Characterization of the interaction between Liver Kinase B1 and Lethal (2) giant larvae in 'Drosophila melanogaster'
Der Verlust von Zellpolarität und die Degeneration der Gewebearchitektur spielen eine wesentliche Rolle für die Entstehung, Wachstum und Ausbreitung von Tumoren.
Insbesondere maligne Karzinomzellen, welche aus Epithelgewebe hervorgehen, zeigen einen Verlust der apikal-basalen Polarität und ein Ablösen vom zellulären Netzwerk.
Die epithelial-mesenchymale Transition, welche die Migration von Epithelzellen ermöglicht, führt zu aggressiveren Tumorphänomenen mit entdifferenzierten Zellen bis hin zu anaplastischen Zelltypen.
Die Masterkinase LKB1 und Lgl, ein Mitglied des Scribble-Komplexes, die beide als Tumorsuppressorproteine bekannt sind, sind essenziell für die Aufrechterhaltung der Zellpolarität.
Vorangegangene Untersuchungen lassen vermuten, dass Lgl ein nachgeschaltetes Phosphorylierungsziel von LKB1 ist und dessen Einflussnahme auf die Zellpolarität unterstützt. Nachfolgend wird die Interaktion beider Proteine im Modellorganismus Drosophila melanogaster weitergehend untersucht
Contactin-associated protein 2 autoantibodies can be associated with multifocal motor-like neuropathy: a case report
Autoantibodies against contactin-associated protein 2 (CASPR2) are usually associated with autoimmune encephalitis and neuromyotonia. Their association with inflammatory neuropathies has been described in case reports albeit all with distal symmetric manifestation. Here, we report a patient who developed distal arm paresis, dominantly of the right arm, over the course of 1 year. Electroneurography showed a conduction block of motor nerve conduction, nerve ultrasonography a swelling of the right median and ulnar nerve and flow cytometry an increase in natural killer (NK cells) in the blood and natural killer T (NKT) cells in the cerebrospinal fluid (CSF), therefore indicating a multifocal motor neuropathy-like (MMN-like) phenotype. CASPR2 autoantibodies were detected in serum and CSF. Through immunotherapy with intravenous immunoglobulins the patient showed clinical and neurographic improvement. We therefore describe the first association of CASPR2 autoantibodies with a MMN-like clinical manifestation, extending the spectrum of CASPR2-associated diseases
Digitale Medien im Mathematikunterricht der Primarstufe 2025:Beiträge zur 8. PriMaMedien-Sommertagung 2025 in Gießen
Seit 2007 tagt die Arbeitsgruppe PriMaMedien im Arbeitskreis Grundschule der Gesellschaft für Didaktik der Mathematik. Die Mitglieder dieser Arbeitsgruppe teilen das Interesse an der Entwicklung, der Konzeption, dem Einsatz und der Analyse digitaler Medien für den Mathematikunterricht in der Primarstufe.
Im Rahmen der Sommertagung 2025 der PriMaMedien-Arbeitsgruppe an der Justus-Liebig-Universität in Gießen fanden sich 30 Personen zusammen, um über Forschungs- und Entwicklungsarbeiten zum Einsatz digitaler Medien im Mathematikunterricht der Primarstufe zu diskutieren. Darüber hinaus wurden auch digital gestützte Lehr-Lernkonzepte in den Blick genommen. Gemäß des ‚Primats der Fachdidaktik’ erfolgte eine Auseinandersetzung mit Forschungsprojekten und Lehr- und Lernangeboten, die sich an den technischen Gegebenheiten und daraus resultierenden fachdidaktischen Potenzialen orientierten.
Im vorliegenden Tagungsband werden Projekte und Arbeiten aus der Forschung, der Schulpraxis und der Lehrkräftebildung dargestellt, die im Rahmen der PriMaMedien-Sommertagung 2025 vorgestellt wurden
Effects of a lifestyle intervention on the biomarkers of oxidative stress in non-communicable diseases: A systematic review
Oxidative stress plays a critical role in the pathogenesis of chronic diseases. Therefore, improvement of oxidative stress status through lifestyle intervention can play a vital role in preventing and treating chronic diseases. This systematic review aims to provide an overview of articles published in the last decade examining the association between lifestyle intervention and oxidative stress biomarkers in the context of non-communicable diseases. The electronic databases PubMed and Web of Science were searched for relevant studies, following the PRISMA (Preferred Reporting of Systematic Reviews and Meta-Analyses) guidelines. This systematic review focused on the four important oxidative stress biomarkers; glutathione (GSH), superoxide dismutase (SOD), catalase, and malondialdehyde. 671 articles were identified, of which nine met the inclusion criteria. A trend emerged, showing that lifestyle modifications that focus on diet and physical health can improve oxidative stress in the form of an increase in superoxide dismutase and CAT levels and a decrease in Malondialdehyde levels in participants with non-communicable diseases (NCDs), GSH levels were not affected. However, the results are difficult to compare because of the heterogeneity of the methods of the biomarkers studied. Our review indicates that oxidative stress can be influenced by lifestyle modifications and may be an effective tool for the prevention and management of non-communicable diseases. This review also elucidated the importance of analyzing multiple oxidative stress biomarkers to evaluate oxidative stress, it further highlights the need to conduct long-term lifestyle intervention studies on oxidative stress biomarkers to understand the connection between oxidative stress biomarkers, NCDs and Lifestyle intervention
Enhancing Volume Precision in Breast Reconstruction: A BMI-Based Model for Predicting Flap Weight in Profunda Artery Perforator Flaps
Background: In recent years, the profunda artery perforator (PAP) flap has gained recognition as a viable technique in autologous breast reconstruction, demonstrating low donor site morbidity and favorable reconstructive outcomes. This study aims to highlight the recent refinements using the PAP flap for breast reconstruction focusing on preoperative volume prediction. Methods: A retrospective study was conducted to evaluate the outcomes of breast reconstruction surgeries using the PAP flap at our institution between May 2018 and December 2022. A total of 114 PAP flaps performed in 96 patients were included. Statistical analysis was performed to analyze flap volume in relation to body mass index (BMI). Surgical details, such as donor site-related complications and patient characteristics, were also collected. Results: BMI was identified as a statistically significant predictor (p < 0.001) for flap weight in the resulting model. The average BMI observed was 23.0, while the mean flap weight was 304.2 g. The predictive model for flap weight was determined as follows: flap weight (g) = (19.252 × BMI) − 143.572. This model underscores the relationship between BMI and flap weight, quantifying the influence of BMI on flap weight prediction. Conclusions: Our study indicates that it is feasible to harvest an adequate volume for breast reconstruction even in patients with a low BMI. To facilitate preoperative prediction of PAP flap volume, we developed an algorithm designed to enhance preoperative planning and estimate the need for supplementary procedures to achieve the desired volume
Evaluation of effectiveness and safety of the large-format pRESET 6-50 thrombectomy stent-retriever in the endovascular treatment of ischemic stroke: real-world experiences from two tertiary comprehensive stroke centers
Objective: Mechanical thrombectomy (MT) has become the standard treatment for acute ischemic stroke (AIS) with large vessel occlusion (LVO). First-pass (FP) reperfusion of the occluded vessel and fewer passes with stent retrievers show improvement in functional outcomes in stroke patients, while higher numbers of passes are associated with higher complication rates and worse outcomes. Studies indicate that a larger size of the stent-retriever is associated with a higher rate of first-pass reperfusion and improved clinical outcomes. In this retrospective study, we investigated the clinical performance of a recently developed and one of the largest stent-retrievers available in the treatment of LVO (pRESET 6-50, phenox GmbH, Bochum). Materials and methods: All consecutive patients with ischemic stroke due to proximal large vessel occlusion treated with MT using the pRESET 6-50 stent-retriever in two tertiary stroke centers between 09/2021 and 07/2022 were included in this study. The reperfusion rate after MT was quantified by the modified thrombolysis in cerebral infarction (mTICI) score, and functional neurological outcome was evaluated with the National Institutes of Health Stroke Scale (NIHSS) score and the major early neurological recovery (mENR) rate after 24 h. Successful FP reperfusion was defined as mTICI ≥ 2b. Successful and complete reperfusion were defined as mTICI ≥ 2b and mTICI ≥ 2c, respectively. Results: In total, 98 patients (52 men and 46 women) with a median age of 75 (range 25–95 years) were included. A total of 70 (72%) patients presented with an occlusion of the middle cerebral artery (MCA) in the M1 segment, 6 (6%) patients with an occlusion of the M2 segment, 17 (17%) patients with an occlusion of the internal carotid artery (ICA), and 5 (5%) patients with an occlusion of the obstructed basilar artery (BA). Successful FP reperfusion was achieved in 58 patients (62%). Successful and complete reperfusion were achieved in 95 (97%) and 82 (83%) patients, respectively. The median National Institutes of Health Stroke Scale (NIHSS) in all treated patients improved from 17 to 7.5. Major early neurological recovery (mENR) was observed in 34 patients (35.1%). Conclusion: MT with the pRESET 6-50 stent-retriever achieves high successful first-pass and final reperfusion rates in patients with AIS and LVO. The results of this study support the thesis to use large-format stent-retriever in proximal vessel occlusion MT whenever feasible in order to improve high FP and final reperfusion rate, which are known predictors of good clinical outcome
