Caracterización de los mecanismos moleculares de translocación de K+ y nuevos reguladores del transportador SlHAK5 en Solanum lycopersicum L
Objectives
K+ is an essential macronutrient for plants, involved in numerous physiological processes. In the current context, where it is necessary to develop varieties with improved nutrient uptake and use efficiency to address agronomic and environmental challenges, understanding the molecular mechanisms regulating K+ uptake and translocation is of particular relevance. Therefore, this doctoral thesis pursued three main objectives: (1) to study the role of the kinase SlCIPK9 in the regulation of the SlHAK5 transporter in tomato; (2) to analyze the function of the SlNRT1.5 transporter in K+ translocation from root to shoot; and (3) to identify new SlCIPK/SlCBL complexes involved in the activation of the SlHAK5 transporter.
Methodology
The above objectives were addressed through a combination of approaches, including heterologous expression assays in yeast and Xenopus laevis oocytes, bifluorescence assays to study protein–protein interactions, generation of knockout plants using CRISPR-Cas, hydroponic cultivation of tomato plants, determination of cation and anion content, pollen germination and tube elongation studies, as well as microscopy techniques.
Conclusions
The experimental work led to the following conclusions:1. slcipk9 mutant plants do not exhibit impaired K+ uptake or growth under low-K+ conditions, suggesting a limited role of SlCIPK9 in K+ nutrition in tomato. 2. SlCIPK9 is predominantly expressed in stamens, and its loss of function led to significantly shorter pollen tubes, revealing a specific role in pollen tube elongation. 3. The pollen phenotype observed in slcipk9 mutants occurs despite normal K+ content, indicating that SlCIPK9 regulates pollen tube elongation via a K+-independent mechanism. 4. SlNRT1.5 mediates K+ efflux when is expressed in Xenopus oocytes and, in tomato plants, it is expressed in multiple tissues, with particularly high expression in the root stele. 5. slnrt1.5 plants accumulate less K+ shoots than WT plants under low external pH (4.5) and limited K+ (0.05 mM) and N (0.5 mM) supply, indicating that SlNRT1.5 is a central component of K+ translocation, most likely by mediating K+ loading into the xylem vessels. 6. Further analyses of slskor slnrt1.5 double mutants reveal an overlapping role of SlSKOR and SlNRT1.5 to shoot K+ accumulation, with SlNRT1.5 playing a more prominent role under low-K+ conditions. 7. The targeted yeast complementation assay identify SlCIPK15 and SlCIPK26 as novel kinases capable of activating the high-affinity K+ transporter SlHAK5. Their functionality was confirmed through Rb+ uptake assays in yeast and BiFC interaction studies in Nicotiana. 8. Studies with slcipk15 knockout plants shows that this kinase plays an important role in pollen tube elongation but not in pollen germination. Unexpectedly, SlCIPK15 acts as a negative regulator of K+ translocation rather than as a positive regulator of SlHAK5-mediated K+ uptake. 9. Functional characterization of slcipk26 knockout lines reveals impaired pollen germination and phenotypes resembling those of slhak5 mutants, suggesting that SlCIPK26 regulates SlHAK5 in reproductive tissues. In contrast, in roots SlCIPK26 does not appear to play a major role in SlHAK5-mediated K+ transport
"Sea alabado por siempre" : la función mistagógica de la oración de alabanza en la autobiografía teresiana. Una aproximación desde la antropología teológica
Objetivos:
La tesis tiene como objetivo general estudiar de forma sistemática las oraciones de alabanza en el Libro de la Vida de Santa Teresa de Ávila, entendiéndolas como portadoras de una teología de la gratuidad divina y como un capítulo fundamental de su mistagogía: la invitación a entrar y profundizar en el Misterio. Entre los objetivos específicos se incluyen: definir un marco teórico sobre el relato de conversión; sintetizar los desarrollos teológicos sobre la gratuidad; mostrar la riqueza del concepto de mistagogía; analizar el contexto histórico y biográfico; sistematizar contenido teológico y recursos retóricos; y comentar las oraciones para evitar diluir su singularidad.
Metodología:
La investigación es teológica, cualitativa e interdisciplinar, con diálogo entre literatura, teología y ciencias humanas. Se ha desarrollado en dos grandes partes: una teórica, abordando los conceptos de conversión, gratuidad y mistagogía; y una empírica, centrada en el análisis del Libro de la Vida. Se han utilizado enfoques sistemáticos y hermenéuticos para desvelar el contenido teológico y los recursos literarios que intensifican la eficacia mistagógica de las oraciones de alabanza.
Resultados y conclusiones:
La tesis demuestra que las oraciones de alabanza no son meros añadidos devocionales, sino recursos mistagógicos que acompañan e invitan al lector al Misterio de Dios. Se ha desarrollado el modelo del “triángulo mistagógico”, interpretando la autobiografía teresiana como un dispositivo pedagógico integral. Se identifican tres modalidades mistagógicas: pedagogía teológica, acompañamiento espiritual e invitación directa al Misterio. Las conclusiones destacan la novedad de este enfoque, su relevancia teológica y la apertura de nuevas líneas para estudios futuros sobre la mistagogía teresiana.Objectives:
The general objective of this dissertation is to systematically study the prayers of praise in Saint Teresa's Book of Life, understanding them as carriers of a theology of divine gratuity and as a fundamental chapter of her mystagogy: the invitation to enter and deepen in the Mystery. Specific objectives include defining a theoretical framework on conversion narrative; synthesizing theological developments on gratuity; showing the richness of the concept of mystagogy; analyzing the historical and biographical context; systematizing theological content and rhetorical resources; and commenting on the prayers to preserve their unique character.
Methodology:
The research is theological, qualitative, and interdisciplinary, engaging in dialogue between literature, theology, and human sciences. It was developed in two main parts: a theoretical one, addressing the concepts of conversion, gratuity, and mystagogy; and an empirical one, focusing on the analysis of the Book of Life. Systematic and hermeneutical approaches were used to reveal the theological content and literary resources that intensify the mystagogical effectiveness of the prayers of praise.
Results and conclusions:
The thesis demonstrates that the prayers of praise are not mere devotional additions but mystagogical resources that accompany and invite the reader into the Mystery of God. The “mystagogical triangle” model was developed, interpreting Teresa's autobiography as a comprehensive pedagogical device. Three mystagogical modalities were identified: theological pedagogy, spiritual accompaniment, and direct invitation to the Mystery. The conclusions highlight the novelty of this approach, its theological relevance, and the opening of new avenues for future studies on Teresian mystagogy
Multi-objective evolutionary feature selection for ensemble learning with random forests in time series forecasting
Time series forecasting is fundamental in numerous domains, including finance, healthcare, energy, and environmental monitoring. However, the high dimensionality of feature spaces can lead to overfitting and reduced interpretability, making feature selection a crucial preprocessing step. This paper proposes a multiobjective evolutionary algorithm for feature selection in time series forecasting, designed to enhance predictive accuracy while improving generalization. The method partitions the dataset, associating each partition with an objective function in the optimization process. By independently selecting relevant feature subsets, it generates a Pareto front of Random Forest models, each trained on a distinct subset of features. These models are then aggregated into a stacking-based ensemble framework, effectively balancing feature relevance and diversity. Additionally, we introduce a feature importance measure based on selection frequency in the non-dominated solutions of the optimization process. To validate our approach, we conduct experiments on real-world forecasting tasks, including air quality prediction in southeastern Spain and Italy and oil temperature forecasting in industrial applications. We also evaluate performance on synthetic datasets of increasing complexity, systematically varying instances, features, seasonality, noise, and trends. The proposed method is compared against conventional Random Forest, a wrapper-based feature selection method with a multiobjective evolutionary search strategy, and several state-of-the-art embedded feature selection techniques for time series forecasting. The results demonstrate that our approach significantly improves forecasting accuracy while mitigating overfitting. By integrating multi-objetive evolutionary optimization, random forest, ensemble learning, and a novel feature importance measure, our method offers a robust, interpretable, and effective feature selection for time series forecasting applications
Emotional intelligence in a language classroom through servicelearning.
Este trabajo pretender inculcar en el alumnado la importancia del bienestar emocional propio y ajeno a través de la metodología del aprendizaje-servicio. Los educandos elaborarán un listado de problemáticas cuya resolución consideren factible desmontando mitos que les hayan socavado su bienestar emocional. El punto de partida será la oralidad y el trabajo colaborativo (individual, pequeño y gran grupo). Esta tarea competencial permitirá romper el hielo entre iguales a comienzo de curso, dando confianza a aquellos alumnos que no tengan el castellano como lengua materna (LM), o bien sean de reciente incorporación, se fomentará la oralidad entre iguales como primer paso para determinar cuál será el objeto del aprendizaje-servicio que desarrollarán posteriormente. Este proyecto responde ante diversos retos: motivar a nuestro alumnado hacia el aprendizaje significativo, desarrollar el espíritu crítico evitando -o al menos dificultando- caer en bulos o noticias falsas que puedan generar comportamientos sexistas o racistas. Se pretende reforzar un aprendizaje significativo y competencial a través de la toma de decisiones, planificando y gestionando el tiempo. Hemos planificado esta experiencia en un tercero se secundaria.Abstract: This research project seeks to develop the importance of their own and others; emotional well-being through the service-learning methodology between students. The learners will proposals their list of problems whose resolution they consider feasible by dismantling myths that have undermined their emotional wellbeing. The starting point will be oral and collaborative work (one on one, small and larger groups). This competence task will allow breaking the ice between peers at the beginning of the course, giving confidence to those students who do not have Spanish as their mother tongue (LM), or are newcomers, oral communication between peers will be encouraged as a first step to determine what will be the object of the service-learning that they will develop later. This project fulfils several challenges: Firstly, motivating our students towards meaningful learning. Secondly, developing the critical spirit of our students avoiding -or at least making it difficult- to fall into hoaxes or false news that may generate sexist or racist behavior. The purpose of this project is to reinforce meaningful and competent learning through decision making, planning and time management. Protocolo de ecografía para diagnóstico precoz del desprendimiento de retina
El desprendimiento de retina (DR) es una emergencia en la que el diagnóstico precoz resulta fundamental para prevenir la pérdida irreversible de la visión. Sin embargo, en presencia de opacidad de los medios, la evaluación del fondo de ojo puede verse limitada, de modo que la ecografía oftálmica se convierte en la principal alternativa diagnóstica para una valoración temprana.
El objetivo de este estudio fue determinar el rendimiento del protocolo Arrixaca, que evalúa los principales hallazgos ecográficos para el diagnóstico diferencial del DR frente a otras patologías de apariencia similar como el desprendimiento de vítreo posterior (DVP) o el hemovítreo (HV).
Se realizó un estudio observacional prospectivo en 127 pacientes con indicación de ecografía oftálmica por sospecha de DR. El examen se realizó utilizando el protocolo Arrixaca, aplicado por tres radiólogos con distintos años de experiencia.
Se comparó la precisión diagnóstica con el gold standard (fondo de ojo o hallazgos quirúrgicos según la indicación clínica) y la concordancia interobservador entre los tres radiólogos.
Los resultados mostraron que el protocolo Arrixaca consiguió un excelente rendimiento con un muy alto porcentaje de acierto, superior tanto al protocolo UM como a los resultados registrados en publicaciones previas, incluso en condiciones de opacidad de medios. Este análisis destaca especialmente al protocolo Arrixaca por su capacidad de minimizar los falsos negativos, un aspecto crítico para proteger la visión del paciente.
Además, la concordancia entre los tres radiólogos fue muy elevada, por lo que el protocolo Arrixaca reveló ser altamente reproducible y sencillo.
En conjunto, los hallazgos confirman que la ecografía oftálmica con el protocolo Arrixaca es una alternativa eficaz y accesible para el diagnóstico de DR, aún en pacientes con opacidad de medios. Su incorporación en los servicios de Radiología podría optimizar la detección precoz del DR y mejorar el pronóstico visual de los pacientes.Retinal detachment (RD) is a medical emergency where early diagnosis is crucial to prevent irreversible vision loss. However, in the presence of media opacity, fundoscopic evaluation may be limited, making ophthalmic ultrasound the primary diagnostic alternative for early assessment.
The aim of this study was to determine the performance of the Arrixaca protocol, which evaluates key ultrasound findings for the differential diagnosis of RD compared to other similar-appearing conditions such as posterior vitreous detachment (PVD) or vitreous hemorrhage (VH).
A prospective observational study was conducted on 127 patients referred for ophthalmic ultrasound due to suspected RD. The examination was performed using the Arrixaca protocol, applied by three radiologists with varying years of experience.
Diagnostic accuracy was compared with the gold standard (fundoscopy or surgical findings, depending on clinical indication), as well as interobserver agreement among the three radiologists.
The results showed that the Arrixaca protocol achieved outstanding performance, with a very high accuracy rate, surpassing both the UM protocol and previously published results, even in cases with media opacity. This analysis particularly highlights the Arrixaca protocol for its ability to minimize false negatives, a critical factor in preserving the patient’s vision.
Additionally, interobserver agreement among the three radiologists was very high, confirming that the Arrixaca protocol is highly reproducible and easy to implement.
Overall, the findings confirm that ophthalmic ultrasound using the Arrixaca protocol is an effective and accessible alternative for the diagnosis of RD, even in patients with media opacity. Its incorporation into Radiology departments could enhance early RD detection and improve visual prognosis for patient
Evaluating Interventions to Mitigate Academic Stress inMedical Students: A Systematic Review
Antecedentes: La formación médica, caracterizada por altas demandas académicas y emocionales, expone a los estudiantes a niveles significativos de estrés. Este estrés puede comprometer su salud mental, manifestándose en síntomas como ansiedad, depresión y burnout, e impactando negativamente en su rendimiento académico. A largo plazo, estas consecuencias pueden afectar la calidad de la atención médica que brindarán en el futuro. Esta revisión analiza intervenciones llevadas a cabo para mitigar los efectos del estrés académico en esta población durantel os últimos 5 años. Métodos: Se realizó una búsqueda siguiendo las directrices PRISMA en las bases de datos PubMed, Web of Science y Scopus. Los términos clave incluyeron "Estudiantes de medicina","estrés académico" e "Intervenciones". Se seleccionaron seis estudios que reportaron intervenciones presenciales y virtuales en esta población y se clasificaron según tres categorías: mindfulness y meditación, yoga y prácticas físicas, y entrenamiento psicológico e inteligencia emocional.Resultados: Cinco estudios evaluaron tres tipos principales de intervenciones: yoga y meditación,intervenciones nutricionales y programas de manejo del estrés. Todas las intervenciones mostraronreducciones significativas en el estrés y mejoras en el bienestar, inteligencia emocional y flexibilidadpsicológica. El yoga y la meditación mejoraron los niveles de estrés percibido y los marcadoresfisiológicos del estrés. Las intervenciones nutricionales con alimentos fermentados, como actobacillus casei, ayudaron a reducir los síntomas gastrointestinales relacionados con el estrés. Los programas de manejo del estrés centrados en estrategias de afrontamiento basadas en problemas aumentaron la autoeficacia y redujeron el malestar psicológico. Conclusiones: Los hallazgos de esta investigación subrayan la necesidad crítica de realizar estudios adicionales sobre intervenciones efectivas para mitigar el estrés académico en estudiantes de medicina en esta línea, mejorando subienestar y capacidad para brindar atención de calidad en el futuro.Background: Medical training, characterized by high academic and emotional demands,exposes students to significant levels of stress. This stress can compromise their mental health,manifesting in symptoms such as anxiety, depression, and burnout, and negatively impacting theiracademic performance. In the long term, these consequences can affect the quality of medical carethey provide in the future. This review analyzes interventions conducted to mitigate the effects ofacademic stress in this population over the past 5 years. Methods: A search was conducted followingPRISMA guidelines in the PubMed, Web of Science, and Scopus databases. Keywords included"medical students," "academic stress," and "interventions." Six studies were selected that reported in-person and virtual interventions in this population and were classified into three categories:mindfulness and meditation, yoga and physical practices, and psychological training and emotional ntelligence. Results: Five studies evaluated three main types of interventions: yoga and meditation,nutritional interventions, and stress management programs. All interventions showed significantreductions in stress and improvements in well-being, emotional intelligence, and psychologicalflexibility. Yoga and meditation improved perceived stress levels and physiological markers of stress.Nutritional interventions with fermented foods, such as Lactobacillus casei, helped reduce stress-related gastrointestinal symptoms. Stress management programs focused on problem-focusedcoping strategies increased self-efficacy and reduced psychological distress. Conclusions: Thefindings of this research underscore the critical need for additional studies on effective interventionsto mitigate academic stress in medical students along these lines, improving their well-being andability to provide quality care in the futur
