    Establishment of cell lines expressing erythrocyte membrane protein 1 (PfEMP1) variants of choice to study Plasmodium falciparum cytoadherence associated functions

    Malaria, caused by a unicellular eukaryotic apicomplexan parasite of the genus Plasmodium, remains a leading cause of morbidity and mortality globally. The pathological most relevant species for humans is Plasmodium falciparum which is responsible for almost all of the malaria related deaths. The symptoms of the disease are exclusively caused by the asexual stages of the parasite’s life cycle, during which the parasites multiply in erythrocytes of the host. Erythrocytes infected with P. falciparum can cytoadhere to various host receptors and ligands on the endothelium to evade the removal of the infected erythrocytes in the spleen. This can lead to blockage and inflammation of blood vessels, cutting off tissue from oxygen support, and resulting in severe forms of the disease, such as cerebral malaria. The interaction of infected erythrocytes with the host´s endothelium is mediated by a large parasite encoded transmembrane protein presented on the erythrocyte surface termed Plasmodium falciparum erythrocyte membrane protein 1 (PfEMP1). PfEMP1 variants possess differing binding properties and are expressed by a multigene family, the var genes. The expression of var genes in the parasite is mutually exclusive so that a parasite expresses only a single var gene. Furthermore, the parasites can switch their var gene expression over time, enabling the parasites to persistently evade the host´s immune system, facilitating long lasting infections. PfEMP1 is considered a key factor for the parasite’s virulence and survival, rendering it as the main virulence factor of P. falciparum parasites. However, the study of specific PfEMP1 variants and var genes has been hampered by PfEMP1s large size and the fact that parasite in vitro cultures are a mix of parasites expressing different PfEMP1s, making them inaccessible to genetic manipulation and molecular analysis. In this thesis, selection linked integration (SLI) was exploited to generate cell lines that express a single HA-tagged PfEMP1 of choice in the entire population of parasites in an in vitro culture. The predominant expression of targeted var genes were validated for transgenic 3D7 and IT4 (formerly FCR3) lab strains by bulk RNA sequencing. The trackability of the HA-tagged PfEMP1s in the cell was demonstrated with immunofluorescence assays. Trypsin cleavage assays confirmed the surface presentation of the targeted PfEMP1 variant in most cases. SLI modified 3D7 and IT4 parasites expressing VAR2CSA or a CD36-binding PfEMP1 variant on the erythrocyte surface were tested and compared for binding against corresponding ligands. This provided empirical evidence that the IT4 strain is superior for the study of PfEMP1-mediated binding while 3D7 parasites only showed poor binding. To increase binding assay throughput and comparability, a semi-automated pipeline was developed for standardized evaluation of binding assays. Furthermore, PfEMP1 variants were activated in IT4 parasites that showed specific binding against the major endothelial cell receptors CD36, ICAM-1 and EPCR, validating the feasibility of this approach. These cell lines are expected to facilitate the study of the binding properties of different PFEMP1 variants without the need to subject the parasites to elaborate receptor or antibody selection procedures. This will enable the comparison of binding capacities and to explore potentially novel interacting receptors of specific PfEMP1 variants. The trackability of the HA-tagged PfEMP1 was further exploited to study PfEMP1 transport into the host cell in 3D7 parasites. In order to do this, an approach to conditionally block protein transport through PTEX, the translocon pumping exported proteins into the host cell, was chosen. When PTEX was blocked, PfEMP1 was no longer exported. The timing of this approach in the parasite blood cycle was chosen in a way to minimize the effect of accessory factors needed for PfEMP1 transport which would indirectly lead to a failure of PfEMP1 export. Overall, these experiments indicated that PfEMP1 is transported by PTEX into the host cell. Next, BioID experiments were conducted to determine the proxiome of PfEMP1 in living parasites. For this the biotin ligase BirA* was fused to three distinct positions of PfEMP1, resulting in three different cell lines. Testing of the cell lines expressing the PfEMP1-BirA* fusions showed that the modified PfEMP1s were predominantly expressed in the parasite cultures and functionally presented on the host cells surface, exhibiting binding against the expected receptors. Large scale BioID experiments with subsequent mass spectrometry analysis of biotinylated proteins identified proteins shown to be important for PfEMP1s transport and function as well as previously characterized and uncharacterized proteins of the Maurer´s clefts, erythrocyte membrane and other host cell structures. The proxiome of the three positions showed marginal differences to each other, indicating that PfEMP1 is not transported as an integral membrane protein. Furthermore, only partial overlap with a Maurer’s clefts proxiome indicated the detected hits were PfEMP1-specific. Notably six exported proteins with unknown function previously not linked to cytoadherence were identified and termed PfEMP1-interacting candidate 1-6 (EMPIC1-6). Subsequently, a modified version of SLI, SLI2, was utilized to introduce a second endogenous modification to the cell lines harboring a SLI-activated var gene. This was used to disrupt candidate proteins identified in the proxiome to evaluate their role in PfEMP1-mediated cytoadherence. The evaluation of PfEMP1 presentation and function with trypsin cleavage assays and binding assays led to the discovery of two novel proteins that are needed for the efficient cytoadherence of P. falciparum parasites despite the fact that their disruption did not ablate PfEMP1 surface transport. In summary, this thesis -by utilizing SLI techniques - successfully generated and validated cell lines expressing specific PfEMP1 variants, which is expected to substantially aid studies on the interaction between these proteins and host cell receptors, var gene switching mechanisms and transport of PfEMP1. This work not only advances our understanding of P. falciparum´s main virulence factor but also opens new avenues to illuminate the cellular mechanisms that enable cytoadherence, leading to a better understanding of the parasite-host interactions.Malaria wird durch einen einzelligen eukaryotischen Apicomplexa-Parasiten der Gattung Plasmodium verursacht und bleibt weltweit eine führende Ursache für Morbidität und Mortalität. Die für den Menschen pathologisch relevanteste Spezies ist Plasmodium falciparum, welche für fast alle Malaria-bedingten Todesfälle verantwortlich ist. Die Symptome der Krankheit werden ausschließlich durch die asexuellen Stadien des Lebenszyklus des Parasiten verursacht. Während dieser vermehrt sich der Parasiten in den Erythrozyten des Wirts. Erythrozyten, die mit P. falciparum infizierte sind, können an verschiedene Rezeptoren und Liganden des Wirts auf den Endothelzellen zytoadhärieren, wodurch diese der Zerstörung durch die Milz entgehen. Dies kann zu Entzündungen bis hin zum Verschluss der Blutgefäße führen, wodurch Gewebe von der Sauerstoffversorgung abgeschnitten wird. Dies führt zu schweren Formen wie zum Beispiel Malaria, einer der schwersten Komplikationen dieser Infektion. Die Interaktion infizierter Erythrozyten mit den Endothelzellen des Wirts wird durch ein großes auf der Oberfläche der Erythrozyten präsentiertes Parasite-Transmembranprotein hervorgerufen, bekannt als P. falciparum erythrocyte membrane protein 1 (PfEMP1). Verschiedene PfEMP1-Varianten besitzen unterschiedliche Bindungseigenschaften und werden von einer Multigenfamilie, den var-Genen, kodiert. Die Expression von var-Genen im Parasiten ist sich gegenseitig ausschließend, das heißt, dass ein Parasit jeweils nur ein einzelnes var-Gen exprimiert. Außerdem verändern die Parasiten ihre var-Gene expression über die Zeit. Dies ermöglicht es den Parasiten, dauerhaft dem Immunsystem des Wirts zu entgehen und langanhaltende Infektionen zu manifestieren. PfEMP1 wird deshalb als Schlüsselfaktoren für die Virulenz und das Überleben des Parasiten angesehen, weshalb dieser als Hauptvirulenzfaktor von P. falciparum-Parasiten bezeichnet werden. Die Untersuchung spezifischer PfEMP1-Varianten und var-Gene wird durch die große Größe von PfEMP1 sowie durch die Tatsache erschwert, dass Parasiten in In-vitro-Kulturen eine Mischung von var-Genen exprimieren. Dies macht sie für genetische Manipulationen und molekulare Analysen schwer zugänglich. In dieser Dissertation wurde das Selection Linked Integration (SLI)-System genutzt, um Zelllinien zu generieren, die in der gesamten Parasitenpopulation einer in-vitro-Kultur ein einzelnes erwünschtes HA-markiertes PfEMP1 exprimieren. Die Expression der Ziel-var-Gene wurde für transgene SLI-Parasitenpopulationen durch Bulk-RNA-Sequenzierung validiert. Die HA-markierten PfEMP1s konnte in der Zelle mit Immunfluoreszenzexperimenten lokalisiert werden und der Oberflächenproteinverdau mit Trypsin bestätigte für die meisten Zelllinien die Oberflächenpräsentation der modifizierten PfEMP1-Variante. Es wurden zwei P. Falciparum Laborstämme mit SLI-modifiziert, 3D7 und IT4 (ehemals FCR3), die VAR2CSA oder eine CD36-bindende PfEMP1-Variante auf der Oberfläche der Erythrozyten exprimierten, wurden auf ihre Bindungseigenschaften verglichen, indem ihre Bindung gegenüber entsprechenden Liganden getestet wurden. Dies lieferte empirische Hinweis dafür, dass der IT4-Parasitenstamm für die Untersuchung von PfEMP1-Bindungen geeigneter ist als der 3D7-Parasitentamm, welcher lediglich schwache Bindung zeigte. Um den Durchsatz und die Vergleichbarkeit von Bindungsexperimenten zu verbessern, wurde eine semi-automatisierte Pipeline für eine standardisierte Auswertung von Bindungsexperimenten entwickelt. Darüber hinaus wurden in IT4-Parasiten PfEMP1-Varianten aktiviert, welche eine spezifische Bindung gegen die prominentesten interagierenden Zellrezeptoren, CD36, ICAM-1 und EPCR zeigten. Dies validierte die Zuverlässigkeit des SLI-Systems für diese Anwendungen. Es ist zu erwarten, dass diese Zelllinien die Erforschung der Bindungseigenschaften verschiedener PfEMP1-Varianten vereinfachen, da diese Parasiten ohne Rezeptor- oder Antikörperselektion generiert wurden. Dies ermöglicht den Vergleich der Bindungskapazitäten und die Erforschung potenzieller neuer interagierender Rezeptoren spezifischer PfEMP1-Varianten. Des Weiteren wurde die Möglichkeit das HA-markierten PfEMP1 in der Zelle zu lokalisieren genutzt, um den Transport von PfEMP1 in die Wirtszelle bei 3D7-Parasiten zu untersuchen. Zu diesem Zweck wurde der Proteinexport durch das PTEX-Translocon, welches exportierte Proteine in die Wirtszelle transloziert, induzierbar blockiert. Bei einer Blockierung von PTEX, wurde PfEMP1 nicht mehr in die Wirtszelle exportiert. Der Zeitpunkt der Blockierung wurde im asexuellen Zyklus so induziert, dass früh exportierte Proteine, die potenziell für den PfEMP1-Transport benötigt werden, nicht beeinflusst wurden, da diese indirekt den Export von PfEMP1 beeinflussen könnten. Insgesamt deuten die Ergebnisse dieser Experimente darauf hin, dass PfEMP1 durch PTEX in die Wirtszelle transloziert wird. Außerdem wurden BioID-Experimente durchgeführt, um Proteine in der Umgebung (Proxiom) von PfEMP1 in lebenden Parasiten zu identifizieren. Dafür wurde die Biotin-Ligase BirA* an drei unterschiedliche Positionen von PfEMP1 fusioniert, wodurch drei verschiedenen Zelllinien hergestellt wurden. Für diese Zelllinien wurde gezeigt, dass die PfEMP1-Varianten mit dem fusionierten BirA* prädominant in der Parasitenkulturen exprimiert und auf der Oberfläche der Wirtszellen präsentiert wurden. Des Weiteren wurden die Bindung der modifizierten PfEMP1 gegen die erwarteten Rezeptoren zeigten. BioID-Experimente im großen Maßstab mit anschließender Massenspektrometrie-Analyse der biotinylierten Proteine identifizierten Proteine, die für den Transport und die Funktion von PfEMP1 relevant sind. Darüber hinaus wurden zuvor charakterisierte und nicht charakterisierte Proteine der Maurer´schen Spalten, der Erythrozytenmembran und anderer Strukturen der Wirtszelle detektiert. Das Proxiom der drei Positionen zeigte nur kleine Unterschiede zueinander, was darauf hinweist, dass PfEMP1 nicht als integrales Membranprotein transportiert wird. Darüber hinaus deutete eine nur teilweise Überlappung mit einem Proxiom der Maurer´schen Spalten darauf hin, dass die detektierten Proteine PfEMP1-spezifisch waren. Dadurch wurden sechs exportierte Proteine mit unbekannter Funktion, die zuvor nicht mit Zytoadhärenz in Verbindung gebracht wurden, identifiziert und als PfEMP1-Interacting Candidate 1-6 (EMPIC1-6) benannt. Des Weiteren wurde eine modifizierte Version von SLI, SLI2, angewandt, um eine zweite endogene Modifikation in den Zelllinien einzubringen, die bereits ein SLI-aktiviertes var-Gene beherbergten. Dies wurde verwendet, um die Funktion von Kandidatenproteine, die im Proxiom identifiziert wurden, zu trunkieren und deren Rolle im Transport und Oberflächenpräsentation von PfEMP1 zu ermitteln. Dies führte zur Identifizierung von zwei neuen Proteinen, die in Oberfächenproteinverdau- und Bindungsexperimenten zeigten, dass diese Kandidaten relevant für die effiziente Zytoadhärenz von P. falciparum-Parasiten sind. Jedoch wurde der Transport von PfEMP1 an die Oberfläche durch die Disruption der Kandidatenproteine nicht beeinflusst. Zusammenfassend hat diese Dissertation - durch die Nutzung von SLI-Techniken - erfolgreich Zelllinien generiert und validiert, die spezifische PfEMP1-Varianten exprimieren Es ist zu erwarten, dass dies die Studien über die Interaktion dieser Proteine mit Wirtszellrezeptoren, var-Gen-Expressionsmechanismen und den Transport von PfEMP1 erheblich vereinfachen wird. Diese Arbeit erweitert nicht nur unser Verständnis des Hauptvirulenzfaktors von P. falciparum, sondern eröffnet auch neue Wege, um die zellulären Mechanismen der Zytoadhärenz zu beleuchten, was dabei helfen wird, die Parasiten-Wirt-Interaktion besser zu verstehen

    Applications of Test Masses in Uniform Circular Motion to Detect Millihertz Gravitational Waves

    In this doctoral dissertation, starting from the first principles of the general theory of relativity, the geodesic deviation of test masses in uniform circular motion is calculated, due to the passage of gravitational waves (GWs) from astrophysical sources. The results of this theoretical analysis are used to conceive of experimental setups that could utilize this principle to detect GWs. These setups are based on, but are not limited by, the current status of technology. Mainly, circular particle accelerators in the mode of storage rings are considered. The major sources of noise for a storage ring gravitational wave observatory (SRGO) are explored, and basic noise estimates are made based on the specifications of existing facilities such as the Large Hadron Collider (LHC). A major part of the dissertation consists of numerical simulations of toy models of an SRGO. The response signal of the GW detector is explored as a function of various model parameters. Errors on estimating the model parameters are found using Markov Chain Monte Carlo (MCMC) methods applied to artificially generated data with added noise. It is shown that an ideal version of such a detector would be sensitive to millihertz GWs by design, and would be able to single-handedly obtain accurate constraints on the GW source parameters. An attempt has been made to incorporate as much physics as currently possible, which covers the essential aspects of reality without over-complicating the models for a first study of this novel experiment concept. The overall focus has been to establish a strong conceptual foundation, to obtain robust qualitative conclusions, and to make order-of-magnitude estimates. Comparisons with the theory and practices of existing GW detectors have been made throughout. Wherever possible, the results are presented in the conventions followed by the GW community at the time of writing this dissertation

    Generate Modelling in High Energy Physics

    Monte Carlo simulations are an important tool in high-energy physics, e.g. to test the predictions of theory models or to infer a priori unknown parameters of the models. However, these simulations demand a substantial amount of computational resources. Thus, this thesis explores the viability of neural-network-based generative models for the CMS experiment at the LHC. The upcoming upgrades of the LHC further challenge the available computing budget of the collaboration in the near future, as these upgrades are expected to substantially increase the number of recorded collisions, necessitating a corresponding expansion in Monte Carlo simulation. For the CMS experiment, the simulation of a single event currently requires approximately two minutes. However, the required time is further expected to at least double, due to upgrades of the CMS detector. This increase is mainly owed to the upgrade of the endcap calorimeters, where the resulting number of channels that need to be simulated will be significantly higher. First, studies on the high-energy physics community JetNet dataset are extensively discussed, and the performance of different generative models is compared. An attention-based information aggregation, which scales linearly with the number of particles in terms of computational complexity, is proposed. Not only does this lead to state-of-the-art results on the JetNet datasets, but also promising results on the CaloChallenge. Finally, the viability of an end-to-end generation approach is studied in a search for Supersymmetry. The semi-leptonic decay of gluinos, produced via pair production, to neutralinos with an intermediate chargino in the decay chain is investigated. In this search, three a priori unknown parameters need to be scanned, which correspond to the masses of the superpartners. Typically, the mass of the intermediate particle is not scanned, since it is not feasible to generate Monte Carlo simulated data for all parameter combinations. The consequences of not scanning the chargino mass when a neural-network-based classifier is used to identify a signal pure region of the phase space for the statistical inference are investigated. Instead of fixing the mass to an arbitrary value, it is explored whether synthetic data from a generative model, which transforms the distributions from different values of the chargino mass into another, improves the statistical significance of the search. In this study, the required integrated luminosity to reach a similar statistical significance is reduced by 20 ± 12%.Monte-Carlo-Simulationen sind in der Hochenergiephysik weitverbreitet, benötigen aber viele Rechenressourcen. In dieser Arbeit wird untersucht, ob generative Modelle auf Basis neuronaler Netze für das CMS-Experiment am LHC verwendet werden können, um die Rechenzeit für die Simulationen zu reduzieren. Die bevorstehenden Erweiterungen des LHC erhöhen die Anforderungen bezüglich Rechenressourcen und Datenmenge. Aktuell dauert die Simulation einer Kollision etwa zwei Minuten, aber es wird erwartet, dass sich diese Zeit durch die geplanten Aktualisierungen verdoppeln oder verdreifachen wird. Eine schnellere Simulationstechnik wird daher benötigt. Die Arbeit konzentriert sich zunächst auf die Untersuchung JetNet-Datensatzes und vergleicht die Qualität und Geschwindigkeit verschiedener generativer Modelle. Eine Informationsaggregationstechnik, die auf Attention basiert und linear mit der Anzahl der Teilchen skaliert, bildet das Herzstück des Modells. Weiterhin kann das Modell mit minimaler Veränderung dazu verwendet werden, die Energieeinträgen aufgrund elektromagnetischer Schauer in Kalorimetern konditioniert zu modellieren. Abschließend wird die direkte Generierung von rekonstruierten Analysedaten am Beispiel einer Suche nach Supersymmetrie untersucht. Hier wird der Zerfall von Gluinos, produziert via Paarproduktion, mit einem intermediären Chargino in der Zerfallskette untersucht. Dabei müssen drei unbekannte Massenparameter gescannt werden. Normalerweise würde in diesem Fall nur Monte-Carlo Datensätze für verschiedene Kombinationen von zwei der drei Parametern generiert, während der Dritte auf die Hälfte der Summe der anderen beiden gesetzt wird. Es wird geprüft, ob ein generatives Modell die Abhängigkeit der Daten auf den letzteren Parameter lernen kann, und ob Samples von dem Modell die statistische Signifikanz der Suche verbessern können. Diese Studie zeigt, dass die notwendige integrierte Luminosität um 20 ± 12% reduziert werden kann durch die Verwendung der synthetischen Daten

    Die Darstellung des Weiblichen in den lateinamerikanischen Chroniken vom XV bis zum XVII Jahrhundert

    La Literatura Hispanoamericana se remonta al encuentro de culturas por parte del Almirante Cristóbal Colón y los pueblos originarios americanos en 1492. Colón y los primeros españoles llevaron una nueva cultura y el germen de la nueva expresión literaria (Oviedo, 1995: 72). La literatura brotó en América con los conquistadores a través de formas básicas como los romances, plegarias, sátiras, canciones populares y otras de mayor importancia como la crónica, el teatro y la poesía popular (Oviedo, 1995: 72). A este grupo heterogéneo de documentos como cartas, relaciones y crónicas que escriben capitanes, soldados, frailes e intelectuales en América o sobre América durante la colonia y la conquista se denomina “Crónicas de Indias”. Este corpus textual que conforman las crónicas de Indias constituye una unidad porque tiene en común el referente, o sea, el descubrimiento y la conquista de Indias y ciertas fronteras cronológico-ideológicas (Mignolo, 1982: 58). Mignolo señala durante el siglo XVI que el vocablo crónica tuvo una trayectoria paralela al vocablo historia (1982: 75). Sostiene que conocer la evolución del término es fundamental porque es el punto de partida para identificar el tipo de discurso al cual se refiere quien lo emplea, ya sea mediante uno o ambos vocablos. El vocablo se origina en la palabra historia (del griego ἱστορία) y “significa también el informe de lo visto o lo aprendido por medio de las preguntas” (Mignolo, 1982: 75). El vocablo no contiene en su definición ningún componente temporal. El vocablo crónica se usaba para denominar el informe del pasado o la anotación de los acontecimientos del presente, fuertemente estructurados por la secuencia temporal. Era como una lista organizada sobre las fechas de los acontecimientos que se desean conservar en la memoria (Mignolo, 1982: 75). Con la llegada a América de los españoles, estos textos precisos y formales evolucionaron y se transformaron en observaciones, fantasías y sugerencias de los cronistas. El discurso de los primeros autores hispanoamericanos presenta características que hacen a estos textos únicos. Los narradores testigos de las crónicas de Indias son narradores autodiegéticos y homodiegéticos. Los letrados alternan elementos factuales y ficcionales. A esta alternancia de los autores, López de Mariscal, la define como el discurso de ficción, y por otro lado, el discurso histórico, o sea, el fáctico (López de Mariscal, 2004: 187). Rolena Adorno presenta un trabajo sobre las Polémicas sobre la posesión de las Indias en las letras hispanoamericanas (2009) y presenta tres argumentos sobre la ficcionalidad y factualidad del papel de las letras coloniales en la literatura latinoamericana. En primer lugar, las crónicas de Indias no solían orientarse por eventos históricos sino por los debates sobre éstas: el derecho o no de imponer el dominio español sobre los nativos. En segundo lugar, estas crónicas se pueden descubrir cómo la literatura de ficción deriva del género de la historia y por último, la centralidad en la tradición cultural hispanoamericana de los debates está todavía presente en obras actuales (Adorno, Rolena, 2009: 68). Los criterios de los textos del corpus de este trabajo son los siguientes: documentos escritos, mononlingües en castellano, excepto la Historia general de las cosas de Nueva España. Se trata de un corpus diacrónico (1492-1609) que tiene como referente común a América. Todos los autores son hombres y españoles a excepción de Cristóbal Colón que es italiano. Según el ámbito social de los autores, se trata de un navegante y comerciante, dos de ellos pertenecen a familias con un cierto origen noble y a su vez son militares, dos son soldados de la conquista y tres son sacerdotes, dos de ellos dominicos y uno franciscano. Las narraciones se efectúan desde diferentes puntos de americanos. Se dividieron a los cronistas en: cronista del encuentro a Cristóbal Colón y el Diario de a bordo (1492); y de la conquista y exploración a Hernán Cortés y las Cartas de Relación (1519-1526); Bernal Díaz del Castillo y la Historia verdadera de la conquista de la Nueva España [1541-1558?] (1632); Bartolomé de las Casas y la Brevísima relación de la Destrucción de las Indias (1552); Álvar Núñez Cabeza de Vaca y la relación de los naufragios y comentarios [1558], Jerónimo de Vivar y la Crónica y relación copiosa y verdadera de los Reinos de Chile [1540-1579] (1905); Bernardo de Sahagún y la Historia general de las cosas de Nueva España [1540-1579]; Reginaldo de Lizárraga y la Descripción del Perú, Tucumán, Río de la Plata y Chile [1602-1609] (1916). En esta tesis se analiza la mirada de los cronistas de las mujeres españolas, indias y mestizas. Se analiza a los cronistas según la representación estereotípica que realizan de las mujeres y se observa si existe la figura femenina en las crónicas y cómo es. La condición de las mujeres ha estado desde hace siglos marcada por su posición de subordinación respecto a la del hombre y por su exclusión de la mayoría de las áreas de actividad vinculadas de una u otra manera al ejercicio de alguna forma de poder dirigida por el discurso teológico, científico y jurídico. Esto trajo el inconveniente que los derechos femeninos no han sido los mismos que los de los varones. Para encontrar estas representaciones en los cronista se plantean tres vertientes siguiendo a Rolena Adorno (1988: 59) a modo de reconstruir la relación entre el discurso caballeresco español y el indianista: 1. se ubica al amerindio que es percibido por el sujeto colonizador con una visión europeizante; 2. se encuentra la figura del indio y de la india, percibidos nuevamente con un visión europeizante, como lector de textos europeos y creador de discursos nativos; y por último, el sujeto colonial amerindio como productor de discursos históricos. Los sujetos cronistas siguen una visión común de la conducta femenina entre el deber ser y el deber estar en la sociedad de corte español. Esto dio lugar a que se reproducirán diferencias entre la imagen de la mujer española y la mujer indígena. Consecuencia de esta diferencia entre la mujer española y la americana, entre otras, será que la conducta esperada que tienen los conquistadores no será la que hallarán en América. Por lo tanto, como parte del proyecto colonizador algunos de los cronistas eclesiásticos querrán imponer sobre las mujeres indígenas patrones de comportamiento europeo. Se analiza la casi cancelación femenina de las crónicas para algunos autores como Cortés y la descripción minuciosa de otros cronistas como Lizárraga o Sahagún. En cuanto a la escritura femenina, aunque no está prevista en el corpus, se puede decir que considerando a Muriel (2000), Lavrin (1990) y otros autores, no constan documentos escrito por mujeres cronistas o al menos que el manuscrito haya sobrevivido a los siglos de principios del siglo XV, por lo tanto, hay una ausencia notable de textos de autoría femenina. Durante el período colonial comprendido por esta tesis que va hasta el siglo XVI, el corpus textual de autoría femenina es incipiente, solo se cuenta con cartas privadas, la carta de Isabel de Guevara a la Princesa Juana, unos textos de la Adelantada Mencía Calderón y como anteriormente dicho, algunos fragmentos de crónicas conventuales que cita Muriel (2000: 44)

    Phytoplankton in the Elbe estuary: New insights into community composition and mixotrophy, from metabarcoding, flow cytometry, and laboratory experiments

    Estuaries, the lower river areas that merge into the oceans, are centers of human lifes, home for a diverse wildlife and important in global carbon cycles. Phytoplankton are the basis of the food chains in estuaries but may suffer from harsh conditions here. For instance, turbidity can darken the water body, but light is needed for photosynthesis. Species that live under such conditions may play an important role because they provide food for zooplankton and take up CO2. However, there is not much knowledge about estuarine life. This is also the case in the Elbe estuary, one of Europe’s largest estuaries, where the lower ca. 70 km are largely unexplored. Even in explored areas of this habitat, certain groups of phytoplankton may have been overlooked so far due to the choice of methods. In our study, we applied molecular tools (metabarcoding) and automated cell counting techniques (flow cytometry) to assess phytoplankton composition in the Elbe estuary. Our focus was to identify important phytoplankton groups in the unmonitored mid to lower estuary and to compare our results from further upstream with available microscopy data. We also carried out laboratory experiments, to assess the ability of phytoplankton from the Elbe estuary to use organic substances (e.g. sugars), a strategy considered mixotrophy. In compliance with former knowledge, phytoplankton communities were overall dominated by diatoms. However, we found that the smallest players among the phytoplankton - the picophytoplankton - and potentially mixotrophic flagellates were more dominant than so far reported. Due to their fragile nature and small size, those groups were likely overlooked in former microscopy-based studies. In our laboratory study, we furthermore found that all included phytoplankton species were able to use organic substances. Our results altogether provide support for the importance of mixotrophy in the Elbe estuary

    Neuromotorische und biomechanische Assoziationen der Deformierbarkeit der Fascia thoracolumbalis

    Mit Faszie wurde ursprünglich ein bindegewebiges Band oder Blatt bezeichnet, das Organe oder Skelettmuskel umhüllt, separiert oder verbindet. In der muskuloskelettalen Medizin wurden die Faszien Jahrhunderte nachrangig betrachtet und werden erst mit Aufkommen der neueren Untersuchungsmethoden, wie dem hochauflösenden Ultraschall, der Bioimpedanzanalyse oder der Magnetresonanztomographie, intensiver erforscht. Seit Mitte der 2000er-Jahre ist eine deutlich steigende Anzahl an Veröffentlichungen über die Faszien und das Fasziensystem zu verzeichnen. In der Zwischenzeit sind grundlegende Erkenntnisse zu ihren Einflüssen auf die Proprio- und Nozizeption oder z. B. zu ihrer Beteiligung an Körperbewegungen vorgelegt worden. Faszien können Kräfte nicht nur lokal, sondern auch über Gelenke hinweg in entferntere Körperabschnitte übertragen. Aufgrund ihrer zahlreichen Innervation werden manche Faszien deshalb von einigen Autoren als eine Art Sinnesorgan beschrieben. Während das Interesse in zahlreichen medizinischen Wissenschaftsdisziplinen seit Jahren anhält, werden Faszien und das Fasziensystem in der Sportwissenschaft vergleichsweise wenig beachtet. Verletzungen in diesem Bereich führen sowohl im Freizeit- als auch im Leistungssport zu erheblichen Leistungseinbußen und können darüber hinaus einen Einfluss auf die Entwicklung und die Aufrechterhaltung muskuloskelettaler Erkrankungen haben. Die Fascia thoracolumbalis (TLF) stellt das zentrale Bindeglied zwischen paraspinaler Muskulatur, der Abdominal- und der oberen Rückenmuskulatur dar. Hier unterstützt sie den M. erector spinae (ES) durch intramuskuläre Drucksenkung bei der Aufrechterhaltung der spinalen Stabilität. Ziel dieser Thesis ist es, neuromotorische und biomechanische Assoziationen der Deformierbarkeit der TLF (TLFD) zu untersuchen. Hierzu wurden fünf Hauptfragestellungen in vier Studien untersucht, die im Rahmen einer kumulativen Dissertation publiziert wurden: (1) Neuromotorische Einflüsse der TLF auf den ES; (2) Assoziationen zwischen sportlicher Aktivität, Faszienmorphologie und der Mikrozirkulation in der TLF und dem ES; (3) Auswirkungen myofaszialer Releasetechniken an der TLF auf deren Mikrozirkulation; (4) Auswirkungen intensiver sportlicher Belastungen auf die TLF und Assoziationen zu verzögert einsetzendem Muskelschmerz (DOMS); (5) die TLF als leistungsbestimmender Faktor im Vergleich von Athlet*innen, Gesunden und Menschen mit akuten lumbalen Rückenschmerzen. Hierzu wurden u. a. hochauflösende dynamische Ultraschalluntersuchungen, die Weißlicht- und die Laser-Doppler-Spektroskopie sowie gyroskopische Bewegungs- und Geschwindigkeitsmessungen durchgeführt. Es zeigte sich, dass im Falle von akuten lumbalen Rückenschmerzen die TLFD die Muskelaktivität des ES zu 82 % granger-kausal vorhersagte, wohingegen ein solcher Effekt bei Gesunden nicht zu beobachten war (1). Weiterhin konnten fasziale Einflussfaktoren auf die Mikrozirkulation der TLF und des ES gefunden werden. In desorganisiertem Fasziengewebe war der Blutfluss gegenüber organisiertem Fasziengewebe um 70 % vermindert (2). Diese Mikrozirkulation stieg nach myofaszialer Releasetherapie um 48,7 % an (3). Es konnte eine Schwellung innerhalb der TLF mit einer Dickenzunahme um 23 % bei DOMS beobachtet werden. DOMS stieg dabei um 74,96 % an (4). Die TLFD korrelierte zudem stark negativ mit der Geschwindigkeit beim Kreuzheben (5), d. h., je deformierbarer die TLF war, neben einem entsprechenden Trainingszustand, desto höhere Geschwindigkeiten konnten beim Kreuzheben erzielt werden. Die Ergebnisse der Untersuchungen der Assoziationen der TLF in dieser Thesis weisen darauf hin, dass die TLF einen entscheidenden Faktor für die Kraftentwicklung des ES darstellt. Mechanismen dahinter könnten eine physiologische intramuskuläre Druckminderung in der paraspinalen Muskulatur durch die Unterstützung der TLF sein, was dem Muskel erlaubt, eine höhere Kraft zu entwickeln. Das macht die TLF auch unter sportwissenschaftlichen Gesichtspunkten zu einer vielversprechenden Struktur, deren Untersuchung hinsichtlich ihres weiteren spezifischen Einflusses in weiterführenden Arbeiten lohnenswert erscheint

    Evaluation und Optimisierung von Nukleosidtriphosphatanalogen als antivirale Prodrugs

    Over the past 40 years, human immunodeficiency virus (HIV) research has evolved from a quest to understand the virus to a multifaceted endeavour aimed at developing effective treatments and prevention strategies. This thesis highlights key milestones in this journey and focuses on recent developments concerning nucleoside reverse transcriptase inhibitors (NRTIs). One such approach lies in the development of robust and flexible prodrug systems that can target not only HIV but also other diseases, like SARS-CoV-2 or Alzheimer’s’ disease. The prodrug strategy was pioneered by MEIER et al., culminating in the development of triphosphate prodrugs (TriPPPro) and -alkyl triphosphate prodrugs. These prodrugs offer a versatile platform for drug delivery, enabling the adaptation of existing NRTIs to multiple pathological targets. This work established methods to evaluate the efficacy and safety of these prodrugs through a responsive workflow that used data acquired from primer extension assays and testing in HIV-infected cell lines to infer new designs for prodrug delivery to yield better overall pharmacological profiles. As a result of the acquired data, four highly active NRTIs were transformed into their prodrugs, alkyl triphosphates and triphosphates. It was found that adjustments according to the biological data led to significant improvements in the pharmacological profiles of thymidine derivatives.In den letzten 40 Jahren hat sich die Forschung zum humanen Immundefizienzvirus (HIV) von einem schlichten Bestreben, das Virus zu verstehen, zu einer vielschichtigen Aufgabe entwickelt. Dabei liegt der Fokus auf der Entwicklung wirksamer Behandlungen und Präventionsstrategien. Diese Arbeit beleuchtet wichtige Meilensteine dieses Weges und konzentriert sich auf die jüngsten Entwicklungen im Bereich Nucleosid Reverse Transkriptase Inhibitoren (NRTIs). Ein solcher Ansatz liegt in der Entwicklung robuster und flexibler Prodrug-Systeme, die nicht nur gegen HIV, sondern auch gegen andere Krankheiten wie SARS-CoV-2 oder Alzheimer wirksam sein können. Dieser Ansatz wurde von MEIER et al. entwickelt und fand seinen Höhepunkt in der Entwicklung von Triphosphat-Prodrugs (TriPPPro) und -Alkyltriphosphat-Prodrugs, welche eine vielseitige Plattform für die Wirkstofffreisetzung bieten und die Anpassung vorhandener NRTIs an verschiedene pathologische Ziele ermöglichen. Diese Arbeit zielt darauf ab, Methoden zur Bewertung der Wirksamkeit und Sicherheit dieser Prodrugs zu erforschen und durch einen reaktionsfähigen Arbeitsablauf die gewonnenen Informationen zu nutzen, um neue Ansätze für die Prodrug-Freisetzung abzuleiten und so insgesamt bessere pharmakologische Profile zu erzielen. Die Kombination hochaktiver NRTIs mit dem anpassungsfähigen Nucleotid-Prodrug-System könnte das Interesse an neuartigen NRTI-Prodrugs gegen HIV erneuern. Im Rahmen dieser Arbeit wurden Methoden zur Bewertung der Wirksamkeit und Sicherheit dieser Prodrugs durch einen anpassungsfähigen Arbeitsablauf entwickelt, bei dem Daten aus Primer-Verlängerungsstudien und Tests in HIV-infizierten Zelllinien verwendet wurden. Daraus folgend sollten neue Designs für Prodrug-Verabreichung abgeleitet werden, die ein besseres pharmakologisches Gesamtprofil zur Folge haben. Als Ergebnis der gewonnenen Daten wurden vier hochaktive NRTIs in ihre Prodrugs, -Alkyltriphosphate und Triphosphate, umgewandelt. Es zeigte sich, dass die Anpassung dieser Systeme auf Grund biologischer Daten zu erheblich Verbesserungen der pharmakologischen Profile der Thymidin-Derivate führte

    Dynamical mean-field theory for solids with strong coupling to the electromagnetic field

    Recent advances in the study of quantum cooperative effects in coupled light-matter systems open up unprecedented pathways to manipulate the properties of materials without external drive. This is due to vacuum fluctuations of the electromagnetic field, which hybridize with matter degrees of freedom even in thermal equilibrium and thereby influence the behavior of the system. In this thesis, we focus on the theoretical study of such photon-induced effects. We first present a collective theory for single-mode models and then apply dynamical mean-field theory (DMFT) to macroscopic solids interacting with a continuum of electromagnetic modes. The collective theory is based on diagrammatic techniques and assumes a linear dipolar coupling to one single cavity mode. It allows expressing the electric susceptibility of the system inside the cavity in terms of the bare matter response. We find that the radiative corrections of the static susceptibility vanish in the thermodynamic limit if the single-particle coupling is finite. Moreover, the formalism proves that nonlinearities in the matter response play a crucial role in affecting the equilibrium state of finite-size systems. As an example, we apply the theory to a simple model of a quantum paraelectric with dipole-dipole interactions and demonstrate that the cavity mode leads to an enhancement of the static electric response in small clusters of material. DMFT, however, is most appropriate for extended solids consisting of a macroscopic number of atoms or molecules. Therefore, we consider a setting, where a continuum of electromagnetic modes gives rise to a non-vanishing total effect even in the thermodynamic limit. The modes correspond to surface plasmon polaritons (SPPs) at a dielectric-metal interface. We study two different model systems: In the first case, we consider a two-dimensional solid that couples to the vacuum fluctuations of the SPPs via a linear dipolar interaction. Within static mean-field approximation, the material exhibits a ferroelectric phase transition that is not affected by the electromagnetic radiation field. Bosonic DMFT provides a more accurate description and reveals that the light-matter interaction enhances the ferroelectric order and stabilizes the ferroelectric phase. In the second case, we study a two-dimensional Hubbard model, which couples to the electromagnetic modes via Peierls phase factors. Even without light-matter interaction, the system may undergo a Mott metal-isulator transition. We follow a diagrammatic approach to incorporate photon-induced effects into the DMFT formalism. Our results suggest that the coupling to the electromagnetic field favors the metallic state over the Mott insulating phase. In summary, this thesis demonstrates that the interplay of light and matter opens up new possibilities to modify the static response of microscopic systems and to control phase transitions in macroscopic solids, even in thermal equilibrium. Moreover, it highlights that DMFT can serve as a valuable theoretical tool to study quantum cooperative effects in systems with strong light-matter interactions

    Artificial Intelligence in Organizations: Managing the Lifecycle of Conversational Agents

    Given the rapid advances of information technology, led by increasing dissemination and possibilities, many areas of science, society and the economy are being impacted upon. In this context, artificial intelligence (AI) has developed into a disruptive technology. Driven by the large availability of data, increased computing power, and algorithmic and technological advances, AI is a decisive factor in the digital transformation of organizations. As a result, companies are spendingsignificant resources on AI-based systems and related applications, e.g., to improve customer engagement and remain competitive. Conversational Agents (CAs), such as chatbots, are an emerging natural language-based application of AI, which provide organizations with an increasingly intelligent, social, learning, integrative, and cost-effective platform to support and automate the delivery of services and information. CAs offer new forms of scalability and availability (24/7), speed, and personalization. However, despite their possibilities, organizations are struggling to realize the full potential of CAs in real-world environments. AI-based CAs pose new challenges due to their unique characteristics. They are not sufficiently represented in current research, which leads to many open questions and opportunities regarding the management of CAs’ lifecycle, especially in regard to different activities in the introduction, design, implementation, operation, and improvement phases. From a broader, organization-wide perspective, the management of AI is an essential task inrealizing the new value propositions. However, knowledge on how to structure AI transformations and effectively manage specific applications is limited, leading to a high failure rate of AI initiatives. There is a need for research to support organizations in systematically developing AI (implementation) competencies in order to master all the complexity of this new technology. By switching viewpoints between organization-wide and system-specific perspectives, this dissertation contributes to the emerging field of (1) AI in organizations in general by providing guidance for navigating, managing, and (re)evaluating AI transformations, and (2) CAs in particular, by providing various forms of validated design knowledge for their lifecycle management. Together, these themes facilitate implications for a broader understanding of AI management in organizational settings

    Climate change impacts: In the perspective of soil organic carbon, above-ground biomass and vegetation shifts (A case study from Nepal)

    Climate change impact has been examined with regard to different aspects of forest ecosystems such as soil organic carbon (SOC), above-ground tree biomass (AGTB) and vegetation shift (broadleaved and coniferous forests). The SOC and AGTB were assessed in terms of their change in amount with future climate change whereas vegetation shift was assessed in terms of shifting altitudinal ranges, natural habitat area and species in future climate change. Nepal was selected as a test region to study the impact of climate change on these aspects due to the large differences in altitude and temperature in the country and also it is one of the most vulnerable countries to climate change. The objectives of this study were, i.e. 1) Are climatic variables important for the estimation of SOC and AGTB? 2) Does future climate change contribute to an increase in SOC and AGTB stock in the forest ecosystem? 3) Does climate change affect the vegetation shift? Multiple linear regression, Random forest and Maximum Entropy (MaxEnt) models were applied to investigate the objectives. The availability of forest resource assessment (2010-2014) data of Nepal, accompanied by topographic and bioclimatic variables, provided the opportunity to study the impacts of climate change on forest ecosystems at a national scale. Climatic variables (temperature and precipitation) show a strong relation with SOC and AGTB. The climatic variables do not only explain SOC and AGTB at present, but they can also predict SOC and AGTB under future climate change scenarios. This study found a higher amount of SOC existed at higher altitudes compared to lower altitudes and the rate of accumulation of SOC increasing at a higher rate with the increase in altitude. The lower latitude has a higher temperature and vice versa. In the projected climate change scenario, i.e. CMIP6, SSP2 4.5 for 2040-2060, the amount of SOC was found to decrease by 3.85% in general with the increasing temperature and precipitation against near current period (1970-2000). In Contrast, the study indicated a positive relation between climatic variables (temperature and precipitation) and AGTB that found the amount of AGTB to increase by 2.96% in general in the same projected climate change scenario against same near current period. Moreover, vegetation shifts from one forest to another are likely to occur over a longer period, influenced by climatic variables. This study found the vegetation shift in terms of areas, i.e. coniferous to broad-leaved forests (1579 km2) and its reverse (232 km2), in terms of altitudinal shift, i.e. 77m higher for broad-leaved forests and 54m lower for coniferous forests, and in terms of species, i.e. broad-leaved forests to coniferous forests and its reverse in the future climate change scenario. The result confirms that a higher amount of SOC stock exists in the forests at a higher altitude compared to the forests at lower altitude. However, the amount of SOC is likely to decrease while the amount of AGTB is likely to increase in the future climate change scenarios. Moreover, the result shows that vegetation shift from coniferous to broad-leaved forest is more dominant than the broad-leaved to coniferous forests and the area of broad-leaved forest will likely to expand while the area of coniferous forest is likely to shrink in the future climate change scenario. Therefore, this study highlights the need to retain SOC amount thus reducing carbon emission from the soil. It also highlights the significance of, particularly, high altitude forest in sequestrating atmospheric carbon in the future climate change scenario. Moreover, the study highlights that the expansion of broad-leaved forests due to vegetation shift may benefit in terms of species diversity, SOC amount and forest resilience and also may affect coniferous forest- dependent people and enterprise due to lower supply of the forest products. Thus, the study suggests to adopt sustainable management of high altitude forests to increase mitigation potential of the forests (increase carbon sequestration and reduce carbon emission) and also suggests to assess adaptation measures for vulnerable communities due to climate change
