Modifizierungen von Titan- und Magnesiumoberflächen mit kontaktbioziden und low-fouling Eigenschaften mittels oberflächeninitiierter Polymerisation
Im Rahmen dieser Doktorarbeit wurde an verschiedenen Modifikationsmethoden für Titan geforscht. Dabei lag der Fokus auf einer oberflächeninitiierten Modifizierung mit Polymeren die eine low-fouling oder antimikrobielle Eigenschaft aufweisen und somit im medizinischen Bereich zu einer Verhinderung der Biofilmbildung verwendet werden können. Dafür wurden die beiden oberflächeninitiierten Polymerisationstechniken SI-ATRP und SI-PIMP evaluiert
Development of high-average-power laser plasma accelerators driven by industrial Yb lasers
Over the past two decades, laser plasma accelerators (LPAs) have emerged as a groundbreaking technology with immense potential for electron acceleration. Their ability to sustain exceptionally high accelerating gradients, on the order of 100 GV/m, and provide electron bunches with only few femtoseconds duration, promises a compact, cost-effective solution for numerous industrial, commercial, and medical applications. Yet, to transition from experimental setups to practical applications, it is crucial to enhance their robustness, reliability, and repetition rate. In this context, industrial-quality Ytterbium:Yttriumaluminium-garnet (Yb:YAG) lasers present an ideal, economically-efficient option, thanks to their inherently small quantum defect, high slope efficiency and high average power.
This thesis explores the feasibility of using industrial Yb:YAG lasers as drivers for plasma accelerators. Typically, these lasers deliver pulses of relatively long duration, extending to the picosecond-level. However, to excite a plasma wave, femtosecond durations are usually required. Hence, the temporal compression of the laser output to a few optical cycles is the first critical aspect under analysis. To address this challenge, an efficient double multi-passcell (MPC) post-compression scheme is employed, achieving the highest-ever compression factor to date for a 10 mJ-level pulse.
In addition to the driver laser, the plasma source plays a key role in the laser-plasma interaction, shaping the plasma density profile. Therefore, an extensive analysis of the most common sources for high-average-power LPA is presented. To overcome the limitations usually faced, a novel microfluidic source is proposed with unique capabilities for precise tailoring of the plasma profile along the laser axis, at the μm level. Its exceptional finetuning ability is demonstrated through a pioneering Bayesian optimisation tool, combining fluid dynamics and particle-in-cell simulations. This novel optimisation approach holds the promise to significantly boost the performances of LPA, particularly in application oriented scenarios.
Finally, the post-compressed laser output is used to demonstrate, for the first time, a plasma wakefield driven by an industrial Yb:YAGlaser. The laser-plasma interaction is thoroughly analysed and a clear path towards the first industrial-laser-drivem electron acceleration is presented
Photocages for time-resolved X-ray crystallography
Die zeitaufgelöste Röntgenkristallographie ist ein aufstrebendes Forschungsgebiet, das die Vorteile der jüngsten Fortschritte in der Detektortechnologie und der hochbrillanten Röntgenquellen von Synchrotrons und Freie-Elektronen-Lasern nutzt. Herausfordernd ist dabei die Anpassung der makromolekularen Kristallographie an einen zeitaufgelösten Experimentiermodus, dessen Hauptforschungsschwerpunkte unter anderem darin liegen, wie die Reaktion im Kristall ausgelöst werden kann. Ein Ansatz zur schnellen und genauen Reaktionsinitiierung im Kristall ist die Bestrahlung der Probe mit Laserlicht. Jedoch reagieren die meisten Proteine nicht von Natur aus auf diese Lichtanregung. Dies kann durch die Einführung von photolysierbaren Verbindungen, sogenannten Photocaging-Gruppen, in das Protein oder sein Substrat erreicht werden. Daher ist es wichtig, neue Photocagetypen für den Einsatz in der zeitaufgelösten Strukturbiologie zu erzeugen und zu charakterisieren. In diesem Projekt wurden eine Reihe von α-Carboxynitrobenzyl-Photocages synthetisiert und mit L-Aspartat verbunden. Die caged-Aspartatverbindugnen waren wasserlöslich, mit hydrolytischen Halbwertszeiten von 10 − 29 h, die sie für eine Reihe von biologischen Anwendungen geeignet erscheinen lassen. Die Quantenausbeute der Aspartatfreisetzung lag zwischen 0,05 und 0,14. α-Carboxynitrobenzyle sind chiral, und ihre Kopplung an L-Aspartat führt bei Verwendung eines Racemats zu einem diastereomeren Gemisch. Einige enantioselektive Routen wurden vorgeschlagen, scheiterten aber letztlich. Stattdessen wird die chirale Auflösung, anstatt chiraler Kontrolle, als ein Weg für zukünftige Arbeiten präsentiert. Mit der zeitaufgelösten UV-Vis-Spektroskopie wurde die Kinetik der α-Carboxynitrobenzyl-Photoreaktionen untersucht. Wichtige Aci-Nitro-Zwischenprodukte wurden mit Zeitkonstanten von 1 − 10 µs oder 1 ms, abhängig von den aromatischen Substituenten, beobachtet. Sub-Nanosekunden-Transienten von 450 fs und 1 ps wurden für α-Carboxy-6-nitropiperonylazetat beobachtet, die auf Schwingungsrelaxation bzw. die S1-Lebensdauer zurückzuführen sind.Time-resolved X-ray crystallography is an emerging research field that takes advantage of recent developments in detector technology and high-brilliance X-ray sources from synchrotrons and X-ray free-electron lasers. Adapting macromolecular crystallography to a time-resolved experimental mode is challenging, with reaction initiation in crystallo a key element that must be addressed. One approach that allows for rapid and accurate triggering of a reaction within a crystal uses laser light irradiation. However, most proteins may not inherently respond to light-initiation. This can be addressed by the introduction of photocaging groups into either the protein or its substrate. Therefore, it is important to both generate and characterise new photocage types for application in time-resolved structural biology. In this project, a number of α-carboxynitrobenzyl photocages were synthesised and joined to L-aspartate. The caged aspartates were water soluble, with hydrolytic half-lives of 10 − 29 h, making them suitable for a range of biological applications. The quantum yields of aspartate release ranged from 0.05 to 0.14. α-Carboxynitrobenzyls are chiral, and their coupling to L-aspartate results in a diastereomeric mixture if a racemate is used. Some enantioselective routes were proposed but failed. Chiral resolution, rather than chiral control, is suggested as an avenue for future work. Time-resolved UV-Vis spectroscopy was used to investigate the kinetics of the α-carboxynitrobenzyl photoreactions. Key aci-nitro intermediates were observed with time constants of 1 − 10 µs or 1 ms, depending on the aromatic substituents. Sub-nanosecond transients were observed for α-carboxy-6-nitropiperonyl acetate: 450 fs and 1 ps, attributed to vibrational relaxation and S1 lifetime, respectively

UV-Exposition multipotenter Dermaler Stammzellen als potentielle Zielzellen für die Entstehung des UV-induzierten malignen Melanoms
Malignant melanoma is the most aggressive and lethal type of skin cancer with rising incidence worldwide. The unequivocal risk factor is exposure to solar and artificial UV radiation. Despite the central role of UV-induced cyclobutane pyrimidine dimers (CPDs), particularly in the formation of UV signature mutations, the precise cellular and molecular mechanisms driving melanomagenesis are far from being understood in detail. Moreover, the cell of origin remains a subject of debate, with discussions centering on whether transformation initiates in mature epidermal melanocytes or stem cell precursors. An alternative theory proposes a damaged extrafollicular dermal melanocyte stem cell as the earliest origin. Within human skin, multipotent neural crest-derived dermal stem cells (DSCs) may serve as a reservoir for epidermal melanocytes. Although DSCs are constantly exposed to solar UV radiation, potentially contributing to the onset of melanoma, there is still insufficient investigation into how DSCs react to UV-induced damage. Therefore, this study aimed to establish workflows for the in vitro investigation of DSCs as potential cell of origin and of their response to UV irradiation. The work focused on examining functional stem cell features and understanding protection mechanisms to determine if UV-induced cell damage poses a higher risk in DSCs.
It was demonstrated that DSCs isolated from human foreskin can serve as a model to study their potential relevance in the genesis of malignant melanoma. Cultivated in specific stem cell medium, primary DSC cultures exhibited neural crest stem cell characteristics, including the expression of relevant markers, the ability to form three-dimensional spheres, and the potential for melanocytic differentiation. However, fibroblast contamination was a serious limitation for certain applications, necessitating the enrichment of DSCs. Immunomagnetic separation with negative selection based on CD90 and alternative non-cell-specific approaches, such as Geneticin treatment or selective detachment, proved unsuitable for DSC purification. Highly enriched DSCs with a good recovery rate were obtained using column-based MACS® positive selection targeting NGFRp75. These purified DSCs maintained viability and differentiation potential, although further validation is needed. This in vitro purification marks a crucial advancement in DSC research.
A comparative study of the DNA damage response after UVA and UVB irradiation revealed that DSCs reacted similarly to fibroblasts and melanocytes, displaying functional DNA repair and apoptosis induction. Protective mechanisms in DSCs were not superior to those of differentiated skin cells. Interestingly, DSCs exhibited an exceptional cell cycle regulation with a lack of growth arrest following UV irradiation, potentially indicating deficiencies in cell cycle checkpoints. This could lead to replication through unrepaired damage, mutations, and an elevated risk of transformation. Although initial evidence hints at a possibly heightened susceptibility of DSCs to UV-mediated carcinogenesis, further research is needed to establish a causal link between this specific radiation response and melanoma development.
UV irradiation predominantly had no impact on stem cell features and epigenetic regulation. Exposure to a single dose of UVA or UVB did not alter the expression of selected melanoma-specific microRNAs or the ability of DSCs to differentiate into melanocytes, neither impairing nor triggering differentiation. The radiation scheme might have been insufficient to induce UV-related effects, requiring multiple irradiations to adequately mimic physiological conditions. Irrespective of irradiation, a set of eight differentially expressed microRNAs allowed a clear distinction between DSCs, melanocytes, and melanoma cells, indicating significant differences in their overall baseline microRNA expression. By contrast, both single and multiple UVB exposures affected the sphere-forming potential of DSCs, resulting in reduced sphere counts. However, uncertainties arose regarding the influence of fibroblasts and cell aggregation in the utilized sphere formation assay, hindering a comprehensive evaluation of the results.
In summary, this study has successfully met its objectives by establishing workflows and methods that enabled an initial in vitro investigation of functional stem cell features and UV protection mechanisms in DSCs. While these findings yield a preliminary assessment of DSCs as potential cell of origin for malignant melanoma, they also raise questions for future investigations that could provide further insights into mechanisms underlying UV-induced melanomagenesis. Understanding how DSCs respond to UV exposure could enhance knowledge of early events in melanoma initiation, potentially improving prevention, early diagnosis, and the development of new treatment strategies
Die Rolle von Arc/Arg3.1 während der frühen Entwicklung bei der Verdrahtung von Hippocampus-Netzwerken
Critical periods are time windows in early development when genetical programs, and environmental and experiential factors confluence to tune functional properties of brain circuits towards their maturation. Concurrently, neural activity, synaptogenesis, and plasticity complete the wiring of hippocampal circuits giving rise to rhythmic activity. Work from our group has previously identified a critical period for hippocampal learning, wherein spontaneous upregulation of Arc/Arg3.1 in the hippocampus during the first postnatal month, permanently influences adult learning and hippocampal oscillations. Here, I propose that Arc/Arg3.1 shapes hippocampal circuits’ wiring and functional maturation during the critical period.
This thesis aimed to delve into the impact of Arc/Arg3.1 on the maturation of synaptic transmission in the hippocampus. My approach was to investigate the molecular, structural, and functional properties of hippocampal circuits in conditional Arc/Arg3.1 knockout (cKO) mice engineered in earlier studies, featuring deletions of the gene at various time points during the first postnatal month. These included a germline KO line, in which the deletion occurs during embryogenesis; an “early-cKO” line, where Arc/Arg3.1 is present during the first postnatal week and the deletion is completed between postnatal days 7-14 (P7-P14); and a “late-cKO” line, where the deletion is completed between P21-P36. I employed a broad range of methods encompassing: in-vitro field recordings, patch-clamp techniques, 3D dendritic reconstructions, immunohistochemistry and quantitative confocal microscopy, electron-microscopy, mass spectrometry-based proteomics, and subcellular fractionation with Western blotting.
The first part of this thesis evaluated the adult KO and cKO mice, at a time point where Arc/Arg3.1 had been fully deleted in the hippocampus and deficits in oscillatory activity and and learning had been observed. The findings presented here revealed an essential role of Arc/Arg3.1 in regulating the temporal dynamics of excitatory synapses in a development-dependent manner, with the most pronounced effects observed upon the earliest deletion. These effects were associated with changes in critical components of the postsynaptic density, including the transmembrane AMPA receptor regulatory proteins (TARPs) and PSD-95. Remarkably, my findings also showed alterations in the inhibitory synaptic transmission, hitherto believed to be independent of Arc/Arg3.1 plasticity, providing a first mechanistic understanding of the oscillatory deficits.
Part II of this thesis described the proteomic profile of the hippocampus in adult WT and Arc/Arg3.1 KO mice, focusing on differences between the CA1, CA3, and DG subfields. The results proved the efficacy of a nanosecond infrared laser (NIRL) ablation method to reliably isolate distinct regions of the mouse brain for subsequent proteomic analysis. Furthermore, I demonstrated that Arc/Arg3.1 regulates the proteomic hippocampal profile in a subfield-specific manner and identified novel exciting candidate proteins regulated by Arc/Arg3.1 under low activity levels. Additionally, enrichment analyses highlighted Arc/Arg3.1’s role in protein transport.
Finally, part III evaluated the role of Arc/Arg3.1 in the development of hippocampal function by examining mice of two and four weeks of age. My findings demonstrated that the kinetics of excitatory synaptic transmission are not altered in any of the KO lines at this stage of development, indicating that the alterations seen in the adult hippocampus develop at a later stage. In contrast to its effects in the adult brain, Arc/Arg3.1’s effects in the developing brain are more pronounced in response to early postnatal deletion, resulting in the active elimination of functional excitatory synapses and possibly the elimination of non-functional inhibitory synapses.
Collectively, this work provides further evidence of the existence of a critical period for the development of hippocampal function and demonstrates that Arc/Arg3.1 plays a vital role in the modulation of this critical period by shaping the wiring of hippocampal circuitry, not only by modulating excitatory synapses but, notably, also inhibitory transmission. My findings open new directions for investigating Arc/Arg3.1-dependent molecular pathways and cellular processes involved in brain wiring and plasticity
A decentralized immigration policy for the Federal Republic of Germany as a contribution to stabilizing shrinking regions
Motivation und Themensetzung:
Die Bundesrepublik Deutschland verfügt über keine tragfähige Strategie, um schrumpfende Regionen langfristig demographisch und strukturell zu stabilisieren. Zugleich wird der Zuzug von Einwandernden aus dem Ausland als Lösungsmöglichkeit gesehen, um dem demographischen Wandel zu entgegnen. Dabei findet Einwanderung fast ausschließlich in die großen urbanen Zentren des Landes hinein statt. Die wenigen bestehenden Politiken für eine räumliche Steuerung von Zuwanderung in schrumpfende Regionen hinein sind jedoch unzureichend ausgeprägt, meist nicht im Sinne der Betroffenen und der Gebietskörperschaften – sowie nur auf Geflüchtete bezogen.
Methoden:
Neben der Auswertung der Literatur zur internationalen guten Praxis dezentral ausgerichteter Einwanderungspolitiken werden in dieser Arbeit explorative Methoden für die Erfassung von individuellen Präferenzen der Eingewanderten erforscht, etwa entsprechend der Bewertung von Fotos mit Ortsbildern oder präferierten Regionen auf kartographischen Darstellungen. Weitere Methoden sind quantitative Befragungen und Fokusgruppendiskussionen zur Ermittlung der Akzeptanz einer Zuwanderung in die genannten Zielregionen hinein, etwa mit Hilfe von potentiellen Matching- und Anreizsystemen sowie von Homeffice-Arbeitsmodellen. Geoinformationssysteme (GIS) werden daneben zum Zweck einer Entwicklung von Indikatoren für ein Matchingsystem eingesetzt.
Ergebnisse:
Eine in Teilen dezentral ausgerichtete Einwanderungspolitik in Deutschland kann zur Stabilisierung schrumpfender Regionen beitragen. Insbesondere in Kombination mit einem mehrstufigen Matchingsystem, das im Sinne der Transparenz ohne Algorithmen mit nachvollziehbaren Indikatoren arbeitet. Und das unter Berücksichtigung individueller Präferenzen für die Verbesserung der Bleibeperspektiven von Eingewanderten in peripheren Regionen. Unter den in diesen Zielregionen lebenden Befragten wurde eine Zustimmung für eine potentielle Ansiedlungsförderung von Eingewanderten vor Ort von 61% ermittelt – unter Angabe verschiedener Prämissen. Die Akzeptanz für eine Ansiedlungsförderung durch potentielle Anreizsysteme wie Steuererleichterungen oder Zuschüsse für die Leerstandsanierungen lag unter den befragten Eingewanderten zudem bei 52-71%. Ebenfalls wurden Perspektiven für Eingewanderte in den Zielregionen, in Homeoffice-Jobs zu arbeiten, überprüft: Mit 58% positiven Haltungen der befragten vorwiegend kleinen und mittelständischen Arbeitgeber*innen aus dem Dienstleistungssektor.
Schlussfolgerungen und Empfehlungen:
In der deutschen Einwanderungspolitik sollte eine durchdachte Komponente dezentraler Lenkung implementiert werden, inklusive freiwilligen Matching- und Anreizsystemen und dem Ziel der Stabilisierung schrumpfender Regionen. Subventionen, Modellprojekte und ein Bundesgesetz für ein Hochskalieren der guten Praxis wären hierfür erforderlich.******************************************************************

Motivation and determination of the topic:
The Federal Republic of Germany lacks an effective strategy to stabilize regions facing long-term demographic and structural decline. At the same time, the influx of immigrants from abroad and resettlement to these regions is seen as a possible solution to mitigate demographic change. However, most immigrants move to big cities and the few existing policies for spatial control of immigration only apply to refugees. These policies are underdeveloped and usually fail to serve the interests of the affected individuals or meet the needs of local authorities and regions.
Methods:
This work consolidates pertinent literature of international good practices conceiving of decentralized immigration policies. Also qualitative explorative methods are utilized and examined to measure individual preferences of immigrants. This involves rating photographs depicting various places, towns or villages, as well as marking preferred regions on maps. Further methods encompass quantitative surveys and focus group discussions. These are aimed at questions regarding acceptance for the reception of immigration within the specified target regions, also regarding potential matching- and incentive systems as well as home office working models. Geographic information systems (GIS) are employed to generate indicators for a matching system.
Results:
An immigration policy in Germany with a partly decentralized approach can help to stabilize shrinking regions. This can be made possible by implementing a multi-stage matching system that works with comprehensible indicators. For transparency towards civil society functioning without help of complex algorithms; also considering individual preferences to enhance the prospects of immigrants staying in their chosen locations. Among respondents residing in the target regions, one questionnaire revealed that 61% were found to be supportive of initiatives aimed at facilitating the settlement of immigrants in their hometowns, under various conditions. The approval rates of potential incentive systems such as tax reliefs, subsidies for the reconstruction and renovation of vacant housing and other systems ranged from 52% to 71% among the surveyed immigrants. Furthermore, the possibility for immigrants to work remotely from home offices in the target regions was investigated, resulting in an overall positive feedback rate of 58% among surveyed small and medium-sized employers in the service sector.
Conclusions and recommendations:
It is recommended that German immigration policy integrates voluntary decentralized distribution components. These involve establishing a matching and an incentive system aimed at stabilizing shrinking regions, particularly small and medium-sized towns. Implementing such a policy would require the provision of subsidies, the establishment of pilot projects, and the enactment of federal legislation to scale up successful initiatives
Social inequalities, (persisten) somatic symptoms and health anxiety
(Persistierende) somatische Symptome und Gesundheitsangst sind ätiologisch wie auch diagnostisch eng verwoben und assoziiert mit hohen individuellen sowie ökonomischen Belastungen. Dennoch ist die Forschung sowie medizinische Versorgung trotz der hohen klinischen Relevanz oftmals fragmentiert und von Stigmatisierung gegenüber betroffenen Patient:innen geprägt. Bestehende Evidenz deutet auf soziale Ungleichheiten in den beiden Gesundheitsoutcomes hin, bislang mangelt es jedoch an einer metaanalytischen Synthese. Die bislang primär unidimensionale Betrachtung sozialer Ungleichheiten vernachlässigt ferner die intersektionale Verschränkung sozialer Dimensionen sowie Erfahrungen struktureller Unterdrückung. In der vorliegenden Dissertation wurden daher erstmalig zwei wichtige (psychosomatische) Gesundheitsoutcomes mit den Konzepten der Gesundheitsgerechtigkeit und Intersektionalität zusammengeführt. Konkrete Ziele waren dabei die vertiefte Untersuchung sozialer beziehungsweise intersektionaler Ungleichheiten in somatischer Symptomschwere und Gesundheitsangst sowie, auf den gewonnenen Erkenntnissen aufbauend, das Ableiten gezielter Maßnahmen für Forschung und Praxis. Dazu wurde zum einen exemplarisch eine systematische Übersichtsarbeit mit Metaanalyse zu sozialen Ungleichheiten in Gesundheitsangst angefertigt. Zum anderen wurden auf Basis der Primärdaten einer deutschlandweiten Bevölkerungsbefragung intersektionale Regressionsmodelle mit Interaktionstermen sowie darauf aufbauend als MAIHDA zu den beiden Gesundheitsoutcomes gerechnet. Die untersuchten intersektionalen Strata wurden aus den sozialen Dimensionen Geschlecht, Migrationsgeschichte und Einkommen gebildet, welche mit (historischer) Unterdrückung in Form von Sexismus, Rassismus und Klassismus/Armut assoziiert sind. Die Dissertation entstand im Rahmen der Studie ‛Social inequalities in aggravating factors of somatic symptom persistence (SOM!;SOC)“, welche Teil der interdisziplinären Forschungsgruppe ‛Persistent SOM!tic symptoms !CROSS diseases – from risk factors to modification (SOM!CROSS)” (FOR 5211) ist. Durch die vertieften Einblicke in den sozialen Kontext (persistierender) somatischer Symptome und Gesundheitsangst trägt die vorliegende Dissertation wesentlich zu den Gesamtzielen der SOMA.SOC Studie wie auch der Forschungsgruppe SOMACROSS bei und sensibilisiert für das wichtige Thema der Intersektionalität in Forschung und Praxis. Die vorliegende Dissertation deckte intersektionale soziale Ungleichheiten in somatischer Symptomschwere und Gesundheitsangst auf. Dabei wurde in der Metaanalyse von 37 Primärstudien die Relevanz sozioökonomischer Benachteiligung bei Gesundheitsangst ersichtlich. Ferner ließen sich aus den Regressionsmodellen intersektionale Ungleichheiten in Gesundheitsangst wie auch somatischer Symptomschwere ableiten, wobei sich ein geringes Einkommen als größter Einflussfaktor erwies. Die signifikant am stärksten von somatischer Symptomschwere wie auch Gesundheitsangst betroffenen intersektionalen Strata bildeten weibliche Personen der ersten Migrationsgeneration mit niedrigem Einkommen sowie männliche Personen der zweiten Migrationsgeneration mit niedrigem Einkommen. Die intersektionalen Effekte konnten am ehesten als additiv eingestuft werden, ein eindeutiger intersektionaler Gradient fand sich nicht. Vor dem Hintergrund der ätiologischen Relevanz negativer Stressoren und der geringen Unterscheidungsgenauigkeit zwischen den intersektionalen Strata erscheinen insbesondere universelle Maßnahmen, abzielend auf strukturelle wie individuelle Stressoren, vielversprechend. Dabei bedarf es zum einen grundlegender sozial- und gesundheitspolitischer Maßnahmen, um die systematische Ungleichverteilung von Macht, Prestige und Ressourcen zu reduzieren und in der Folge Gesundheitsgerechtigkeit zu fördern. Zum anderen bedarf es konkreter Maßnahmen für die klinische Versorgung, um gezielt intersektionale Ungleichheiten in somatischer Symptomschwere und Gesundheitsangst zu reduzieren. Multimodale Untersuchung des cerebello-thalamo-kortikalen Netzwerks beim Tremor der Parkinsonerkrankung
Background: Parkinson's disease (PD) is characterised by motor symptoms, including tremor, which significantly impair patients' quality of life. Recent research has shifted focus to understanding the pathophysiology of tremor as a network phenomenon, specifically involving the basal ganglia-thalamo-cortical (BTC) and cerebello-thalamo-cortical (CTC) networks. It has been found that the CTC network, particularly the cerebellum, is involved in maintaining and increasing tremor amplitude.
Methods: This thesis employed a multimodal approach that integrated structural and functional magnetic resonance imaging (MRI), computational modelling, and non-invasive brain stimulation to analyse the dynamics of the cerebellum and CTC network in PD and tremor. Four sequential studies focused on morphological analyses, network dynamics, stimulation protocols, and device development.
Results: Significant changes were observed in the volume of cerebellar lobule VIIb, strongly correlating with the severity of PD tremor, suggesting its potential as a biomarker. Disruptions in brain equilibrium due to PD were noted throughout the brain and within the CTC network, accompanied by an increased influence of the cerebellum in the diseased state. These findings were complemented by optimizing the montage of cerebellar stimulation for efficiency and safety, recommending a 5x5 sponge electrode placed 2 cm lateral to the inion and a 5x7 counter electrode on the jaw. Based on these insights, a comprehensive experimental protocol for effective, closed-loop, phase-adaptive, non-invasive cerebellar transcranial alternating current stimulation (tACS) was developed. Concurrently, a novel device was designed and developed that integrates all necessary functions to achieve significant real-time alignment between tACS and tremor, ensuring both safety and efficacy.
Conclusion and Significance: The extensive multimodal study of the CTC network confirms its significant role in the tremor physiology of PD, both structurally and dynamically. The increased influence of the cerebellum on the hierarchical organization of the brain in PD underscores its suitability as a target for non-invasive brain stimulation to restore a healthy brain equilibrium. The optimized stimulation pattern and the designed closed-loop, phase-adaptive tACS device enable targeted modulation of cerebellar oscillatory activity. This thesis represents an endeavour to advance the existing knowledge on PD tremor pathophysiology and creates opportunities for personalized, non-invasive treatment strategies for this symptom.Hintergrund: Die Parkinsonerkrankung (PD) ist durch motorische Symptome gekennzeichnet, einschließlich Tremor, der die Lebensqualität der Patienten erheblich beeinträchtigt. Diese Ergebnisse wurden durch die Optimierung der Montage der cerebellären Hirnstimulation in Bezug auf Effizienz und Sicherheit ergänzt, indem eine 5x5 Schwammelektrode 2 cm seitlich des Inions und eine 5x7 Gegenelektrode am Kiefer platziert wurden. Basierend auf diesen Erkenntnissen wurde ein umfassendes experimentelles Protokoll für eine effektive, closed-loop, phasenadaptive, nicht-invasive cerebelläre phasenadaptive transkranielle elektrische Wechselstromstimulation (tACS) entwickelt. Parallel dazu wurde ein neuartiges Gerät entworfen und entwickelt, das alle notwendigen Funktionen integriert, um eine signifikante Echtzeit-Anpassung zwischen tACS und Tremor zu erreichen und sowohl Sicherheit als auch Effizienz zu gewährleisten.
Schlussfolgerung und Bedeutung: Die umfassende multimodale Untersuchung des CTC-Netzwerks bestätigt seine bedeutende Rolle in der Tremorphysiologie bei PD, sowohl strukturell als auch dynamisch. Der erhöhte Einfluss des Cerebellums auf die hierarchische Organisation des Gehirns bei PD unterstreicht seine Eignung als Ziel für nicht-invasive Hirnstimulation, um ein gesundes Hirngleichgewicht wiederherzustellen. Das optimierte Hirnstimulationsprotokoll und das entworfene closed-loop, phasenadaptive tACS-Gerät ermöglichen eine gezielte Modulation der cerebellären oszillatorischen Aktivität. Diese Arbeit trägt zum aktuellen Wissen über die Tremorphysiologie bei PD und zur Entwicklung neuer, personalisierter, nicht-invasiver Behandlungsstrategien für dieses Symptom bei
