    Targeting Trypanothione Metabolism in Trypanosomatids

    [EN] Infectious diseases caused by trypanosomatids, including African trypanosomiasis (sleeping sickness), Chagas disease, and different forms of leishmaniasis, are Neglected Tropical Diseases affecting millions of people worldwide, mainly in vulnerable territories of tropical and subtropical areas. In general, current treatments against these diseases are old-fashioned, showing adverse effects and loss of efficacy due to misuse or overuse, thus leading to the emergence of resistance. For these reasons, searching for new antitrypanosomatid drugs has become an urgent necessity, and different metabolic pathways have been studied as potential drug targets against these parasites. Considering that trypanosomatids possess a unique redox pathway based on the trypanothione molecule absent in the mammalian host, the key enzymes involved in trypanothione metabolism, trypanothione reductase and trypanothione synthetase, have been studied in detail as druggable targets. In this review, we summarize some of the recent findings on the molecules inhibiting these two essential enzymes for Trypanosoma and Leishmania viability.S

    Social vulnerability assessment for flood risk analysis

    This article belongs to the Special Issue Flood Risk Assessments: Applications and Uncertainties[EN] This paper proposes a methodology for the analysis of social vulnerability to floods based on the integration and weighting of a range of exposure and resistance (coping capacity) indicators. It focuses on the selection and characteristics of each proposed indicator and the integration procedure based on the analytic hierarchy process (AHP) on a large scale. The majority of data used for the calculation of the indicators comes from open public data sources, which allows the replicability of the method in any area where the same data are available. To demonstrate the feasibility of the method, a study case is presented. The flood social vulnerability assessment focuses on the municipality of Ponferrada (Spain), a medium-sized town that has high exposure to floods due to potential breakage of the dam located upstream. A detailed mapping of the social vulnerability index is generated at the urban parcel scale, which shows an affected population of 34,941 inhabitants.     Characterization of the Gene Encoding S-adenosyl-L-methionine (AdoMet) Synthetase in Penicillium chrysogenum; Role in Secondary Metabolism and Penicillin Production

    [EN] The filamentous fungus Penicillium chrysogenum (recently reidentified as Penicillium rubens) is used in the industrial production of the β-lactam antibiotic penicillin. There are several mechanisms regulating the production of this antibiotic, acting both at the genetic and epigenetic levels, the latter including the modification of chromatin by methyltransferases. S-adenosyl-L-methionine (AdoMet) is the main donor of methyl groups for methyltransferases. In addition, it also acts as a donor of aminopropyl groups during the biosynthesis of polyamines. AdoMet is synthesized from L-methionine and ATP by AdoMet-synthetase. In silico analysis of the P. chrysogenum genome revealed the presence of a single gene (Pc16g04380) encoding a putative protein with high similarity to well-known AdoMet-synthetases. Due to the essential nature of this gene, functional analysis was carried out using RNAi-mediated silencing techniques. Knock-down transformants exhibited a decrease in AdoMet, S-adenosyl-L-homocysteine (AdoHcy), spermidine and benzylpenicillin levels, whereas they accumulated a yellow-orange pigment in submerged cultures. On the other hand, overexpression led to reduced levels of benzylpenicillin, thereby suggesting that the AdoMet synthetase, in addition to participate in primary metabolism, also controls secondary metabolism in P. chrysogenum.SIJunta de Castilla y Leó

    [ES] El Comité para la Eliminación de todas las formas de discriminación contra la Mujer (CEDAW) de la ONU, ha condenado recientemente a España por violencia obstétrica. La violencia obstétrica es un término que hace referencia a las malas prácticas profesionales en el parto y posparto, y que ha sido identificada como un tipo de violencia sistémica que aparece en todos los países, independientemente de factores como el estatus social, capacidad económica o nivel de estudios. Este tipo de violencia está arraigada en prácticas profesionales tradicionales y en una falta de consideración de la mujer como sujeto de derechos. En la contribución se estudian cuatro de los casos que llegaron al seno del CEDAW en los que se describen dichas prácticas profesionales sufridas por mujeres españolas y que conllevaron la condena a España por falta de protección de los derechos sexuales y reproductivos de la mujer

    Current and promising novel drug candidates against visceral leishmaniasis

    [EN] Leishmaniasis is a group of zoonotic diseases caused by a trypanosomatid parasite mostly in impoverished populations of low-income countries. In their different forms, leishmaniasis is prevalent in more than 98 countries all over the world and approximately 360-million people are at risk. Since no vaccine is currently available to prevent any form of the disease, the control strategy of leishmaniasis mainly relies on early case detection followed by adequate pharmacological treatment that may improve the prognosis and can reduce transmission. A handful of compounds and formulations are available for the treatment of leishmaniasis in humans, but only few of them are currently in use since most of these agents are associated with toxicity problems such as nephrotoxicity and cardiotoxicity in addition to resistance problems. In recent decades, very few novel drugs, new formulations of standard drugs or combinations of them have been approved against leishmaniasis. This review highlights the current drugs and Combinations that are used medical practice and recent advances in new treatments against leishmaniasis that were pointed out in the recent 2 nd Conference, Global Challenges in Neglected Tropical Diseases, held in San Juan, Puerto Rico in June 2018, emphasizing the plethora of new families of molecules that are bridging the gap between preclinical and first-in-man trials in next future.SIMinisterio de Economía y CompetitividadJunta de Castilla y Leó

    [ES] El Instituto de Biotecnología de León (INBIOTEC) es un Centro Tecnológico que surgió en León en 1991 para promocionar las actividades de investigación, control de calidad y desarrollo tecnológico de interés para las entidades vinculadas a la biotecnología con las que colabora. INBIOTEC inició su andadura bajo la dirección del Prof. Juan Francisco Martín, catedrático de Microbiología de la Universidad de León (ULE), quien, tras su jubilación, cedió el testigo en 2011 al Prof. Elías F. Rodríguez Ferri Catedrático de Sanidad Animal de la ULE) en la dirección gerente y al Prof. Rafael Balaña Fouce Catedrático de Toxicología de la ULE) en la dirección científica. Posteriormente, en 2016, la dirección del Instituto recayó en el Dr. Carlos Barreiro (Investigador de INBIOTEC). El Instituto de Toxicología de Castilla y León (INTOXCAL) nació en 1995, estrechamente ligado a la actividad industrial de León y al mundo de la ciencia. En el año 2010, se unió a INBIOTEC para crear juntos un centro de referencia tecnológica en León, donde, actualmente, constituye el área de Analítica de INBIOTEC. Tradicionalmente, INBIOTEC se ha dedicado al estudio de todos los aspectos relacionados con la obtención de metabolitos microbianos de interés industrial en el área farmacéutica, agroalimentaria o medioambiental. Esto incluye la selección de cepas productoras, la mejora de condiciones de cultivo, el estudio de la regulación etabólica, análisis ómicos, el escalado a nivel semi-industrial y la purificación de los compuestos de interés. Así, su participación en contratos directos con empresas, los servicios analíticos a terceros y el desarrollo de proyectos competitivos a nivel europeo, nacional y regional utilizando hongos, levaduras y bacterias, principalmente actinobacterias, le han permitido solicitar más de 15 patentes y realizar más de 400 publicaciones científicas en sus 25 años de existencia.S

    [ES] El dolor de espalda resulta uno de los principales problemas de salud a nivel mundial debido a su alta prevalencia (Lo et al., 2021; Meucci et al., 2015). Alrededor del 70-80% de la población general sufren dolor de espalda en algún momento de su vida (Vera-García et al., 2015). En muchos casos el dolor de espalda resulta inespecífico e inclasificable y acaba por resultar una afección de tipo crónico (Meucci et al., 2015). Uno de los principales factores de riesgo para el desarrollo de dolor de tipo lumbar es el sedentarismo (Mahdavi et al., 2021). Investigaciones recientes demuestran que la actividad sedentaria aumenta el riesgo de padecer dolor lumbar tanto en adultos como en niños y adolescentes (Mahdavi et al., 2021). Entre los niños y adolescentes la actividad sedentaria más común esta asociada a los largos periodos de sedestación en las aulas (McGill et al., 2006). Concretamente, diversos autores han investigado la influencia de la postura de sedestación mantenida sobre el desarrollo de dolor lumbar y cervical, acortamiento de la musculatura isquiotibial e incluso disminución de la calidad de vida (López et al., 2021; Recalde et al., 2023). En la actualidad existe un gran número de investigaciones sobre la higiene postural en las aulas y las estrategias de mejora y prevención (Borrego et al., 2018). Entre estas estrategias se incluyen programas de actividad física que buscan mejorar la condición física de los estudiantes (Flores-Paredes, 2020; Herbert, 2022; Neil- Sztramko et al., 2021). Algunas de las herramientas más usadas dentro de estos programas son los fisiobalones o también llamados “fitball” o, en español, fisiobalones (Behm et al., 2010; Pasco y Félix, 2018). El fisiobalón resulta una herramienta útil debido a su inestabilidad la cuál ayuda a la mejora del control motor, propiocepción y activación muscular tanto en jóvenes como en adulto

Alcaldes, Presidente de la Diputación Provincial, Sr. Vocal del Consejo General del Poder Judicial, Sr. Subdelegado del Gobierno, Delegado del Gobierno en Castilla y León; autoridades civiles, militares e institucionales de distinto orden que nos acompañan en este acto. Tengo el privilegio de presentar la siguiente laudatio en favor del Profesor que lo ha sido de esta Universidad, D. Dionisio Llamazares Fernández. Dionisio Llamazares, nació el seis de junio de 1936 en la pedanía de Yugueros, en La Ercina. Es el menor de los hijos de una familia humilde. Gracias a sus dotes y al impulso del maestro de la localidad, accedió a los estudios secundarios y, a pesar de las dificultades económicas, a cursar Derecho, obteniendo el título de Licenciado y, poco después, el de Doctor; ambos con las más altas calificaciones, este último, además, premio “José Fernández” a la mejor tesis doctoral defendida en la Universidad de Oviedo durante el bienio 66-68. Dos becas le permitieron continuar sus investigaciones en Estrasburgo y Munich y, a la postre, ejercer la condición de Profesor en las sedes de Oviedo, San Sebastián y León de la Universidad ovetense. En León, será Director del Colegio Universitario desde mediados los años setenta, época de la que se ha destacado su capacidad para incorporar relevantes figuras docentes e investigadoras, su tenacidad para dotar al Colegio de adecuados medios materiales y bibliográficos, así como la consolidación y promoción del profesorado local. Creó una muy valiosa colección de monografías con el apoyo de la Caja de Ahorros Provincial bajo la denominación “Colegio Universitario de León” y formó parte de la Comisión que exitosamente dio lugar a la transformación de los Centros de León en Universidad.     B- and T-Lymphocyte Attenuator Targeting Protects Against the Acute Phase of Graft Versus Host Reaction by Inhibiting Donor Anti-Host Cytotoxicity

B-and T-lymphocyte attenuator (BTLA) functions as a coinhibitory/costimulatory molecule that belongs to the immunoglobulin superfamily and exhibits a pattern of expression restricted to the hematopoietic compartment. Engagement of BTLA by its ligand, herpes virus entry mediator (HVEM), delivers negative signals to T cells, whereas engagement of HVEM receptor on T cells by surface BTLA expressed on other immune cells costimulates T activation. Previous work has reported that parental donor BTLA knock-out or HVEM knock-out T cells adoptively transferred into nonirradiated F1 recipient mice survived poorly, and the rejection of host hematopoietic cells was attenuated compared with F1 recipients receiving wild-type T cells. Methods. Parent into nonirradiated immunocompetent F1 murine model of acute graft versus host reaction, which is induced with the adoptive transfer of splenocytes from donor B6 mice (H-2 b) into F1 recipients (BALB/c×B6, H-2 a/b), was used as an experimental approach to test the therapeutic effect of targeting BTLA during the course of an allogeneic immune response. Results. We herein provide evidence that administration of an anti-BTLA monoclonal antibody leads to significant reduction of donor anti-host allogeneic immune response against bone marrow and thymus during the acute phase of graft versus host reaction in a parent into nonirradiated F1 murine model of alloreactivity. Anti-BTLA protection against donor anti-host hematopoietic cell rejection correlated with impaired anti-host cytotoxic T-lymphocyte activity than reduction in T-cell number infiltrating host tissues. Conclusions.     Penicillium chrysogenum var. halophenolicum, a new halotolerant strain with potential in the remediation of aromatic compounds in high salt environments

    [EN] A halotolerant phenylacetate-degrading fungus Penicillium CLONA2, previously isolated from a salt mine at Algarve (Portugal), was identified as a variant of P. chrysogenum using the ITS-5,8S rDNA and the D1/D2 domain of 28S rDNA sequences. The metabolic features and genetic characteristics suggest that this strain belongs to a subgroup of P. chrysogenum, named var. halophenolicum. The presence of the penicillin biosynthetic cluster was proven by Southern hybridizations using probes internal to the pcbAB and penDE genes and sequencing of the pcbAB-pcbC intergenic region. However the pcbAB-pcbC divergent promoter region contained 20 point modifications with respect to that of the wild type P. chrysogenum NRRL1951. The CLONA2 strain produced non-aromatic natural penicillins rather than benzylpenicillin in a medium containing potassium phenylacetate (the precursor of benzylpenicillin) and was able to grow well on phenylacetatic acid using it as sole carbon source. Due to the ability of P. chrysogenum CLONA2 to degrade aromatic compounds, this strain may be an interesting organism for aromatic compounds remediation in high salinity environments.SIUniversidade Nova de LisboaEuropean Unio
