Rotationally stable screw anchor (RoSA) vs. Gamma3 Nail (G3N) in pertrochanteric femoral fractures: A functional outcome analysis
Diese Publikationspromotion fokussiert sich auf die Untersuchung des funktionellen Outcomes ≥ ein Jahr nach PFF. In diese Studie wurden insgesamt 43 Patienten inkludiert. 20 Patienten wurden mit einem RoSA und bei Bedarf additiver Trochanterabstützplatte (TAP) in der RoMed Klinik, Bad Aibling versorgt. 23 Patienten erhielten eine intramedulläre Osteosynthese in der Asklepios Klinik St. Georg. Mittels zwei funktioneller Scores (SF-36, New Mobility Score) wurde das funktionelle Outcome erfasst und analysiert. Zudem wurden intraoperative, sowie postoperative Komplikationen binnen 1 Jahr dokumentiert. Hinsichtlich des funktionellen Outcomes zeigte sich eine zahlenmäßige, leichte, dennoch nicht signifikante Überlegenheit des G3N. Die Komplikationsrate lag für beide Implantate (G3N 9%, RoSA 10%) im Bereich der, in der Literatur dokumentierten. Im RoSA Kollektiv kam es in 2 Fällen zu einem Ausbruch der Platte am Femurschaft. Ein cut-out wurde in keinem Fall dokumentiert. In der Literatur ist die größte Gruppe an Komplikationen, die der implantatassoziierten Komplikation. Dartunter stellt das cut-out im Bereich des Femurkopfes eine der häufigsten Komplikationen dar. Das Prinzip des RoSA beruht auf der theoretisch stabileren Verankerung der Schraub-Klingen-Kombination im Kopf-Hals Fragment. Die biomechanische Herausforderung des RoSA liegt in der Verankerung des Implantats am Femurschaft, während der G3N diese im Sinne eines Rohr-in-Rohr Prinzips aufnimmt. Die Weiterentwicklung des RoSA zu einem intramedullären Implantat, mag die biomechanischen Vorteile der Implantate vereinen und zu einer weiteren Reduktion der implantatassoziierten Komplikationen führen. Zusammenfassend zeigen beide Kollektive hinsichtlich des funktionellen Outcomes keine signifikanten Unterschiede. Bei instabilen PFF ist entsprechend unserer Ergebnisse dennoch ein intramedulläres Implantat von Vorteil
Sex- and species-specific CD99 expression of T cells and its implications in multiple sclerosis
Differences in immune responses between women and men are leading to a strong sex bias in the incidence of autoimmune diseases that predominantly affect women, such as MS. MS manifests in more than twice as many women, making sex one of the most important risk factors. However, it is incompletely understood which genes contribute to sex differences in autoimmune incidence. To address that, we conducted a gene expression analysis in female and male human spleen using the GTEx project dataset to identify differentially expressed genes between women and men. We identified the transmembrane protein CD99 as one of the most significantly differentially expressed genes with marked increase in men. CD99 has been reported to participate in immune cell transmigration and T cell regulation, but sex-specific implications have not been comprehensively investigated.
Here, we found higher CD99 gene expression in male human spleens compared to females and confirmed this expression difference on protein level on the surface of T cells and pDCs. Hormones were ruled out as the cause by in vitro assays and ex vivo analysis of trans men samples. In CSF, CD99 was higher on T cells compared to blood. Of note, male MS patients had lower CD99 levels on CD4+ T cells in the CSF, unlike controls. By contrast, female and male mice had similar CD99 expression and Cd99-deficient mice showed equal susceptibility to EAE compared to WTs. Functionally, CD99 increased upon human T cell activation and inhibited T cell proliferation after blockade. Accordingly, CD99-deficient Jurkat cells showed decreased cell proliferation and cluster formation, rescued by CD99 reintroduction.
Our results demonstrate that CD99 is sex-specifically regulated in healthy individuals and MS patients and that it is involved in T cell costimulation in humans but not in mice. CD99 could potentially contribute to MS incidence and susceptibility in a sex-specific manner
Erkennen von Lernbarrieren in Textvignetten - ErLe - Entwicklung und Validierung eines quantitativen Verfahrens zur Beurteilung situationsspezifischer Fähigkeiten
Theoretischer Hintergrund: Die professionelle Wahrnehmung unterrichtsrelevanter Situationen gilt als Schlüsselkompetenz von Lehrkräften. Aus sonderpädagogischer Perspektive sind insbesondere solche Situationen relevant, in denen es zu Störungen des Lehr-Lern-Prozesses kommt, in denen also Lernbarrieren entstehen könnten. Potenzielle Lernbarrieren zu erkennen, wird damit zu einer wesentlichen Aufgabe von Lehrkräften.
Die Analyse von Lehr-Lern-Situationen muss über die Beobachtung individueller Bedingungen, die zu Lernbarrieren beitragen können, hinausgehen und auch alle anderen Bedingungen in den Blick nehmen (Langner & Jugel, 2019; Ricken, 2017; Vock & Gronastaj, 2017). Merkmale der Klassenführung und Lernunterstützung gelten dabei als besonders relevant (Holodynski et al., 2017). Die Beschreibung und Erfassung der Kompetenz Lernbarrieren zu erkennen, die in der fehlenden oder mangelhaften Umsetzung von Klassenführungs- und Lernunterstützungsmerkmalen liegen, fehlen aber bisher als Ansatz.
Vorgehen: In der vorliegenden Arbeit werden ausgehend von der Unterrichtsqualitätsforschung Merkmale der Klassenführung und Lernunterstützung herausgearbeitet. Deren fehlende oder fehlerhafte Umsetzungen werden als potenzielle Lernbarrieren verstanden und operationalisiert. Die Kompetenz, diese zu erkennen, wird auf der Basis des Kompetenzmodells von Blömeke et al. (2015a) als situationsspezifische Fähigkeit beschrieben und betont u. a. die bewusste Fokussierung und Wahrnehmung von Unterrichtsbedingungen und die Beurteilung hinsichtlich der förderlichen oder hinderlichen Auswirkung auf die Lehr-Lernsituation.
In vier Entwicklungsschritten wird ein Verfahren entwickelt, bei dem anhand der Beurteilung von Textvignetten die Kompetenz zum Erkennen von Lernbarrieren erfassbar werden soll. Textvignetten und Items werden theoriegeleitet konstruiert, erprobt und auf der Basis der gewonnenen Daten sowie mehrerer Expertendiskussionen variiert und weiterentwickelt. Die Stichproben der vier Entwicklungsschritte umfassen insgesamt 935 Lehrkräfte und Studierende verschiedener Lehramtsstudiengänge aus verschiedenen Fachsemestern. Daten zu den Beurteilungen der Vignetten werden deskriptiv und anhand non-parametrischer Verfahren ausgewertet.
Ergebnisse: Im ersten Entwicklungsschritt wird erprobt, wie spezifisch Impulse durch die Vignette und das Beurteilungsformat sein müssen, damit Beurteilungsprozesse hin-sichtlich der Klassenführung und Lernunterstützung in angemessenem Schwierigkeitsgrad in Gang gesetzt werden. Dazu werden zwei unterschiedliche Vignettenentwürfe mit offenen und geschlossenen Beurteilungsimpulsen eingesetzt. In den Ergebnissen zeigt sich, dass weniger spezifische offene Impulse nicht dazu führen, dass Studierende Unterrichtsbedingungen fokussieren, sondern vor allem Bedingungen des Kindes als Ursache für problematische Lehr-Lern-Situationen in Betracht ziehen. In der weiteren Entwicklung werden deshalb Vignetten eingesetzt, in denen konkrete Unterrichtssituationen beschrieben werden, die Lernbarrieren in der Klassenführung und Lernunterstützung enthalten, und die anhand von Items zur Klassenführung und Lernunterstützung zu beurteilen sind. Für diese Variante zeigt sich im ersten Entwicklungsschritt eine akzeptable Reliabilität und es deuten sich Gruppenunterschiede zugunsten der Sonderpädagogikstudierenden an, wobei Schwierigkeitsgrade noch nicht angemessen sind.
Im zweiten Entwicklungsschritt werden Vignette und Items inhaltlich, sprachlich und methodisch überarbeitet und erprobt. Die Ergebnisse liefern Hinweise darauf, dass vor allem Kontextinformationen zur Vignette einen starken Einfluss auf die Beurteilung haben und die beschriebene Unterrichtssituation eher holistisch als differenziert beurteilt wird. Gruppenunterschiede zugunsten höherer Fachsemester zeigen sich zwischen Studieren-den des 2. und 6. Bachelorsemesters.
Der dritte Entwicklungsschritt hat zum Ziel, Schwierigkeitsgrade über die Darstellung der Merkmale in den Vignetten zu beeinflussen und differenziertere Beurteilungen anzuregen. Dies wird umgesetzt, indem auf der Basis realer Unterrichtsbeispiele vier neue Vignetten bei gleichbleibenden Konstruktionskriterien entwickelt und validiert werden. Neben Lernbarrieren werden auch gelingende Situationen in den Vignetten dargestellt. In den Ergebnissen zeigen sich zufriedenstellende mittlere Schwierigkeitsgrade und zufriedenstellende Reliabilitäten für drei der vier Vignetten. Die Vignetten selbst haben einen starken Einfluss auf die Beurteilungen und werden eher nach Gesamteindruck als differenziert beurteilt. Gruppenunterschiede zeigen sich nur im Vergleich der Lehrkräfte mit Studierenden, Lehrkräfte beurteilen Lernbarrieren zutreffender, gelingende Situationen aber weniger zutreffend als Studierende.
Der vierte Entwicklungsschritt hat zum Ziel, noch stärker eine Differenzierung zu betonen und Einflüsse auf die Vignettenbeurteilung zu überprüfen. Dazu werden die Vignetten nochmals überarbeitet und validiert, gelingende Situationen und Lernbarrieren werden in insgesamt ausgewogener Anzahl dargestellt, innerhalb der Vignetten aber in der Anzahl variiert. Die Gesamtwahrnehmung der Klassenatmosphäre wird zusätzlich erhoben. Außerdem wird in einer Teilstichprobe eine Variation der Instruktion erprobt. Die Ergebnisse zeigen, dass mittlere Schwierigkeitsgrade und Varianzen zufriedenstellend sind, Items zu Barrieren und gelingenden Situationen aber nicht differenziert beurteilt werden und getrennte Skalen bilden. Die Vignetten werden in allen Gruppen eher holistisch beurteilt – Lehrkräfte urteilen insgesamt eher negativ und erzielen so höhere Werte für Barrieren. Studierende der Allgemeinpädagogik urteilen eher positiv und erzielen deshalb höhere Werte für Gelingendes. Während die Urteile der Lehrkräfte weitgehend unabhängig von der Einschätzung der Klassenatmosphäre und der Vignettenkonstruktion sind, zeigen sich vor allem bei Bachelorstudierenden starke Zusammenhänge der Beurteilungen mit der Einschätzung der Klassenatmosphäre. Ein relevanter Zusammenhang mit der Variation der Instruktion zeigt sich nicht.
Fazit: Das entwickelte textvignettenbasierte Verfahren eignet sich, um unterschiedliche Beurteilungen von Lernbarrieren und von gelingenden Situationen reliabel zu erfassen und Gruppenunterschiede abzubilden. Die Expertise, Gelingendes und Misslingendes differenziert zu beurteilen und das gleichbleibend gut in allen vier Vignetten, zeigt sich in keiner der Teilstichproben. Eine solche Expertise scheint sich ohne spezifische Lerngelegenheit nicht zu entwickeln. Beurteilungen sind in allen Gruppen eher holistisch und ab-hängig von Gesamteindrücken und unterschiedlichen schwerpunktmäßigen Fokussierungen auf misslingende oder gelingende Prozesse. Die Vignetten selbst haben unabhängig von den gezielten Konstruktionskriterien und -variationen einen starken Einfluss auf die Beurteilungen.Theoretical background: The professional perception of teaching-relevant situations is considered a key competence of teachers. From a special education perspective, situations are particularly relevant in which disruptions to the teaching-learning process occur and in which learning barriers could arise. Recognizing potential barriers to learning therefore becomes an essential task for teachers.
The analysis of teaching-learning situations must go beyond the observation of individual conditions that can contribute to learning barriers and also take all other conditions into account (Langner & Jugel, 2019; Ricken, 2017; Vock & Gronastaj, 2017). Features of classroom management and learning support are considered particularly relevant (Holodynski et al., 2017). However, the description and recording of the competence to recognize learning barriers that lie in the absence or inadequate implementation of classroom management and learning support features is still missing as an approach.
Approach: In this work, characteristics of classroom management and learning support are identified based on teaching quality research. Their missing or incorrect implementations are understood and operationalized as potential learning barriers. The competence to recognize this is based on the competence model by Blömeke et al. (2015a) described as a situation-specific ability and emphasizes, among other things, the conscious focus and perception of teaching conditions and the assessment of the beneficial or hindering impact on the teaching-learning situation.
In four development steps, a process is being developed in which the competence to recognize learning barriers can be determined by assessing text vignettes. Text vignettes and items are constructed based on theory, tested and varied and further developed on the basis of the data obtained and several expert discussions. The samples from the four development steps were taken from a total of 935 teachers and students during various teacher training courses from different semesters. Data on the assessments of the vignettes are evaluated descriptively and using non-parametric methods.
Results: In the first development step, it is tested how specific impulses must be provided by the vignette and the assessment format so that assessment processes with regard to class management and learning support are initiated with an appropriate level of difficulty. For this purpose, two different vignette designs with open and closed assessment impulses are used. The results show that less specific open impulses do not lead to students focusing on teaching conditions, but rather considering the child's conditions as the cause of the problematic teaching-learning situation. In further development, vignettes will therefore be used in which concrete teaching situations are described, which contain learning barriers in class management and learning support, and which can be assessed using items on class management and learning support. In the first development step, this variant shows acceptable reliability and there are group differences in favor of special education students, although levels of difficulty are not yet appropriate.
In the second development step, the vignette and items are revised and tested in terms of content, language and methodology. The results provide evidence that contextual information about the vignette has a strong influence on the assessment and that the teaching situation described is assessed more holistically than differentiated. Group differences in favor of higher semesters can be seen between students in the 2nd and 6th Bachelor semesters.
The third development step aims to influence levels of difficulty through the representation of the characteristics in the vignettes and to encourage more differentiated assessments. This is implemented by developing and validating four new vignettes based on real teaching examples while maintaining the same design criteria. In addition to learning barriers, successful situations are also presented in the vignettes. The results show satisfactory average levels of difficulty and satisfactory reliabilities for three of the four vignettes. The vignettes themselves have a strong influence on the assessments and are judged more on the basis of an overall impression than on a differentiated basis. Group differences only become apparent when comparing teachers with students; teachers judge learning barriers more accurately, but successful situations less accurately than students.
The fourth development step aims to emphasize differentiation even more and to examine influences on the vignette assessment. For this purpose, the vignettes are revised and validated again, successful situations and learning barriers are presented in a balanced number overall, but the number varies within the vignettes, and the overall perception of the class atmosphere is recorded. In addition, a variation of the instructions is tested in a subsample. The results show that medium levels of difficulty and variance are satisfactory, but items about barriers and successful situations are not assessed differentiated and form separate scales. The vignettes are assessed more holistically in all groups - teachers judge them more negatively overall and thus achieve higher scores for barriers. General education students are more likely to judge things positively and therefore achieve higher scores for success. While the teachers' judgments are largely independent of the assessment of the class atmosphere and the vignette construction,
there are strong connections between the assessments and the assessment of the class atmosphere, especially among bachelor's students. There is no relevant connection with the variation of the instruction.
Conclusion: The text vignette-based method developed is suitable for reliably recording different assessments of learning barriers and successful situations and for depicting group differences. The expertise in assessing successful situations and learning barriers in a differentiated manner and doing so consistently well in all four vignettes is not evident in any of the sub-samples. Such expertise does not appear to develop without specific learning opportunities. Assessments in all groups are more holistic and dependent on overall impressions and different focuses on failing or successful processes. The vignettes themselves have a strong influence on the assessments, regardless of the targeted design criteria
Investigating Nanostructured Materials by Multi-modal in situ X-ray Methods
Understanding the structural transformations affecting nanomaterials is essential to rationally improve their properties and functionalities. Despite recent advances in several X-ray investigation routines, the transformation phenomena affecting nanomaterials entail structural modifications on several length scales that cannot be covered by a single analytical method. In this thesis, we establish a multi-modal X-ray scattering setup to collect structural information spanning from fractions of Å to hundreds of nanometers via simultaneous acquisition of small-angle X-ray scattering (SAXS) and X-ray total scattering (TS). We then apply the method to investigate the model syntheses of Pd, Cu, and CuPd nanocrystals in solution, and to further elucidate the degradation of photo- electrochemical (PEC) CuBi2O4 thin films during operation.
We reveal that a Pd(II)-amino initial complex directly converts into Pd nanocrystals without formation of intermediates, in agreement with the classical nucleation model. We employ the pair-distribution function (PDF) analysis of the in situ TS signal and complementary in situ X-ray absorption spectroscopy (XAS) during the synthesis of Cu nanocrystals to reveal a multi-step conversion of an initial Cu(II)-amino complex into a partially reduced Cu(I) organometallic intermediate prior to the nucleation of polycrystalline Cu2O seeds. We further demonstrate via PDF and SAXS that individual Cu2O seeds fuse together to form larger Cu2O nanocrystals, which slowly convert into metallic Cu. We thus show that the synthesis of Cu nanocrystals follows a complex non-classical crystallization pathway.
Both in situ XAS and PDF suggest that alloyed CuPd nanocrystals form via preliminary formation of Pd-rich seeds, which promote the direct reduction of the Cu(I)-amino intermediate into the metallic form without formation of the Cu2O phase. PDF analysis further indicates that the CuPd nanocrystals nucleate and grow as multi-twinned icosahedra. Furthermore, SAXS data reveals that the as-formed CuPd icosahedra assemble within the reaction medium into large face-centered cubic (fcc) supercrystals due to the entropic forces arising by the ordering of the solvent molecules into lamellae.
We reveal by both PDF and SAXS that the fast decrease in the performances of CuBi2O4 electrodes correlates directly with the formation of a metallic Bi phase. We also identify additional degradation phenomena, such as the emergence of metallic Cu and the dissolution of the electrode in contact with the electrolyte, which further affect the CuBi2O4 thin film activity and morphology. Overall, our study shows that the multi- modal acquisition of PDF, SAXS, and XAS significantly enhances our understanding on the transformation mechanisms affecting nanomaterials and poses the methodological basis to investigate the fabrication and operation of a wide range of nanostructured materials.Das Verständnis der strukturellen Veränderungen von Nanomaterialien ist von entscheidender Bedeutung für die Verbesserung ihrer Eigenschaften und Funktionalitäten. Trotz der jüngsten Fortschritte in verschiedenen Röntgenmethoden können strukturelle Veränderungen auf mehreren Längenskalen nicht mit einer einzigen analytischen Methode erfasst werden. In dieser Arbeit etablieren wir einen multimodalen Röntgenstreuungsaufbau, um strukturelle Informationen von Bruchteilen von Å bis zu Hunderten Nanometern durch das simultane Messen von Röntgenkleinwinkelstreuung (Small Angle X-ray Scattering, SAXS) und Röntgentotalstreuung (Total Scattering, TS) zu erhalten. Wir verwenden den multimodalen Ansatz, um die Synthese von Pd-, Cu- und CuPd-Nanokristallen in Lösung sowie die Zersetzung von photoelektrochemischen (PEC) CuBi2O4-Dünnschichten in-situ bzw. operando zu untersuchen.
Wir zeigen, dass ein initialer Pd(II)-Aminkomplex direkt, ohne Zwischenprodukt, in Pd-Nanokristalle umwandelt wird. Wir nutzen die Paarverteilungsfunktion (Pair Distribution Function, PDF) des TS-Signals mit komplementärer Röntgenabsorptionsspektroskopie (X-ray Absorption Spectroscopy, XAS), um die mehrstufige Umwandlung eines initialen Cu(II)-Aminkomplexes in eine teilweise reduzierte Cu(I)-Aminspezies vor der Nukleationskeimbildung von polykristallinen Cu2O-Keimen aufzuzeigen. Des Weiteren demonstrieren wir mittels PDF und SAXS, dass einzelne Cu2O-Keime miteinander verschmelzen und größere Cu2O-Nanokristalle bilden, die sich langsam in metallisches Cu umwandeln. Folglich zeigen wir, dass die Synthese von Cu-Nanokristallen einem komplexen, nicht-klassischen Kristallisationsweg folgt.
Sowohl die in-situ XAS- als auch die in-situ PDF-Analyse deuten darauf hin, dass sich legierte CuPd-Nanokristalle durch die vorherige Bildung von Pd-reichen Keimen bilden, die die direkte Reduktion des Cu(I)-Amin-Zwischenprodukts in die metallische Form ohne Bildung der Cu2O-Phase fördern. Die PDF-Analyse deutet außerdem darauf hin, dass die Nanokristalle als mehrfach verzwillingte Ikosaeder keimen und wachsen. Darüber hinaus zeigen SAXS-Daten, dass sich die gebildeten CuPd-Ikosaeder im Reaktionsmedium aufgrund der entropischen Kräfte durch die Anordnung der Lösungsmittelmoleküle in Lamellen, zu großen kubisch flächenzentrierten Superkristallen zusammensetzen.
Wir zeigen mit PDF sowie mit SAXS, dass der schnelle Leistungsabfall von CuBi2O4- Elektroden direkt mit der Bildung einer metallischen Bi-Phase korreliert. Wir identifizieren zusätzliche Degradationsphänomene, wie die Entstehung von metallischem Cu und die Auflösung der Elektrode im Kontakt mit dem Elektrolyten, welche die CuBi2O4- Dünnschichtaktivität und Morphologie weiter beeinträchtigen. Insgesamt zeigt unsere Studie, dass die multimodale Erfassung von PDF, SAXS und XAS unser Verständnis der Umwandlungsmechanismen von Nanomaterialien erheblich verbessert. Unser Ansatz stellt die methodische Grundlage für die Untersuchung der Herstellung sowie der Funktionsweise eines breiten Spektrums von nanostrukturierten Materialien
The role of conventional dendritic cell subsets in T-cell activation in Ebola virus infection
Severe Ebola virus disease (EVD) is characterized by excess, dysregulated T cell activation and high levels of inflammation. In conflict with these clinical observations, in vitro studies described that Ebola virus (EBOV) infection of monocyte-derived dendritic cells (moDCs), results in suppression of T cell activation due to inhibition of DC activation. However, it is unknown how other DC subsets distinct from moDCs respond to EBOV infection. Therefore, we assessed the response of FMS-like tyrosine kinase 3 (Flt3)-dependent, conventional mouse DCs (cDCs) to EBOV infection, and studied their CD8 T-cell activation capacity in a DC-T cell co-culture system utilizing a recombinant EBOV expressing the model antigen ovalbumin.
Our data indicate that EBOV infection of mouse cDCs, results in activation rather than inhibition, promotes antigen cross-presentation and thus, leads to high levels of CD8 T cell activation. With that, we propose a mechanistic explanation for the excess T cell activation observed in severe human EVD
Effects of the Chemical Environment on the Optical Properties of Semiconductor Nanostructures
Semiconductor nanostructures have an enormous potential to revolutionize the technology for electronic and optoelectronic devices. In recent decades, the chemical synthesis of semiconductor nanostructures, which is essential for their use on a large scale, has developed rapidly. As a result, the implementation of nanostructures in applications, such as optoelectronic devices, is now increasingly becoming the focus of research. Part of this research is to understand and control the properties of nanostructures in their entirety to unlock their full potential. Critical to the successful use of nanostructures in devices is a comprehensive understanding of how their properties are affected by their environment.
This thesis examines the influence of the chemical environment on the optical properties, particularly the photoluminescence, of semiconductor nanostructures. This question is not only interesting for applications in sensor technology, but also represents a part of the necessary basic knowledge that is of central importance for the use of nanostructures in general. Two types of nanostructures, nanowires and quantum dots, were analyzed using confocal spectroscopy.
Colloidal CdSe nanowires (or quantum wires) were prepared wetchemically. To explore the influence of the chemical environment on the optical properties, measurements were performed in flow channels and gas-flow setups. In particular, the influence of oxygen on the photoluminescence and the Raman scattering was investigated. Both irreversible and reversible effects were observed and analyzed. Based on the findings, models were constructed to identify and adequately describe the effects. This led to the elucidation of photoinduced processes, such as the activation of photoluminescence and other changes in the emission and Raman spectra. The irreversible photoinduced activation of photoluminescence could be attributed to the formation of oxides and the concomitant elimination of surface defects in the form of unpassivated selenium. The reversible changes in the spectra, observed both in the form of additional blue-shifted emission and shifts in the Raman bands, could be attributed to the action of oxygen as a reversible electron acceptor.
In addition, the influence of oxygen on the photoluminescence properties of quantum dots was investigated. Here, the focus was on the reversible processes and in particular the blinking of the photoluminescence was investigated. A detailed analysis of the experimental data and the comparison with simulated data revealed multiple different effects of oxygen on the photoluminescence of quantum dots. The studies showed that oxygen acts as an electron acceptor, neutralizing particles and thus stabilizing the photoluminescence of quantum dots excited at high excitation powers. On the other hand, the effect of oxygen on the photoluminescence blinking of quantum dots with thin shells suggests that oxygen can also act as a centre for non-radiative recombination
Unravelling climate conditions preceding the End-Permian Mass Extinction: Insights from latest Permian interannual tropical variability and extreme events
In the present, anthropogenic greenhouse gas emissions are dramatically changing the climate and threatening ecosystems. Evidence of extinction in the present makes it essential that we learn from past analogues such as the End-Permian Mass Extinction, the Earth’s most severe mass extinction. During the End-Permian Mass Extinction, more than 90% of species were extinguished by intense climate change driven by large-scale volcanic greenhouse gas emissions. Previous studies exploring the End-Permian Mass Extinction focus on geological timescales of 10s or 100s of thousands of years. These studies therefore overlook the most relevant annual and decadal timescales of environmental change at which ecosystems respond. In this thesis, I use the Max-Planck Institute Earth System Model to investigate the dominant-mode of interannual variability and patterns of marine extreme events in the latest Permian climate, to better understand the conditions preceding the End-Permian Mass Extinction.
I contribute towards bridging the disparity of timescales by identifying patterns of short-term variability in tropical sea surface temperatures and their implications for marine biota. For the first time, I reveal the dominant-mode of interannual tropical variability in the latest Permian as an atmosphere-ocean feedback system centred in the equatorial West Panthalassic Ocean. This mode of variability is characterised by a periodicity of 2-4 years and affects the patterns of precipitation, temperature, mixing and transport across the tropical Tethys and Panthalassic Oceans. I relate the pattern of surface tropical marine heatwaves to the leading-mode of variability and determine maximum marine heatwave durations of up to 22 months and intensities of up to 5.9◦ C in the equatorial East Panthalassic.
I explore the spatial pattern of the deadly trio of marine stressors (ocean warming, acidification and deoxygenation) across the major shelf regions of the latest Permian and their relationship with evidence of higher extinction severity at extratropical latitudes. I find that contrary to previous studies, hypoxia is unlikely to drive extinction across high latitude shallow shelf regions and rather poses the greatest stress to the tropical Tethys Ocean. Uniquely among the latest Permian shelf regions, the East Panthalassic shelf is likely exposed to the full deadly trio. The southern high latitudes are exposed to intense marine heatwaves but both northern and southern high latitudes have a similar expected intolerance to pH extremes. My results support previous studies that suggest the extratropical extinction selectivity described by the fossil record may primarily be explained by ocean acidification rather than warming.
My results clearly indicate that the End-Permian Mass Extinction occurred earlier and faster in response to the climate perturbation than previously described. I suggest that better understanding of regional mass extinction triggers in the past can be applied to aid conservation efforts in the present. This can be achieved through a better comprehension of the evolving spatial patterns of environmental stress in response to climate change.Gegenwärtig verändern die vom Menschen verursachten Treibhausgasemissionen das Klima drastisch und bedrohen ganze Ökosysteme. Angesichts der Hinweise auf das gegenwärtige Artensterben ist es von entscheidender Bedeutung, dass wir aus den Analogien der Vergangenheit lernen, wie z. B. aus dem Massenaussterben gegen Ende des Perms, dem schwersten Massenaussterbeereignis in der Erdgeschichte. Während des Massenaussterben gegen Ende des Perms wurden mehr als 90% der Arten durch intensive Klimaveränderungen ausgelöscht, die durch großflächige vulkanische Treibhausgasemissionen verursacht wurden. Bisherige Studien zur Erforschung des Massenaussterben gegen Ende des Perms konzentrieren sich auf geologische Zeiträume von 10- oder 100-tausend Jahren. Diese Studien vernachlässigen daher die bedeutenden jährlichen und dekadischen Zeitskalen der Umweltveränderungen, auf die die Ökosysteme reagieren. In dieser Doktorarbeit verwende ich das Erdsystemmodell des Max-Planck-Instituts, um den vorherrschenden Modus der interannualen Variabilität und die Muster mariner Extremereignisse im Klima des jüngsten Perms zu untersuchen, um die Verhältnisse vor dem Massenaussterben gegen Ende des Perms besser zu verstehen.
Ich trage dazu bei, die Diskrepanz der Zeitskalen zu überbrücken, indem ich die Muster der kurzfristigen Variabilität der tropischen Meeresoberflächentemperaturen und ihre Auswirkungen auf die Meeresbiota identifiziere. Erstmals zeige ich den dominanten Modus der interannualen tropischen Variabilität im jüngsten Perm als ein atmosphärisch-ozeanisches Rückkopplungssystem auf, das im äquatorialen westlichen Panthalassa Ozean zentriert ist. Dieser Modus der Variabilität ist durch eine Periodizität von 2 bis 4 Jahren gekennzeichnet und beeinflusst die Muster von Niederschlag, Temperatur, Durchmischung und Transport über den tropischen Tethys- und Panthalassischen Ozean. Ich bringe das Muster der tropischen marinen Hitzewellen an der Oberfläche mit dem führenden Modus der Variabilität in Verbindung und identifiziere maximale marine Hitzewellen mit einer Dauer von bis zu 22 Monaten und einer Intensität von bis zu 5,9◦ C im äquatorialen östlichen Panthalassikum.
Ich analysiere das räumliche Muster des tödlichen Trios mariner Stressfaktoren (Ozeanerwärmung, Ozeanversauerung und Ozeansauerstoffentzug) in den wichtigsten Schelfregionen des jüngsten Perms und untersuche ihre Beziehung zu den Anzeichen für ein stärkeres Aussterben in außertropischen Breitengraden. Im Gegensatz zu früheren Studien komme ich zu dem Ergebnis, dass es unwahrscheinlich ist, dass Hypoxie das Aussterben in flachen Schelfregionen in hohen Breitengraden vorantreibt; vielmehr stellt sie die größte Bedrohung für den tropischen Tethys-Ozean dar. Unter den Schelfregionen des jüngsten Perms ist das Östliche Panthalassa Schelf wahrscheinlich dem gesamten tödlichen Trio ausgesetzt. Die südlichen hohen Breiten sind intensiven marinen Hitzewellen ausgesetzt, aber sowohl die nördlichen als auch die südlichen hohen Breiten weisen eine ähnliche erwartete Intoleranz gegenüber pH-Extremen auf. Meine Ergebnisse unterstützen frühere Studien, die darauf hindeuten, dass die im fossilen Datensatz beschriebene Selektivität des außertropischen Aussterbens in erster Linie durch die Versauerung der Ozeane und nicht durch die Erwärmung erklärt werden kann.
Meine Ergebnisse deuten darauf hin, dass das Massenaussterben gegen Ende des Perms als Reaktion auf die Störung im Klima früher und schneller stattfand als bisher angenommen. Ich schlage vor, dass ein besseres Verständnis der Auslöser regionaler Massenaussterben in der Vergangenheit zur Unterstützung von Naturschutzmaßnahmen in der Gegenwart genutzt werden kann. Dies kann durch ein besseres Verständnis der sich entwickelnden räumlichen Muster der Umweltbelastung als Reaktion auf den Klimawandel erreicht werden
Soziale Beziehungen und gesundheitsbezogene Lebensqualität von Menschen mit chronischen Wunden
Chronische Wunden stellen in alternden Gesellschaften eine wachsende Herausforderung dar. Neben körperlichen Belastungen können sie mentale und psychosoziale Beeinträchtigungen verursachen. Mit dem multidimensionalen Konstrukt der gesundheitsbezogenen Lebensqualität (HRQoL) können diese Beeinträchtigungen aus Sicht der Betroffenen erfasst werden. Soziale Beziehungen können hinsichtlich sozialer Kontakte, sozialer Partizipation und sozialer Unterstützung betrachtet werden. Ziel der vorliegenden Dissertation war es, HRQoL und soziale Beziehungen von Menschen mit chronischen Wunden zu beschreiben, den wundspezifischen Fragebogen Wound-QoL zu validieren und Zusammenhänge zwischen diesen beiden Konstrukten zu analysieren sowie Veränderungen in sozialen Beziehungen zu untersuchen. Zudem sollte der wundspezifische Fragebogen Wound-QoL validiert werden. Dafür wurde zunächst eine systematische Literaturrecherche zu sozialen Beziehungen bei Menschen mit chronischen Wunden durchgeführt. Anschließend wurde in einer europäischen Studie HRQoL beschrieben und der wundspezifische Fragebogen Wound-QoL validiert sowie Aspekte sozialer Beziehungen quantitativ und qualitativ erfasst.
Die Ergebnisse zeigen Beeinträchtigungen der wundspezifischen HRQoL in allen Bereichen, insbesondere hinsichtlich Psyche und Alltagsleben. Der eingesetzte Wound-QoL weist größtenteils gute psychometrische Eigenschaften auf. Basierend auf den Ergebnissen der Item-Response-Theorie wird eine neue Auswertungsmethode vorgeschlagen. Diese neue Auswertungsmethode könnte insbesondere in der internationalen Forschung eine genauere Messung der HRQoL ermöglichen, da sie methodische Probleme berücksichtigt und verringert.
Die subjektiv wahrgenommene soziale Unterstützung wurde von den Befragten insgesamt als hoch eingeschätzt, jedoch mit großen interindividuellen Unterschieden. Eine besondere Bedeutung haben Familienangehörige sowie Lebenspartnerinnen und -partner, die sowohl emotionale und alltägliche Unterstützung leisten als auch pflegerische Tätigkeiten übernehmen. Hier sollten einerseits Edukationsprogramme implementiert werden, um die nicht-professionelle Wundversorgung zu unterstützten und damit die Qualität der Versorgung zu erhöhen. Andererseits sollten Interventions- und Unterstützungsmaßnahmen angeboten werden, um körperliche und mentale Belastungen der Angehörigen zu verringern. Insbesondere bei Personen mit geringer sozialer Unterstützung durch den Familien- und Freundeskreis können professionelle Pflegefachkräfte eine wichtige Rolle einnehmen und eine Quelle emotionaler Unterstützung sein. Einrichtungen und Konzepte, bei denen Menschen mit chronischen Wunden zusammenkommen und sowohl professionelle Pflege erhalten als auch in den Austausch mit anderen kommen, könnten helfen, das soziale Wohlbefinden dieser Personen zu erhöhen. Durch eine regelhafte Erhebung in der Versorgung können soziale Beziehungen sowohl als Auslöser von Belastungen als auch als Outcome von Behandlungen betrachtet und evaluiert werden
Die vernachlässigte Rolle der evolutionären Anpassung von Phytoplankton in marinen Ökosystemmodellen — Implikationen für Modellvorhersagen und Perspektiven für die Zukunft
Marine ecosystems perform functions that are vital to global climate and human life, including the long-term storage of atmospheric carbon and food production. Despite their small size, phytoplankton are key players in marine ecosystem functioning, not only as primary producers, but also as a crucial component of biogeochemical cycles and climate-relevant feedback loops. Global warming, however, leads to a rapid reorganization of phytoplankton communities, with severe consequences for ecosystem functioning. To reliably assess future changes in ecosystem functioning, we need robust approaches.
While sedimentary records and marine monitoring data provide information on the past to present state of an ecosystem, albeit on different temporal scales, laboratory and mesocosm experiments allow to study marine organisms and communities under future levels of warming. Still, experiments can neither replicate the complexity of real ecosystems nor provide realistic rates of environmental change. Ecosystem models can fill this gap and are therefore a very well-suited tool for estimating future species- to ecosystem-level changes. However, while ecosystem models are increasingly being used to assess future changes in ecosystem functions such as net primary production and carbon export, their projections are inconsistent and differ not only in magnitude but also in direction. Since model projections represent a valuable resource for political decision-making, improving their informative value is crucial.
This thesis aims to help improve model projections of future ecosystem change by addressing two major uncertainties in current ecosystem models. First, most ecosystem models ignore the high adaptive potential of phytoplankton, which can be relevant on perennial time scales. We show that phytoplankton adaptation can significantly change simulated phytoplankton and ecosystem dynamics under both present-day and future climate conditions. Second, most ecosystem models are developed for ecosystems in their current state, so that they may no longer be applicable when ecosystem structure has changed in the future. We present a conceptual framework that makes use of data from sediment archives to develop evolutionary ecosystem models that are equally applicable to the past, the present, and, by implication, the future. This work not only emphasizes the need to revise current ecosystem models, especially those relevant for political decisions, but also provides approaches for solutions
