    Magnetron sputtering of magnetic thin film materials for spintronics

    Spintronics explores the utilization of the electron spin in addition to the charge in solid-state devices, with the goal of enhancing functionality beyond conventional electronics. An essential component for many spintronic devices are magnetic thin film, and consequentially the fabrication of such films often marks the first step in the research of novel materials or towards their integration into applications. In this thesis, the film growth and the properties of two different magnetic thin film materials, grown by magnetron sputtering, are investigated. The first part of the thesis is focused on MnAl. This ferrimagnetic alloy features a high magnetocrystalline anisotropy, making it attractive for application as rare-earth free permanent magnet or in various spintronic applications as a perpendicular magnetic anisotropy (PMA) material. However, so far, the magnetothermal transport properties of MnAl, i.e. the anomalous Nernst effect (ANE), are still unknown. Here, we will report on the successful growth of MnAl thin films, and their magnetothermal transport properties. For the fabrication, MgO substrates with a Cr buffer layer were utilised, and several deposition parameter were optimised to achieve a chemically ordered, highly texturised MnAl. By magnetometry measurement as well as measurements of the anomalous Hall effect (AHE), we demonstrate the PMA and find magnetic properties comparable to other MnAl thin films. Finally, the magnetothermal transport properties, especially the ANE, are investigated. We find that the ANE features the signature of PMA, and the anomalous Nernst coefficient is comparable to other PMA materials. With that, we establish MnAl thin films as a potential candidate for spintronic or spincaloritronic applications. The second part of the thesis is centred around the ferrimagnetic insulator of aluminium substituted yttrium iron garnet (YAlIG). Magnetic garnets, in particular yttrium iron garnet (YIG), are widely used materials in spintronic or magnonic experiments. However, experiments at elevated temperatures around the Curie temperature have proven to be challenging. In the framework of this thesis, we aim to reduce the Curie temperature of YIG by substitution with Al. To this end, YAlIG thin films are prepared by radio frequency (rf) sputtering at room temperature in combination with an ex situ annealing step, and we demonstrate a high structural quality of the epitaxially grown thin film. The systematic reduction of the Curie temperature with increasing Al substitution is confirmed by magnetometry measurements. Upon characterisation by broadband ferromagnetic resonance, we find that the magnetic anisotropy is highly dependent on the substrate material, and a Gilbert damping comparable to other garnet thin films. Therefore, YAlIG thin films provide a material platform for spintronic and magnonic experiments across different magnetic phases

    Systematische Untersuchungen zur Thermodynamik und Stabilität ausgewählter Metallborhydride der 1. bis 5. Gruppe des Periodensystems der Elemente

    Im Rahmen der Dissertation wurden die Borhydride von Ca, Sr, Ba, Y, La, Zr und Hf hergestellt und deren thermodynamische Daten durch kalorimetrische Messungen ermittelt. Weiterhin wurden DFT-Berechnungen und verschiedene Korrelationen zur Ergänzung der experimentellen Datenbasis eingesetzt. Es folgte ein Vergleich aller ermittelten Daten mit abgeschätzten Werten. Auf Basis der bestimmten thermodynamischen Daten erfolgten umfangreiche Berechnungen zum Zersetzungsverhalten der Boranate, welche mit den experimentellen Befunden verglichen wurden. Weiterhin wurden Gleichgewichtsberechnungen zur Rehydrierbarkeit der reinen Boranate genutzt, um Aussagen zu deren Eignung als Wasserstoffspeichermaterialien abzuleiten.     Entwicklung einer Strategie für eine qualitätsgesicherte Evolution dynamischer Simulationsmodelle

    Die Diplomarbeit untersucht die Qualitätsbewertung von dynamischen Simulationsmodellen zur Prozessunterstützung in modernen Industrieanlagen. Es wird in Anlehnung an die ISO 25010 ein Qualitätsmodell zur Bewertung von Softsensormodellen zur Online-Prozessüberwachung erstellt. Methoden zur Bewertung von Softwarequalität werden auf die Qualitätsbewertung von Simulationsmodellen übertragen. Der Fokus liegt hierbei auf der Funktionalität und Zuverlässigkeit der Simulationsmodelle. Anhand des Qualitätsmodells wird ein Prototyp zur teilautomatisierten Qualitätsprüfung implementiert. Das wichtigste Merkmal des Prototyps ist die Ausgabe von Handlungsempfehlungen zur Evolution der Simulationsmodelle. Anhand eines Softsensormodells zur Überwachung der Biomassekonzentration in einem Fermentationsprozess und einer Fallstudie wird der Prototyp evaluiert.This thesis investigates the quality assessment of dynamic simulation models for process support in modern industrial plants. Based on ISO 25010, a quality model for the assessment of soft sensor models for model-based process monitoring is developed. For this, methods for evaluating software quality are transferred to the quality of simulation models. The focus is on the functionality and reliability of the simulation models. The quality model is used to implement a prototype for semi automated quality testing. The most important feature of the prototype is the output of recommendations for the evolution of the simulation models. The prototype is evaluated using a soft sensor model for online estimation of biomass concentration in a fermentation process and a case study

    Mental health and perceived stress coping competence in old age high-risk groups during the COVID-19 pandemic

    With the onset of the coronavirus pandemic in 2020 in Germany, the population faced a mentally challenging time. Governmental measures like social distancing measures, lockdowns and quarantines were implemented to reduce the spread of the coronavirus and save as many lives as possible. People of old age were soon identified as high risk group for a lethal course of disease and as in particular need of protection. Mental health professionals feared an aggravation of mental distress and mental pathology. Against this background, this dissertation aimed to investigate mental coping and the progression of mental health outcomes in old and oldest-old risk groups over the course of the COVID-19 pandemic, as well as associated sociodemographic and psychosocial factors associated with it. In our first study, we investigated the perceived stress coping competence (self-efficient coping vs. helplessness-associated coping) as well as perceived worries about and perceived threat by the COVID-10 pandemic and associated sociodemographic and psychosocial factors in the oldest-old population in Germany in the early phase of the pandemic. Linear multivariate regression models revealed that in our sample of n=197 people aged 78 to 100 years old higher levels of resilience were linked to higher perceived self-efficient coping. Male gender compared to the female gender, and a medium educational level in reference to a low educational level, was associated with less helplessness-associated coping. Loneliness showed associations with both, self-efficient and helplessness-associated coping. Higher levels of anxiety and depressive symptoms were linked to higher scores of helplessness-associated coping. Regarding perceived worries about the pandemic, binary logistic regression models showed that male gender and higher perceived social support were linked to more worries about the pandemic. Despite the higher risk for a severe course of disease for people of old age, older age was associated with less perceived personal threat by the coronavirus. Furthermore, more reported depressive symptoms were linked to less perceived threat, and higher reported somatization symptoms were associated with higher personal threat by the virus. Therefore, mental health factors seem to shape the way oldest-old individuals perceive the COVID-19 pandemic in partly unexpected ways, and resilience may illustrate an important factor when targeting more functional ways of coping with stress. Moreover, people of oldest-old age experiencing greater loneliness, high depressive and anxiety symptoms could be supported by adapting their coping skill repertoire to foster the feeling of self-efficiency when coping with stress. In our second and third study, we analyzed the development and progression of mental health factors like depressive, anxiety and somatization symptoms. First, in study 2 with n=156 people of oldest-old age (range= 78 to 97 years) Wilcoxon t-tests showed an increase of anxiety, depressive and somatization symptoms from 2021 to 2021 with generalized regression models revealing that higher levels of symptomatology in 2020 were associated with higher symptom burden in 2021. Higher perceived social support in 2020 was linked to less depressive symptoms in 2021. Building on this findings, we then conducted multilevel mixed-effects generalized linear models to analyzed data over three points of measurement from 2020, 2021 and end of 2021/beginning of 2022 with n=135 participants (age range= 78 to 97 years). Results showed that after a first increase of depressive and anxiety symptoms from 2020 to 2021, symptomatology levels stayed relatively stable from 2021 to 2021/2022. Moreover, older age and higher reported worries were associated with higher levels of anxiety symptoms. Greater perceived social support was linked to less anxiety and depressive symptoms over time. Resilience was associated with less depressive symptoms. Since pre-existing heart conditions illustrated an additional risk factor, next to an older age (60+ years), for a serious and lethal course of coronavirus disease, the fourth study additionally focused on an old age sample with cardiovascular risk profiles. The aim was a comparison of this cardiovascular risk group with the general old age population in terms of symptomatology levels and psychosocial factors associated with it. We analyzed data of n=1236 participants with n=618 showing a cardiovascular risk profile, drawn from the AgeWell.de study, and n=618 from a representative survey of the old age general population with an age range of 64 to 81 years in the beginning of the COVID-19 pandemic. Results showed that the cardiovascular risk group showed higher depressive symptom levels compared to the general population. Multivariate regression models revealed that in the cardiovascular risk group higher perceived social support was linked to less depressive and anxiety symptoms. In the general old age population perceived social support was associated with depressive symptoms, social support and anxiety symptoms showed no such association. Higher perceived worries were associated with higher reported anxiety symptoms in the general population only. Perceived resilience was linked to less depressive and anxiety symptoms in both groups. In conclusion, the oldest-old individuals showed an overall good mental health in the long term despite a first increase in anxiety and depressive symptomatology within the first year. Fostering perceived social support and perceived resilience may play an important role in preventing and reducing depressive symptomatology in times of crises like the pandemic.:Table of Contents I 1.     Same same but different, but still same: Dieser Spruch ist an sich selbst ein Meme und beschreibt gleichzeitig das Thema dieser Arbeit, in der die Wiederverwertung und Neudeutung klassischer Kunstwerke in modernen Formaten und Kontexten behandelt wird. Memes greifen oft visuelle und inhaltliche Elemente klassischer Kunst auf, interpretieren sie aber durch Community-Strukturen neu verändern ihren Kontext. Die Arbeit untersucht Parallelen und Unterschiede zwischen Meme-Kultur und Aneignungskunst, um die Verbindung von Hochkultur und Subkultur zu beleuchten. Dabei werden sowohl die Funktionsprinzipien von Memes als auch ihre Verbreitung und visuelle Ästhetik analysiert. Anhand ausgewählter Beispiele wird aufgezeigt, wie klassische Kunst in die Meme-Kultur integriert wird und welche Bedeutung diese Transformationen haben

    On the use of an endorectal balloon in ultrahypofractionated MR-guided radiotherapy of prostate cancer

    Die ultrahypofraktionierte MRT-gestützte Strahlentherapie (MRIgRT) ist eine Alternative zur normo- oder hypofraktionierten Bestrahlung beim lokalisiertem Prostatakarzinom. Mit der erhöhten Dosis pro Fraktion bei der ultrahypofraktionierten Strahlentherapie ist eine präzise Dosisappliaktion von großer Bedeutung, um eine Tumorkontrolle mit minimalen Nebenwirkungen zu erreichen. Ziel dieser Studie war es, zwei Patientenkohorten zu untersuchen, um die Auswirkungen der Verwendung eines Rektumballons (ERB) auf die Positionsstabilität der Prostata und der Samenblasen (SV) während der ultrahypofraktionierten MRIgRT beim lokalisierten Prostatakarzinom zu analysieren. Die erste Kohorte bestand aus zehn Patienten, die zwischen September 2022 und Mai 2023 am Universitätsklinikum Dresden in der Klinik und Poliklinik für Strahlentherapie und Radioonkologie mit einem ERB an einem Unity MR-LINAC (Elekta) ultrahypofraktioniert bestrahlt wurden. Die zweite Kohorte umfasste zehn Patienten, die im gleichen Zeitraum ohne ERB an der Universitätsklinik für Strahlentherapie Tübingen ebenfalls an einem Unity MR-LINAC (Elekta) mittels ultrahypofraktionierter MRIgRT behandelt wurden. Die Dosis auf die Prostata und die auf SV betrug für beide Kohorten 39.20 Gy in 7 Fraktionen, mit einem gleichzeitig integrierten Boost von 42.60 Gy auf die Prostata. Insgesamt wurden für beide Kohorten zwei anatomische MRT-Bilder pro Fraktion, bestehend aus dem täglichen Planungs-MRT (PL) für die erneute Planung gemäß dem adapt-to-shape workflow (ATS) und dem MRT-Scan nach der Behandlung (PT), zur Anpassung der Konturen [Monaco 5.51.11-Behandlungsplanungssystems (Elekta)] verwendet. Anschließend wurden der Massenmittelpunkt für die Prostata und die SV bestimmt, und die Positionsabweichungen zwischen PL-MRT und PT-MRT berechnet. Die Abweichungen wurden in den Richtungen links-rechts (LR, x), superior-inferior (SI, y), anterior-posterior (AP, z) sowie für die euklidische Distanz (d) ausgewertet. Fälle mit d>0.4 cm wurden visuell auf mögliche Patientenbewegungen überprüft und gegebenenfalls ausgeschlossen. Zur Klassifizierung von zufälligen und systematischen Fehlern wurden der Gruppenmittelwertfehler (M) und die Standardabweichungen (SD) der zufälligen (σ) und systematischen Fehler (∑) berechnet. Für die Kohorte aus Dresden, welche mit einem ERB behandelt wurde, wurden insgesamt 44 Behandlungsfraktionen ausgewertet. Die durchschnittliche Behandlungszeit betrug 43,5 Minuten. Die Positionsabweichungen der Prostata in dieser Gruppe zeigten im Mittel eine Verschiebung von M=0.0 [-0.5–0.3] cm für LR, 0.2 [-0.2–1.4] cm für SI, 0.2 [-0.2–0.5] cm für AP und 0.3 [0.0–1.4] cm für d. Für die Positionsabweichung der Samenblasen betrug die durchschnittliche Verschiebung 0.0 [-0.2–0.3] cm für LR, 0.1 [-0.2–0.5] cm für SI, 0.2 [-0.1–0.7] cm für AP und 0.3 [0.1–0.8] cm für d. Für die Kohorte aus Tübingen, welche ohne einem ERB behandelt wurde, wurden insgesamt 69 Behandlungsfraktionen ausgewertet. Die durchschnittliche Behandlungszeit betrug 30 Minuten. Die Positionsabweichungen der Prostata in dieser Gruppe zeigten im Mittel eine Verschiebung von M=0.0 [-0.4– 0.23] cm für LR, 0.1 [-0.7– 1.1] cm für SI, 0.0 [-0.6 – 0.9] cm für AP und 0.26 [0.0 – 1.2] für d. Für die Positionsabweichung der Samenblasen betrug die durchschnittliche Verschiebung 0.0 [-0.21 – 0.4] cm für LR, 0.0 [-1.1 – 0.8] cm für SI, 0.1 [- 0.7– 0.9] cm für AP und 0.3 [0.1–0.8] cm für d. Die Bewertung dieser Ergebnisse zeigte, dass die Prostatabewegung in der SI Richtung in der Gruppe mit ERB grenzwertig signifikant größer war (0.15 cm gegenüber 0.05 cm, p=0,052). Auch in der AP Richtung war die Bewegung in der Gruppe mit ERB signifikant größer (0.17 cm gegenüber -0.01 cm, p=0.009). Bei den Samenblasen zeigte die Gruppe mit ERB eine stärkere Bewegung in der SI Richtung (0.1 cm gegenüber 0.0 cm, p=0.039). Darüber hinaus war die Variabilität der Bewegungen in der Gruppe ohne ERB höher (0.2 cm gegenüber 0.1cm) was sich in einer signifikanten Differenz der absoluten Bewegungen widerspiegelte (p=0.014) Insgesamt wird aus den Ergebnissen deutlich, dass die intrafraktionale Bewegung der Prostata und SV in beiden Gruppen, mit oder ohne ERB, minimal war. Die Studie legt somit nahe, dass der ERB bei der ultrahypofraktionierten MRIgRT nicht notwendig ist, da er keine signifikanten Vorteile hinsichtlich der Positionsstabilität der Prostata und SV bietet

    Characterization of the DEAD-box RNA helicase DDX3X and its role in ribonucleoprotein granule assembly

    Ribonucleoprotein (RNP) granules are membraneless compartments that form inside the cyto- or nucleoplasm by the process of phase separation. RNP granules play essential roles in cell organization and physiology and their misregulation can have detrimental effects including protein misfolding and aggregation. Even though our knowledge about RNP granules has increased greatly in the past years there are still many open questions about the mechanisms involved in their formation, dissolution, and properties. There are indications that RNA helicases could play a crucial role in regulating the properties of RNP granules. Many RNA helicases can be found in different types of RNP granules. Furthermore, they have the ability to bind and remodel secondary structures of RNA, one of the main components of RNP granules. This gives them the potential ability to influence RNP granule formation, dissolution, and properties. In my PhD thesis, I analysed the human DEAD-box helicase DDX3X, which is known to be present in stress-inducible RNP granules. For this, I established a purification method for DDX3X-WT as well as two cancer related-variants of DDX3X. By biochemical analysis, I could demonstrate that the cancer-related variants are partially or fully defective in their enzymatic activity. I could show that DDX3X forms condensates at physiologically relevant conditions in vitro and that the phase separation propensity is independent on the enzymatic activity. However, analysis of the material properties of condensates formed by DDX3X-WT and the two cancer-related variants in different conditions revealed significant differences. This indicates that the enzymatic activity of DDX3X is relevant for the material properties of these condensates. Furthermore, I set up an in vitro system to mimic stress-inducible RNP granules, using G3BP1 and RNA. This assay revealed that condensates formed by G3BP1 in the presence of heat-aggregated RNA exhibit solid-like features. I could show that DDX3X-WT, but not the cancer-related variants, collaborates with G3BP1 and promotes a solid to liquid phase transition of these solid-like condensates. Taken together, this suggests that the RNA helicase DDX3X can regulate the material properties of RNP granules using its enzymatic activity.     Einleitung: Wegen der hohen Prävalenz und der schwerwiegenden gesundheitlichen, sozialen und ökonomischen Folgen des weltweiten Alkoholkonsums ist die Erforschung besserer medikamentöser Behandlungsoptionen der Alkoholabhängigkeit von entscheidender Bedeutung. Klinische Studien für alle potentiell wirksamen pharmakologischen Ansatzpunkte durchzuführen, erscheint jedoch nicht realistisch. Mit dem „Testsystem für Medikamente bei Alkoholabhängigkeit“ (TEMA) wurde eine Methode entwickelt, die bestimmte präklinische Paradigmen in ein humanes Laborexperiment überträgt. Neben der Abbildung suchttypischen Verhaltens können damit auch verschiedene Kontingenzen zwischen Suchtverhalten und Verstärkung modelliert und eine bessere experimentelle Kontrolle über Alkoholexposition ermöglicht werden. Das TEMA sollte in der TEMANX-Studie validiert werden. Dazu sollte nachgewiesen werden, dass der µ-Opiat-Rezeptorantagonist Naltrexon gegenüber Placebo die Bereitschaft reduziert, in einem Laborexperiment Arbeit für Alkohol zu leisten. In der vorliegenden Arbeit wurden die Daten der TEMANX-Studie ausgewertet. Methoden: 46 nicht alkoholabhängige aber riskant trinkende Freiwillige (6 weiblich), darunter 17 Träger des OPRM1 A118G-Polymorphismus, führten zwei Experimente mit intravenöser Alkoholselbstverabreichung (ASA) durch. Das erste Experiment wurde vor, das zweite nach mindestens 7 Tagen randomisierter doppelblinder Behandlung mit Naltrexon bis 50 mg pro Tag oder Placebo durchgeführt. Jede Alkoholinfusion erforderte vorherige Arbeit in einer Aufmerksamkeitsaufgabe, deren Anforderungen gemäß einem progressiven Arbeitsschema im Verlauf exponentiell gesteigert wurden. Die Dynamik der Alkoholexposition und die steigende Arbeitsanforderung ermöglichte die Abbildung von einer initialen Anstiegsphase mit schnell steigenden Blutalkoholkonzentrationen (BAK), gefolgt von einer Plateauphase. In letzterer konnte die BAK trotz kontinuierlicher Arbeit für Alkohol nicht weiter erhöht, sondern ein Abfall der BAK nur verlangsamt werden. Ergebnisse: Personen, die mit Naltrexon behandelt wurden, reduzierten ihre kumulative Arbeitsleistung für Alkohol (cWFA) stärker als Teilnehmer in der Placebogruppe. Dieser Unterschied in der primären Zielgröße war in den vorab definierten Analysen der gesamten 150 Minuten der ASA, nicht statistisch signifikant (p = 0,471, Cohen’s d = 0,215). In den sekundären Zielgrößen zeigten sich ebenfalls keine signifikanten Unterschiede. Die Naltrexon-Serumspiegel korrelierten mit der Veränderung der Naltrexon-induzierten cWFA (r = −0,53; p = 0,014). Separate explorative Analysen ergaben, dass Naltrexon die cWFA während der Anstiegsphase signifikant reduzierte, nicht aber während der Plateauphase. Phasenabhängigtraten Korrelationen zwischen der cWFA und der Veränderungen der subjektiven Maße von alkoholinduzierter Stimulation, Wohlbefinden und Verlangen auf, welche durch Naltrexon modifiziert wurden. Naltrexon veränderte weder das reale Trinkverhalten noch das Alkohol-Craving. Der OPRM1-Polymorphismus konnte die Wirkung von Naltrexon nicht vorhersagen. Die Studienmedikation wurde gut vertragen und die TEMA-Prozeduren waren praktikabel und veranlassten keinen Teilnehmer zum Studienabbruch. Diskussion: Aufgrund eines starken Placeboeffekts konnte die Überlegenheit von Naltrexon gegenüber Placebo in den vorab geplanten Analysen nicht validiert werden. Unter Berücksichtigung der Dosis-Wirkungs-Beziehung der Naltrexon-Serumspiegel sowie aufgrund des Naltrexoneffekts in der Anstiegsphase, während die BAK anstieg, ist dennoch ein Medikamenteneffekt anzunehmen. Die phasenabhängigen Korrelationen der cWFA und den subjektiven Maßen legen nahe, dass die WFA während Anstiegsphase positiv und während der Plateauphase möglicherweise negativ verstärkt wurde. Die Ergebnisse zeigten sich trotz der verhältnismäßig kleinen Stichprobe nicht alkoholabhängiger freiwilliger Probanden und obwohl Naltrexon weder die Trinkmenge noch das Craving im richtigen Leben veränderte. Das TEMA stellt damit das erste Laborparadigma dar, das in der Lage ist, verschiedene Trinkmotivationen innerhalb eines Experiments zu modellieren. Deswegen hat es das Potential, in Zukunft schnell und effektiv potentielle Medikamente zu überprüfen, deren klinischer Wirkmechanismus nicht gut bekannt ist und zur Entscheidung beizutragen, ob es sich lohnt bestimmte neue Substanzen in großen klinischen Studien weiter zu untersuchen.     Characterisation of the expression and function of immune relevant molecules for disease progression in hematologic and solid tumors

    Cancer immunotherapy has revolutionized the treatment of tumor patients. However, only a limited number of patients developed a long-term response. Therefore, the identification of biomarkers is urgently required for the effective stratification of patients with solid and hematopoietic tumors, in order to select the most suitable therapy for the patient. These biomarkers can be single molecules, like RNAs, proteins or proteoglycans and frequently are direct mediators or a result of changes in the tumor microenvironment (TME). These include miRNAs, small non coding RNAs (20-24nt), which have a multivalent role and involved in the post transcriptionally control of multiple immune relevant targets thereby contributing to immune escape and tumor progression. Various miRNAs have been found to be deregulated in malignant lesions and might serve as diagnostic, prognostic and predictive biomarkers. Due to the large number of these small RNAs, which could regulate the expression of various molecules, their analysis is a challenge. Therefore, the aim with this PhD thesis is to identify novel biomarkers based on their functional role in immune responses utilizing immune modulatory miRNAs in melanoma and myelodysplastic syndromes (MDS). Relevant miRNAs were identified based on their ability to bind on the target checkpoint molecules of interest (PD-L1, CTLA-4) through the miTRAP method. The ability of these miRNAs to downregulate their target molecules was validated through flow cytometry and changes in the molecules’ function in vitro were assessed. Selected targets were quantified by qPCR and their relation to clinicopathological characteristics and the composition of the tumor immune cell infiltrate of melanoma patients were determined. In silico analysis was utilized for the identification of deregulated miRNAs involved in common pathways of MDS. MiRNAs targeting the NF-kB pathway components TLR4, MYD88, IRAK1, TRAF6 and NFκB1 were quantified in formalin-fixed, paraffin-embedded bone marrow biopsies (BMBs) of healthy controls and MDS patients. The expression levels of the TLR4 ligand, biglycan, were evaluated in MDS/AML cell lines and BMBs, while changes in the BM immune infiltrate were analysed via multi-spectral imaging. Inflammasome components were evaluated in vitro by Western blot analysis and validated in biopsies of MDS patients via multi-spectral imaging. The BGN-mediated pyroptosis was inhibited by drugs or CRISPR/Cas9 knock-out and changes in morphology and the cell population composition determined by flow cytometry and microscopy. Six (6) PD-L1 and seven (7) CTLA-4 specific miRNAs were identified capable of downregulating their respective target molecules, while dual reporter assay confirmed their binding to the 3’ UTR of their targets. Downregulation mediated by the selected miRNAs was sufficient to increase cytotoxicity (PD-L1) and reduce the trogocytotic potential (CTLA-4) of transfected cells. The PD-L1 miRNA signature was capable of predicting melanoma patients’ relapse when combined with CD8+ T cell infiltration and PD-L1 expression, while the CTLA-4 miRNA expression correlated with changes in the CD4+ T cell subset in melanoma patients before and after treatment with anti-CTLA-4 antibodies. The expression of 8 inflammation related miRNAs showed a differential expression pattern between healthy donors and MDS/sAML patients, while the generation of the infla-miR signature increased its diagnostic and prognostic potential. BGN was identified as a mediator of inflammation in MDS along with the miRNAs, showing BGN-mediated pyroptosis in the in vitro model used and in the BMBs. Recombinant BGN was capable of disrupting hematopoiesis in vitro, while the effect could be reversed by inhibitors of the inflammasome pathway and antibodies. Knockdown of BGN in MDS cells induced myeloid differentiation, while patients with high levels of BGN in their bone marrow were deemed detrimental for MDS patients. Identification of immune modulatory miRNAs can allow us to effectively stratify patients, when analysed in parallel with immune infiltration and could be an intervention tool to restore dysregulated pathways in the context of cancer. Moreover, changes in the regulation of miRNAs can indicate systematic changes in the tumor microenvironment and direct us towards the identification of novel biomarkers.
