LactoTruite, a project to increase the resilience of trout farms towards the emergence of lactococcosis in Brittany
International audienceThe aquaculture sector must develop strategies to adapt towards the consequences of global warming, particularly to face the increase of infectious pathologies.. Lactococcosis is a bacterial disease frequently affecting fish farms of the Mediterranean countries, with severe clinical signs at water temperature above 17°C. During summer 2023, Brittany trout farms where strongly affected by a lactococcosis outbreak, generating up to 50% of mortality in some farms and an associated economic loss of several millions of euros. AimsThe aim of this project is to bring short and mid-terms solutions to the fish farmers to understand and fight against the pathology and limit its impact in the future. MethodologyThe first task will be dedicated to an epidemiological study of the disease and its etiological agent. The aims will be to understand how the disease occurs in fish farm, to identify bacterial species involved, their virulence factors and antibioresistance profiles, and to develop robust and discriminant diagnosis tools. The second task will be focused on the optimization of vaccinal and therapeutic (antibiotherapy) schemes using experimental in vivo approachesDuring the third task, the heritability of genetic resistance of rainbow trout against lactococcosis but also another important bacterial diseases, flavobacteriosis, will be evaluated and correlate with growth traits, allowing to estimate potential survival gains obtainable through trout selection. Moreover, we will investigate the immune mechanisms commonly implemented in fish surviving to these both bacterial diseases , with the objective to to evaluate the possibility of leading a genetic selection based on the immune robustness of the fish.Conclusion Epidemiological and descriptive data coupled with the development of concrete solutions will allow to better understand and fight against the lactococcosis pathology that is enhanced by the climate evolution. These tools will help to preserve the trout farming sector and maintain its competitiveness
Longitudinal monitoring of racehorse welfare during preparation for the jump racing season
International audienceRacing preparation in young racehorses is a pivotal phase marked by intense physical, psychological, and environmental adaptation. Monitoring welfare during this period is essential to protect animal well-being, as well as to support performance and public acceptability of the sport. Structured protocols for assessing the welfare of racehorses have recently been developed, but their use in field conditions remains limited. This study aimed (1) to evaluate the feasibility of implementing a welfare assessment protocol in a professional French jump racing yard and (2) to monitor longitudinal changes in physical and behavioural welfare indicators during the training of young racehorses preparing for the jump racing season. Sixteen two- to three-year-old racehorses (50% Thoroughbreds, 50% French Non-Thoroughbred racehorses) were assessed at three time points over a four-month period: October 2023 (T0), November 2023 (T1), and February 2024 (T2), prior to the start of the jump racing season. Horses were housed in the same yard, trained and managed consistently by the same staff, and observed in their usual environment without altering their routine or workload. Each welfare parameter was consistently assessed by the same trained, non-blinded equine veterinarian across visits. Environmental assessment, physical health parameters, facial expression using the horse grimace scale (HGS), and human–horse relationship tests (approach tests) were systematically performed in the afternoon, during calmer periods after training and outside feeding times, while behavioural time budgets via instantaneous scan sampling were always observed across full days (6a.m. - 6p.m.). Repeated measures were analysed using linear and logistic mixed-effects models. All horses had access to high-quality forage ad libitum, clean water, and appropriate bedding. However, turnout was absent, and most horses had only visual social contact (67.5%). Body condition score declined significantly at T1and T2, particularly in horses with shorter prior training duration, suggesting physiological adaptation to conditioning. External and internal lip commissure lesions were present in 65% and 75% of observations, respectively, raising concerns about potential equipment-related discomfort. Horse grimace scores increased significantly from T0 to T2, suggesting a rise in discomfort or fatigue. Approach test responses did not vary significantly over time. While behavioural time budgets remained globally stable at group level, notable inter-individual variation was observed. Horses with physical social contact had significantly lower HGS, suggesting a potential beneficial effect of social interaction on discomfort or pain expression. However, paradoxically, these horses also exhibited significantly more stereotypic behaviours. This longitudinal field-based study confirms the feasibility and relevance of structured assessments to monitor welfare during racehorse training. The protocol enabled repeated, individualised monitoring under real-life conditions and highlighted several welfare indicators evolving between the early training phase and the start of the racing season. While causal inference is limited, these findings illustrate how structured welfare assessments can be integrated into routine veterinary visits, offering practical tools for trainers and veterinarians to monitor horses’ adaptation during training and to strengthen welfare standards across the racing sector
Contribution au développement d'un atlas multimédia numérique de sémiologie : système nerveux
L’apprentissage de la sémiologie médicale vétérinaire nécessite une exposition à de nombreux cas cliniques, souvent insuffisante en formation initiale. Pour y répondre, Oniris, rejointe depuis 2023 par les ENVF, a lancé la création de SemioVet, un site web conçu comme un glossaire illustré de signes cliniques enrichi d’explications physiopathologiques et de quiz pour en faciliter la reconnaissance et l’interprétation. L’objectif de ce travail était de créer le contenu de SemioVet pour l’examen du système nerveux des ruminants. Au total, 114 signes cliniques ont été recensés et 92 nouvelles fiches de signes cliniques élaborées. Seules 47 fiches ont été illustrées et 228 quiz relatifs à la sémiologie nerveuse ont été réalisés. Pour 15 fiches de signes jugés les plus observés, du contenu supplémentaire a été rédigé tels que les mécanismes physiopathologiques, les réflexes cliniques et des exemples de maladies associées. Bien que le site soit encore en développement, il représente un outil prometteur pour l’enseignement vétérinaire, en permettant aux étudiants de clarifier les définitions et de visualiser diverses manifestations d’un même signe, notamment pour les signes rares comme ceux de l’examen du système nerveux. Ce travail a relevé l’importance de la propédeutique, en reprenant les étapes de l’examen clinique pour n’oublier aucun signe clinique. Il a aussi confirmé la difficulté à synthétiser certains mécanismes physiopathologiques et réflexes cliniques. L’ajout de multimédia reste à réaliser, renforçant la nécessité d’enregistrer les signes cliniques nerveux observés dans les CHUV. Le projet sera également étendu à d’autres espèces, demandant l’implication d’enseignants spécialisés, afin de parvenir à un consensus sur les définitions et d’enrichir l’illustration de la diversité des manifestations cliniques
Investigating control-centred results after uncentred principal component analysis
International audienceIn sensory evaluation, principal component analysis (PCA) is often used to explore differences between products. In some studies, there is one control product (e.g. a reference or benchmark) and many test products, where test-control paired differences are of primary interest. We discovered two equivalent ways to investigate these results using PCA. The first is a centred PCA of column-centred test-control paired comparisons, which includes both test-control paired differences and control-test paired differences. The second is an uncentred PCA of a control-centred matrix. We show why these two approaches are equivalent. We also show the truncated total bootstrap method, which is used to investigate uncertainty, yields equivalent results in both solutions. The uncentred PCA of a control-centred matrix is more computationally efficient and facilitates interpretations by situating the control product at the origin of score plots. The proposed methods are illustrated using a data set from a trained sensory panel for a control product and nine test product formulations

Ingénierie génétique de chlamydomonas reinhardtii pour une approche vers l'expression de protéines et l'assemblage de vecteurs viraux AAV
The microalgae C. reinhardtii is a model organism because its genome has been characterised and genetic tools developed. The production of recombinant proteins in microalgae is an emerging field, which is not yet as mature as the expression systems classically used, such as animal or bacterial cells. In particular, the literature remains unclear as to the genetic modifications that are actually required in the gene of interest in order to obtain optimal and stable expression of the desired protein. In the first part of this thesis, we determined the most favourable genetic combination for the expression of the recombinant mCherry protein model. This enabled the protein to be produced at a maximum level of 19 mg/L, the highest yield in the literature to date. The genetically modified microalgae were kept in liquid culture, which proved to be scalable.On the other hand, there are currently no publications reporting the expression of AAV-derived proteins in microalgae with the aim to produce recombinant AAV gene therapy viral vectors, Evauating the feasibility of AAV production in microalgae would provide a partial answer to the current challenges of AAV manufacturing. The second part of this thesis work is therefore exploratory and innovative with the aim. To produce in C. reinhardtii AAV2 main, viral proteins of large size and complex assembly, Rep and Cap. The work carried out in this thesis has led to the accumulation of new knowledge on the genetic engineering of microalgae for bioproduction of biotherapeutic products for human healthcare.La microalgue C. reinhardtii est un organisme modèle en raison de son génome qui a été caractérisé et dont les outils génétiques ont été développés. La production de protéines recombinantes dans les microalgues est un domaine émergent, encore peu mature par rapport aux systèmes d’expression classiquement utilisés comme les cellules animales ou bactériennes. En particulier, la littérature reste floue quant aux modifications génétiques réellement indispensables à réaliser dans le gène d’intérêt afin d’obtenir une expression optimale et stable de la protéine souhaitée. Ainsi la première partie de ce travail de thèse a permis de déterminer la combinaison génétique la plus favorable pour l’expression d’une protéine recombinante modèle, mCherry. Les séquences optimisées ont permis de produire cette protéine à un taux maximum de 19 mg/L, un rendement qui est à ce jour le plus élevé décrit dans la littérature. Les microalgues ainsi modifiées génétiquement ont été maintenues en culture liquide ce qui révèle une possible montée en échelle. D’autre part, à l’heure actuelle, il y a peu de données quant à la faisabilité de l’expression dans les microalgues de protéines complexes et de grandes tailles telles que celles dérivées des virus adéno-associées (AAV) dans le but de produire des vecteurs recombinants pour la thérapie génique. En effet, la bioproduction d’AAV dans les microalgues permettrait de répondre en partie aux enjeux actuels de bioproduction d’AAV. La deuxième partie de ce travail de thèse a ainsi eu un caractère exploratoire et innovant. L’objectif de produire, dans C. reinhardtii les protéines virales principales de l’AAV2 Rep et Cap dont l’assemblage est complexe. L’ensemble des travaux de cette thèse ont permis d’accumuler de nouvelles connaissances quant au génie génétique des microalgues à des fins de production de biothérapies en santé humaine
MEK inhibition prevents human skin graft rejection by promoting CD8+TCF1+ over CD8 effector T cells
International audiencePharmacological MEK inhibition might be an innovative approach to complete the immunosuppressive regimen that enables solid organ transplantation. While MEK inhibitors like trametinib are approved in oncology, their immunomodulatory properties remain poorly investigated in the context of organ transplantation, especially in human context. Using a human skin transplantation model in NSG mice reconstituted with third-party human PBMCs, we evaluated the effects of trametinib on graft survival and the human allogeneic immune response. MEK inhibition significantly prolonged graft survival without reducing graft infiltrate, while preserving the human epidermal tissue. Single-cell RNA sequencing of splenic cells revealed that MEK inhibition impaired CD8+ T cell differentiation into effector phenotypes, favoring an accumulation of CD8+ TCF1+ stem-like cells. Additionally, MEK inhibition supported CD4+ T cell homeostasis by maintaining IL-7R expression. These findings suggest that MEK inhibition may simultaneously control the alloimmune response and support immune recovery, highlighting its potential in solid organ transplantation
Study of the Relationship Between Electrical Conductivity, Batter–Crumb Transition, and Leavening Powders During the Ohmic Baking of Pound Cake
International audienceOhmic heating (OH) is a promising technology in the food industry, with electrical conductivity (EC) playing a crucial role in its effectiveness. This study aimed to correlate the evolution of EC during pound cake baking using OH with the degree of starch gelatinization (DSG) and assess the impact of two sodium acid pyrophosphate leavening acids—SAPP10 and SAPP40—on EC. Baking was conducted in a prototype OH cell, and EC was measured in two ways: during baking as a function of increasing temperature, and at specific baking times and temperatures at the cake center. The latter was measured using impedancemetry at room temperature and correlated with the DSG determined by differential scanning calorimetry. Key results showed a clear negative correlation between EC and the DSG in the center of the OH‐baked cake. Specifically, as the DSG increased from 4% at 60°C to about 48% at 76°C, EC decreased from 390 μS/cm to approximately 89 μS/cm, respectively. Importantly, the addition of different baking powders did not significantly affect EC evolution. These findings suggest that the relationship between EC and starch gelatinization in cakes is influenced by moisture content and ion mobility during baking. This study highlights the potential of EC monitoring as a tool to track internal changes in cakes during the OH process, offering valuable insights into how baking conditions can influence final product characteristics like texture and quality
Validation de la gastro-résistance de gélules fabriquées à partir du kit Appropharm™ à destination de transplantation de microbiote fécal
Les entéropathies inflammatoires chroniques du chien sont responsables de troubles digestifs invalidants, tant pour l’animal que pour son propriétaire. Elles sont fréquemment associées à une dysbiose intestinale. La transplantation de microbiote fécal (TMF) constitue une modalité thérapeutique de plus en plus utilisée. Elle consiste à administrer les selles d’un donneur sain afin de rétablir l’équilibre de la composition et du fonctionnement du microbiote intestinal du chien malade. Lorsque le transplant est administré par voie orale, le passage dans l’estomac peut compromettre la viabilité des bactéries ensemencées. La confection de gélules gastro-résistantes apparaît donc nécessaire afin de protéger le transplant de l’acidité gastrique. L’objectif de cette étude était d’évaluer la capacité des gélules confectionnées avec le kit Appropharm™ à franchir l’estomac et à s’ouvrir dans le duodénum sans exposition de leur contenu à l’acidité gastrique. Pour cela, un produit de contraste a été placé dans des gélules fabriquées selon le même protocole que celui utilisé pour la conception des gélules de TMF. Ces gélules ont été administrées à six chiens de gabarits différents, et leur transit digestif a été suivi par radiographie. Les résultats montrent que la quasi-totalité des gélules s’ouvrent prématurément dans l’estomac, quel que soit le chien, ce qui suggère une efficacité limitée de la gastro-résistance obtenue avec le kit Appropharm™. Ces observations amènent à s’interroger sur la pertinence de l’objectif de gastro-résistance dans ce contexte et, le cas échéant, sur les moyens à mettre en œuvre pour améliorer le franchissement pylorique
Etude retrospective de 2018 à 2024 des cas de Rhodococcose chez le poulain : facteurs de risque et facteurs pronostiques
La rhodococcose équine est une maladie infectieuse à tropisme respiratoire causée par Rhodococcus equi, qui affecte de nombreux élevages de manière endémique ou sporadique. Elle constitue un enjeu économique et sanitaire important en raison de son impact sur la survie des poulains et leur future carrière sportive. L’objectif de ce travail était d’analyser rétrospectivement les facteurs de risque et les facteurs pronostiques de la rhodococcose dans un élevage équin en Normandie entre 2018 et 2024, afin d’identifier des leviers de prévention ou d’amélioration de la prise en charge, adaptés aux réalités de terrain. L’étude a finalement été réalisée entre 2020 et 2024, et 220 poulains ont été inclus. Deux facteurs de risque ont été mis en évidence : la période de naissance (janvier-février et avril), en désaccord partiel avec la littérature, et l’utilisation d’une litière accumulée en 2023, en accord avec les données publiées. Le sexe des poulains, l’administration de plasma après un apport colostrale de mauvaise qualité, le colostrum de mauvaise qualité ou encore la densité de population n’ont pas donné de résultats significatifs dans cette étude. Le pronostic vital apparaît favorable, avec une mortalité inférieure à 1 % lorsque la prise en charge est précoce, en accord avec les études antérieures. En revanche, le pronostic sportif n’a pas pu être établi de manière significative, du fait du recul insuffisant et de limites méthodologiques. Ces résultats mettent en évidence l’importance d’une méthodologie standardisée incluant un suivi systématique des poulains et l’utilisation de la PCR. Il serait intéressant de considérer d’éventuelles co-infections, d’intégrer une analyse génétique et de constituer des cohortes plus larges, afin d’améliorer la compréhension et la maitrise de cette pathologie complexe
