    Silver Microparticle and Submicron Wire - Polylactic Acid Composites for Additive Manufacturing

    This thesis explores the incorporation of silver microparticle and submicron wire additives into thermoplastic filament feedstock for fused filament fabrication (FFF) to create multifunctional three-dimensional (3D) printable composites. The impact of silver microparticle and submicron wire additives on mechanical behavior along with antibacterial effect of the silver microparticle and submicron wire additives on printed objects were assessed. Composite FFF filaments were fabricated by solution processing, granulation, and extrusion. Differential Scanning Calorimetry (DSC) was conducted to measure the glass transition and melting point temperatures of the composite filaments for 3D printing. The effect of the additive addition on the thermal properties and tensile mechanical performance was measured. Scanning Electron Microscopy (SEM) was used to analyze the composite microstructure and fracture behavior. The reduction in the growth of Escherichia coli (E. coli) was measured after exposure to FFF-printed composite test specimens with additive concentrations ranging from 0.0 to 10.0 weight percent

    Low dissolved oxygen, hypoxia, is a prevalent stressor in aquatic environments arising from natural and anthropogenic events. Included in the biological responses to hypoxia are changes in gene expression that are coordinated by the hypoxia-inducible factor (HIF) family of transcription factors. The paradigm for HIF signaling is that alpha subunits are degraded during normoxia, but degradation is suppressed at low oxygen, leading to the accumulation of HIF1alpha, dimerization with beta subunits, and regulation of target genes transcription. In fish, there is evidence that mRNA levels also increase during hypoxia, but these measurements are from experiments without corresponding protein levels. Therefore, in this study we addressed two questions. Do levels of HIF1alpha mRNA increase after short-term hypoxic exposure? Do levels of HIF1alpha mRNA correlate with protein levels measured in the same tissues? Fundulus grandis, a small, abundant, estuarine fish, was exposed to approximately 1 mg/l O2 for 6 or 24 h, after which RNA was extracted from liver, gills, ovary, and skeletal muscle. HIF1alpha mRNA was quantified by quantitative PCR. The results show that there were no changes in HIF1alpha mRNA under these conditions, even when HIF1alpha protein levels were significantly elevated. Our results support a role for protein stabilization, rather than new transcription in the initial response of fish to low oxygen, in accord with the paradigm developed in mammalian cells

    Co-option of the yolkless Oocyte Receptor for CRISPR/Cas9 may Induce Transient Sterility in Female Drosophila grimshawi flies

    Evolutionary developmental biology has historically used generalist model organisms like Drosophila melanogaster. The Hawaiian Drosophilidae radiation, which occurred just 25 million years ago, is an ideal lineage for work in Evo-Devo. Hawaiian flies make up 25% of the world’s Drosophila species and extreme diversity is seen throughout the group. D. grimshawi is a generalist Hawaiian picture-wing fly that has served as a model for the Hawaiian Drosophilidae radiation. However, D. grimshawi’s power and use as a model organism is constrained by our lack of ability to manipulate its genome. In this work, I attempted to edit the genome of D. grimshawi for the first time using a novel CRISPR/Cas9 delivery method known as ReMOT Control. However, my preliminary results may suggest that use of the DmP2C ligand for delivery of the CRISPR/Cas9 components causes injected D. grimshawi females to become transiently sterile, preventing genome editing from occurring with ReMOT Control

    A Machine Learning Method for the Prediction of Melt Pool Geometries Created by Laser Powder Bed Fusion

    A machine learning model is created to predict melt pool geometries of Ti-6Al-4V alloy created by the laser powder bed fusion process. Data is collected through an extensive literature survey, using results from both experiments and CFD modeling. The model focuses on five key input parameters that influence melt pool geometries: laser power, scanning speed, spot size, powder density, and powder layer thickness. The two outputs of the model are melt pool width and melt pool depth. The model is trained and tested by using the k fold cross validation technique. Multiple regression models are then applied to find the model that produces the least amount of error. Verification of the ML model was achieved by comparing the model results with experimental results and CFD results given the same parameter values throughout the models. The ML model results are consistent with the experimental and CFD results

These causes were used to create a concept matrix surrounding the medicalization of fatness that illustrated how these concepts are defined relationally, as well as how they combine to form the medical conception of the “obesity epidemic”. The concept matrix carries an indirect influence upon weight stigma that can lead to weight discrimination when fat folks seek out healthcare
