    α-Gal immunization positively impacts Trypanosoma cruzi colonization of heart tissue in a mouse model

    Chagas disease, caused by the protozoan parasite Trypanosoma cruzi, is a significant endemic infectious disease in Latin America and is spreading in the U.S. and Europe with the presence of its insect transmission vector. No approved vaccine against Chagas disease exists. We describe a vaccine candidate based on a carbohydrate found on the T. cruzi cell surface, linked in the vaccine to a virus-like particle that provides a strong and focused immune response. Mice were immunized and challenged with the Trypanosoma cruzi parasites from two strains (Y and Colombian). Vaccination conferred substantial protection of mice against infection, compared with the unvaccinated group. Vaccinated animals presented low parasitemia, increased production of pro-inflammatory cytokines IL-12 and IFN-gamma, decreased cardiomyocyte damage, and rapid clearance of parasite nests from heart tissue. These effects were especially significant at time points modeling chronic disease, an important consideration for this pathogen. We, therefore, believe this is a valuable path to pursue in the development of vaccines against Chagas disease. Chagas disease, caused by the parasite Trypanosoma cruzi, is considered endemic in more than 20 countries but lacks both an approved vaccine and limited treatment for its chronic stage. Chronic infection is most harmful to human health because of long-term parasitic infection of the heart. Here we show that immunization with a virus-like particle vaccine displaying a high density of the immunogenic alpha-Gal trisaccharide (Q beta-alpha Gal) induced several beneficial effects concerning acute and chronic T. cruzi infection in alpha 1,3-galactosyltransferase knockout mice. Approximately 60% of these animals were protected from initial infection with high parasite loads. Vaccinated animals also produced high anti-alpha Gal IgG antibody titers, improved IFN-gamma and IL-12 cytokine production, and controlled parasitemia in the acute phase at 8 days post-infection (dpi) for the Y strain and 22 dpi for the Colombian strain. In the chronic stage of infection (36 and 190 dpi, respectively), all of the vaccinated group survived, showing significantly decreased heart inflammation and clearance of amastigote nests from the heart tissue

    High-throughput profiling of metabolites has been used to identify metabolic changes in murine models as a response to the infection by the parasitic trematode Schistosoma. These investigations have contributed to our understanding on the pathogenesis of this tropical neglected disease, with a potential of helping diagnosis. Here, our study aimed to investigate the application of gas chromatography–mass spectrometry (GC/MS) on the profiling of urine metabolites from mice carrying infections by Schistosoma mansoni. Two larval infection doses created distinctive infection intensities in mice, whereby the heavily infected animals were found to release 25 times more eggs in faeces than lightly infected animals. Over 200 urine metabolites were identified from these animals by GC/MS, following two complementary derivatisation methods. A list of 14 individual metabolites with altered relative abundances between groups were identified. Most of the altered metabolites showed a trend of increased abundances in response to infection intensity, indicating host-specific metabolic alterations as a result of the disease. Hippurate, a metabolite which concentration is intimately modulated by the gut microbiota, was found to be highly correlated to infection intensity. Our study showed that urine metabolic profiling by GC/ MS can distinguish non-infected animals from those carrying light and heavy infections by S. mansoni, revealing metabolites associated to the infection and providing insights on the pathogenesis of schistosomiasis

    A dependência tecnológica do Sistema Único de Saúde (SUS), expressa no elevado déficit da balança comercial do Complexo Econômico-Industrial da Saúde (CEIS) brasileiro, prejudica a sustentabilidade do Sistema Nacional de Saúde, em virtude do alto custo de materiais importados, que podem ser majorados em situações de desvalorização cambial. Também reduz a capacidade de resposta do SUS em situações emergenciais, em que aumentos críticos na demanda, aliados a medidas protetivas governamentais que restringem exportações, geram indisponibilidade de materiais. Essa dependência está diretamente relacionada à fragilidade da base produtiva nacional, incapaz de atender a demanda, principalmente nos materiais de maior densidade tecnológica. Essa fragilidade dificulta a transformação do conhecimento científico, cuja geração tem se fortalecido, em novos produtos ou tecnologias inovadoras. Um dos métodos utilizados para fomentar o desenvolvimento da base produtiva nacional é o uso do poder de compra estatal, tanto por meio da disponibilização de condições diferenciadas para fornecedores locais, quanto pela indução à inovação, utilizando-se como atrativo o potencial de aquisição governamental. Este estudo analisa a prática das compras realizadas pela Fiocruz no período 2015-2020, com o objetivo de identificar pontos críticos que subsidiem a implementação ou ajustes de estratégias de utilização do poder de compra estatal para o fortalecimento da base produtiva do CEIS. Os dados comprovam que a instituição é altamente dependente de produtos e insumos importados, principalmente em suas unidades produtivas, Bio-Manguinhos e Farmanguinhos. Evidencia-se, também, que as compras junto a fornecedores estrangeiros ocorrem em maior proporção nas modalidades licitatórias em que se permite maior direcionamento da compra a fornecedores específicos. Com base nesses dados, e considerando-se os recentes retrocessos nas normas regulatórias que inviabilizam a aplicação das margens de preferência, sugeriu-se a revisão dos processos relacionados à definição de requisitos de compra, tais como padronização ou escolha de marcas específicas, e a própria especificação dos materiais, aliada a uma maior interação institucional com o mercado privado local, buscando identificar compatibilidades de demanda e potencial produtivo para a criação de soluções inovadoras. Sugere-se, ainda, que essas ações sejam realizadas por estrutura específica, a fim de evitar conflitos entre a função primária e derivada dos processos de compras.The technological dependence of the Unified Health System (UHS), expressed in the high trade deficit of the Brazilian Health Economic-Industrial Complex (HEIC), undermines the sustainability of the National Health System, due to the high cost of imported materials, which can be increased in situations of currency devaluation. It also reduces the UHS capacity to respond in emergency situations, where critical increases in demand, combined with governments protective measures that restrict exports, generate unavailability of materials. This dependence is directly related to the fragility of the national productive base, incapable of meeting the demand, especially in materials with higher technological density. This weakness hampers the transformation of scientific knowledge, whose generation has been strengthened, into innovative products or technologies. One of the methods used to foment development of the national productive base is the use of state purchasing power, both by providing differentiated conditions for local suppliers, and by inducing innovation, using the potential for government acquisition as an attraction. This study analyses the practice of purchases carried out by Fiocruz in the 2015-2020 period, aiming to define critical points that subsidize the implementation, or adjustments, of strategies related to the use of the state purchasing power to strengthen the productive base of the HEIC. The data proves that the institution is highly dependent on foreign products, mainly in its production units, Bio-Manguinhos and Farmanguinhos. It is also evident that purchases from foreign suppliers occur in greater proportion in the bidding modalities that allow greater targeting of specific suppliers. Based on these data, and considering the recent regression in regulatory standards that impedes the application of preference margins, it was suggested to review the processes related to the definition of purchase requirements, such as standardization or choice of specific brands, and the specification of materials itself, combined with greater institutional interaction with the local private market, seeking to identify compatibilities between demand and productive potential for the creation of innovative solutions. It is also suggested that these actions be carried out by a specific structure, to avoid conflicts between the primary and derived functions of procurement processes

    Identification of the sex chromosome system in a sand fly species, Lutzomyia longipalpis s.l.

    In many animal species, sex determination is accomplished by heterogamety i.e., one of the sexes produces two types of gametes, which upon fertilization will direct the development toward males or females. Both male (“XY”) and female (“ZW”) heterogamety are known to occur and can be easily distinguished when the sex-chromosomes are morphologically different. However, this approach fails in cases of homomorphic sex chromosomes, such as the sand fly Lutzomyia longipalpis s.l. (Psychodidae, Diptera), which is the main vector of visceral leishmaniosis in Brazil. In order to identify the heterogametic sex in L. longipalpis s.l., we did a whole-genome sequencing of males and females separately and used the “Y chromosome Genome Scan” (YGS) method to find sex-specific sequences. Our results, which were confirmed by PCR, show that L. longipalpis s.l. has XY system. The YGS method can be especially useful in situations in which no morpho logical difference is observed in the sex-chromosomes or when fresh specimens are not readily available

    Antecedentes: Faltam informações sobre o papel do uso crônico de hidroxicloroquina durante o SARS-CoV-2 surto. Nosso objetivo foi comparar a ocorrência de COVID-19 entre pacientes com doenças reumáticas em uso de hidroxicloroquina com indivíduos da mesma família que não tomaram o medicamento durante as primeiras 8 semanas de infecção viral comunitária. transmissão no Brasil. Métodos: Esta análise transversal de linha de base é parte de um estudo multicêntrico observacional de 24 semanas envolvendo 22 ambulatórios acadêmicos brasileiros. Todas as informações sobre os sintomas do COVID-19, epidemiológicos, clínicos e os dados demográficos foram registrados em uma plataforma específica baseada na web por meio de ligações telefônicas de médicos e estudantes de medicina. A COVID-19 foi definida de acordo com os critérios do Ministério da Saúde (BMH) do Brasil. Os testes de Mann-Whitney, Qui-quadrado e Exato de Fisher foram utilizados para análise estatística e dois Modelos de Regressão Logística Final binários de Wald teste foram desenvolvidos usando um método retroativo para a presença de COVID-19. Resultados: De 29 de março a 17 de maio de 2020, um total de 10.443 participantes foram inscritos, incluindo 5.166 (53,9%) pacientes com doença reumática, dos quais 82,5% tinham lúpus eritematoso sistêmico, 7,8% artrite reumatóide, 3,7% Síndrome de Sjögren e esclerose sistêmica de 0,8%. No total, 1822 (19,1%) participantes relataram sintomas de fu dentro nos 30 dias anteriores à inscrição, dos quais 3,1% preencheram os critérios do BMH, mas sem diferença significativa entre.Background: There is a lack of information on the role of chronic use of hydroxychloroquine during the SARS-CoV-2 outbreak. Our aim was to compare the occurrence of COVID-19 between rheumatic disease patients on hydroxychloroquine with individuals from the same household not taking the drug during the frst 8 weeks of community viral transmission in Brazil. Methods: This baseline cross-sectional analysis is part of a 24-week observational multi-center study involving 22 Brazilian academic outpatient centers. All information regarding COVID-19 symptoms, epidemiological, clinical, and demographic data were recorded on a specifc web-based platform using telephone calls from physicians and medical students. COVID-19 was defned according to the Brazilian Ministry of Health (BMH) criteria. Mann–Whitney, Chisquare and Exact Fisher tests were used for statistical analysis and two binary Final Logistic Regression Model by Wald test were developed using a backward-stepwise method for the presence of COVID-19. Results: From March 29th to May 17st, 2020, a total of 10,443 participants were enrolled, including 5166 (53.9%) rheumatic disease patients, of whom 82.5% had systemic erythematosus lupus, 7.8% rheumatoid arthritis, 3.7% Sjögren’s syndrome and 0.8% systemic sclerosis. In total, 1822 (19.1%) participants reported fu symptoms within the 30 days prior to enrollment, of which 3.1% fulflled the BMH criteria, but with no signifcant diference between
