Myeloid Neoplasms With Erythroid Predominance and Excess Blasts in Young Adults Exhibit Distinct Genetic Profiles
International audienceThe evolution of acute myeloid leukemia (AML) classifications has progressively shifted the diagnostic focus toward genetic criteria. Nevertheless, morphology remains a key element in clinical practice, often serving as the initial trigger for additional molecular investigations. The diagnosis of acute erythroleukemia (AEML), initially defined by the FAB group, is no longer recognized as a distinct entity in the latest WHO and ICC classifications. Some studies have indicated that AEML shares similarities with myelodysplastic neoplasms, including a high frequency of TP53 mutations and adverse karyotypes. Here, we conducted a retrospective analysis in adults with AEML defined using historical morphologic criteria (≥ 50% erythroid precursors and ≥ 20% blasts among non‐erythroid cells). In contrast to older patients, young adults (18–60 years) exhibit unique genetic profiles including a high prevalence of normal karyotypes (65%), NPM1 (35%) and UBTF (23%) mutations. AEML morphology in NPM1 ‐mutated cases did not impact clinical outcomes but was associated with specific molecular features, including an enrichment of WT1 and cohesin gene mutations. In this age group, our findings support that morphologically defined AEML often corresponds to AML according to current genetic criteria, consistent with recent classification systems that prioritize molecular features over morphology
On small analytic relations
International audienceWe study the class of analytic binary relations on Polish spaces, compared by the notions of continuous reducibility or injective continuous reducibility. In particular, we characterize when a locally countable Borel relation is E0 xi (or Pi 0 xi), for xi >= 3, by providing a concrete finite antichain basis. We give a similar characterization for arbitrary relations when xi = 1. When xi = 2, we provide a concrete antichain of size continuum made up of locally countable Borel relations minimal among non-E02 (or non-Pi 02) relations. The proof of this last result allows us to strengthen a result due to Baumgartner in topological Ramsey theory on the space of rational numbers. We prove that positive results hold when xi= 2 in the acyclic case. We give a general positive result in the not necessarily locally countable case, under another suitable acyclicity assumption. We provide a concrete finite antichain basis for the class of uncountable analytic relations. Finally, we deduce from our positive results some antichain basis for graphs, of small cardinality (most of the time 1 or 2)
Inclusive national innovation systems: rethinking institutions in the light of inclusion imperatives
SH1International audienceNational innovation systems (NISs) have been important in the literature since the 1990s for highlighting the institutional performance of economies and promoting economic development. Inclusion in systemic innovation activities is an emerging area of research. However, the definition of inclusion within innovative activities remains unclear and is associated with numerous forms and characteristics depending on the context visited. Our work highlights the conceptual gap that exists around the notion of inclusive innovation by characterising three forms of inclusion in relation to innovation activities. We thus set out, in the form of a typology, three distinct framings which enable us to identify three different levels associated with specific institutional mechanisms and forms of inclusion. This typology makes it possible to identify appropriate innovation policies, depending on how inclusive innovation is characterised (low, medium, and high). It also helps to clarify the inclusive nature of innovation in NIS approaches

MR-guided Laser Interstitial Thermal Therapy mesencephalotomy for medically intractable malignant pain
International audienceStereotactic mesencephalotomy is a neurosurgical technique designed to sever spinothalamic pain transmission pathways for medically intractable pain. This report presents the first case of Magnetic Resonance-guided Laser Interstitial Thermal Therapy (MRgLITT) mesencephalotomy for severe malignant pain due to metastatic melanoma. The procedure significantly reduced the patient's pain, with a postoperative visual analog scale (VAS) score decreasing from > 7 to < 3. No adverse effects were observed. The case underscores the potential of MRgLITT mesencephalotomy as a precise, minimally invasive option for pain management in palliative care settings
Intranasal spike and nucleoprotein fusion protein-based vaccine provides cross-protection and reduced transmission against SARS-CoV-2 variants

Next-generation vaccines are needed for broader protection and blocking virus transmission. Here, we rationally designed an original nasal subunit vaccine composed of a fusion protein (SwFN) made of Wuhan spike and nucleoprotein combined with biocompatible mucosal nanocarriers (Nc). In mouse model, the nasal Nc-SwFN vaccine elicited multivalent serum and mucosal neutralizing antibodies. Robust spike and nucleoprotein cross-reactive immunity against variants was induced with a predominant phenotype of resident memory T cells in the lungs. Moreover, Nc-SwFN led to protective responses against Wuhan and Delta infection in relevant models with an absence of morbidity, mortality, and virus dissemination in the lungs and brain. Finally, Nc-SwFN drastically reduced host-to-host transmission. These promising results underscore the advantages of the nasal Nc-SwFN approach as a broad-spectrum vaccine candidate against current and emerging SARS-CoV-2 variants
Faut-il protéger les nourrissons des ondes radiofréquences ?
International audienceCes dernières années, l’usage des technologies sans fil s’est intensifié, exposant même les populations les plus jeunes comme les enfants aux ondes radiofréquences (RF) de manière fréquente, précoce et souvent passive via leur environnement.L’Agence nationale de sécurité sanitaire de l’alimentation, de l’environnement et du travail (Anses) a publié plusieurs rapports sur ce sujet, notamment en 2013 et 2016 [1], [2], soulignant que les enfants sont plus vulnérables que les adultes en raison de leurs spécificités morphologiques et physiologiques ainsi que leur durée d’exposition prolongée. Cette vulnérabilité est particulièrement marquée au cours des premières années de vie, du fait de l’immaturité biologique et de la croissance rapide. C’est dans ce contexte que cet article se propose d’explorer les effets des RF sur les jeunes enfants, en se concentrant particulièrement sur la période de la naissance à 3 ans.Malgré ces préoccupations, peu d’études se concentrent sur l’impact des RF envers les nourrissons. La majorité des recherches considère que l’âge de la première utilisation du téléphone mobile se situe rarement avant 7 ans, bien que 5,5 % des enfants déclaraient, dès 2013, l’avoir utilisé avant cet âge [3]. Pourtant, l’exposition précoce aux RF augmente avec la démocratisation de l’usage des appareils et équipements communiquant à distance, par exemple la multiplication des jouets connectés ou l’ubiquité environnementale des réseaux de la téléphonie mobile. Cette situation interroge sur les effets potentiels des RF et la nécessité d’une protection spécifique des nourrissons. Ainsi cet article s’appuie sur le rapport de l’Anses publié en 2016 [2] sur la « radiofréquence et santé des enfants » pour examiner les implications de ces effets sur la santé infantile et la nécessité d’une protection accrue
Intensity vs. Volume in Professional Soccer: Comparing Congested and Non-Congested Periods in Competitive and Training Contexts Using Worst-Case Scenarios
International audienceBackground: Understanding the balance between intensity and volume during training and competition is crucial for optimizing players' performance and recovery in professional soccer. While worst-case scenarios (WCSs) are commonly used to assess peak match demands, little is known about how the time spent within WCS thresholds varies across congested and non-congested periods, especially when considering differences in playing time. This study examines the time spent at different percentages of WCSs during congested and non-congested periods for players with lower and higher playing times throughout training sessions and matches. Methods: Data were collected from a professional soccer team across a congested and non-congested match period. Twenty players were divided into two groups based on playing time: the top 10 playing times (PT 1-10) and the bottom 10 playing times (PT 11-20). WCS thresholds for total distance (TD) and the distance covered above 20 km•h -1 (D20) were quantified in 10% increments, starting from 50% and increasing up to >100%. The time spent at each threshold was compared between periods and groups for the integrated soccer exercises performed during all training sessions. Repeated measures of ANOVA were used to analyze differences between playing time groups and periods. Results: During training, players spent significantly more time within the 50-90% WCS TD and WCS D20 thresholds during non-congested periods compared to congested periods (p < 0.05). However, no significant differences were observed in the time spent for >90% of the WCSs between periods (p > 0.05). Both PT 1-10 and PT 11-20 groups exhibited similar patterns of WCS achievement, with small effect sizes observed for a few indicators. Conclusion: Coaches should design training sessions that replicate or exceed match demands, particularly during non-congested periods. Future strategies should integrate larger-sided games with longer durations and dissociated contents to better individualize and optimize training loads, especially for non-starters.</div

Quantification of Urine and Plasma Levels of Extracellular Vesicles in a Cohort of Kidney Transplant Recipients and Chronic Kidney Disease Patients
International audienceExtracellular vesicles (EVs) have a key role in intercellular communication. We hypothesized that EVs are biomarkers of nephropathy or kidney allograft rejection. We screened patients with chronic kidney disease (CKD) and kidney transplant (KT) recipients. We measured the urine and plasma levels of total EVs overall and EV subpopulations (positive for podocalyxin, aquaporin-1, CD133, CD144, CD19, CD3, CD16, CD56, or CD41). We included 92 patients with CKD, 70 KT recipients, and 33 healthy volunteers. In CKD, the total urine EV concentration was correlated positively with the estimated glomerular filtration rate (eGFR), but none of the subpopulations was identified as a potential biomarker of nephropathy. Among the KT recipients, 30 had good allograft function and 40 had allograft disease (13 with antibody-mediated rejections (ABMR), 12 with T-cell-mediated rejection (TCMR), and 15 with allograft dysfunction). Patients with ABMR had low plasma levels of EVs derived from B-cells, T-cells, and endothelium (p = 0.003, 0.009, and 0.005, respectively). Patients with TCMR had a low urine level of EVs derived from endothelium (p = 0.05). EVs derived from B-cells, T-cells, and endothelium might be biomarkers of kidney allograft rejection. However, we did not identify biomarkers of nephropathy in CKD
