Epigenetic consequences of early-life infection in a mouse model
Environmental exposures during susceptible early-life developmental periods can have the ability to model and shape individuals immunological responses in adulthood. This has been partly demonstrated in animal models, highlighting the long-term consequences of early-life exposure to bacterial infection in adulthood but the mechanisms driving and maintaining the immune early life programming are not yet fully understood. In this thesis, we investigated the epigenetic mechanisms, mainly DNA methylation, controlling the establishment/maintenance of early-life programming by bacterial and viral exposure in mice. Initially, it was important to carefully consider the sequencing method which would deliver a genome-wide DNA methylation profile of high quality to be able to identify LPS-programming specific methylation signature. For this purpose, we investigated the cutting pattern of restriction enzymes used for reduced representation CpG sequencing in order to control the number of dynamically regulated CpG sites interrogated. CpGs in CGI and shelf/shore could be enriched by the enzymes MspI, HhaI and BstUI, particularly in gene bodies for all genomic regions, promoters (TSS1500, TSS200), intra- (1st exon, gene body, 3’UTR, 5’UTR) and inter-genic regions. The enzyme HpyCH4IV mainly enriched CpG elements in the open sea for all genomic elements. This study allowed us to make an informed decision about the enzymes best fitted to identify DNA methylation patterns associated with LPS and H1N1-programming.
To understand the mechanisms controlling the establishment/maintenance of early-life programming in mice, we characterised LPS-programmed lymphocytes in vivo and in vitro. A single exposure to LPS at post-natal day 14 elicited stable, sex specific, long-term hypo
responsiveness of both the in-vivo and ex-vivo immune response to a homotypic LPS re-exposure in adulthood. In addition, both the HPA and HPG axes were concurrently programmed with blunted corticosterone after an acute stress and reduced circulating testosterone levels. Genome wide DNA methylation analysis identified a similar number of hyper- and hypo-methylated loci in LPS-programmed mice, spread across the genome specifically in intragenic regions. The programming associated phenotype was trans-generationally inherited in an oscillatory manner to the two subsequent generations. This suggests compensation efforts from the offspring phenotype to match and best adapt to their living environment. Several sperm miRNAs were found differentially expressed in LPS-programmed males, suggesting a probable route for the transmission of programming to next generations. Next, we adopted a similar approach to investigate the consequences of neonatal exposure to influenza virus. Viral respiratory tract infections are highly prevalent during early-life and have a long-lasting, profound, impact on both neurodevelopment and the subsequent risk for developing allergy and asthma. Little is known about the long-term effects on the innate immune system. BALB/c mice exposed to Influenza A (H1N1, A/Puerto Rico/8/1934) at PND14 had increased serum IL-6, MIP-β and RANTES (p<0.05) when re-exposed to H1N1 in adulthood, confirming long-term immune programing.
Similar effects were observed after re-stimulating H1N1 programed mice with polyI:C and homotypic polyI:C programming and re-stimulation, suggesting preferential programming of the innate immune system. Reduced-representation epigenome sequencing identified a network of
methylation changes common to both early life polyI:C and H1N1 programming, leaving a mechanistic ‘trace’ that remained visible throughout life. Taken together our data suggest that neonatal exposure to infections induces an early activation of the innate immune system which coincides with cellular developmental peaks, hence epigenetically and transcriptionally programming cellular functions until adulthood. Early-life infection with either LPS or H1N1 was shown to lead to profound DNA methylation and transcriptional re-programming of immune cells. The pathway and network analysis of differentially methylated genes revealed that the immune system was at the centre of a number of transcriptionally modified clusters, in accordance with the diverse consequences of early-life LPS programming on the immune, neurological and hormonal systems

Shifting aspect or elevation? The climate change response of ectotherms in a complex mountain topography
Climate change is expected to cause mountain species to shift their ranges to higher elevations. Due to the decreasing amounts of habitats with increasing elevation, such shifts are likely to increase their extinction risk. Heterogeneous mountain topography, however, may reduce this risk by providing microclimatic conditions that can buffer macroclimatic warming or provide nearby refugia. B/ordering the Anthropocene: Inter- and Transdisciplinary Perspectives on Nature-Culture Relations
In und mit diesem Themenheft möchten wir zur produktiven Grenzarbeit einladen und das Verhältnis von Natur und Kultur im Anthropozän kritisch ausleuchten. Vor einigen Jahren wurde der Begriff Anthropozän als Bezeichnung für die gegenwärtige erdgeschichtliche Epoche, in der der Mensch als die zentrale Triebkraft für die globalen Veränderungen ökologischer Systeme angesehen wird, von Paul Crutzen vorgeschlagen. Diese Epoche zeichnet sich sowohl durch das Verschwimmen von Grenzlinien zwischen Gesellschaft und Natur, Wissenschaft und Politik als auch durch verstärkte Grenzziehungen zwischen sozialen Gruppen, Lebensstilen sowie Globalem Norden und Globalem Süden aus. Mit diesem Themenheft möchten wir einen Anstoß geben, Grenzphänomene in den Beziehungen zwischen Natur und Gesellschaft auszuloten, die bislang wenig im Mittelpunkt der Border Studies standen. Die Herausforderungen und Probleme des Anthropozäns bedürfen eines grenzüberschreitenden Denkens und Forschens, das zu einer neuen Reflexivität und einem neuen Engagement anregt, wozu das multidisziplinäre Feld der Border Studies beitragen kann.In and with this thematic issue we would like to invite you to engage in productive boundary work and to critically examine the relationship between nature and culture in the Anthropocene. A few years ago, the term Anthropocene was proposed by Paul Crutzen as a term for the current geological epoch, in which humankind (the ‘anthropos’) is seen as the central driving force for global changes in ecological systems. This epoch is characterized by the blurring of boundaries between society and nature, science and politics, as well as by the increased drawing of boundaries between social groups, lifestyles, and the Global North and Global South. With this issue, we would like to give an impetus to explore boundary phenomena in the relationship between nature and society, which up to now have not been the focus of Border Studies. The challenges and problems of the Anthropocene require cross-border thinking and research that stimulates a new reflexivity and commitment, to which the multidisciplinary field of Border Studies can contribute.Dans et avec ce cahier thématique, nous aimerions vous inviter à vous engager dans un travail frontalier productif et à examiner de manière critique la relation entre la nature et la culture dans l'Anthropocène. Validation of the vegetative and generative state of growth of vines in varying management and stress systems
Grundlage der Arbeit stellten zwei Arbeitshypothesen dar, die es zu überprüfen galt. Die beiden Arbeitshypothesen wurden aufgrund einer Beobachtung eines Phänomens im Freiland formuliert, da bei scheinbar unterschiedlich starkem Reblausbesatzes im Boden kein direkter Zusammenhang zu einer schädigenden Wirkung dieses Stressphänomens (Reblausbefall) hergestellt werden konnte. Im Gegenteil traten die schädigenden Wirkungen des Reblausbefalls in Form von Rückgangserscheinungen eher in Rebflächen auf, die einer mangelhaften Bewirtschaftung oder Bestandsführung unterlagen. Zur Klärung dieses Phänomens wurden daher die beiden (A., B.) folgenden Hypothesen aufgestellt, die es zu überprüfen galt:
A.: Die in mit Unterlagsrebsorten der Kreuzung V. berlandieri x V. riparia zu beobachtenden Rückgangserscheinungen in Rebanlagen mit Reblausvorkommen werden nicht allein durch die Saugtätigkeit der Reblaus an den Rebwurzeln verursacht; eine direkte Korrelation zwischen Reblausdichte und Ausmaß der Rückgangserscheinungen an infizierten Reben besteht nicht. B.: Das Ausmaß möglicher Rückgangserscheinungen ist abhängig von der Bewirtschaftung des Rebbestands, wobei der Bodenbewirtschaftung eine maßgebliche Rolle zukommt.
Um diese Hypothesen zu überprüfen wurde ableitend von den Zielen der Arbeit ein Reblausbonitursystem zur Erfassung der Reblausabudanz im Freiland im Boden erarbeitet. Die besonderen Schwierigkeiten bei der Erfassung des Reblausbefalls im Boden aufgrund des stark variierenden Wurzelsystems der Rebe und dem ebenfalls stark schwankenden Reblausbefalls an der Wurzel der Rebe wurde mit dem Reblausbonitursystem (Grabungen) Rechnung getragen, sodass ein valides System zur Beurteilung des Reblausbefalls auch im jahreszeitlichen Verlauf erstellt worden konnte. Damit wurde ein Reblausbonitursystem geschaffen, das auch in praktischer Anwendung durchführbar ist und somit mit seiner höheren möglichen Replikationsrate bei der Probenentnahme dem stark variierenden Charakter des Reblausbefalls im Boden Rechnung trägt.
Um Rückgangserscheinungen in einem Weinberg nachhaltig analysieren zu können, wurde ein Wuchsbonitursystem evaluiert. Dieses System beurteilt alle Reben einer Rebfläche innerhalb eines einfachen für den Menschen kognitiv sehr gut verarbeitenden Bewertungssystem zum Wuchs der Reben. Damit ist eine Erfassung einer sehr großen Datenmenge zur Beurteilung des Wuchses auf einem Rebfeld möglich. Die Korrelation des mit dem System ermittelten Wuchses zu vielen Leistungsparametern der Rebe und den Gegebenheiten auf den Versuchsflächen zeigt die hohe Validität des Wuchsbonitursystems auf. Auch konnten Wuchsunterschiede in sehr differierenden Versuchsfeldern mit dieser Methode deutlich belegt werden. Damit wurde deutlich, dass mit dieser Wuchsboniturmethode die Rückgangserscheinungen in einer Rebfläche erkannt und analysiert werden konnten. Somit konnten mit den beiden Instrumenten der Wuchs- und Reblausbonitur die beiden Arbeitshypothesen dahingehend belegt werden, dass unter Verwendung der Unterlagensorten der Kreuzungskombination V. berlandieri x V. riparia kein Zusammenhang zwischen der Reblausdichte im Boden und Rückgangserscheinungen der Rebe vorliegen. Darüber hinaus konnte belegt werden, dass in Erweiterung dieser Erkenntnis das Ausmaß der Rückganserscheinungen bei Reben maßgeblich von der Art der Bewirtschaftung abhängt und somit deutlich abzutrennen ist vom Einfluss des Reblausbefalls. Vor allem konnte durch die vorliegende Arbeit unter Einbeziehung des Wuchsbonitursystems eine Methode erarbeitet werden, die in Zukunft bei der Beurteilung von Fernerkundungsdaten ein nützliches Instrument darstellt, um die wirklichen Gegebenheiten hinsichtlich des Wuchses der Reben auf Weinbergsflächen mit den berechneten Werten zu Wuchsindizes zu korrelieren.The work was based on two working hypotheses that had to be tested. The two working hypotheses were formulated based on field observations which showed that infestation intensity of grape phylloxera on grape roots cannot be directly linked to the level of plant damage. Moreover, the negative effects of phylloxera infestations on grape growth and health were more frequent and succinct in vineyards subject to poor vineyard management especially in regards to soil cultivation and management. In order to clarify this phenomenon, the following two working hypotheses (A., B.) were formulated:
A.: Decline of vines based on V. berlandieri x V. riparia rootstocks and infested by phylloxera, are not only caused by the feeding action of phylloxera on the roots; there is no direct correlation between phylloxera abundance and the extent of decline in infected vines.
B.: The degree of decline depends on vineyard management, with soil management playing a key role.
In order to test these hypotheses, a phylloxera-assessment system was developed to determine phylloxera abundance and infestation intensities on roots under field conditions. The particular difficulties in recording phylloxera infestations are due to the widely varying root system of the vines as well as the and the strongly fluctuating phylloxera infestations on the roots of the vines. These inhomogeneous occurrences – of grape roots in soil and phylloxera on roots - were especially taken into account during the development of the root sampling scheme used for the phylloxera assessment system. - Thus, a valid system for assessing phylloxera infestation intensities was developed which also allows for assessment of the seasonal fluctuations. The phylloxera assessment system developed was designed in a way making it applicable also in the agricultural practise and accounting for the widely varying character of root systems as well as phylloxera infestation on roots.
In order to be able to analyse long-lasting signs of vine decline in a vineyard, a growth assessment system was developed and evaluated. This system assesses all vines in a vineyard using a visual evaluation system for vine growth and health-. This allows for a very large amount of data to be recorded to assess vine growth and health in a vineyard. Vine growth and health determined with the system was correlated to many performance parameters. The correlations to the conditions of different trial sites and vineyard management regimes showed the high validity of the growth assessment system. Differences in growth in very different experimental fields and plots could also be clearly demonstrated with this method.
Thus, the two working hypotheses could be substantiated with the two instruments of the vine growth and phylloxera assessment systems. For the rootstock varieties V. berlandieri x V. riparia it could be shown that there is no connection between the phylloxera abundance on roots and symptoms of decline of vines. In addition, it could be demonstrated that the extent of growth depressions in vines largely depends on the type of cultivation and vineyard management and that damage can therefore be clearly separated from the sole influence of phylloxera. Above all, the present work, including the growth assessment system, was able to develop a method that will be a useful instrument in future for the assessment of remote sensing data in order to correlate the real conditions regarding the growth of the vines in vineyards with the calculated values for growth indices
The Big Mess Construction: Forms and Functions in Present-Day English
The present thesis is devoted to a construction which defies generalisations about the prototypical English noun phrase (NP) to such an extent that it has been termed the Big Mess Construction (Berman 1974). As illustrated by the examples in (1) and (2), the NPs under study involve premodifying adjective phrases (APs) which precede the determiner (always realised in the form of the indefinite article a(n)) rather than following it.

Previous research on the construction is largely limited to contributions from the realms of theoretical syntax and a number of cursory accounts in reference grammars. No comprehensive investigation of its realisations and uses has as yet been conducted. My thesis fills this gap by means of an exhaustive analysis of the construction on the basis of authentic language data retrieved from the British National Corpus (BNC). The corpus-based approach allows me to examine not only the possible but also the most typical uses of the construction. Moreover, while previous work has almost exclusively focused on the formal realisations of the construction, I investigate both its forms and functions.
It is demonstrated that, while the construction is remarkably flexible as concerns its possible realisations, its use is governed by probabilistic constraints. For example, some items occur much more frequently inside the degree item slot than others (as, too and so stand out for their particularly high frequency). Contrary to what is assumed in most previous descriptions, the slot is not restricted in its realisation to a fixed number of items. Rather than representing a specialised structure, the construction is furthermore shown to be distributed over a wide range of possible text types and syntactic functions. On the other hand, it is found to be much less typical of spontaneous conversation than of written language; Big Mess NPs further display a strong preference for the function of subject complement. Investigations of the internal structural complexity of the construction indicate that its obligatory components can optionally be enriched by a remarkably wide range of optional (if infrequent) elements. In an additional analysis of the realisations of the obligatory but lexically variable slots (head noun and head of AP), the construction is highlighted to represent a productive pattern. With the help of the methods of Collexeme Analysis (Stefanowitsch and Gries 2003) and Co-varying Collexeme Analysis (Gries and Stefanowitsch 2004b, Stefanowitsch and Gries 2005), the two slots are, however, revealed to be strongly associated with general nouns and ‘evaluative’ and ‘dimension’ adjectives, respectively. On the basis of an inspection of the most typical adjective-noun combinations, I identify the prototypical semantics of the Big Mess Construction.
The analyses of the constructional functions centre on two distinct functional areas. First, I investigate Bolinger’s (1972) hypothesis that the construction fulfils functions in line with the Principle of Rhythmic Alternation (e.g. Selkirk 1984: 11, Schlüter 2005). It is established that rhythmic preferences co-determine the use of the construction to some extent, but that they clearly do not suffice to explain the phenomenon under study. In a next step, the discourse-pragmatic functions of the construction are scrutinised. Big Mess NPs are demonstrated to perform distinct information-structural functions in that the non-canonical position of the AP serves to highlight focal information (compare De Mönnink 2000: 134-35). Additionally, the construction is shown to place emphasis on acts of evaluation. I conclude the construction to represent a contrastive focus construction.
My investigations of the formal and functional characteristics of Big Mess NPs each include analyses which compare individual versions of the construction to one another (e.g. the As Big a Mess, Too Big a Mess and So Big a Mess Constructions). It is revealed that the versions are united by a shared core of properties while differing from one another at more abstract levels of description. The question of the status of the constructional versions as separate constructions further receives special emphasis as part of a discussion in which I integrate my results into the framework of usage-based Construction Grammar (e.g. Goldberg 1995, 2006).Die vorliegende Arbeit befasst sich mit einer Konstruktion, welche so deutlich von den Regularitäten der prototypischen englischen Nominalphrase abweicht, dass sie auch unter dem Namen Big Mess Construction bekannt wurde (Berman 1974). Die Beachtung, welche die Konstruktion in der bisherigen Forschung erfahren hat, beschränkt sich hauptsächlich auf Beiträge aus dem Bereich der theoretischen Syntax und einzelne lückenhafte Beschreibungen in Grammatiken. Eine systematische Analyse der Konstruktion und ihrer Verwendung steht bislang aus. Die vorliegende Dissertation unterzieht die Konstruktion einer umfassenden Untersuchung auf der Grundlage authentischer Sprachdaten, die dem British National Corpus (BNC) entnommen wurden. Nebst einer Reihe qualitativer Analysen werden insbesondere auch quantitative Ergebnisse herangezogen, die es mir gestatten, über die möglichen Vorkommnisse der Konstruktion hinaus auch deren häufigste und somit typische Verwendungen zu beleuchten. Während bisherige Beschreibungen fast ausschließlich die formalen Realisierungen der Konstruktion untersucht haben, widme ich mich sowohl ihren Formen als auch ihren Funktionen.
Es wird gezeigt, dass die Konstruktion eine hohe Flexibilität aufweist, wenngleich die jeweils möglichen Verwendungen sich deutlich in ihrer Häufigkeit unterscheiden. Beispielsweise divergieren die unterschiedlichen Gradadverbien, welche die Konstruktion einleiten können, deutlich in ihrer Frequenz. Entgegen gängiger Annahmen der bisherigen Forschung stellen diese Gradadverbien keine gänzlich geschlossene Gruppe dar. Weiter wird verdeutlicht, dass die Konstruktion in ihrem Vorkommen keinesfalls als hochgradig spezialisiert angesehen werden kann, jedoch bestimmte Präferenzen für bestimmte syntaktische Funktionen und Texttypen erkennbar werden. Ferner wird ersichtlich, dass die obligatorischen Komponenten der Konstruktion durch eine Vielzahl optionaler Strukturelemente ergänzt werden können.
Untersuchungen der Realisierungen der obligatorischen aber lexikalisch variablen Elemente (Kopf der AP und Kopfsubstantiv) erlauben Rückschlüsse auf die Produktivität der Konstruktion. Des Weiteren wird mit Hilfe der Methoden der Collexeme Analysis (Stefanowitsch und Gries 2003) und Co-varying Collexeme Analysis (Gries und Stefanowitsch 2004b, Stefanowitsch und Gries 2005) aufgezeigt, dass die zwei Slots eine starke Präferenz für ‚Dimension‘ Adjektive und evaluative Adjektive sowie für general nouns aufweisen. Ausgehend von den Resultaten zu den häufigsten Adjektiv-Substantiv Kombinationen erfolgt eine qualitative Analyse der typischen Semantik der Konstruktion.
Die funktionalen Analysen konzentrieren sich auf Faktoren aus zwei Bereichen. Zunächst wird Bolingers (1972) Hypothese überprüft, dass die abweichende Wortfolge, die der Konstruktion zugrunde liegt, mit dem Principle of Rhythmic Alternation (z.B. Selkirk 1984: 11, Schlüter 2005) in Zusammenhang zu bringen sei. Es wird aufgedeckt, dass rhythmische Prinzipien die Verwendung der Konstruktion zu einem gewissen Grad kodeterminieren, als Erklärungsansatz jedoch nicht ausreichen. In einem nächsten Schritt widme ich mich diskurspragmatischen Funktionen. Wie erkennbar wird, kommt der Big Mess Construction die Funktion zu, den Fokus des übergeordneten Satzes über die markierte Wortstellung herauszustellen (vgl. De Mönnink 2000: 134-35) und evaluative Akte hervorzuheben. Abschließend wird die Konstruktion der Gruppe der focus fronting Konstruktionen zugeordnet.
In den Untersuchungen zu den Formen und Funktionen der Big Mess Construction werden stets auch vergleichende Ergebnisse zu einzelnen Versionen der Konstruktion präsentiert (z.B. zu den As Big a Mess, Too Big a Mess und So Big a Mess Constructions). Es wird ermittelt, dass die Versionen einen geteilten Grundbestand an typischen Charakteristika aufweisen, während auf spezifischeren Beschreibungsebenen auch individuelle Nutzungsprofile erkennbar werden. Die Frage nach dem Status der einzelnen Versionen wird ferner als Teil einer separaten Diskussion aufgegriffen, in der ich die Möglichkeit der Integration meiner Ergebnisse in die Konstruktionsgrammatik (z.B. Goldberg 1995, 2006) aufzeige
Bordering in Pandemic Times: Insights into the COVID-19 Lockdown
In the course of the COVID-19 pandemic, borders have become relevant (again) in political action and in people's everyday lives within a very short time. This was especially true for the inhabitants of border regions, whose cross-border life worlds were suddenly irritated by closed borders and police controls. However, the COVID-19 pandemic also led to an increased evidence of social, cultural, economic, health and mobility boundaries beyond national borders, which raised pressing questions about social inequalities. The authors shed light on these dynamics from the perspective of territorial borders, social boundaries and (dis)continuities in border regions through a variety of thematic and spatial approaches. The critical observations and scientific comments were made during the lockdown in April and May 2020 and provide insights into the events during the global pandemic.Au cours de la pandémie COVID-19, les frontières ont (re)pris de l'importance dans l'action politique et dans la vie quotidienne en très peu de temps. C'était particulièrement vrai pour les habitants des régions frontalières, dont le quotidien transfrontalier était soudainement irrité par la fermeture des frontières et les contrôles de police. Toutefois, la pandémie de COVID-19 a également entraîné une perception accrue des frontières sociales, culturelles, économiques, sanitaires et de mobilité au-delà des frontières nationales, ce qui a soulevé des questions pressantes sur les inégalités sociales. Les auteurs éclairent ces dynamiques sous l'angle des frontières territoriales, des démarcations sociales et des (dis)continuités dans les régions frontalières par le biais de diverses approches thématiques et spatiales. Les observations critiques et les commentaires scientifiques ont été faits pendant le confinement en avril et mai 2020 et donnent un aperçu des événements pendant la pandémie mondiale.Im Zuge der COVID-19-Pandemie sind Grenzen binnen kürzester Zeit im politischen Handeln und im Alltag der Menschen (wieder) relevant geworden. Dies betraf in besonderem Maße die Bewohner*innen von Grenzregionen, deren grenzüberschreitende Lebenswelten plötzlich von geschlossenen Grenzen und polizeilichen Kontrollen irritiert wurden. Doch auch jenseits von Staatsgrenzen führte die COVID-19-Pandemie zu einer verstärkten Wahrnehmung von sozialen, kulturellen, wirtschaftlichen, gesundheitlichen und mobilitätsbezogenen Grenzen, die drängende Fragen nach gesellschaftlichen Ungleichheiten aufwarfen. Die Autor*innen beleuchten diese Dynamiken aus dem Blickwinkel von territorialen Grenzen, sozialen Grenzziehungen und (Dis)Kontinuitäten in Grenzregionen über vielfältige thematische und räumliche Zugänge. Die kritischen Beobachtungen und wissenschaftlichen Kommentare sind während des Lockdown im April und Mai 2020 entstanden und geben Einblicke in das Zeitgeschehen während der globalen Pandemie
