Solid-Phase Parallel Synthesis of Dual Histone Deacetylase-Cyclooxygenase Inhibitors

    No full text
Multi-target drugs (MTDs) are emerging alternatives to combination therapies. Since both histone deacetylases (HDACs) and cyclooxygenase-2 (COX-2) are known to be overexpressed in several cancer types, we herein report the design, synthesis, and biological evaluation of a library of dual HDAC-COX inhibitors. The designed compounds were synthesized via an efficient parallel synthesis approach using preloaded solid-phase resins. Biological in vitro assays demonstrated that several of the synthesized compounds possess pronounced inhibitory activities against HDAC and COX isoforms. The membrane permeability and inhibition of cellular HDAC activity of selected compounds were confirmed by whole-cell HDAC inhibition assays and immunoblot experiments. The most promising dual inhibitors, C3 and C4, evoked antiproliferative effects in the low micromolar concentration range and caused a significant increase in apoptotic cells. In contrast to previous reports, the simultaneous inhibition of HDAC and COX activity by dual HDAC-COX inhibitors or combination treatments with vorinostat and celecoxib did not result in additive or synergistic anticancer activities

Triage in Times of COVID-19: A Moral Dilemma

    No full text
We present evidence from choice experiments on hypothetical triage decisions in a pandemic. Respondents have to decide who out of two patients gets ventilation. Patients are described in terms of attributes such as short-term survival chance, long-term life expectancy, and their current ventilation status. Attributes are derived from the ethical discourse among experts regarding triage guidelines during the COVID-19 pandemic and differ in the extent to which they are salient from a utilitarian or deontological perspective. Empirically, we find that although nonexperts agree with experts in prioritizing utilitarian attributes in triage decisions, nonexperts also consider the adherence to the norm of wearing face masks as particularly relevant. Furthermore, our study supports Greene and colleagues’ dual-process model of moral judgment; we find that utilitarian attributes are more decisive for respondents with a greater inclination toward utilitarianism and for respondents with a greater tendency toward reflection

Entwicklung eines Demonstrators zur autonomen, maritimen und robotergestützten E-Fuel-Betankung

    No full text

A New Risk Assessment Model for Unexploded Underwater Military Munitions

    No full text
This study addresses the need for improved risk assessment (RA) during explosive ordnance disposal (EOD) at sea by highlighting the environmental and operational complexities of handling unexploded ordnance (UXO) in the marine environment. It discusses the increasing demand for offshore EOD due to the construction of offshore wind parks and initiatives proactively addressing UXO dump sites. Conducting EOD work professionally and diligently to prevent accidents and environmental contamination will, hence, continue to be of paramount importance. In an effort to understand the RA tools that are available to EOD experts, this work reviews ten RA methods and identifies a methodological gap, thereby revealing the need for a new EOD RA method. Consequently, it introduces a novel method named the Risk Assessment Model for Unexploded underwater Military Munitions (RUMMs). This model is based on a directed graph and considers properties and aspects of the UXO, the environment, and the EOD procedure. RUMMs, which can be used during early EOD planning states or immediately before UXO handling, calculates the probability of an undesired detonation, the consequence of an undesired detonation, and the complexity of the EOD operation. The model is extensively explored by means of local sensitivity analysis and Monte Carlo simulation to demonstrate its reliability. Subsequently, RUMMs is tested with twenty EOD operations, which yield plausible model outputs that are in line with expectations and real-world decisions. The model's consistent and reproducible results, unaffected by fatigue or time pressure, add a new layer of reliability to EOD decision-making.:1 Introduction 1.1 Offshore UXO in a Multi-Use and Multiple Stressor Environment 1.2 Focus, Scope, and Purpose of this Dissertation 1.3 Research Hypotheses 1.4 Structure of the Dissertation 2 The Issue of Offshore Unexploded Ordnance 2.1 Background and Context 2.2 UXO Properties 2.3 Potential Effects of the Presence of UXO 2.4 Conclusion—The Case for Explosive Ordnance Disposal 3 Offshore Explosive Ordnance Disposal 3.1 EOD frameworks 3.2 EOD procedure According to the Selected Framework 3.3 EOD in Practice 3.4 Environmental Properties Affecting EOD 3.5 Conclusion—The Case for EOD Risk Assessment 4 Review of Risk Assessment Methodologies 4.1 Risk—A Brief Overview 4.2 Preparation of the Offshore UXO and EOD Risk Assessment Review 4.3 Offshore UXO Risk Assessment Review 4.4 Conclusion—The Case for a Novel Method for EOD RA 5 Development and Description of the EOD RA Model 5.1 Background of the EOD RA Model 5.2 Development Process of RUMMs 5.3 Undesired Events 5.4 Exceptions, Inherent Risk and Residual Risk 5.5 Risk Receptors 5.6 The Two Model Layouts 5.7 RUMMs Factors 5.8 Omitted Factors 5.9 Model Outputs 5.10 Other RUMMs Functionalities 5.11 Model Limitations 5.12 Conclusion—The Case for Model Exploration and Testing 6 Methodological Exploration and Application 6.1 Local Sensitivity Analysis 6.2 Monte Carlo Simulation 6.3 Application of RUMMs 6.4 Revision of the Gap Analysis 6.5 The Future of RUMMs 6.6 Conclusion—The Case for Real World Application 7 Conclusion 7.1 Recap of the Research Hypotheses 7.2 Outlook on RUMMs 7.3 Recommendations for UXO and EOD Research 7.4 RUMMs’ Response to Other Demands 7.5 RUMMs and the Way Forward in Offshore EO

Sustainability of wastewater management: A socio-ecological system (SES) approach for textile industry in Bangladesh with a focus on Resource Nexus

    No full text
The dissertation examines the sustainability of managing textile wastewater within the textile value chain (TVC), emphasizing resource interplay, environmental impacts, and the integration of the Resource Nexus framework. The studies of this dissertation analyze tangible environmental resources (e.g., water, energy, material), intangible environmental resources (e.g., technological and organizational training), and governance resources (e.g., regulations, monitoring, research & development) dimensions, including sustainable practices, compliance, and stakeholder perceptions in the context of textile wastewater management (TWM). The studies of this dissertation utilize a combination of qualitative methods, such as thematic analysis of interviews and focus group discussions (FGDs), alongside quantitative approaches, including survey data analysis and structural equation modeling (SEM). Qualitative data offer insights into stakeholder perceptions and regulatory frameworks, while quantitative data provide empirical evidence of associations between resource management strategies, environmental governance, and water quality. The dissertation on the sustainability of TWM highlights the significance of stakeholder engagement for effective regulation and monitoring; however, there's a notable gap in fully exploring diverse stakeholders' perspectives and their roles in decision-making processes. While acknowledging the importance of community livelihoods and ecosystem health, some studies inadequately assess the socio-economic impacts and ecological consequences of wastewater discharge on neighboring communities, overlooking conflicts arising from resource usage and discharge. Additionally, although proposing frameworks like the Resource Nexus, there is a lack of quantitative justification, hindering informed decision-making and policy formulation. Addressing these gaps is essential for enhancing the studies' relevance and applicability to improving TWM and achieving sustainable transformation. Furthermore, the dissertation significantly contributes by identifying gaps, providing empirical evidence, and suggesting policy interventions for sustainable TWM. It highlights disparities between compliant and non-compliant industries, emphasizes stakeholder engagement, promotes eco-technological innovations, and advocates for collaborative efforts among policymakers, industry stakeholders, and communities. These insights contribute to a comprehensive understanding of the Resource Nexus approach and offer actionable guidance for stakeholders aiming to address the challenges of sustainable TWM. These studies are crucial for addressing the sustainability challenges in textile wastewater management, particularly in contexts like Bangladesh. The studies of this dissertation offer insights into the complexities of resource management, stakeholder engagement, regulatory compliance, and the need for sustainable practices, ultimately contributing to achieving Sustainable Development Goals (SDGs) related to clean water, industry innovation and infrastructure, and sustainable communities, enhancing environmental performance, and fostering socio-ecological resilience. These findings contribute to a holistic understanding of the Resource Nexus approach and offer actionable insights for policymakers, industry stakeholders, and researchers aiming to address the challenges of TWM sustainably.:Summary I Table of Contents III List of Figures IV List of Tables V Abbreviations VI 1 Introduction 1 2 Research articles of the cumulative dissertation 5 Overview of the research articles 5 2.1.1 Implications and limitations 11 2.1.2 Outlook for future research 12 Resource nexus oriented decision making along the textile value chain: The case of wastewater management (Article 1) 14 Sustainability transformation in the textile industry – the case of wastewater management (Article 2) 17 Resource Nexus for decision-making: An industry-community synergistic approach for textile wastewater management (Article 3) 20 Improving water quality for environmental performance: The role of resource-based decision-making in textile wastewater management (Article 4) 22 3 References 26 4 Acknowledgements 33 5 Appendix A: Article 1 35 6 Appendix B: Article 2 83 7 Appendix C: Article 3 109 8 Appendix D: Article 4 13

And can it tell the story of the city? Seemingly dull grey street concrete tells the story of world history - and filigree mosaics are reminiscent of the Wittenberge town architect Friede Bruns, who continued the city planning 100 years ago. Street trees are remnants of the industrial era and the cross of the Protestant town church has been carved from centuries-old street planks. Nothing is ordinary here - all the more reason for us to learn to appreciate the seemingly self-evident. The perspectives in this book are very diverse: from historians to construction workers, from citizens to urban developers, everyone who works, walks on and remembers the streets of Wittenberge on a daily basis has their say.:Oliver Hermann Grußwort S. 9 Marcel Steller Vorwort S. 10-11 Marcel Steller Digital ins Mittelalter – Eine Einführung S. 12-25 Marcel Steller Barfuß mit allen Sinnen S. 26-37 Felix Oldörp Barfuß die Stadt erkunden S. 38-45 Steffi Gottwald Straßen und Plätze prägen den Charakter einer Stadt S. 46-53 Andreas Grieswald Wie werden Wittenberges Gehwege sauber? Premiere Sa 01. Dez 201
