Postoperative opioid-free analgesia in dogs undergoing tibial plateau leveling osteotomy: a feasibility study
3International audienceObjectives: This study was designed to prospectively evaluate the feasibility of an opioid-free anesthesia protocol and describe the quality of recovery and management of postoperative analgesia in dogs after a tibial plateau leveling osteotomy (TPLO).Methods: In total, 20 dogs presented for TPLO were included. After premedication with intravenous (IV) medetomidine (0.005-0.007 mg/kg) and midazolam (0.2 mg/kg), the dogs were anesthetized using ketamine (2 mg/kg) and propofol and maintained with isoflurane and ketamine CRI (0.6 mg/kg/h). Sciatic and femoral nerve blocks were performed with bupivacaine 0.5% (0.087 +/-0.01 and 0.09 +/-0.02 mL/kg, respectively). Meloxicam (0.2 mg/kg IV) was administered intraoperatively, after osteotomy. Fentanyl (0.002 mg/kg IV) was administered intraoperatively, as rescue analgesia in the case of sustained increase in cardiorespiratory variables. Two pain scores (French 4A-VET and Glasgow short form) were performed at conscious sternal recumbency and 2, 4, 6, 8, 12, and 20 h after extubation and compared to baseline using a Friedman test followed by a Nemenyi post-hoc test. The time taken for the first food intake and urination was reported.Results: Intraoperative opioid-free anesthesia was feasible in 11 dogs, whereas 9 dogs received fentanyl once during arthrotomy. No opioid postoperative rescue analgesia was required. Food intake occurred within 6 h, and all dogs were discharged after 24 h without any complication.Conclusion: Total opioid-free postoperative analgesia was achieved in all dogs, with adequate recoveries. Although opioid-free anesthesia was feasible in 55% of the population, a single dose of fentanyl was necessary in 45% of the dogs during arthrotomy

Dynamic combinatorial chemistry for the multiplexed identification of glyco-dyn[n]arenes in an anti-adhesive strategy against Pseudomonas aeruginosa
Carbohydrate-protein interactions are of prime importance in cell-cell communication, signal transduction, cancer, bacterial or viral infection. Chemists have designed multivalent systems to mimick these recognition phenomena and provide potent ligands of these proteins with foreseen therapeutic applications. Dynamic combinatorial chemistry provides access to a library of chemical species in equilibrium through reversible covalent bonds. This strategy can be readily applied to the rapid and efficient identification of multivalent glycoclusters by introducing a protein into the equilibrating library for the selection of the fittest glycocluster for this protein. 1,4-Dithiophenols conjugated to monosaccharides were equilibrated into dynamic combinatorial libraries providing a diverse mixture of glycoclusters. Selection of the best ligand for different lectins (ConA, LecA and LecB from Pseudomonas aeruginosa) could increase the concentration of glyco-dyn[3]arenes and glyco-dyn[4]arenes. A key aspect of this strategy is that multiplexing can be readily achieved by using two building blocks (galactosylated and fucosylated 1,4-dithiophenols) to interrogate several lectins at once in a single experiment. These macrocyclic glycoclusters could be synthesized, isolated, then evaluated as ligands of the lectins and displayed nanomolar dissociation constants. Furthermore, while no toxicity could be detected against human cells or bacteria, their evaluation as anti-adhesive agents could be confirmed through a virulence assay on human A549 lung epithelial cells

Embodied collaborations: Using virtual reality and spatialized sound to share sensible experience
International audienceThis article proposes to plunge into reactivated experience : a methodology designed to locate and use new paths of mediation to share an embodied and sensorial experiences and discourses. This way of doing collaborative research involves an immersive apparatus which produces images and sounds broadcasted through a virtual reality headset. It plays on the trouble created by these mediations and their in‐motion constitution in order to produce a reflexive discussion about mundane experiences. It is part of a broader research project about how inhabitants of Lyon and Madrid's districts endure heat waves in urban context
Méthodologies d'aide à la décision chez le patient traité par anticoagulant oral dans une maladie thrombo-embolique à partir d'études observationnelles
The long-term aim of this thesis is to provide physicians with a strategy for estimating the risk of haemorrhage in 2 types of population: those with atrial fibrillation and those with venous thromboembolism. This strategy is based on statistical evaluation models derived from cohort studies. The first part of this thesis was devoted to a review of the literature on the pathophysiological mechanisms surrounding thrombosis, embolism and atherosclerosis. The modes of action and evidence of clinical efficacy of oral anticoagulants were researched, analysed and compared. The results of the SCORE study conclude this first part. The second part focused on validating the use of haemorrhagic risk scores in a cohort of primary care patients, and then exploring other previously unidentified risk factors (CACAO study). The third part aimed to determine the role of Covid-19 in the occurrence of thromboembolic events, based on an ancillary study of a randomized controlled trial in primary care.Les travaux de cette thèse ont pour but, à long terme, de mettre à disposition du médecin une stratégie d’estimation du risque hémorragique chez 2 types de population, celle atteinte par une fibrillation atriale et celle porteuse d’une thromboembolie veineuse. Cette stratégie se base sur des modèles d’évaluation statistique à partir d’études de cohorte. La première partie de cette thèse a été consacrée à une revue de littérature concernant les mécanismes physiopathologiques entourant la thrombose, l’embolie, l’athérosclérose. Les modalités d’actions et les preuves d’efficacité clinique des anticoagulants oraux ont été recherchées, analysées et comparées entre eux. Les résultats de l’étude SCORE concluent cette première partie. La seconde partie s’est attachée à valider l’utilisation de scores de risques hémorragiques sur une cohorte de patients en soins primaires puis d’explorer d’autres facteurs de risque non identifiés jusqu’alors (Etude CACAO). La troisième partie visait à déterminer quelle était la place de la Covid-19 dans la survenue des évènements thromboemboliques à partir de l’analyse ancillaire d’un essai contrôlé randomisé en soins primaires
Remote islands as natural laboratories: human–food association increases attraction to humans and novelty exploration in a seabird
1/3International audienceIncreased attraction to humans and their objects often arises after repeated and positive human–wildlife encounters (e.g. food provided in tourist settings). The causes of this ‘over-attraction’, which may result from a learned association between humans and food, are still poorly studied in wild animals. Understanding the influence of humans on animals’ responses is yet crucial to prevent negative effects (e.g. aggression). We presented three novel objects to two groups of free-ranging brown skuas ( Catharacta antarctica ssp. lonnbergi ) in the remote sub-Antarctic, where their habitats show no or minimal human disturbance. Skuas in one group (Verte) had previously participated in repeated food-rewarded behavioural and cognitive tasks with a human experimenter; skuas in the other group (Ratmanoff) had never done so. Objects consisted of (i) one natural-food-resembling object (plastic fish), (ii) one anthropogenic food object (real cake slice), and (iii) one anthropogenic non-food object (yellow glove). Verte group skuas approached the human experimenter and pecked significantly more and sooner at novel objects. Human–food association may have thus resulted in increased attraction to humans and novelty exploration in previously naive brown skuas, making this species a useful model for investigating the consequences of experience with humans on wildlife behaviour
