    Statistical optics methods for generating, simulating, and characterising emission from relativistic electron beams

    Synchrotron radiation sources and free-electron lasers have revolutionised the study of matter and biological systems by providing unique combinations of radiation pulse characteristics. Although these two types of facilities differ fundamentally in terms of pulses time scales, coherence properties, and (peak) brilliance, each offers an exceptional mix of these features. The combination of coherence, short pulse duration, and high photon flux, characteristic of both types of sources, delivers unparalleled experimental capabilities across a wide range of photon energies. However, these advanced capabilities also introduce new challenges in both the characterisation and numerical modelling of this radiation. Improving our understanding of radiation diagnostics techniques and developing new simulation methods could pave the way for innovative approaches to enhance pulse properties. I begin my thesis by exploring the potential for developing new theoretical models for describing partially coherent sources, with a particular focus on radiation from relativistic electron beams. To this end, I propose a set of numerically efficient algorithms designed to simulate individual statistical instances of the radiation field. These algorithms are highly versatile and efficient, capable of simulating radiation from a range of sources, from fully incoherent to partially coherent systems, including thermal sources, synchrotron radiation, and even free-electron laser radiation with complicated Wigner function distributions. In particular, one of the developed algorithms offers a new perspective on synchrotron radiation. Building on this, I propose a method for measuring the coherence length of synchrotron radiation at linear accelerator facilities, such as the European XFEL, using a non-interferometric approach. By detecting radiation directly after monochromatization and employing auto-correlation analysis, the coherence length can be determined without the need for an interference scheme. This measurement also reveals that the monochromatized transverse distribution of synchrotron radiation consists of spikes. In practice, this method serves as a novel diagnostic tool for determining the transverse electron beam size at free-electron laser facilities, leveraging the van Cittert-Zernike theorem to relate the transverse coherence length to the electron beam size. Next, I focus on the virtual source structure of undulator radiation from a single electron. Although undulator radiation in resonance is typically considered to have a single waist at the virtual source position, closer inspection reveals finer details. There are two characteristic lengths associated with undulator radiation: the overall device length and the period length. These scales correspond to the radiation formation lengths and are directly related to the diffraction size of the radiation. In this part, I examine the less-studied peculiarities of the virtual source associated with the undulator period length. Finally, I describe a numerical code for simulating synchrotron radiation generation in the presence of waveguide effects. These effects typically arise in the infrared frequency range and are influenced by the metallic elements of the beamlines, which are always present. I conducted various cross-checks of the developed code, comparing it with the Synchrotron Radiation Workshop code for free-space scenarios and with analytical expressions for practical cases involving circular waveguides. This research contributes to the development of radiation beamlines for THz pump -- X-ray probe experiments at free-electron laser facilities. The final part of the thesis includes numerical calculations for an iris waveguide designed for THz radiation transport through the beamlines of free-electron lasers

    Multiviszerale Resektionen und Gefäßresektionen bei viszeralen und retroperitonealen Sarkomen: Auswirkungen auf die Rezidivrate und das Gesamtüberleben

    Das Ziel dieser Arbeit ist, die chirurgische Behandlung retroperitonealer und viszeraler Sarkome besser zu verstehen. Dabei sollen Parameter identifiziert werden, die die langfristige Prognose bzw. die Rate an Lokalrezidiven sowie das Gesamtüberleben der Patienten beeinflussen. Nicht nur die Heterogenität der über 100 verschiedenen Sarkomentitäten und die geringen Fallzahlen machen diese Tumore zu komplexen Herausforderungen. Auch die enorme Größe der retroperitoneal liegenden Sarkome und der schwierige chirurgische Zugang sorgen dafür, dass viele Patienten ein Rezidiv entwickeln und an ihrer Erkrankung versterben. Die vollständige Resektion des Sarkoms stellt den einzigen kurativen Ansatz dar und multiviszerale Resektionen sind zum Standard der Behandlung geworden. Dabei galt die Beteiligung eines großen Gefäßes lange als Kontraindikation für einen operativen Eingriff. In dieser Arbeit wurde ein besonderes Augenmerk auf Patienten geworfen, bei denen eine multiviszerale Resektion oder eine Gefäßresektion durchgeführt wurde. Für die Bearbeitung haben wir eine ausführliche Datenbank zur retrospektiven Analyse der allgemeinchirurgischen Patienten mit retroperitonealen und viszeralen Sarkomen des Universitätsklinikums Hamburg- Eppendorf (UKE) angelegt. Im Beobachtungszeitraum 2014-2020 wurden 111 Patienten identifiziert, die wegen eines retroperitonealen (73 Fälle) oder viszeralen Sarkoms (38 Fälle) operiert wurden. Von diesen Patienten haben 71,2% eine multiviszerale Resektion erhalten und bei 18% war eine Gefäßresektion notwendig. Mit dem Ansatz der multiviszeralen Resektion zum Erreichen eines suffizienten Sicherheitsabstandes konnten wir 5 Jahre nach der Operation ein Gesamtüberleben von 59,46% bei einer Rezidivrate von 58,42% erreichen. Unsere multivariate Analyse hat gezeigt, dass das Grading einen negativen Einfluss sowohl auf die Rezidivrate als auch auf das Gesamtüberleben hat. Außerdem zeigten auch der randbildende Resektionsstatus sowie schwere Morbidität in Form von Re-Operationen einen signifikant negativen Einfluss auf das Überleben der Patienten. Neben diesen Ergebnissen gibt die deskriptive Aufarbeitung des Kollektivs einen guten Überblick über die wichtigsten Aspekte unserer Sarkompatienten. Die große Schwierigkeit dieser Arbeit ist die geringe Anzahl an Patienten. Dies führt, gemeinsam mit den Schwächen einer retrospektiven Datenbearbeitung dazu, dass die meisten Ergebnisse nicht signifikant oder repräsentativ sind. Dieser Schwierigkeit könnte in Zukunft mit prospektiven Modellen und längeren Beobachtungszeiträumen begegnet werden. Auch die Beurteilung der Lebensqualität bei multiviszeralen Resektionen im Rahmen der Sarkomerkrankungen ist eine interessante und wichtige Thematik, die im Zuge einer prospektiven Befragung angegangen werden könnte. The aim of this study is to enhance the understanding of the surgical treatment of retroperitoneal and visceral sarcomas. The objective is to identify parameters that influence the rate of local recurrences, distant metastasis and the overall survival of patients. The complexity of these tumors arises not only from the heterogeneity of over 100 different sarcoma entities but also from the limited case numbers. Additionally, the considerable size of retroperitoneally located sarcomas and the challenging surgical access contribute to the development of recurrences and mortality in afflicted patients. Complete resection of the sarcoma remains the sole curative approach, with multivisceral resections becoming the standard treatment. The involvement of a major vessel was traditionally considered a contraindication for surgical intervention. This study specifically focuses on patients who underwent multivisceral resection or vascular resection. An extensive database for the retrospective analysis of general surgical patients with retroperitoneal and visceral sarcomas at the University Medical Center Hamburg-Eppendorf (UKE) was established for this purpose. During the observation period from 2014 to 2020, 111 patients were identified who underwent surgery for retroperitoneal (73 cases) or visceral sarcomas (38 cases). Among these patients, 71.2% underwent multivisceral resection, and 18% required vascular resection. Employing the approach of multivisceral resection to achieve an adequate safety margin, we achieved an overall survival of 59,46% and a recurrence rate of 58,42% five years post-operation. Our multivariate analysis revealed that tumor grading negatively influenced both the recurrence rate and overall survival. Furthermore, margin-negative resection status and severe morbidity in the form of reoperations also significantly negatively impacted patient survival. In addition to these findings, the descriptive analysis of the cohort provides a comprehensive overview of the key aspects of our sarcoma patients. The major challenge of this study lies in the limited number of patients. Together with the limitations of retrospective data processing, this results in many outcomes being non-significant or not representative. Addressing this challenge in the future could involve prospective models and longer observation periods. Evaluating the quality of life in the context of multivisceral resections for sarcoma diseases is also an interesting and important theme that could be explored through a prospective survey

    The effect of electromagnetic fields on tendinopathies. Study on the effect of a singular application of high-energy pulsed electromagnetic fields

    Tendinopathien sind Erkrankungen, die durch Degeneration, Trauma oder Überlastung entstehen. Sie sind oft langwierig und neigen zu Rezidiven. Die aktuelle Therapie besteht in einer Kombination aus Orthesen, Physiotherapie, ggf. NSAR und physikalische Verfahren wie Ultraschall oder Elektrotherapie. Auch gepulste elektromagnetische Felder (PEMF)-Therapie hat bereits Einzug in die S2k-Leitlinie zur Epicondylitis erreicht. Jedoch gibt es aktuell keine Empfehlung für PEMF-Therapie bei Tendinopathien, da die Studienlage unzureichend ist. Neuere Studien haben jedoch gezeigt, dass hier möglicherweise doch positive Effekte vorliegen können und die PEMF-Therapie im Verlauf reevaluiert werden sollte. Dies basiert auf Grundlagenstudien, die einen Effekt auf verschiedene Signalwege beschrieben haben, u.a. Ca2+-Signalkaskade, Stickstoffmonoxidsignalkaskade, Heat-Shock-Protein(HSP)-70-Expression. Die dabei genutzten PEMF-Energien liegen in den bisherigen Studien zumeist in einem Bereich von ein bis maximal 300mT. Unsere Studie ist die erste, die eine Effektanalyse einer hochenergetischen PEMF-Therapie (750mT) auf Tendinopathien durchgeführt hat. Die dafür konzipierte Studie wurde als monozentrische klinisch kontrollierte Studie mit einfacher Verblindung (für Probanden und statistische Auswertung) angelegt. Die Probandenanzahl wurde mittels Poweranalyse (2 Gruppen, 5 Messzeitpunkte, Effektstärke = 0,25, Signifikanzniveau = 0,05, Power 90%) auf 26 Probanden errechnet, in Erwartung eines Drop-Outs von 25% wurden 34 Probanden in die Studie eingeschleust. Die Probanden wurden zufällig auf zwei Gruppen (Verum vs. Placebo) aufgeteilt. Für die Verumgruppe konnten 17 Probanden in die statistische Auswertung (kein Ausfall) aufgenommen werden. In der Placebogruppe wurden 4 Probanden ausgeschlossen. Für eine Inklusion in die Studie mussten die Probanden volljährig und geschäftsfähig sein, sowie eine klar lokalisierbare Tendinopathie aufweisen. Ausschlusskriterien waren eine gestörte Schmerzwahrnehmung, eine vorliegende Krebserkrankung, Schwangerschaft oder Implantate. Für die Messung der Schmerzen wurde die Druckschmerzschwelle (PPT) gewählt. Bei der PPT handelt es sich um eine sehr gut validierte und mit hoher Interklassenkorrelation ausgestattete objektivierbare Messmethode für Schmerzen. Zur Anwendung der PPT wird eine Druckplatte auf den Schmerzpunkt aufgelegt, bei Erhöhung des Drucks kann bei Schmerzangabe die Kraft in Newton vom Gerät abgelesen werden. Um die Wirkung unserer Behandlung auch laborchemisch nachzuweisen, wurde das HSP 70 im Blut der Probanden bestimmt. HSP 70 ist eine Proteinfamilie die entscheidend in der Faltung und den Transport von Proteinen über die Zellmembran beteiligt ist und so Entzündungsreaktionen, Zellregeneration und Apoptoseraten beeinflusst. Nach der Baselineuntersuchung (PPT und HSP 70) wurde den Probanden der Verumgruppe eine 10-minütige PEMF-Therapie mit 750mT appliziert. Der Placebogruppe wurde eine Therapie simuliert. Unmittelbar nach Therapie erfolgte eine zweite PPT-Bestimmung, sowie eine Bestimmung der PPT und HSP 70 Werte nach einer Stunde, einem Tag und sieben Tagen. Die statistische Auswertung erfolgte für die kategorialen Daten (Geschlecht und ICD-10-Diagnsoe) mittels absoluter und relativer Häufigkeiten. Zur Analyse von Baseline-Gruppenunterschieden wurde ein Chi-Quadrat-Test (inkl. Cramers-V) durchgeführt. Hier zeigten sich keine Baselineunterschiede. Für die kontinuierlichen Daten (Größe, Gewicht, BMI und abhängige Variablen PPT und HSP 70) erfolgte eine Auswertung über Mittelwert und Standardabweichung, zur Untersuchung von Baseline-Gruppenunterschieden wurde der Student's-T-Test durchgeführt. Auch hier zeigten sich keine Baselineunterschiede. Bei vorliegender Normalverteilung wurde für die abhängigen Variablen eine 2-way-rANOVA-Analyse durchgeführt. Bei signifikantem Ergebnis wurde einer Bonferroni-Prozedur durchgeführt, bei Verletzung der Spharizität eine Greenhouse-Geisser-Korrektur. Im Ergebnis konnte eine signifikante Zunahme der Druckschmerzschwelle in der Verumgruppe über die Zeit um im Mittel 110% (p<0,001) festgestellt werden. Für die Placebogruppe konnte eine Zunahme der Druckschmerzschwelle über die Zeit von etwa 20% beobachtet werden. Dies entsprach in etwa dem erwarteten Placeboeffekt und war nicht signifikant. Für HSP 70 konnte keine signifikante Veränderung festgestellt werden. Elektrotherapeutische Ansätze müssen aufgrund der hohen Energie und damit einhergehender Induktionsströme in der Bewertung der unmittelbaren Ergebnisse berücksichtigt werden. Langzeiteffekt über einen Tag oder gar eine Woche können bei Elektrotherapie aber nicht beobachtet werden.Tendinopathies are diseases caused by degeneration, trauma or overloading. They are often protracted and tend to recur. Current therapy consists of a combination of orthoses, physiotherapy, possibly NSAIDs and physical procedures such as ultrasound or electrotherapy. Pulsed electromagnetic field (PEMF) therapy has also been included in the S2k guideline on epicondylitis. However, there is currently no recommendation for PEMF therapy for tendinopathies, as the study situation is insufficient. However, recent studies have shown that there may be positive effects and that PEMF therapy should be re-evaluated over time. This is based on basic studies that have described an effect on various signaling pathways, including the Ca2+ signaling cascade, nitric oxide signaling cascade and heat shock protein (HSP)-70 expression. The PEMF energies used in previous studies are mostly in the range of one to a maximum of 300mT. Our study is the first to analyse the effect of high-energy PEMF therapy (750mT) on tendinopathies. The study designed for this purpose was set up as a monocentric clinically controlled study with single blinding (for subjects and statistical analysis). The number of subjects was calculated using power analysis (2 groups, 5 measurement times, effect size = 0.25, significance level = 0.05, power 90%) to 26 subjects; 34 subjects were enrolled in the study in anticipation of a drop-out of 25%. The subjects were randomly divided into two groups (verum vs. placebo). For the verum group, 17 subjects were included in the statistical analysis (no drop-out). In the placebo group, 4 subjects were excluded. To be included in the study, the subjects had to be of legal age, legally competent and have a clearly localizable tendinopathy. Exclusion criteria were impaired pain perception, an existing cancer, pregnancy or implants. The pressure pain threshold (PPT) was chosen to measure pain. The PPT is a very well validated and objectifiable measurement method for pain with a high interclass correlation. To apply the PPT, a pressure plate is placed on the pain point; when the pressure is increased, the force in Newtons can be read off the device when the pain is indicated. In order to prove the effect of our treatment in the laboratory, HSP 70 was determined in the blood of the test subjects. HSP 70 is a protein family that plays a decisive role in the folding and transport of proteins across the cell membrane and thus influences inflammatory reactions, cell regeneration and apoptosis rates. After the baseline examination (PPT and HSP 70), a 10-minute PEMF therapy with 750mT was applied to the subjects in the verum group. Therapy was simulated for the placebo group. Immediately after therapy, a second PPT determination was carried out, as well as a determination of the PPT and HSP 70 values after one hour, one day and seven days. The statistical analysis was carried out for the categorical data (gender and ICD-10-diagnosis) using absolute and relative frequencies. A chi-square test (incl. Cramers-V) was performed to analyse baseline group differences. No baseline differences were found here. The continuous data (height, weight, BMI and dependent variables PPT and HSP 70) was analysed using the mean and standard deviation; the Stundent's T-test was used to examine baseline group differences. No baseline differences were found here either. In the case of a normal distribution, a 2-way ANOVA analysis was carried out for the dependent variables. If the result was significant, a Bonferroni procedure was carried out; if sphericity was violated, a Greenhouse-Geisser correction was carried out. The result showed a significant increase in the pressure pain threshold in the verum group over time by an average of 110% (p<0.001). For the placebo group, an increase in the pressure pain threshold over time of around 20% was observed. This corresponded approximately to the expected placebo effect and was not significant. No significant change was observed for HSP 70. Electrotherapeutic approaches must be considered in the evaluation of the immediate results due to the high energy and associated induction currents. However, long-term effects over a day or even a week cannot be observed with electrotherapy

    Chemogenetic Manipulation of Microglia: Effects on neuronal structure and function

    Microglia, the resident immune cells of the central nervous system (CNS), play a critical role in maintaining homeostasis, immune surveillance, and modulating neuronal function. They are highly dynamic cells capable of responding to environmental changes by altering their morphology and function. Recent research has highlighted that microglia are not only involved in immune responses but also play a pivotal role in synaptic plasticity, synaptic pruning, and overall cognitive processes. Activation of microglia, whether by inflammatory processes or chronic diseases, has profound consequences on neurons and synapses, leading to alterations in synaptic strength, connectivity, and ultimately cognitive functions. However, the detailed mechanisms underlying microglia-neuron communication remain under active investigation. Traditional methods to activate microglia, such as pharmacological agents like lipopolysaccharide (LPS) or genetic approaches including global gene knockout models, can be non-specific and affect multiple cell types, making it difficult to isolate the direct role of microglia in neuronal modulation. These methods also typically lack precise timing, further complicating our understanding of microglia's direct influence. To address these challenges, I took a more targeted approach by directly activating microglia using a chemogenetic tool to target the Gq signaling pathway. In this dissertation, I employed a chemogenetic strategy with the tamoxifen-inducible Cre-ER loxP system to selectively express Gq-DREADD (Designer Receptors Exclusively Activated by Designer Drugs) in microglia. This approach allowed us to activate microglia in a controlled and targeted way, avoiding unwanted effects on other brain cells. The activation of the Gq pathway led to changes in microglial morphology, specifically in reduced branching complexity and increased intracellular calcium levels suggesting a shift in their functional state. Additionally, the chronic Gq-DREADD activation induced a decrease in the number of excitatory synapses on CA1 pyramidal neurons., which was found to be dependent on brain-derived neurotrophic factor (BDNF) signaling. As well as a reduction in long-term potentiation (LTP) in the hippocampus, which is a key mechanism for synaptic plasticity and memory. Despite these significant changes at the synaptic level, the mice with activated microglia initially performed well in learning and memory tasks, such as locating a hidden platform in the Morris water maze. However, in the following days, their ability to retain spatial information declined compared to controls, indicating that while learning was unaffected, the stability of the memories formed was compromised. Our findings show that selective activation of microglia can negatively impact synaptic plasticity and memory retention without inducing other overt symptoms of sickness. This method offers a valuable tool for studying the effects of microglial activation on neuronal function with high specificity and precise timing

    The development and conceptualisation of statistics and economics are closely linked. Historically, a variety of different concepts of statistics have been advocated by different (schools of) economists, ranging from the use of statistics as a mere means of mass observation and description to the application of complex mathematical models from which causalities were inferred. The use of statistics depended on how economists defined the purpose of economic studies as well as their understanding of the type of knowledge that can and should be generated in economics. Throughout history, different fields of application dominated the discussion and further development of concepts of statistics. In the 19th century, statistics developed in particular as researchers turned their attention to the quantitative study of the “society”. At the beginning of the 20th century, the study of business cycles encouraged the advancement of statistics. This dissertation examines two examples of the conception and use of statistics in the 1860s/70s and the second half of the 1920s by economists in the tradition of German historical economics. Georg Friedrich Knapp (1842-1926) was a central figure of the German Younger Historical School of Economics. His “historical-realistic” concept of economics and the social sciences that envisaged a factual, objective and apolitical investigation of the economy and the society was shaped in particular by his early work on statistics (1865-1874). While he favoured the holistic and descriptive understanding of German statistics, which accounted for the historical peculiarity of empirical observations and was shared by the economists of the Younger Historical School, he distanced himself in part from the “historical-ethical” research programme of its leader, Gustav Schmoller (1838-1917). Knapp demanded the extension of quantitative statistical observation even to such spheres, like the investigation of morality, which seemed to be reserved mainly to philosophical investigations and refrained from normative presuppositions to empirical observation, like the idea of an ongoing cultural progress. In order to enhance the scientific rigour of statistical studies in German economics, he developed a formal-mathematical theory of population statistics that provided a systematic framework for ordering and analysing population data. Knapp applied this approach of organising knowledge even to his monetary theory, in which he provided a taxonomy of historical monetary constitutions and adhered to the requirements of scientific rigour that he developed in his formal treatises on statistics. This “statistical mode” of scientific investigation pervaded his entire work. Since the late 1910s, statistical business cycle research internationally developed into a dominant field of economics. Its results were made accessible to official and private stake-holders via a rising number of statistical barometers. In 1926, Kurt Singer (1886-1962) and the renowned German business cycle researcher Arthur Spiethoff (1873-1957) introduced a regular statistical business cycle reporting to the Hamburg-based economic weekly “Wirtschaftsdienst. Weltwirtschaftliche Nachrichten” that consisted of two complementary pillars – the “Wirtschaftsbarometer” and the reports “Zur Lage”. Spiethoff’s business cycle theory provided the main analytical and interpretive framework for the business cycle reporting of the Wirtschaftsdienst between 1926 and 1930. His theory was based on a comprehensive empirical study of business cycles in the 19th and early 20th century up to World War I. From this study, Spiethoff derived a small number of empirical indicators that described the typical course of the cycle. These indicators were prioritised in the Wirtschaftsbarometer and the reports Zur Lage for the assessment of the overall economic situation. When the data series of the Wirtschaftsbarometer deviated from the expected typical course, the authors at the Wirtschaftsdienst reflected on the weaknesses of the suggested empirical indicators, but still decidedly adhered to Spiethoff’s general approach and his explanation of business cycles. Singer demarcated the business cycle analysis of the Wirtschaftsdienst from other approaches that were applied in Germany since the mid-1920s. In 1926, he sharply criticised the work of the “Institut für Konjuntkurforschung” in Berlin, led by Ernst Wagemann (1884-1956) and thereby initiated a debate with Adolf Löwe (1893-1995), who headed the department for business cycle research at the “Königliches Institut für Seeverkehr und Weltwirtschaft” in Kiel, about the appropriate way to conduct business cycle research. Singer’s critique and his demarcation appear to be exaggerated against the background of similarities in the model cycle and the assessment of the overall economic situation in the publications of the IfK and the Wirtschaftsdienst. Strategic considerations and personal interests are part of the explanation of Singer’s attack on Wagemann and the IfK. A further important explanation lies in substantial differences in the methodological standpoints of all three involved parties. In fact, the dispute between Singer and Löwe can be regarded as being embedded in a more general controversy about the reorganisation and reorientation of German economics after World War I and the breakdown of the German Younger Historical School of Economics. With favouring an inductive and holistic business cycle theory, which refrained from mathematical-statistical reasoning, Spiethoff and Singer adhered closest to the tradition of German statistics and German historical economics. Wagemann aligned the work of the IfK to American empiricism and focused on formal-statistical reasoning. Löwe’s endeavour to combine deductive theorising with mathematical-statistical testing corresponded to the vision of upcoming econometrics. The latter two were thus moving further away from German historical economics and towards an understanding of economics as a formal and exact science

    Die Aufklärung hochauflösender dreidimensionaler Strukturen von Proteinen mittels Röntgenstrukturanalyse erfordert Proteinkristalle zur Signalverstärkung. Proteinkristalle wurden in lebenden Zellen aller Domänen beobachtet. Ergänzend zu konventionellen Kristallisationsmethoden wurde in der vorliegenden kumulativen Arbeit ein systematischer Ansatz für die Kristallisation rekombinanter Proteine in lebenden Insektenzellen verfeinert und ausgeweitet. Dabei wurde insbesondere die Detektion der Kristalle in cellulo verbessert, indem eine Kleinwinkelstreuung/ Pulverdiffraktions Methode zum Screenen von Zellkulturen entwickelt wurde. Dafür wurden mit rekombinanten Baculoviren infizierte Zellkulturen an einer Hintergrund-optimierten Synchrotron-Strahlführung vermessen und mit Mockvirus infizierten und mit uninfizierten Insektenzellkulturen verglichen. Sind Kristalle in der Zellkultur vorhanden, so liegen bei dieser Messung Beugungssignale in zufälliger Orientierung vor. Dadurch werden auf dem Detektor schwache unvollständige Debye-Scherrer Ringe aufgenommen, die durch Summation mehrerer Bilder, radialer Mittlung und Hintergrundsubtraktion in 1D-Plots der Streuwinkelkurve als Peaks erscheinen. Es konnte gezeigt werden, dass bei einer minimalen Kristallbildungsrate von unter einem bis etwa sechs Prozent ausreicht, um innerhalb von Sekunden Proteinmikrokristalle in der Zellkultur zu identifizieren. Die minimal notwendige Kristallbildungsrate ist v.a. abhängig vom expremierten Zielgen. Um die Effizienz und Sensitivität der Methode bestimmen zu können wurden dafür Proteine gewählt, die in cellulo deutlich mit Lichtmikroskopie sichtbare Kristalle bilden. Die Dynamik der Kristallbildung in den Zellen wurde im Ensemble untersucht. Dadurch konnte der optimale Zeitpunkt für das serielle Kristallographieexperiment ermittelt werden. Serielle Kristallographie (SX) Beugungsdatenaufnahme mittels einer Synchrotron Strahlungsquelle und Freie Elektronenlasern wurde in cellulo - also ohne vorherige Extraktion und Reinigung der Kristalle aus den Zellen - etabliert, da sich gezeigt hat, dass kein erhöhtes Hintergrundsignal bei Datenaufnahme in lebenden Zellen auftritt, und die umgebende Zelle die Proteinkristalle vor etwaigen Schäden durch das Isolationsverfahren und vor Dehydration schützt. Am Synchrotron wurde anhand verschiedener Konstrukte der Proteine N. crassa HEX-1 und T. brucei Inosinmonophosphatdehydrogenase (TbIMPDH) die Datenaufnahme in MicroMeshes (MiteGen) mit einem helikalen Linienscan gezeigt und die Strukturen gelöst. Auch die Datenaufnahme bei Raumtemperatur (RT) wurde demonstriert, wobei es durch die Verwendung von CrystalDirectTM Platten nach Beschichtung mit Poly D Lysin möglich war, sowohl eine rekombinante Bakulovirus (rBV) Infektion, Kristallwachstum als auch die Datenaufnahme in cellulo und in situ durchzuführen. Dies vermeidet Schäden an potentiell fragilen Kristallen. Diffraktionsdatensammlung bei RT kann alternative Konformere aufdecken und ermöglicht automatisierbare Datenaufnahme an bis zu 96 Proben ohne Trägerwechsel. SX in cellulo am Freie Elektronenlaser LCLS wurde mit einem fixed-target Verfahren erfolgreich demonstriert. Im Vergleich zur Probenlieferung mit einem Liquid Jet entfallen die Risiken von Verstopfung einer Düse oder Sedimentieren der Probe im Reservoir. Somit kann eine hohe Trefferquote erzielt werden. Interessanterweise ermöglicht Kristallisation in der lebenden Zelle die Bindung von genuinen Kofaktoren, welche in physiologischer Konzentration in den Kristall diffundieren oder kokristallisieren. Wird auch die Datenaufnahme in intakten Zellen durchgeführt, können diese Kofaktoren bei ausreichend starker Bindung und Datenqualität identifiziert werden. Auf diese Weise konnte gezeigt werden, dass die inhibierende Konformation der TbIMPDH ein Phosphat an der IMP Bindungsstelle inkorporiert, und in den kanonischen Nukleotidbindungsstellen GDP und ATP mit einer alternativen Konformation. Für die Lösung der Struktur nach erfolgreicher Diffraktionsdatensammlung gibt es die Möglichkeiten des molekularen Ersatzes, sei es mit einer bereits experimentell gelösten homologen Struktur oder seit AlphaFold2 auch anhand eines in silico Modells. Um Bias zu vermeiden wurde angestrebt schwere Atome in intrazelluläre Kristalle einzubringen, anhand derer de novo Phasierung getestet werden könnte. Es wurden erfolgreich TbIMPDH Konstrukte mit einem Lanthanid-Bindetag (LBT) intrazellulär kristallisiert. Diese Kristalle zeigten eine geringere Stabilität. SAD-Phasierung von Test-Diffraktionsdaten der TbIMPDH mit terminalem LBT war nicht möglich, weil Terbium intrazellulär nicht ausreichend spezifisch bindet und sich nach Kristallisolation die Auflösung der Diffraktionsdaten durch Terbium-Soaking reduziert. Als alternativen Ansatz wurde die Inkorporation von Selen als Selenomethionin während der Genexpression getestet. Durch rBV Infektion der mit einem sehr hohen Virustiter (MOI 10) konnte eine erhöhte Inkorporation von Selen in den Kristallen mittels Massenspektrometrie für Neurospora crassa HEX-1 nachgewiesen werden. Dieser Ansatz ist vielversprechend für eine zukünftige SAD-Phasierung, sollte jedoch für weitere Proteine validiert werden. Die Resultate dieser kumulativen Dissertation leisten einen Beitrag zum Methodensprektrum der Strukturbiologie, indem die in cellulo Kristallisation für eine größere Gruppe Wissenschaftler durch vereinfachte Detektion und serielle Datenaufnahme in intakten Insektenzellen als Ergänzung zu konventioneller in vitro Kristallisation zugänglich gemacht wird.Elucidation of high-resolution three-dimensional protein structures via X-ray crystallography requires protein crystals for signal enhancement. Protein crystals have been observed in living cells from all domains of life. This cumulative dissertation refines and expands a systematic approach for crystallization of recombinantly introduced proteins in living insect cells, as a complementary method to conventional crystallization approaches. In particular, detection of crystals in cellulo has been optimized by development of a small angle X-ray scattering/ powder diffraction synergistic method to screen cell cultures. To this end, recombinant baculovirus infected cell cultures were measured at a low-background optimized synchrotron beamline. Their X-ray scattering profile was compared to that of mockvirus-infected and uninfected insect cells. If protein crystals are irradiated, they are in random orientation within the cells. Consequently, weak incomplete Debye-Scherrer rings are recorded, which by summation of multiple detector images, radial averaging, and background subtraction show as peaks in the 1D-plots of the scattering curve. It could be shown that a crystallization efficiency in cells of less than one to six per cent suffices to identify within seconds if there are protein microcrystals within a cell culture. This is largely dependant on the expressed target protein. To this end, proteins were chosen that form clearly visible crystals identifieable by light microscopy. Thus, the efficiency and sensitivity of the established method could be elucidated. Additionally, the dynamic behaviour of crystal growth within the cell ensemble was investigated. This enabled determination of the optimal timepoint after infection for a serial crystallography (SX) experiment. It became clear that data collection within living cells, meaning in cellulo with no prior extraction and purification of crystals, does not increase background noise. Going further, the surrounding cell even protects protein crystals from dehydration and damage during crystal extraction. In this work, SX data collection in cellulo was established at synchrotron radiation sources and using X-ray free-electron laser (XFEL) radiation. With synchrotron radiation, efficient data collection on MicroMeshes (MiteGen) with helical line scans is demonstrated on different constructs of the proteins Neurospora crassa HEX-1 (NcHEX-1) and Tb inosine 5’-monophosphate dehydrogenase (TbIMPDH), solving the structures. In addition, data collection at room temperature (RT) is demonstrated. Using CrystalDirectTM plates with a poly D-lysine coating for enhanced cell adhesion, everything from cell infection, crystal growth up to data collection in the intact cells can be done in situ. This way, any damage on potentially fragile intracellular crystals is prevented. Collecting at RT can also uncover potential alternative conformers. The method enabels automatable data collection on up to 96 sampels without changing the mount. Successful SX in cellulo on XFELs is demonstrated with a fixed-target approach on micro-patterned silicon chips. Compared to liquid jets, risks of nozzle clogging or crystal sedimentation in the sample reservoir are eliminated, reducing sample consumption and increasing hitrates. Interestingly, crystallization inside cells enables binding of genuine co-factors abundant at physiological concentrations. Ligands can either diffuse into or co-crystallize with the crystal. Collecting diffraction data in viable insect cells can lead to identification of such co-factors, provided sufficiently strong binding and data quality. This way it can be shown that the inactive conformation of TbIMPDH incorporates phosphate at the IMP binding site, as well as GDP and ATP in alternate conformations at canonical nucleotide binding sites. Solving the structure after successful data collection can be done by phase retrieval with molecular replacement. Phases can be transfered from a experimentally phased homologue, or more recently from in silico models generated f.i. with AlphaFold2. In order to prevent bias, it is still desirable to introduce heavy atoms into intracellular protein crystals, which later could be used for experimental de novo phasing. Towards this end, TbIMPDH constructs with terminal lanthanide binding tags (LBT) were successfully crystallized in cellulo, unfortunately leading to increased crystal fragility compared to the TbIMPDH in cellulo crystals without LBT. Moreover, unspecific binding of the lanthanide terbium to other moelcules within the cell was confirmed, and, terbium soaking reduced resolution of diffraction data. As an alternate approach, the incorporation of selenium in the form of selenium methionine was tested on different targets during gene expression. It was hypothized and proven by mass spectrometry for NcHEX-1 that a high virus titer (MOI 10) results in an increased selenium incorporation. The potential of this approach for a future phasing via SAD needs to be validated on additional proteins. Results of this cumulative dissertation contribute to the method spectrum of structural biology, opening the approach of in cellulo crystallization to a wider group of researchers.     A transforming Arctic: Unraveling Climate Change Impacts on Sea-Ice Evolution and Oceanic pCO2

    The Arctic undergoes profound transformations due to global warming, with climate change impacts varying at regional levels. While many changes, such as declining sea ice and alterations in ocean biochemistry, are often discussed based on the pan-Arctic average, their local implications and intensity variations across regions have been largely overlooked, despite their importance for stakeholders. This thesis aims to help shift the perspective towards understanding regional changes in the Arctic by investigating two crucial indicators affected by a warming climate: Arctic sea-ice coverage and surface ocean partial pressure of carbon dioxide (pCO2 ). The first is of interest to a variety of stakeholders, particularly to those interested in changes along the coast–ice transition zone, such as the shipping industry and indigenous people. The latter is a key factor in the exchange of CO2 between ocean and atmosphere and is, therefore, decisive for the acidification of the oceans. The sensitivity of sea-ice area to near-surface air temperature changes, i. e. how strongly sea-ice area diminishes for a given rise in temperature, exhibits a high seasonal dependence, characterized by significant variability in summer and low variability in winter. This study reveals that the transition between summer and winter, along with the observed low variability in winter, can be attributed to the geographic blocking of the sea-ice edge by surrounding land masses. By quantifying the timing of the blocking, the analysis links the timing changes to rising global temperatures. The findings indicate that as the timing shifts and the season during which the ice edge is blocked shortens (by around 7 days per tenth degree of global warming on average), adjacent seasons will experience heightened sensitivity in sea-ice area. Particularly, sensitivities in areas along the coasts of the high Arctic Ocean will undergo a sudden change in the future. Expanding the sensitivity analysis to a regional scale, the study provides new perspectives on how the sea ice in individual Arctic regions responds to global warming, anticipating future changes. The East Siberian Sea, the Chukchi Sea, and the Laptev Sea are identified as regions likely to lose their summer sea ice first. The Barents Sea is projected to become the first region to lose its remaining winter sea ice, ultimately becoming ice-free year-round. Shifting focus to surface ocean pCO2, two data products estimating surface ocean pCO2 to fill the sparse observations in the Arctic are used to investigate the evolution of surface ocean pCO2 in the Arctic domain. Both products reveal consistent increases in most regions over the last two decades. However, substantial differences in the magnitudes of inter- and intra-annual changes are observed between the two datasets. Separating the spatial and temporal variability in the pCO2 via EOF analysis reveals the dominant drivers of changes in the pCO2 in different domains of the Arctic. The study identifies sub-Arctic domain changes primarily related to the seasonal solar irradiance cycle, while high Arctic pCO2 changes are dominated by changes in sea-ice cover. Examining seasonality, shifts and changes in intra-annual amplitude are noted already in the historical record, particularly north of Canada, indicating potential impact on ecosystems. This thesis underscores the necessity of taking a regional perspective on Arctic climate change. By systematically analyzing and quantifying the blocking effect for the first time, it provides a foundation for understanding sea-ice area changes in the high Arctic, benefiting stakeholders interested in Arctic Ocean transformations, particularly in coastal regions. Additionally, the pCO2 analysis offers insights into the drivers of pCO2 in the Arctic Ocean, serving as a benchmark to enhance future data products, crucial for investigating regional changes in the Arctic surface ocean carbon cycle. These findings contribute to the groundwork for future research in the Arctic.Die Arktis ist aufgrund der globalen Erwärmung tiefgreifenden Veränderungen unterworfen, wobei sich die Auswirkungen des Klimawandels je nach Region stark unterscheiden. Während viele Veränderungen, wie der Rückgang des Meereises und Veränderungen in der Meeres-Biochemie, oft auf pan-arktischer Ebene diskutiert werden, blieben ihre lokalen Auswirkungen und Unterschiede in deren Intensität zwischen den Regionen lange Zeit weitgehend unbeachtet, obwohl sie für verschiedene Interessensgruppen von großer Bedeutung sind. Diese Dissertation soll dazu beitragen, den Fokus auf das Verständnis regionaler Änderungen in der Arktis zu verlagern, indem sie zwei wesentliche Indikatoren untersucht, die von einem wärmeren Klima betroffen sind: die Meereisbedeckung der Arktis und der Partialdruck von Kohlendioxid (pCO2 ) an der Meeresoberfläche. Erstere ist von Interesse für eine Vielzahl von Interessensgruppen, insbesondere für diejenigen, die an Veränderungen entlang der Küsten-Eis-Übergangszone interessiert sind, wie z. B. die Schifffahrtsindustrie und indigene Bevölkerungsgruppen. Letztere ist ein Schlüsselfaktor für den Austausch von CO2 zwischen Ozean und Atmosphäre und entsprechend maßgeblich entscheidend für die Versauerung der Ozeane. Die Sensitivität der Meereisfläche gegenüber Veränderungen in der oberflächennahen Lufttemperatur, d. h. wie stark sich die Meereisfläche bei gegebenem Temperaturanstieg verringert, zeigt eine hohe saisonale Abhängigkeit, die durch starke Variabilität im Sommer und geringe Variabilität im Winter gekennzeichnet ist. Diese Studie zeigt, dass der Übergang zwischen Sommer und Winter sowie die geringe Variabilität im Winter auf das Blockieren des Meereises durch umliegende Landmassen zurückzuführen ist. Dies wird in der Arbeit als “geographic muting” oder auch “geographic blocking” bezeichnet. Durch die Quantifizierung des Zeitpunkts dieses Blocking-Effekts wird eine klare Relation zwischen dem Timing dem globalen Temperaturanstieg festgestellt. Die Ergebnisse deuten darauf hin, dass mit der Verschiebung des Timings und der Verkürzung der Saison, über welche das Meereis blockiert ist, (im Durchschnitt etwa 7 Tage pro Zehntelgrad globaler Erwärmung) benachbarte Jahreszeiten eine erhöhte Sensitivität der Meereisfläche zeigen werden. Insbesondere Sensitivitäten in küstennahen Regionen werden zukünftig einer plötzlichen Verände rung unterliegen. Durch Ausweiten der Sensitivitätsanalyse auf regionale Ebene zeigt die Studie neue Perspektiven auf, wie das Meereis in einzelnen Regionen der Arktis auf die globale Erwärmung reagiert, und antizipiert zukünftige Änderungen. Die Ost-Sibirische See, die Tschuktschensee und die Laptewsee werden als Regionen identifiziert, die mit großer Wahrscheinlichkeit zuerst ihr Sommer-Meereis verlieren werden. Die Barentssee wird dagegen voraussichtlich die erste Region sein, die ihr verbleibendes Winter-Meereis verliert und letztendlich das ganze Jahr über eisfrei sein wird. Bei der anschließenden Betrachtung des oberflächennahen Partialdrucks von Kohlendioxid (pCO2 ) werden zwei Datensätze verwendet, welche pCO2 aus lückenhaften Beobachtungen schätzen, um die Entwicklung des pCO2 in der Arktis zu untersuchen. Beide Datensätze zeigen über die letzten zwei Jahrzehnte eine konsistente Zunahme des pCO2 für die meisten Regionen. Dabei werden jedoch erhebliche Unterschiede im Ausmaß der zwischenjährlichen und innerjährlichen Veränderungen zwischen den beiden Datensätzen beobachtet. Indem räumliche und zeitliche Variabilität im pCO2 mittels EOF-Analyse separiert werden, lassen sich die dominierenden Treiber jener Veränderungen identifizieren. Die Studie zeigt, dass Veränderungen im subarktischen Bereich hauptsächlich dem saisonalen Zyklus der Sonneneinstrahlung zugeordnet werden können, während in der hohen Arktis Prozesse dominieren, die mit Änderungen in der Meereisbedeckung in Verbindung stehen. Bei der Untersuchung der Saisonalität, lassen sich Hinweise auf Verschiebungen und Änderungen in der saisonalen Amplitude bereits in den historischen Daten feststellen (insbesondere nördlich von Kanada), was potenzielle Auswirkungen auf die dortigen Ökosysteme vermuten lässt. Diese Dissertation unterstreicht die Notwendigkeit, eine regionale Perspektive auf den arktischen Klimawandel einzunehmen. Indem der Blocking-Effekt erstmals systematisch analysiert und quantifiziert wird, legt diese Arbeit die Grundlage für das Verständnis von Veränderungen in der Meereisfläche in der hohen Arktis und dürfte somit von Nutzen für Interessengruppen sein, für welche die Transformationen des Arktischen Ozeans, insbesondere in Küstenregionen, von Bedeutung sind. Darüber hinaus bietet die pCO2-Analyse Einblicke in die Treiber des pCO2 im Arktischen Ozean und dient somit auch der Verbesserung zukünftiger Datensätze, die für die Untersuchung regionaler Veränderungen im Kohlenstoffkreislauf der arktischen Meeresoberfläche entscheidend sind.     Das Lebensmittel Fisch hat seit Jahrtausenden eine große Bedeutung für den Menschen, aufgrund seiner Nährstoffzusammensetzung, der globalen Verfügbarkeit und teilweise leichten Zugänglichkeit. Fisch als lebenswichtige Quelle der menschlichen Ernährung steht aufgrund der Überfischung vor Herausforderungen, was die Entwicklung der Aquakultur und nachhaltiger innovativer Fütterungsstrategien anregt. Die Arbeiten dieser Dissertation konzentrierten sich auf eine verringerte Abhängigkeit von Fischmehl und Fischöl in der Fischhaltung durch den Einsatz von Mikroorganismen-biomasse als alternativem Rohstoff. Ziel war es, durch neue Futtermittel eine vergleichbare oder bessere Fischfiletqualität als herkömmliche Fischfilets zu erreichen. Nach Auswahl der Mikroorganismen folgte die Entwicklung verschiedener Futtermittel, welche in einer zweistufigen Studie an Zandern (Sander lucioperca) und Regenbogenforellen (Oncorhynchus mykiss) getestet wurden. Die Zusammensetzung des finalen Futters umfasste die Arten Rhodotorula glutinis, Crypthecodinium cohnii und Arthrospira sp., womit die Hälfte des herkömmlichen Fischmehl- und Fischölgehalts im Futtermittel ersetzt werden konnte. Die an Zander und Regenbogenforellen durchgeführten Untersuchungen zeigten, dass das auf Mikro-organismen basierende Futtermittel das Wachstum und die Qualität der Speisefische aufrechterhält, die mit herkömmlichen Futtermitteln für Regenbogenforellen vergleichbar sind. Beim Zander besteht weiterhin Optimierungsbedarf, da das Futtermittel zu einer verminderten Wachstumsleistung führte. Die sensorische Bewertung der Fischfilets beider Spezies ergab nur geringfügige Unterschiede in Textur, Geruch und Geschmack, was auf das Potenzial für eine breite Marktakzeptanz hinweist. Die Arbeiten betonen die Bedeutung neuer, nachhaltiger Futtermittelzutaten und deren Potential in der Aquakultur.The fish as a food has a huge importance to human nutrition for thousands of years, due to its nutrient composition, global availability and, in some cases, easy accessibility. Fish as an essential source of human nutrition faces challenges due to overfishing, giving rise to the development of aquaculture and sustainable alternatives, such as innovative fish feed ingredients. The studies in this dissertation focused on reducing the dependence on fishmeal and fish oil by using a microorganism biomass mixture as alternative ingredient. The studies presented aimed for a comparable or better quality of fish fillet than conventional fish fillet by using the new feed. After the selection of microorganisms, various feed formulations were developed, which were further investigated in a two-stage study on pike-perch (Sander lucioperca) and rainbow trout (Oncorhynchus mykiss). The composition of the final feed included the species Rhodotorula glutinis, Crypthecodinium cohnii, and Arthrospira sp., which were able to replace half of the conventional fishmeal and fish oil content in feed. The studies showed that the microorganism-based feed maintains growth and quality of rainbow trout that was comparable to conventional rainbow trout feeds. There is still a need for optimization when it comes to pike-perch, as the feed led to reduced growth performance. Sensory evaluation of pike-perch and rainbow trout fillets revealed minor differences in texture, odor, and taste, indicating potential for wider market acceptance.
