    Understanding Terrorist Use of Explosives in the United States

    Improvised explosive devices (IEDs) remain a weapon of choice for terrorists and violent extremists, presenting a sustained threat to U.S. homeland security. Using public court records pertaining to U.S. federal charges, NCITE researchers have counted 98 incidents occurring from January 2009 through May 2024 in which individuals motivated by a violent extremist ideology plotted or attempted an IED within the United States. A close look at the data reveals an evolving threat landscape. Within the first five months of 2024, ideologically motivated IED incidents had already reached a five year annual high. Analysis of the data indicates that U.S.-based violent extremists see value in using IEDs against a wide range of local targets, especially critical infrastructure and crowded spaces. Moreover, as a tactical instrument, the IED is appealing to would-be attackers across the violent extremist ecosystem

    As expressed in the original 2008 report, Research Challenges in Combating Terrorist Use of Explosives in the United States, improvised explosive devices (IEDs) are generally easy to develop, difficult to combat, and cause disproportionate physical and psychological harm to the citizenry. Research, development, test, and evaluation options to assist domestic counter-IED efforts are plentiful, easily overwhelming the ability of government and industry to fund. This updated report outlines 10 challenge areas where concentrated research can be most beneficial when combating IED use in the homeland and is a summation of interagency efforts to analyze operational capabilities and gaps, as well as their associated research needs

    Comparative Transcriptomics of Candida albicans: Identifying Species-Specific Filamentation Genes Dependent on Environment

    Candida albicans is a human commensal and opportunistic pathogen that lives in the gastrointestinal and genitourinary tracts with infections ranging from superficial to lethal systemic disease. In the United States, Candida infections are the fourth leading cause of nosocomial bloodstream infections with a 40% mortality rate. Candida albicans species can exist in several morphological states of yeast-like budding, pseudohyphae, and hyphae (filamentous). The ability to switch morphologically is closely linked to pathogenesis and much of our understanding to identify these mechanisms has been studied in Candida albicans type strain, SC5314. Other clinical C. albicans strains are genetically and phenotypically divergent from this type of strain, particularly in their ability to filament. In our study, we are examining the filamentation transcriptome of a non-SC5314 clinical strain of C. albicans. We will compare this transcriptome to our already-published filamentation transcriptome of SC5314 to identify filamentation-related gene regulation signatures that are conserved species-wide. Clinical isolate P87 was grown in several filamentation inducing and two control conditions. Filamentous cells were extracted and purified for RNA seq. We used different bioinformatic approaches to prepare the data for read alignment and quantification of gene and transcript levels. We will describe the RNA seq workflow methodology, and highlight challenges and outcomes associated with each step in quantifying gene expression. This data will identify a species-independent, media-independent, filamentation transcriptome for C. albicans

    Reliability of a Novel Force Control Task in a Population with No Lower Extremity Injury

    Background: Athletes who return to cutting and pivoting sports after anterior cruciate ligament reconstruction (ACLR) have a 15-fold greater risk of sustaining an anterior cruciate ligament (ACL) injury compared to previously uninjured individuals. Neuromuscular assessments during rehabilitation can identify deficits in individuals after ACLR and may be useful as screening tools for ACL re-injury risk. Recently, a neuromuscular force control task has been developed that differentiates control strategies between high-performance athletes, recreational athletes, individuals with an ACL deficiency, and individuals following ACLR. Those with an ACL injury exhibited diminished motor control compared to uninjured individuals. However, the reliability of this methodology has yet to be established. Objective: The purpose of this study was to determine the reliability of the novel force control task in a population with no lower extremity injury. Methods: Thirteen individuals completed the force control task for medial/lateral (ML) and anterior/posterior (AP) directions for both legs on two separate occasions (1 week apart). While standing on force plates, the participants generated shear forces bi-directionally to the beat of a metronome set at 60 bpm and tried to align the slider on screen with stationary indicators set at 50% of the participant’s maximal force in each direction. The largest Lyapunov exponent (LyE) was calculated from the force profiles for each measure. Intraclass correlation coefficient (ICC) with 95% confidence intervals, standard error of measurement (SEM), and minimum detectable change (MDC) were calculated to assess the reliability. Results: The ICC values (r=0.78-0.92) across all measures were considered good-excellent, and the Right ML direction was statistically significant (r= 0.92, 95% CI= 0.75-0.98, p=0.025). Conclusions: This study evaluated the reliability of a novel force control task. Our results indicate that the force control task measures force control variability with good to excellent reliability, with the highest reliability in the Right ML direction
