    A Revista Fitos é um periódico científico interdisciplinar, em acesso aberto, sem custos para autores e de difusão gratuita, com publicação em fluxo contínuo, organizado, gerenciado e editado pelo Centro de Inovação em Biodiversidade e Saúde (CIBS), do Instituto de Tecnologia em Fármacos (Farmanguinhos), da Fundação Oswaldo Cruz. Tendo a inovação em Biodiversidade e Saúde como escopo, o periódico publica artigos científicos inéditos e originais abordando a pesquisa, o desenvolvimento tecnológico e a inovação em Biodiversidade e Saúde e as inter-relações entre suas diversas dimensões. Os manuscritos deverão ser redigidos em português, inglês ou espanhol, do tipo: artigo de pesquisa, artigo de revisão, relato de experiência, comunicação breve, ensaio, perspectiva, resenha, carta e monografia de espécies da biodiversidade brasileira. Link da revista: https://periodicos.fiocruz.br/pt-br/revista-fitosO presente estudo trata-se de uma revisão sistemática sobre as possíveis interações medicamentosas do uso de plantas medicinais e fitoterápicos incluídos na Relação Nacional de Medicamentos Essenciais (RENAME). Para isso, foram pesquisadas as interações entre medicamentos alopáticos e as plantas medicinais: alcachofra, aroeira, babosa, cáscara sagrada, espinheira-santa, garra do diabo, guaco, hortelã pimenta, isoflavona de soja, plantago, salgueiro e unha de gato. Foram identificados inicialmente, 637 artigos, com uma seleção final de 61 artigos, desses os mais encontrados foram revisões de literatura (52%), estudos in vitro (23%), estudos em animais (11%), relatos de caso (7%), estudo descritivo observacional (5%) e ensaio clínico (2%). Verificou-se interações como, diminuição da absorção de medicamentos, interferência com enzimas do citocromo P450 e efeitos sinérgicos levando a toxicidade. Foram descritas diversas interações, tais como: varfarina e anticoagulantes com babosa, garra do diabo, guaco, isoflavonas de soja, plantago e salgueiro; diuréticos com alcachofra, babosa, cáscara sagrada e plantago; corticoides com babosa, cáscara sagrada e plantago; digitálicos ou digitoxinas com babosa, cáscara sagrada e plantago; anti-inflamatórios não esteroidas com garra do diabo e plantago. Verifca-se que o uso de fitoterápicos exige cautela a fim de evitar repercussões graves no que tange a possíveis interações.The present study is a systematic review on the possible drug interactions of the use of medicinal plants and herbal medicines included in the National List of Essential Medicines (RENAME). For this, the interactions between allopathic medicines and medicinal plants were investigated: artichoke, mastic, aloe, cascara sagrada, espinheira-santa, devil's claw, guaco, peppermint, soy isoflavone, plantago, willow and cat's claw. Initially, 637 articles were identified, with a final selection of 61 articles, of which the most found were literature reviews (52%), in vitro studies (23%), animal studies (11%), case reports (7%), descriptive observational study (5%) and clinical trial (2%). Interactions such as decreased drug absorption, interference with cytochrome P450 enzymes and synergistic effects leading to toxicity were observed. Several interactions have been described, such as: warfarin and anticoagulants with aloe, devil's claw, guaco, soy isoflavones, plantago and willow; diuretics with artichoke, aloe, cascara sagrada and plantago; corticosteroids with aloe, cascara sagrada and plantago; digitalis or digitoxins with aloe, cascara sagrada and plantago; non-steroidal anti-inflammatory drugs with devil's claw and plantain. It is verified that the use of herbal medicines requires caution in order to avoid serious repercussions in terms of possible interactions

Entretanto, para isso é necessário compreender como as plantas medicinais são utilizadas na comunidade. O objetivo do estudo foi conhecer o perfil de uso das plantas medicinais em Alfenas, Minas Gerais. Um questionário padronizado foi aplicado a uma amostra (n=422) da população em locais públicos. Trata-se de um estudo descritivo. Considerou-se os seguintes critérios de inclusão: ser maior de 18 anos de idade, residir no município sede do estudo e ter utilizado plantas medicinais em algum momento da vida. Os dados foram coletados por meio de entrevistas, tabulados com dupla conferência e analisados por estatística descritiva. Verificou-se que 61,8% dos participantes relataram utilizar ocasionalmente as plantas medicinais no tratamento de doenças. As plantas medicinais mais utilizadas pela população foram: hortelã (21%); erva-cidreira (17%); camomila (7,3%); alfavaca (4,3%) e boldo (4,1%). Os principais motivos de uso referidos foram: calmante (21%), resfriado (10,6%), gosto pessoal (9,2%) e alimentação (8,8%). Espera-se que os resultados sirvam para o planejamento de ações e consolidação dessa prática no cuidado em saúde em Alfenas, servindo de modelo para outros municípios brasileiros.The Ministry of Health has supported the implementation of phytotherapy in primary care. However, necessary to understand how medicinal plants are used in the community. The objective of the study was to know the profile of use of medicinal plants in a city in the south of Minas Gerais. A standardized questionnaire was applied to a sample (n = 422) of the population in public places. This is a descriptive study. The following inclusion criteria were considered: being over 18 years of age, living in the municipality where the study was conducted and having used medicinal plants at some point in life. The collected data were tabulated with double checking and analyzed using descriptive statistics. It was found that 61.8% of participants reported occasionally using medicinal plants to treat diseases. The medicinal plants most used by the population were: mint (21%); lemon balm (17%); chamomile (7.3%); lavender (4.3%) and boldo (4.1%). The main reasons for use mentioned were: soothing (21%), cold (10.6%), personal taste (9.2%) and food (8.8%). It is hoped that the results will serve to plan actions and consolidate this practice in health care in Alfenas, serving as a model for other Brazilian municipalities

    The search for biomarkers and treatments in Chagas disease: insights from TGF-Beta studies and immunogenetics

    Produção científica do Laboratório de Genômica Funcional e Bioinformática (vigente até jan/2023).Produção científica do Laboratório de Inovações em Terapias, Ensino e Bioprodutos.The anti-inflammatory cytokine transforming growth factor beta (TGF-β) plays an important role in Chagas disease (CD), a potentially life-threatening illness caused by Trypanosoma cruzi. In this review we revisited clinical studies in CD patients combined with in vitro and in vivo experiments, presenting three main sections: an overview of epidemiological, economic, and clinical aspects of CD and the need for new biomarkers and treatment; a brief panorama of TGF-β roles and its intracellular signaling pathways, and an update of what is known about TGF-β and Chagas disease. In in vitro assays, TGF-β increases during T. cruzi infection and modulates heart cells invasion by the parasite fostering its intracellular parasite cycle. TGF-β modulates host immune response and inflammation, increases heart fibrosis, stimulates remodeling, and slows heart conduction via gap junction modulation. TGF-β signaling inhibitors reverts these effects opening a promising therapeutic approach in pre-clinical studies. CD patients with higher TGF-β1 serum level show a worse clinical outcome, implicating a predictive value of serum TGF-β as a surrogate biomarker of clinical relevance. Moreover, pre-clinical studies in chronic T. cruzi infected mice proved that inhibition of TGF-β pathway improved several cardiac electric parameters, reversed the loss of connexin-43 enriched intercellular plaques, reduced fibrosis of the cardiac tissue, restored GATA-6 and Tbox-5 transcription, supporting cardiac recovery. Finally, TGF-β polymorphisms indicate that CD immunogenetics is at the base of this phenomenon. We searched in a Brazilian population five single-nucleotide polymorphisms (-800 G>A rs1800468, -509 C>T rs1800469, +10 T>C rs1800470, +25 G>C rs1800471, and +263 C>T rs1800472), showing that CD patients frequently express the TGF-β1 gene genotypes CT and TT at position -509, as compared to noninfected persons; similar results were observed with genotypes TC and CC at codon +10 of the TGF-β1 gene, leading to the conclusion that 509 C>T and +10 T>C TGF-β1 polymorphisms are associated with Chagas disease susceptibility. Studies in genetically different populations susceptible to CD will help to gather new insights and encourage the use of TGF-β as a CD biomarker

    Curine ameliorates lipopolysaccharide-induced acute lung injury by downregulating the TLR4/MD-2/NF-κB(p65) signaling pathway

Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq) .Acute lung injury is characterized by alveolar-capillary barrier rupture, neutrophil-mediated airway inflammation, and tissue hypoxia. Since no effective pharmacotherapy is currently available, alternatives to improve its condition are mandatory. Recent studies demonstrated that curine, a bisbenzylisoquinoline alkaloid from Chondrodendron platyphyllum (A.St.-Hil.) Miers, Menispermaceae, with anti-allergic and anti-inflammatory properties, inhibited lipopolysaccharide-mediated acute pulmonary response in mice. Therefore, this study aimed to investigate curine’s mechanism of action in a lipopolysaccharide-induced acute lung injury model. Curine inhibited the recruitment of inflammatory cells to the bronchoalveolar lavage fluid, mainly dependent on neutrophil migration, and restored the pulmonary architecture by reducing edema, vascular permeability, and the total protein content as well as the wet/dry ratio of the lung. Curine also decreased the TNF-α, IL-1β, and IL-6 production through downregulating the toll-like receptor 4 receptor expression and the nuclear factor-kappa B (p65) phosphorylation. In silico analysis demonstrated that curine made hydrophobic interactions with the Leu78, Ile80, Phe121, Ile124, Phe126, and Ile 152 amino acids of the hydrophobic cavity of the MD-2 receptor. This curine interaction presented stability during the simulation, remaining linked to the active site, indicating an antagonistic interaction with the molecular complex lipopolysaccharides/toll-likereceptor/4 myeloid differentiation factor 2

Saiba mais sobre a Rede Genômica Fiocruz em: http://www.genomahcov.fiocruz.br/The pandemic caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has impacted public health and the world economy and fueled a worldwide race to approve therapeutic and prophylactic agents, but so far there are no specific antiviral drugs. Understanding the biology of the virus is the first step in structuring strategies to combat it, and in this context several studies have been conducted with the aim of understanding the replication mechanism of SARS-CoV-2 in vitro systems. In this work, studies using transmission and scanning electron microscopy and 3D electron microscopy modeling were performed with the goal of characterizing the morphogenesis of SARS-CoV-2 in Vero-E6 cells. Several ultrastructural changes were observed—such as syncytia formation, cytoplasmic membrane projections, lipid droplets accumulation, proliferation of doublemembrane vesicles derived from the rough endoplasmic reticulum, and alteration of mitochondria. The entry of the virus into cells occurred through endocytosis. Viral particles were observed attached to the cell membrane and in various cellular compartments, and extrusion of viral progeny took place by exocytosis. These findings allow us to infer that Vero-E6 cells are highly susceptible to SARS-CoV-2 infection as described in the literature and their replication cycle is similar to that described with SARS-CoV and MERS-CoV in vitro models
