The Investigation of the Chemical Composition and Applicability of Gold Nanoparticles Synthesized with Amygdalus communis (Almond) Leaf Aqueous Extract as Antimicrobial and Anticancer Agents
The current work's main objective was to determine the chemical composition of Amygdalus communis (AC) leaf extract and examine the antibacterial and cytotoxic properties of biosynthesized gold nanoparticles (AuNPs). The chemical composition of AC leaf extract was determined using LC-ESI/MS/MS to detect compounds that may be responsible for the reducing, stabilizing, and capping steps in the synthesis of nanoparticles and their biological activities. The AC-AuNPs were spherical, with a particle size lower than 100 nm and a face-centered cubic structure. The EDX spectrum confirmed the formation of AuNPs and a negative zeta potential value (-27.7 mV) suggested their physicochemical stability. The in vitro cytotoxic efficacy of the AC-AuNPs against colorectal adenocarcinoma (Caco-2), glioma (U118), and ovarian (Skov-3) cancer cell lines and human dermal fibroblasts (HDFs) was evaluated by MTT assay. CaCo-2 cell proliferation was effectively inhibited by the AC-AuNPs at concentrations between 25 and 100 g mL-1. The AC-AuNPs exerted preeminent antimicrobial activity against Bacillus subtilis with an MIC of 0.02 μg/mL, whilst good activity was shown against Staphylococcus aureus bacteria and Candida albicans yeast with an MIC of 0.12 μg/mL. Ultimately, the results support the high antibacterial and anticancer potential of biosynthesized AuNPs from AC leaf extract
Tracing the protoliths of the garnet amphibolitic and retrogressed eclogitic slices and a conceptual tectonic model for their emplacement onto the Central Menderes Massif, Turkey: New geochemical data and laser ablation-inductively coupled plasma-mass spectrometry U-Pb zircon and rutile ages
The age, emplacement, and metamorphic history of the garnet amphibolitic and retrogressed eclogitic slices in the Menderes Massif (Eastern Aegean) have been a matter of debate since the 1990’s. Late Cretaceous garnet amphibolitic and retrogressed eclogitic slices in the Bozdağ and Çine nappes show low-angle tectonic contacts with the surrounding Early Cambrian meta-siliciclastics in the Alaşehir-Yahyaalcı and late Neoproterozoic basement rocks in the Birgi and Tire-Çamlıca Klippe in the northern Menderes Massif. Lower and upper intercept ages on the discordia diagrams of the garnet amphibolitic slices are 82 ± 230 Ma and 554 ± 14 Ma, respectively, in the Yahyaalcı-Alaşehir area of the Bozdağ Nappe; and 96 ± 260 Ma and 550 ± 13 Ma, respectively, in the Camlica-Tire Klippe area of the Çine Nappe. Retrogressed eclogitic slices give dates of 81 ± 13 Ma lower intercept and 546 ± 13 Ma upper intercept discordant ages in the Yenişehir-Kiraz area of the Çine Nappe. Lower intercept age of 81 ± 13 Ma is supported by two spots dated on metamorphic zircons showing positive Eu patterns yielding 86.0 ± 1.3 Ma concordant age. Zircon U-Pb dating on the oscillatory zoning spots are dated as 537.5 ± 1.6 Ma (mean square weighted deviation [MSWD] = 1.5, n = 41) for the garnet amphibolic slices in the Yahyaalcı-Alaşehir area (Bozdağ Nappe), 539.0 ± 1.1 Ma (MSWD = 1.4, n = 56) for the garnet amphibolite slices in the Camlica-Tire Klippe area (Çine Nappe), and 536.6 ± 2.3 Ma (MSWD = 1.7, n = 32) for the retrogressed eclogitic slices (Çine Nappe). These ages combined with Eu negative anomalies on the dated spots are evaluated as representing their magmatic crystallization ages of the protoliths. Garnet isopleth intersections and pseudosections gave pressure-temperature conditions of 14 kbar and ~680 °C for the retrogressed eclogites, whereas garnet amphibolites display disequilibrium in whole rock scale. We propose that the age of their protoliths likely correlate with the Cadomian riftrelated Early Cambrian meta-mafic dykes that were subducted beneath the İzmir-Ankara-Erzincan Neotethyan lithosphere during the Late Cretaceous. The new rutile U-Pb age of 30.1 ± 2.0 Ma supports that the tectonic slices of the İzmir-Ankara oceanic lithosphere and metamorphosed Tauride-Anatolide continental margin were emplaced onto the Menderes Massif to generate the “main Menderes metamorphic terrane” during the latest Paleocene and early Eocene
A correlation coefficient-based feature selection approach for virus-host protein-protein interaction prediction
Prediction of virus-host protein-protein interactions (PPI) is a broad research area where various machine-learning-based classifiers are developed. Transforming biological data into machine-usable features is a preliminary step in constructing these virus-host PPI prediction tools. In this study, we have adopted a virus-host PPI dataset and a reduced amino acids alphabet to create tripeptide features and introduced a correlation coefficient-based feature selection. We applied feature selection across several correlation coefficient metrics and statistically tested their relevance in a structural context. We compared the performance of feature-selection models against that of the baseline virus-host PPI prediction models created using different classification algorithms without the feature selection. We also tested the performance of these baseline models against the previously available tools to ensure their predictive power is acceptable. Here, the Pearson coefficient provides the best performance with respect to the baseline model as measured by AUPR; a drop of 0.003 in AUPR while achieving a 73.3% (from 686 to 183) reduction in the number of tripeptides features for random forest. The results suggest our correlation coefficient-based feature selection approach, while decreasing the computation time and space complexity, has a limited impact on the prediction performance of virus-host PPI prediction tools
Forecasting of COVID-19 Fatality in the USA: Comparison of Artificial Neural Network-Based Models
The first death caused by the novel coronavirus in the USA was declared on February 29, 2020, in the Seattle area in Washington state. Forecasting the number of deaths has great importance in terms of public psychology and strategic decisions to be taken by the government. There are several data-driven models in the literature to predict the deaths in the USA caused by COVID-19. However, most of them are based on a few variables of the data for forecasting. From this point of view, this study provides an artificial neural network (ANN)-based approach by considering 12 different variables for forecasting the cumulative deaths caused by COVID-19 in the USA. The proposed ANN structure was trained with three algorithms, namely scaled conjugate gradient algorithm, Levenberg-Marquardt algorithm and Bayesian regularization algorithm. These three forecasting models were constructed on 13 parameters such as 12 inputs and one output. The sensitivity and performance of the proposed forecasting models were analyzed and compared by using indices mean absolute error, mean absolute percentage error, correlation coefficient (R-value), sum square error, variance account for, mean square error and root-mean-square error. Results show that the forecasting model with Bayesian regularization performs better than other models for forecasting the cumulative deaths due to COVID-19 in the USA
In vitro anti-carcinogenic effect of andarine as a selective androgen receptor modulator on MIA-PaCa-2 cells by decreased proliferation and cell-cycle arrest at G0/G1 phase
Pancreatic cancer (PC) continues to be devastating due to its highly malignant nature and poor prognosis. The limited benefits of the chemotherapeutic drugs and increasing resistance pose a critical challenge to overcome and warrant investigations for new therapeutic agents. Several preclinical and clinical studies have suggested a possible role of the androgen receptor (AR) signaling pathway in PC development and progression. Nevertheless, the studies are limited and inconclusive in explaining the molecular link between AR signaling and PC. Selective androgen receptor modulators (SARMs) are small molecule drugs with high affinity for the androgen receptor. SARMs elicit selective anabolic activities while abrogating undesired androgenic side effects. There is no study focusing on the utility of SARMs as inhibitors of PC. Here, we report the first study evaluating the possible anti-carcinogenic influences of andarine, a member of the SARMs, on PC. The data we presented here has illustrated that andarine repressed PC cell growth and proliferation via cell cycle arrest at G0/G1 phase. Gene expression analysis revealed that it downregulates CDKN1A expression level accordingly. Furthermore, we established that the anti-carcinogenic activity of andarine is not mediated by the PI3K/AKT/mTOR signaling pathway, a crucial regulator of cell survival. Our findings suggest that andarine might be considered as a prospective drug for PC
Conjunctival Impression Cytology and Tear Film Changes in Sarcoidosis: A Subjective and Objective Diagnosis Study
Objectives: To evaluate sarcoidosis-induced tear film changes using subjective and objective diagnostic tests, particularly conjunctival impression cytology (IC), and to compare the results with healthy individuals.
Materials and methods: This study evaluated clinical data collected between January 2019 and January 2021 from 57 right eyes of 57 sarcoidosis patients without ocular involvement (Group 1) and 33 right eyes of 33 healthy individuals with similar demographic characteristics (Group 2). The Schirmer I test, tear break-up time (TBUT), fluorescein staining, and conjunctival IC were all performed as part of the conjunctival and corneal examinations following a thorough ophthalmological examination. The Ocular Surface Disease Index (OSDI) was used to assess subjective ocular symptoms.
Results: The mean ages in Groups 1 and 2 were 49.26±3.18 and 51.91±2.89 years, respectively (p=0.720). The mean Schirmer I test, TBUT, and OSDI scores differed significantly (p<0.05 for all), with Group 1 having a significantly higher percentage of dry eyes than Group 2. Group 1 had significantly higher Nelson's grading system grades than Group 2 based on conjunctival IC analysis (p=0.001). There were no significant differences in visual acuity (p=0.17) or intraocular pressure (p=0.14) between groups.
Conclusion: Sarcoidosis patients had significantly higher Nelson grades in conjunctival IC, as well as significantly higher percentages of dry eye as determined by the Schirmer I test, TBUT, and OSDI. Reduced tear quantity and quality may destabilize the tear film layer, resulting in a variety of ocular symptoms
A Turkish Study to Identify the Discharge Learning Needs of Spinal Surgery Patients
PURPOSE: The aim of this study was to identify the learning needs of spinal surgery patients before hospital discharge. METHODS: This cross-sectional study consisted of 117 spinal surgery patients admitted to the neurosurgery department between October 2019 and March 2020. Data were collected using a descriptive information form, visual analog scale, and the Patient Learning Needs Scale. Data were analyzed using descriptive statistics, Mann-Whitney U and Kruskal-Wallis tests, and Spearman correlation analysis. RESULTS: The mean age of the participants was 54 years, 54.7% were male, and 59% underwent surgery because of spinal disc herniation. The mean Patient Learning Needs Scale score was 188.74. The primary learning needs of the patients were related to the dimensions of activities of living, medication, treatment, and complications, whereas the feelings related to condition were the least-demanded dimension of learning needs. Sex and occupation were the primary factors influencing learning needs. CONCLUSION: The level of learning needs in spinal surgery patients was relatively high. Therefore, discharge education may be planned in line with the learning needs and priorities of these patients, and sex and occupation may be considered while planning discharge education
Tetrachloromethane Induced Acute Liver Injury in Mice: Biochemical and Histopathological Study of the Hepatoprotective Effect of Hep-X Standardized Botanical Dietary Supplement
This work was aimed at investigating the hepatoprotective effect of Hep-X, a polyherbal formulation containing Silybum marianum L. (Milk thistle), Rosmarinus officinalis L. (Rosemary), Curcuma longa L. (Turmeric) and Fumaria officinalis L. (Fumitory) as standardized botanical dietary supplement, on mice with CCl4-induced acute liver injury. The total phenolic and flavonoid contents of Hep-X were determined as 0.125 and 0.528 mg/mL, respectively. The quercetin content was determined as 50 µg/mL using HPLC analysis. The total antioxidant capacity showed correlation between the Hep-X concentration and percentage inhibition of free radicals. Hep-X was administered orally at 25, 50 and 100 mg/kg b.w./day against CCl4-induced hepatotoxicity in mice. The hepatic damage was measured using blood biochemical parameters. Animals upon Hep-X treatment exhibited better drug effIcacy in certain blood parameters than silymarin-treated mice. Also, Hep-X administration significantly ameliorated the liver damage by suppressing iNOS expression and apoptosis as well as by recovery of the histological structure. The obtained results suggest that Hep-X is able to significantly alleviate the hepatotoxicity induced by CCl4 in mice, which can be due to antioxidant properties of the polyherbal formulatio
Tourism activities' role on destination choice and post-visiting behavior: Evidence from Istanbul as a cultural destination
This study aims to (1) validate a list of tourism activities in Istanbul, (2) evaluate the importance of tourism activities on a cultural destination choice, and (3) explain the impact of tourism activities on post-visiting behavior. A preference list for activities was generated by in-depth interviews were conducted on travel agencies and tourist guides organizing cultural tours, besides cultural tour programs. The research was conducted on European tourists visiting Istanbul as a cultural destination of Turkey in 2019. A purposive sampling method was used. Selection criteria are in the top six European tourists visiting Istanbul according to their nationality and staying at least one night in the destination. Data were collected by a three-part questionnaire. The results indicated that European tourists attach importance to tourism activities on destination choice. Hence, visiting activities are the most priority activities on the choice of Istanbul as a cultural destination while festivals and sports activities are the least. By the way, European tourists prefer different tourism activities according to their nationality. The results indicated that preference for tourism activities has a positive impact on tourists' post-visiting behaviors
Using ultraviolet dermoscopy in diagnosing scabies
The latest generation ultraviolet (UV) dermatoscopes, which emit UV light at a wavelength of 365 nm and enlarge lesions, are practical devices that can facilitate the diagnosis and follow-up of some dermatological diseases with fluorescence that can be observed in skin lesions. In 305 patients, 468 tunnels were evaluated: first in polarized mode and later in UV mode. The recorded samples were compared one-to-one by the same dermatologist. Due to the study's design, images were examined in three stages: tunnel borders, tunnel content and mite appearance. In UV mode, the entire body of mites gives a bright reflex along with an oval-shaped diagnostic clue well beyond the delta sign. The borders of the tunnel reflect brightly in UV mode, with borders that are more sharply visible than in polarized mode. Although the tunnel content cannot be clearly distinguished in polarized mode, especially in people with hyperkeratotic palms, or 'working hands' (e.g. farmers, mechanics and construction workers), with the bright reflex, the tunnel borders in UV mode give a bright reflection and provide a clear view of tunnels. Tunnel content gives a bright reflection in UV mode. Our results show that UV dermatoscopes provide more effective and clearer images than polarized dermatoscopes in the diagnosis of scabies
