    Analysis of cell junctions in JAG1-deficient cochleae

    Presented on April 12, 2024 at the University of Rochester.Abstract: During cochlear development, Notch signaling plays an important role in determining the fates of hair cells and supporting cells. JAG1, a Notch ligand, is involved in lateral induction, the first of the two crucial roles Notch signaling has in the sensory regions. The JAG1 ligand is localized to supporting cells and, unlike other Notch ligands, continues to be expressed in the mature cochlea. However, deleting JAG1 early in development leads to fused stereocilia in the inner hair cells of the cochlea beginning at P(8), accompanied by hearing loss in the mice at 6 weeks of age indicated by raised ABR levels. Because JAG1 is localized to supporting cells, and the phenotype presents in hair cells, these results suggest a possible signaling interaction defect between hair cells and supporting cells. The three types of junctions in the mammalian cochlea are gap junctions which are involved in signal transmission, adherens junctions which are involved in calcium dependent intercellular adhesion, and tight junctions which serve as selectively permeable barriers between neighboring cells. Tight junctions and adherens junctions will be looked at using antibody staining. Primary antibodies for N-cadherin, which is localized to inner hair cells, and E-cadherin, which is localized to outer hair cells and Henson’s cells will be used to examine adherens junctions. Primary antibodies for ZO-1 and Occulin (OCC), which are both membrane proteins, will be used to examine tight junctions. I will discuss ways to optimize conditions for antibody staining of junction markers, to continue investigating potential abnormalities in hair cell-supporting cell interactions in the developing mouse cochlea. After antibody optimization, cochleae of different time points from JAG1-deficient mice will be stained with junction markers and imaged using confocal microscopy to look for alterations in expression or localization, indicating abnormalities in cell junctions.</p

    A groundbreaking text on the history of the use of patents in architecture.Although patents existed in Renaissance Italy and even in Confucian thought, it was not until the middle third of the nineteenth century that architects embraced the practice of patenting in significant numbers. Patents could ensure, as they did for architects' engineering brethren, the economic and cultural benefits afforded by exclusive intellectual property rights. But patent culture was never directly translatable to the field of architecture, which tended to negotiate issues of technological innovation in the context of the more abstract issues of artistic influence and formal expression. In Prior Art, scholar Peter Christensen offers the first full-scale monographic treatment of this complex relationship between art and invention.Christensen's method, a site-oriented approach steeped in multinational and multilingual archival work, is geared toward unifying fractured global histories of architectural patents through the distinct union of architectural, cultural, and legal history.     Using Virtual Reality to Study Spatial Mapping and Threat Learning

    This data is from a virtual reality threat conditioning study comparing those that learned the contingencies of the paradigm (learners) and those who did not. Participants collected flowers in a virtual environment one at a time, and after collecting a flower, they rated on a scale (0-9) how likely they were to get a shock (9 being more likely). Unknown to participants, half of the environment predicted shock (at reinforcement rates 35%, 50%, and 60%), while the other half was "safe". Participants also collected and returned objects to their original location 4 times each. 2 objects were located in the "safe" half, two were located in the dangerous half. The distance between where the participant placed the object and it's original location was measured. State and trait anxiety scores were measured before (state and trait) and after (state only) the conditioning task. Participants also completed the Raven's Progressive Matrices before the task. During the task, skin conductance level was collected as participants approached a flower (SCL) and after they collected the flower and rated the likelihood of shock but before receiving a shock (SCR). At the end of the task, participants were asked to name the objects they collected and their locations.     This presentation material supports a workshop presented at the 2024 RIT & UR Staff Conference.This lightning talk is about sharing the RCL Data Set Grant Program. This student-oriented service aims to help UR undergraduates and graduate students in the Arts, Sciences, and Engineering, the Warner School of Education, and Simon Business School obtain non-traditional data sets for their research studies and create data from their projects. Since this grant program launched in 2019, we have offered grants to 28 recipients from the Economics Dept and Political Science Dept. Earth and Environmental Sciences, Computer Science, and Simon Business School with a total amount of $34,910. Please come and listen to how this program works.</p
