The impact of organisation culture in the retention and career growth of female artisans in the automotive sector in Gqeberha
There have been many initiatives that have been implemented to motivate women to enter and remain in the engineering field in South Africa and across the world. However, research has shown that the numbers of women representation in the engineering sector is still very low. It was the aim of this study to explore essential themes that can be adopted to improve career growth and retention of women in the engineering automotive industry sector of Gqeberha. The study further seeks to formulate a set of key organisational culture attributes that needs to be incorporated into organisations to improve female artisans’ career advancement and retention. The research took a positivist/postposivist paradigm, with a quantitative research approach using a probability sampling method. A structured self-administered questionnaire was distributed electronically to female respondents employed in the automotive companies located within Gqeberha with the aim of obtaining information to achieve the research objectives, targeting a population of female artisans employed in these organisations. Seventy-five (75) questionnaires were distributed, forty-one (41) were completed and returned, which represents a 54.67% response rate. The research found that there is a significant relationship between organisational culture, career advancement and employee retention. The main themes that emerged from the study of employee retention were sense of security, transparency and innovation. While training and development opportunities were found to be important for career advancement. Developing a socio-economic framework for assessing the effectiveness of Expanded Public Works Programmes (EPWP): The case of the Prosopis mesquite Working for Water clearing project in the Northern Cape Province, South Africa
The EPWP functions as a bridge between unemployment and entry into the labour market by providing work readiness skills training to its beneficiaries who receive below-market rate stipends for the short- term duration of their participation. The EPWP combines service delivery issues with social development objectives by promoting intensive manual labour in its projects. As a social protection strategy, public works programmes cater to those who do not meet the criteria to receive government social grants. As one of the programmes under the EPWP dealing with the control and eradication of invasive alien plants, the Working for Water (WfW) programme also uses intensive manual labour methods for clearing alien plant species. Although the clearing successes of WfW are well documented, the programme has focused little attention to the longer-term livelihood impacts of the temporary work and skills training provided to beneficiaries. This study suggests this could be due to a lack of the appropriate indicators to measure these outcomes. Therefore, an evaluation framework for environmental public works projects is proposed, which consists of outcome indicators to track the livelihood impact of the work experience and skills training on the beneficiaries post-participation, since the aim of these EPWP interventions is to improve beneficiaries’ labour market outcomes. The Northern Cape province’s Prosopis mesquite clearing project was used as the case study to develop and test the evaluation framework. The outcome indicators were informed by the key stakeholders’ interviews and the beneficiaries’ survey, specifically since the beneficiaries were well placed to give feedback on the benefits of the work experience and training post-participation. The combined strengths of the Sustainable Livelihoods Approach and the Capability Approach were useful for formulating the outcomes indicators, while the indicators for the inputs, activities and outputs were formulated from the key stakeholder interviews and online EPWP reports. A mixed methods approach was used and primary data were collected through key stakeholder interviews with the Prosopis mesquite clearing project managers and an online survey with some of the beneficiaries. Online EPWP reports and records obtained from WfW were used as secondary data. Data analysis used RStudio, Microsoft Excel and GraphPad Prism. The data analysis and evaluation framework indicators constituted the results section and aimed to highlight the factors that managers should focus on to achieve the desired livelihood outcomes. The proposed outcome indicators can be used to gauge the effectiveness of environmental public works’ social development interventions. The results revealed that the project budget fluctuations resulted in the Working for Water managers adopting a myopic view in administering the workdays and skills training, which diminished the livelihood impact of the Prosopis mesquite clearing project to merely a ‘make work’ project with no observable longer-term livelihood benefits. The selection input indicators and their utilisation during project activities needs to be better aligned with the desired longer-term livelihood outcomes that these environmental public works projects seek to achieve, mainly that of preparing beneficiaries for jobs in the labour market.Thesis (PhD) -- Faculty of Commerce, Economics and Economic History, 202
Contrasting effects of climate change on the invasion risk and biocontrol potential of the invasive Iris pseudacorus L. between Northern and Southern Hemisphere
Iris pseudacorus is both a prized ornamental and an invasive aquatic plant that tends to grow dense monospecific stands, displacing the local vegetation and altering the hydrology of freshwater ecosystems. Originally from Europe, this species has historically invaded North America, China and Japan, and more recently spread through Argentina, South Africa and Australasia, where it is now a target for biological control. Field surveys within its native range have led to the selection of three candidate biocontrol agents. Prioritizing the best candidates for different regions constitutes a critical step, which could save significant time and resources before further cost-intensive experimental studies are conducted. Climate change is seldom taken into consideration in the prioritization process. In this regard, climatic suitability can be used to model the potential distributions of weeds and their candidate agents, both in space and time, thus allowing to identify areas at risk of invasion and predict where agents will be able to establish long-term. Accordingly, the objectives of this work were (i) to predict I. pseudacorus invasions and range shifts in the context of climate change; (ii) to identify wetland areas most at risk of invasion under present and future climatic conditions; and (iii) to prioritize the best suite of candidate biocontrol agents for different invaded ranges, worldwide. To do so, we modelled the present and future (2040–2060) climatic suitability of I. pseudacorus and its candidate agents using the software MaxEnt. Our results highlight a clear distinction between predictions for the Northern and Southern Hemispheres. In North America and eastern Asia, the area climatically suitable for I. pseudacorus is expected to increase and shift northwards. As for its biocontrol agents, very low suitability is predicted across these regions, further decreasing under future climatic conditions. On the other hand, climatically suitable areas for the plant in South America, southern Africa and Australasia are predicted, on average, to reduce in response to climate change. A decrease in climatic suitability is also expected for its candidate biocontrol agents which, however, would still maintain a significant range overlap with their host. These results can be used to prioritize areas most at risk of invasion and identify which combination of candidates could potentially provide the best level of control across different invaded ranges
The heterologous expression and in vitro biochemical characterization of the Hsp70 escort protein 1 and mitochondrial Hsp70 partner proteins of the Trypanosoma brucei parasite and humans
The 70 kDa family of heat shock proteins (Hsp70) plays a central role in the maintenance of cellular proteostasis, with paralogues occurring in all the major compartments of the eukaryotic cell. Hsp70s act in conjunction with proteins known as co-chaperones, as part of the larger molecular chaperone network. In the mitochondrion, Hsp70 (mtHsp70) is responsible for the import of proteins synthesized in the cytosol, protein folding in the matrix and the maintenance of the iron-sulphur cluster. In human cells mtHsp70 (HSPA9) is also referred to as mortalin, as the knockdown of the protein leads to cell mortality. Trypanosoma brucei is the causative agent of sleeping sickness in humans and nagana in animals. In the T. brucei parasite there are three identical mtHsp70 (TbmtHsp70) proteins that are produced, forming part of the Hsp70 machinery that is essential for parasite survival. In humans, the levels of HSPA9 are often elevated in non-communicable diseases such as cancer and neurodegeneration. Despite their vital cellular roles, mtHsp70s are characteristically prone to self-aggregation. The binding of the Hsp70 escort protein (Hep1) is required to prevent the aggregation of mtHsp70 proteins, enabling the proteins to function. In many non-communicable diseases, mtHsp70 and other molecular chaperones such as heat shock protein 90 (Hsp90) are being investigated as potential drug targets. Existing anti-trypanosomal drugs for treating sleeping sickness are toxic, having adverse side effects that are potentially lethal. Investigations into Hsp70s, and other molecular chaperones, form part of the research into the discovery of novel and efficacious therapeutics. This is the first study to characterise Hep1 and investigate its partnership with mtHsp70 in T. brucei. The overall aim of this study was to comparatively assess the T. brucei and human mtHsp70/Hep1 partnerships. The putative T. brucei Hep1 (TbHep1) orthologue was analysed in silico, and it was found to possess a zinc finger domain consisting of anti-parallel β-sheets that are characteristic of canonical Hep1 proteins, whilst the N-terminal domain was unstructured. Based on sequence analysis, the regions outside of the zinc finger domains lacked conservation. Despite the lack of sequence conservation, the N- and C-terminal regions of TbHep1 shared segments of similarity with Hep1 orthologues of other kinetoplastid and trypanosomal orthologues. The same held true for the N- and C-termini of human Hep1 (HsHep1) when compared to other Hep1 orthologues of mammalian origin. Biochemical analysis revealed TbmtHsp70 and HSPA9 to be prone to self-aggregation, which was reduced by co-expression with TbHep1 and HsHep1, respectively. Recently Hep1 proteins have been determined to be present in the cytosol. In this study, TbHep1 and HsHep1 also interacted with the cytosolic Hsp70s, HSPA1A and TbHsp70, by preventing their thermally induced aggregation and stimulating their ATPase activities. TbHep1 and HsHep1 also suppressed the thermally induced aggregation of the model substrates malate dehydrogenase and citrate synthase, independently of Hsp70. To date, only two Hep1 orthologues, HsHep1 and LbHep1, have been found to function in a similar manner to a J-protein co-chaperone by stimulating the ATPase activities of their partner mtHsp70 proteins. In this study, TbHep1 stimulated the ATPase activity of TbmtHsp70. HsHep1 also stimulated the ATPase activity of TbmtHsp70. However, the mechanism of action still needs to be determined. This study also explored the potential of the Hep1 orthologues to be functionally activated by oxidative stress, which is prevalent in mitochondria. The abilities of TbHep1 and HsHep1 to reduce the thermally induced aggregation of malate dehydrogenase were enhanced under oxidative conditions. Disrupting the function of Hep1 has been found to eventually lead to cell death, and given the critical role played by mtHsp70 in the cell, this partnership could be exploited as a potential drug target. In conclusion, this study demonstrated that TbHep1 and HsHep1 functionally interact with mtHsp70s, whilst also possessing independent chaperone activities that are also potentially influenced by the environmental redox state.Thesis (PhD) -- Faculty of Science, Biotechnology Innovation Centre, 202
A digital transformation framework for municipalities in the Eastern Cape
Digital transformation (DT) integrates technology into all business areas, fundamentally changing how businesses operate and deliver value to their clients. It is also a cultural change that requires organisations to continually challenge the status quo, experiment and get comfortable with change. Smart Municipalities in South Africa have various challenges concerning integrating services and information flow among departments within the municipalities, thus preventing the creation of value for the citizens. The ongoing digital transformation revolutionises urban development and governance, offering opportunities for enhanced citizen services, resource optimisation, and sustainable growth. However, the complexity of implementing smart city initiatives demands a structured approach. Due to urbanisation, municipal management in developing nations must deal with issues like increased energy consumption, pollution, the disposal of toxic wastes, resource depletion, ineffective management of urban infrastructures, ineffective planning processes, overloaded transportation networks, endemic congestion, as well as social inequality and socioeconomic disparity. The extent of digital adoption in the South African province of the Eastern Cape has provided municipalities with a learning opportunity to change into data-driven Smart Municipalities in order to increase productivity, growth, employment, and the need for people with a variety of skills. This study required a review of the literature to examine digital transformation, Smart Municipalities, Smart Municipalities at national and international levels, and Smart Municipality models. The literature review also helped to create a Smart Municipality conceptual model. Data for this study were collected in four Eastern Cape municipalities, namely: DR. AB. Xuma, Enoch Mgijima, Intsika Yethu Local municipalities, and their critical stakeholder Chris Hani District Municipality. The population was all municipal workers, including Municipal Managers, Councillors, Executive Management, ICT officials, Unit Managers, Computer Users and other municipal employees who were using IT to do their job responsibilities within the municipalities. This research study followed a positivistic approach using quantitative methods of analysis. Accordingly, the conceptual model was hypothesised and tested using a questionnaire to collect data and the data were statistically analysed. The study followed a descriptive research designwith a quantitative approach. Various statistical analyses were conducted, including descriptive statistics, reliability and validity metrics of the research instrument, correlation coefficients and Exploratory Factor Analysis (EFA). South Africa has provided municipalities with a learning opportunity to change into data-driven Smart Municipalities to increase productivity, growth, and employment supported by the need for people with a variety of skills. A Value Alignment Smart City Stakeholder (VASCS) model was created in previous research and identified nine dimensions of a smart city, namely Smart Economy, Smart Environment, Smart Governance, Smart Living, Smart Mobility, Smart Organisation, Smart People, Smart Policies and Smart Technology. Due to lack of DT framework and guidelines that guides municipalities on how they can adopt Smart Municipality concept, the objective of this study was to propose and evaluate a Value Alignment Smart Municipality Stakeholder (VASMS) model for municipalities in the Eastern Cape. The nine dimensions were empirically evaluated by stakeholders in four Eastern Cape municipalities. The results depict a change towards the importance of developing a Smart Municipality in the four municipalities, while following appropriate planning, designing, management, operations and governance guidelines. The respondents regarded the smart policy dimension as the most important dimension in the proposed VASMS model. This study further provided practical guidelines for the adoption of smart municipalities in the Eastern Cape Province of South Africa. This proposed VASMS model aims to address the unique challenges faced by municipalities in the Eastern Cape as they embark on their digital transformation journey. It outlines a stepby- step process encompassing strategic planning, technological integration, stakeholder engagement, and governance restructuring. Moreover, it emphasises the importance of fostering a collaborative ecosystem between government agencies, private sector partners, and citizens. By offering this VASMS model, this research contributes to the advancement of Smart Municipality initiatives in the Eastern Cape, ultimately paving the way for more efficient, resilient, and citizen-centric urban environments.Thesis (MCom) -- Faculty of Science, School of Computing Sciences, 202
Insights into the relative abundance, demographics and behaviour of sea turtles along the east coast of South Africa
Sea turtles are ecologically important species that have endured a history of overexploitation. Combined with their long, complex life cycles and ongoing anthropogenic pressures, they remain conservation-dependent. Conservation efforts worldwide focus on sea turtle nesting beach protection, however, comprehensive management strategies require an understanding of their lives at sea which are often spent in distant waters. This understanding is hindered by the relative challenges of in-water studies. South Africa has also focused research and conservation efforts on nesting adult female loggerheads (Caretta caretta) and leatherbacks (Dermochelys coriacea), largely overlooking the species or age classes remaining at sea. Green turtles (Chelonia mydas), hawksbills (Eretmochelys imbricata) and non-breeding loggerheads are frequently stranded and rehabilitated by aquaria, and observed by recreational divers on reefs, suggesting year-round residency along the country’s entire seaboard. Recent technological advancements in photo-identification methods for sea turtles, underwater photography and internet connectivity allowing even recreational divers to become citizen scientists now place us on the verge of an exciting opportunity to unravel the cryptic in-water ecology of these species. The overall aim of this study was to conduct the first assessment of relative abundance, demographics, distribution, and behaviour of sea turtles on reefs within the iSimangaliso and Aliwal Shoal Marine Protected Areas (MPAs) on the east coast of South Africa. A robust photo-identification database was established from research dives, voluntourism and citizen science contributions over a two-year period. From 1057 sightings in the two MPAs, a total of 227 individual sea turtles were identified constituting three species (green turtles: n = 120; hawksbills: n = 46; loggerheads: n = 61). Their relative abundance was proportional to the overall sizes of the South West Indian Ocean (SWIO) populations (green turtles > hawksbills > loggerheads > leatherbacks). However, sightings per unit effort (SPUE) showed that loggerheads were more prevalent than hawksbills in iSimangaliso, likely due to its proximity to this loggerhead rookery. Reefs supported mixed size aggregations with juvenile to adult-sized green turtles (straight carapace lengths, SCLs of 44.9–99.2 cm), whilst hawksbills were mostly juveniles or subadults (SCL range: 37.4–73.4 cm) and loggerheads were mostly adult-sized (SCL range: 66.9–81 cm). Adult sex ratios were slightly female-biased but not significantly different from 1:1. The longest minimum residence periods recorded for individual subadult green and hawksbill turtles (676 and 675 days respectively) and adult-sized loggerheads (621 days) were suggestive of residency. Resident sea turtle activities were typical; they wereobserved foraging, resting, cleaning and interacting year-round, whilst only loggerheads and leatherbacks were observed in breeding behaviour (e.g., patrolling and mating) just after the seasonal arrival of transient individuals. This study thus reveals that the east coast of South Africa hosts regionally important resident and transient sea turtle aggregations with some of the highest SPUE results recorded for the region. This first attempt at monitoring foraging grounds using a variety of techniques and stakeholder groups can serve as the baseline assessment for future work and the foundation for a long-term in-water monitoring programme, allowing for expansion along the entire coastline.Thesis (MSc) -- Faculty of Science, School of Environmental Sciences, 202
Enhancing employee engagement and wellbeing whilst implementing remote working throughout the COVID-19 pandemic
The COVID-19 pandemic has disrupted the way in which companies conduct their operations. Companies that were characterised by centralised brick and mortar offices had to quickly adapt to a remote work setting to ensure the continuation of operations. This has created difficulties in ensuring that operations continue as per normal without impacting productivity and profitability. Employees are key to organisations in ensuring that a competitive advantage is maintained and achieved. Competitors will be able to copy products, service and processes but they will find it difficult in duplicating the manner in which their employees conduct their daily work activities. Employee engagement and well-being were key for successful organisations pre-COVID-19 in differentiating themselves from competitors. The aim of this study is to assist companies in identifying the drivers of employee engagement and their impact on well-being whilst implementing a remote work setting. The study will more specifically investigate a proposed theoretical model towards increasing employee engagement and well-being. The model proposed that employee engagement and well-being will be impacted by conciliation, cultivation, confidence, compensation and communication. The empirical component of the study was conducted by administering an electronic survey to 110 employees. The survey was administered to a private company that has a national footprint in South Africa. The main findings of this study confirmed that the factors proposed in the model will have a significant impact on employee engagement and that employee engagement will impact well-being. The study found additional variables such as communication personal, communication infrastructure, employer care and intrusion of work into private life that will impact employee engagement. Furthermore, it was found that all the drivers will influence employee well-being. The findings found that employee engagement and well-being will be interdependent on each other. Best practices in the use and exchange of microorganism biological control genetic resources
The Nagoya Protocol actions the third objective of the Convention on Biological Diversity and provides a framework to effectively implement the fair and equitable sharing of benefits arising out of the use of genetic resources. This includes microorganisms used as biological control agents. Thus biological control practitioners must comply with access and benefit-sharing regulations that are implemented by countries providing microbial biological control agents. A review of best practices and guidance for the use and exchange of microorganisms used for biological control has been prepared by the IOBC Global Commission on Biological Control and Access and Benefit-Sharing to demonstrate commitment to comply with access and benefit-sharing requirements, and to reassure the international community that biological control is a very successful and environmentally safe pest management strategy that uses biological resources responsibly and sustainably. We propose that best practices include the following elements: collaboration to facilitate information exchange about the availability of microbial biological control agents and where they may be sourced; freely sharing available knowledge in databases about successes and failures; collaborative research with provider countries to develop capacity; and production technology transfer to provide economic opportunities. We recommend the use of model concept agreements for accessing microorganisms for scientific research and non-commercial release into nature where access and benefit-sharing regulations exist and where regulations are not restrictive or do not exist. We also recommend a model agreement for deposition of microbial biological control agents into culture collections
