Violence in the affective-sexual trajectories of young gay men: "new" settings and "old" challenges

A literatura brasileira sobre violência contra homens gays, geralmente, tem como foco a discriminação sofrida por esta população, mas pouco se problematiza a violência que eles sofrem nas relações familiares devido à sua orientação sexual, ou ainda, em suas relações ditas como "namoro" ou "ficar". Com o objetivo de discutir a presença de violências durante a trajetória afetivo-sexual dos jovens, este estudo qualitativo realizou entrevistas em profundidade a partir de um roteiro semiestruturado. Os resultados mostraram que há múltiplas faces de violência que ocorrem durante a infância e adolescência nas relações familiares, perpassando nos seus relacionamentos afetivo-sexuais na adolescência e juventude, incluindo violências sexuais, físicas, psicológicas e institucionais. As redes de apoio dos jovens são limitadas, como exemplo, a poucos amigos e ao acesso de blogs na internet. Nenhum profissional de saúde foi citado pelos jovens como fonte de ajuda. Destaca-se também a necessidade de debater a prevenção da violência e promoção da saúde destes jovens, ampliando o olhar para as várias formas contemporâneas de se relacionar intimamenteThis paper discusses the phenomenon of violence in the affective-sexual trajectories of young, cisgender gay men, from popular strata, in the metropolitan region of Rio de Janeiro, Brazil. Brazilian literature on violence against gay men generally focuses on the discrimination suffered by this population. However, the violence they are submitted to among family relationships due to their sexual orientation, or even their relationships known as "dating" or "hookup", is hardly discussed. This qualitative study used in-depth interviews based on a semi-structured guide to discuss violence during the young gay men's affective-sexual trajectory. The results evidenced multiple faces of violence during childhood and adolescence in family relationships, spanning their affective-sexual relationships in adolescence and youth, including sexual, physical, psychological, and institutional violence. Support networks are limited, for example, to a few friends and access to blogs on the Internet. No health professional was cited as a helping source. Caracterização fenotípica e funcional de células da imunidade inata no estabelecimento da malária pulmonar murina
A malária é uma doença infecto-parasitária causada por protozoários do gênero Plasmodium, e é um dos principais problemas de saúde pública mundial. A parasitose pode evoluir para formas graves com complicação pulmonar associada à síndrome do desconforto respiratório agudo (MA-SDRA). Infecções por P. falciparum, P. vivax, P. ovale ou P. knowlesi podem levar ao quadro respiratório grave de MA-SDRA em humanos. Nestes, assim como nos modelos experimentais murinos, a MA-SDRA é caracterizada pelo aumento da permeabilidade do endotélio da microvasculatura pulmonar e inflamação. Entretanto, pouco se sabe sobre os mecanismos imunoregulatórios associados à patogênese da MA-SDRA. O objetivo deste estudo foi investigar a cinética de recrutamento das células do sistema imune inato para o pulmão, e o perfil de polarização dos macrófagos alveolares após a infecção de camundongos C57BL/6 e BALB/c com Plasmodium berghei ANKA (PbA). Os camundongos C57BL/6 infectados apresentaram disfunção pulmonar e edema, este evidenciado pelo aumento no peso do órgão e no conteúdo de proteínas no lavado broncoalveolar (BAL) e interstício pulmonar. Os animais BALB/c parasitados, entretanto, apresentaram discreto aumento no peso do órgão e de proteínas quantificadas no BAL, alterações que não se refletiram em dano à função pulmonar. As subpopulações de células mieloides (monócitos inflamatórios, macrófago alveolar, neutrófilos e/ou eosinófilos) presentes no BAL e no tecido pulmonar foram analisadas por citometria de fluxo Observamos diferenças no número percentual e/ou total de monócitos inflamatórios e neutrófilos, em ambos os compartimentos, ao longo da infecção em animais BALB/c e C57BL/6. Embora com cinética diferenciada, os animais parasitados, de ambas as linhagens, apresentaram redução no percentual e número total de macrófagos alveolares ao longo da doença. Interessantemente, os macrófagos alveolares de camundongos C57BL/6 exibiram maior expressão de CD206 e de MHC classe II e, redução no percentual de células expressando a enzima iNOS. Além disso, foi detectado aumento na atividade da enzima arginase e de óxido nítrico, no BAL dos animais C57BL/6 infectados. Nenhuma variação foi notada na população de eosinófilos ao longo da infecção em ambas as linhagens. Análise do perfil de citocinas revelou, no BAL dos animais BALB/c e C57BL/6, níveis aumentados de TNF e IFN. Entretanto, no BAL dos camundongos C57BL/6 foi detectado um maior desbalanço quando relacionado com a IL-10, já nos camundongos BALB/c não observamos diferenças estatísticas. Em conclusão, nosso estudo mostrou que camundongos C57BL/6 e BALB/c apresentam uma dinâmica de populações celulares diferenciada no pulmão e distinta plasticidade dos macrófagos alveolares. Estes eventos podem estar associados ao desenvolvimento de complicações pulmonares observadas nos animais C57BL/6 e BALB/cMalaria is an infectious-parasitic disease caused by a protozoa of the genus Plasmodium, and is one of the main public health problems worldwide. The parasitosis may progress to severe forms with pulmonary complications associated with acute respiratory distress syndrome (MA-ARDS). Infection with P. falciparum, P. vivax, P. ovale or P. knowlesi can lead to severe respiratory symptoms of MA-ARDS in humans. In murine models of malaria, MA-SDRA is characterized by increased permeability of the pulmonary microvasculature endothelium and inflammation. However, little is known about the immunoregulatory mechanisms associated with the pathogenesis of MA-ARDS. The aim of this study was to investigate the kinetics of lung innate immune cell recruitment and the polarization profile of alveolar macrophages, after infection of C57BL/ 6 and BALB/c mice with Plasmodium berghei ANKA (PbA). C57BL/6 infected-mice showed pulmonary dysfunction and edema as attested by increased organ weight and protein content in the bronchoalveolar lavage (BAL) and pulmonary interstitium. In contrast, BALB/c mice showed a small increase in the organ weight and total proteins in the BAL, alterations that did not interfere with lung function. Subpopulations of myeloid cells (inflammatory monocytes, alveolar macrophages, neutrophils and/or eosinophils) present in the BAL and in the lung tissue were analyzed by flow cytometry We observed differences in the percentage and/or total number of inflammatory monocytes and neutrophils, in both compartments, in BALB/c and C57BL/6 mice, throughout the infection. Infected mice of both strains, although exhibiting different kinetics, showed reduction in the percentage and total number of alveolar macrophages, throughout the disease. Interestingly, alveolar macrophages of C57BL/6 mice exhibited higher expression of CD206 and MHC class II as well as reduction in the percentage of positive cells for iNOS enzyme. In addition, an increase in the activity of the enzyme arginase and nitric oxide was detected in BAL of the infected C57BL/6 mice. No changes were noticed in the eosinophil population over the course of infection in both strains. Cytokine analysis in BAL from animals BALB/c and C57BL/6 revealed increased levels of TNF and IFN. However, in the C57BL/6 mice, a greater imbalance was detected when related to IL-10, in BALB/c mice, we did not observe statistical differences. In conclusion, our study showed that C57BL/6 and BALB/c mice have different cellular dynamism in the lung and distinct alveolar macrophages polarization. These events may be associated with the development of pulmonary complications observed in animals C57BL/6 and BALB/c2022-05-2
A Sanger-based approach for scaling up screening of SARS-CoV-2 variants of interest and concern

Several tools and protocols have been shared since the beginning of the COVID-19 pandemic, but they need to be timely adapted to cope with SARS-CoV-2 evolution. Although whole-genome sequencing (WGS) of the virus genetic material have been widely used, it still presents practical difficulties such as high cost, shortage of available reagents in the global market, need of a specialized laboratorial infrastructure and well-trained staff. These limitations result in genomic surveillance blackouts across several countries. Here we propose a rapid and accessible protocol based on Sanger sequencing of a single PCR fragment that is able to identify and discriminate all SARS-CoV-2 variants of concern (VOCs) identified so far, according to each characteristic mutational profile at the Spike-RBD region (K417N/T, E484K, N501Y, A570D). Twelve COVID-19 samples from Brazilian patients were evaluated for both WGS and Sanger sequencing: three from P.2, two from P.1 and seven from B.1.1 lineage. All results from the Sanger sequencing method perfectly matched the mutational profile of VOCs and non-VOCs described by WGS. In summary, this approach allows a much broader network of laboratories to perform molecular surveillance of SARS-CoV-2 VOCs and report results within a shorter time frame, which is of utmost importance in the context of rapid public health decisions in a fast evolving worldwide pandemic.2030-03-2

El artículo discute el papel que juega la representación en la construcción del imaginario colectivo en relación a este delito, abordando la noción de cultura de la violación, la forma en que el delito de violación es representado en los medios de comunicación, así como los mitos perpetuados por los enfoques adoptados. Subsidies for Paleoparasitology: analysis of samples from an archaeological site in Northeastern Brazil and comparison of quantitative methods
Considerando que o interesse em melhor conhecer a relação parasi-to/hospedeiro/ambiente desde as primeiras civilizações, levou à indagações peculiares que cul-minaram na criação da Paleoparasitologia, e que esta apresenta a Patoecologia e Paleoepidemi-ologia como modernas abordagens de estudo, distintas, porém convergentes, o presente estu-do objetivou analisar paleoparasitologicamente amostras de sedimentos e coprólitos provenien-tes do Sítio Arqueológico Pedra do Tubarão, Pernambuco, Brasil e com igual relevância, veri-ficar, experimentalmente, a eficiência de técnicas quantitativas aplicadas em abordagens popu-lacionais. As amostras inéditas provenientes do sítio foram analisadas com vistas a confirmar a existência de parasito e diagnosticá-lo morfometricamente. Na fase experimental, três metodo-logias internacionalmente reconhecidas, americana, inglesa e coreana, foram aplicadas em co-prólito experimental e tiveram a eficiência de seus algoritmos testada, segundo a aplicação de um parâmetro, realizado em pool de fezes antes da dessecação extrema. Foram diagnosticados 70 ovos bem preservados de Spirometra sp. nas amostras oriundas no sítio arqueológico, resul-tado que conduziu à discussão de elementos arqueobioculturais desse achado. Resultados estes que levam à inferências, sob o prisma da Patoecologia, que o grupo populaci-onal que ocupava o sítio Pedra do Tubarão poderia estar potencialmente em risco de infecção zoonótica por Spirometra sp., e que a manutenção deste parasito, provavelmente de origem felina, no cotidiano dos indivíduos por longo período, corrobora com potencial chance de de-senvolvimento de esparganose como desfecho mais desfavorável para tal infecção. Na replica-ção dos métodos de quantificação de ovos por grama das escolas elencadas, foram aplicados testes estatísticos que nortearam a discussão da eficiência destes, bem como discutidos pontos peculiares de suas exequibilidades. O experimento comparativo possibilitou concluir que den-tre as três metodologias aplicadas, a americana e a coreana foram as que mais se aproximaram do parâmetro, sendo recomendadas como elegíveis para realização de mais estudos que visem o estabelecimento de quantificação \201Cpadrão ouro\201D em análises paleoepidemiológicas populaci-onais em sítios com boa preservação.Since the first civilizations, the desire to better understand the host-parasite-environment relationship has led, over the years, to peculiar questions that culminated in the creation of Paleoparasitology, which in turn presents Pathoecology and Paleoepidemiology as its modern study approaches, which are distinct, though converging. In this context, this study aimed to conduct a paleoparasitological analysis of sediment and coprolite samples from the Archaeolog-ical Site of Pedra do Tubarão in Pernambuco, Brazil, and, equally important, to verify experi-mentally the efficiency of quantitative techniques applied in population approaches. The new samples from the site underwent rehydration, sedimentation, and microscopic analysis to con-firm the parasite\2019s existence and diagnose it morphometrically. The experimental phase consist-ed of stages that applied three internationally recognized methodologies (American, English, and Korean) in equal rates of experimental coprolite and tested the efficiency of their algo-rithms, according to the parameter application, as control technique, performed in the stool pool before extreme desiccation. Seventy well-preserved eggs of Spirometra sp. were diagnosed in the samples from the archaeological site, a result that led to discussing several archaeobiocultur-al elements of Pathoecology in this finding. When replicating the methods of quantification of eggs per gram of the listed international schools, statistical tests were applied, which guided the discussion about their efficiency, as well as the description and discussion of specific points of their feasibility. Parasitological results of the samples led to inferences, from the perspective of Pathoecology, which considered biotic and abiotic conditions found in the area, in addition to the subsistence habits of this population, and that the population group that occupied the Pedra do Tubarão site could potentially be at risk of zoonotic infection by Spirometra sp. The preservation of this par-asite, probably of feline origin, in the daily life of individuals for a long period, corroborates the potential of developing sparganosis as the most unfavorable outcome for such infection. Aimed at obtaining subsidies for the application of paleoepidemiological methodologies, the compara-tive experiment performed allowed to conclude that among the three methodologies applied, the American and the Korean were the ones that came closest to the parameter, being recommended as eligible for further studies seeking to establish \201Cgold standard\201D quantification in paleoepide-miological population analyzes in well-preserved

Historical Biogeography and the Evolution of Hematophagy in Rhodniini (Heteroptera: Reduviidae: Triatominae)
The The Rhodniini tribe is one of the five tribes in the subfamily Triatominae and is notorious for its domestic blood-sucking pests and vectors of Trypanosoma cruzi across Latin America. The human and economic costs of the Chagas disease in the American tropics are considerable, and these insects are of unquestionable importance to humans. We used mitochondrial rDNA (16S), nuclear ribosomal RNA (28S) and wingless (Wg) sequences to perform phylogenetic analysis to derive trees based on parsimony and maximum likelihood. Nucleotide sequences were used in molecular-clock analyses to estimate time divergence between species of Rhodniini. The potential distribution of each species was modeled and compared with Kappa statistic. Multivariate niches with bioclimatic variables were used to describe differences between the species using discriminant analysis. The results of this study indicate that the Rhodniini originated 17.91 Mya ago. Rhodnius domesticus is the oldest species having its origin at 9.13 Mya. Rhodniini are closely related to Salyavatinae that are specialist termite predators and diverged from this subfamily 30.43 Mya. Most species are clearly allopatric and have distinct bioclimatic niches. The colonization of bromeliads, palms trees and bird nests represent important events for the speciation of these taxa. The hematophagous habit can be described as a scenario where Rhodniini’s ancestor could be pre-adapted for the invasion of bromeliads, palm trees, and bird nests where they would find significant water availability and thermal damping. These environments are widely used by vertebrate inquilines that would be the source of food for the species of Rhodniini. BCG vaccination to reduce the impact of COVID-19 in healthcare workers: Protocol for a randomised controlled trial (BRACE trial)

to unrelated pathogens. This off-target
effect could reduce
the impact of emerging pathogens. As a readily available,
inexpensive intervention that has a well-established
safety
profile, BCG is a good candidate for protecting healthcare
workers (HCWs) and other vulnerable groups against
COVID-19.
Methods and analysis This international multicentre phase
III randomised controlled trial aims to determine if BCG
vaccination reduces the incidence of symptomatic and severe
COVID-19
at 6 months (co-primary
outcomes) compared with
no BCG vaccination. We plan to randomise 10 078 HCWs
from Australia, The Netherlands, Spain, the UK and Brazil in
a 1:1 ratio to BCG vaccination or no BCG (control group). The
participants will be followed for 1 year with questionnaires
and collection of blood samples. For any episode of illness,
clinical details will be collected daily, and the participant will be
tested for SARS-CoV-
2
infection. The secondary objectives are
to determine if BCG vaccination reduces the rate, incidence,
and severity of any febrile or respiratory illness (including
SARS-CoV-
2),
as well as work absenteeism. The safety of BCG
vaccination in HCWs will also be evaluated. Immunological
analyses will assess changes in the immune system following
vaccination, and identify factors associated with susceptibility
to or protection against SARS-CoV-
2
and other infections.
Ethics and dissemination Ethical and governance approval
will be obtained from participating sites. Results will be
published in peer-reviewed
open-access
journals. The final
cleaned and locked database will be deposited in a data
sharing repository archiving system.
