Quantitative proteomic analysis of plasma membranes from the fish pathogen Saprolegnia parasitica reveals promising targets for disease control
Get PDF[EN] The phylum Oomycota contains economically important pathogens of animals and plants, including Saprolegnia parasitica, the causal agent of the fish disease saprolegniasis. Due to intense fish farming and banning of the most effective control measures, saprolegniasis has re-emerged as a major challenge for the aquaculture industry. Oomycete cells are surrounded by a polysaccharide-rich cell wall matrix that, in addition to being essential for cell growth, also functions as a protective “armor.” Consequently, the enzymes responsible for cell wall synthesis provide potential targets for disease control. Oomycete cell wall biosynthetic enzymes are predicted to be plasma membrane proteins. To identify these proteins, we applied a quantitative (iTRAQ) mass spectrometry-based proteomics approach to the plasma membrane of the hyphal cells of S. parasitica, providing the first complete plasma membrane proteome of an oomycete species. Of significance is the identification of 65 proteins enriched in detergent-resistant microdomains (DRMs). In silico analysis showed that DRM-enriched proteins are mainly involved in molecular transport and β-1,3-glucan synthesis, potentially contributing to pathogenesis. Moreover, biochemical characterization of the glycosyltransferase activity in these microdomains further supported their role in β-1,3-glucan synthesis. Promoter Engineering Reveals the Importance of Heptameric Direct Repeats for DNA Binding by Streptomyces Antibiotic Regulatory Protein–Large ATP-Binding Regulator of the LuxR Family (SARP-LAL) Regulators in Streptomyces natalensis
Get PDF[EN] The biosynthesis of small-size polyene macrolides is ultimately controlled by a couple of transcriptional regulators that act in a hierarchical way. A Streptomyces antibiotic regulatory protein–large ATP-binding regulator of the LuxR family (SARP-LAL) regulator binds the promoter of a PAS-LuxR regulator-encoding gene and activates its transcription, and in turn, the gene product of the latter activates transcription from various promoters of the polyene gene cluster directly. The primary operator of PimR, the archetype of SARP-LAL regulators, contains three heptameric direct repeats separated by four-nucleotide spacers, but the regulator can also bind a secondary operator with only two direct repeats separated by a 3-nucleotide spacer, both located in the promoter region of its unique target gene, pimM. A similar arrangement of operators has been identified for PimR counterparts encoded by gene clusters for different antifungal secondary metabolites, including not only polyene macrolides but peptidyl nucleosides, phoslactomycins, or cycloheximide. Here, we used promoter engineering and quantitative transcriptional analyses to determine the contributions of the different heptameric repeats to transcriptional activation and final polyene production. Optimized promoters have thus been developed. Deletion studies and electrophoretic mobility assays were used for the definition of DNA-binding boxes formed by 22-nucleotide sequences comprising two conserved heptameric direct repeats separated by four-nucleotide less conserved spacers. La solicitud de la prestación por jubilación de un trabajador que ha prestado servicios en España y Bélgica genera un conflicto respecto a las bases reguladoras de la misma y la aplicación de los Reglamentos Comunitarios o el Convenio bilateral suscrito entre ambos países en esta materia. Este artículo analiza los mecanismos creados por los Reglamentos Comunitarios y otras normas como los Convenios bilaterales de Seguridad Social suscritos por España para evitar que los trabajadores migrantes se vean perjudicados por esta situación.[EN] The rise in labour migration flows on a European level also affects the field of Social Security. The contribution record of migrant workers is interrupted in the country they leave. Applying for the retirement benefit of an employee who has worked in Spain and Belgium creates a conflict with respect to the regulatory bases of retirement and the application of EU regulations or the bilateral agreement signed between both countries in this matter. This article analyses the tools created by EU regulations and other regulations, such as bilateral social security agreements signed by Spain, to prevent migrant workers from beingjeopardised by this situation.S
Biosynthesis of Astaxanthin as a Main Carotenoid in the Heterobasidiomycetous Yeast Xanthophyllomyces dendrorhous
Get PDF[EN] Carotenoids are organic lipophilic yellow to orange and reddish pigments of terpenoid nature that are usually composed of eight isoprene units. This group of secondary metabolites includes carotenes and xanthophylls, which can be naturally obtained from photosynthetic organisms, some fungi, and bacteria. One of the microorganisms able to synthesise carotenoids is the heterobasidiomycetous yeast Xanthophyllomyces dendrorhous, which represents the teleomorphic state of Phaffia rhodozyma, and is mainly used for the production of the xanthophyll astaxanthin. Upgraded knowledge on the biosynthetic pathway of the main carotenoids synthesised by X. dendrorhous, the biotechnology-based improvement of astaxanthin production, as well as the current omics approaches available in this yeast are reviewed in depth.S

Initial Training of Primary Education Teachers in Nutrition: Review of Contents of Spanish University Curricula Based on ESD

habits, which are crucial for promoting balanced diets throughout life. This research aims to analyze
the nutritional training of teachers by reviewing the presence of nutritional education content in
their university education. The methodology employed is a descriptive documentary analysis
incorporating both qualitative and quantitative components, utilizing a document analysis of the
teaching guides (280 documents) from primary education degrees offered in Spain (67 universities).
The results reveal significant deficiencies in the nutritional content of the teaching guides, highlighting
gaps in the initial training of teachers. Among the inadequately addressed topics are food sovereignty,
food security, diseases related to poor nutrition, guidelines for healthy eating, and types of diets that
promote good health and respect the socioeconomic environment. Therefore, to meet some of the
challenges proposed by the 2030 Agenda and the SDGs, it would be advisable to provide primary
education degree students at Spanish universities with greater knowledge of nutrition-related aspects.S

Get PDF[ES] La colección Adonáis es, con diferencia, la más longeva dentro del panorama de la edición de poesía en la España de posguerra, y una de las más influyentes desde su creación en 1943. Aunque el grueso del catálogo de la colección está compuesto por obras originales en español, esta también ha dado cabida a la publicación de poesía traducida de diversas lenguas, mayoritariamente europeas, incluyendo algunas peninsulares, como el gallego o el catalán, pero también otras escasamente traducidas durante esta época. Dada la fecha en la que arrancaría Adonáis, la publicación de las obras de la colección se vio afectada necesariamente por el filtro de la censura franquista. A fin de comprender la recepción por parte de los censores de las traducciones editadas por Adonáis en la dictadura y los primeros años de la transición, este articulo analiza el catálogo completo de las obras traducidas publicadas en la colección durante el periodo comprendido entre 1943 y 1983, ano este último en el que se generaron los últimos expedientes de censura de libros. A tal efecto, la parte central de este análisis es el estudio de un catálogo que recopila información exhaustiva sobre todos los títulos traducidos de la colección durante esa época, tanto datos relativos a su publicación como otros relacionados con su tratamiento censorio, con el objetivo de entender los procesos de traducción, edición y recepción. Asimismo, se examina cuantas de estas obras fueron retraducidas tras este periodo, a fin de comprobar si dichos títulos circulan en versiones no censuradas. A fin de realizar este estudio, se comenzará con una explicación del contexto histórico en el que se enmarca la producción editorial de la colección, incluyendo un breve repaso de su creación y evolución y de la composición de sus equipos editoriales. A continuación se detalla la metodología utilizada en la recopilación y posterior análisis del catálogo y se comentan varios datos de publicación clave para entender su trayectoria. Posteriormente, se examinan los informes y las calificaciones censorias disponibles relacionados con los títulos de la colección, así como correspondencia clave entre censores y editores para entender la relación existente entre ambas partes. El estudio de estos aspectos pone de relieve tanto la importancia de Adonáis en la conformación del mercado editorial de posguerra como el papel fundamental de determinados miembros del equipo editorial en el tratamiento que recibirá la colección por parte de la censura franquista
