    Cannabisin B induces autophagic cell death by inhibiting the AKT/mTOR pathway and S phase cell cycle arrest in HepG2 cells

    This study investigates the anticancer properties of cannabisin B, purified from hempseed hull, in HepG2 human hepatoblastoma cells. The results indicate that cannabisin B significantly inhibited cell proliferation by inducing autophagic cell death rather than typical apoptosis. Cell viability transiently increased upon the addition of a low concentration of cannabisin B but decreased upon the addition of high concentrations. Cannabisin B-induced changes in cell viability were completely inhibited by pre-treatment with 3-methyladenine (3-MA), indicating that the induction of autophagy by cannabisin B caused cell death. Additionally, cannabisin B induced S phase cell cycle arrest in a dose-dependent manner. Moreover, cannabisin B was found to inhibit survival signaling by blocking the activation of AKT and down-stream targets of the mammalian target of rapamycin (mTOR). These findings suggest that cannabisin B possesses considerable antiproliferative activity and that it may be utilised as a promising chemopreventive agent against hepatoblastoma disease.     When uncertainty meets life: The effect of animacy on probability expression

    Everyone faces uncertainty on a daily basis. Two kinds of probability expressions, verbal and numerical, have been used to characterize the uncertainty that we face. Because our cognitive concept of living things differs from that of non-living things, and distinguishing cognitive concepts might have linguistic markers, we designed four studies to test whether people use different probability expressions when faced with animate or inanimate uncertainty. We found that verbal probability is the preferred way to express animate uncertainty, whereas numerical probability is the preferred way to express inanimate uncertainty. The "verbal-animate" and "numerical-inanimate" associations were robust enough to persist when tested with forced-choice response patterns regardless of the information (e.g., equally likely outcomes, frequencies, or personal beliefs) used to construct probabilities of events. When the response pattern was changed to free-responses, the associations were evident unless the subjects were asked to write their own probability predictions for vague uncertainty. Given that the world around us consists of both animate (i.e., living) and inanimate (i.e., non-living) things, "verbal-animate" and "numerical-inanimate" associations may play a major role in risk communication and may otherwise be useful for practitioners and consultants

    The effects of central pro-and anti-inflammatory immune challenges on depressive-like behavior induced by chronic forced swim stress in rats

    Although increasing evidence demonstrates that both chronic stressors and inflammatory immune activation contribute to pathophysiology and behavioral alterations associated with major depression, little is known about the interaction effect of central inflammatory immune activation and stress on depressivelike behavior. Our previous work has shown that 14-day chronic forced swim stress induces significant depressive-like behavior. The present investigation assessed whether pro-inflammatory cytokine and anti-inflammatory cytokine challenges have differential interaction effect on depressive-like behavior induced by chronic forced swim stress in rats. The pro-inflammatory and anti-inflammatory immune challenges were achieved respectively by central administration of lipopolysaccharide (LPS), a proinflammatory cytokine inducer, and interleukin-10 (IL-10), an anti-inflammatory cytokine. It was found that either central LPS treatment alone or chronic forced swim stress alone significantly induced depressive-like behavior, including reduced body weight gain, reduced saccharin preference and reduced locomotor activity. However, there was no significant synergistic or additive effect of central LPS treatment and stress on depressive-like behavior. LPS treatment did not exacerbate the depressive-like behavior induced by forced swim stress. Nevertheless, IL-10 reversed depressive-like behavior induced by forced swim stress, a finding indicating that IL-10 has antidepressant effect on behavioral depression induced by stress.     Effect of 45-day simulated microgravity on the evaluation of orally reported emergencies

    Accurate evaluation of emergencies is a critical concern in long-duration space flights. Accordingly, we studied the effect of 45 days of -6 degrees head-down bed rest - a model that simulates the conditions in microgravity environments - on the evaluation of orally reported emergencies. Sixteen male participants listened to corresponding emergency scenarios and assessed the severity of these situations eight times before, during and after bed rest. The results revealed a recency effect': compared with emergency descriptions in the order of serious to mild, those framed in the reverse order were judged to be more serious. However, the severity ratings did not vary with time spent in the simulated microgravity environment. These findings are similar to those observed in a regular environment on Earth, indicating that the design principles of information presentation for situations on Earth may also be extended to designs intended for outer space. Practitioner Summary: A recency effect was found in the evaluation of orally reported emergencies under simulated microgravity conditions. The design principles of information presentation for situations on Earth may also be extended to designs intended for outer space

    The Beijing Twin Study (BeTwiSt): A Longitudinal Study of Child and Adolescent Development

    Rates of emotional and behavioral problems among children and adolescents in China are increasing and represent a major public health concern. To investigate the etiology of such problems, including the effects and interplay of genes and environment, the Beijing Twin Study (BeTwiSt) was established. A representative sample of adolescent twins in Beijing (N = 1,387 pairs of adolescent twins, mostly between the ages of 10 and 18 years) was recruited and assessed longitudinally. Data collection included the following: emotional and behavioral problems (e.g., depressive symptoms, anxiety, delinquency, drinking, and smoking); family, peer, and school environments; stress; social and academic competence; cognitive traits (e.g., emotion suppression, rumination, and effortful control); and saliva samples for DNA genotyping and sequencing. The combination of quantitative and molecular genetic approaches and the timeliness of the project, with the sample residing in a region with a rapidly changing economic and cultural climate, are particular strengths of this study. Findings from this study are expected to help understanding of the etiological mechanisms underlying child and adolescent normal and abnormal development in regions undergoing substantial social, cultural, and economic changes

    Task complexity matters: The influence of trait mindfulness on task and safety performance of nuclear power plant operators

    People with high level of trait mindfulness are more likely to maintain an open and present-focused awareness and attention. Whereas a positive link between trait mindfulness and well-being has been established, its influence on real-world performance has not been fully addressed. In Study 1, we validated the Freiburg Mindfulness Inventory (FMI) in a Chinese sample (N = 294) and found that a two-dimensional solution (a presence factor and an acceptance factor) best fit the data. In Study 2, using this validated scale, we directly investigated the influence of trait mindfulness on task and safety performance. In a sample of 136 Chinese nuclear power plant operators, it was found that trait mindfulness interacted with task complexity to influence performance. For high-complexity-task holders (the control room operators), the presence factor was positively related to their task and safety performance; for the low-complexity-task holders (the field operators), the presence factor had no influence on safety performance but a negative influence on task performance. The acceptance factor did not have any meaningful influences. These results suggest that the benefit of being mindful outweighs its cost for complex but not simple tasks. Implications are discussed.     Contingent capture can occur at specific feature values: Behavioral and electrophysiological evidence

    The notion that attentional top-down control can be tuned to a stimulus feature is widely accepted. Although previous studies suggested that the stimulus-driven attentional capture could be contingent on top-down attentional control settings, it was uncertain whether contingent capture can occur at a specific feature value. Three experiments were conducted to address this issue using both behavioral and ERPs measures. Participants were required to respond to one color singleton in the search display (target) but refrain from responding to the search display containing another color singleton (nontarget). When target and nontarget belonged to different color categories (Experiment 1), only the target-color cue and within category irrelevant-color cue elicited the significant cue validity effect (i.e. RTs were shorter when the target was presented at the same location as the preceding cue rather than at a different location); they also lead to a robust N2pc effect, indicative of attention-capture. In addition, these two cue types had similar attention-capturing capacity. However, when target and nontarget belonged to the same color category (Experiments 2 and 3), only the target-color cue elicited the significant cue validity effect and the robust N2pc effect. The same within category irrelevant-color cue no longer elicited the cue validity effect, and the N2pc effect was also attenuated.     Environmental and behavioral controls of the expression of clozapine tolerance: Evidence from a novel across-model transfer paradigm

    Repeated administration of antipsychotic drugs induces a sensitization-like or tolerance-like effect in many behavioral tasks, including the conditioned avoidance response (CAR) and the phencyclidine (PCP)-induced hyperlocomotion, two rodent models with high predictive validity for antipsychotic activity. This study investigated the impacts of contextual and behavioral variables on the expression of clozapine tolerance using a recently validated across-model transfer paradigm (Zhang and Li, 2012 [1]). Male Sprague-Dawley rats were first repeatedly treated with clozapine (2.5-10.0 mg/kg, sc) in the CAR model or PCP (1.6 mg/kg, sc)-induced hyperlocomotion model for five consecutive days. They were then tested for the expression of clozapine tolerance in another model for another 5 days. Finally, all rats were switched back to the original model and tested again for the expression of clozapine tolerance. When tested in the PCP model, rats previously treated with clozapine in the CAR model did not show an immediate weaker inhibition of PCP-induced hyperlocomotion than those treated with clozapine for the first time, but showed a significantly weaker inhibition over time. In contrast, when tested in the CAR model, rats previously treated with clozapine in the PCP model showed an immediate weaker disruption of avoidance response than those treated with clozapine for the first time, but this weaker effect diminished over time. These results suggest that the expression of clozapine tolerance is strongly modulated by the test environment and/or selected behavioral response. Clozapine tolerance and its situational specificity may be related to the drug's low extrapyramidal motor side effect, its superior therapeutic efficacy and/or emergence of clozapine withdrawal syndrome.     Prospective memory predicts medication management ability and correlates with non-adherence to medications in individuals with clinically stable schizophrenia

    Background: Prospective memory (PM), the ability to remember to carry out an intended action in the future, is thought to relate closely to everyday functioning, such as medication adherence. PM impairment in schizophrenia may contribute to unintentional medication non-adherence. This study aimed to examine the relationship between PM, medication management and medication adherence. Methods: Eighty-two stable patients with schizophrenia underwent assessment for PM and medication management ability by laboratory measures at baseline. Clinical symptoms, other neuropsychological functions and risk factors known to associate with non-adherence were also measured. Linear regression was used to identify predictors of medication management ability at baseline. Actual medication adherence was assessed three months later and logistic regression was used to identify predictors of non-adherence. Results: In the linear regression model, time-and event-based PM together accounted for 72.3% of the variance in the performance of medication management at baseline. At three-month follow-up, the non-adherent group performed significantly more poorly in time-and event-based PM, and had poorer insight, more severe symptoms and poorer ability to manage medications, as compared to the adherent group. In the logistic regression model, insight and PANSS general score significantly predicted non-adherence in the community. Time- and event-based PM moderated the predictive power of insight and PANSS general score. Conclusions: Our results support that PM performance robustly predicts medication management ability and may influence medication adherence in the community by moderating the effect of insight and symptom severity. Optimizing PM performance may improve medication adherence in the community, particularly for unintentional non-adherence. (C) 2013 Elsevier B. V. All rights reserved

    The neural correlates of reward-related processing in major depressive disorder: A meta-analysis of functional magnetic resonance imaging studies

    Background: A growing number of functional magnetic resonance imaging (fMRI) studies have been conducted in major depressive disorder (MDD) to elucidate reward-related brain functions. The aim of this meta-analysis was to examine the common reward network in the MDD brain and to further distinguish the brain activation patterns between positive stimuli and monetary rewards as well as reward anticipation and outcome. Methods: A series of activation likelihood estimation (ALE) meta-analyses were performed across 22 fMRI studies that examined reward-related processing, with a total of 341 MDD patients and 367 healthy controls. Results: We observed several frontostriatal regions that participated in reward processing in MDD. The common reward network in MDD was characterized by decreased subcortical and limbic areas activity and an increased cortical response. In addition, the cerebellum, lingual gyms, parahippocampal gyrus and fusiform gyrus preferentially responded to positive stimuli in MDD, while the insula, precuneus, cuneus, PFC and inferior parietal lobule selectively responded to monetary rewards. Our results indicated a reduced caudate response during both monetary anticipation and outcome stages as well as increased activation in the middle frontal gyrus and dorsal anterior cingulate during reward anticipation in MDD. Limitations: The reward-related tasks and mood states of patients included in our analysis were heterogeneous. Conclusions: Our current findings suggest that there exist emotional or motivational pathway dysfunctions in MDD during reward-related processing. Future studies may be strengthened by paying careful attention to the types of reward used as well as the different components of reward processing examined. (C) 2013 Elsevier B.V.
