    Detection of a SARS-CoV-2 variant of concern in South Africa

    Continued uncontrolled transmission of SARS-CoV-2 in many parts of the world is creating conditions for substantial evolutionary changes to the virus1,2. Here we describe a newly arisen lineage of SARS-CoV-2 (designated 501Y.V2; also known as B.1.351 or 20H) that is defined by eight mutations in the spike protein, including three substitutions (K417N, E484K and N501Y) at residues in its receptor-binding domain that may have functional importance3,4,5. This lineage was identified in South Africa after the first wave of the epidemic in a severely affected metropolitan area (Nelson Mandela Bay) that is located on the coast of the Eastern Cape province. This lineage spread rapidly, and became dominant in Eastern Cape, Western Cape and KwaZulu–Natal provinces within weeks. Although the full import of the mutations is yet to be determined, the genomic data—which show rapid expansion and displacement of other lineages in several regions—suggest that this lineage is associated with a selection advantage that most plausibly results from increased transmissibility or immune escape

Em 2011, consolidou o acesso aberto à produção científica, por meio do seu Repositório Institucional ARCA e em 2014 lançou a sua Política de Acesso Aberto ao Conhecimento, fortalecendo o acesso livre e gratuito ao conhecimento produzido pela instituição. Em 2017, criou o Grupo de Trabalho em Ciência Aberta, voltado para assessorar a elaboração do Termo de Referência: gestão e abertura de dados para pesquisa na Fiocruz, que subsidiou a elaboração da Política de gestão, compartilhamento e abertura de dados para pesquisa da FIOCRUZ, aprovada em setembro de 2020. Esses documentos apontam para a necessidade de se criar Planos de Gestão de Dados (PGD) alinhados aos princípios FAIR (Findable, Accessible, Interoperable, Reusable). A adoção de um PDG nas práticas científicas serve tanto para atender às exigências das agências de fomento e das revistas científicas, bem como um instrumento para o pesquisador criar estratégias de coleta, armazenamento, validação, segurança, preservação e compartilhamento dos seus dados, durante todo o ciclo de vida da pesquisa

    Evaluation of the compulsory notification strategy of acts of violence committed against the elderly in a Hospital in the Eastern Health Region of the Federal District

    Introdução: A violência contra pessoa idosa é um problema de saúde pública com previsão de notificação compulsória pelos profissionais de saúde. Apesar de constituir uma diretriz legal, a notificação da violência pelos profissionais envolvidos no acolhimento e assistência à população idosa ainda é uma prática pouco adotada nos serviços, o que concorre para a sua subnotificação. Esta atividade, se exercida de forma sistemática e com qualidade, pode contribuir para acionar a rede de proteção social e ampliar a oferta de serviços de proteção e de assistência na esfera pública. Esse estudo apresenta uma avaliação da estratégia de notificação compulsória dos atos de violência praticados contra a pessoa idosa em um Hospital da Região Leste de Saúde do Distrito Federal e teve como base normativa o Estatuto do Idoso, publicado em 2003. Metodologia: Trata-se de um estudo descritivo, com recorte temporal, utilizando método de natureza qualitativa. Foram empregadas as dimensões conformidade/fidelidade, priorizando a subdimensão de disponibilidade de insumos, e a qualidade técnica, por meio da subdimensão resolutividade para verificar se o componente "notificação" da vigilância da violência foi implantado de acordo com a previsão legal estabelecida. Resultados: A subnotificação dos casos de abusos contra esse segmento no HRL indica que a qualidade do acolhimento e do registro das informações é insuficiente para identificação dos casos de violência e sua notificação. Na análise dos entrevistados, a subnotificação pode ser atribuída à indisponibilidade da Ficha de Notificação nos setores, ao desconhecimento da obrigatoriedade do registro, a indefinição de fluxo interno das informações coletadas e a inexistência de capacitações voltadas para a instituição desta prática no âmbito hospitalar. Conclusão: A ausência do principal insumo para a realização do registro pode contribuir com a subnotificação dos casos suspeitos ou confirmados de violência, além de fragilizar o monitoramento dessas ações para o provimento de subsídios necessários ao acionamento das normas de proteção ao idoso na qual a unidade hospitalar tem um papel estratégico.Introduction: Violence against the elderly is a public health problem with mandatory notification by health professionals. Despite constituting a legal guideline, the notification of violence by professionals involved in welcoming and assisting the elderly population is still a little adopted practice in services, which contributes to its underreporting. This activity, if carried out systematically and with quality, can contribute to trigger the social protection network and expand the offer of protection and assistance services in the public sphere. This study presents an evaluation of the compulsory notification strategy of acts of violence committed against the elderly in a Hospital in the Eastern Health Region of the Federal District and was based on the Statute of the Elderly, published in 2003. Methodology: It is about a descriptive study, with a time frame, using a qualitative method. The compliance/fidelity dimensions were used, prioritizing the sub-dimension of availability of inputs, and technical quality, through the resoluteness sub-dimension, to verify whether the "notification" component of the surveillance of violence was implemented in accordance with the established legal provision. Results: The underreporting of cases of abuse against this segment in the HRL indicates that the quality of reception and recording of information is insufficient to identify cases of violence and their notification. In the analysis of the interviewees, underreporting can be attributed to the unavailability of the Notification Form in the sectors, the lack of awareness of the mandatory registration, the lack of definition of the internal flow of collected information and the lack of training aimed at the institution of this practice in the hospital environment. Conclusion: The absence of the main input for the registration can contribute to the underreporting of suspected or confirmed cases of violence, in addition to weakening the monitoring of these actions to provide the necessary subsidies to trigger the norms of protection for the elderly in which the unit hospital has a strategic role

    Reenacting the Birth of a Function: Functional Divergence of HIUases and Transthyretins as Inferred by Evolutionary and Biophysical Studies

    Transthyretin was discovered in the 1940s, named after its ability to bind thyroid hormones and retinol. In the genomic era, transthyretins were found to be part of a larger family with homologs of no obvious function, then called transthyretin-related proteins. Thus, it was proposed that the transthyretin gene could be the result of gene duplication of an ancestral of this newly identified homolog, later found out to be an enzyme involved in uric acid degradation, then named HIUase (5-hydroxy-isourate hydrolase). Here, we sought to re-enact the evolutionary history of this protein family by reconstructing, from a phylogeny inferred from 123 vertebrate sequences, three ancestors corresponding to key moments in their evolution-before duplication; the common transthyretin ancestor after gene duplication and the common ancestor of Eutheria transthyretins. Experimental and computational characterization showed the reconstructed ancestor before duplication was unable to bind thyroxine and likely presented the modern HIUase reaction mechanism, while the substitutions after duplication prevented that activity and were enough to provide stable thyroxine binding, as confirmed by calorimetry and x-ray diffraction. The Eutheria transthyretin ancestor was less prone to characterization, but limited data suggested thyroxine binding as expected. Sequence/structure analysis suggests an early ability to bind the Retinol Binding Protein. We solved the X-ray structures from the two first ancestors, the first at 1.46 resolution, the second at 1.55 resolution with well-defined electron density for thyroxine, providing a useful tool for the understanding of structural adaptation from enzyme to hormone distributor

    Scaling effect in COVID-19 spreading: The role of heterogeneity in a hybrid ODE-network model with restrictions on the inter-cities flow

Understanding the clinical characteristics, interactions with the environment, and the variables that favor or hinder its dissemination help the public authorities in the fight and prevention, leading for a rapid response in society. Using models to estimate contamination scenarios in real time plays an important role. Population compartments models based on ordinary differential equations (ODE) for a given region assume two homogeneous premises, the contact mechanisms and diffusion rates, disregarding heterogeneous factors as different contact rates for each municipality and the flow of contaminated people among them. This work considers a hybrid model for covid-19, based on local SIR models and the population flow network among municipalities, responsible for a complex lag dynamic in their contagion curves. Based on actual infection data, local contact rates (β) are evaluated. The epidemic evolution at each municipality depends on the local SIR parameters and on the inter-municipality transport flow. When heterogeneity of β values and flow network are included, forecasts differ from those of the homogeneous ODE model. This effect is more relevant when more municipalities are considered, hinting that the latter overestimates new cases. In addition, mitigation scenarios are assessed to evaluate the effect of earlier interventions reducing the inter-municipality flux. Restricting the flow between municipalities in the initial stage of the epidemic is fundamental for flattening the contamination curve, highlighting advantages of a contamination lag between the capital curve and those of other municipalities in the territories

    Assessing the antigenicity of different VP3 regions of infectious bursal disease virus in chickens from South Brazil

    Infectious bursal disease (IBD), also known as Gumboro disease, is a viral infection that causes mortality and immunosuppression in chickens (Gallus gallus). VP2 and VP3 are the major structural viral capsid components and are the most immunogenic proteins of IBD virus (IBDV). Reliable diagnostic tests using VP2 and VP3 produced in heterologous systems are important tools to control this infection. One advantage of an IBD diagnostic based on VP3, over those that use VP2, is that VP3 has linear epitopes, enabling its production in bacteria. We tested the suitability of recombinant VP3 (rVP3) as a diagnostic reagent in an enzyme-linked immunosorbent assay (ELISA). Compared with a commercial test, rVP3 ELISA showed high sensitivity and specificity as a diagnostic tool for vaccinated animals. In addition, rVP3, but not the commercial ELISA, was able to detect antibodies in nonvaccinated chickens, probably developed against circulating IBDV strains. It was possible the assessment of VP3 regions antigenicity using chicken antisera. The full-length recombinant VP3 can be used to assess post vaccination immunological status of chickens and its production is feasible and inexpensive. The evaluation of VP3 regions as candidates for general use in the diagnosis of IBD in chickens should be conducted with caution.     Lack of Evidence of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) Spillover in Free-Living Neotropical Non-Human Primates, Brazil

    O coronavírus 2 da síndrome respiratória aguda grave (SARS-CoV-2), o agente da doença de coronavírus 2019 (COVID-19), é responsável pela pior pandemia do século XXI. Como todos os coronavírus humanos, o SARS-CoV-2 se originou em um reservatório de vida selvagem, provavelmente de morcegos. Como SARS-CoV-2 se espalhou por todo o mundo em humanos, ele se espalhou para infectar uma variedade de animais não humanos espécies em ambientes domésticos, fazendas e zoológicos. Além disso, uma ampla gama de espécies, incluindo uma macaco neotropical, provaram ser suscetíveis à infecção experimental com SARS-CoV-2. Juntas, essas descobertas levantam o espectro do estabelecimento de novos ciclos enzoóticos do SARS-CoV-2. Para avaliar a exposição potencial de primatas não humanos de vida livre ao SARS-CoV-2, amostramos 60 macacos neotropicais vivendo nas proximidades de Manaus e São José do Rio Preto, dois hotspots para COVID-19 no Brasil. Nossos testes moleculares e sorológicos não detectaram evidências de infecção por SAR-CoV-2 entre essas populações. Embora este resultado seja tranquilizador, os esforços sustentados de vigilância de vida selvagem vivendo em estreita associação com populações humanas é garantido, dada a natureza estocástica de eventos de transbordamento e as enormes implicações do transbordamento do SARS-CoV-2 para a saúde humana.Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the agent of coronavirus disease 2019 (COVID-19), is responsible for the worst pandemic of the 21st century. Like all human coronaviruses, SARS-CoV-2 originated in a wildlife reservoir, most likely from bats. As SARS-CoV-2 has spread across the globe in humans, it has spilled over to infect a variety of non-human animal species in domestic, farm, and zoo settings. Additionally, a broad range of species, including one neotropical monkey, have proven to be susceptible to experimental infection with SARS-CoV-2. Together, these findings raise the specter of establishment of novel enzootic cycles of SARS-CoV-2. To assess the potential exposure of free-living non-human primates to SARS CoV-2, we sampled 60 neotropical monkeys living in proximity to Manaus and São José do Rio Preto, two hotspots for COVID-19 in Brazil. Our molecular and serological tests detected no evidence of SAR-CoV-2 infection among these populations. While this result is reassuring, sustained surveillance efforts of wildlife living in close association with human populations is warranted, given the stochastic nature of spillover events and the enormous implications of SARS-CoV-2 spillover for human health
