License to Kill? - Disease Eradication Programs May Not be in Line with the Convention on Biological Diversity
Global human population growth is associated with many problems, such asrnfood and water provision, political conflicts, spread of diseases, and environmental destruction. The mitigation of these problems is mirrored in several global conventions and programs, some of which, however, are conflicting. Here, we discuss the conflicts between biodiversity conservation and disease eradication. Numerous health programs aim at eradicating pathogens, and many focus on the eradication of vectors, such as mosquitos or other parasites. As a case study, we focus on the "Pan African Tsetse and Trypanosomiasis Eradication Campaign," which aims at eradicating a pathogen (Trypanosoma) as well as its vector, the entire group of tsetse flies (Glossinidae). As the distribution of tsetse flies largely overlaps with the African hotspots of freshwater biodiversity, we argue for a strong consideration of environmental issues when applying vector control measures, especially the aerial applications of insecticides.rnFurthermore, we want to stimulate discussions on the value of speciesrnand whether full eradication of a pathogen or vector is justified at all. Finally, we call for a stronger harmonization of international conventions. Proper environmental impact assessments need to be conducted before control or eradication programs are carried out to minimize negative effects on biodiversity

The Role of DNA Methylation: Linking Exposure to Phenotype
DNA methylation, through 5-methyl- and 5-hydroxymethylcytosine (5mC and 5hmC) is considered to be one of the principal interfaces between the genome and our environment and it helps explain phenotypic variations in human populations. Initial reports of large differences in methylation level in genomic regulatory regions, coupled with clear gene expression data in both imprinted genes and malignant diseases provided easily dissected molecular mechanisms for switching genes on or off. However, a more subtle process is becoming evident, where small (<10%) changes to intermediate methylation levels were associated with complex disease phenotypes. This has resulted in two clear methylation paradigms. The latter "subtle change" paradigm is rapidly becoming the epigenetic hallmark of complex disease phenotypes, although we were currently hampered by a lack of data addressing the true biological significance and meaning of these small differences. The initial expectation of rapidly identifying mechanisms linking environmental exposure to a disease phenotype led to numerous observational/association studies being performed. Although this expectation remains unmet, there is now a growing body of literature on specific genes, suggesting wide ranging transcriptional and translational consequences of such subtle methylation changes. Data from the glucocorticoid receptor (NR3C1) has shown that a complex interplay between DNA methylation, extensive 5"UTR splicing and microvariability gives rise to the overall level and relative distribution of total and N-terminal protein isoforms generated. Additionally, the presence of multiple AUG translation initiation codons throughout the complete, processed, mRNA enables translation variability, hereby enhancing the translational isoforms and the resulting protein isoform diversity; providing a clear link between small changes in DNA methylation and significant changes in protein isoforms and cellular locations. Methylation changes in the NR3C1 CpG island, alters the NR3C1 transcription and eventually protein isoforms in the tissues, resulting in subtle but visible physiological variability. Implying external environmental stimuli act through subtle methylation changes, with transcriptional microvariability as the underlying mechanism, to fine-tune the total NR3C1 protein levels. The ubiquitous distribution of genes with similar structure as NR3C1, combined with an increasing number of studies linking subtle methylation changes in specific genes with wide ranging transcriptional and translational consequences, suggested a more genome-wide spread of subtle DNA methylation changes and transcription variability. The subtle methylation paradigm and the biological relevance of such changes were supported by two epigenetic animal models, which linked small methylation changes to either psychopathological or immunological effects. The first model, rats subjected to maternal deprivation, showed long term behavioural and stress response changes. A second model, exposing mice to early life infection with H1N1, illustrated long-term immunological effects. Both models displayed subtle changes within the methylome. Suggesting/Indicating that early life adversity and early life viral infection "programmed" the CNS and innate immune response respectively, via subtle DNA methylation changes genome-wide. The research presented in this thesis investigated the ever-growing roles of DNA methylation; the physiological and functional relevance of subtle small DNA methylation changes genome-wide, in particular for the CNS (MD model) and the immune system (early life viral infection model) ; and the evidence available, particularly from the glucocorticoid of the cascade of events initiated by such subtle methylation changes, as well as addressing the underlying question as to what represents a genuine biologically significant difference in methylation.DNS-Methylierung durch 5-Methyl- und 5-Hydroxymethylcytosin (5mC und 5hmC) gilt als eine der Hauptschnittstellen zwischen dem Genom und unserer Umwelt und hilft phänotypische Variationen in der menschlichen Population zu erklären. Erste Berichte über große Unterschiede im Methylierungsgrad in regulatorischen Regionen des Genoms, gepaart mit Genexpressionsdaten, sowohl in geprägten Genen, als auch malignen Erkrankungen, lieferten leicht zu entschlüsselnde molekulare Mechanismen zum Ein- und Ausschalten von Genen. Aktuell wird jedoch auch ein subtilerer Prozess ersichtlich, bei dem kleine (<10%) Veränderungen der intermediären Methylierungsgrade mit komplexen Krankheitsphänotypen in Verbindung gebracht werden können. Diese beiden Ansätze haben zu zwei Paradigmen geführt. Das letztgenannte Paradigma der "subtilen Veränderungen", entwickelt sich derzeit zum epigenetischen Kennzeichen für Phänotypen komplexer Krankheiten, allerdings noch eingeschränkt durch einen Mangel verfügbarer Daten, mit denen die echte biologische Signifikanz und Bedeutung dieser kleinen Unterschiede adressiert werden könnten. Die anfängliche Erwartung, rasch Mechanismen identifizieren zu können, die bestimmte Umweltbelastungen mit Krankheitsphänotypen koppeln, führte zur Veröffentlichung zahlreicher Beobachtungs- und Assoziationsstudien. Obwohl die eigentliche Erwartung nicht erfüllt werden konnte, verfügen wir durch diese Studien nun über eine weitreichende Menge an Information zu spezifischen Genen, die auf weitreichende transkriptionale und translationale Konsequenzen solcher subtiler Methylierungsänderungen hindeutet. In diesem Zusammenhang haben Daten zum Glukokortikoidrezeptor (NR3C1) gezeigt, dass ein komplexes Wechselspiel zwischen DNS-Methylierung, extensivem 5"UTR-Spleißen und Mikrovariabilität das Gesamtniveau sowie die relative Verteilung der erzeugten Gesamt- und N-terminalen Protein-Isoformen beeinflusst. Die Anwesenheit von mehreren AUG-Translationsinitiationscodons in der gesamten verarbeiteten mRNA ermöglicht Translationsvariabilität, wodurch die Diversität des Translations-Isoformen und die resultierenden Protein-Isoformen erhöht wird. Dies stellt eine eindeutige Verbindung zwischen kleinen DNS-Methylierungänderungen und signifikanten Veränderungen der Protein-Isoformen dar. Methylierungsänderungen in der NR3C1-CpG-Insel verändern die NR3C1-Transkription und schließlich die Protein-Isoformen in den Geweben, was zu einer dezenten, aber sichtbaren physiologischen Variabilität führt. Dies wiederum impliziert, dass externe Umweltreize durch subtile Methylierungsänderungen, zusammen mit der Transkriptionsmikrovariabilität als zugrundeliegendem Mechanismus, den gesamten NR3C1-Proteinspiegel feineinstellen. Die ubiquitäre Verteilung von Genen mit ähnlicher Struktur wie NR3C1 in Kombination mit einer zunehmenden Anzahl von Studien über die dezente Methylierungsänderungen in spezifischen Genen mit weitreichenden transkriptionellen und translationalen Konsequenzen, suggeriert eine genomweite Verbreitung von feinen DNS-Methylierungsänderungen und Transkriptionsvariabilität. Das subtile Methylierungsparadigma und die biologische Relevanz solcher Veränderungen wird durch zwei epigenetische Tiermodelle unterstützt, die kleine Veränderungen der Methylierung entweder mit psychopathologischen oder immunologischen Effekten verbinden. Das erste Modell, in dem Ratten mütterlicher Deprivation ausgesetzt werden, zeigt langfristige Veränderungen des Verhaltens und der Reaktion auf Stress. Ein zweites Modell, in dem juvenile Mäuse mit H1N1 Viren infiziert werden, zeigt langfristige immunologische Effekte. Beide Modelle beschreiben subtile Veränderungen innerhalb des Methyloms, und suggerieren, dass Belastungen und virale Infektionen in juvenilen Lebensphasen durch subtile DNS-Methylierungänderungen genomweit das ZNS und die angeborene Immunantwort "programmieren". Die in dieser Arbeit vorgestellte Forschung untersucht die stetig wachsende Rolle der DNS-Methylierung; die physiologische und funktionelle Relevanz subtiler Änderungen der DNA-Methylierung genomweit, insbesondere für das ZNS (Mütterliche Deprivation in Ratten) und das Immunsystem (Virale Infektion juveniler Mäuse); die bereits verfügbaren Daten, insbesondere zu Glukokortikoiden innerhalb der Eventkaskaden, die durch subtile Veränderungen der Methylierung ausgelöst werden; sowie darüber hinaus die Frage, was ein echten, biologisch signifikanten Unterschied in der Methylierung darstellt
Movement and Persistence: On the History of Jewish Emancipation in the Grand-Duchy of Mecklenburg-Schwerin, 1813-1869
Mit der Rechtsentwicklung der jüdischen Minderheit wird ein wesentliches Problem der Herausbildung der modernen Gesellschaft angesprochen. Am Beispiel des strukturell rückständigen Mecklenburgs läßt sich eindrucksvoll sehen, wie langwierig und widersprüchlich die gleichberechtigte Aufnahme einer nichtchristlichen Bevölkerungsgruppe in einem Territorium war, dessen ritterschaftlichen Eliten ein konfessionelles Selbstverständnis von Staat und Gesellschaft hatten. Anders aber als zu erwarten war, gab es im Rahmen der Landespolitik durchaus Chancen für eine dauerhafte Emanzipation der Juden, als die Landesgesetzgebung das Großherzogtum zweimal - 1813 und 1849 - mit an die Spitze der Emanzipationspolitik in Deutschland brachte. Die scharfen Einschnitte machen Mecklenburg zu einem Muster, das in besonderer Weise das Auf und Ab des jüdischen Emanzipationsprozesses in den deutschen Ländern symbolisiert. Wie die Arbeit zeigt, spielte christlich hergeleitete Judenfeindschaft im protestantischen Mecklenburg wohl eine große Rolle; die Ursachen für die Widerstände gegen eine Emanzipation der Juden aber waren eher dort zu suchen, wo konservative ständische Eliten nicht bereit waren, ihren Sonderstatus zugunsten einer strukturell offenen Gesellschaft aufzugeben. Hinsichtlich der Quellenbasis konnte der Autor auf eine Vielzahl zufällig erhalten gebliebener jüdischer Selbstzeugnisse zurückgreifen, so daß biographische Skizzen entstanden, die vielschichtige Einblicke in das sich wandelnde Selbstverständnis mecklenburgischer Juden zulassen und den überraschend großen Eigenanteil der jüdischen Führungsschicht am Prozeß ihrer rechtlichen Gleichstellung sichtbar machen.The development of the Jewish minority\'s legal position poses a major problem in the emergence of modern society. The structurally backward region of Mecklenburg provides an impressive example of how the inclusion of a non-Christian population group was both long-winded and fraught with inconsistencies, as the leading noble elites harbored a religious self-image of their state and socieity. Yet the territorial legislation did open chances for a lasting emancipation of the Jews on at least two occasions - 1813 and 1849 - when it brought the Grand-Duchy into the avant-garde of emancipaton policies in Germany. Sharp rifts, however, turned Mecklenburg into a prime example of the ups and downs in the Jewish emancipation process. As this study shows, the traditional Christian anti-Judaism probably played a major role in Protestant Mecklenburg; at the same time, resistance against emancipating the Jews was mainly found where conservative elites in the estates system were unwilling to give up their special status in favour of a structurally open society. The present study can draw on a large number of Jewish ego-documents, allowing for biographical sketches and complex insights into the changing self-images of Jews in Mecklenburg. They reveal that the leading strata of Jewish society were actively engaged in the process towards legal equality
Das neue Tarifeinheitsgesetz
Mit dem am 10. Juli 2015 in Kraft getretenen „Gesetz zur Tarifeinheit“ wird der durch das Bundesarbeitsgericht im Jahre 2010 aufgegebene Grundsatz der Tarifeinheit durch den Gesetzgeber reaktiviert. Das Ziel des Gesetzes ist es, die Funktionsfähigkeit der Tarifautonomie durch die Auflösung von Tarifkollisionen zu sichern. Schon das Gesetzgebungsverfahren wurde durch vielstimmige Gutachten und Stellungnahmen begleitet, die sich nicht nur mit der verfassungsrechtlichen Problematik der gesetzlichen Regelung auseinandersetzen, sondern auch mit deren Anwendung in der tarifrechtlichen Praxis. Denn mit dem Instrument der Verdrängung eines geltenden Tarifvertrags im jeweiligen Betrieb nach dem Mehrheitsprinzip in Fällen einer Tarifkollision hat der Gesetzgeber zugleich in vielerlei Hinsicht „Neuland“ betreten.

Effiziente PDE beschränke Formoptimierung in Formenräumen
Shape optimization is of interest in many fields of application. In particular, shape optimization problems arise frequently in technological processes which are modelled by partial differential equations (PDEs). In a lot of practical circumstances, the shape under investigation is parametrized by a finite number of parameters, which, on the one hand, allows the application of standard optimization approaches, but, on the other hand, unnecessarily limits the space of reachable shapes. Shape calculus presents a way to circumvent this dilemma. However, so far shape optimization based on shape calculus is mainly performed using gradient descent methods. One reason for this is the lack of symmetry of second order shape derivatives or shape Hessians. A major difference between shape optimization and the standard PDE constrained optimization framework is the lack of a linear space structure on shape spaces. If one cannot use a linear space structure, then the next best structure is a Riemannian manifold structure, in which one works with Riemannian shape Hessians. They possess the often sought property of symmetry, characterize well-posedness of optimization problems and define sufficient optimality conditions. In general, shape Hessians are used to accelerate gradient-based shape optimization methods. This thesis deals with shape optimization problems constrained by PDEs and embeds these problems in the framework of optimization on Riemannian manifolds to provide efficient techniques for PDE constrained shape optimization problems on shape spaces. A Lagrange-Newton and a quasi-Newton technique in shape spaces for PDE constrained shape optimization problems are formulated. These techniques are based on the Hadamard-form of shape derivatives, i.e., on the form of integrals over the surface of the shape under investigation. It is often a very tedious, not to say painful, process to derive such surface expressions. Along the way, volume formulations in the form of integrals over the entire domain appear as an intermediate step. This thesis couples volume integral formulations of shape derivatives with optimization strategies on shape spaces in order to establish efficient shape algorithms reducing analytical effort and programming work. In this context, a novel shape space is proposed.Formoptimierung ist in vielen Anwendungsbereichen von großem Interesse. Insbesondere entstehen Formoptimierungsprobleme in technologischen Prozessen, die mit Hilfe von partiellen Differentialgleichungen (PDEs) modelliert werden. In vielen praktischen Anwendungen ist die zu untersuchende Form durch endlich viele Parameter charakterisiert. Einerseits ermöglicht dies die Anwendung von Standardoptimierungsansätzen, andererseits wird der Raum der erreichbaren Formen unnötig begrenzt. Einen Weg dieses Dilemma zu umgehen liefert das Formenkalkül. Bisher wurden die auf dem Formenkalkül basierenden Formoptimierungprobleme jedoch hauptsächlich mit Gradientenabstiegsverfahren gelöst. Die im Allgemeinen vorhandene Unsymmetrie der zweiten Formableitung beziehungsweise der Form-Hesse-Matrix ist ein Grund hierfür. Generell sind Formenräume keine linearen Räume, wodurch eine große Lücke zwischen Formoptimierung und der herkömmlichen PDE-beschränkten Optimierung entsteht. Diese Lücke gilt es zu schließen. Falls kein linearer Raum in Frage kommt beziehungsweise vorhanden ist, dann fällt die nächst beste Wahl auf eine Riemannsche Mannigfaltigkeit. Möchte man auf Riemannschen Mannigfaltigkeiten optimieren, so muss man mit dem Riemannschen Form-Gradienten beziehungsweise der Form-Hesse-Matrix arbeiten. Die Riemannsche Form-Hesse-Matrix besitzt im Gegensatz zu der herkömmlichen Form-Hesse-Matrix die häufig gewünschte Eigenschaft der Symmetrie. Solch eine Hesse-Matrix gibt Auskunft über die Wohlgestelltheit eines Optimierungsproblems und definiert die hinreichenden Optimalitätsbedingungen. Im Allgemeinen wird sie verwendet, um gradientenbasierte Formoptimierungsverfahren zu beschleunigen. Diese Arbeit befasst sich mit PDE-beschränkten Formoptimierungsproblemen, welche als Optimierungsprobleme auf Riemannschen Mannigfaltigkeiten betrachtet werden, um effiziente Methoden auf Formenräumen für diese bereitzustellen. Es werden Lagrange-Newton, sowie quasi-Newton Ansätze für PDE-beschränkte Probleme in dieser Arbeit formuliert. In diesen Methoden wird die Hadamard-Form der Formableitung verwendet. Diese Hadamard-Form ist ein Oberflächenintegral und oftmals sehr aufwendig und zeitintensiv herzuleiten. Als Zwischenergebnis dieser Herleitung erhält man ein Volumenintegral, welches aus mehreren Gesichtspunkten viel attraktiver ist als ein Oberflächenintegral. Daher beschäftigt sich diese Arbeit zusätzlich mit der Frage, wie man dieses Volumenintegral in den in dieser Arbeit entwickelten Formoptimierungsmethoden verwenden kann, um Effizienz durch Reduzierung analytischer Arbeit und des Programmieraufwandes zu erreichen. Der Aufsatz analysiert den Film "Karl May" von Hans Jürgen Syberberg, welcher am 18.10.1974 in München uraufgeführt wurde. Der Film bildet den mittleren Teil der Film-Trilogie, die Syberberg mit "Ludwig " Requiem für einen jungfräulichen König" begonnen und mit "Hitler, ein Film aus Deutschland" abgeschlossen hatte. Die Filmbiografie "Karl May", produziert von Bernd Eichinger und finanziert von der TMS Film GmbH (München) sowie Zweites Deutsches Fernsehen (ZDF) (Mainz), stellt den alternden Schriftsteller Karl May, gespielt von Helmut Käutner, in den Mittelpunkt - einen Autor, der sich an seinen eigenen Legenden psychisch abarbeitete und nach literarischer Anerkennung strebte. rn- Der Aufsatz gibt einen Vortrag wieder, der von Annette Deeken auf einer Tagung der Karl May-Gesellschaft am 1. A Machine Learning Method for Co-Registration and Individual Tree Matching of Forest Inventory and Airborne Laser Scanning Data
Determining the exact position of a forest inventory plot—and hence the position of the sampled trees—is often hampered by a poor Global Navigation Satellite System (GNSS) signal quality beneath the forest canopy. Inaccurate geo-references hamper the performance of models that aim to retrieve useful information from spatially high remote sensing data (e.g., species classification or timber volume estimation). This restriction is even more severe on the level of individual trees. The objective of this study was to develop a post-processing strategy to improve the positional accuracy of GNSS-measured sample-plot centers and to develop a method to automatically match trees within a terrestrial sample plot to aerial detected trees. We propose a new method which uses a random forest classifier to estimate the matching probability of each terrestrial-reference and aerial detected tree pair, which gives the opportunity to assess the reliability of the results. We investigated 133 sample plots of the Third German National Forest Inventory (BWI, 2011"2012) within the German federal state of Rhineland-Palatinate. For training and objective validation, synthetic forest stands have been modeled using the Waldplaner 2.0 software. Our method has achieved an overall accuracy of 82.7% for co-registration and 89.1% for tree matching. With our method, 60% of the investigated plots could be successfully relocated. The probabilities provided by the algorithm are an objective indicator of the reliability of a specific result which could be incorporated into quantitative models to increase the performance of forest attribute estimations

A Satellite-Based Imaging Instrumentation Concept for Hyperspectral Thermal Remote Sensing
This paper describes the concept of the hyperspectral Earth-observing thermal infrared (TIR) satellite mission HiTeSEM (High-resolution Temperature and Spectral Emissivity Mapping). The scientific goal is to measure specific key variables from the biosphere, hydrosphere, pedosphere, and geosphere related to two global problems of significant societal relevance: food security and human health. The key variables comprise land and sea surface radiation temperature and emissivity, surface moisture, thermal inertia, evapotranspiration, soil minerals and grain size components, soil organic carbon, plant physiological variables, and heat fluxes. The retrieval of this information requires a TIR imaging system with adequate spatial and spectral resolutions and with day-night following observation capability. Another challenge is the monitoring of temporally high dynamic features like energy fluxes, which require adequate revisit time. The suggested solution is a sensor pointing concept to allow high revisit times for selected target regions (1"5 days at off-nadir). At the same time, global observations in the nadir direction are guaranteed with a lower temporal repeat cycle (>1 month). To account for the demand of a high spatial resolution for complex targets, it is suggested to combine in one optic (1) a hyperspectral TIR system with ~75 bands at 7.2"12.5 -µm (instrument NEDT 0.05 K"0.1 K) and a ground sampling distance (GSD) of 60 m, and (2) a panchromatic high-resolution TIR-imager with two channels (8.0"10.25 -µm and 10.25"12.5 -µm) and a GSD of 20 m. The identified science case requires a good correlation of the instrument orbit with Sentinel-2 (maximum delay of 1"3 days) to combine data from the visible and near infrared (VNIR), the shortwave infrared (SWIR) and TIR spectral regions and to refine parameter retrieval
