moderating role variable, it was revealed that self-efficacy had a moderating role in the effect of political skill on role conflict and role overload among the sub-dimensions of role stress. The findings were discussed in the last part of the study, and suggestions for future research were presented

    FİNANSAL GEVŞEKLİK TÜRLERİNİN FİRMA PERFORMANSINA VE HİSSE SENEDİNE OLAN ETKİSİNİN İNCELENMESİ

    Bu çalışmada, finansal gevşeklik türlerinin, finansal performansa ve hisse senedi fiyatına olan etkisi araştırılmıştır. Bu doğrultuda, 2013-2021 yılları arasında, BİST 30 Endeksi’nde faaliyet gösteren firmaların verileri incelenmiştir. Analizde kullanılacak bağımsız değişkenler; en çok kullanılan finansal gevşeklik türleri olan; mevcut gevşeklik, geri kazanılabilir gevşeklik ve potansiyel gevşekliktir. Analizde kullanılacak bağımlı değişkenler ise; aktif karlılığı, özsermaye karlılığı, Tobin Q oranı, piyasa değeri defter değeri oranı ve hisse senedi fiyatıdır. Uygulamada, ilgili yıllara ait verilere panel veri analiz yönetimi uygulanmıştır. İlgili analizin yapılmasında, Stata 14 paket programı kullanılmıştır. Literatürdeki çalışmalar incelendiğinde, Türkçe yayın olarak bir adet doktora tezi bulunduğu ve mevcut literatürün yurt dışı yayınlar ağırlıklı olduğu, ayrıca literatürdeki çalışmalarda analize finansal performans ölçümlerinin katıldığı, fakat hisse senedi fiyatlarının dahil edilmediği görülmüştür. Bu doğrultuda hem BİST30 Endeksi üzerinde analiz yapılmış hem de analize firmalara ait hisse senedi fiyatları dahil edilmiştir. Yapılan panel veri analizi sonucunda, finansal gevşeklik türlerinin bilanço temelli finansal oranları anlamlı olarak etkilediği görülmüştür
